Abstract:
A library of pea (Pisum sativum L) genomic DNA in bacteriophage EMBL3 was screened for seed storage protein genes of the legumin and vicilin families. Three genomic clones were isolated. One of the clones was found to contain a gene in the Leg A sub-family which was designated Leg E. The nucleotide and predicted amino acid sequence of Leg E were compared to those of Leg A. The coding sequences of both genes are strongly homologous with only 9 bases difference out of 1560 bases. A second genomic clone contained two genes from the Leg J subfamily. Leg J and Leg K. The clone was shown to overlap with a genomic clone isolated previously, JC5 (Gatehouse et al. 1988). Strong homology was found between the Leg K and Leg J sequences. The Leg K gene is predicted to be pseudogene, due to the conversion of the ATG methionine start codon to a GTG valine codon and the presence of a stop codon in the 5' end of the coding sequence in the reading frame predicted by the first subsequent start codon. A genomic library was constructed for Arabidopsis thaliana, using EMBL3 as a vector to sub-clone Sau3AI partially digested Arabidopsis genomic DNA. About 8 x 10(^4) random clones were obtained when the ligated vector DNA and insert were in vitro packaged. The Arabidopsis gene library was screened for clones containing sequences encoding the cell wall protein extensin, using a rape (Brassica napus L extensin cDNA as a probe. Six clones were isolated, two of which were restriction mapped. One of them was partially sequenced. This clone did not contain an extensin gene homologous to the probe sequence, and only contained a short extensin-like sequence which was responsible for the observed hybridisation. The putative gene may represent another type of protein, since it was expressed in the root of Arabidopsis and Brassica napus L, as shown by "Northern" blots which were probed with labelled DNA from the clone.

Functional screening of environmental genomic libraries permits the identification of clones expressing activities of interest without requiring prior knowledge of the genes responsible. In this study, protocols were optimized for the construction of mid-size DNA insert, inducible expression environmental genomic plasmid libraries for this purpose. A library with a mean insert size of 5.2 kilobases was constructed with environmental DNA isolated from surface ocean water collected at Hawaii Ocean Time-series station ALOHA in plasmid cloning vector pMCL200 under the inducible control of the PLAC promoter. To begin to evaluate the utility of such libraries for gene expression-based screens, this library was screened phenotypically for clones expressing genes that confer fluorescence or distinctive coloration on colonies of host Escherichia coli cells, and results were compared to those for a fosmid library constructed from the same marine microbial DNA sample. Ecologically relevant sequences were identified in both libraries, and each was observed to offer both advantages and disadvantages. Results of this study suggest that mid-size insert plasmid libraries under the control of inducible promoters can provide a useful and complementary approach for both functional screening and shotgun sequencing of environmental genomic libraries.

Abstract:
The aim of the project was to determine the sequence of the gene encoding glucose-6-phosphate dehydrogenase (G6PDH) in the industrially used, but genetically little explored yeast Candida utilis. In the first strategy, a C. utilis DNA library was constructed in the expression vector phage lambda gt11, and was screened immunologically with antiserum raised against C. utilis G6PDH. The second strategy involved construction of libraries in the plasmid vector pTZ18R and screening with a mixed oligonucleotide probe. Two methods were developed that allow DNA, of appropriate quality for the construction of DNA libraries, to be prepared from C. utilis cells. Both involve centrifugation of C. utilis lysates through caesium trifluoroacetate gradients. A rabbit antiserum was raised against a commercial preparation of C. utilis G6PDH. It reacted with Western blotted C. utilis G6PDH, and non-specific binding of the antiserum to Western blotted E. coli protein was reduced by treatment with an E. coli extract. Antiserum pretreated in this way was used for immunological screening. Sequence information was determined for the inserts of 7 phage identified during immunological screening. None of the insert sequences was considered to contain part of the C. utilis G6PDH coding sequence. One putative positive colony was identified during the screening of the pTZ18R libraries. Sequence determination showed the insert to contain a region complementary to 16 bases in one of the 17-base long probe oligonucleotides. This insert was considered not to contain part of the C. utilis G6PDH coding sequence. Time did not permit further work which, using the methods developed, should now permit determination of the C. utilis G6PDH gene sequence.

The vine mealybug, Planococcus ficus (Signoret), is a severe pest of grapevine in many grape and wine producing countries around the world. It is renowned not only for the considerable damage it infers to grapevine of its own accord, but in particular for its role in transmitting deleterious viral diseases such as grapevine leafroll disease, Kober stem grooving, Shiraz disease and corky bark. Incidentally, it is an exceptionally tenacious antagonist of grapevine, being resistant to both chemical and biological control mechanisms. As a result, finding an effective strategy for P. ficus control has become a main priority of viticultural industries worldwide. Possible implementation of biotechnological approaches to pest management has resulted in a need for P. ficus genetic data - of which there are currently very little available. The transcribed genes of an organism can be captured in a cDNA library, and the sequences of the various transcripts can then be characterized. In this study altogether five cDNA libraries were constructed from the transcribed sequences of Planococcus ficus (Signoret). Instrumental to their construction was the identification of an RNA extraction protocol that provided large quantities of high quality RNA from mealybugs. The five cDNA libraries were the result of a set of modifications to the Creator™ SMART™ cDNA Library Construction Kit (used for Primary Library construction), and differed mainly with regards to range of insert sizes they contain. Whereas an abundance of short fragments were found in the Primary Library (42% of screened inserts 60.5 kb, and 20% >1 kb), the Fractionated Libraries contained inserts of specific size ranges that were more-or-less equally represented. The broadest size range was found in Fractionated Library 4, for which a uniform distribution over the range 0.25 kb - 4 kb was observed. Average insert sizes of Fractionated Libraries 1 to 4 were estimated at 0.25 kb, 0.5 kb, 1 kb and 2 kb respectively. These results demonstrated the importance of using a protocol designed to circumvent the bias towards incorporation of shorter transcripts in cDNA libraries. Although the libraries were not exhaustively analyzed, the outcome of a pilot investigation indicated that 41% of the submitted sequences had matches in the non-redundant database of the National Center for Biotechnology Information (NCBI, E-value 6 10-5), and that approximately 82% of these were of insect origin. Moreover, two potential targets for an RNAi-mediated approach to P. ficus pest control were identified. With one exception, these sequences seemed to be unique to arthropods. Future research needs to investigate the efficiency by which these sequences are able to constrain P. ficus proliferation, and their suitability for grapevine transformation.

