AbstractStable isotope labeling by amino acids in cell culture (SILAC) combined with high-resolution mass spectrometry is a quantitative strategy for the comparative analysis of (sub)proteomes. It is based on the metabolicincorporation of stable isotope-coded amino acids during growth of cells or organisms. Here, complete labeling of proteins with the amino acid(s) selected for incorporation needs to be guaranteed to enable accurate quantification on a proteomic scale. Wild-type strains of baker’s yeast (Saccharomyces cerevisiae), which is a widely accepted and well-studied eukaryotic model organism, are generally able to synthesize all amino acids on their own (i.e., prototrophic). To render them amenable to SILAC, auxotrophies are introduced by genetic manipulations. We addressed this limitation by developing a generic strategy for complete “native” labeling of prototrophic S. cerevisiae with isotope-coded arginine and lysine, referred to as “2nSILAC”. It allows for directly using and screening several genome-wide yeast mutant collections that are easily accessible to the scientific community for functional proteomic studies but are based on prototrophic variants of S. cerevisiae.

Abstract Research into plant genetics often requires the suppression or complete knockout of gene expression to scientifically validate gene function. In addition, the phenotypes obtained from gene suppression can occasionally have commercial value for plant breeders. Until recently, the methodological choices to achieve these goals fell into two broad types: either some form of RNA-based gene silencing; or the screening of large numbers of natural or induced random genomic mutations. The more recent invention of gene editing as a tool for targeted mutation potentially gives researchers and plant breeders another route to block gene function. RNAi is widely used in animal and plant research and functions to silence gene expression by degrading the target gene transcript. Although RNAi offers unique advantages over genomic mutations, it often leads to the formation of a genetically modified organism (GMO), which for commercial activities has major regulatory and acceptance issues in some regions of the world. Traditional methods of generating genomic mutations are more laborious and uncertain to achieve the desired goals but possess a distinct advantage of not being governed by GMO regulations. Gene editing (GE) technologies have some of the advantages of both RNAi and classical mutation breeding in that they can be designed to give simple knockouts or to modulate gene expression more subtly. GE also has a more complex regulatory position, with some countries treating it as another conventional breeding method whilst the EU defines GE as a technique of genetic modification and applies the normal GMO authorization procedures. This chapter explores the pros and cons of RNAi alongside other methods of modulating gene function.

Organophosphorus compounds (OP) are found in environmental objects and food products. Due to their high toxicity and inhibition of cholinesterase activity, it is necessary to control residual amounts of OP. The most common methods for determining OP are gas and liquid chromatography with various detection methods. However, chromatographic analysis is lengthy, requires complex sample preparation and expensive equipment, which limits its use for screening a large number of samples and continuous monitoring of the content of OP. To detect the OP, it is necessary to use High Throughput Screening methods, using simple, fast and inexpensive analysis methods. Currently, immunochemical methods are increasingly used to determine OP. These methods are based on the recognition of the analyte (antigen) by specific receptors (antibodies) with the formation of the antigen-antibody complex and the measurement of the analytical signal generated by the immunochemical test system in response to complex formation, which leads to high sensitivity and specificity of the analysis.

ASTM Standard C1161 is often used to determine fracture performance of piezoelectric materials. The basic geometry of C1161 is a four-point beam subjected to mechanical loading. When single crystal PMN-PT was recently subjected to this test unexpected results were observed. Fracture did not occur at the defects incorporated into the specimens nor did failure occur at locations of where maximum stress or strain was predicted by mechanical analyses.
