Murray, Lydia Soraya. "Dissecting the mechanisms of disease of COL4A1 and COL4A2 mutations." Thesis, University of Glasgow, 2014. http://theses.gla.ac.uk/5143/.

Full text
Abstract:
The best characterised collagen IV disease is Alport syndrome, caused by α3.α4.α5(IV) mutations. Mutations affecting α1(IV)2α2(IV) can cause diseases, such as haemorrhagic stroke, aneurysm, haematuria, proteinuria, cataracts, vascular tortuosity and iris hypoplasia. Recent data has shown that α and COL4A2 mutations may be more common in the population than previously anticipated. However, the mechanisms of disease of all collagen IV mutations are poorly understood. The aim of this thesis was to investigate the disease mechanism in a selection of COL4A1, COL4A2 and COL4A5 mutations. Collagen IV is folded in the endoplasmic reticulum (ER) then secreted to provide the major structural component of the basement membrane (BM). As such, a combination of ex situ biopsies, in vitro cell cultures and an in vivo mouse model provided an excellent platform for investigating the intracellular and extracellular effects of mutant trimers. Primary dermal fibroblasts containing COL4A2 and COL4A5 mutations, and a Col4a1 mouse model were treated with the FDA approved chemical chaperone phenyl-4-butyric acid (PBA) to determine its effects on intracellular ER retention of trimers (which can activate ER stress), extracellular trimer incorporation into the BM, and disease phenotypes. In vitro and ex situ investigation of a COL4A2 mutation suggested that intracellular α1(IV)2α2(IV) accumulation and ER stress associated with severe BM defects and the disease. Intracellular accumulation and ER stress were reduced by PBA treatment, and treatment of a Col4a1 mouse model displayed elevated α1(IV)2α2(IV) incorporation onto the BM. In vivo PBA treatment did not exacerbate any defects, rescued some disease phenotypes, and had no effect on others. In addition, one of five COL4A5 mutations analysed in vitro displayed intracellular α3.α4.α5(IV) accumulation and ER stress that were ameliorated by PBA. Collectively these data suggest that ER stress and/or reduced α1(IV)2α2(IV) BM incorporation underlie some collagen IV pathologies, and provide novel evidence that they may be amenable to chemical chaperone therapy. More investigation is now needed to further dissect the relative contributions of ER stress and BM defects to collagen IV diseases.

Turner, Adam. "Functional Interrogation of the COL4A1/COL4A2 and SMAD3 Coronary Artery Disease Loci." Thesis, Université d'Ottawa / University of Ottawa, 2016. http://hdl.handle.net/10393/35248.

Full text
Abstract:
Coronary artery disease (CAD) is the leading cause of death worldwide caused by a complex array of environmental and genetic factors. In recent years, the CARDIoGRAM Consortium has identified dozens of novel CAD-associated loci, including COL4A1/COL4A2 and SMAD3. The goals of this project were two-fold. First, to identify functional CAD-associated SNPs (single nucleotide polymorphisms) at these loci and investigate how these common polymorphisms alter CAD risk. Second, to determine if the COL4A1/COL4A2 and SMAD3 loci display both biological interaction and statistical interaction in the context of atherosclerosis. COL4A1 and COL4A2 are critical components of vascular basement membranes and have many additional roles in the vessel wall. SMAD3 is a transcription factor that is a key mediator in the canonical TGFβ signaling pathway. In the first section of this thesis, I show that COL4A1/COL4A2 and SMAD3 display biological interaction in that the TGFβ-mediated upregulation of COL4A1 and COL4A2 is dependent on SMAD3. Furthermore, we were able to identify a COL4A2-SMAD3 SNP pair that displayed a highly significant statistical interaction for CAD association, highlighting that perturbations of the SMAD3/type IV collagen signaling axis contribute to the pathogenesis of atherosclerosis. The second section details the characterization of a novel functional CAD-associated SNP at the SMAD3 locus. The rs17293632 SNP, highly linked to the rs56062135 index SNP reported by CARDIoGRAM, disrupts a conserved AP-1 binding site in intron 1 of the SMAD3 gene. rs17293632 lies within a strong enhancer in many cell types, including arterial smooth muscle cells. The minor, protective allele (T) at rs17293632 disrupts binding of AP-1 proteins, lowers the activity of this enhancer and lowers SMAD3 mRNA levels in humans in both whole blood and carotid plaque tissue. Altogether, AP-1 regulation of SMAD3 enhancer activity suggests a novel regulatory mechanism relevant to the pathogenesis of CAD. The third section summarizes efforts to characterize the mechanisms whereby the independent rs4773144 and rs9515203 SNPs at the COL4A1/COL4A2 locus associate highly with CAD. We were unable to identify causal SNPs at the COL4A1/COL4A2 locus, highlighting the challenges of post-GWAS characterization of some CAD-associated loci. The experiments with SNPs at COL4A1/COL4A2 demonstrate the challenges in defining mechanisms whereby noncoding DNA variants can lead to common disease.

