Full textColon cancer cells and T cells regulate central carbon metabolism to meet their anabolic needs. In KRAS and BRAF tumors, metabolic reprogramming is a premise to support rapid proliferation. In T cells, the mitochondrial T cell activation inhibitor (TCAIM) is known to affect mitochondrial morphology but its effect on cellular metabolism is not well understood. Via mathematical modelling, I investigate the differential regulation of closely related cell lines. I present the first mathematical model for colon cancer and T cell metabolism, unraveling differential regulation between related cell lines. The model shows that CaCO2-BRAFV600Ecells are mostly downregulated compared to CaCO2-KRASG12Vand CaCO2-control. Additionally, it demonstrates the critical role of monocarboxylate transporter (MCT), especially for CaCO2-KRASG12V. Concerning T cells, I compare wild-type T cells to homozygous TCAIM T cells. This unveils that TCAIM homozygous cells have a mostly downregulated TCA cycle, validated by RNASeq data, and are less metabolically active than wild-type T cells. Furthermore, if the glycolytic flux is not sufficient to support lactate export and biomass production, the model reveals that the TCA cycle is reversed as it requires less regulation. Taken together, this work presents a novel approach to integrate data referring to metabolic and genetic regulation of metabolism. On this basis, we can now better discriminate the metabolic capacity of CaCO2-control, CaCO2-BRAFV600E, CaCO2-KRASG12V, wildtype CD8 T cells, and homozygous TCAIM CD8 T cells.

Le projet prend en compte la recherche historique et archiviste pour démontrer la nature interartistique de la poésie moderniste au Canada entre 1930 et 1960. En démontrant que les peintures, sculptures, et conceptions et illustrations de livres par les artistes Canadiens qui connaissaient et travaillaient de près avec les poètes modernistes du Canada ont informé et affirmé les sujets et l'aesthetique de la poésie, cette thèse de doctorat soutient que la di-mension sociale de la litérature moderniste au Canada soit considérée. Chapître un enquête sur la relation personnelle entre la poète Dorothy Livesay et la peintre Emily Carr et révèle les convergences aesthétiques et thématiques de leurs pein-tures et poèmes comme ils se rapportent au paysagisme et aux émotions. Chapître deux traite sur le sujet des petites revues de Montréal dans les années 1940 comme étant des sites interartistiques de collaboration parmi des artistes et des poètes et soutient que les peintures par ces artistes fournissaient des modèles de l'agence humaine pour les poètes. Full textWithin the world of library collection and preservation, the focus has historically been on the dual preservation of physical form and intellectual knowledge as represented through symbols (text), images, metadata, and organization of information. Certainly, the philosophy of the library to "enhance learning and ensure access to information for all" (ALA 2008) is paramount in directing resources and collection development in response to the needs of the user community. However, research and educational material may be found beyond the explicit materiality of books and the text they contain; understanding and recognizing spiritual and cultural properties implicitly located within certain library materials may provide additional fields of research, as well as learning and collaboration opportunities. Using adjunct literature within the field of museum studies and ethnography, this paper proposes to discuss how simultaneous recognition of intangible and tangible properties of certain objects is relevant to the library. The special collections library is perhaps the best place where such considerations can be made - it is a library dedicated to preserving not only copies of each work, but variant copies whose tangible and intangible properties may vary from one another - though some examples may apply to academic or public libraries as well. The scope of this undertaking will include a discussion of the adjunct and related literature from the museum world, identification of books, or types of books that have spiritual or cultural values to be considered, collection of cultural or religious viewpoints related to identified materials from members of one of those communities that create or use these items (Buddhist), and discussion of how such findings can inform the ongoing operations of special collections libraries and their mission as cultural memory institutions.

Full textFocusing on the self-appraisal, I explore some of the impression management (IM) tactics and interpersonal metadiscourse (MD) markers used by writers to manage the impressions they give off. Of particular interest is the extent to which (a) patterns of self-presentation, and (b) language background (NS/NNS) contribute to the affective response of the reader.
The self-appraisals, collected from 41 students taking a required undergraduate writing course, were grouped according to the affective response of independent readers. Patterns of IM and MD use were compared across positive- and negative-affect groups, and across NSs and NNSs.
The findings reveal distinct differences between affect groups in patterns of IM and MD use. Interestingly, NS/NNS differences did not contribute to the affective response of the reader, thus challenging the assumption that NS linguistic form is requisite to pragmatic competence.