Technological advancements in the second half of the century have significantly affected academic library institution. Libraries are undergoing a transition period and constantly updating their system to adapt to those advancements. However, library building is slow to adapt to the changes. This study examines those changes in academic libraries and buildings based on two main variables. namely human beings. and the medium. through which the information is transmitted The invention of first. electronic, and then digital systems significantly changed these two variables. The resulting library building is shaped by the new human Behavior and the form of information-mediums in a technology-oriented environment. The result of this study is 'five governing principles,' which explain the position of those two variables in a complex web of relations created by technological advancements. The same principles also explain the role of different variables in the future of the library building typology.

Buildings can be built that use no energy from external power grids. They can be essentially carbon neutral in their environmental load, and they can be built and operated at fair market values. Although this may sound like a dream of the future, the World Business Council for Sustainable Development (WBCSD) is working toward making that dream a reality today. The Council announced on March 26, 2006 (Environment News Service) that it is forming an alliance of global companies to ensure that by 2050 all new buildings meet these standards.1The profession of architecture, at the beginning of the 21st century, finds itself in a position where architectural philosophies and methodologies must respond to the world changing events that are impacting our planet. This creative project outlines some of the methodologies used to create a carbon-neutral public library. Within a public library, architects respond to two of the primary concerns of this young century; the global need for a clean, livable planet and the proliferation of digital media and methods that are driving globalization as we are beginning to know it.The proposed new Union Township Branch of the Anderson Public Library seemed a logical subject for this Creative Project because much of the past work of our firm and much of my personal experience involved the design of public libraries. The carbon neutral concepts evolved gradually, after much study, and the goal of this creative project is that this particular library and many other sustainable libraries in the future will benefit from the concepts and methodologies outlined here, as those new buildings approach carbon neutrality.

En aquest context, la síntesis de nous lligands quirals és essencial per descobrir bons sistemes catalítics en catàlisi asimètrica. Aquesta tesi s'ha centrat en el desenvolupament de tres noves lligandoteques quirals heterodadores i la seva aplicació a la substitució al·lílic catalizada per paladi. Es tracta de lligandoteques estables a l'aire, de fàcil manipulació i que s'han sintetizats en poques etapes a partir de productes d'elevada disponibilitat i baix preu. Combinant estudis teòrics i espectroscòpia de RMN, hem pogut afinar racionalment els lligands, millorar l'enantioselectivitat i identificar les espècies responsables dels resultats catalítics. A més a més, els productes de substitució al·lílic resultants es van utilitzar amb èxit per a la síntesi de molècules més complexes.

Se trata de ligandotecas estables al aire, de fácil manipulación y que se han sintetizado en pocas etapas a partir de productos de elevada disponibilidad y bajo precio. Combinando estudios teóricos y espectroscopia de RMN, hemos podido afinar racionalmente a los ligandos, mejorar la enantioselectividad e identificar las especies responsables de los resultados catalíticos. Además, los productos de sustitución alílica resultantes se utilizaron exitosamente para la síntesis de moléculas más complejas.
The constant demand for enantiomerically pure compounds (drugs, agrochemicals, additives ...) has driven the development of asymmetric catalysis using chiral organometallic compounds as catalysts. In this context, the synthesis of new chiral ligands is essential to discover good catalytic systems in asymmetric catalysis. This thesis has focused on the development of three new heterodonating chiral ligand libraries and their application to Pd-catalyzed allylic substitutions. These are ligand libraries stable in the air, easy to handle and have been synthesized in a few steps from readily available products and at low price. Combining theoretical studies and NMR spectroscopy, we were able to rationally fine-tune the ligands, improve enantioselectivity, and identify the species responsible for catalytic performance. In addition, the resulting allylic substitution products were successfully used for the synthesis of more complex molecules.

