Darmkrebszellen und T-Zellen regulieren ihren zentralen Kohlenstoffmetabolismus um
ihren anabolen Bedarf zu erfüllen. Tumorzellen mit einer KRAS- oder BRAF-Mutation
zeigen ein schnelles Wachstum, welches eine Umprogrammierung des Metabolismus vor
aussetzt. Der mitochondriale T-Zellen-Aktivierungsinhibitor (TCAIM) ist bekannt dafür
die mitochondriale Zellstruktur zu beeinflussen. Der Einfluss auf den Metabolismus nicht
klar.
In dieser Arbeit präsentiere ich erstmalig ein mathematische Model des zentralen Kohlen
stoffmetabolismus in Darmkrebszellen und T-Zellen. Mithilfe dieses Modells analysiere
ich, wie sich die Regulation in ähnlichen Zelllinien unterscheidet. In Bezug auf die Darm
krebszellen vergleiche ich BRAF-(CaCO2-BRAFV600E), KRAS-(CaCO2-KRASG12V) mu
tierte Zelllinien mit einer Basiszelllinie (CaCO2-control) und zeige, dass der Kohlenstoff
metabolismus in BRAF-mutierten Zellen im Vergleich zu den beiden übrigen Zelllinien
herabreguliert ist. Das Modell bestätigt außerdem, dass der Monocarboxylattransporter
(MCT) in den Darmkrebszellen eine wichtige Rolle, insbesondere in den KRAS mu
tierten Zellen, spielt. In T-Zellen zeigt der Vergleich von Wildtypzellen (CD8 T-Zellen)
mit TCAIM homozygoten Zellen (TCAIM homozygote CD8 T-Zellen), dass der Kohlen
stoffmetabolismus in zweiteren überwiegend herabreguliert und weniger aktiv ist. Diesen
Effekt konnte ich durch die Analyse von RNASeq-Daten der jeweiligen Zelltypen bestä-
tigen. Des Weiteren stelle ich fest, dass sich der Tricarbonsäurezyklus umkehrt, wenn
durch die Glykolyse nicht ausreichend Laktat exportiert und die Biomasseproduktion
unterstützt werden kann.
Meine Arbeit stellt damit insgesamt einen neuartigen Ansatz zur Integration von Meta
bolomik und RNAseq Daten dar, um die Regulation des zentralen Kohlenstoffmetabo
lismus zu verstehen.Colon cancer cells and T cells regulate central carbon metabolism to meet their anabolic needs. In KRAS and BRAF tumors, metabolic reprogramming is a premise to support rapid proliferation. In T cells, the mitochondrial T cell activation inhibitor (TCAIM) is known to affect mitochondrial morphology but its effect on cellular metabolism is not well understood. Via mathematical modelling, I investigate the differential regulation of closely related cell lines. I present the first mathematical model for colon cancer and T cell metabolism, unraveling differential regulation between related cell lines. The model shows that CaCO2-BRAFV600Ecells are mostly downregulated compared to CaCO2-KRASG12Vand CaCO2-control. Additionally, it demonstrates the critical role of monocarboxylate transporter (MCT), especially for CaCO2-KRASG12V. Concerning T cells, I compare wild-type T cells to homozygous TCAIM T cells. This unveils that TCAIM homozygous cells have a mostly downregulated TCA cycle, validated by RNASeq data, and are less metabolically active than wild-type T cells. Furthermore, if the glycolytic flux is not sufficient to support lactate export and biomass production, the model reveals that the TCA cycle is reversed as it requires less regulation. Taken together, this work presents a novel approach to integrate data referring to metabolic and genetic regulation of metabolism. On this basis, we can now better discriminate the metabolic capacity of CaCO2-control, CaCO2-BRAFV600E, CaCO2-KRASG12V, wildtype CD8 T cells, and homozygous TCAIM CD8 T cells.

