Abstract:
ABSTRACT The mesophile Acinetobacter sp. strain BD413 and the extreme thermophile Thermus thermophilus HB27 display high frequencies of natural transformation. In this study we identified and characterized a novel competence gene in Acinetobacter sp. strain BD413, comA, whose product displays significant similarities to the competence proteins ComA and ComEC inNeisseria and Bacillus species. Transcription of comA correlated with growth phase-dependent transcriptional regulation of the recently identified pilin-like factors of the transformation machinery. This finding strongly suggests that comA is part of a competence regulon. Examination of the genome sequence of T. thermophilus HB27 led to detection of a comA/comEC-like open reading frame (ORF) which is flanked by an ORF whose product shows significant similarities to the Bacillus subtilis competence protein ComEA. To examine whether these two ORFs, designated comEC andcomEA, are implicated in natural transformation of T. thermophilus HB27, both were disrupted by using a thermostable kanamycin resistance marker. Natural transformation in comEC mutants was reduced 1,000-fold, whereas in comEA mutants the natural transformation phenotype was completely eliminated. These results strongly suggest that both genes, comEC and comEA, are required for natural transformation in T. thermophilus HB27. Several transmembrane α-helices are predicted based on the amino acid sequences of ComA in Acinetobacter sp. strain BD413 and ComEC in T. thermophilus HB27, which suggests that ComA and ComEC are located in the inner membrane and function in DNA transport through the cytoplasmic membrane.

ABSTRACT The Spx protein of Bacillus subtilis exerts both positive and negative transcriptional control in response to oxidative stress by interacting with the C-terminal domain of the RNA polymerase (RNAP) alpha subunit (αCTD). Thus, transcription of the srf operon at the onset of competence development, which requires the ComA response regulator of the ComPA signal transduction system, is repressed by Spx-αCTD interaction. Previous genetic and structural analyses have determined that an Spx-binding surface resides in and around the α1 region of αCTD. Alanine-scanning mutagenesis of B. subtilis αCTD uncovered residue positions required for Spx function and ComA-dependent srf transcriptional activation. Analysis of srf-lacZ fusion expression, DNase I footprinting, and solid-phase promoter retention experiments indicate that Spx interferes with ComA-αCTD interaction and that residues Y263, C265, and K267 of the α1 region lie within overlapping ComA- and Spx-binding sites for αCTD interaction. Evidence is also presented that oxidized Spx, while enhancing interference of activator-RNAP interaction, is not essential for negative control.

Aims: To assess swelling after surgical extraction of impacted mandibular third molar using two different techniques i.e. comma incision and standard Wards incision. Study Design: Cross sectional study. Setting: Dental OPD of Isra Dental College, Isra University. Period: February to August 2017. Materials and Methods: A sample of 70 patients with impacted third molar was selected by non-probability purposive sampling. Patients were distributed in either conventional technique or by coma shaped incision groups. All procedure was carried out at the Department of Oral surgery, Isra Dental College Hospital. Post-operative measurement of swelling was measured on 1st, 3rd and 7th day respectively. Results: Mean ± SD was noted as 28.70 ± 3.89 years respectively. Age distribution was from 20 – 35 years. Gender distribution in group A was 11 (31.4%) males and 24 (68.6%) females while in group B was 17 (48.5%) males and 18 (51.5%) females respectively with X2 = 2.14 and p= 0.22. Swelling was measured after surgical extraction at day 1, day 3 and day 7. Conclusion: The results of this study showed that the swelling was to some extent reduced in the Coma incision in comparison to the standard Ward`s incision. Further research with newer flap designs like the comma design, which might lessen the post-operative problems, ought to be measured in the extraction of impacted third molar surgery.

Conclusion: In conclusion, the Coma incision was preferable over theconventional method- the standard Ward`s incision because of lesser degree of trismus seen.Further research with newer flap designs and the comma design should be preferred by theclinicians for the extraction of impacted third molar surgery.