The Grande Bibliothèque du Québec (GBQ), the merger of the National Library of Québec with the Central Municipal Library of Montréal in a ninety-million dollar construction project which opened in the spring of 2005, serves as the point of departure and as a model and metaphor for reflection on the significance of libraries in the cultural life of a society and in the construction of social identity. The province of Québec is unique, not only because it is the sole province in Canada where the citizens are a French language majority, but also because it is the only province to have established its own national library. The Bibliothèque nationale du Québec and other significant libraries around the world collect and preserve memory in ways that create a context for cultural recall. Government and religious leaders have, for a long time, recognized the potential that libraries have in influencing popular thought and ideas of citizens. In societies where democratization of information is fostered, libraries are promoted as a source of pride and cultural achievement in buildings that are constructed as architectural monuments. In war-torn regions or in areas under authoritarian control, library materials are censured and cultural epochs are erased or destroyed. The heated debates that took place at the turn of the late 19th and early 20th century over the creation of a public or municipal library in Montréal and in the struggle between religious and secular forces over control and direction of public reading characterize a lengthy discourse that parallels the development of the Bibliothèque nationale du Québec and of public libraries in Québec. The various stages of evolution and current metamorphosis of the Bibliothèque nationale / Grande Bibliothèque can be viewed as a reflection of the evolution and metamorphosis of society and cultural memory in Québec throughout the nineteenth century to the present.

Advanced technology is often considered to be the property of those individuals or corporations based in the richest and most powerful nations, or the rich and powerful individuals or corporations that can be found in the world's poorest countries. It is typically the case that the most disadvantaged populations do not have equal access to the internet, laptops or personal workstations, fabricating machines, or any of the many software programs or hardware components that are available to many of the world's workers and citizens. There is a tremendous gap between those with access to advanced technology and those without.The aim of this thesis is to address this gap by speculating on the potential of implementing an advanced software application—Building information Modeling (BIM)--into several low technology settings that exist in the author's home country of Nepal.To advance this question, a number of research tactics were engaged: field research was conducted in Nepal during the summer of 2006; advanced digital applications, including Revit and BIM software, were studied and used both as a graduate student and an intern in an architectural firm in Atlanta, Georgia; and the non-governmental organization READNepal was contacted and studied, as was the architectural office with which they collaborate in Katmandu.Three different sites in Nepal were selected to highlight the differences present in the rural areas in terms of cultural, social, and physical qualities, as well as to consider the different "cultures of building" and levels of technological sophistication in the three settings.In time, "trajectories" were developed that link the digital tools readily available to the READNepal offices in New York and Nepal, with the architect's office in Katmandu, and with the contractors and day laborers (who are often illiterate) in the countryside. In this sense, it is suggested that through the introduction of basic software and hand-held devices, information can flow not only from the funding organization's office in New York to the architect, or from the architect to the job site, but from the job site to the architect and READNepal office as well. In this way, communication is improved and the architecture has the potential to be more localized. Power is distributed and knowledge flows not only from top-to-bottom, but from bottom-to-top, from day laborer in rural Nepal to architect in Kathmandu to New York office worker.The thesis offers not a concrete solution but rather an understanding that while potential exists, there is much more to be done to bridge the gap that exists between clients, architects, and constructors in societies where gaps exist between those who have access to high technology and those who do not.

Abstract:
Classrooms, libraries, student unions, and university campuses shape students’ learning experiences. These physical learning spaces set the stage for college student engagement and academic performance. Most of the research about the role of physical spaces in learning lacks the student perspective. The goal of this study was to offer a student-centered vision of ideal learning spaces. Students are the learners for whom learning spaces are designed, and this thesis examines the way students of one summer class at Oklahoma Baptist University conceptualized and interacted with their learning spaces. Data collection included surveys of the students, a focus group with members of the class, participant observation in the classroom, and interviews with students and the professor. Students viewed physical spaces as the backdrop for human action and chose spaces that supported their learning styles and goals. Students described supportive spaces as warm, purposefully crafted spaces, and full of other people who were seriously pursuing the same goals. This thesis explores the ways students conceptualized and interacted with learning spaces as a network of support for their learning and provides recommendations for the design of learning spaces that facilitate this support.

Aujourd’hui, les bibliothèques universitaires (BU) en Syrie sont confrontées à une évolution de leur environnement liée en grande partie à la mutation de l’enseignement supérieur et l’arrivée de nouvelles technologies. Ainsi, les BU se trouvent dans l’obligation de s’interroger sur la pertinence de leurs offres de services. En effet, les besoins et les attentes des universitaires ne sont plus les mêmes qu’auparavant. Ces bibliothèques doivent, par conséquent, suivre cette évolution et répondre à leurs nouveaux besoins pour pouvoir survivre dans un contexte où elles ne sont plus la seule source d’information. L’objectif de cette thèse est de contribuer à la réflexion sur l’adéquation entre l’offre et les besoins. Pour y parvenir, des enquêtes quantitative et qualitative ont été menées auprès d’étudiants, d’enseignants, de dirigeants et de bibliothécaires dans cinq universités syriennes. Cette enquête a été réalisée dans le but de connaître la situation actuelle des BU et des besoins informationnels de leurs publics. Nous avons, ensuite, élargi et diversifié la gamme de services proposés aux universitaires tout en partant de leurs besoins tant exprimés qu’implicites
Today, in Syria, university libraries are facing an actual change in their environment. This change is largely due to the evolution of higher education and the advent of new technologies. In this context, university libraries have to question the relevance of their services. Indeed, information needs have evolved and are no longer the same as before hence libraries need to adapt to evolution by meeting new needs in a context where they are no longer the only source of information. This thesis aims at contributing to reflection on adequacy between supply and service needs. To achieve this goal, a quantitative and a qualitative survey was conducted among students, professors, managers and librarians in Syria to have a clear understanding of the current situation of university libraries and information needs of their publics. We then expanded and diversified the group of services and provided the academics with the basis of their stated and implied needs

