Nitrile Hydratase (NHase) is a bacterial enzyme which catalyzes the hydrolysis of organic nitriles to their corresponding amides. The active site of NHase contains Fe (III) or Co(III) ions in an unusual coordination sphere containing 2 amide N atoms and three S donors. The research described herein is an attempt to model this active site using two different model systems: one that generates metal-coordinated imines, and one that generates metal-coordinated amides. Using 2,2’-dithiodibenzaldehyde and derivatives of 2,2’-dithiodisalicylic acid as reactants, several new and interesting metal coordinated imine and metal coordinated amide complexes have been synthesized. The synthesis and crystallization of several new organic and ligand species is discussed as well. The synthesis of a Co complex that resembles the active site of NHase is reported herein, along with several other new organometallic species. Also, two projects using computational chemistry are reported: one modeling trispyrazolylborate (Tp) complexes of iron and copper, and one modeling iron, manganese, and copper complexes of pentadentate imine/phenolate ligands. Complexes of Tp ligands with relatively bulky substituents in the 3-position are subject to isomerization through a borotropic shift. Computational chemistry has shed light on the energetics behind the borotropic shift. Calculations on the CO stretching frequency of several different TpCu(I)CO complexes have been carried out in an effort to match experimental data and generate predictive values for several other species not yet synthesized. This stretching frequency can generally be directly related to the electron withdrawing/donating character of the Tp ligand attached to the metal center. Pentdentate imine/phenolate ligands can coordinate to transition metals in a number of isomers. Computational chemistry has been used to shed light on the structural preferences of the iron, manganese, and copper species. These calculations were able to match the experimental data for crystallographically characterized species, and the metal complexes were shown to have specific structural preferences depending on the identity of the metal used.<br>Thesis (Ph.D.)--Wichita State University, College of Liberal Arts and Sciences, Dept. of Chemistry<br>"December 2007."

Nous avons synthetise et caracterise deux nouvelles familles de porphyrines dont les proprietes se distinguent par la nature des substituants portes par le macrocycle. La porphyrine portant deux unites intercalantes de type acridine est capable d'interagir avec l'adn. L'intercalation des acridines dans l'adn place le centre reactif (la porphyrine) a proximite de sa cible. Des coupures simple brin et double brin de l'adn ont pu etre provoquees apres activation de la porphyrine sous forme d'un complexe de manganese. Les experiences de coupures ont ete analysees par electrophorese. La deuxieme partie de ce travail est la conception de ligands dont la structure se compose d'une tetraphenylporphyrine portant des sites de coordination de type catechol. Les ligands se distinguent par le nombre (de un a quatre) et la position (en meta ou en ortho du substituant meso-phenyle) des groupements catechols portes par le macrocycle. La substitution en ortho, conduit a quatre atropoisomeres , #2#2, #3 et #4. L'orientation selective des groupements catechols a ete confirmee a l'etat solide par radiocristallographie. En solution, une etude prealable par rmn a mis en evidence un processus d'isomerisation conduisant principalement a l'atropoisomere le plus stable.

The certified thesis is available in the Institute Archives and Special Collections.<br>Includes bibliographical references (p. 75, 77).<br>Because of the increasingly globalized world we live in, companies today are very interested in going overseas to develop and utilize global engineering resources. By doing so, they hope to take advantage of new global product development (GPD) enablers and motivators such as the internet, new collaborative information technology tools, access to new markets, and the increasing availability of low-cost engineering talent. While globalization has significantly decreased barriers so that more companies are hurrying to move engineering activities to its global sites, it is no secret that GPD teams pose significant coordination challenges. Cost savings from lower labor rates abroad can easily be eaten up by the increased coordination costs required to manage overseas interactions between local and global activities. This paper introduces a model that maps a project's coordination structure to help managers decide which activities should be allocated to a global site and which ones should be kept at home. It introduces a new multi-site coordination matrix based on the Design Structure Matrix and an optimization model that chooses where to locate activities to minimize project coordination costs.<br>(cont.) A key principle the model relies on is the modularization of activities at each site for efficient organization design. A partir d’une enquête qualitative, s’appuyant sur des entretiens semi-directifs auprès de professionnels occupant des postes à responsabilité dans les milieux de la presse et de la publicité sur le web, nous analysons la construction du marché et ses logiques de fonctionnement. Ces dernières interrogent la continuité du modèle publicitaire de la presse sur ce média. Notre objectif est de comprendre la manière dont la valorisation marchande de l’audience y évolue pour les sites issus de la presse imprimée. Les modes de quantification et de qualification de l’audience se transforment et marquent l’affrontement entre une logique issue de la publicité média et une autre relative au marketing direct, tirant parti du caractère interactif de la publicité en ligne. Trois dimensions complémentaires structurent notre travail : l’évolution de la relation marchande entre les éditeurs et les annonceurs et la manière dont elle est outillée (partie 1) ; l’inscription du marché dans un contexte socioéconomique imposant des contraintes spécifiques et une tension autour de modes de qualification alternatifs du produit publicitaire (partie 2) ; le rôle joué par l’appropriation des innovations techniques par certains acteurs ainsi que la mesure dans laquelle cette appropriation détermine, au moins en partie, l’évolution de l’activité publicitaire (partie 3)<br>The online advertising market is a complex, unstable and evolutive one. Our thesis describes and explains the way this market works for online news publishers from traditional news organisations. From a qualitative survey, based on semi-structured interviews with professionals in positions of responsibility in online news organisations and online advertising industry, we analyze the market construction and its operating logic. The latter questions the continuity of the news organisations advertising model online. Our goal is to understand how operates the audience markets valuation online for those traditional news market players. The audience quantification and qualification process are changing and show us conflicting logics between a logic imported from offline media advertising and another one from direct marketing, taking advantage of the interactive nature of online advertising. Three distinct but complementary points structure our work: the evolution of the market relationship between publishers and advertisers and how it is equipped (part 1); the inclusion of the market in a socio-economic context which imposes specific constraints and a tension between alternative modes of advertising product’s qualification (part 2); the role played by the appropriation of technological innovations by some players as well as the extent to which this appropriation determine, at least in part, the advertising business dynamics (part 3)