Following this examination, a more traditional design method was used to create a Montessori School that would draw inspiration from its position between the earth and sun to guide its architecture. This thesis examines the phenomenon of colour as a means of expression of meaning in spatial contexts. The nature of the underpinning project involved paintings and their integration with an architectural setting. Judgements made-in-situ by users of the building and an expert focus group (architects, designers and fine artists) were comparatively analysed for variance in interpretations of meaning, taking into consideration their experience with colour as a medium of expression. Commonalities and differences in the responses of colour amongst and between the various groups were also analysed. To achieve this a combination of questionnaires, semi-structured interviews, and focus group meetings were used as data sources. The researcher used her experience as a painter to create two large paintings (11.5m x 2m each), which were installed in the public space of the Manchester Bridgewater Concert Hall over a period of four months. One painting was predominantly blue and green, the other was predominantly yellow and red. The installation had two phases, in which the respective paintings were each installed separately and accompanied by a corresponding lighting scheme. Colours were separated into two temperature groups - warm and cold. However, previous research findings had indicated that responses to these two groups of colour differ along other dimensions also. What had not been established by previous research, and was examined in this project, was whether these indicative differences would apply when colour is approached as part of an holistic environmental meaning rather than in isolation. The integration of paintings and colour into the architectural setting made it possible for multiple layers of experience to be examined. Meaning was extracted from the relationship between colour and the perception of two dimensions of space - pictorial space depicted in the paintings and the architectural space. The relationship between both was also explored which allowed the confirmation of previous findings and the analysis of the variables which need to be addressed when dealing with colour for paintings in real architectural environments. The thesis describes the author's conceptual model based on a combination of this empirical evidence and theoretical framework developed from the existing interdisciplinary body of knowledge on colour. The thesis also discusses how relationships between the aesthetic and psychological categories were established. It contributes to the field by demonstrating how the subjectivity of the perceptual experience can be translated into the expression of meaning along cognitive and affective dimensions within the context of a real-life application of colour in space. Additional to the written thesis a short audiovisual provided in both video and CD Rom, was created to show both the making of the paintings and their installation at the Bridgewater Concert Hall.

This research investigates the creative use of expressive drapery and textiles, and in particular the line of washing, in three films that have rich displays of fabrics and costume: The Piano (dir. Jane Campion, 1993), The Governess (dir. Sandra Goldbacher, 1998) and Girl with a Pearl Earring (dir. Peter Webber, 2003); and provides the inspiration for a series of short films that explore positive and negative aspects of clotheslines and textiles. The research areas of Film Costume, Fashion Studies and Art History each provide relevant context and background to the study of the clothes and linens in the three films under consideration. Significant themes that are considered include: cinema’s ambivalent relationship with costume; dress, undress and the male gaze; representations of historical and character-coded clothing; the expression of fantasy and desire through textiles and costume; and the contribution of art history towards creating an understanding of fabrics that denote a separation between the ordinary and extraordinary and between reality and an imaginary sphere. Mary Ann Doane’s study of the gaze at the interface of the interior and exterior of the home in the ‘woman’s film’ leads her to consider the frequent portrayal of women waiting by or looking through windows in relation to Freud’s theory of The Uncanny and leads me to recognize that the line of washing in cinema also denotes the limits of a woman’s space within the grounds of her home and marks a formal boundary between the familiar and the unknown. Similarly, the line of washing reflects notions of The 5 Uncanny in displays that portray tensions between the opposing themes of the seen and unseen, the spoken and unspoken, of presence and absence and of purity and contamination. The idea of familiar clothes and linens displayed on the line of washing as alternately comforting and disconcerting has become the focus of my practice. Some of the works have been shown individually during the course of this study in group and open art exhibitions, and a solo exhibition at Galeri Caernarfon 15th January – 24th February 2017 showed the collected short films and supporting material resulting from the research.