The roles of the person who works in a school library, as well as their title - librarian, teacher-librarian, library teacher, library media specialist, school librarian, library media teacher - have undergone countless revisions since the first official school libraries opened their doors in the early 1900s. Although school library media specialists (LMSs) have struggled to negotiate their identities in public K12 education for decades, this "identity crisis" seems to have reached a critical point due to changes in U.S. learning environments brought about by federal legislation, the implementation of standards-based teaching, the emphasis on standardized criterion referenced testing, and the proliferation and ubiquitous use of computers and the Internet as information sources. Although teacher identity has been thoroughly studied, the ways in which LMSs describe themselves in their professional role and how their identities change from pre-service to in-service are rarely investigated. Using Holland, Lachicotte, Skinner, and Cain’s (1998) theory of identity and their concept of figured worlds as a framework, this study explores how four novice LMSs negotiated the identities made available to them in the figured worlds of their public K12 school environments. The following questions guided the study: 1. How do novice library media specialists describe their professional identities? a. How does personal history inform the construction of professional identity of novice library media specialists? b. How do novice library media specialists negotiate identity within the figured worlds of public K12 schools? The four participants were first year LMSs recruited from a large urban school district in the southeastern United States. Data collection took place over the course of the 2011-2012 school year and included in-depth interviews, document analysis, journal responses, and observations. Findings indicate that the figured worlds in which novice library media practitioners begin their careers are often shaped by the experiences that faculty, administrators, and students have had with previous LMSs and bear significant influence on the identities afforded new LMSs as well as their own experiences with LMSs prior to their preparation programs.

Abstract:
Aim. Most of the literature related to the study of library and information science tends to spring from a modern, positivist paradigm. Only a small number of theorists, working in what some describe as “the periphery” of library and information science (Trosow 2001), explore the ways that postmodern thought can be applied to the study of libraries. This thesis looks at issues related to a particular public library through both modern and postmodern perspectives. The insights evoked raise questions about many of the basic tenets of public librarianship. Scope. The subject of this study is the Vancouver Island Regional Library, a large integrated public library system on the west coast of Canada. The researcher is also the Executive Director. The underlying conceptual framework includes the idea that modern and postmodern thought are reflective of different epistemologies and play out differently within the contemporary social condition. The research is grounded in a qualitative, constructivist research tradition. Mailed questionnaires, individual interviews, and group interviews, using open-ended questions, are used to elicit opinions from board members, qualified librarians, local branch heads, and customers. Comments that seem to indicate a modern or postmodern perspective about such things as truth, knowledge, belief, the knowing subject, language, binary pairs, the role of the author, the organization of knowledge, the increase in information, professionals, power, the panoptic principle, moral panics, significant groups, feminism, culture, premodern phenomena, work, and bureaucracy, as they relate to the Vancouver Island Regional Library, are interpreted hermeneutically. This interpretation, it is argued, evokes new and interesting insights into the Regional Library that would not be found through scientific analysis. A second cycle of hermeneutic interpretation evokes scepticism about many of the modern, positivist tenets of public librarianship that one participant suggests are “inculcated” into librarians. Questions are raised about such long-standing beliefs as the need for the librarian’s neutral stance; the necessity of the balanced collection; the public library’s role in supporting intellectual freedom; the intrinsic difference between fiction and non-fiction; the scientific nature of the Dewey Decimal Classification system; the superiority of professional librarians; the possibility of complete, correct reference transactions; the need to charge penalties to ensure materials are returned on time; the belief that the female nature of the workforce is not significant; the effectiveness of library bureaucracy; the need for increased tax support for public libraries; and traditional ways of planning for the future of public libraries. A number of pragmatic strategies are suggested. Conclusion. The principal conclusion of the research is that postmodern thought can offer new insights into the Vancouver Island Regional Library. By implication it can help librarians and library and information science theorists understand other public libraries differently and, in so doing, assist public libraries adapt to the postmodern social condition.

Reality TV is the most popular genre of television programming today. The number of reality television shows has grown exponentially over the last fifteen years since the premier of The Real World in 1992. Although reality TV uses styles similar to those used in documentary film, the “reality” of the shows is questioned by critics and viewers alike. The current study focuses on the “reality” that is presented to viewers and how that “reality” is created and may differ from what the participants of the shows experience. I appeared on two reality shows, Faking It and That's Clever, and learned a great deal as a participant observer. Within the study, I outline my experience and demonstrate how editing changed the reality I experienced into what was presented to the viewers. O'Connor's (1996) representation context web serves as a model for the realities created through reality television. People derive various benefits from watching reality TV. Besides the obvious entertainment value of reality TV, viewers also gather information via this type of programming. Viewers want to see real people on television reacting to unusual circumstances without the use of scripts. By surveying reality TV show viewers and participants, this study gives insight into how real the viewers believe the shows are and how authentic they actually are. If these shows are presented as reality, viewers are probably taking what they see as historical fact. The results of the study indicate more must be done so that the “reality” of reality TV does not misinform viewers.