Pihlajamaa, T. (Tero). "New tools for the study of an old collagen:characterization of the human COL9A1, COL9A2 and COL9A3 genes and production of human type IX collagen as a recombinant protein." Doctoral thesis, University of Oulu, 2000. http://urn.fi/urn:isbn:9514257359.

Full text
Abstract:
Abstract Type IX collagen is a quantitatively minor component of cartilage collagen fibrils. Although a few mutations have been associated with multiple epiphyseal dysplasia, recent evidence suggests involvement of type IX collagen in a wider spectrum of phenotypes. The functional role of this molecule remains undetermined, in part due to difficulties in obtaining high amounts of intact protein. To facilitate more efficient mutation screening and comparison of the genomic organization of the human genes encoding the α1(IX), α2(IX) and α3(IX) polypeptides, their genomic structures were characterized. Complete nucleotide sequences were determined for the COL9A2 and COL9A3 genes along with sequences for all the exon boundaries in the COL9A1 gene. Putative transcription control elements were identified and the alternative promoter region was characterized in the human and mouse COL9A1 genes. Mutation screening was performed for the COL9A3 gene and two apparently neutral 9-bp deletions within the COL1 domain were identified. These are the first deletions within a triple-helical domain of any collagen that are not associated with a disease phenotype. An insect cell expression system with an exogenous source of prolyl 4-hydroxylase was used to produce heterotrimeric human type IX collagen. The recombinant protein consisted of the three a chains in a 1:1:1 ratio and showed correct folding and high thermal stability. Up to 10 mg of secreted protein could be purified from a litre of culture medium. The expression system was used to analyze the chain association of type IX collagen in cellulo. Although the chains are capable of homotrimerization, a preference for heterotrimer formation was noted. The neutral deletion was characterized further using the insect cell system. Mutant α3(IX) chains carrying a deletion of one Gly-X-Y triplet were shown to form correctly folded heterotrimers with the wild-type α1(IX) and α2(IX) chains. The results suggest a function for the NC2 domain in neutralizing the effect of the deletion. This work provides a novel means for the analysis of type IX collagen mutations and their protein-level effects, and should enable future studies to be made of the structure-function relationship in type IX collagen.

Grigorova, Fialka. "Characterization of the Role Nuclear Bmp2 (nBmp2) Plays in Regulating Gene Expression." BYU ScholarsArchive, 2011. https://scholarsarchive.byu.edu/etd/3196.

Full text
Abstract:
The nBmp2 protein was first identified in a DNA affinity chromatography/mass spectrometry screen designed to detect proteins that interact with a cartilage-specific enhancer element (called D/E) from the type XI collagen gene Col11a2. The transcription factor SOX9, a protein from the Sox (SRY-related HMG box) family, binds to and activates gene expression from this enhancer. nBmp2 has no transcriptional activity of its own on this enhancer, but when co-transfected with SOX9 it increases SOX9's activation of D/E nearly 2-fold. SOX9 also activates cartilage-specific enhancer elements from the Col2a1, Col27a1, and Col9a1 genes. The purpose of this project was to determine 1) whether nBmp2 similarly effects SOX9-dependent expression from these enhancers, and 2) whether it does so by binding (either directly or indirectly) to the Col2a1, Col27a1, and Col9a1 enhancers. The work described in this thesis has shown that nBmp2 increases luciferase levels produced from three enhancer/reporter plasmids, but it does so without binding directly to the enhancers. This work has opened up a new area of exploration into the function of the novel protein nBmp2 to examine its potential effects on a variety of different gene regulatory processes.

Bethune, Richard Alan. "Profiling white-collar criminals : what is white-collar crime, who perpetrates it and why?" Thesis, University of Edinburgh, 2015. http://hdl.handle.net/1842/10609.

Full text
Abstract:
Following a period of resurgence in academic interest in the subject over the last 30-40 years, white-collar crime has found greater prominence within criminology. Efforts over this period have however failed to produce a single satisfactory and agreed-upon definition, a consistent and coherent body of research, and a single theory which can account for all forms of white-collar crime. This thesis aims to address certain shortcomings in the current state of white-collar crime theory and understanding. Part 1) addresses the issues of both conceptual definition of white-collar crime and specific behaviours as proscribed within the Legislation. Part 2) examines current criminological theory and research on individual differences (arguably the biggest gap in current knowledge in the area of white-collar crime); it examines the origins and current state of offender profiling in crime prevention, before Part 3) presents original research on establishing offence-specific white-collar criminal profiles based on demographic, sociological, psychological, organisational and motivational factors. Part 4) examines why certain individuals may perpetrate certain crimes in certain situations, beginning with a review of those few white-collar crime specific theories that do exist, before reviewing traditional sociological theories and attempting to apply them to white-collar crime; finally in Part 5) a new conceptual framework for white-collar crime is presented, which is referred to as the theory of ‘Differential Assimilation’. I bring together each of these chapters and situate the thesis within current research and literature, summarising how it engages and contributes to the field of white-collar crime. I include suggestions for the practical application of certain white-collar crime prevention techniques within organisations.