Many Gram-positive and Gram-negative bacteria possess natural competence mechanisms for DNA capture and internalization. In Bacillus subtilis, natural competence is absolutely dependent upon the presence of a seven-gene operon known as the comG operon (comGA–G). In species of Streptococcus, this function has been described for a four-gene operon (comYA–D in Streptococcus gordonii and cglA–D in Streptococcus pneumoniae). In this study, a nine-orf operon (named comYA–I) required for natural competence in Streptococcus mutans was identified and characterized. Orf analysis of this operon indicates that the first four Orfs (ComYA–D) share strong homology with ComYA–D of S. gordonii and CglA–D of S. pneumoniae, the fifth to seventh Orfs (ComYE–G) match conserved hypothetical proteins from various species of Streptococcus with ComYF possessing a predicted ComGF domain, the eighth Orf (ComYH) shows a strong homology to numerous DNA methyltransferases from restriction/modification systems, and the ninth Orf (ComYI) is homologous to acetate kinase (AckA). RT-PCR analysis of the orf junctions confirmed that all nine orfs were present in a single transcript, while real-time RT-PCR analysis demonstrated that these orfs were expressed at a level very similar to that of the first orf in the operon. Mutations were constructed in all nine putative orfs. The first seven genes (comYA–G) were found to be essential for natural competence, while comYH and comYI had reduced and normal natural competence ability, respectively. Analyses of S. mutans comY–luciferase reporter fusions indicated that comY expression is growth-phase dependent, with maximal expression at an OD600 of about 0·2, while mutations in ciaH, comC and luxS reduced the level of comY expression. In addition, comY operon expression appears to be correlated with natural competence ability.

Comza es un proyecto que planea llevarse a cabo a partir de 2016, teniendo el año 2015 como el período para su preparación. Este proyecto consiste, a grandes rasgos, en la implementación de una plataforma en línea que agilice el proceso de cotización al que se enfrentan hoy en día los compradores en general. Esta agilización responde a problemas inherentes dentro de este proceso. Como se verá más adelante, estos problemas se relacionan principalmente con los costos de búsqueda y la ausencia de información perfecta, para que un determinado consumidor pueda optar por la mejor alternativa en forma transpareante. En este mundo, Comza viene a plantearse como una solución factible para los problemas y costos de búsqueda que sufren los agentes económicos. Si bien estos costos han disminuido con la implementación de internet, todavía son importantes y traen consigo significantes efectos para los compradores y empresas, como se demostrará en esta tesis.

Abstract:
The aim of this thesis is to contribute to the study of African-Caribbean cultural and religious practices. Research in this area has tended to focus on Cuban Santeria, Haitian Vodou, Trinidadian Shango/Orisha/Spiritual Baptist and Jamaican Myal/Kumina practices. There has been little research on Guyanese religious and cultural practices. Kean Gibson's Comfa Religion and Creole Language in a Caribbean Community (2001) appears to be the most complete study of Guyanese Comfa. Comfa is the generic term used for the manifestation of spirits. Anyone who becomes spiritually possessed on hearing the beating of drums is said to 'ketch comfa'. Comfa practitioners recognise a pantheon of seven ethnic spirits: African, Amerindian, Chinese, Dutch, English, (East) Indian and Spanish. These groups have been historically associated with Guyana. Spirit possessions are stereotypically defined to reflect the ethnicity of each spirit. Inspired by Ema Brodber's method of combining sociological research with creative writing, this thesis is organised in three parts to reflect an interdisplinary methodology. Firstly, I combine Gibson's sociological account of Cornfa with the works of writers and postcolonial critics, namely Wilson Harris, Edward Brathwaite, Stuart Hall and Antonio Benitez-Rojo to consider Cornfa's significance to Guyanese cultural identity. I use Harris's ideas in his essay History Fable and Myth to argue that, as myth and art, Comfa has the potential to transform the recurrent image of despair, racial division and political violence that impact Guyana's cultural psyche. Secondly, I explore four texts by Caribbean writers to highlight the social, cultural and historical significance of spirit possession/spiritual practices and the way Caribbean spiritual traditions can be used as literary aesthetic. The third part of the thesis engages the foregoing analyses and theoretical considerations to write a novella with Cornfa as the central theme. The novella aims to demonstrate Comfa's potentiality as a literary and cultural resource.