Flowering is a fundamental process in the life cycle for plant. This process is marked by vegetative to reproductive apical meristem conversion, due to interactions between several factors, both internal and external to plant. Therefore, eight subtractive libraries were constructed using apical meristem induced or not induced for two contrasting species: Solanum lycopersicum cv. Micro-Tom and Solanum pimpinellifolium. Several cDNAs were identified and among these, were selected two cDNAs: one homologous cDNA to cyclophilin (LeCYP1) and the other to Auxin repressed protein (ARP). It has observed that LeCYP1 and ARP genes are important in the developmental process to plants. In silico analysis, were used several databases with the exclusion criterion E-value <1.0x10-15. As a result, conservation was observed for proteins analyzed by means of multiple alignments and the presence of functional domains. Then, overexpression cassettes were constructed for the ARP cDNA in sense and antisense orientations. For this step, it was used the CaMV35S promoter. The cDNA orientation (sense or antisense) in relation to the promoter was determined by restriction enzymes and sequencing. Then, this cassette was transferred to binary vector pZP211 and these cassettes were transferred into Agrobacterium tumefaciens LBA4404. S. lycopersicum cv. Micro-Tom (MT) and MT-Rg1 plants were transformed. In addition, seedlings were subjected to hormone treatments using a synthetic auxin (- naphthalene acetic acid) and cyclosporin A (cyclophilin inhibitor) treatments and it was found that the hormone treatment there were changes in development of lateral roots pattern, probably related to decreases in auxin signaling caused by reduction of LeCYP1 in MT-dgt plants while cyclosporin A treatments, there was a slight delay in flowering in cv. MT plants. Furthermore, assay with real-time PCR (RT-qPCR) were done for expression level analysis from LeCYP1 and ARP in order to functionally characterize these sequences in tomato plants.

The development of library Consortia in Zimbabwe was necessitated by the need to reduce subscription costs and to widen access to electronic resources as well as implement new technologies among academic libraries. The development of Zimbabwe University Library Consortium (ZULC) and College and Research Library Consortium (CARLC) enabled libraries to cooperate and collaborate in building capacity to support teaching, learning and research through access to quality scholarly information. The trajectory of consortia development in Zimbabwe since 2002 has however been characterised by a focus on the academic sector to the exclusion of other types of libraries. The future development of library consortia in Zimbabwe can be better envisioned when correlated with the country’s national development agenda. While not made explicit, this agenda is underpinned by the idea of access to information. This study investigated how the benefits of the existing library consortia can be harnessed to promote the achievement of Zimbabwe’s national development goals. More specifically, it examined the ways that the development paths of ZULC and CARLC can be transformed to support the country’s national development agenda and programmes. This culminated in a model that will accelerate and guide the future development of its library consortia to facilitate a supporting developmental role. The novel aspect of this study is that it seeks to integrate library consortia into the national development plans of a developing country and to extend their benefits as widely as possible. An extensive literature review of the characteristics, models, and development of consortia in selected countries was complemented by an empirical mixed-method component that generated data through interviews, questionnaires, observation, and the analysis of key documents. A special feature of the study is a detailed analysis of the successes and challenges of library consortia in other countries and in the Southern African region to supplement the empirical data that informs the proposed model. The main finding is that a model with a multi-type structure and a National Coordinating Committee is best to transform the development paths of Zimbabwe’s academic library consortia to support the country’s national development agenda. The model’s key elements are finance, structure, governance, functions, and special features.

Public libraries are mainly employing women and have female managers. According to theories based upon so-cial construction we create our roles together in our society. The role of a manager has for a long time been viewed as the role of a man. The objective of my thesis is to gain knowledge on how a few women that work as managers in public libraries perceive and describe their roles. A parallel aim is to let a few future librarians de-scribe their managers and their own views on leadership, with a focus on gender issues. My questions are the following, how do the women construct their roles as managers of public libraries?, how do the female students describe the role as a female manager of a public library, and their own managers?, can stereotype views and attributes be distinguished in my informants tales on leadership and gender?The starting point of this thesis is theories of social construction of Peter Berger and Thomas Luckmann, the thought that we shape our world through social interaction, mainly through our language. The theories of Yvonne Hirdman, regarding gender power structure, offers the political backdrop for my thesis. Alice Eagly has re-searched around leader stereotypes, leadership styles and gender. From her and her peers I take with me theories that claim that the leader stereotype is gendered male.The material I have used to answer my questions are interviews with ten informants, all of them women working at public libraries, five of them managers and five students. I have conducted structured interviews with open questions that I have then looked at by means of a method where different factors are intertwined, mainly language, social situations, political factors and theory. All these then used to interpret the empirical evidence. The basis of the method is interpretation and reflection.I found that my informants construe an image of the role as a woman and a manager of a public library as a complex role that does not fall into one unambiguous gender stereotype. They describe their one manager roles and their managers with attributes and characteristics that in recent research has been assigned to both genders. I could nevertheless identify the use of strategies that according to Eagly et al is used by women in manager roles to prevent them from suffering negative criticism or other consequences solely because of the combination of their gender and position.

MicroRNAs (miRNAs) are a class of small regulatory RNAs, which act as post-transcriptional modulators of gene expression. Because of their critical roles in gene expression, genome-wide profiling of miRNAs, by qRT-PCR or array, has become an essential step in understanding cellular biology. In this thesis, we have developed a novel method for constructing miRNA libraries, as an initial step toward genome-wide miRNA characterization. We purified and cloned miRNAs by HPLC from several cell lines. Individual clones were sequenced, and identified by bioinformatics. It was estimated that about 30-40% of the clones from each library represented authentic miRNAs.