This thesis describes a new approach to the study of high-redshift star-formation and its environments that can be applied to large high-redshift surveys. Instead of relying on spectroscopy or narrow-band photometry to study galaxy line emission in detail, the properties of large emission line galaxy (ELG)populations are estimated from broadband photometry by measuring colour-residuals against colours drawn from a set of line-free stochastic burst models-based on (Bruzual & Charlot, 2003). Simulated star-formation histories drawn from semi-analytic and adaptive-mesh-refinement codes were converted into mock galaxy colours, but neither could-span the range of observed galaxy colours at high redshift. Instead, an existing set of exponentially declining star-formation models with stochastic bursts was used, because it closely spanned the range in observed galaxy colours in the bandsthat were line-free at each redshift. Small colour offsets were measured between the models and the observations, corresponding to the equivalent widths (EWs)of Hα, [OIII] and [OII]. In this way, I measure the rest-frame Equivalent Widths of the Hα, [OIII]and [OII] emission lines as they are redshifted through all filters from CANDELS(near-continuous U to 4.5μm coverage) for a large sample of galaxies from z=0.1up to z=5. This approach relies solely on the line-free models, a set of existing reliable photometric redshifts, and a colour cut (B−K < 2 or equivalent) to select only the dust-free young objects (the majority of identified emission-line galaxies). Once correctly identified, I apply this method to the CANDELS-UDS photometry to characterise the properties of Emission-Line Galaxies (ELGs) through these lines. I find that in this sample the Hα and [OIII] ELG fraction with EW > 150Årises from < 5% at z < 1 up to 40% at z > 2. The co-moving ELG density rises from 5 to 30 ×10 −4 /Mpc −3 at z=2.3. The evolution of median Hα EW with redshift is consistent with results from HiZELS and 3D-HST yielding median EW ∼ M 0.25 (1+z) 1.75 up to z=2.3, from which it departs to values of 450Å atz=4.3. [OIII] remains weaker than Hα for z < 3 and matches its values above that redshift. [OIII] also displays a larger fraction of extreme EWs than Hα. [OII], while correctly identified, never becomes as extreme as the other two lines lines, even when corrected for the evolving continuum. This is evidence of an increasing [OIII]/[OII] ratio with increasing z through-out this sample. While these results agree with spectroscopic and narrow-band surveys, the use of the deeper broadband filter coverage enables a systematic measurement of the increasingly prevalent high EWs ( > 500Å) in galaxies at every redshift spanning the 10 8 to 10 10.5 M range. Subsequently, this method was applied to all the other CANDELS fields (GOODS-South and North, COSMOS and EGS) and further corroborates these results. These results further show that EW dependence on mass is steeper for [OIII] than for Hα. Line EWs are then converted into luminosities for the three lines and fitting formulas are obtained, displaying L Hα ∼(1+z) 3.2 M 0.45−0.6log(1+z), with similar results for the other lines. L Hα is converted into star-formation rate and specific star-formation rate (sSFR). sSFR at low-z aligns approximately with the main sequence (with a steeper dependence in mass), but at high-redshift sSFR remains above the main sequence by a factor of 2 and rising towards medians SFR=100/Gyr around log(M/M )=9, showing a departure of the main sequence of star formation at lower masses log(M/M ) < 9.5. The SFRD of ELGs is 1% at low redshift, but rises to 30% at z=4.5. The L [OIII] /L Hα ratio is used to estimate L [OIII] /L Hβ and the ionization parameter q, for which the median atz > 0.5 stays approximately constant at 10 8 cm/s, and increases with mass. Using the L [OIII] /L [OII] ratio and q, median metallicity is shown to be sub-solar, and can be tentatively estimated for z > 0.5 to be Z/Z ∼0.3. The errors are large, but this could also mean a large range in metallicity from Z to 0.1Z . L [OIII] /L [OII] rises with sSFR as shown in the literature. This method shows great potential to survey emission-line-derived physical quantities for large galaxy populations with a low computational footprint, which could be particularly useful for pixel-by-pixel EW imaging. It is also flexibile, which allows it to be applied to any future deep multi-broadband fields.

In order to meet the increasing demand for spectrum to support future DoD test and evaluation (T&E) requirements in an environment of decreasing spectrum availability, the DoD requires new spectrum aggregation and spectrum management technologies. Advanced radio technologies that sense and aggregate non-contiguous blocks of spectrum into a larger communication channel capable of supporting higher user data rates are needed. Moreover, the T&E community must more effectively manage and use the spectrum that is available to it, and utilize new paradigms in which spectrum is dynamically allocated on a non-interference basis to multiple, concurrent users and in a way that meets both planned and unplanned changes to mission requirements, and real time channel conditions, factoring in both Federal and non-Federal users of spectrum.
The objective of the Adaptive Spectrum Aggregation and Management (ASAM) project is to develop technologies that will sense available spectrum, aggregate white space within fragmented frequency bands to create communication channels, and dynamically allocate those channels across multiple range users using algorithms that increase spectrum utilization and efficiency while applying policy constraints. The ASAM project is sponsored by the Test Resource Management Center (TRMC) and was awarded in 2016 through the National Spectrum Consortium.
This paper will describe the ASAM project and technical challenges associated with its development.

