Abstract:

Abstract:
AbstractThis article presents the case for a rethinking of the academy’s approach to sacred space through a demonstration of the way that a focus on unskilled actors reconfigures established approaches and interpretations. The article opens with an auto-ethnographic account of the powerful effect of Shimla’s Tara Devi temple on personal wellbeing and from this starting point spirals out to explore how Tara (and her sacred places) are connected to wellbeing both in the Himalayan region of Shimla and beyond. Through this process, arguments that I have previously made, concerning both the relation of sacred places to happiness (2010) and the way that sacred places operate in Himalayan North India (2012), are significantly complicated, leading to a reappraisal of the role that unskilled actors play in the constitution of sacred space. The article concludes by drawing these ethnographic reflections and theoretical considerations together to develop a key set of recommendations that call for policy-makers to engage sensitively with sacred places in the contemporary, post-secular city.

Abstract:

Abstract:
The texture, colour and sensory properties of non-fat yoghurts prepared with different tara gum concentrations (0.25, 0.50 and 1.0 gL-1) and different tara gum (TG) with buttermilk powder (BP) combinations (0.25+10, 0.50+10 and 1.0+10 gL-1) were investigated during storage. While addition of 0.25 and 0.50 gL-1 of tara gum caused an increase in firmness, consistency, cohesiveness, viscosity index of yoghurt samples, use of 1 gL-1 of tara gum and combinations of tara gum with buttermilk powder led to a decrease in these properties. During the storage period, increasing the concentration of tara gum resulted in significant decrease in L* and a* values, significant increase in b* value in yoghurt. Sensory properties of non-fat yoghurts supplemented with 0.25 and 0.50 gL-1 did not significantly differ from those of control yoghurts. In contrast, use of 1 gL-1 of tara gum and combinations of tara gum and buttermilk powder caused a significant decrease of sensory properties of non-fat yoghurt.

Purpose The purpose of this paper is to study the ability of commercial tara powder to convert rust into iron tannate and evaluate their use as raw material for the formulation of water based rust converter. Design/methodology/approach Water-borne acrylic primers were formulated with tara powder and aqueous tara extract and applied on steel rusted by three different methods. The conversion of rusted steel by tara tannins was studied by X-ray diffraction spectroscopy, infrared spectroscopy and scanning electron microscopy. The acrylic primers, containing hydrolysable tannins, were coated with alkyd finish and were evaluated in comparison to commercial systems in accelerated corrosion tests. The corrosion inhibition effects of tara powder on mild steel in 0.1M NaCl were studied by DC electrochemical techniques. Findings Tara tannin converts rust to ferric tannate and increases the magnetite content of rusted steel. The water-based acrylic primer formulated with aqueous extract of tara, alkyd-coated finish, showed performance equivalent to pure alkyd system. Research limitations/implications The chlorides content in the commercial tara powder can be screened the beneficial effect of hydrolysable tannins to convert rust. Furthermore, the water-based rust converter formulated with acrylic resin may be sensitive to salt contamination of rust. Originality/value Hydrolysable tannins from commercial tara powder have not been studied yet in its application to the development of rust converters. A water-based primer formulated with commercial tara powder developed for the effective treatment of rusted surfaces can be of interest as an environmentally friendly to current commercial approaches.

Tara gum is a natural hydrocolloid obtained by a mechanical process from the endosperm of Tara tree seeds. In this study, chemical composition and structure of tara polysaccharide gum were characterized by fourier transform infrared spectroscopy (FT-IR) and gas chromatograph-mass spectrometer (GC-MS). Its rheological properties were also discussed. The results showed that tara gum had typical characteristics of plant polysaccharides, which was mainly consisted of galactose and mannose (mass ratio of galactose/mannose was 4.2:1), as well as a small amount of arabinose, glucose and xylose. The viscosity of tara gum solution was increased with the increase of its concentration. The tara gum solution demonstrated a higher viscocity at the shear rate lower than 80 Hz, but a higher elasticity at the shear rate higher than 80 Hz.