Entre le début du xviiie siècle et les années 1970, soit durant plus de deux siècles et demi, il s'est noué dans les Alpes Briançonnaises un ensemble de relations entre une ressource, le charbon, et une population d'hommes, paysans et montagnards. Cette étude s'attache à faire le récit de ces relations, à parcourir la chaîne des associations au moyen desquelles les paysans et le charbon se sont simultanément inventés, concourant à la constitution d'une socio-nature, concrétisée dans des formes spécifiques d'exploitation, les charbonnières, qui ont cohabité durablement avec les petites mines industrielles installées progressivement dans la bassin. Le réseau de l'activité charbonnière paysanne tient à quatre associations successivement explorées : la relation au charbon et sa naturalisation comme charbon-qui-convient ; l'investissement " communaliste " de la forme d'État de la concession ; la stabilisation d'un dispositif sociotechnique qui offre une troisième voie à l'alternative socio-économique à laquelle sont soumises les population montagnardes : émigrer ou rester et subir le " bagne " de la mine industrielle ; enfin une relation instrumentale à la technologie, c'est-à-dire l'équipement pratique et discursif de la mine rationnelle et industrielle porté par les ingénieurs des mines. Dans ce récit, on ne rencontrera pas de structures sociales, pas de systèmes techniques et bien sûr pas leurs agents théoriques autres que ceux que mobilisent les personnages du récit, dans les filets desquels ils se prennent, avec lesquels ils composent, auxquels ils se soumettent, qu'ils utilisent comme guide pour l'action ou sur les terres desquels ils braconnent. Mobilisant les cadres d'analyse des sciences studies, de la sociologie pragmatique et de la micro-histoire, cette étude propose la relation singulière d'un moment de la société, considéré dans son individualité historique.

This study aims to recognize how virtual teams should be coordinated and led in organizations that operate at multiple sites. To fulfill the purpose of the study, a literature review on previous research on virtual teams was implemented. Further, a qualitative study including interviews with leaders at different levels at Ericsson was conducted to gain empirical data on how leadership in virtual teams works. The interviews were semi-structured with the purpose of not directing the leaders towards only bringing up the challenges that emerged in the literature review but to also bring up new perspectives. The major challenges that virtual teams face, thus challenges that the leaders must somehow address, can be divided into three areas: communication, coordination and location. Concerning communication, it is important that leaders of virtual teams set the means for communications, define ground rules and make sure that the right kind of communication is used for the right purpose. Further, it is important to communicate more often than if the team would have been co-located and actively try to build trust and cohesion within the team by being transparent and honest. To meet face to face with the team members, especially in the beginning when the team is created, is important both for understanding (virtual communication becomes easier afterward) and for the building of trust and cohesion. In the area coordination it is important for the leader to clearly define the roles of each team member and how the roles as well as the team efforts contribute to a larger picture. There is no perfect distribution that is applicable to all virtual teams, each team must find their balance concerning how many team members that should be positioned at each site. In general, to push responsibilities down in the hierarchy is a suitable role structure for virtual teams. The location area includes challenges such as different languages, cultures and collaboration across different time zones. It is important to have an operative language that all team members understand and master. When creating a new team, it is important that the leader or the person creating it, is aware of how cultural differences impact, so that the challenges can be addressed properly, and the benefits of diversity as well as the expanded labor pool, can be utilized. Working across different time zones brings benefits such as more hours of the day can be used, but it also includes challenges, such as limited real-time communication. Thus, the optimal time overlap is difficult to define, but generally, leaders prefer involving fewer time zones and large time overlaps.

Construction industry is one of the main pillars of the economy of Hong Kong. Over the years, Hong Kong construction industry has earned a reputation for the rapid construction of quality high-rise first class building. It is a common practice of the main contractors to sublet most of their works to subcontractors in the HK building projects. The percentage gross value of main contract work performed by subcontractors increased from 57 per cent to 67 per cent during 1981 to 2005 according to government statistics for 2006 (Census and Statistics Department, 2007). It is anticipated that more numbers of subcontractors would be involved due to the rapid development of high-rise buildings in the last decade. As a result, the role of the main contractor has been gradually transformed from a constructor to a manager of subcontractors of the projects. The performance of the subcontractors is one of the most important factors governing project performance. In recent years, there are increasing complaints from subcontractors that they cannot performance effectively and efficiently due to poor site coordination by main contractors. An average of 35.10 per cent of productivity wasted due to site coordination problems caused by main contractors was stated by the respondents of a questionnaire survey. The aim of this thesis is to formulate relationships in terms of multiple regression equations to explain how the performance of subcontractors is affected by the critical site coordination problems caused by main contractors, and to develop framework to improve site coordination.

La parésie, l’hyperactivité musculaire et la rétraction des tissus mous sont les principaux mécanismes physiopathologiques responsables de la dégradation de la marche dans l’hémiparésie. Lors de la prise en charge du patient hémiparétique, le questionnement du clinicien s’articule autour de la part de responsabilité de chaque mécanisme, et de l’impact des traitements sur l’organisation globale du mouvement et la qualité de la marche. L’analyse de la coordination inter-segmentaire, utilisant la mesure de la phase relative continue dans le plan sagittal, apporte des éléments de réponse à cette problématique clinique. La quantité et les variations du déphasage entre les segments des membres inférieurs dans les phases du cycle de marche décrivent les stratégies de coordination motrice. Des paramètres issus de la phase relative continue inter-segmentaire, apportent des informations spécifiques quant à l’origine physiopathologique des déficits cinématiques pour aider au choix du traitement, à l’incidence des conditions de réentraînement de la marche, et aux bénéfices d’un traitement. Cette analyse, complémentaire aux évaluations cliniques réalisées dans la pratique courante, met en évidence la restriction de possibilités de modulation par les mécanismes physiopathologiques dans le membre inférieur parétique, et des mécanismes de compensation dans le membre inférieur non-parétique. Les résultats de ce travail pourraient inciter les cliniciens à déterminer les stratégies de coordination motrice, pour définir les traitements permettant au mieux de réduire les anomalies de la marche des patients présentant une lésion motrice du système nerveux central<br>Paresis, muscle hyperactivity and soft tissue retraction are the main mechanisms responsible for the gait disturbance in hemiparesis. In the rehabilitation management of hemiparetic patients, clinicians try to determine the responsibility of each mechanism, and to quantify the impact of treatments on movement organization and gait efficiency. Inter-segmental coordination analysis, using measurement of the continuous relative phase in the sagittal plan, can assist in reaching these objectives. The amount of dephasing between lower limb segments, in each phase of the gait cycle, sheds light on the coordination pattern. Relevant parameters of the inter-segmental continuous relative phase may reveal specific information such as the predominance of neurological and orthopedic factors implied in the kinematic deficits, the impact of various gait rehabilitation conditions, or treatment-related benefits. This analysis, complementary to routine clinical examination, may also disclose specific motor deficits in the paretic lower limb, and compensatory strategies at work in the non-paretic lower limb. The findings reported in this thesis may encourage rehabilitation clinicians to carefully study coordination patterns, which may help define optimized treatments to lessen gait impairment in spastic paresis