Feesey, Terrence James. "An investigation of variables influencing the experience of unemployment for blue collar and white collar workers." Thesis, University of British Columbia, 1987. http://hdl.handle.net/2429/26811.

Abstract:
This study was designed to probe the experience of white collar unemployment. Some research results suggest that white collar people have an easier time with unemployment than do blue collar people while other findings suggest the contrary. A questionnaire format instrument was designed to record self-reported changes of an affective and behavioural nature in a sample of 66 white collar and 24 blue collar unemployed adults. It was hypothesized that on the whole, the blue collar sample would report a more difficult response to unemployment than the white collar sample. It was further hypothesized that after an unspecified period of time the unemployed white collar sample would become passive and depressed. Twelve variables focusing on learned helplessness, self-esteem, depression, locus of control, social interaction, time structure, personal meaning and perceived measures of health and finances were recorded and intercorrelated in this relationship study. Correlation matrices were constructed for the general sample, the white collar and the blue collar sub-samples. Reliability and validity coefficients of the instrument were calculated on each variable and were found to be acceptable for the purpose of this study. The relationships among the variables supported the notion that generally, the people in the blue collar unemployed sample experienced more difficulty with unemployment than did those people in the white collar sample. The white collar sample subjects did not, however, show a significant disposition toward passivity and depression as a function of time. Instead, the data suggested the presence of a second white collar subgroup who appeared to be experiencing great personal difficulties regardless of the duration of their unemployment. It was suggested that the appearance of a bi-modal white collar sample was the result of the sampling technique, and further that these results may reflect the state of the real world. This position is offered as a possible justification for the contradictory white collar unemployment findings in the past.<br>Education, Faculty of<br>Educational and Counselling Psychology, and Special Education (ECPS), Department of<br>Graduate

APRS was selected as the protocol for position, transmission, and recovery. This allows users to benefit from any existing APRS enabled devices. The beacon was designed by attempting to optimize operational longevity and minimize size. Consequently, the system is implemented on a single board and enclosed for protection. As the system must be comfortable for the mammal, it was manufactured from lightweight components and enclosed in a plastic housing. To attach the case to the mammal, it is connected to a flexible, zig-zag, wearable antenna, which functions as a collar.

Rannikmäe, Kristiina. "Genetic associations with sporadic cerebral small vessel disease." Thesis, University of Edinburgh, 2017. http://hdl.handle.net/1842/23585.

Full text
Abstract:
Background: Cerebral small vessel disease (SVD) causes substantial cognitive, psychiatric and physical disabilities. Despite its common nature, SVD pathogenesis and molecular mechanisms remain poorly understood, and prevention and treatment are probably suboptimal. Identifying the genetic determinants of SVD will improve understanding and may help identify novel treatment targets. The aim of this thesis is to better understand genetic associations with SVD through investigating its pathological, radiological and clinical phenotypes. Methods: To unravel the genetic associations with SVD, I used three complementary approaches. First, I performed a systematic review looking at existing intracerebral haemorrhage (ICH) classification systems and their reliability, to help inform future studies of ICH genetics. Second, I performed a series of systematic reviews and meta-analyses, investigating associations between genetic polymorphisms and histopathologically confirmed cerebral amyloid angiopathy (CAA). Third, I performed meta-analyses of existing genome-wide datasets to determine associations of >1000 common single nucleotide polymorphisms (SNP) in the COL4A1/COL4A2 genomic region with clinico-radiological SVD phenotypes: ICH and its subtypes, ischaemic stroke and its subtypes, and white matter hyperintensities. Results: The reliability of existing ICH classification systems appeared excellent in eight studies conducted in specialist centres with experienced raters, although these existing systems have several limitations. In my systematic evaluation of CAA genetics, meta-analyses of 24 studies including 3520 participants showed robust evidence for a dose-dependent association between APOE ɛ4 and histopathological CAA. There was, however, no convincing association between APOE ɛ2 and presence of CAA in a meta-analysis of 11 studies including 1640 participants. Meta-analyses of five studies including 497 participants showed, contrary to an existing popular hypothesis, that while APOE 4 may increase the risk of developing severe CAA vasculopathy, there is no clear evidence to support a role of ɛ2. There were few data about the role of APOE in hereditary CAA, but in the three studies that had looked at this, there was no evidence for an association between APOE ɛ4 and CAA severity. There were too few studies and participants to draw firm conclusions about the effect of non-APOE ε2/ε3/ε4 genetic polymorphisms on CAA, but there were positive associations with TGF-β1, TOMM40 and CR1 genes in four studies. Finally, in my meta-analyses of the COL4A1/COL4A2 genomic region, three intronic SNPs in COL4A2 were associated with SVD phenotypes: significantly with deep ICH, and suggestively with lacunar ischaemic stroke and WMH. Conclusions: I have shown that while existing ICH classification systems appear to have very good reliability, further research is needed to determine their performance in different settings. For large population-based prospective studies of ICH genetics, anatomical systems are likely to be more feasible, scalable and appropriate, although they have limitations and will need to be further developed. Using systematic reviews and meta-analyses, I have confirmed a dose-related association between APOE ɛ4 and histopathological CAA, but also demonstrated that, despite popular acceptance, there is insufficient data to draw firm conclusions about the association with APOE ɛ2. I found some positive associations with CAA in other genes, which merit replication in further larger studies, and showed that there is currently insufficient data about the role of APOE in hereditary CAA. Finally, I identified a novel association between a locus in a known hereditary SVD gene – COL4A2 – and sporadic SVD. This highlights a new and successful approach for selecting candidate genes and can be expanded in future studies to include other known hereditary SVD genes.