This dissertation offers the first comprehensive examination of the diverse inte-ractions and collaborations among Canadian modernist poets and artists, as well as the aesthetic, thematic, and idiomatic relationships between their poems and works of art. The project incorporates archival and historical research to demon-strate the interartistic nature of modernist poetry in Canada between 1930 and 1960. By establishing that paintings, sculptures, and book designs and illustrations by Canadian artists who knew and worked closely with Canadian modernist poets informed and affirmed the content and aesthetics of the poetry, the dissertation argues for a consideration of the social dimension of literary modernism in Cana-da. Chapter One investigates the personal relationship between the poet Dorothy Livesay and the artist Emily Carr and reveals the aesthetic and thematic conver-gences of their paintings and poems as they relate to landscape and affect. Chapter Two deals with the Montreal little magazines of the 1940s as interartistic sites of collaboration among artists and poets and argues that the artists' paintings provided models of human agency for the poets. Chapter Three considers the small press movement and gallery space of the 1950s as similar sites of interartistic collaboration and contact; it suggests that this contact inspired Canadian modernist poets to translate the aesthetic and thematic tensions of Canadian art into their poems. Chapter Four concerns both the poetry and visual art of P.K. Page-Irwin and argues that the poet addressed an ongoing aesthetic conflict in her poetry through the visual arts. Where the first chapter examines the relationship between a single poet and a single artist, the second and third chapters analyze the dynamics of groups of artists and poets working closely together on little magazines and on small press publications and encountering each others' work within the space of a gallery. The final chapter considers the work of a poet who is also a visual artist. This framework reveals the diversity of interartistic relationships that flourished throughout the rise of modernism in Canada.Cette thèse de doctorat offre le premier examen approfondi des diverses interactions et collaborations parmi les poètes et les artistes modernes du Canada et les relations aesthétiques, thématiques, et idiomatiques entre leurs poèmes et leurs oeuvres d'art. Within the world of library collection and preservation, the focus has historically been on the dual preservation of physical form and intellectual knowledge as represented through symbols (text), images, metadata, and organization of information. Certainly, the philosophy of the library to "enhance learning and ensure access to information for all" (ALA 2008) is paramount in directing resources and collection development in response to the needs of the user community. However, research and educational material may be found beyond the explicit materiality of books and the text they contain; understanding and recognizing spiritual and cultural properties implicitly located within certain library materials may provide additional fields of research, as well as learning and collaboration opportunities. Using adjunct literature within the field of museum studies and ethnography, this paper proposes to discuss how simultaneous recognition of intangible and tangible properties of certain objects is relevant to the library. The special collections library is perhaps the best place where such considerations can be made - it is a library dedicated to preserving not only copies of each work, but variant copies whose tangible and intangible properties may vary from one another - though some examples may apply to academic or public libraries as well. The scope of this undertaking will include a discussion of the adjunct and related literature from the museum world, identification of books, or types of books that have spiritual or cultural values to be considered, collection of cultural or religious viewpoints related to identified materials from members of one of those communities that create or use these items (Buddhist), and discussion of how such findings can inform the ongoing operations of special collections libraries and their mission as cultural memory institutions.

Knowing how to promote oneself in written self-promotional genres is a vital, yet neglected, component of non-native speaker (NNS) communicative competence.Focusing on the self-appraisal, I explore some of the impression management (IM) tactics and interpersonal metadiscourse (MD) markers used by writers to manage the impressions they give off. Of particular interest is the extent to which (a) patterns of self-presentation, and (b) language background (NS/NNS) contribute to the affective response of the reader.
The self-appraisals, collected from 41 students taking a required undergraduate writing course, were grouped according to the affective response of independent readers. Patterns of IM and MD use were compared across positive- and negative-affect groups, and across NSs and NNSs.
The findings reveal distinct differences between affect groups in patterns of IM and MD use. Interestingly, NS/NNS differences did not contribute to the affective response of the reader, thus challenging the assumption that NS linguistic form is requisite to pragmatic competence.

Background Omni-channel retailing has developed as an extension of multi-channel retailing. The difference between the concepts is the level of integration, where an omni-channel requires seamless and complete channel integration compared to a multi-channel where the level of integration varies from non-existing to high. Today’s customers expect an integrated shopping experience where they can combine the different channels according to their preferences and omni-channel is a way for retailers to meet this need. Purpose The purpose of this thesis is to explore the concept of omni-channel retailing and its implementation in retail firms operating in the Swedish market. Method This study is based on an interpretive approach and takes the form of an exploratory multiple case study through the investigation of four retailers. Qualitative data was collected through semi-structured interviews and the findings were analyzed abductively. Conclusion The conclusion of this thesis is that the investigated retailers still have, more or less, a long way to go in their integrative efforts. The retailer that has come the furthest in their effort has only implemented nine out of fifteen omni-channel criteria. Three main incentives for implementing an omni-channel strategy were identified. These are to respond to customer demands, to optimize the individual sales channels and to gain a competitive advantage. Additionally, several challenges arise as the retailer moves towards an omni-channel. These include technological investments, change management, channel conflict, return management and consistency. Furthermore, five potential sources of these challenges were identified as technological requirements, organizational changes, sales accreditation policies, in-store returns and finally the franchise business model.

Arabic literature in Israel has evaded critical attention, or has been treated as an uncomplicated part of Palestinian national culture, on a quest for unification and an identity that was devastated in 1948. This dissertation complicates that narrative through close readings of short stories by five Arab citizens of Israel—Imil Habibi, Muhammad ‘Ali Taha, Muhammad Naffa‘, Hanna Ibrahim, and Zaki Darwish—between 1967 and 1983. Focusing on the relationship between geography and fiction, I suggest that literary constructions of “place” and “space” by these authors reveal a range of cultural negotiations that break down entrenched dyads: Palestinian yet Israeli; Palestinian on the one hand, Israeli on the other; spared exile, but suffering occupation. Instead, these writers evoke the hybrid and ambivalent experiences produced in the paradoxical spaces of Israeli-Palestinian life. I develop an analytical framework that incorporates geographic and literary theory. I use the work of humanists such as Gaston Bachelard, Yi-Fu Tuan, and Edward Casey to suggest that literature mediates geography in a way that communicates belonging, alienation, or personal and collective meaning. The framework is bolstered with the work of postcolonial theorists such as Homi Bhabha, along with historical and political sources, to capture the contextual resonance of the texts. After laying out these theoretical guidelines, I offer a historical account of Arabic literature in Israel and embark on four analytical chapters. Chapter Two explores Imil Habibi’s portrayals of anxiety around post-1967 Palestinian reunions. Chapter Three focuses on the themes of Muhammad ‘Ali Taha’s Palestinian collective identity in Israel. Chapter Four takes up the theme of “the land” in the works of Muhammad Naffa‘ and Hanna Ibrahim, in the context of 1970s land expropriations. Chapter Five explores a long story by Zaki Darwish and its depiction of the body’s phenomenological relation to the homeland. Rather than portraying counter-narratives that suggest a binary of “Israeli” and “Palestinian” always at odds, these authors portray the spaces and characters in between. They disclose the anxieties of finding a sense of place in the context of a dispersed Palestinian nation, geopolitical uncertainty, social marginalization within the state, and the subtle geographies of a historic homeland that both is—and is not—one’s own.

