Resumo: O objetivo desse estudo é fazer um rastreio da história do gosto do leitor-comum, do leitor-autor e do leitor-adaptador através dos séculos, e apresentar a possibilidade de diálogo estético e histórico internacional por meio da análise da natureza da dramaturgia e suas relações com as outras artes -ópera, música e dança-, e com aspectos da encenação e da carpintaria teatral, reflexões baseadas na teoria da "estética da recepção", conforme Thomas Bleicher, Hannelore Link, Alois Wierlacher, Dietrich Krusche e Reinold Werner. Para isso, será feito uma análise e pesquisa da viagem internacional através dos continentes de The Beggar's Opera, de John Gay (1728), uma moderna saga de anti-heróis, que ultrapassou a barreira da temporalidade e dos idiomas, e que reflete um processo de odisséia satírica, iniciado na Londres georgiana do início do século XVIII, com chegada triunfal até o século XX, com "recepções produtivas": Die Dreigroschenoper [A Ópera de Três Vinténs], de Bertolt Brecht (1928), na Berlim weimariana da década de 20; A Ronda dos Malandros, de Carla Civelli e Maurício Barroso, na São Paulo e seu mecenato industrial na passagem dos anos 40 aos 50; e Ópera do Malandro, de Chico Buarque (1978), no Rio de Janeiro militar dos anos 70, obras com nuances de nacionalismo, cosmopolitismo e universalismo, apoiadas nas idéias da incompreensível exploração humana do ciclo predatório (homo homini lupus) e da desumanizada exploração capitalista.
Abstract: This dissertation was carried out to investigate the history of bookishness of the common reader, the author-reader, and the adapter-reader troughtout centuries, and to present a possibility of international esthetic and historical dialog by analyzing the nature of dramaturgy and its relationship to other arts - opera, music and dance - and to aspects of stanging and to aspects to stanging and theater carpentry, reflections based on the theory of "esthetic reception" according to Thomas Bleicher, Hannelore Link, Alois Wierlacher, Dietrich Krusche e Reinold Werner. With that purpose in mind, one will carry out an analysis and research of the international travel around the continents of The Beggar's Opera, by John Gay (1728) a modren saga of antagonists, which outreached the limits of temporality and foreign languages, and which reflects a process of satiric odyssey, started in Georgian London at the beginning of the 18 th century, triumphally reaching the 20 th century, with "productive receptions": Die Dreigroschenoper [A Ópera de Três Vinténs], by Bertold Brecht (1928), in Weimar Berlin in the 1920 s, A Ronda dos Malandros, by Carla Civelli and Maurício Barroso, in São Paulo and its industrial patronage along the 1940s and 1950s, and Ópera do Malandro, by Chico Buarque (1978), in Rio de Janeiro military uproar along the 1970s, works with hues of nationalism, cosmopolitanism and universalism, supported by ideas of the incomprehensible human exploitation of the predatory cycle (homo homini lupus) and the ruthless capitalistic exploitation.

Resumo: As funções do coro na comédia de Aristófanes é o título e o tema desta tese que investiga o coro no gênero que foi praticado na Grécia antiga. Nossa investigação pautou-se no princípio de que os textos da Comédia Antiga que chegaram à nossa época foram compostos para serem postos em cena e, por isso mesmo, têm peculiaridades que devem ser levadas em consideração no momento da análise. Os textos das onze comédias de Aristófanes preservadas registram um roteiro ficcional que pode ser recuperado apenas pela leitura, como é comum em todo texto teatral, mas, além disso, os enredos das peças deixam gravadas as estratégias cênicas de realização concreta da obra em um espaço teatral específico. Assim, a partir do texto, é possível depreender tanto o enredo quando elementos da performance. Quando observamos o coro neste gênero, notamos que, nestes dois aspectos da comédia, ele desempenha importantes funções: na narrativa, ele assume diferentes papéis actanciais de acordo com as exigências do roteiro ficcional da peça; e, na performance, assume funções técnicas que viabilizam a apresentação da peça em um espaço concreto. A estrutura narrativa das obras foi estudada pelo viés teórico da semiologia do teatro. Para o estudo das funções do coro na performance das comédias, foram levadas em consideração as condições materiais dos festivais teatrais do século V a.C. - espaço amplo da orquestra, aglomeração no theatron, restrição a, no máximo, cinco atores para desempenhar todas as personagens da peça - e, a partir dessas exigências concretas, observou-se que o coro é um elemento estrutural do gênero frequentemente usado pelo poeta para solucionar os problemas ligados às dificuldades espaciais próprias da realização teatral da Atenas do século V a.C.
Abstract: The functions of the chorus in the Aristophanes comedy is the title and the main subject of this thesis which study the chorus in the genre practiced in the Ancient Greece. Our investigation grounded in the principle the texts of the Ancient Comedy that survived until our time were made to be put in scene and for that reason they have some singular characteristics that needed to be considered in the analysis. The texts of the eleven preserved comedies of Aristophanes show a fictional script that can be recovered only by reading, as usual in every theatric text, but, more than that, the plot of the plays keep record the scenic strategies applied in its concrete realization. So, from the text it is possible to comprehend the plot and elements of the performance. When we focus the chorus in this genre we can notice that - in theses two aspects of comedy - it has important functions: in the narrative it took different actantial roles according to the demands of the script of the play; and in the performance it took some technical roles that make possible the presentation of the play in a concrete space. The narrative structure of the plays was studied using as theoretical support the theater semiology. To study the functions of the chorus in the performance of the comedies the material resources in the theater festivals during the 5th century B.C. were considered - and from theses concrete demands, we could see that the chorus is a structural element of the genre frequently used by the poet to solve some problems caused by space difficulties characteristic of the theatral realization in the 5th century B.C. Resumo: O objetivo desta tese de doutorado é a análise das categorias do cômico na obra do poeta mineiro Carlos Drummond de Andrade. Como sua produção é acima de tudo cerebral e não busca sentimentalismos ou provocar a emoção através das palavras, o poeta trata do cômico de uma maneira muito particular. Usando um trabalho minucioso com as palavras que se deixa transparecer nos versos cuidadosamente escritos, Drummond consegue criar um tipo específico de cômico, seja através da linguagem coloquial, prosaica, seja através de termos e construções rebuscadas. A primeira está presente na maioria de seus poemas, e só deixa espaço para o rebuscamento em algumas obras pontuais, consideradas, por alguns críticos, como um retrocesso neoclássico. Embora o estudo da teoria do cômico em Drummond tenha recebido a atenção da maioria dos críticos, sua análise, muitas vezes, não vai além de alguns parágrafos, capítulos ou estudos pontuais de alguns poemas, ou seja, partes que integram um estudo maior, voltado a outra temática. Este estudo, ao debruçar-se, especificamente sobre a análise do cômico em suas origens, seus efeitos e suas construções na obra poética do autor, viu a necessidade de definir suas categorias, como a ironia e o humor, bem como suas formas de expressão, como a sátira, o chiste e a paródia a fim de que, não tendo a pretensão de trazer-lhes uma conceituação definitiva, a análise dos poemas seja realizada de modo satisfatório, à luz delas, ampliando os estudos acerca da lírica reflexiva do poeta e das formas de comicidade por ele empregadas
Abstract: The objective of this thesis is the analysis of the categories of comedy in the work of the poet Carlos Drummond de Andrade. As his production is above all brain and do not look for sentimentality or search emotion through words, the poet comes to the comic in a very particular way. Using a detailed work with words that make it clear in verses carefully written, Drummond manages to create a specific type of comic, whether through colloquial language, prose, either through the terms and ornate buildings. That is present in most of his poems, and only leaves room for the occasional pretensions in some works, considered by some critics as a throwback neoclassical. Although the study of the theory of comedy in Drummond has received the attention of most critics, their analysis often does not go beyond a few paragraphs, chapters or specific studies of some poems, or parts of a larger study, focused the other subject. This study, as look into, specifically, on the analysis of the comic in its origins, its effects and its buildings in the poetic works of the author, saw the need to define their categories, as irony and humor, and its forms of expression, as satire, the wit and parody in order that, having no claim to bring a definitive conceptualization, the analysis of the poems be performed satisfactorily in the light of them, extending the studies of the poet's reflective lyric and about the ways of humor employed by him