Abstract:
Reorganization of the actin cytoskeleton is essential to numerous cellular processes including cell locomotion and cytokinesis. This actin remodeling is regulated in part by Rho family GTPases. Previous studies implicated Trio, a Dbl-homology guanine nucleotide exchange factor with two exchange factor domains, in regulating actin cytoskeleton reorganization, cell motility and cell growth via activation of Rho GTPases. Trio is essential for mouse embryonic development and Trio-deficiency is associated with abnormal skeletal muscle and neural tissue development. Furthermore, genetic analyses in Caenorhabditis elegans and Drosophila demonstrate a role for trio-like genes in cell migration and axon guidance. Herein we characterize a novel Trio-binding protein, Tara, that is comprised of an N-terminal pleckstrin homology domain and a C-terminal coiled-coil region. Trio and Tara associate as assessed by the yeast interaction-trap assays and mammalian co-immunoprecipitation studies. Ectopically expressed Tara localizes to F-actin in a periodic pattern that is highly similar to the pattern of myosin II. Furthermore, a direct interaction between Tara and F-actin is indicated by in vitro binding studies. Cells that transiently or stably overexpress Tara display an extensively flattened cell morphology with enhanced stress fibers and cortical F-actin. Tara expression does not alter the ability of the cell to attach or to initially spread, but rather increases cell spreading following these initial events. Tara stabilizes F-actin structures as indicated by the relative resistance of Tara-expressing cells to the F-actin destabilizer Latrunculin B. We propose that Tara regulates actin cytoskeletal organization by directly binding and stabilizing F-actin, and that the localized formation of Tara and Trio complexes functions to coordinate actin remodeling.

Abstract:
AbstractGenes of the Drosophila Polycomb and trithorax groups (PcG and trxG, respectively) influence gene expression by modulating chromatin structure. Segmental expression of homeotic loci (HOM) initiated in early embryogenesis is maintained by a balance of antagonistic PcG (repressor) and trxG (activator) activities. Here we identify a novel trxG family member, taranis (tara), on the basis of the following criteria: (i) tara loss-of-function mutations act as genetic antagonists of the PcG genes Polycomb and polyhomeotic and (ii) they enhance the phenotypic effects of mutations in the trxG genes trithorax (trx), brahma (brm), and osa. In addition, reduced tara activity can mimic homeotic loss-of-function phenotypes, as is often the case for trxG genes. tara encodes two closely related 96-kD protein isoforms (TARA-α/-β) derived from broadly expressed alternative promoters. Genetic and phenotypic rescue experiments indicate that the TARA-α/-β proteins are functionally redundant. The TARA proteins share evolutionarily conserved motifs with several recently characterized mammalian nuclear proteins, including the cyclin-dependent kinase regulator TRIP-Br1/p34SEI-1, the related protein TRIP-Br2/Y127, and RBT1, a partner of replication protein A. These data raise the possibility that TARA-α/-β play a role in integrating chromatin structure with cell cycle regulation.

The article examines the situation of Tara uyezd and service people who lived in the town of Tara in the first third of the 18th century. The research relies on such archival materials as responses of officials of Tara uyezd who completed to Prof. G.F. Miller’s questionnaire, books and tables of Siberian prikaz on the servitors in Siberia, and materials from the Senate fund. The article provides data on the town of Tara and Tara uyezd in the 1730s. Tara had a near-border position. A large Oirat state -Dzungar Khanate - was located to the south of Tara, and noble Oirat nomads collected tribute from the Turkic population of the uyezd. The reforms of Peter I made profound changes in the social world of Siberia. In the first third of the 18th century, the composition of the uyezd’s population was significantly altered. A new social group raznochintsy was formed of the relatives of servitors and clergy, and a large part of Tara’s service people were transferred to the garrison regiments of Siberia and the Orenburg governorate. The conflict between Tara’s horse Cossacks and captain Yakov Cheredov is indicative and important for understanding the service in favor of the state at that time. The Cheredovs were a deep-rooted clan of Russian service people who had lived in Tara since its founding. The Cheredovs held a number of important offices in Tara, and many of them became Boyar scions and nobles. After the 1722 Tara Rebellion, in which the Cheredovs played a significant role, they lost their privileged position and became raznochintsy . The ‘old’ service people who were nobles, Boyar scions and Cossacks remained the main military force in Siberian uyezds after the reforms. However, their dependence on the state increased. New garrison regiments in the region were formed in the 1730s, mostly of ‘old’ service people.