Mestrado em Biomedicina Farmacêutica<br>This report describes my experience of nine months as a trainee of a CRO (Eurotrials, Scientific Consultants), as well as a trainee of a clinical research site (Clinical Academic Center – Braga, Association).
This document describes the European framework about clinical research and the Portuguese situation compared to similar countries. The activities developed during this internship are also described. These activities are divided in two phases.
The first one occurred in Eurotrials, Scientific Consultants, a CRO specialized in clinical research and scientific advice. The first weeks were dedicated to intensive self-training needed to perform CTA tasks. These tasks included qualification, initiation and monitoring activities related to clinical trials, as well as the development of a quality management system.
The second phase took place on 2CA-Braga, a clinical research center located in Hospital of Braga. Clinical studies coordination was the main focus of this second phase of my internship, as well as negotiation of clinical studies agreements. I had also the opportunity to participate in “1as Jornadas de Investigação Clínica e Inovação” (1st Clinical Investigation and Innovation Conference) organized by 2CA-Braga.
Globally, this internship was a great opportunity to get knowledge and experience in the implementation and management of clinical trials, in a CRO and clinical research site perspectives. These two perspectives provided an interesting overview about the scientific needs of different players involved in clinical research.
To conclude, this internship strengthened the knowledge acquired from my academic background, which make me able to face and overcome new challenges in the clinical research area.<br>Este relatório descreve a minha experiência de nove meses enquanto estagiária numa CRO (Eurotrials, Consultores Científicos) e num centro de investigação clínica (Centro Clínico Académico – Braga, Associação).
Este documento começa por analisar o enquadramento europeu da investigação clínica e, posteriormente, a situação portuguesa comparativamente a países similares. De seguida, são descritas as atividades desenvolvidas no âmbito deste estágio, as quais foram divididas em duas fases.
A primeira fase desenrolou-se na Eurotrials, Consultores Científicos, uma CRO especializada em investigação clínica e consultoria científica. As primeiras semanas foram dedicadas ao treino e à formação necessários para o desempenho de tarefas como assistente de estudos clínicos. Estas tarefas incluíram a participação em várias atividades de qualificação, iniciação e monitorização de estudos clínicos, assim como o desenvolvimento e aperfeiçoamento de um sistema de gestão de qualidade.
A segunda vertente decorreu no 2CA-Braga, um centro de investigação clínica localizado no Hospital de Braga. Nesta fase, o estágio focou-se essencialmente na coordenação de estudos clínicos, assim como, em atividades relacionadas com o processo de revisão/negociação de contratos financeiros. Para além disso, tive a oportunidade de participar nas “1as Jornadas de Investigação Clínica e Inovação” organizadas pelo 2CA-Braga.
Globalmente, o estágio traduziu-se numa excelente oportunidade para ganhar conhecimentos e experiência nas tarefas associadas a projetos e serviços no âmbito da implementação e gestão de estudos clínicos, na perspetiva de uma CRO e de um centro de investigação clínica. A integração destas duas perspetivas permitiu-me identificar e confluir necessidades específicas de diferentes players envolvidos na investigação clínica.
Concluindo, este estágio permitiu-me aplicar e aperfeiçoar os conhecimentos adquiridos durante a minha formação académica tornando-me capaz de enfrentar e transpor novos desafios na área da investigação clínica.