Abstract:
This thesis attempts to redress the drought of work on Dickens on comedy, which is surprising considering how often Dickens is thought of as a comic writer. The thesis uses Dickens to demonstrate problems with and resistance to existing theorizations of laughter, and attempts to develop a new way of thinking about laughter through Dickens. The thesis begins with a theoretical section, which is a discussion of existing discussions of laughter followed by an attempt to develop a new way of thinking about laughter by making use Alain Badiou’s concept of the ‘event.’ The thesis then moves to Section Two, in which these ideas are discussed alongside Dickens’s novels. Chapter Four attempts to show in a general way how Dickens and these discussions of laughter belong together, and how a certain moment in the nineteenth century that Dickens was a unique part of shows that new ways of discussing laughter are needed. Chapter Five argues that laughter in Dickens is not natural or spontaneous but part of constructing an idea of natural spontaneity. Pickwick Papers, it is argued, is the novel of retroactive causes, showing how laughter can create ideas of ‘nature’ which then appear to explain social behaviour such as laughter itself. Chapter Six tackles the relationship between laughter and anxiety. It argues that laughter creates order by ‘dealing’ with anxiety, but that this order it produces is profoundly unstable and has new anxieties. Barnaby Rudge is the novel which shows this in its particular historical context. The final chapter argues that Dickens’s writing can be called ‘comic’ in the terms that have been established throughout the thesis. Discussing Great Expectations, it argues that laughter is a plotting force that creates narratives and structures. Finally, the conclusion discusses changes that may have happened to laughter in the nineteenth century and what it means to find ourselves laughing at Dickens’s texts today.

Abstract:
A Amazônia, durante toda a sua história, teve como principal via de transporte a rede fluvial. A construção da rodovia BR-364 representou um marco para a região, visto que propiciou, a partir de então, o acesso às demais regiões brasileiras via terrestre. Propiciou a vinda de muitos migrantes de diversas partes do Brasil em direção à Amazônia. A Ponta do Abunã é uma das áreas que tem sua construção desde os períodos áureos da borracha amazônica, ainda no século XIX, mas que tem a chegada de um fluxo maior de migrantes principalmente a partir no final da década de 1970, acompanhando a construção da BR-364. A área até então é tida como pertencente ao Estado do Acre, pelo menos por parte deste Estado e da população que passa a residir naquela localidade. A indefinição quanto a que unidade federativa pertencia e tinha que se submeter, criou alguns problemas de ordem prática, como a questão de horários, já que o Estado acreano encontra-se uma hora a menos de diferença com o Estado de Rondônia. Na década de 1980 começam as discussões sobre a possessão da área, de um lado o Acre, do outro Rondônia. Passados cerca de dez anos, após algumas averiguações, foi decidido judicialmente que aquele território sempre pertenceu a Rondônia e que o Acre deveria retirar todos os órgãos instalados na localidade. Mesmo com a decisão, a relação com o Acre continuou, principalmente devido à maior proximidade geográfica, já que a Ponta do Abunã encontra-se mais distante da cidade de Porto Velho, além da existência de uma barreira geográfica - o Rio Madeira. Daí, analisar o território e as identidades que se produzem e se forjam nele são importantes para compreendermos como na Ponta do Abunã, ao longo de sua história, têm sido produzidas identidades territoriais.
The Amazon, throughout its history, has had the fluvial system as the main means of transportation. The opening of BR-364 Road represented a mark to the region as it provided access to the rest of Brazilian areas and it also made migration possible to several places towards the Amazon region. Ponta do Abunã village is one of the areas that has been developed since the rubber aural times, in the XIX century, and the settlement of a high flux of migrants from different areas around Brazil, mainly by the end of 1970, following the construction of BR-364 Road. The area, since then, has been considered as belonging to the State of Acre, by villagers and dwellers live there. Uncertainty regarding what State, Acre or Rondônia, that region would belong to has raised some geographical matters related to the time zone, as the State of Acre is one hour behind the State of Rondônia. In the 1980s, a discussion about the possession of the area between Acre and Rondônia came out and ten years later, it was legally decided that the State of Acre should all the public services installed in Ponta do Abunã Village. Despite that decision, commercial trades continued mainly because Ponta do Village is farther from the capital city, Porto Velho, than from Rio Branco, the capital city of Acre, and besides, because of the geographical barrier: the Madeira river. Indeed, analyzing the territory and its features that have been set is relevant to understand how, throughout its own history, that region has acquired territorial identities, as it was settled by migrants who initially founded an Acreanian territory, and later lived in a Rondonian territory - Ponta do Abunã Village. In doing so, it is noticeable the great role that the National State and the boundaries play on the construction of the territory.