Mt. Tara is one of the most important centres of Balkans and European ecosystems and species diversity. They represent a unique example of well preserved forests in south eastern Europe with numerous endemic and relict species of flora and fauna. In this floristic diversity of Mt Tara of the greatest interest is the Serbian (Pancic's) spruce Picea omorika. Mt. Tara NP is characterized by specific geomorphologic, hydrologic, geologic, soil and climatic features. The geographical information system (GIS) that we have created has proved an excellent tool for the spatial planning strategy in assessment and conservation of all natural characteristics of Mt. Tara NP, and is helpful to Park management for sustainable use of landscape resources. GIS of Mt. Tara NP includes data on natural, artificial and management themes.

Abstract:

Abstract:
Tara Duncan, 1994, was established as a terrestrial genus of New Zealand talitrid amphipods, with the type species Orchestia sylvicola Dana, 1852. It was recently discovered to be a junior homonym of Tara Peckham & Peckham, 1886, a genus of salticid spiders (Araneae, Salticidae, Amycinae, Astiini). Tara Peckham & Peckham, 1886, is a valid name in current use for a small endemic genus of Australian jumping spiders recently revised by Zabka (1987). Tara Duncan, 1994, appears in the literature at least three times: in the original description of the genus (Duncan 1994), in identification keys to the terrestrial amphipods of New Zealand (Fenwick & Webber 2008), and in the recent inventory of New Zealand biodiversity (Webber et al. 2010). The new name, Dana, is proposed to replace Tara Duncan, 1994.

Abstract:
The spatiotemporal regulation of E-cadherin expression is important during body plan development and carcinogenesis. We found that Tara (Trio-associated repeat on actin) is enriched in cadherin-based adherens junctions (AJs), and its knockdown in MDCK cells (Tara-KD cells) significantly decreases the expression of E-cadherin. Tara-KD activates Rac1 through the Trio RhoGEF, which binds to E-cadherin and subsequently increases the phosphorylation of p38 and Tbx3, a transcriptional E-cadherin repressor. Accordingly, the decrease in E-cadherin expression is abrogated by ITX3 and SB203580 (specific inhibitors of Trio RhoGEF and p38MAPK, respectively), and by dephosphomimetic Tbx3. Despite the decreased E-cadherin expression, the Tara-KD cells do not undergo an epithelial–mesenchymal transition and remain as an epithelial cell sheet, presumably due to the concomitant up-regulation of cadherin-6. Tara-KD reduces the actin-belt density in the circumferential ring, and the cells form flattened cysts, suggesting that Tara functions to modulate epithelial cell sheet formation and integrity by up-regulating E-cadherin transcription.

Abstract:
Oil extracted from the seeds ofCaesalpinia spinosa(common name: tara) can be used in a number of applications. In the present study, tara seed oil was obtained by ultrasonic extraction. The effects of different solvents, particle sizes in the ground seed samples, extraction times, ultrasonication powers, extraction temperatures, and liquid–solid ratios on the yield of tara seed oil were investigated. The yield from the ultrasonic extraction was compared with that from traditional Soxhlet extraction. The results showed that ultrasonic extraction could be completed in a shorter time with reduced solvent consumption. The yield of tara seed oil increased with increasing ultrasonication power and extraction temperature. Gas chromatography was used to analyze the fatty acid compositions of the oils extracted by the two methods. The fatty acid compositions of the oils from both extraction methods were similar, which indicates that ultrasonic extraction is a viable alternative means of extraction. It is a rapid, efficient, and simple method for production of lipids from tara seeds.

Abstract:
Photosynthesis evolved in the ocean more than 2 billion years ago and is now performed by a wide range of evolutionarily distinct organisms, including both prokaryotes and eukaryotes. Our appreciation of their abundance, distributions, and contributions to primary production in the ocean has been increasing since they were first discovered in the seventeenth century and has now been enhanced by data emerging from the Tara Oceans project, which performed a comprehensive worldwide sampling of plankton in the upper layers of the ocean between 2009 and 2013. Largely using recent data from Tara Oceans, here we review the geographic distributions of phytoplankton in the global ocean and their diversity, abundance, and standing stock biomass. We also discuss how omics-based information can be incorporated into studies of photosynthesis in the ocean and show the likely importance of mixotrophs and photosymbionts.