Abstract:
Psychopathy has become a highly researched personality disorder in order to better understand criminal and violent behavior (Hare, 1993). Measures of psychopathy have proven to be useful tools in predicting outcomes of institutionalized populations by predicting future dangerousness (Hare, 1999). While several experts in the field of psychopathy allude to the idea of the successful psychopath and their presence in the corporate world (Hare, 1993; Babiak & Hare, 2006), very little research has been done in this area. The current study builds upon the small amount of empirical research by testing hypotheses regarding the relationship between psychopathic personality traits and intentions to engage in white collar crime. Using a sample of 181 university students, psychopathic personality traits were measured using the Psychopathic Personality Inventory - Revised (PPI-R). In addition, scales were developed to measure attitudes toward white collar offending and vignettes were constructed to measures intentions to engage in white collar crime. Four relationships are of primary focus: 1.) Do psychopathic personality traits account for variability in attitudes toward white collar crime?; 2.) Do attitudes toward white collar crime correlate with intentions to engage in white collar crime?; 3.) Are psychopathic personality traits related to intentions to offend and?; 4.) Do attitudes toward offending mediate the relationship between psychopathy and intentions to offend? A major finding is that the Self-Centered Impulsivity factor of the PPI-R accounts for a significant amount of variance in intentions to engage in white collar crime and environmental crime. Additional relationships between psychopathy, attitudes, and intentions are also discussed.

Abstract:
Thesis (MScEng)--Stellenbosch University, 2012.<br>ENGLISH ABSTRACT: This project targeted the development of a new tracking collar, trap telemetry system and supporting infrastructure, to aid researchers from the Cape Leopard Trust. Previously used collar products had all proven insu cient in some capacity and remote monitoring of trap sites was also required. Tracking collars are used to identify the movement patterns of the leopards and through the resulting research, assist in protecting this threatened species. In the development of the tracking collar and trap telemetry system, a high level system design was rst formulated, identifying major components that would be required. Alternative methods for implementation were then considered and the most optimal chosen. Two di erent modes of communication with the collar were envisioned and designed for. These would be used to transmit logged coordinates obtained from a GPS receiver back to researchers. A VHF terrestrial radio link was investigated, but an Iridium Satellite based solution was ultimately selected. An Iridium Satellite communications system was also used for transfer of trap state data. Ultimately, a working trap telemetry system was delivered for use by researchers. The tracking collar system had progressed to a working prototype, requiring miniaturisation and packaging before deployment. A possible packaging solution was also identi ed. The trap telemetry system, although displaying certain de ciencies, provided a capability previously unavailable to researchers. With further development, there is potential for the tracking collar to provide accurate satellite tracking and communications in a mass and price combination not previously available.<br>AFRIKAANSE OPSOMMING: Hierdie projek het as doel die ontwikkeling van 'n nuwe opsporingshalsband, 'n lokval telemetriese stelsel en die nodige ondersteunende infrastruktuur daarvoor. Abstract:
This thesis will handle the gendered wage gap in the blue-collar sectors and how the blue-collar unions collective organization (from here on referred to as LO) has handled this issue. In the Swedish labour market the unions and the employers’ organizations negotiate terms and conditions of employment that result in legally binding collective agreements. Within the blue-collar sector wages are to a large extent negotiated on this level and the blue-collar unions coordinate within LO on which questions to focus on in the negotiations with the employers. But during 2015 the unions within LO were not able to agree and the coordination failed (Medlingsinstitutet, MI, 2017). The main issue was disagreement on how to handle low wages and value discrimination. To understand the split during 2015, the reader needs to understand the background and the deeply rooted norms within LO and how the Swedish labour market is organized. Therefore this thesis is divided into three parts. The first part describes historical aspects of how the Swedish labour market has developed with focus on women’s salaries. The second part looks at how LO has chosen to handle the gendered wage gap during the years 2004-2012. The last part will be a critical discourse analysis of articles handling the failed coordination in 2015.