Resumo: O teste do cometa é utilizado para estimativa de dano ao DNA nos protocolos de avaliação do risco toxicológico. É rápido, de baixo custo, de fácil realização, seguro e pode ser aplicado em diversos tecidos. Corantes fluorescentes são os mais empregados para o ensaio. É também descrita a utilização de sais de prata com vantagens operacionais, carecendo de sistemas automatizados validados para esta análise. Neste trabalho, desenvolveram-se dois algoritmos automatizados para a análise do cometa corado pela prata. Foram realizados ensaios do cometa para sete grupos com dois camundongos Balb/c machos cada, submetidos ao tratamento intraperitoneal com soro fisiológico (G1), e três doses crescentes de genotóxicos conhecidos: MNU (N-nitroso-N-metilurea) (G2 - 5 μg/g, G3 - 25 μg/g, G4 - 50 μg/g) e DEN (N-nitrosodietilamina) (G5 - 20 μg/g, G6 - 80 μg/g, G7 - 180 μg/g). As lâminas dos testes do cometa foram confeccionadas a partir do sangue total e coradas pela prata e SYBR Gold. As coradas pela prata foram analisados por três avaliadores e pelos algoritmos automatizados. Aqueles corados pela fluorescência foram analisados pelo software Comet IV®. Os avaliadores apresentaram alta correlação de suas estimativas. Os algoritmos se correlacionaram fortemente com a análise dos avaliadores, principalmente com os escores obtidos pela classificação visual. Houve alta concordância na avaliação da sua repetitividade. Os algoritmos apresentaram resultados semelhantes, não indicando superioridade entre si para a análise do teste. Todos os métodos avaliados se mostraram capazes de diferenciar as concentrações dos genotóxicos utilizados. Porém, os algoritmos desenvolvidos não apresentaram correlação com os resultados obtidos pelo sistema do teste do cometa corado pela fluorescência, sugerindo que os métodos de coloração discriminem diferentes estruturas... Abstract: The comet assay is used to estimate DNA damage in the protocols for toxicological risk assessment. It's fast, inexpensive, easily performed, safe and can be applied in most tissues. Fluorescent dyes are frequently used for testing. Silver salts have been reported with some operational advantages; however automated systems for its analysis were not sufficiently validated for this staining. In this study, we have developed two algorithms for automated analysis of the comet assay silver stained. The assay was performed for seven groups of two (Balb/c) mice each, treated with intraperitoneal saline (G1) and three different genotoxic doses: MNU (N-nitroso-N-methylurea) (G2 - 5 μg/g, G3 - 25 μg/g, G4 - 50 μg/g) and DEN (N-nitrosodiethylamina) (G5 - 20 μg/g, G6 - 80 μg/g, G7 - 180 μg/g). The slides of the comet assay were made with total blood and were stained with silver and SYBR Gold. The silver stained slides were analyzed by three evaluators and the automated algorithms. Those stained by fluorescence were analyzed through software Comet IV®. Evaluators showed high correlation of their estimates. Both algorithms were correlated strongly with the evaluators' analysis mainly on the scores obtained by visual classification. Moreover, their agreement was high when assessed by repetitivity. The algorithms showed similar results, demonstrating no superiority to each other for the analysis of the test. All methods evaluated proved able of differentiating genotoxic concentrations used. However, the algorithms were not correlated with the results obtained by the automated system of comet assay fluorescence stained, suggesting that the staining methods linkage different genomic structures or with different affinities. The automated algorithms developed proved valid for the direct analysis of comet assay silver stained, allowing their use in genotoxicity research

Resumo: A mutagenicidade é um efeito tóxico que danifica o material genético da célula, causando alterações no DNA ou no cromossomo. Existem inúmeras substâncias presentes em complexos de efluentes domésticos e industriais que podem causar sérios prejuízos no material genético. Testes biológicos de toxicidade e genotoxicidade são indispensáveis para estudos de monitoramento, que tem por finalidade avaliar a extensão da poluição ambiental e as reações dos organismos vivos frente a esta poluição. Águas que recebem descargas de diferentes fontes poluidoras, como efluentes industriais e urbanos, na sua maioria, são impactadas por misturas complexas compostas de químicos diversos. O córrego Santa Gertrudes e rios associados recebem efluentes industriais, de uma série de indústrias cerâmicas, e domésticos do município de Santa Gertrudes, SP. Os efluentes cerâmicos possuem efeito tóxico, sobretudo devido à presença de metais resultantes do processo de esmaltação dos revestimentos cerâmicos. As espécies Allium cepa e Oreochromis niloticus têm se mostrado bastante eficiente na avaliação da genotoxicidade ambiental. Neste trabalho foi avaliada a qualidade da água dos córregos Santa Gertrudes, Caju, Itaqui e do ribeirão Claro, por meio de testes desenvolvidos com estes bioindicadores. Pelos resultados obtidos, pudemos observar que os compostos presentes nas águas destes rios apresentam potencial tóxico, citotóxico, genotóxico e mutagênico para os dois organismos-teste. O teste com bulbos de A. cepa mostrou uma inibição no desenvolvimento das raízes, devido às substâncias tóxicas presente em águas coletadas após o despejo de efluentes. As amostras dos pontos correspondentes ao local de emissão de efluentes também induziram elevados índices de AC e MN em células meristemáticas de A. cepa, para todas as coletas realizadas. Abstract: Mutagenicity is a toxic effect that damages the genetic material of the cell, causing alterations in either the DNA or the chromosome. There are several substances present in complexes of domestic and industrial effluents that may cause serious damage to the genetic material. Biological tests of toxicity and genotoxicity are essential for monitoring studies, which aims at assessing the extension of the environmental pollution and the reaction of the living organisms to this pollution. Waters that receive discharges from different sources of pollution, such as urban and industrial effluents, are mostly impacted by complex mixtures composed by several chemicals. Santa Gertrudes stream and associated rivers receive domestic effluents from the municipality of Santa Gertrudes (SP, Brazil) and industrial effluents from several ceramic industries. The ceramic effluents have toxic effects mainly due to the presence of metals resulting from the enameling process of ceramic coating. Allium cepa and Oreochromis niloticus species are very efficient in evaluating environmental genotoxicity resulting from pollution. In this study, the quality of the water of the Santa Gertrudes, Caju, Itaqui and Claro streams was assessed by tests developed with these bioindicators. Based on the results, we could observe that the compounds present in the waters are potencialy toxic, cytotoxic, genotoxic and mutagenic to both test organisms. The tests with bulbs of A. cepa showed an inhibition in root development due to the toxic substances present in the waters collected after the discharge of the effluents. Likewise, the samples from the effluent emission sites induced high indices of CA and MN in meristematic cells of A. cepa for all the collections made. The collection site located after the confluence of the Santa Gertrudes and Claro streams presented an increase in the alteration indices, especially for the August... Resumo: Os antioxidantes, principalmente os encontrados na dieta, são agentes responsáveis pela inibição e redução dos danos oxidativos causados pelas espécies reativas nas células. A cisplatina (cDDP), um potente agente antineoplásico utilizado com frequência no tratamento de tumores sólidos, tem seu uso clínico limitado devido aos efeitos adversos como nefrotoxicidade, genotoxicidade e supressão da medula óssea. Como a geração de espécies reativas pode resultar em danos ao DNA pela ação destas espécies ou indiretamente via produtos de degradação da peroxidação lipídica, nosso estudo avaliou o possível efeito protetor da adição da polpa de maracujá amarelo na dieta de ratos espontaneamente hipertensos (SHR) e Wistar normotensos, contra a nefrotoxicidade e genotoxicidade induzida pela cDDP. Os resultados obtidos nas condições experimentais estabelecidas neste trabalho mostraram que a cDDP promoveu um aumento dos níveis das substâncias reativas aos ácido tiobarbitúrico, bem como decréscimo nos níveis de glutationa nos rins dos animais SHR, reforçando a hipótese que a peroxidação lipídica está relacionada com o mecanismo de nefrotoxicidade. O tratamento com doses múltiplas da polpa de maracujá amarelo não alterou os outros parâmetros da função renal no período analisado. Embora o efeito protetor da polpa de maracujá contra os danos induzidos ao DNA em células renais e hepáticas não tenha sido evidente nos resultados obtidos pelo Ensaio do Cometa, a polpa foi eficaz na redução de micronúcleos induzidos pela cDDP. Assim, umas das hipóteses para a redução da pressão arterial sistólica observada neste trabalho, poder ser atribuída à presença de alguns compostos fenólicos e outros ainda não identificados na polpa. Visto que, a interação entre os fitoquímicos presentes na polpa de maracujá demonstraram eficácia na redução da mutagenicidade e efeitos protetores antioxidantes.
Abstract: The antioxidants, particularly those found in the diet, are responsible for inhibiting and reducing the damage caused by reactive species in the cells. Cisplatin (cDDP), a potent anticancer agent often used in the treatment of solid tumors, has limited clinical use due to adverse effects such as nephrotoxicity, genotoxicity and myelosuppression. As the generation of reactive species can result in DNA damage by direct action of this or indirectly by degradation products of the lipid peroxidation, our study evaluated the potential protector of adding yellow passion fruit pulp to the diet of spontaneously hypertensive rats (SHR) and normotensive wistar rats, against the nephrotoxicity and genotoxicity induced by cDDP. The results showed that the cDDP promoted an increase in the levels of thiobarbituric acid reactive substances and a decrease in the levels of glutathione in the kidneys of the animals, reinforcing the hypothesis that lipid peroxidation be related to the mechanism of nephrotoxicity. The treatment with multiple doses of passion fruit pulp did not change the others parameters of renal function in the schedule. Although the protective effect of passion fruit pulp against the damage caused to the DNA of kidney and hepatic cells has not been evident in the results obtained by Comet assay, the pulp was effective in the reduction of micronuclei induced by cDDP. One of the hypotheses for the reduction in systolic blood pressure observed in the present study attributed this effect to the presence of some phenolic compounds and others not yet identified in the passion fruit pulp. Furthermore, the interaction between the phytochemicals of the pulp demonstrated effectiveness in reducing the mutagenic and antioxidant effects.