Abstract:
Abstract This paper examines Tara Rai’s Chhapamar Yuwatiko Diary [‘A Diary of a Young Guerrilla Girl’] (2010) – a memoir which describes a 15-year-old girl’s experience of first armed encounter, subsequent detainment, and release from the custody towards the end of the Maoist war in Nepal. We analyze the author’s narrative of adversity and distress, using thematic analysis. Three themes, namely, (1) perception of impending death, (2) severe stress reactions, and (3) gradual recovery are found in temporal succession. In a subsequent analysis, we examine using content analysis the personal, group, and socio-political factors linked to these dominant themes to understand the dynamics associated with Rai’s understanding of personal experience, and adjustment to violence. Discussion of the findings orient the readers of this narrative not only to how Rai’s perception of her trauma experience changes but also to how this account can inform the way people negotiate the trauma of war.

Abstract:
The progress of rigor-mortis and the post-mortem changes in three species of freshwater eelsduring storage at room temperature (30° to 35°C) and ice (0°C) were studied. Rigor-mortis in fish sample atroom temperature, started 1hr after spiking and it reached to a maximum of 76%, 57.14% and 100% within 4hrs after death in Mastacembelus armatus (Baim), Macrognathus aculeatus (Tara baim) and inMastacembelus pancalus (Guchi), respectively. In ice stored eels rigor also started within one hour of spikingand gradually reached to a maximum of 88.89% within 3 hrs in Baim and 85.71%, 100% in Tara baim and inGuchi respectively within 2 hrs. Rigor continued for 17, 14 and 18 hrs in Baim, Tara baim and Guchi, respectively and then started to relax. The pH of the muscle was 6.84, 6.80 and 6.92 immediately after catchin Baim, Tara baim and in Guchi, respectively which decreased gradually with the lapse of storage periodregardless of the storage temperature. The organoleptic quality of eels during ice storage was assessed on thebasis of sensory parameters. Baim was found in acceptable conditions for 24 days whereas Tara baim andGuchi were in acceptable condition for 28 days in ice storage. Percent moisture content increased but proteinand lipid contents decreased in almost all the fish samples with the lapse of storage period but a little changewas observed in ash content. The TVB-N, peroxide and NPN value remained within the recommended valueup to 24 days in Baim and 28 days in Tara baim and Guchi. The initial bacterial loads were 6.5×103,6.2×103 And 6.3×103 CFU/g in Baim, Tara baim and in Guchi, respectively. At the end of the 24 days ofice storage, bacterial load increased in all the fish samples.

Planta Medica International Open 7, no. 01 (April 2020): e34-e44. http://dx.doi.org/10.1055/a-1141-0151.

Abstract:
AbstractThe skin color is imparted by the pigment melanin produced in the melanosomes of melanocytes, through the catalytic action of melanogenesis enzymes tyrosinase, tyrosinase-related protein 1, and dopachrome tautomerase. Disruptions in the melanogenesis process may result to hypopigmentation, as observed in cutaneous postinflammatory conditions. Here, the bioactivity of tara tannin, specifically on melanogenesis, was evaluated in vitro using human epidermal melanocytes (HEM) and B16F10 murine melanoma cells in order to determine the possibility that it may be used as a treatment against hypopigmentation. The melanin content of tara tannin-treated B16F10 cells and the expression level of melanogenesis enzymes and melanosome transport proteins were determined. To elucidate the underlying mechanism of tara tannin’s effect on melanogenesis, DNA microarray analysis was performed. Tara tannin significantly increased melanogenesis in both murine and human pigment cell models by upregulating melanogenesis-associated enzymes’ (tyrosinase, tyrosinase-related protein 1, and dopachrome tautomerase) protein and mRNA expression levels, as well as the melanosome transport proteins (myosin Va and RAB27A) expression, both attributed to increased microphthalmia-associated transcription factor (MITF) expression. Global gene expression analysis results revealed the modulation of genes (p≤0.05; fold-change ≥2.0 and ≤−2.0) that are under the transcriptional regulation of MITF and genes relevant for MAPK signaling, metabolic pathways, and cell cycle. Tara tannin has a significant effective melanogenesis-promoting effect, making it a potential therapeutic agent against hypopigmentation disorders. This is the first report on the melanogenesis regulatory effect of tara tannin in vitro.