Des ligands binaphtyle dérivés contenant des groupes donneurs P"=" O ont été utilisés pour la synthèse rationnelle de composés de coordination à propriétés multiples avec des unités M〖(hfac)〗_3. L'oxyde chiral de bisphosphine L a agi comme un ligand chélate donnant des espèces racémiques monomères de formule [〖{Ln(hfac)_3 L}〗_3] (Ln= Eu,Dy and Yb). Ces complexes ont été caractérisés structurellement et leurs propriétés physiques ont été étudiées à l'état solide. Le composé [〖{Eu(hfac)_3 L}〗_3] présentait une luminescence centrée sur le métal. Différemment, le ligand L n'a pas sensibilisé l'émission de luminescence pour [〖{Dy(hfac)_3 L}〗_3]. Cependant, ce dernier affichait un comportement SMM induit par l’application d’un champ magnétique. Le complexe [〖{Yb(hfac)_3 L}〗_3] était un exemple de field-induced SMM chiral et luminescent. Pour les espèces à base de Dy(III)- et Yb(III)-, l'aimantation s'est relâchée via un processus Raman similaire sous l'effet d'un champ magnétique externe. Tous ces composés sont sublimés lorsqu'ils sont chauffés à pression réduite. Ensuite, la chimie de coordination des ligands bisphosphates énantiopurs dérivés du binaphtyle (S)/(R)-L^n (n=1,3) and (S,S,S)/(R,R,R)-L^n (n=2,4) a été étudié. La réaction de ces ligands avec des quantités équimolaires des précurseurs métalliques [M(hfac)_3 (H_2 O)_2] (M=Y,Eu,Dy and Yb) a donné des polymères de coordination monodimensionnel et énantiopurs. Avec les ligands (S)/(R)-L^n (n=1,3), deux espèces polymorphes différentes pourraient être cristallisées en changeant les conditions de réaction et la nature de l'ion métallique. composés à base d’ion Dy(III) ont manifesté un comportement field-induced SMM et une émission de luminescence. La corrélation magnéto-optique et les résultats des calculs ab initio sont présentés. Le complexe 〖[Dy(hfac)_3 {(S)-L^1}]〗_n a montré de contributions multiples de la relaxation d'aimantation malgré la présence d'un seul centre cristallographique de Dy(III). La solubilisation des polymères de coordination 〖[M(hfac)_3 {(S)/(R)-L^1}]〗_n a provoqué une réorganisation structurale en espèces monomères de formule [M(hfac)_3 {(S)/(R)-L^1}]. Ceci a été démontré par spectroscopie NMR et calculs DFT. De manière similaire à l'état solide, le complexe [Dy(hfac)_3 {(S)-L^1}] a présenté un comportement field-induced SMM à plusieurs contributions. Les processus régissant la relaxation d'aimantation de 〖[Dy(hfac)_3 {(S,S,S)-L^2}]〗_n et 〖[Dy〖(hfac)〗_3 {(S)-L^3 }]〗_n ont été étudiés plus en détail en appliquant une stratégie de dilution magnétique et d'enrichissement isotopique avec (_ ^163)Dy(III) (I=1⁄2) et (_ ^164)Dy(III) (I=0). Malgré la minimisation des interactions dipolaires et l'absence de spin nucléaire, une forte dépendance de l'aimantation par le champ a toujours été observée. Les ligands (S)/(R)-L^n (n=1,3) et (S,S,S)/(R,R,R)-L^n (n=2,4) ont sensibilisé efficacement le luminescence des espèces à base d'Eu(III). Leur nature énantiopure a favorisé l'émission de CPL à la fois en solution et à l'état solide. Enfin, le comportement SMM induit par le champ et l'émission de CPL ont été observés dans le même composé en utilisant des centres Yb(III). L'utilisation du ligand L^5 à base de TTF et des unités chirales Yb〖{(R)/(S)"-" facam}〗_3 a donné les dimères énantiopurs 〖[Yb〖{(R)/(S)"-" facam}〗_3 (L^5)]〗_2. Le fragment TTF conférait une activité redox. L'application d'un champ statique modéré a révélé une lente relaxation de l'aimantation. L'excitation directe des états ILCT de L^5 a sensibilisé la luminescence centrée sur le métal. De plus, des émissions NIR-CPL en solution et à l'état solide ont été détectées. Le complexe 〖[Yb〖{(R)/(S)"-" facam}〗_3 (L^5)]〗_2 était un SMM redox induit par champ chiral affichant une émission CPL. The chiral bisphosphine oxide L acted as a chelate ligand giving monomeric racemic species of formula [〖{Ln(hfac)_3 L}〗_3] (Ln= Eu,Dy and Yb). These complexes were structurally characterized and their physical properties were studied in solid state. The compound [〖{Eu(hfac)_3 L}〗_3] exhibited metal-centred luminescence. Conversely, the ligand L was not able to sensitise luminescence emission for [〖{Dy(hfac)_3 L}〗_3]. However, the latter displayed field-induced SMM behaviour. The complex [〖{Yb(hfac)_3 L}〗_3] was an example of a chiral luminescent field-induced SMM. For both Dy(III)- and Yb(III)-based species, the magnetization relaxed via a similar Raman process under the effect of an external magnetic field. All these compounds sublimated when heated at reduced pressure. Subsequently, the coordination chemistry of the enantiopure binaphthyl-derived bisphosphate ligands (S)/(R)-L^n (n=1,3) and (S,S,S)/(R,R,R)-L^n (n=2,4) was studied. Reaction of these ligands with equimolar quantities of the metal precursors [M(hfac)_3 (H_2 O)_2] (M=Y,Eu,Dy and Yb) yielded enantiopure 1D-coordiantion polymers. With ligands (S)/(R)-L^n (n=1,3), two different polymorphic species could be crystallised by changing reaction conditions and nature of the metal ion. The Dy(III)-based compounds manifested field-induced SMM behaviour and luminescence emission. Magneto-optical correlation and results from ab initio calculations are presented. The complex 〖[Dy(hfac)_3 {(S)-L^1}]〗_n showed multiple contributions of the magnetization relaxation despite the presence of a single crystallographic Dy(III) centre. Solubilization of the coordination polymers 〖[M(hfac)_3 {(S)/(R)-L^1}]〗_n caused a structural reorganization to monomeric species of formula [M(hfac)_3 {(S)/(R)-L^1}]. This was demonstrated by NMR spectroscopy and DFT calculations. Similarly to the solid state, complex [Dy(hfac)_3 {(S)-L^1}] exhibited a multi-contribution field-induced SMM behaviour. The processes governing the magnetization relaxation of 〖[Dy(hfac)_3 {(S,S,S)-L^2}]〗_n and 〖[Dy〖(hfac)〗_3 {(S)-L^3 }]〗_n were further investigated by applying a strategy of magnetic dilution and isotopic enrichment with (_ ^163)Dy(III) (I=1⁄2) and (_ ^164)Dy(III) (I=0). Despite the minimisation of the dipolar interactions and the absence of nuclear spin, a strong field dependence of the magnetization was still observed. The ligands (S)/(R)-L^n (n=1,3) and (S,S,S)/(R,R,R)-L^n (n=2,4) efficiently sensitised the luminescence of the Eu(III)-based species. Their enantiopure nature promoted CPL emission in both solution and solid state. Finally, field-induced SMM behaviour and CPL emission were observed in the same compound by employing Yb(III) centres. The use of the functionalized TTF-based ligand L^5 and chiral Yb〖{(R)/(S)"-" facam}〗_3 units gave the enantiopure pair of dimers 〖[Yb〖{(R)/(S)"-" facam}〗_3 (L^5)]〗_2. The TTF fragment conferred redox activity. The application of a moderate static field revealed slow relaxation of the magnetization. Direct excitation of the ILCT states of L^5 sensitised the metal-centred luminescence. Moreover, both solution and solid state NIR-CPL emission were detected. The complex 〖[Yb〖{(R)/(S)"-" facam}〗_3 (L^5)]〗_2 was a redox chiral filed-induced SMM displaying CPL emission. Together with the Yb(III)-based complexes coordinated by the ligands (S)/(R)-L^n (n=1,3) and (S,S,S)/(R,R,R)-L^n (n=2,4), these are the first documented solid state NIR-CPL emissive examples for molecular complexes