Abstract:
This exploratory research examines the wage effects of a variety of measures thought to be important in the employment income determination process. It does this by using ordinary least squares multiple regression techniques to construct wage equations for two samples of blue-collar workers in Montreal. Blue-collar wage equations are constructed for: (i) the Montreal labour market and (ii) the Montreal food processing industry.<br>The results of this study show that Montreal blue-collar wages are determined by a variety of factors. Most importantly, however, are the wage effects exhibited by a worker's gender, union membership status and whether or not a worker has received a promotion from his/her present employer. These factors show statistically significant effects on the wages of blue-collar workers in the Montreal labour market as well as on the wages of blue-collar workers in the Montreal food processing industry. The present research also finds that certain factors have distinct wage effects on the different samples of blue-collar workers examined. Data from the present study indicates that the wages of Montreal blue-collar workers are influenced primarily by: (i) on-the-job training, (ii) promotions received, (iii) job opportunities available, (iv) an interruption in one's studies, (v) trade union membership and (vi) gender. While the wages of blue-collar workers employed specifically in the Montreal-area food processing industry are affected principally by: (i) the use of computers, (ii) the use of machines, (iii) promotions received, (iv) a varied work experience, (v) trade union membership, (vi) marital status and (vii) gender. Abstract:
This thesis explores the interactions between women and men who work in highly-skilled blue-collar trades. The aim of this research is to describe women's perceptions and responses to their on-the-job communicative interactions with male co-workers, supervisors and union officials. small focus groups were conducted to produce rich narrative data that was audio recorded for later use by the researcher. The researcher met with the four subjects for three sessions. The interviews lasted three hours each. The researcher also conducted follow-up interviews by phone to clarify subjects' responses. The subjects were provided with an interview schedule of questions prior to the interview. This thesis seeks to identify women's perceptions of male and female differences in communication, perceived problematic communicative interactions and women's responses to perceived differences. This thesis also explores the possible correlation between women's sense of self-esteem and interactions with males on the job. Finally, subjects were interviewed to determine what strategies, if any, are used by women to work more effectively in a predominately male work environment. It was found that this sample of women reported several perceived differences between male and female communication styles and that some differences are problematic. The subjects reported that difficult interactions may result in feelings of anger, frustration, anxiety, hostility or sadness. Finally, the subjects offered several strategies for coping in nontraditional jobs.

Abstract:
One person - one vote; on Election Day in a representative democracy, everyone with the right to vote is equal. However, this occurs in Sweden once every four years. Between those occasions, we cannot be certain that political influence is equal until this has been tested. In this thesis, I will investigate whether Swedish democracy is responding to blue- and white-collar families equally. Do they have equal influence over public policy? And what consequences do the result have for some theories of democracy? With quantitative research, by replicating large parts of the research done by Martin Gilens and his research team, this thesis will be able to test if political influence is equal between these two groups. This allows us to test some of the theories and earlier research conclusions of Swedish democracy. No one, that I am aware of, has published something regarding this type of research in Sweden before. Gilens &amp; Page’s (2014) research found that in the United States, economic elites and interest groups had substantial influence on public policy whereas average citizens had little or none - confirming economic elite domination and biased pluralism theories. By analysing 182 policy proposals, I find no clear proof, but strong indications that white- collar families have more influence on policy-making than blue-collar families. Future research would most likely be able to draw more confident conclusions if there was a greater data sample. If these indications are correct, then this has implications for some theories of American and Swedish democracy.

Abstract:
The purpose of this study was to examine if blue- and white collar workers differ in self-rated job-satifaction  and  also to find out if job-satisfacaction can be predicted by gender, age, period of employment and union affiliation. The study was performed at a state university in southern Sweden. The sample consisted of 80 employees from four different departments, the accounting department, the department of human reosurces, the IT department and the service department. Of these were 26 blue collar-workers and 54 white collar-workers based on union affiliation. The questionnaire was about psychosocial work environment (PAK) and is based on the test of Sigvard Rubenowitz. The test measures job-satisfaction by five different scales, self-control, work management, working community, work incentive and work load. Statistical analysis with t-test and multiple regression analysis revealed a significant difference between blue collar workers and white collar workers where white collar-workers felt more work incentive, more self-control and felt a higher work load. The result also revealed that period of employment was a significant predictor for work load and age was a significant predictor for work management and work incentive.