Technical support for products after the sale is commonplace in today’s businesses. Original Equipment Manufacturers (OEMs) provide technical support to their dealer channel for resolution of complex product issues. Technical support staffing levels can vary by product type, product complexity, and production volumes, and case volumes. This research seeks a better understanding of appropriate staffing levels between three product lines for one OEM. The objective of this paper is to develop a model for monthly and weekly average case volumes for the three product lines, based off of historical case volume data. This data is used to predict a product line’s (platform’s) workload based off the month of the year. The output of each platform’s monthly case volume is then used in an optimization model to determine optimal staffing levels for each platform, based off the time of the year. The models developed for each platform use a linear relationship which regresses workload on a set of binary variable for the months of the year. Each of the models developed provided statistically significant coefficients for months which contain the platform’s highest workload. The outputs from these models are used in a mixed integer nonlinear programming optimization model to determine staff level of full time equivalent (FTE) employees at each platform. Each of the three scenarios utilized in this research provide similar trends and staffing levels for each of the three product lines. Results of this research are of interest for the management of technical support staffing.

Full textThis thesis focuses on the relationship between Dora Carrington and Lytton Strachey and offers three models for reading their unconventional relationship. Carrington was in love with the homosexual Strachey and the two lived together at Tidmarsh, and later Ham Spray House, for more than fourteen years. The three models make extensive use of primary sources, namely the letters and diaries of Carrington and Strachey. Furthermore, I draw on two seminal biographies of Carrington and Strachey written by Gretchen Gerzina and Michael Holroyd respectively. The first model I examine is a form of pederasty. I argue that, soon after they met, Carrington and Strachey began a friendship which was based on his educating her in a variety of ways. He served as a mentor both intellectually and sexually. Strachey was familiar with the concept of pederasty as a result of his involvement with the Cambridge Conversazione Society, better known as the Apostles, and used his knowledge to induct a rather naïve Carrington into new ways of thinking. This pederastic relationship also allowed Carrington a certain amount of freedom as it enabled her to pursue her art without the demands a heterosexual male would make of her. The second model for reading their relationship is that of parody. While Carrington and Strachey’s relationship resembles a heteronormative relationship, it can, at times, be read as parodic. I argue that they both subvert heteronormativity in humorous ways as a means to critique their parents’ Victorian marriages and to interrogate notions of masculinity vi and femininity. I discuss the roles they played within their domestic environment, and pay particular attention to how this intersected with Carrington’s artistic endeavours. This parodying of heteronormativity was, I suggest, also one of the only ways they could find of expressing the love they felt for one another. The last model I offer draws on theories of kinship. I examine how Carrington and Strachey resorted to familial constructions of descent as a means to veil the love they had for one another and to avoid criticism and ridicule from the Bloomsbury group and beyond. When they established a home at Tidmarsh, they altered their form of kinship to utilise principles of alliance. However, another shift took place with the introduction of Ralph Partridge, Carrington’s husband, and I argue that the terms they used to address each other changed to constructions, once again, of descent, at least until the dissolution of the Carrington-Partridge marriage. Carrington and Strachey’s relationship is often viewed as unconventional and she is often depicted as being utterly subservient towards him. However, the three models I have used demonstrate that their love was mutual. The models also reveal their relationship to be quite conventional in the manner in which Carrington and Strachey expressed their love for one another and how these expressions of love developed during the different phases of the life they spent together.