L'acide tout-trans retinoique, principal derive actif de la vitamine a, presente des activites pleiotropiques comme l'inhibition de la proliferation cellulaire, l'induction de la differenciation cellulaire ou la regulation de l'apoptose. Il est donc utilise dans le traitement de certaines affections cutanees ou de certains cancers. L'activite des retinoides est controlee par des recepteurs nucleaires, rars et rxrs (, et ), qui regulent l'activation transcriptionnelle de genes cibles specifiques. Le developpement de nouveaux retinoides a permis d'aboutir a la synthese d'agonistes et d'antagonistes specifiques et selectifs des differentes classes de rars et des rxrs avec des applications therapeutiques mieux definies. La cristallographie du domaine de liaison au ligand de rar a permis de mettre en evidence la conformation et les acides amines de la poche de liaison de l'acide retinoique. Nous avons principalement cible notre etude sur les trois residus de tyrosine du domaine de liaison au ligand de rar et notamment sur le role determinant de la tyrosine 277. Nous avons note une inactivation dose dependante de la liaison rar-#3ht-ra apres traitement du recepteur au tetranitromethane, reactif specifique des tyrosines. Grace au couplage hplc-spectrometrie de masse, nous avons observe une protection par l'acide retinoique contre la nitration de la tyrosine 277. La mutation de ce residu par une alanine engendre une diminution de l'affinite de liaison de l'acide retinoique et de ligands synthetiques et reduit considerablement l'activite transactivatrice du recepteur. D'autres residus d'acides amines de la poche de liaison au ligand de rar ont egalement ete etudies. Les mutants obtenus ont des affinites de liaison et des activites de transactivation differentielles selon le ligand utilise. Ces acides amines semblent donc impliques dans le mecanisme d'activation agoniste et antagoniste de rar.

In this class of proteins, an ATPase component (e.g., ClpA) binds, unfolds, and translocates protein substrates into a protease component (e.g., ClpP) where degradation occurs. This thesis addresses the communication between ClpA and ClpP required to efficiently translocate and degrade protein substrates. Synchrotron hydroxyl radical footprinting is used to show that the ClpA D2 loop, located in the pore of ClpA, is in an "up" conformation when ATP is bound and assumes a "down" conformation when ADP is bound. These results provide the first direct experimental support for a nucleotide-dependent D2 loop conformational change previously proposed to mediate substrate translocation. Synchrotron footprinting also shows that the ClpP N-terminal loops undergo a conformational change, upon the binding of ClpA, from a closed, pore-blocking position, to an open, pore-free position. Complementary kinetic studies show that deletion of the ClpP N-terminus (ClpPAN) accelerates the degradation rate of large peptide substrates 5-15 fold, indicating that opening of the pore is functionally important. Furthermore, unlike ClpAP and wild-type ClpP, ClpPAN shows a distinct slow phase of product formation that is eliminated with the addition of hydroxylamine, suggesting that deletion of the N-terminal loops stabilizes the acyl-enzyme intermediate. Additionally, size-exclusion chromatography and kinetic studies are used to show that ClpP alone can processively degrade a full-length protein substrate in the absence of ClpA, albeit at a 2000-fold slower rate, and furthermore that the size distribution of ClpP-generated peptide products is strikingly similar to the size distribution of ClpAP-generated peptide products.<br>(cont.)Both distributions contain peaks at integral multiples of 7-8 amino acids, consistent with a mechanism in which ClpP controls product sizes by alternating between translocation in steps of 7-8 amino acids and proteolysis. Les systèmes multi-agents permettent la simulation de phénomènes complexes difficilement descriptibles de manière analytique. Cette approche repose souvent sur la coordination d'agents dont l'ensemble des actions et interactions entraînent l'émergence du phénomène à simuler. Lorsqu'il s'agit de reproduire le comportement humain, il faut parfois composer avec une coordination compétitive et être capable de reproduire des comportements anticipatifs. Dans cette thèse, nous nous sommes intéressés à la problématique de reconnaissance et d'anticipation du contexte de coordination. Nos travaux se basent sur une approche de l'anticipation proposée par Rosen: « l'anticipation préventive » qui consiste à adapter son comportement courant de manière à éviter un certain nombre d'états indésirés. Nous proposons une formalisation de ce concept basée sur la manipulation et le traitement de réseaux de contraintes. Le modèle décisionnel sous-jacent utilise des techniques de propagation de contraintes permettant à chaque agent d'inférer les effets d'une action et de détecter ainsi un certain nombre d'états indésirés. L'algorithme proposé est générique et permet de filtrer, en amont de la coordination, toutes actions conduisant à l'un des états non désirés du système. Implémentée dans le cadre d'une simulation de trafic développée à l'INRETS: ARCHISIM, notre proposition permet d'introduire des comportements opportunistes plus réalistes sans risque d'apparition d'interblocage au centre des carrefours. This approach is often based on the coordination of agents whose actions and interactions involve the emergence of the phenomenon to be simulated. When the aim is to mimic the human behaviour, it is sometimes necessary to use a competitive coordination and to be able to reproduce anticipatory behaviours. This thesis addresses the issue of recognition and anticipation of coordination context. Our work is based on an approach of the anticipation suggested by Rosen: « preventive anticipation » which consists in adapting its actual behaviour to avoid some undesired states. We propose a formalization of this concept based on constraints networks processing. The underlying decisional model uses constraints propagation techniques which allow each agent to gather the effects of an action and thus to detect some undesired states. The proposed algorithm is generic and filters out, before any coordination, all actions leading to one of the non-desired states of the system. Implemented within the framework of a simulation of traffic developed at INRETS and called ARCHISIM, our proposal enable to introduce opportunistic behaviours without any risk of having deadlock in the center of the intersection. The various evaluations validate the emergence of traffic phenomena and confirm the relevance of the approach

Biomimetic catalysis is an important research field, as a better understanding of nature´s powerful toolbox for the conversion of molecules can lead to technological progress. [FeFe]-hydrogenases are very efficient catalysts for hydrogen production. These enzymes play a crucial role in the metabolism of green algae and certain cyanobacteria. Their active site consists of a diiron complex that is embedded in an interactive protein matrix. In this thesis, two pathways for mimicking the outer coordination sphere effects resulting from the protein matrix are explored. The first is the construction of model complexes containing phosphine ligands that are coordinated to the iron center as well as covalently linked to the bridging ligand of the complex. The effect of such linkers is an increased energy barrier for the rotation of the Fe(CO2)(PL3)-subunit, which potentially could stabilize a terminal hydride that is an important intermediate in the proton reduction cycle. The second pathway follows the incorporation of [FeFe]-hydrogenase active site model complexes into metal-organic frameworks (MOFs). Resulting MOF-catalysts exhibit increased photocatalytic activity compared to homogenous references due to a stabilizing effect on catalytic intermediates by the surrounding framework. Catalyst accessibility within the MOF and the influence of the framework on chemical reactivity are examined in the work presented. Furthermore, an initial step towards application of MOF-catalysts in a device was made by interfacing them with electrodes. The work of this thesis highlights strategies for the improvement of biomimetic model catalysts and the knowledge gained can be transferred to other systems mimicking the function of enzymes.