Abstract:
Gottfredson and Hirschi's general theory of crime (1990) has received a great deal of empirical examination in the criminology, yet the application of this theory to white-collar crime offenders has not received a great deal of attention. Research that has been conducted in the realm of white-collar crime has yielded mixed support for low self-control in explaining such offenses (Simpson and Piquero, 2002; Reed and Yeager, 1996; Langton et al., 2006; Blickle, 2006). The current study seeks to supplement the literature by focusing not simply on the direct causal links between self-control and white-collar offending, but also exploring how attitudes play a role between self-control and intentions to engage in white-collar crime. Specifically, this study examines whether attitudes towards environmental offending mediate and moderate the relationship between self-control and intentions to engage in environmental white-collar crime. The results indicated that attitudes toward environmental offending did have a mediating effect, but the effect of attitudes did not significantly vary as a function of self-control. Subsequently, simple slopes analysis found that the effect of attitudes was only significant among those with average and high levels of self-control. Implications for the general theory of crime and future directions for white-collar crime research are discussed.

Abstract:
In this work we have studied the aperiodic exchange modulation on the Kondo necklace model. Wang, Chuanlin. "Retrofitting of infilled RC frames using collar jointed masonry." Thesis, University of Leeds, 2017. http://etheses.whiterose.ac.uk/15961/.

Full text
Abstract:
Masonry is a composite material made of masonry units bonded together with mortar. A large number of historical buildings constructed using masonry can be found all over the world. Little or no seismic loading was considered when they were built. Therefore, masonry structures often need to be retrofitted or strengthened. This research proposed a new strengthening approach using a collar-jointed technique. Namely, the approach is implemented by building another masonry wall parallel to the existing single-leaf wall and bonding the two together using a mortar collar joint. Furthermore, collar-jointed masonry wall construction is also a popular construction system in reality. This thesis considers two different types of collar wall strengthening applications: pre- and post-damaged walls. The results found out that the pre-damaged strengthening could improve the lateral resistance by about 50% while the post-damaged retrofitting can only restore the initial strength. A simplified micro-scale finite element model for fracture in masonry walls was developed. The mortar joints and the brick-mortar interfaces are taken to have zero-thickness. The bricks were modelled as elastic elements while the brick-mortar interfaces were represented using a Mohr-Coulomb failure surface with a tension cut-off and a linear compression cap. One feature of the research was to identify the material parameters for the constitutive model. The material parameters were tuned by minimizing the difference between the experimental and numerical results of a single leaf wall panel. The model was then validated by assigning the parameters to the single-leaf masonry wall as well as to the double-leaf wall to predict its mechanical behaviour. Good agreement with experimental results was found. Furthermore, masonry is also widely used in the form of infill panels within RC frames. Therefore, the collar-jointed technique has also been extended and applied to the infilled RC frame. The numerical results showed that the collar-jointed technique could provide some benefits to the composite structure.

Thesis: S.M. in Technology and Policy, Massachusetts Institute of Technology, School of Engineering, Institute for Data, Systems, and Society, September, 2020<br>Cataloged from student-submitted PDF of thesis.<br>Includes bibliographical references (pages 52-53).<br>Increasing rates of skill obsolescence amongst white-collar workers resulting from rapid technological and organizational shifts within firms, coupled with an insufficient supply of workers proficient in technical areas such as software engineering and data science, has meant that firms are beginning to look to reskilling their incumbent workers as a means of filling skill gaps. This aim of this study was to examine the process and outcomes of efforts made by a US-based diversified global insurance company between 2016 and 2018 to reskill and reassign approximately 300 employees in the face of changing technological and market demands. More specifically, this involved running an immersive coding program for employees with little or no prior coding experience, with the ultimate aim of finding them new job roles in technical areas such as software engineering and data science. Quantitative analysis of longitudinal employee data, combined with qualitative interviews with program participants and administrators of the program, was used to assess the wide-ranging impacts that program had both on the firm and its workforce. Results suggest that the firm benefitted immensely from the retention of business-related knowledge and a low turnover rate of program graduates compared to externally hired software engineers. The majority of program participants also benefited from receiving free technical skills training, resulting in improved career prospects, however inefficiencies in the onboarding process for placing code school graduates on to new technical teams meant that some unfortunate participants were unfairly disadvantaged. Overall, this study provides encouraging evidence for the potential for white-collar reskilling programs to bring about positive outcomes for both firms and their workers, with several lessons learned for future implementations of similar programs.<br>by Frank Ryan.<br>S.M. in Technology and Policy<br>S.M.inTechnologyandPolicy Massachusetts Institute of Technology, School of Engineering, Institute for Data, Systems, and Society

