Full textFillings in Conservation of gypsum plaster artifacts are frequent, given the material fragility. In order to stabilize the structure or the aesthetic when lacunas are occurring, non-original materials are applied: fresh paste is modelled in situ, or a cast-apart piece is fixed with adhesive after drying. Depending on deontological and technical criteria, a large range of materials can be used. To better understand them, a research was conducted with a multidisciplinary methodology involving analytical techniques of the conservation speciality. At first, statues from the gypsothèque and XIXth century frames were selected from the Louvre museum to be studied, combining in situ observations of fillings with documentary information from literature and archives. Several types of filling were identified such as material similar to the original and formulations with additives to ensure the distinction of treated parts. From those data, gypsum plaster at several water/powder ratios, with additions of calcium carbonate and titanium dioxide were selected for characterization. In a second phase, micro-structural aspects were examined by observations in optical and scanning electron microscopies (SEM). Elements were identified using energy dispersive spectroscopy (SEM-EDS). Thirdly, rheological aspects of suspensions were characterized and measured. Parallels between the consistencies of formulations were established, allowing to range the textures from paste to fluid suspensions, and their most suited use as modelling or casting formulations. In a fourth phase, mechanical properties at solid state were evaluated by measuring ultimate strengths through compressive and flexural tests, alongside the identification of E modulus. At last, to evaluate the stability of fillings under museum conditions, three artifacts, treated between 2015 and 2017, were monitored by high definition 3D scanning and images correlation. One of the artifacts was also equipped with LVDT displacement sensors to complete the evaluation.By relating physical, chemical, and multi-scale structural aspects of the formulations at liquid and dried state, the study of fillings can guide professionals in the selection of fillers according to specific conditions of intervention. The developed protocol can also be reused to study other filling formulations, artefact materials, typologies or specialities of Cultural Heritage

Full textThe overall objective was to assess the genetic diversity of a collection of native potato varieties originating from the island of Chiloe, to characterize the resistance of these varieties to late blight (Phytophthora infestans) and to inquire about the status of in-situ conservation. The following specific topics was developed: a) conservation in-situ of native Solanum within the great isle of Chiloe and its impact on diversity, b) evaluation of the morphological diversity of the native potatoes that we have collected c) assessment of genetic diversity of the collection by the mean of microsatellite and AFLP markers, d) characterization of field and in-vitro resistances to late blight. Based on the results of the surveys, we can conclude that in-situ maintenance of native potato diversity i is not well preserved, due to strong social and economic changes on the island of Chiloe. There is a clear process of replacing native varieties for commercial cultivars in response to market conditions. Native varieties are present in 80.5% of the farms of Chiloe, but the diversity of varieties has been significantly reduced. The results morphological diversity showed the formation of groups under the popular name and local attributes assigned by the farmers themselves. Molecular evaluation of the collection (SSRs and AFLP) reveals a high degree of genetic diversity. Both markers were consistent in classifying Solanum fernandezianum as the more distant genotype compared to all the others. The SSRs allowed the estimation of the polymorphic information content for seven loci which values ranged between 0.63 to 0.89. Both types of markers, did not provide the same groupings among the accessions. The SSR-based study showed a low differentiation between native potatoes and improved cultivated varieties. The analysis of diversity based on the AFLP was not inconsistent with this result because despite containing only one cultivar (Desiree), the cultivar was grouped with the native varieties. In relation to resistance to late blight, most accessions of native potato fall into the ranks moderately resistant to moderately susceptible

Full textActivity reports of the French Blood Establishment (EFS) indicate a growing demand for Labile Blood Products (LBP) as red blood cells (RBC), platelets and plasma. To ensure the vital demand of labile blood products (LBP), it’s essential to optimize the logistics related with the collection of blood components. To deal with this situation, the EFS Auvergne-Loire carry out a reflection in order to use more efficiently the collection devices in fixed and mobile sites, to improve the quality of service offered to the donor and the efficiency of human resources. In this context we have developed in this thesis operational tools for (i) modeling of blood collection devices (ii) The regulation of flows donors (iii) Planning of bloodmobile collections.The method analysis of collection devices is based on techniques of discrete event simulation. A preliminary modeling of donors’ flow in fixed and mobile collection systems using Petri nets was conducted. For the regulation of flow of donors, i.e. the optimal capacity planning and appointment scheduling of blood collections, two approaches were considered: (a) Simulation based-optimization.(b) Mathematical Programming: Mixed integer nonlinear programming (MINLP) based on queuing networks and mathematical programming representation of discrete event systems. For planning of bloodmobile collections. Two models have been developed: (a) At the tactical level: Mixed integer linear programming (MIP) to determine the weeks in which the mobile collection must be organized in order to ensure the regional self-sufficiency of RBC.