Les clusters octaédriques de molybdène (Mo) et de rhénium (Re) présentent un intérêt croissant pour des applications dans plusieurs domaines tels que la santé ou l’énergie. Ceci en raison de la richesse de leurs propriétés physico-chimiques et structurales (ex. : absorption/émission de lumière, oxydation réversibles et modulables). L’introduction d’un second métal dans la composition du cluster constitue un excellent moyen pour i) modifier et contrôler les propriétés physicochimiques du cluster et ii) générer de nouvelles propriétés. Avant cette étude, seuls quelques travaux consacrés principalement à la synthèse et aux études structurales de composés hétérométalliques octaédriques étaient rapportées dans la littérature. La principale difficulté qui a limité le développement de ce champ de recherche était la formation simultanée et la co-cristallisation de plusieurs clusters hétérométalliques avec des rapports Mo/Re différents ou bien pour un même rapport, de plusieurs isomères. Ce problème a été résolu dans ce travail en utilisant la séparation post-synthétique basée sur les différences des comportements redox et de solubilité des chalcogénures à clusters hétérométalliques. Cela nous a permis d’étudier individuellement la structure, les caractéristiques spectroscopiques et les propriétés redox de clusters bien définis de type [Re₆₋ₓMoₓSe₈(CN)6]ⁿ⁻(x = 1, 2, 3). Ces travaux fournissent une série de clusters Mo/Re capables de plusieurs transitions électrochimiques dans une fenêtre étroite de potentiels, accompagnés d'une modification de leur spectre optique. Ils pourront par exemple être utilisés pour la réalisation de capteurs<br>Octahedral cluster compounds of molybdenum and rhenium are of growing interest due to the set of promising properties caused by intrinsic characteristics of the cluster core, such as photoluminescence, reversible redox transitions preserving the geometry of the cluster core, and magnetism. A change in the composition of the metal core with the formation of heterometallic cluster can serve as an excellent tool for controlled modification of the physicochemical properties of cluster and introducing new features non-typical for homometallic analogues. Prior to the start of this study, only a few works devoted mainly to the synthesis and study of the structure of octahedral heterometallic compounds were presented in the literature. The main difficulty hampering the further development of this field was the formation and co-crystallization of several heterometallic clusters of different metal ratio in the same reaction conditions. This problem was successfully solved using the post-synthetic separation based on the deviations in the redox character and salts solubility of Re/Mo chalcogenide clusters of different metal ratio. It allowed us the individual study of the structure, spectroscopic characteristics, and redox properties of well-defined heterometallic cluster complexes [Re₆₋ₓMoₓSe₈(CN)6]ⁿ⁻ (x = 1, 2, 3). The present research provided a series of Re / Mo clusters capable of several electrochemical transitions in a narrow potential window, accompanied by a change in the optical spectrum of the cluster complex. Ce travail concerne la gestion et la coordination d'un ensemble d'unités de production réparties en différents sites et entre lesquelles s'échangent des flux de produits. Le problème consiste à trouver comment répartir dans le temps les productions correspondant à des commandes de produits entre les différentes unités de production, certains produits ou composants pouvant être produits dans plusieurs de ces unités de production. Cette répartition est faite en tenant compte des capacités de production des différents sites, avec des objectifs de minimisation de critères globaux. Ce problème est modélisé comme un problème de satisfaction de contraintes (CSP). Un langage de programmation par contraintes mettant en œuvre des méthodes nouvelles issues des recherches dans le domaine des CSP est utilisé pour la résolution. Différentes stratégies de recherche de solutions sont proposées et classées. L’ajout de périodes de taille variables permet de limiter la combinatoire du problème et de respecter la précision des données (commandes à plus ou moins long terme). La prise en compte au niveau planification de certaines contraintes du niveau ordonnancement est effectuée grâce à une approche intégrée planification et ordonnancement multi-site. L’intégration des résultats et du logiciel issus de ce travail a été effectuée sur un logiciel a vocation industrielle dans le cadre d'un projet européen.