Lipase (triacylglycerol acylhydrolase; EC 3. 1. 1. 3), an enzyme which catalyzes the hydrolysis of triacylglycerol to glycerol and fatty acids, has been widely used in fat, food ingredients, detergents and biotechnology industries. Sesame(Sesamun indicum L.), belongs to the Pedaliaceae family, is one of most important oilseed crops in the world. Sesame seed possesses more than 50% oil content in dry weight. The storage triacylglycerols (TAGs) is used as energy source for germination and subsequent post-germinative growth. Because more than 50% of oil in sesame seed is exhausted after sprouting for two days, it is suggested that the lipase exhibits highest activity during this stage. The purpose of this study is to construct cDNA libraries from germinating sesame seeds and to clone the lipase genes for functional analysis of gene expression. Sesame seed extracts from different time points after imbibition are subjected for assaying lipase activity. It is found that lipase activity increases after imbibition and the maximum activity is detected at 16 hours after imbibition. Detection of lipase gene expression and cloning of partial cDNAs at different time points are conducted by PCR cloning by the use of designed plant lipase degenerate primers. Three partial lipase/esterase cDNA sequences are cloned, and the result of sequence alignment shows that they may be different lipase/esterase genes. Therefore, cDNA libraries of 8 and 12 hours after imbibition are constructed and used to clone full-length lipase/esterase cDNA sequences. Expression analysis of three lipase genes could be further studied.

A major goal of synthetic biology is to engineer synthetic gene circuits with desired behaviors. Up to now, however, it still requires extensive and iterative work. The main obstacle is the lack of some well-characterized biological parts and design methods. By the identification technique, the biological parts can be well-characterized to build a useful library for engineering a novel gene circuit. Hence how to build a useful library of biological parts for engineering synthetic gene circuit in vitro and in silico is an important topic from the viewpoint of synthetic biology. Promoters and RBSs are regarded as a lumped component in this study so that the promoter-RBS component can help us construct useful promoter-RBS libraries by the identification technique. The promoter-RBS libraries can be easily used to engineer a synthetic gene circuit before laborious process of trial-and-error. In general, there exist 3 kinds of promoter-RBS components in synthetic biology, hence 3 kinds of promoter-RBS libraries are constructed in this study, i.e., constitutive, repressor-regulated and activator-regulated promoter-RBS libraries.We provide the construction procedure for the constitutive, repressor-regulated, and activator-regulated promoter-RBS libraries. After building 9 promoter-RBS libraries, we provide the characteristic indexes of these libraries through their mathematical models, a systematic method is proposed to help us select the adequate promoter-RBS component set. Finally, we provide a library-based search method to help us quickly select the most adequate promoter-RBS component set from promoter-RBS libraries. The proposed library-based search method can reduce the number of trial-and-error experiments in selecting an adequate promoter-RBS component set for a synthetic gene circuit, and then a gene circuit can be predictably implemented by a systematic design method before trial-and-error experiments.

Numerous nucleoside analogs play an important role in acting as anti-viral, anti-tumor and nuclear imaging agents. However, there are a variety of new viruses and various mutants appear, which may results in resistance or inefficiency of anti-viral activity. In this study, as the starting material, uridine was derivatized to afford 4'-α/β-aminomethyl uridine analogs followed by coupling with various acids for construction of libraries via amide-bond formation. 2',3',5'-hydroxyl groups of uridine were first protected with TBDMSCl, followed by restoring 5'-OH group with HOAc. After oxidation with EDC, the aldehyde could be produced. Treatment with aqueous HCHO and NaOH and subsequent reduction with NaBH4, the 4'α-C-branched chain could be generated. Attempt to introduce azido group by nucleophilic substitution of the tosyl group was unsuccessful even under 110 oC. Whereas triflate proved to be an excellent leaving group, only cyclized product 2',4'-BNA was obtained when one free hydroxy moieties were reacted with lithium azide. This was confirmed by comparing with the reported NMR spectra, which showed only singlet for all the three protons (coupling constant was zero) on C1', C2' and C3'. Ditriflates could be prepared when two equivalents of triflic anhydride was used. Repeating the treatment with lithium azide, the desired azido groups could be introduced and no cyclized product was observed. The steric interference raised by the second trifluoromethan -sulfonyl group was proposed to account for the unfavorable nucleophilic attack of 6'-OTf on 2'-TBDMS. Removal of TBDMS group was easily achieved using TBAF, which was followed by transformation of the azido groups to the diamino groups under reduction conditions of hydrogen gas and Pd/C. As the target compound 4',4'-bis-aminomethyl uridine analog was in hand, further coupling with various carboxylic acids to provide the compound libraries through amide linkage are performable. Relevant biological assay studies to find out the potential compound are in progress.

At present, it is still appealing to have promoter-RBS libraries for systematic design of synthetic genetic circuit in Cyanobacteria for a variety of application. In the Cyanobacterium Synechococcus sp. strain PCC 7942, we set up three kinds of promoter-RBS libraries, i.e., constitutive, repressor-regulated and activator-regulated promoter-RBS libraries, for systemic design of synthetic gene circuits in Cyanobacteria. In order to engineer synthetic gene circuit into Cyanobacterium Synechococcus sp. PCC 7942, we provide the flow chart to constructing constitutive, repressor-regulated, and activator-regulated promoter-RBS libraries based on promoter-RBS intensities through experimental data, i.e., , , and , respectively. By the parameter identification technique based on dynamic model, a promoters-RBS is regarded as a characteristic part to be selected for gene circuit design with a desired behavior in Cyanobacteria, and regulated by different biological parts in blue-green algae. A design example with desired cell population control is given to illustrate the design procedure of synthetic circuit in Cyanobacteria by the proposed promoter-RBS library-search method and is validated by experimental data in vivo. For the convenience of design, we provide a systematic method for a genetic circuit to control part’s intensity to achieve some desired behavior in Cyanobacterium Synechococcus sp. PCC 7942. Consequently, we believe that the proposed library-based search method can help user select an adequate set of promoter-RBS parts to realize various kinds of application in rapidly developed synthetic biology.