Resumo: Na década de 80, em São Paulo, atores grupos de teatro e diretores começaram a aproximar-se da máscara e da técnica do palhaço, circence para ampliar os recursos da interpretação. Essa busca efetivou-se tanto com os grupos procurando o circo, como tambem com a vinda de professores e diretores do estrangeiro, que já trabalhavam nessa vertente, para o Brasil. Entre eles, destaca-se o diretor italiano Franco Zigrino. Na capital Paulista ele desenvolveu cursos com máscara, além de realizar montagens de espetáculos. Nesse processo formaram-se vários profssionais que deram continuidades à aproximação da atuaçao teatral com máscaras e hoje são referências importantes no teatro paulistano. No primeiro momento, esta pesquisa investiga as origens e características da pedagogia das máscaras e sua contribuição para o teatro, auxiliando na formaçãode atores. Em seguida, realiza uma busca através de documentos oficiais, matérias de jornais e entrevistas com seus alunos e atores a respeito da passagem do diretor Francesco Zigrino no país e as principais características deixadas pelo seu trabalho. Como parte final, são colocadas reflexões acerca destas influências, que resultam em considerações sobre as características da pedagogia trazida pelo italiano e como elas foram absorvidas e adaptadas à realidade histórica e cultural do Brasil. As considerações finais enfantizam as influências do trabalho do diretor italiano na formação de atores.
Abstract: In the 1980's in São Paulo, actors, theatre groups and directors began to get closer to studyings masks and to the technique of the circus clown, so as to expand acting resources. This search was carried out both with the groups in search of the circus, and also with the teachers and directors arriving in Brazil from abroad, who had already worked with this art. Among them was the Italian director Francesco Zigrino. In the capital of São Paulo he developed courses on masks while also producing shows. Many professionals were trained in this process, and continued the work of bringing together treatrical acting and masks. Today, they have become important references on the São Paulo theatre scene. As a starrer, this research investigates the origins and characteristicas of the pedagogy of masks and its contribution th the theatre, while also helping to develop actors. Then, it searches through official documents, news paper articles and interviews with Francesco's students and actors about the journey of the director through Brazil, and the main characterístics that his work has left. Finally, there will be some reflection on those influences that lead to considerations as to the characteristics of the pedagogy brough by the Italian director, and as this was absorved and adapted to the historical and cultural reality of Brazil. The final considerations emphasize the influences of the work of the Italian director in the formation of actors.

Resumo: Devido às crescentes expansões demográficas e industriais observadas nas últimas décadas, o meio ambiente tem recebido uma carga significativamente crescente de efluentes domésticos, industriais e agrícolas, causando impactos severos nos ecossistemas e um potencial comprometimento à saúde humana. Dentre os efluentes domésticos, podemos citar uma gama de poluentes, tais como químicos de diversas categorias, além de contaminações por agentes biológicos diversos. Já os efluentes industriais contêm poluentes orgânicos e/ou inorgânicos, dependendo da atividade industrial. Baseando-se nestes dados, este trabalho teve como objetivo investigar, por meio de ensaios biológicos com dois organismos-teste, a possível presença de contaminantes com potencial citotóxico, genotóxico e mutagênico, que são despejados ao longo do rio Preto, inclusive na Represa Municipal de São José do Rio Preto. O material biológico utilizado neste estudo constituiu-se de sementes de Allium cepa (cebola) e peixes da espécie Oreochromis niloticus (Tilápia). Coletas de águas foram realizadas, sazonalmente, nos meses de agosto de 2006 e 2007 (estação seca) e março de 2007 e 2008 (estação chuvosa), em seis pontos distintos: Ponto 1 (P1), 8 km antes do represamento; Ponto 2 (P2), 1 km antes do represamento; Ponto 3 (P3), local de despejo do esgoto; Ponto 4 (P4), margem oposta do despejo do esgoto; Ponto 5 (P5), saída do represamento; Ponto 6 (P6), 1 km após o represamento. Análises químicas foram realizadas para todas as coletas realizadas. Para a realização do estudo, 100 sementes de Allium cepa foram submetidas à germinação, em placa de Petri, em amostras de águas coletadas nos seis diferentes pontos do rio Preto, em água ultra pura (controle negativo) e em uma substância reconhecidamente aneugênica (Trifluralina - controle positivo), sempre à temperatura ambiente... Abstract: Due to increasing population and industrial expansion observed in recent decades, the environment has received a significant increased burden of domestic industrial and agricultural sewerage, which can cause severe impacts on ecosystems, and a potential damage to human health as well. A wide range of harmful pollutants can be found in domestic effluent, such as chemicals from various categories, in addition to contamination by various biological agents. On the other hand, industrial effluents contain organic and / or inorganic pollutants, depending on industrial activity. Based on these data, this study aimed to investigate, by means of biological tests with two test-organism, the possible presence of contaminants with cytotoxic, genotoxic and mutagenic potential, which are dumped along the Preto river, an important river that flows in the region of Sao Jose do Rio Preto/SP. The biological material used in this study consisted of seeds of Allium cepa (onion) and one specie of fish (Tilapia: Oreochromis niloticus). Water samples were taken seasonally in August 2006 and 2007 (dry season) and March 2007, and 2008 (rainy season), in six distinct sites: Site 1 (S1), 8 km before the damming, Site 2 (S2), 1 km before the damming, Site 3 (S3), place of sewerage discharge; Site 4 (S4), opposite margin of sewage discharge, Site 5 (S5), end of the damming; Site 6 (S6) 1 km after damming. Chemical analyses were performed for all collected samples. Abstract:
This thesis focuses on the sudden proliferation of a highly popular genre in French cinema over the past two decades. While genre cinema, French and otherwise, and especially comedy have historically been under-examined in France, this study contributes to a gradual shift towards the application of more traditionally Anglophone cultural and gender studies approaches to the analysis of French cinema. It also looks at the question of generic evolution and the relationship between domestic practice and Hollywood paradigms, including the way in which the national cinema is being reconceptualised in the face of an increasingly transnationally conceived film market. Major lines of enquiry thus concern both the rom-com’s French specificity, including the extent to which the genre is ‘naturalised’ and/or exported, and its representational politics, particularly with regard to gender. Is it appropriate to see the rom-com as a feminised genre in France and, if so, in what sense? How does the genre negotiate major contemporary social shifts, particularly those linked to gender? Notable here are the extension of singlehood and the increased acceptability of female autonomy; the concomitant decline of marriage; and the increased range of ‘acceptable’ gender roles today - especially the rise of the professional woman, which in turn has implications for masculinity and for the family unit. Do films broadly reflect and embrace such shifts or do they resist, critique or downplay them? Finally, how are these questions inflected by considering production and reception contexts, as well as the issue of gendered authorship? In seeking to answer such questions, the thesis encompasses analysis of both formal aspects of the genre and specific narrative details. In so doing it hopes to pinpoint some of the ways in which the French rom-com has been mediating perceptions of key social trends over the past two decades, both at home and ultimately globally.