This essay describes coordination of the work environment on construction sites where many entrepreneurs share workplace. As employer, the entrepreneur has main responsibility for the workers safety on the construction site, the project supervisor is responsible for the work environment, and to coordinate the work environment a coordinator of the construction work environment is appointed. The purpose of this essay is to describe how the work environment is ensured on a shared workplace. In order to answer the essays questions I’ve used a juridical method to interpret the law. Current law and regulations has been studied, as well as the industry's rules and regulations. The juridical method is complemented by social science methodology by the use of an empirical survey consisting of interviews with a coordinator of the construction work environment, and subcontractors on a construction site. The questions of this essay concerns the project supervisor’s and the entrepreneur’s safety responsibilities of the working environment, and how they ensure their joint work environment. And also the requirements imposed on the entrepreneur’s coordination responsibilities, and if the working methods differ when the workers are posted. A construction site is a complicated workplace with many companies working side by side. Requirements for cooperation and consultation are the responsibility of all employers who have employees engaged on the same construction site. Entrepreneurs who post workers in Sweden have the same employer responsibilities as other entrepreneurs, and shall ensure the work environment of the posted worker.<br>Denna uppsats handlar om samordningen av arbetsmiljön på byggarbetsplatser där flera entreprenörer delar arbetsplats. Som arbetsgivare har en entreprenör huvudansvaret för de anställdas arbetsmiljö, på byggarbetsplatsen är det byggherren som ansvarar för arbetsmiljön, och för att samordna parternas arbetsmiljö utses en byggarbetsmiljösamordnare. Studiens syfte är att deskriptivt redogöra för hur arbetsmiljö säkerställs på en gemensam arbetsplats. För att besvara uppsatsens frågeställningar har juridisk metod använts genom att tolka gällande rätt. Förutom lag och förordningar har även böcker i ämnet använts, samt branschens regler och bestämmelser. Den juridiska metoden kompletteras med samhällsvetenskaplig metod genom att vidga studien med empiriskt material i form av intervjuer med en byggarbetsmiljösamordnare, samt ett urval underentreprenörer på en byggarbetsplats. Uppsatsens frågeställningar berör förhållandet mellan byggherrens och entreprenörers arbetsmiljöansvar och hur de arbetar för att säkerställa den gemensamma arbetsmiljön. This thesis is concerned with the study of biomedical scientific research work that is intensely distributed, i.e. socially distributed across multiple institutions, sites, and disciplines. Specifically, this PhD probes the ways in which scientists co-operating on multi-sited crossdisciplinary projects, design, use and maintain information-based resources to conduct and coordinate their experimental activities. The research focuses on the roles of information artefacts, i.e. the tools, media and devices used to store, track, display, and retrieve information in paper or electronic format, in helping the scientists integrate their activities to achieve concerted action. To examine how scientists in globally distributed settings organise and co-ordinate their scientific work using information artefacts, a multi-method multi-sited study informed by different ethnographic perspectives was conducted focused on a large European crossdisciplinary translational research project in nanodiagnostics. Situated interviews with project scientists, participant observations and participatory learning exercises were designed and deployed. From the data analysis, several abstractions were developed to represent how the joined utilisations of key information artefacts support the co-ordination of experimental activities. Subsequently, a framework was developed to highlight key interactional strategies that need to be managed by experimenters when using artefacts to organise their work cooperatively. This framework was then used as a guiding device to identify innovative ways to design future digital interactive systems to support the co-ordination of intensely distributed scientific work. From this study, several key findings came to light. We identify the role of the experimental protocol acts as a co-ordinative map that is co-designed dynamically to disseminate various instantiations of experimental executions across sites. We have also shed light on the ways the protocol, the lab book and the material log are used jointly to support the articulation of scientific work. The protocol and the lab book are used both locally and across co-operating sites to support four repeatability and reproducibility levels that are key to experimental validation. The use of the local protocol / lab book dyads at each site is further integrated with that of a centralised material log artefact to enable a system of exchange of scientific content (e.g. experimental processes, intermediate results and observations) and experimental materials (both physical materials and key information). We have found that this integration into a co-ordinative cluster supports awareness and the articulation of experimental activities both locally and across remote labs. From this understanding, we have derived several sensitising tensions to frame the strategies that scientific practitioners need to manage when designing their multi-sited experimental work and technologists should consider when designing systems to support them: (1) formalisation / flexibility; (2) articulability / local appropriateness; (3) scrutiny / tinkering; (4) accountability / applicability; (5) traceability / improvisation and (6) lastingness / immediacy. Lastly, based on these tensions, we have suggested a number of implications for the design of interactive information artefacts that can help manage both local and multi-sited co-ordination in intensely distributed scientific projects.

Les hydrazines monosubstituees rhnnh#2 sont oxydees par la pghs en diazene rn=nh avec formation de la pghsfe(ii) et du complexe pghsfe#i#i-diazene. L'oxydation de ce complexe conduit a un nouveau complexe a liaison fe-c de l'enzyme: pghsfe#i#i#i-r. La formation de ces complexes montre que la pghs possede un site actif ouvert. Les phenylhydrazones constituent une nouvelle classe de substrats suicide de la fonction dioxygenase de la pghs. Les -azohydroperoxydes correspondants sont formes, puis transformes en -azoalcools par la fonction peroxydase de la pghs. L'oxydation de ces -azoalcools par la pghsfe(iii) en cetone ou aldehyde avec la formation de n#2, de radicaux ph#. Et de la pghsfe(ii), conduit a un complexe pghsfei#i#i-ph. Les inhibitions de la pghs par les hydrazines monosubstituees et par les phenylhydrazones precedemment decrites sont vraisemblablement dues a la formation des radicaux r#. Et des complexes pghsfe#i#i#i-r. Le spectre de resonance raman de la pghs contenant de l'heme natif est caracteristique d'une espece dont le fer est hexacoordine et spin fort. Une structure pghsfe#i#i#i-aquo est proposee pour l'enzyme native. Le spectre de resonance raman de l'enzyme reconstituee montre la presence d'une nouvelle espece dont le fer est hexacoordine et spin faible et dont la structure pourrait etre: pghsfe#i#i-histidine distale. La proximite de l'histidine distale est demontree par la formation d'une liaison hydrogene entre l'histidine distale et le groupement carbonyle en milieu acide. Les spécificités du secteur de la construction engendrent un contexte de travail coopératif que nous pouvons qualifier d'incertain par nature. L'activité de chantier se caractérise par un mode de production in situ. Dès lors, nombreux sont les dysfonctionnements qui peuvent apparaître. Nous citerons, par exemple, les problèmes liés à la nature du sol, aux intempéries ou encore ceux qui sont propres aux interactions entre les intervenants (ex. fourniture en matériaux, interfaces entre les corps de métier...). Aussi, la coordination repose sur un mélange subtil entre interactions implicites et explicites, où l'organisation prend de multiples configurations (hiérarchique, adhocratique ou transversale), et où la qualité du processus collectif repose sur l'autonomie et le sens des responsabilités de chacun des intervenants. Nous faisons l'hypothèse qu'un tel contexte est largement fondé sur la notion de confiance, car celle-ci a la capacité de réduire la perception du risque et de permettre l'action dans un environnement marqué par de nombreuses incertitudes. En conséquence, nous suggérons un rapprochement entre les outils d'assistance à la coordination et la notion de confiance, et nous proposons une nouvelle approche du pilotage de l'activité collective à partir de la représentation de la confiance. Ce travail de doctorat se structure autour de la notion de «confiance dans le bon déroulement de l'activité». Nous suggérons que celle-ci est dépendante de chacune des dimensions de l'activité collective : sa progression, les acteurs chargés de son exécution, les ouvrages en résultant (et leur difficulté de mise en œuvre), ainsi que les documents nécessaires à sa réalisation. Aussi, notre méthode consiste d'abord en l'identification des divers critères de confiance, consolidés par une étude de terrain. Puis, nous établissons un modèle mathématique destiné à évaluer cette confiance à partir des informations issues d'un contexte de coopération. Sur base de ces éléments, s'en suit la proposition d'un prototype dénommé Bat'iTrust, reposant sur une architecture logicielle multi-vues et orientée services. Pour l'utilisateur, la navigation au sein de ce prototype est guidée par un tableau de bord centré sur le concept de confiance. Enfin, la validation de cette proposition repose sur des enquêtes et une phase d'expérimentation qui nous ont permis de confronter nos résultats à des sujets expérimentaux représentatifs du domaine.