COMMA-LIM (Cologne Model of the Middle Atmosphere - Leipzig Institute for Meteorology) ist ein 3D-mechanistisches Gitterpuktsmodell, welches sich von ca. 0 bis 135 km in logarhitmischen Druckkordinaten z = -H ln(p=p0) erstreckt, wobei H=7 km und p0 den Referenzdruck am unteren Rand bezeichnet. Die vertikale Auflösung von COMMA-LIM wurde auf 48 Schichten erhöht. Zugleich wurde die Beschreibung des Strahlungsprozesses verbessert, zusammen mit den Beiträgen zur Temperaturbilanz durch atmosphärische Wellen und Turbulenz. Weitere Veränderungen betreffen die numerische Realisation der horizontalen Diffusion und des Filterproblems. Die Beschreibung ist unterteilt in den dynamischen Teil und die Strahlungsbeträge. Die jahreszeitlichen Klimatologien werden vorgestellt und diskutiert
COMMA-LIM (Cologne Model of the Middle Atmosphere - Leipzig Institute for Meteorology) is a 3D-mechanistic gridpoint model extending up from 0 to 135 km with a logharithmic vertical coordinate z = -H ln(p=p0), where H=7 km and p0 is the reference pressure at lower boundary. The resolution of the 24 layer version has been increased to 48 layers and several improvements are made in the parameterisation of radiative processes, heating/cooling due to atmospheric waves and turbulence, as well as in the numerical realization of the horizontal diffusion and filtering. This description is divided into the section describing the changes in the dynamical part and the modifications in radiation routines. After all, the seasonal climatologies will be shown and discussed to demonstrate what the COMMA-LIM is capable of reproducing

Abstract:
Sclerotinia sclerotiorum est un champignon pathogene qui infecte le colza (brassica napus l. ) en colonisant d'abord le petale. L'ablation genetique du petale par expression d'une molecule cytotoxique pourrait permettre de lutter contre ce champignon. Pour que la structure de la plante ne soit pas alteree, le gene codant pour cette molecule doit etre place sous le controle d'un promoteur specifique du petale. Aucun gene decrit dans la litterature n'est exprime specifiquement dans le petale jeune entier. Par soustraction et hybridation differentielle, nous avons mis en evidence une famille de deux genes, bnp4 et bnp8. Ces genes sont exprimes fortement dans le petale mais egalement plus faiblement dans les etamines. Une experience d'hybridation in situ a montre que ces genes etaient exprimes dans les couches epidermiques du petale. Une analyse de leur region promotrice respective, par deletions et fusions a la sequence codante du gene rapporteur gus, a permis de montrer que le fragment ( 299, + 24) du gene bnp4 confere une specificite stricte dans le petale chez 40% des transformants d'arabidopsis thaliana obtenus. L'expression gus est localisee principalement dans l'epiderme. Ces genes codent une proteine membranaire putative de la membrane plasmique ou du reticulum endoplasmique. Une analyse phylogenique basee sur la sequence d'un fragment de la sequence codante des genes de colza a montre une forte conservation de la sequence parmi les differentes brassicacees etudiees. Le profil d'expression de ces genes est egalement bien conserve parmi ces especes. La molecule cytotoxique employee est la barnase. Des strategies a double composantes par complementation biochimique de deux peptides inactifs sont actuellement a l'etude dans la levure avant de le mettre en application chez les plantes.