Abstract:
There is a tendency amongst scholars to view comedic elements common to Roman Comedy, such as the tricky subordinate or the nagging wife, as part of a developing Western Comedic Tradition. The appearance of these characters in Medieval Japanese Kyōgen, a comedic art-form unconnected with Western Comedy, challenges this viewpoint and suggests that they are part of a wider comedic identity. This thesis compares and contrasts Roman Comedy and Kyōgen through their techniques of fear-alleviation, exploring the manner in which each culture addresses social anxieties. The first chapter explores the comedic master-slave/servant relationship through the medium of the tricky slave/servant. It examines how the motif of the tricky subordinate is used to alleviate contemporary fears of authority figures. Chapter 2 considers the other half of this relationship, focusing on authority’s fear of rebellion and how this is addressed through the loyal and/or stupid slave/servant. Chapter 3 explores the depiction of religious and supernatural figures in the two comedic forms and examines the methods by which these awe-inspiring beings are portrayed humorously and rendered harmless. The fourth chapter reflects on the treatment of illness in Roman Comedy and Kyōgen. It discusses how contemporary social anxieties regarding blind men (Medieval Japan) and the stigma of insanity and epilepsy (Rome) are alleviated through the humorous comedic depiction of blind and insane/epileptic characters. Chapter 5 explores the comedic presentation of professional figures. This chapter contrasts the boastful character of the comedic soldier of Roman Comedy with the braggart priest of Kyōgen. In Chapter 6, the focus is on the misogynistic treatment of wives in both comedic art-forms. This chapter explores contemporary fear of wives and how this fear is alleviated through their negative portrayal in comedy. This thesis finds that there is a strong correlation between Roman Comedy and Kyōgen, both in the types of social anxiety which they seek to alleviate and the methods by which they seek to accomplish this. It also finds that the motif of the tricky subordinate and the nagging wife are not just Western phenomena but that they are also present in the Eastern Comic Tradition. The comparison of Roman Comedy with Kyōgen, an unrelated comedic form, leads to an enhanced understanding of the role which these characters play in alleviating social anxiety. It also enables the consideration of stock characters in Roman comedy from a wider viewpoint, presenting an opportunity for scholars to re-evaluate characters such as the tricky subordinate and the nagging wife as products of a wider, universal comic tradition.

Abstract: The present research aims at recording and understanding the trajectory of the Projeto Comédia Popular Brasileira (CPB) and, consequently, of the Fraternal Companhia de Arte e Malas-Artes, through the characters created during the 15 years of esthetical research (1993-2008). The objects of this analysis are the specific projects developed at each phase of the CPB Project, their respective plays and spectacles, the conception of the characters and actors' work and formation as well as their relation with their main target: the audience. During its first phase (1993-1997) - SEE, Fraternal creates Brazilian-type characters, influenced by the comedians Martins Pena, Artur Azevedo and Ariano Suassuna and reestablishing the dialogue with the fixed types of commedia dell'arte. In its second phase (1998-2001) - HEAR, the character-types give space to characters inspired by medieval popular parties/festivals, based on the study of Mikhail Bakhtin. Finally, in its third phase (2002-2008) - IMAGINE, acrobat actors introduce the characters through narration and acting. In this period, the Society deepens its practice through the scenic interplay between characters, actors and narrators for the dramaturgical and interpretive construction of its spectacles, which was inspired by Bertolt Brecht e Luigi Pirandello. The choice of characters as interlocutors of Brazilian people history and wishes reflected, since the very beginning, the specific world and popular culture view taken by Fraternal which is opposed to the dominant class' one. Abstract:
This research explores psychological therapists’ experiences of humour in sessions with clients diagnosed with a terminal illness. In considering the extensive research uncovered involving humour and death, comparatively little was found in the field of terminal illness, humour and the psychological therapies, and none specifically on therapists’ experiences of these phenomena. Bruner’s (1991, 2004) narrative approach is used to examine six psychological therapists’ experiences which elicited: participating therapists’ personal experiences of humour compared to those experiences with clients; how preconceptions of working in terminal care shaped their experiences of humour once they were experienced therapists; the nature of working with terminally ill clients; the nature of humour as a hindrance and/or help; the differences between humour with clients in terminal settings compared to other settings; and finally, what therapists have learned through their experiences. Analysis of the findings is conducted by looking at both the content and structure of participants’ narratives, paying close attention to character, plot, temporality, and situatedness. The nature of this research and the findings and their implications are discussed and critiqued, before further research is recommended and concluding remarks are made.

Abstract:
The late Middles Ages saw the development in Europe of increasingly complex, ambitious, and self-conscious forms of creative literature. In the works of poets such as Dante, Petrarch and Chaucer new models of authorship and poetic identity were being explored, new kinds of philosophical and aesthetic value attributed to literary discourse. But these creative developments also brought with them new dangers and tensions, a sense of guilt and uncertainty about the value of creative literature, especially in relation to the dominant religious values of late medieval culture. In this thesis I explore how these doubts and tensions find expression in Chaucer’s poetry, not only as a negative, constraining influence, but also as something which contributes to the shape and meaning of poetry itself. I argue that as Chaucer develops his own expansive, questioning poetics in The House of Fame and The Canterbury Tales, he problematises the principle of allegory on which the legitimacy of literary discourse was primarily based in medieval culture and the final fragments of The Canterbury Tales see Chaucer struggling, increasingly, to reconcile the boldness and independence of his poetic vision with the demands of his faith. This struggle, which emerges most strongly and polemically in the final fragments, I argue, runs in subtle and creative forms throughout the whole of Chaucer’s work. By seeing Chaucer in this light as a poet not of fixed, but of conflicted and vacillating intentions – a poet productively caught drawn between ‘game’ and ‘earnest’, radical ironies and Boethian truths – I attempt to account, in a holistic manner, for the major dichotomies that characterise both his work and its critical reception.