Nous y analyserons donc la création des bibliothèques universitaires modernes au Québec à travers les activités de leur principal organisme de concertation, le Sous-comité des bibliothèques de la Conférence des recteurs et des principaux des universités du Québec (CREPUQ), de sa création en 1967 jusqu’en 1975. Nous relèverons ainsi que le capital symbolique des bibliothèques est en fait fonction de leur capacité à appuyer la double mission des universités, la recherche et l’enseignement, par leurs ressources et leur expertise. Dans ce contexte, le sous-comité des bibliothèques adoptera une stratégie visant la reconnaissance de cette expertise, l’optimisation de l’utilisation des ressources documentaires de ses membres et l’augmentation de leurs crédits budgétaires. Il défendra également l’autonomie des bibliothèques face à diverses politiques émergeant de l’État et de la CREPUQ. Enfin, nous noterons la prépondérance de la coopération dans l’habitus du microcosme des bibliothèques universitaires et conclurons que, si les bibliothèques réussissent généralement à atteindre leurs objectifs lorsque l’enjeu relève directement de leurs compétences, elles échoueront lorsque leurs intérêts demandent un investissement financier important.
In the late 1960s, at a time when the State was investing heavily in higher education, librarians had to meet a greater need for documentary resources than ever before. To answer this need, they established a cooperative network, thus consolidating the microcosm of Quebec university libraries. This paper is an exploratory study of this microcosm evolution in the academic field, using Pierre Bourdieu's sociological concepts of field, habitus and capital. In this Master's thesis, we analyze the creation of modern Quebec university libraries through the activities of their main consultative body, the Conference of Rectors and Principals of Quebec Universities’ (CREPUQ) Libraries subcommittee, from its creation in 1967 to 1975. We theorize that the symbolic capital of libraries depends on their capacity to support, through their resources and expertise, the universities’ research and teaching mission. In this context, the library subcommittee will adopt a strategy to promote its members’ expertise, to optimize the use of their resources and to increase their budget. It will also defend the libraries autonomy against various policies adopted by the State and the CREPUQ. We conclude by noting the predominance of co-operation in the university libraries microcosm's habitus and by highlighting the fact that, if libraries generally succeed in achieving their objectives when it falls within their competencies, they systematically fail when their strategies require a significant financial investment.

Abstract:
The diploma thesis deals with unrealised projects of new buildings or refurbishments of libraries in the Czech Republic. It is notionally split into two parts. The theoretical part contains the reasons, why to build library buildings today and actual requierements on these specific kind of buildings. The most important czech and foreign documents on this topic are presented. One specific part is dealing with funding of library high-priced projects and also examples of successfull projects. The main focus of work is a summary of six public libraries focussed on their poor space conditions. After this, gathered data are equated with each other, with recommended values in Standard of Public Information Library Services and with comparable data of libraries placed in new or reconstructed buildings. In conclusion, the author questions subjectively the future of these six libraries. Part of the thesis is also extensive supplement with photos and graphic visualizations of unrealised projects.

Abstract:
Specialised subject gateways have become an essential tool for locating and accessing digital information resources, with the added value of organisation and previous evaluation catering for the needs of the varying communities using these. Within the framework of a research project on the subject, a software tool has been developed that enables subject gateways to be developed and managed. General guidelines for the work were established which set out the main principles for the technical aspects of the application, on one hand, and on aspects of the treatment and management of information, on the other. All this has been integrated into a prototype model for developing software tools. The needs analysis established the conditions to be fulfilled by the application. A detailed study of the available options for the treatment of information on metadata proved that the best option was to use the Dublin Core, and that the metadata set should be included, in turn, in RDF tags, or in tags based on XML.

As the increasing popularity of Internet, personal computers, and the evolution of information communication technology (ICTs), students' learning styles has also gradually changed for a variety of needs for knowledge and learning. Knowledge building, as an important concept, describes what a learning community needs to accomplish in order to create knowledge. It also refers to the process of creating new cognitive artifacts based on the common goals, group member discussions, and synthesis of ideas within community. Nevertheless, during the learning process of Knowledge building, there are some problems raised while guiding students to keep their in-depth discussion. In this study, we attempt to find out effective methodologies based on the assistance of library support, to promote learners’ participation, learning achievement and information literacy skills. First, this study introduces a way in which librarians participate in students’ learning process as tutor. Second, the research was conducted by quasi-experiment design; by which an on-line Knowledge Forum for students’ knowledge building learning environment is lunched. Discussion, learning activities and learning materials are delivered through Knowledge Forum. Researchers ask students choose one of four topics (Digital divide, Group dynamic, Attitudes and behavior change, Online persona) to discuss on Knowledge Forum. Students were asked to adopt concept mapping to construct their mind-map and knowledge flow in class. In addition, their learning performance was evaluated based on the process of knowledge mapping (3 times), the content of discussions, tasks, reports and grades. Their works of concept map were analyzed by UCINET, visualizing students’ mind changing, how they organized their knowledge, and to identify the positive effects on students’ performance. Moreover, the study also adopts various data gathering methodologies, such as questionnaires as well as the interaction records between librarians and students for overall understanding the Knowledge construction process. The results showed that librarians’ involvement in learning activities do help students’ improvement in knowledge constructing level, keeping their reflective process in-depth. The data also showed that students using concept maps in the knowledge building learning process as a sound way to assist their tasks accomplishment. This research suggests, in the future, there could be more successful collaboration, such as Knowledge building, among librarians, students, and teachers in different types of educational scenarios. Both educators and students will not only understand the importance of the Knowledge construction for better learning performance but also aware librarians as well as their service as an indispensable role in better education setting.