Abstract:
Economic and tax crimes account for a significant proportion of criminal activity and result in considerable economic damage. In Germany, two of the most prominent offenders in this area in recent years were Thomas Middelhoff and Uli Hoeneß, both of whom served jail sentences. Taking advantage of the widespread media coverage of both cases, the following paper draws on economic theory to compare prison terms with fines. It argues that fines are preferred from an economic perspective and can therefore be considered a useful firstchoice punishment in cases involving white-collar and tax crimes. The paper sees itself in this regard as a plea.

This dissertation explores one white working-class family’s hopes, fears, illusions, and tensions related to social mobility. I tell stories from my experiences as a first-generation college student, including: ethnographic fieldwork; interviews with my family, community members, and former teachers; and narratives from other working-class academics to provide an in-depth, evocative, and relational look at mobility. I explore the roots of vulnerability in my family and how I was socialized into understanding belonging and worthiness in particular ways, and how this socialization influences my feelings of belonging and worthiness in the academy. The goal of this research is bridging – past and present selves, working-class and academic cultures, work and family – for me and my family and other first-generation students and their families.

Abstract:
This work investigates the characteristics of masculinity that are at the symbolic heart of China’s economic success, and of which the figure of the white-collar man is emblematic. Based on fieldwork observations, interview and media publications, it examines the gendered practices, aspirations and attitudes of men who identify with or aspire to white-collar status alongside discursive representations of the Chinese white-collar man, interrogating the links between practice and discourse. Drawing on various approaches to theorizing subjectivity, it argues that white-collar masculinity is performed in ways that suggest both radical shifts and continuities in understandings of gender, which challenge the prevalent teleological narrative of China’s modernization. The first chapter sets the scene for white-collar masculinity in the reform era and discusses fieldwork methodologies. Chapter two sets out the theoretical framework adopted to analyse the gendered white-collar subject, and examines academic literature on masculinities in China. Chapter three examines the ‘body culture’ of informants, and how they ‘bring themselves’ to white-collar discourse through attention to their bodies in areas of daily life such as dress, movement and hygiene. Chapters four and five look respectively at the production of corporate masculinity both inside and outside the office, through an exploration of business and leisure practices, and their overlap. Chapter six takes a close look at the young white-collar man as (heterosexual) boyfriend and husband and the final chapter investigates sexualisations of young urban middle-class males, and comments on their transformative possibilities.

Abstract:
This thesis highlights the importance of workers’ perceptions of and attitudes to sexual harassment. Past researchers have found that a variety of individual factors (age, gender, gender role, and past experiences of sexual harassment), and organisational factors (gender ratio, sexual harassment policies and the role of employers), correlate with the incidence of sexually harassing behaviours. Two studies presented in this thesis extend this research and were designed to investigate how these factors relate to workers’ attitudes towards and perceptions of sexual harassment. Study one investigated 176 workers from a large, white-collar organisation. Study two sampled 75 workers from a smaller, blue-collar organisation. By comparing two different workplaces the effect of the organisational climate was investigated. Individuals from Study two experienced more sexual harassment, were more tolerant of sexual harassment and perceived less behaviour as sexual harassment compared with individuals from Study one. The organisational context was found to affect the way in which organisational and individual factors related to workers' attitudes to and their experiences of sexual harassment. However, the factors that influenced workers’ perceptions of sexual harassment were stable across both studies. Although workers’ attitudes to and their perceptions of sexual harassment were significantly correlated, they were influenced by different factors. Overall, workers’ perceptions of sexual harassment were influenced by their attitudes, the behavioural context, and the gender of the victim and perpetrator. In contrast, attitudes to sexual harassment appeared to be more strongly influenced by individual factors, such as age, gender, gender role, past experiences of sexual harassment, and perceptions of management’s tolerance of sexual harassment. The broader implications of these findings are discussed and recommendations for future research are suggested.

Abstract:
In the literature on the emergence of the welfare state, the strength of trade unions and the organized working class is often touted as the primary driving force behind the welfare state project. Furthermore, much of the previous literature has tended to assume union homogeneity across countries, federations, industries and professions. What is conspicuously lacking from the current political science literature is a systematic analysis of real-world trade unions’ choice of labor market advocacy focus. Using a qualitative approach and studying both published union material as well as conducting a number of elite interviews with high-level union officials, this thesis studies the degree to which Swedish trade unions’ labor market policy preferences are defined by the union members’ labor market risk exposure and whether the union adheres to white-collar or blue-collar unionism. While the conclusions indeed suggest that labor market risk and blue-collar/white-collar unionism do have a systematic impact on cartain aspects of trade unions’ labor market advocacy, future “large N” studies utilizing alternative methodological approaches will be required to draw more easily generalizable conclusions.