This thesis serves as a preliminary design study for the combination of a flight-proven ion optics system (SWIM) with a conversion surface to create a small energetic neutral atom (ENA) sensor. It is planned to use this sensor as ENA sensor for the DFP-SCIENA instrument on Comet Interceptor. Due to the nature of the Comet Interceptor mission (ESA F-class mission with a maximum launch mass of 1000 kg) the development time for a new sensor that meets the size and weight restrictions is limited. The proposed combination of SWIM with a conversion surface is based on a proven ion optics design and should result in a compact sensor design. The main goal of this thesis was to simulate different conversion surface designs and evaluate their compatibility with the SWIM instrument. During this process the different designs were optimised based on the intermediate simulation results. The simulation process was performed by using SIMION to calculate particle trajectories.  In the end, two different conversion surface designs yielded promising results. With both designs detailed simulations and data analysis were conducted to determine the different properties of the two designs. One of these designs was chosen to be further investigated for use on the Comet Interceptor mission.

Resumo: A exposição do homem às substâncias danosas, sejam estas substâncias de origem natural ou sintética, vem crescendo durante as últimas décadas. Esta exposição é capaz de induzir diversos efeitos deletérios nos organismos expostos, provocando diversas doenças e até mesmo a morte. Da mesma forma que aumenta a quantidade de substâncias promotoras de impactos ambientais, também crescem as pesquisas que buscam por novas substâncias que sejam capazes de proteger os organismos destes efeitos deletérios. Esse trabalho tem como objetivo avaliar a atividade citotóxica, genotóxica e antigenotóxica, mutagênica e antimutagênica de venenos de Hymenoptera (especificamente da abelha Apis mellifera e da vespa Polybia paulista), em diferentes concentrações, utilizando para essa avaliação o sistema teste de HepG2. Foi utilizado o ensaio do MTT para se avaliar a citotoxicidade tanto do veneno de A. mellifera como de P. paulista. As concentrações consideradas não citotóxicas (1, 5 e 10μg/mL de veneno de vespa e 0,1, 0,05 e 0,01 μg/mL do veneno de abelha) foram utilizadas para se avaliar o potencial genotóxico (ensaio do cometa) e mutagênico (teste do micronúcleo). Estas concentrações não citotóxicas mostraram-se genotóxicas e mutagênicas para o sistema teste utilizado. Foram utilizadas outras concentrações, mais baixas que as utilizadas nos testes de genotoxicidade e mutagenicidade (1ng/mL, 100pg/mL e 10pg/mL de veneno de vespa e 10pg/mL, 1pg/mL e 0,1pg/mL para o veneno de abelha), para a realização dos testes de antigenotoxicidade e antimutagenicidade com células HepG2. As concentrações utilizadas nesses ensaios mostraram que ambos os venenos, ao invés de inibirem e/ou diminuirem o efeito genotóxico e mutagênico da substância metilmetano sulfonato, aumentaram ainda mais os danos causados por esta substância... Abstract: Human exposure to harmful substances, natural or synthetic, is increasing in the last decades. This exposure is able to induce several deleterious effects on the exposed organisms, causing several diseases and even death. As the amount of substances that promote environmental impacts increases, researches seeking for new substances that are able to protect the organisms against these deleterious effects have also increased. This study aimed to evaluate the cytotoxic, genotoxic and antigenotoxic, mutagenic and antimutagenic of Hymenoptera venoms (specifically of the bee Apis mellifera and the wasp Polybia paulista), in different concentrations, using for this evaluation the HepG2 test system. The MTT assay was used to assess the cytotoxicity the venom of A. mellifera and P. paulista. The concentrations considered non cytotoxic (1, 5 and 10μg/mL of the wasp venom and 0.1, 0.05 and 0.01 μg/mL of the bee venom) were used to evaluate the genotoxic (comet assay) and mutagenic potential (micronucleus test). These non cytotoxic concentrations were genotoxic for the test system used. Other concentrations were used, lower than the ones used in the genotoxicity and mutagenicity tests (1ng/mL, 100pg/mL and 10pg/mL of the wasp venom and 10pg/mL, 1pg/mL and 0,1pg/mL for the bee venom), in order to perform the antigenotoxicity and antimutagenicity tests with HepG2 cells. The concentrations used in these assays showed that both venoms, instead of inhibiting and/or decreasing the gentoxic and mutagenic effects of the substance methylmethane sulfonate, increased even more the damages caused by this substance. In order to verify which would be the compound responsible for the effects registered for the venoms, it was also evaluated the effect of phospholipase A2 (PLA2 from A. mellifera)... Los movimientos voluntarios del cuerpo son controlados por el sistema nervioso central y periférico a través de la contracción de los músculos esqueléticos. La contracción se inicia al liberarse un neurotransmisor sobre la unión neuromuscular, iniciando la propagación de un biopotencial sobre la membrana de las fibras musculares que se desplaza hacia los tendones: el Potencial de Acción de la Unidad Motora (MUAP). La señal electromiográfica de superficie registra la activación continua de dichos potenciales sobre la superficie de la piel y constituye una valiosa herramienta para la investigación, diagnóstico y seguimiento clínico de trastornos musculares, así como para la identificación de la intención movimiento tanto en términos de dirección como de potencia. En el estudio de las enfermedades del sistema neuromuscular es necesario analizar el nivel de actividad, la capacidad de producción de fuerza, la activación muscular conjunta y la predisposición a la fatiga muscular, todos ellos asociados con factores fisiológicos que determinan la resultante contracción mioeléctrica. Además, el uso de matrices de electrodos facilita la investigación de las propiedades periféricas de las unidades motoras activas, las características anatómicas del músculo y los cambios espaciales en su activación, ocasionados por el tipo de tarea motora o la potencia de la misma. El objetivo principal de esta tesis es el diseño e implementación de protocolos experimentales y algoritmos de procesado para extraer información fiable de señales sEMG multicanal en 1 y 2 dimensiones del espacio. Dicha información ha sido interpretada y relacionada con dos patologías específicas de la extremidad superior: Epicondilitis Lateral y Lesión de Esfuerzo Repetitivo. También fue utilizada para identificar la dirección de movimiento y la fuerza asociada a la contracción muscular, cuyos patrones podrían ser de utilidad en aplicaciones donde la señal electromiográfica se utilice para controlar interfaces hombre-máquina como es el caso de terapia física basada en robots, entornos virtuales de rehabilitación o realimentación de la actividad muscular. En resumen, las aportaciones más relevantes de esta tesis son: * La definición de protocolos experimentales orientados al registro de señales sEMG en una región óptima del músculo. * Definición de índices asociados a la co-activación de diferentes músculos * Identificación de señales artefactuadas en registros multicanal * Selección de los canales mas relevantes para el análisis  Extracción de un conjunto de características que permita una alta exactitud en la identificación de tareas motoras Los protocolos experimentales y los índices propuestos permitieron establecer que diversos desequilibrios entre músculos extrínsecos del antebrazo podrían desempeñar un papel clave en la fisiopatología de la epicondilitis lateral. Los resultados fueron consistentes en diferentes ejercicios y pueden definir un marco de evaluación para el seguimiento y evaluación de pacientes en programas de rehabilitación motora. Por otra parte, se encontró que las características asociadas con la distribución espacial de los MUAPs mejoran la exactitud en la identificación de la intención de movimiento. Lo que es más, las características extraídas de registros sEMG de alta densidad son más robustas que las extraídas de señales bipolares simples, no sólo por la redundancia de contacto implicada en HD-EMG, sino también porque permite monitorizar las regiones del músculo donde la amplitud de la señal es máxima y que varían con el tipo de ejercicio, permitiendo así una mejor estimación de la activación muscular mediante el análisis de los canales mas relevantes.
Voluntary movements are achieved by the contraction of skeletal muscles controlled by the Central and Peripheral Nervous system. The contraction is initiated by the release of a neurotransmitter that promotes a reaction in the walls of the muscular fiber, producing a biopotential known as Motor Unit Action Potential (MUAP) that travels from the neuromuscular junction to the tendons. The surface electromyographic signal records the continuous activation of such potentials over the surface of the skin and constitutes a valuable tool for the diagnosis, monitoring and clinical research of muscular disorders as well as to infer motion intention not only regarding the direction of the movement but also its power. In the study of diseases of the neuromuscular system it is necessary to analyze the level of activity, the capacity of production of strength, the load-sharing between muscles and the probably predisposition to muscular fatigue, all of them associated with physiological factors determining the resultant muscular contraction. Moreover, the use of electrode arrays facilitate the investigation of the peripheral properties of the active Motor Units, the anatomical characteristics of the muscle and the spatial changes induced in their activation of as product of type of movement or power of the contraction.The main objective of this thesis was the design and implementation of experimental protocols, and algorithms to extract information from multichannel sEMG signals in 1 and 2 dimensions of the space. Such information was interpreted and related to pathological events associated to two upper-limb conditions: Lateral Epicondylitis and Repetitive Strain Injury. It was also used to identify the direction of movement and contraction strength which could be useful in applications concerning the use of biofeedback from EMG like in robotic- aided therapies and computer-based rehabilitation training.In summary, the most relevant contributions are:§The definition of experimental protocols intended to find optimal regions for the recording of sEMG signals. §The definition of indices associated to the co- activation of different muscles. §The detection of low-quality signals in multichannel sEMG recordings.§ The selection of the most relevant EMG channels for the analysis§The extraction of a set of features that led to high classification accuracy in the identification of tasks.The experimental protocols and the proposed indices allowed establishing that imbalances between extrinsic muscles of the forearm could play a key role in the pathophysiology of lateral epicondylalgia. Results were consistent in different types of motor task and may define an assessment framework for the monitoring and evaluation of patients during rehabilitation programs.On the other hand, it was found that features associated with the spatial distribution of the MUAPs improve the accuracy of the identification of motion intention. What is more, features extracted from high density EMG recordings are more robust not only because it implies contact redundancy but also because it allows the tracking of (task changing) skin surface areas where EMG amplitude is maximal and a better estimation of muscle activity by the proper selection of the most significant channels.