Des plantes de colza transgéniques ont été obtenues en utilisant deux stratégies de transformation différentes. La régénération et la caractérisation de plantes issues de racines transformées par Agrobacterium rhizogenes est décrite dans la première partie. Une deuxième partie est consacrée à l'étude de la transformation directe de protoplastes par électroporation. La mise au point de cette technique a été réalisée sur des protoplastes de tabac par utilisation d'un dosage d'expression transitoire. L'application de l'électroporation aux protoplastes de colza a permis d'obtenir des plantes transgéniques exprimant un gène de résistance à la kanamycine. Différentes applications agronomiques possibles de ces méthodes de transformation sont envisagées pour le colza
Transgenic rapeseed plants have been obtained using two transformation procedure. The regeneration and caracterization of rapeseed plants derived from hairy roots induced by Agrobacterium rhizogenes are describe in the first part. The second part is devoted to the study of direct gene transfer by electroporation. This technique was optimized on tobacco protoplasts using a transient expression assay. The application of this technique to rapessed protoplasts made it possible to obtain transgenic rapeseed plants resistant to the antibiotic kanamycin. Possible agronomy applications of this strategy of transformation to rapeseed improvement are considered

La bnd22 est une proteine induite au cours d'une contrainte hydrique progressive chez le colza. Cette proteine s'accumule fortement dans les jeunes feuilles de la plante au stade rosette. La synthese et l'accumulation de la bnd22 dans ces feuilles sont correlees avec la tolerance de celles-ci a la contrainte hydrique, caracterisee par le maintien du contenu relatif en eau, le maintien de la teneur en proteines solubles et l'inhibition de l'activite proteolytique foliaire endogene. Ces resultats constituent un argument en faveur d'une activite potentielle d'inhibiteur de proteases chez la bnd22, suggeree par des travaux anterieurs sur la base d'homologies de sequence avec les inhibiteurs de la famille de kunitz. La bnd22 a ete purifiee par chromatographie d'echange d'anions et par chromatographie en phase inverse. Elle a ete caracterisee sur le plan structural et une activite inhibitrice de proteases a ete recherchee. Elle presente une masse, mesuree par spectrometrie de masse, d'environ 19 kda. Sa structure secondaire, determinee par dichroisme circulaire et predite d'apres sa sequence en acides amines, est composee aux deux tiers de structure non periodique ainsi que d'un tiers de feuillet beta, analogue a celle de certains inhibiteurs de la famille de kunitz. Les mesures d'activite proteolytiques en presence de bnd22, effectuees sur les proteases endogenes foliaires ainsi que sur quatre proteases exogenes d'origines differentes, n'ont pas permis de mettre en evidence une inhibition significative

Abstract:
Lake Arakhley is located within the Lake Baikal basin in Eastern Siberia, Russia. The area is characterized by continental subarctic climate with considerate diurnal temperature range, long cold dry winters and short hot summers with more precipitation occurring during the latter half of the summer. Climatic shifts in high water years and low water years result in morphometric changes in the lake and in the chemical and physical parameters of the ecosystem. During low water years, concentrations of ammonium nitrogen and nitrite nitrogen are decreased, whereas nitrate concentration increases. High water years feature average concentrations of ammonium ions 1.5–2 times higher than the values of recent dry years. Redundancy analysis (RDA) of abiotic factors and biotic community indicated that the community structure shows the greatest correlation with physical and chemical parameters of water and biogenic elements (nitrites, ammonium, phosphates) along the first axis, and with the lake depth and transparency along the second axis. Changes in abiotic factors induce functioning and formation of characteristic communities of the primary producers in the trophic structure of the ecosystem. During low water years, with increased level of autochthonous organic matter, Lindavia comta dominance is observed, while during high water years, with increased allochthonous organic matter Asterionella formosa appeared as dominant. Currently, during low water years, the hydrophytes community is monodominant and composed of Ceratophyllum demersum. Meanwhile, such species indicating eutrophic conditions as Myriophyllum sibiricum, Potamogeton pectinatus are found in the lake vegetation.