Technical support for products after the sale is commonplace in today’s businesses. Original Equipment Manufacturers (OEMs) provide technical support to their dealer channel for resolution of complex product issues. Technical support staffing levels can vary by product type, product complexity, and production volumes, and case volumes. This research seeks a better understanding of appropriate staffing levels between three product lines for one OEM. The objective of this paper is to develop a model for monthly and weekly average case volumes for the three product lines, based off of historical case volume data. This data is used to predict a product line’s (platform’s) workload based off the month of the year. The output of each platform’s monthly case volume is then used in an optimization model to determine optimal staffing levels for each platform, based off the time of the year. The models developed for each platform use a linear relationship which regresses workload on a set of binary variable for the months of the year. Each of the models developed provided statistically significant coefficients for months which contain the platform’s highest workload. The outputs from these models are used in a mixed integer nonlinear programming optimization model to determine staff level of full time equivalent (FTE) employees at each platform. Each of the three scenarios utilized in this research provide similar trends and staffing levels for each of the three product lines. Results of this research are of interest for the management of technical support staffing.

Traditionally, pluripotent stem cells are derived from preimplantation embryos and fetal germ cells, which give rise to embryonic stem cells (ESCs) and embryonic germ cells (EGCs), respectively, In contrast, induced pluripotent stem cells (iPSCs) are derived from somatic cells upon overexpression of defined transcription factors such as Oct4, Sox2, Klf4 and c-Myc. Despite their origin from different cell types, all of these pluripotent stem cell lines share the ability to self-renew indefinitely in culture while retaining the capacity to differentiate into derivatives of all three germ layers. Because pluripotent cells provide a useful tool in basic research and cell therapy, it is critical to understand the molecular similarities and differences between ESCs, EGCs and iPSCs.
The studies presented in this thesis aim to address the equivalence of different pluripotent cell types. In the first study, we performed a systematic comparison of DNA methylation and gene expression patterns between isogenic mouse ESCs and EGCs. Surprisingly, we found that global DNA methylation patterns were indistinguishable between ESC and EGC lines of the same sex, while female cell lines exhibited global hypomethylation compared to male cell lines. Mechanistically, upregulation of the X-linked gene, dual specificity phosphatase 9 (Dusp9) in female cells attenuated MAP kinase signaling, resulting in global DNA hypomethylation via the reduction of Dnmt3a and Dnmt3b protein levels. In the second study, we compared isogenic, transgene-free hESC and hiPSC lines to determine whether molecular differences exist between hiPSC and hESC lines when controlling for genetic background and reprogramming methodology. Strikingly, transcriptional variation between different genetic backgrounds was greater than variation observed between cell types (i.e., hiPSCs compared to hESCs). Moreover, the few transcriptional differences observed between isogenic hESC and hiPSC lines had no apparent functional consequences and these genes were not identified during the comparison of a larger set of independently derived non-isogenic hESC/hiPSC lines. We conclude that hESCs and hiPSCs are highly similar on a transcriptional and functional level and cannot be distinguished by a defined gene expression signature.
Together, our data demonstrated that sex rather than cell type of origin drives global epigenetic and transcriptional patterns in conventional mouse pluripotent cell lines. These results provide fundamental insights into the epigenetic regulatory mechanisms that govern pluripotency. Additionally, our comparison of isogenic hiPSCs and hESCs supports the view that cellular reprogramming technologies faithfully reset the transcriptional pattern of somatic cells and establish a pluripotent state that is molecularly and functionally equivalent to embryo-derived stem cells. These findings may provide the basis for future mechanistic studies and help to translate iPSC technologies into a therapeutic setting.Medical Sciences