This study is aimed at offering an overview of the entire dramatic oeuvre of VidaOgnjenović to date within the wider context of the contemporary Serbian dramaturgy and literarytradition. This objective entails an analysis of the literary-dramatic procedure used, as well as thethematic-motif determinants, distinctive features in regard to genre and style, linguisticexpression, characterisation, attitude towards historical, political, ideological, cultural, social andother societal phenomena. In order to accomplish the objective of research thus formulated, itwas necessary to form a singular coordinate system. One coordinate axis represents majorauthors and their representative works, which bear crucial significance for the development ofthe contemporary Serbian drama from World War II to the first decade of this century, while theother axis represents literary tradition as an unambiguous point of origin and creative inspiration.The study is divided into three organically related units: ‘Contemporary Serbian drama’,‘Dramatic oeuvre of Vida Ognjenović’ and ‘Literary tradition as creative inspiration’.

Abstract:
The volatile and noble gas inventories of comets are poorly understood relative to asteroids. Interplanetary dust particles (IDPs) and micrometeorites are thought to originate predominantly from comets, therefore they provide a means of investigating cometary volatiles which can be compared to unequilibrated meteorites and help us understand the relationship between comets and asteroids. In preparation for Xe analyses, we have determined the bulk elemental compositions, H, C, N and O isotopic compositions and carbonaceous content of 26 IDPs, in order to correlate any Xe components with particular, possibly primitive cometary, characteristics. The IDPs as a whole show great heterogeneity in their mineralogy and isotopic compositions, in accordance with other studies. We observed some extreme isotopic enrichments in D and 15N, including correlations between localised D enrichments and C/H ratios, leading to a revision of the organic endmember components devised by previous authors. The Raman parameters we report also agree with other studies on the highly disordered nature of the carbon in IDPs relative to chondrites. We observed two unusual IDPs containing previously unreported features; including a Pb-chromate and large Ti-oxide grain in a highly refractory IDP, and an extremely isotopically anomalous terminus particle in a hybrid IDP; which could be an interstellar grain impact. Perhaps the most interesting particle we analysed here is composed of 50 wt% C, is highly N-rich and contains a large abundance of presolar silicates, some of which appear to be GEMS. This C-rich “ultra-carbonaceous” IDP could provide an important link to the organic material in primitive chondrites and comets. We also report the first measurement of Xe in three individually analysed IDPs, one of which is only 8 microns in diameter, and thirteen AMMs up to 150 microns in diameter. The AMMs all yielded gas, with some enabling Xe isotopic compositions to be roughly determined. These were mostly a mixture of trapped Q, solar wind and terrestrial air, with some showing 129I-derived, presolar HL and fission-derived Xe influence, similar to the range in isotopic compositions observed in chondrites. The 132Xe concentrations in the AMMs also span the range observed in chondrites. The ultra-carbonaceous, N-rich and highly isotopically anomalous IDP has yielded a 132Xe concentration, normalised to chondritic IOM content, in the same range as IOM in primitive chondrites. This suggests a possible common Q-carrier distribution in comets and asteroids, therefore further indicating wide-scale mixing in the solar nebula.

Founded in 1836, Union Theological Seminary sought to be a school where moderate theological views could be taught in the heart of America’s most important city, New York City. Scholars have recognized Union’s role in the fundamentalist-modernist controversy of the 1920s, but few have noticed its impact in Chinese Protestantism. From 1911-1949, 196 Union alumni - an average of five students for each graduating class - went to China. Thirty-nine of these former students were Chinese nationals. Many of these Chinese graduates became key leaders in China’s Young Men’s Christian Association, the Christian colleges in Republican China and the Three-Self Patriotic Movement. Men such as Y.T. Wu (Wu Yaozong), K.H. Ting (Ding Guangxun), Timothy Tingfang Lew (Liu Tingfang), William Hung (Hung Ye), and Andrew Y.Y. Tsu (Zhu Yuyue) provide interesting case studies into how Union influenced Chinese Christianity. I apply insight from two sociologists, James Davison Hunter and Mark Granovetter, to new archival findings to contend that Union Seminary developed a dense network of influential Chinese and American Christians in the Sino-Foreign Protestant Establishment. The New York seminary acted as a social hub where Christian leaders established and strengthened their relational ties. Over time these ties formed a dense social network that influenced Sino-American relations and Chinese Protestantism. Union advanced the indigenization of Christianity in twentieth century China, helping “Christianity in China” become “Chinese Christianity. Union's role in China has been largely unnoticed until now.

Abstract:
The core concern of my thesis is with shifting the focus from the description on how innovation is done (predominantly STS accounts of innovation in-the-making) to what designers do with conceptions of innovation. The thesis is based on ethnographic fieldwork within a group of interaction designers of Milan. Despite the different conceptions and traditions of innovation that these designers bring in – the artistic and technological ones – I observed that a design-centered conception of innovation is reproduced, as well as the idea that plans and intentions precede things. However, another key idea of my fieldwork is the importance designers give to imagining things as they might be, rather than focusing on how things are. This is where different models of action, planned and open ones coexist in creative ways: it is these processes that the ethnography details.