Abstract:
Problem  Following the expanded competition when it comes to attraction and retention of skilled workers, hence white-collar workers, organizations need to better adapt to the rapid changes in the labor market. Due to the lack of knowledge about the recent Generation Z and their perceptions and values regarding work, the Person-Organization (P-O) fit has been applied in order to gain a deeper knowledge of what attracts and retains individuals of Generation Z, in order for companies to better meet their needs.   Purpose Due to the lack of research done on Generation Z, the purpose of this study is to gain a greater understanding of the values and perceptions of Generation Z and how they operate in the white-collar business sector. Moreover, this creates a greater insight for organizations in order to help them better target the right talent pool and gain a greater understanding of Generation Z that has come to change the labor market.   Method In order to gain a greater knowledge and insight about Generation Z, a qualitative study has been performed to better understand the perception of this Generation. In-depth interviews have been conducted with Generation Z that have or are currently operating in the white-collar business sector in order to meet the purpose of this study. Semi-structured interviews were conducted in order to allow for a fruitful discussion of their values and perceptions and the likely impacts they consider when joining an organization.   Conclusion Generation Z indicated that certain aspects are to consider in the decision-making process of applying or taking a job. Indeed, their own values, beliefs and work-personal life matter to a great extent and is to be matched with the company. This study comes to the result of six determined factors influencing this decision-making process: small enterprises, flexibility trust and freedom, work/private life environmental factors, organizations values/ethics, do what they love, digital natives.

L'importance du rôle des clones séminifères dans le programme d'amélioration du bananier est discutée<br>Two species are involved in the genetic background of edible bananas Musa acuminata Colla and M. B albisiana Colla (2n = 22). Inter or intra-specific differences do not exist between nuclear DNA amounts of 3 clones of the 2 species, determined by Feulgen cytophotometry. Nuclear DNA content amounst to 2. 7 pq per 2C nucleus. Meiotic chromosome pairing is good between both species at 2n, 3n and 4n levels. In the present state of knowledge, no significant difference can be shown between the 2 genomes. Chromosome changes reduce fertility. They result from hybridization between individuals of different geographic areas. Rate of spherical tetrads and pollen fertility are significantly correlated, which could give a special meaning to the orientation of the 2 homeotypic mitosis of the dyad. Female fertility of chromosome origin is increased in edible clones by embryo-sac development or fertilization failure and by abnormal relationships between embryo and endosperm. The wild tetraploids induced by colchicine treatment of seedlings have a slower growth rate and reduced numbers of female flowers and a lower female fertility. A mean of 3 potential quadrivalents are formed in the PMC. Tetraploid male fertility is good with a possibility of morphological separation of haploid and diploid gametophytes. Wild bananas interest in breeding scheme is discussed

Abstract:
Restructuring and widespread workplace closures have exposed white-collar workers to unprecedented levels of job insecurity and unemployment. This thesis describes two studies which examined effects on health of job insecurity and job loss in a longitudinal study of 10,308 white-collar British civil servants. Study 1 examined the consequences of short and long-term job insecurity generated by the transfer of work to agencies which were periodically under threat of privatisation. Relative to unaffected controls, self-reported morbidity increased significantly among respondents expecting transfer to an agency. Physiological measurements, such as blood pressure and body mass index, deteriorated significantly among respondents working in an agency. Increases in morbidity were greater in men than women. Study 2 examined the effects of job insecurity and job loss for respondents in one complete Civil Service department sold to the private sector. Data from three time points were analysed: 2 years pre-privatisation (anticipation phase); 3 months pre-privatisation (pretermination phase); and Ilh years post-privatisation. During the anticipation phase, compared with controls, respondents from the department undergoing privatisation experienced an overall increase in self-reported morbidity. By the pre-termination phase adverse changes were also seen in physiological measures. Postprivatisation, respondents were stratified by employment status. Overall, respondents in secure employment enjoyed better self-reported health than those in any other group. Unemployed men and insecurely employed women had the poorest self-reported health. Psychological disorder was significantly associated with insecure employment in both sexes and unemployment in men. Some evidence of health selection out of the workforce was found, but adjustment for preexisting morbidity demonstrated adverse effects on health in excess of those attributable to selection. Changes in work characteristics and health-related behaviours explained little of the effect of employment status on health. Further research on the employment statushealth relationship is required.