Thesis (MA (English))--University of Stellenbosch, 2007.This thesis focuses on the relationship between Dora Carrington and Lytton Strachey and offers three models for reading their unconventional relationship. Carrington was in love with the homosexual Strachey and the two lived together at Tidmarsh, and later Ham Spray House, for more than fourteen years. The three models make extensive use of primary sources, namely the letters and diaries of Carrington and Strachey. Furthermore, I draw on two seminal biographies of Carrington and Strachey written by Gretchen Gerzina and Michael Holroyd respectively. The first model I examine is a form of pederasty. I argue that, soon after they met, Carrington and Strachey began a friendship which was based on his educating her in a variety of ways. He served as a mentor both intellectually and sexually. Strachey was familiar with the concept of pederasty as a result of his involvement with the Cambridge Conversazione Society, better known as the Apostles, and used his knowledge to induct a rather naïve Carrington into new ways of thinking. This pederastic relationship also allowed Carrington a certain amount of freedom as it enabled her to pursue her art without the demands a heterosexual male would make of her. The second model for reading their relationship is that of parody. While Carrington and Strachey’s relationship resembles a heteronormative relationship, it can, at times, be read as parodic. I argue that they both subvert heteronormativity in humorous ways as a means to critique their parents’ Victorian marriages and to interrogate notions of masculinity vi and femininity. I discuss the roles they played within their domestic environment, and pay particular attention to how this intersected with Carrington’s artistic endeavours. This parodying of heteronormativity was, I suggest, also one of the only ways they could find of expressing the love they felt for one another. The last model I offer draws on theories of kinship. I examine how Carrington and Strachey resorted to familial constructions of descent as a means to veil the love they had for one another and to avoid criticism and ridicule from the Bloomsbury group and beyond. When they established a home at Tidmarsh, they altered their form of kinship to utilise principles of alliance. However, another shift took place with the introduction of Ralph Partridge, Carrington’s husband, and I argue that the terms they used to address each other changed to constructions, once again, of descent, at least until the dissolution of the Carrington-Partridge marriage. Carrington and Strachey’s relationship is often viewed as unconventional and she is often depicted as being utterly subservient towards him. However, the three models I have used demonstrate that their love was mutual. The models also reveal their relationship to be quite conventional in the manner in which Carrington and Strachey expressed their love for one another and how these expressions of love developed during the different phases of the life they spent together.

Guayule (Parthenium argentatum Gray), a semidesert shrub was evaluated as a potential rubber crop. Since polyploid guayule reproduces by apomixis, progeny of individual plants should duplicate the characteristics of their parent. This study was conducted to estimate variations within and among progeny families from single-plant selections. Ten progeny per family were individually evaluated for plant height, width, fresh and dry weight of clipped branches, rubber and resin content, and yield. Leaves of selected plants were analyzed by gradient polyacrylamide gel electrophoresis for esterase and peroxidase to estimate genetic relatedness between parents and progenies. Negative correlation was between parents and progenies for most characters, but none were statistically significant from zero. This suggests a low heritability for the characters measured. Esterase and peroxidase detected variation in banding between parents and progenies and among progenies. This study suggested that out-crossing and meiotic reduction have occurred among apomictic guayule lines.

Thesis (MA)--Stellenbosch University, 2003.ENGLISH ABSTRACT: This research assignment deals with the breakdown in the Clausewitzian concept of the modern trinitarian structure of war. Martin Van Creveld in his book entitled, “The Transformation of War” written in 1991, discusses ‘Future War’ and the way in which wars will be fought. It will not be the highly technical interstate kind of war the West has been preparing for, but rather low intensity conflict where the lines between state, soldier and civilian become blurred, society becomes a war zone and the conflict becomes a more direct experience for the people. Colombia is a prime example of where this is occurring and the most salient manifestation of the low intensity conflict is the blurring of the lines between war and crime. There are left wing guerrillas fighting for social justice for the dispossessed population, but their tactics resemble crime and the government views them as terrorists. They run a self-sufficient organisation, one of the most profitable insurgent groups in the world largely funded through kidnap ransom payments. The right-wing paramilitaries are on a quest to cleanse Colombian society of the guerrillas and assassinate suspected guerrilla sympathisers. To complicate issues, both insurgent groups are involved in the drug trafficking trade, whether it be directly or by way of taxing land on which coca is grown. In this situation, war and crime have become inextricably linked and a distinction between the two is impossible on both practical and conceptual levels. However, if it is not crime and it is not war, but a complicated melange of the two, a new framework for analysis is required in order to attempt a solution.

Radio-loud active galactic nuclei (AGN) are one class of objects associated with accretion activity onto supermassive black holes in centers of massive galaxies. They are believed to be in a radiatively-inefficient accretion mode with low accretion rate. To understand this accretion mode, it is important to measure its radiative output at high energies (> 13.6eV), which can be traced through optical emission lines. However, little is known about their true radiative output. This is because no correlation between optical emission-line and radio luminosity has been found for the majority of low-luminosity radio AGN, which are often classified as low-excitation radio galaxies, or Fanaroff-Riley Class I (FR-I) radio galaxies. We demonstrate that most of the line emission found in these galaxies is not powered by the central AGN, but likely powered by some old stellar population. Only when this component is subtracted or otherwise taken into account can we estimate the true line emission associated with the AGN. These emissions may show interesting correlations with the radio luminosities in some cases.