Der Einsatz der Vitamin B 1 Vorstufe Benfotiamin hat sich im Tiermodell durch Verhinderung oder gar Aufhebung typischer diabetischer Folgeschäden wie Ne- phropathie, Retinopathie und Neuropathie ausgezeichnet. Diese Wirkung wird unter anderem der Aktivitätssteigerung des Enzyms Transketolase zugeschrie- ben, welches auch bei Dialysepatienten ohne diabetische Grunderkrankung sup- primiert ist. Das Ziel der vorliegenden Arbeit war, die Auswirkungen einer ora- len Benfotiaminsubstitution auf den Stoffwechsel von Langzeithämodialysepati- enten zu untersuchen. Die 15 rekrutierten Patienten mit und ohne Diabetes mel- litus erhielten über einen Zeitraum von 2 Monaten eine Dosis von 300 mg/d Benfotiamin, die in den folgenden 2 Monaten bis maximal 450 mg/d gesteigert wurde. Um einen Eindruck über den Verlauf der Entzündungssituation und des oxidativen Stresses zu gewinnen, wurden im Patientenvollblut AGEs und pro- inflammatorische Zytokine gemessen. Außerdem wurden peripheren Lympho- zyten mit Hilfe des alkaline Comet-Assay und des Mikrokerntestes auf DNA- Schädigungen analysiert. In beiden Patientengruppen lässt die Senkung der Mi- krokernraten den Schluss zu, dass Benfotiamin DNA-Schäden und somit eventu- ell das Krebsrisiko reduziert. Dieses vielversprechende Ergebnis korreliert jedoch nicht mit dem Resultat des Comet-Assay. Da hier der relative DNA-Schaden ten- dentiell ansteigt, sollte es Ziel weiterer Studien sein, diesen Sachverhalt an ei- nem größeren Patientenkollektiv mit Kontrollgruppen zu überprüfen. Eventuell ist letzteres Testsystem wegen seiner hohen Sensitivität in diesem Fall nicht op- timal geeignet. Außerdem sollte gezielt auf die beobachtete schnellere und stär- kere Mikrokernsenkung der diabetischen Patienten eingegangen werden, da die- se in der vorliegenden Studie zahlenmäßig unterrepräsentiert waren. Positiv zu bewerten ist der leichte CRP-Abfall sowie der Anstieg des Gesamtproteins und Albumin im Serum, was auf eine Reduktion der Mikroinflammation und oder eine verbesserte Ernährungssituation hinweist. Entgegen der Erwartung ließ sich in dieser Studie keine Reduktion der zirkulierenden AGEs und AOPPs im Serum erzielen. Um eine Reduktion des oxidativen Stresses besser beurteilen zu können, sollten in Folgestudien direkte und leicht veränderliche Marker wie der Glutathionspiegel verwendet werden. Zusammenfassend reduzierte Benfotiamin bei Hämodialysepatienten mit und ohne Diabetes mellitus DNA-Schäden in peripheren Lymphozyten bei unver- änderter Inflammationssituation und steigerte die Plasmaproteinkonzentration. Dies wurde eventuell durch Reduktion von oxidativem Stress und oder Beein- flussung seiner Ursachen wie Reduktion von Urämietoxinen erreicht.Weitere kli- nische Studien sind notwendig, um dieses vielversprechende Medikament in der täglichen Praxis einsetzen zu können. Besonders vorteilhaft ist seine gute Verträg- lichkeit auch in hoher Dosierung. Darüber hinaus soll das Präparat auch neuro- patische Schmerzen reduzieren, die sich bei Dialysepatienten häufig manifestie- ren, und wirkt somit multikausal
It has been shown in animal models, that Benfotiamine, a precursor of the vitamine B1, prevents typical complications of diabets, like nephropathy, retinopathy and neuropathy. This effect is attributed to the increased activity of the enzyme trankelotase. The latter is also suppressed in patients of the hemodialysis program who are not diabetic. The goal of this thesis was to show the effects of an oral administration of benfotiamine on longterm hemodialysis patients. Fifteen patients were treated with 300 mg per day of benfotiamine which was increased in the following two months to 450 mg per day. The patient group consisted of a sub-group of diabetic and non-diabetic individuals. Advanced glycation endproducts (AGEs) and proinflammatory cytocines were measured in patients full-blood to show the impact on the inflammation and the oxidative stress situation. The DNA-damage in peripheral lymphocytes was determined using the alkaline comet-assay and the micronucleus-assay. The rate of micronuclei was diminished in both patient groups which could be attributed to the reduction of DNA-damage by benfotiamine and so eventually to a reduced risk of cancer. However, this result does not agree with the comet-assay experiments. The relative DNA-damage increased in the course of the study and so seems to be unaffected by the benfotiamine therapy. This may be attributed to the high sensitivity of the comet-assay technique. Therefore, further investigations with a bigger patient group in a double-blind study are necessary. Additionally, there should be a greater focus on diabetic patients that showed a faster and increased reduction of micronuclei which were underrepresentated in this study. The slight reduction of CRP and the increased protein and serum-albumine concentration correlates to a better nutritional status. On the other hand, the increasing neopterine and interleukine 6 level do not agree to the changes in the inflammatory situation. Against all expectations there was no reduction of AGEs and AOPPs in patients serum. Following studies should focus on rapidly changing direct markers like the glutathione level. In summary, benfotiamine reduces DNA-damage in peripheral lymphocytes in hemodialysis patients with or without diabetes. The plasma protein concentration was increased but unexpectedly the inflammatory situation was stable. These effects may be due to a reduction of oxidative stress or its causes like diminished ureamic toxines. One of Benfotiamines advantages is its good tolerance, even in increased dosages. Furthermore it seems to diminish neuropathic pain which is frequent in hemodialysis patients. However, more clinical studies are neccessary for a use in daily practice

Abstract:
Research accentuates that end-users‘ noncompliance with information security policy (ISP) is a key concern for government just as it is for the private sector. Although awareness and training programmes are important factors impacting employees‘ intentions to comply with an organisation‘s ISP, it can be argued that there is insufficient empirical evidence to support this assertion. To address this gap, this study seeks to expand research on ISP compliance by focusing on attitudes as targets of change. A research model based on the Theory of Planned Behaviour was proposed to illustrate the influence of ISP awareness training on end-users‘ attitudes towards complying with their organisation‘s ISP. Relevant hypotheses were developed to test the research conceptualisation. A survey and an experiment was undertaken to collect the data from a sample of 173 end-users of a single government organisation in one province. The data was captured and analysed using a Statistical Package for Social Sciences (SPSS). Furthermore, Structural Equation Modelling (SEM) was used to test whether the overall model appears to be a good fit to support the hypotheses. The reliability, validity, and model fit were found to be statistically significant, and three out of five research hypotheses were supported. Overall this study contributes to the existing body of knowledge by providing an understanding of the methods that can be used to encourage end-users‘ ISP compliance behaviour through an attitudinal shift, thereby targeting end-users‘ attitude as a means to improve information security policy compliance. Implications of the findings are further discussed in the paper.