Abstract:
This thesis concerns itself with the notion of a networked or cooperating library and the discursive means by which it is constructed in documents related to the formation of a national strategy of libraries in Sweden. I study the draft as presented by the national library, Kungliga Biblioteket (KB), in early 2018, the comments and advice submitted by different parts of the library sector during the fall of 2018 as well as the final proposition handed over to the Minister of Culture in March 2019. The study focuses on what defines or constitutes a networked library, on the significance of a networked library for the perceived identity of libraries and library professionals and finally, on what grounds a networked library is ascribed legitimacy or is subject to criticism. This is worth investigating because of the calls for increased collaboration and coordination of different parts of the library sector and its implications for the professional identity as well as the wider understanding of what a library is and what a library does. The method of approach is a thematic document analysis informed by discourse studies. Discursive strategies are used as the main analytic tool. Strategies are adopted from the critical vein of discourse analysis (CDA) but are applied here in a way consistent with poststructuralist discourse theory and its ontological assumptions. The results show that the library sector is going through a dilemma of redefining, adjusting, modernizing, and broadening the meaning of libraries or keeping with a long-standing tradition that not always corresponds with the library practices of today. This tradition is subject to changing conditions – not least due to the digital transformation – as much as it remains a firm ground for legitimizing the professional field. A networked library requires a stale image to be left behind in favor of a collaborative library infrastructure, extending its meaning towards a library identity which adheres to the conditions of a networked world. In conclusion, a networked library is found in the middle-ground between a traditional identity and creating a new story about libraries.

Abstract The purpose of this research is to understand English teachers’ attitudes and needs for the practice of e-Learning. It also aims to discuss the mode for establishing a cooperative relationship with the English teachers while constructing English e-Learning environment in school libraries. A questionnaire drafted according to related literature helps to produce a more objective analysis of the matter. Using the stratified sampling method, the study chose English teachers of junior high schools in Taipei City as survey subjects. The data from the questionnaire are analyzed by packaged statistics software SPSS. The results indicate that (1)The English teachers have positive attitudes on the application of e-Learning in English classes, but the appropriate timing, application events and the integration methods for the teaching materials should be adjusted;(2)A multitude of lively e-Lesson media and complementary extra-curriculum materials are utilized by English teachers, however it is uncommon to find teachers making use of self-made multi-media instruction materials;(3)7 needs for constructing English e-Learning physical environment are generalized: Encouragement for Participation, Group Negotiation, Better Facilities, Technical Assistance, Practical In-service Training, Adjustment of Curricular Affairs, and Resource Integration;(4)The modular functions of the English e-Learning platform adheres to the principle of “complementing school syllabus and improving learning effectiveness”;(5) Most English teachers are willing to assist with the construction of English e-Learning environment;(6) With the missions of teaching support and resource integration, school libraries need to construct a multi-directional English learning environment under a cooperative relationship with English teachers. Finally, based on the results, this study makes some suggestions as the reference for the promotion of English e-Learning in the junior high schools.

This study investigates, within the current health care situation, the interrelationship of the user, resources and tool in the design of a prototype WELLNESS database-driven web site. A shift has taken place in health care, in which the base of conventional medicine has broadened to integrate other systems, practices and worldviews. These include complementary and alternative medicine, health promotion, disease prevention and wellness. Emphasis is placed on the need to take personal responsibility for one's own health and wellness. The global burden of chronic disease, reaching epidemic proportions, is increasingly linked to risk factors resulting from personal lifestyle choices. The growing evidence of the user's need to make personal, informed, lifestyle choices and their reliance on the Web for health information, required investigation. WELLNESS, a specific orientation to health and wellness, formed the framework within which the user and resources were defined and the tool designed. The user was profiled as the WELLNESS health information seeker, hereby contributing significantly to an understanding of the user in this new context. The user profile informed the establishment of resource selection criteria and tool design. The identification of WELLNESS content selection criteria, within a five-dimensional model, was required to ensure quality, relevant and credible resources. The tool is comprised of the WELLNESS thesaurus and WELLNESS database-driven web site. The WELLNESS thesaurus was constructed based on a combination of relevant thesauri. It will be used as an indexing tool. An investigation of existing health information web sites highlighted the importance of designing a specific WELLNESS database-driven web site. A database host was identified against which the original study's conceptual schema was assessed. A low-fidelity prototype web site was designed as the interface between the WELLNESS health information seeker and the database of WELLNESS health information resources. This study has epidemiological, philosophical, epistemological, sociological and psychological relevance. The provision of access to WELLNESS health information resources, made available in the WELLNESS database-driven web site, for personal, informed lifestyle choice by the WELLNESS health information seeker could potentially contribute to the reduction of the global burden of chronic disease.