In this thesis, I investigated how human vision processes colour and luminance information to enable perception of our environment. I first tested how colour can alter the perception of depth from shading. A luminance variation can be interpreted as either variation of reflectance (patterning) or variation of shape. The process of shape-from-shading interprets luminance variation as changes in the shape of the object (e.g. the shading on an object might elicit the perception of curvature). The addition of colour variation is known to modify this shape-from-shading processing. In the experiments presented here I tested how luminance driven percepts can be modified by colour. My first series of experiments confirmed that depth is modulated by colour. I explored a larger number of participants than previously tested. Contrary to previous studies, a wide repertoire of behaviour was found; participants experienced variously more depth, or less depth, or no difference. I hypothesised that the colour modulation effect might be due to a low-level contrast modulation of luminance by colour, rather than a higher-level depth effect. In a second series of experiments, I therefore tested how the perceived contrast of a luminance target can be affected by the presence of an orthogonal mask. I found that colour had a range of effects on the perception of luminance, again dependant on the participants. Luminance also had a similar wide range of effects on the perceived contrast of luminance targets. This showed that, at supra-threshold levels, a luminance target's contrast can be modulated by a component of another orientation (colour or luminance defined). The effects of luminance and colour were not following a particular rule. In a third series of experiments, I explored this interaction at detection levels of contrast. I showed cross-interaction between luminance target and mask but no effects of a colour mask.

To do so, we will investigate interdisciplinary performances, synesthesia (union of perceptions) and the different ways in which painting and music can relate to each other.

This thesis demonstrates the colonizing impact of multiculturalism on in-between subjects. In-between subjects are defined as individuals whose identities form at the interstices of two or more cultures. Using evidence from the narratives of The Namesake, The In-Between World of Vikram Lall, and Londonstani, this research describes a set of structurally embedded cultural concepts – rooting, marking, deference, and communication. These concepts contribute to the scripting of the essentialized identity roles that limit the recognition, inclusion, and participation of in-between subjects. Further, these conditions impose a double-bind on in-between subjects; in performing essentialized identities, in-between subjects deny their own free participation and expression, but to not perform an essentialized identity would mean sure exclusion and marginalization. The evidence points to a possible solution, however, in the form of safe spaces and relationships where difference is unscripted and alterity fills the space between Self and Other.Ce travail démontre les effets colonisant de la multiculturalisme sur les "in-betweens." Les in-betweens sont des individus ayant une identité constituer au croisement de deux ou plusieurs cultures. This thesis uses mixed methods and datasets to explore how political news is perceived and shared within and across party lines in the context of the 2016 US presidential elections. We begin by examining the impact of political context versus article content on the reader through a crowdsourced study, and follow up with a large scale analysis of story sharing on the social platform Twitter to find cases where popularity transcends political affiliation. In Part One, we look at reader reactions. We investigate the question of trust in political news by performing a study online. We look at the impact of content features (reading level of the article) versus context clues (media brands) to find that political affiliation and brand outweigh all other aspects. In the second part of this thesis, we focus on reader actions. In particular, we look at how political news stories from the same time period are shared on the social media platform Twitter. As we found party loyalty and media brand perceptions to be significant influences on the reader's opinion of news, we are especially interested in examining emotional features that cause stories to become popular beyond political boundaries. Together, these two parts hope to form a more complete view of factors affecting and driving readers in an election cycle that is heavily influenced by media coverage, both traditional and new.

The major component of complex genomes is repetitive elements, which remain recalcitrant to characterization. Using maize as a model system, we analyzed whole genome shotgun (WGS) sequences for the two maize inbred lines B73 and Mo17 using k-mer analysis to quantify the differences between the two genomes. Significant differences were identified in highly repetitive sequences, including centromere, 45S ribosomal DNA (rDNA), knob, and telomere repeats. Genotype specific 45S rDNA sequences were discovered. The B73 and Mo17 polymorphic k-mers were used to examine allele-specific expression of 45S rDNA in the hybrids. Although Mo17 contains higher copy number than B73, equivalent levels of overall 45S rDNA expression indicates that transcriptional or post-transcriptional regulation mechanisms operate for the 45S rDNA in the hybrids. Using WGS sequences of B73xMo17 doubled haploids, genomic locations showing differential repetitive contents were genetically mapped, revealing differences in organization of highly repetitive sequences between the two genomes. In an analysis of WGS sequences of HapMap2 lines, including maize wild progenitor, landraces, and improved lines, decreases and increases in abundance of additional sets of k-mers associated with centromere, 45S rDNA, knob, and retrotransposons were found among groups, revealing global evolutionary trends of genomic repeats during maize domestication and improvement.