Full textENGLISH ABSTRACT: This research assignment deals with the breakdown in the Clausewitzian concept of the modern trinitarian structure of war. Martin Van Creveld in his book entitled, “The Transformation of War” written in 1991, discusses ‘Future War’ and the way in which wars will be fought. It will not be the highly technical interstate kind of war the West has been preparing for, but rather low intensity conflict where the lines between state, soldier and civilian become blurred, society becomes a war zone and the conflict becomes a more direct experience for the people. Colombia is a prime example of where this is occurring and the most salient manifestation of the low intensity conflict is the blurring of the lines between war and crime. There are left wing guerrillas fighting for social justice for the dispossessed population, but their tactics resemble crime and the government views them as terrorists. They run a self-sufficient organisation, one of the most profitable insurgent groups in the world largely funded through kidnap ransom payments. The right-wing paramilitaries are on a quest to cleanse Colombian society of the guerrillas and assassinate suspected guerrilla sympathisers. To complicate issues, both insurgent groups are involved in the drug trafficking trade, whether it be directly or by way of taxing land on which coca is grown. In this situation, war and crime have become inextricably linked and a distinction between the two is impossible on both practical and conceptual levels. However, if it is not crime and it is not war, but a complicated melange of the two, a new framework for analysis is required in order to attempt a solution.
AFRIKAANSE OPSOMMING: Hierdie navorsingsprojek gaan oor die ineenstorting van die Clausewitziaanse begrip van die moderne Trinitariese oorlogstruktuur. In sy boek, getiteld "The Transformation of War" wat in 1991 geskryf is, bespreek Martin van Creveld die 'toekomstige oorlog' en die wyse waarop oorloë gevoer staan te word. Dit sal nie die hoogs tegniese interstaatlike soort oorlog wees waarvoor die Weste hom voorberei nie, maar eerder 'n lae intensiteitskonflik waar die lyne tussen die staat, soldaat en burgerlike ineenvloei; die gemeenskap word 'n oorlogsone en die konflik word 'n direkte ervaring vir die bevolking. Ces concepts contribuent à la création des personnages hyper-culturelles, dont l'effectualisation limite la reconnaissance, l'inclusion, et la participation des in-betweens. This thesis uses mixed methods and datasets to explore how political news is perceived and shared within and across party lines in the context of the 2016 US presidential elections. We begin by examining the impact of political context versus article content on the reader through a crowdsourced study, and follow up with a large scale analysis of story sharing on the social platform Twitter to find cases where popularity transcends political affiliation. In Part One, we look at reader reactions. We investigate the question of trust in political news by performing a study online. We look at the impact of content features (reading level of the article) versus context clues (media brands) to find that political affiliation and brand outweigh all other aspects. In the second part of this thesis, we focus on reader actions. In particular, we look at how political news stories from the same time period are shared on the social media platform Twitter. As we found party loyalty and media brand perceptions to be significant influences on the reader's opinion of news, we are especially interested in examining emotional features that cause stories to become popular beyond political boundaries. Together, these two parts hope to form a more complete view of factors affecting and driving readers in an election cycle that is heavily influenced by media coverage, both traditional and new.

The major component of complex genomes is repetitive elements, which remain recalcitrant to characterization. Using maize as a model system, we analyzed whole genome shotgun (WGS) sequences for the two maize inbred lines B73 and Mo17 using k-mer analysis to quantify the differences between the two genomes. Significant differences were identified in highly repetitive sequences, including centromere, 45S ribosomal DNA (rDNA), knob, and telomere repeats. Genotype specific 45S rDNA sequences were discovered. The B73 and Mo17 polymorphic k-mers were used to examine allele-specific expression of 45S rDNA in the hybrids. Although Mo17 contains higher copy number than B73, equivalent levels of overall 45S rDNA expression indicates that transcriptional or post-transcriptional regulation mechanisms operate for the 45S rDNA in the hybrids. Using WGS sequences of B73xMo17 doubled haploids, genomic locations showing differential repetitive contents were genetically mapped, revealing differences in organization of highly repetitive sequences between the two genomes. In an analysis of WGS sequences of HapMap2 lines, including maize wild progenitor, landraces, and improved lines, decreases and increases in abundance of additional sets of k-mers associated with centromere, 45S rDNA, knob, and retrotransposons were found among groups, revealing global evolutionary trends of genomic repeats during maize domestication and improvement.