In 'Reading Between the Lines', I investigate the literary representations that, building on the influential writings of Mountstuart Elphinstone and WaIter Scott, created an 'Afghanistan of the mind' over a century of British engagement. Using the quantitative methodologies of book history, my thesis sheds light on the public responses to three successive Anglo-Afghan wars, and the reception of the writings to which these gave rise. By this means, I trace the rise and fall of readers' interest in Afghanistan through six studies of individual authors. The first chapter examines the media storm around the writings of Florentia Sale, eyewitness to the disasters of the First Anglo-Afghan War, whose bestselling Journal was published by John Munay. Chapter two considers John William Kaye, and his journalistic and fictional reshaping of the war, culminating in his authoritative History. The third chapter considers George Alfred Henty's use of Afghanistan and the Second Anglo-Afghan War in his historically-based adventure stories for boys, which is contrasted with the more subtle imaginative representations in Rudyard Kipling's 'The Man Who Would Be King' and Kim, explored in chapter four. Investigating literary responses in the 1920s and 30s, I study two relatively unknown authors, to demonstrate the pervasive urge for eyewitnesses to the Third Anglo-Afghan War and its aftermath to tell their story. In chapters five and six, Martin Louis Alan Gompel1z (pseud. Ganpat) and Alice Pennell (nee Sorabji), are scrutinized through a detailed biographical, bibliographical and textual study, using material from Blackwood's and John MU11'ay archives, giving the most complete account of their writings to date. Using these six authors, and the success or failure of their writings, I critically examine the various influences of the press, publishers, and book trade on the rise and fall of public appetites for narratives of Afghanistan.

We present the effective temperatures (T-eff), metallicities, and colours in Sloan Digital Sky Survey (SDSS), Two Micron All Sky Survey, and Wide-field Infrared Survey Explorer filters, of a sample of 3834 late-K and early-M dwarfs selected from the SDSS Apache Point Observatory Galactic Evolution Experiment (APOGEE) spectroscopic survey ASPCAP (APOGEE Stellar Parameters and Chemical Abundances Pipeline) catalogue. We confirm that ASPCAP T-eff values between 3550 < T-eff < 4200 K are accurate to similar to 100 K compared to interferometric T-eff values. In that same T-eff range, ASPCAP metallicities are accurate to 0.18 dex between -1.0 <[M/H]< 0.2. For these cool dwarfs, nearly every colour is sensitive to both T-eff and metallicity. Notably, we find that g - r is not a good indicator of metallicity for near-solar metallicity early-M dwarfs. We confirm that J - K-S colour is strongly dependent on metallicity, and find that W1 - W2 colour is a promising metallicity indicator. Comparison of the late-K and early-M dwarf colours, metallicities, and T-eff to those from three different model grids shows reasonable agreement in r - z and J - K-S colours, but poor agreement in u - g, g - r, and W1 - W2. Comparison of the metallicities of the KM dwarf sample to those from previous colour-metallicity relations reveals a lack of consensus in photometric metallicity indicators for late-K and early-M dwarfs. We also present empirical relations for T-eff as a function of r - z colour combined with either [M/H] or W1 - W2 colour, and for [M/H] as a function of r - z and W1 - W2 colour. These relations yield T-eff to similar to 100 K and [M/H] to similar to 0.18 dex precision with colours alone, for T-eff in the range of 3550-4200 K and [M/H] in the range of -0.5-0.2.

My thesis defends the cognitive value of a close reading of literary fiction which I believe has been overlooked in the philosophical literature. I first outline an account of literary fiction in terms of the standard features of both literature and fiction. The diverse philosophical positions which hold that reading literary fiction ‘improves the mind’ attribute some significance to literariness but do not say how the literary features of literary fiction help to develop cognitive gain. An explanation is required in order to meet anti-cognitivist scepticism. I rule out the view that cognitive gain is irrelevant to our aesthetic appreciation of literature. My thesis locates cognitive gain in the Verstehen tradition and identifies five relevant senses of ‘understanding’ as the cognitive value at stake. The case is made that reading literary fiction as literature stimulates the relevant senses of understanding; in the course of the discussion I meet objections from elitism and subjectivity. I argue that a reader’s engagement with literary devices stimulates the five senses of understanding and supply examples from: irony, particular detail and precise phrasing, metaphor, play with perspective, ambiguity and repetition. Contrary to the contention that dominates the current debate, that a reader gains knowledge from fiction, I argue that it is the way readers gain understanding from literature that is more significant to cognitive value. Finally, I argue that the relevant senses of understanding may be transferred to an extra-textual context and so bridge the gap between understanding the text and understanding the world beyond the text.