Den här studien undersöker yrkesverksamma teckenspråkstolkars syn på likheter och skillnader mellan på-platstolkning och distanstolkning. Undersökningen behandlar också samordning och teckenspråkstolkars uppfattning av begreppet. Materialet består av videoinspelade intervjuer som sedan analyserats utifrån uppsatsens frågeställningar. Teckenspråket är ett visuellt språk och innehåller på så vis grammatiska delar som inte återfinns i talat språk, exempelvis teckenrummet framför den som tecknar. I denna studie undersöks vad som förändras i tolkningen när det sker via en skärm och på så vis blir tvådimensionellt i stället för tredimensionellt för mottagaren. Även påverkan på tolkningen av att tolkanvändarna inte befinner sig i samma fysiska rum som tolken tas upp i undersökningen. Resultatet visar att distanstolkning begränsar teckenspråkets referenssystem i det tredimensionella rummet, men även användandet av kroppsspråket i ett tolkmedierat samtal. Vid på-platstolkning kan tolken vrida sig mot en tolkanvändare för att diskret få kontakt med denne, men vid tolkning på distans fungerar inte denna kroppsliga orientering som resurs på samma sätt med tanke på att skärmen är tvådimensionell och att tolkanvändaren inte befinner sig i samma rum som tolken.<br>This study examines professional sign language interpreters view on similarities and differences between interpreting on site and remote interpreting. The study also examines coordination and the understanding of the word by sign language interpreters. The material for this study consists of videorecorded interviews which have been analysed based on the study’s research questions. Sign language is a visual language and therefore has grammatical differences to spoken language, for example the signing space in front of the person signing. This study examines what parts of interpreting that change when it is performed remotely and therefore becomes two- instead of three-dimensional for the recipient. The study also investigates how the interpreting is affected when the interpreter users are not in the same physical room as the interpreter. The results show that remote interpreting limits the reference system as well as how bodily orientation is used as a resource in an interpreted conversation. The interpreter can turn towards an interpreter user during on-site interpreting in order to get their attention discreetly, however, in remote interpreting this body turn does not work in the same way, seeing as the screen is two dimensional and that the interpreter user is not in the same physical room as the interpreter.

response to its market growing and competitiveness. However this evolving path has faced
many obstacles such as cultural barriers and the lack of efforts to achieve quality at the
Brazilian construction site. As one of the causes of this facts the lack of integration between
construction and the designing process generates waste, pathologies, work redone and
improvised changes originated by incompatible projects which were taken by incapacitated
personnel at an inconvenient moment. At the same time, the great amount of information
generated on the designing or construction phases is lost due to the lack of an effective
coordination of these activities. From this scenario Souza (2001), has developed two
management methods in her doctorate thesis named Preparation for the Execution of
Buildings (known as PEO) and the Pro-active Coordination (known as CPA), these methods
are based on the application of the processes adopted by the French construction industry on
Brazilian construction sites. These methodologies aim to establish the integration of the
project phases by a set of actions and activities implemented by a system of meetings where
the main objective is to discuss the projects and the planning of the construction site seeking
to obtain a better quality and better outcome on the productive process. These methodologies
aim to bring expressive improvements on the productivity, as well as on the lowering of
waste, on the settlement of goals, on planning actions in accordance with all the agents
involved and mainly on the reduction of costs allied to higher quality standards. Thus is an
objective of this research to implement the Preparation on the Execution of Buildings (PEO)
seeking to understand the management of the designing process and its interface with the
construction site. The methodology applied on the development of the analysis is classified as
qualitative and it will be carried by the practical application of the research and the
evaluation of its results. Thus, the study was divided into two stages, one of them intended for
a pilot study of PEO in Company A, and the other to the implementation of PEO on the
company B. This way, the method was developed from a system meetings entitled structured
in three different stages: the release of PEO; the development of PEO and the completion of
the intensive phase of PEO, which were performed over a period from two to three months.
The data analysis was structured upon five main aspects which were evaluated during the
implementation of the method on the studied companies. These aspects were: the actions
taken during the PEO phase, the development of the actions and the participation of the
agents; the internal structure of the companies analyzed; The management and design
coordination and the contract form of the contractors, architects and engineers. The results
obtained demonstrated the PEO method's effectiveness and viability while a booster on the
improvement of project management. Therefore, it can be concluded that the PEO is feasible
to be applied in building construction companies, since this method can contribute to the
integration of the design phases and construction, as well as to the improvement of the
communication between the agents involved in the building construction field.<br>Nos últimos 20 anos, o setor da construção civil no Brasil evoluiu significativamente em
resposta ao crescimento da competitividade no mercado. Entretanto, nesse processo de
evolução ainda têm sido encontradas barreiras culturais e organizacionais, e, sobretudo,
dificuldades nos esforços à qualidade nos canteiros de obras brasileiros. Aliada a este quadro,
a falta de integração entre as atividades de construir e projetar provoca uma série de
desperdícios, patologias, retrabalhos e alterações improvisadas decorrentes de projetos não
compatibilizados, ora por ausência de detalhes nos projetos, ora por decisões tomadas por
pessoas não capacitadas em momentos inadequados. A partir dessa realidade, Souza (2001)
desenvolveu dois métodos de gestão nomeados Preparação da Execução de Obras (PEO) e
Coordenação Pró-Ativa (CPA) em sua tese de doutoramento, por meio da transposição desses
métodos adotados na gestão da interface projeto-execução de obras na França para a realidade
dos canteiros de obras brasileiros. Esses métodos visam à integração das etapas de projeto e
obra a partir de um conjunto de ações e atividades realizadas por meio de reuniões
sistematizadas voltadas à discussão do projeto, ao planejamento e à organização do futuro
canteiro de obras, a fim de obter melhor qualidade e desempenho no processo produtivo.
Dessa forma, este trabalho tem como objetivo geral implementar o método da preparação da
execução de obras (PEO) a fim de avaliar a interface entre a gestão do processo de projeto e a
execução do canteiro de obras. A metodologia adotada para o desenvolvimento deste trabalho
é classificada como qualitativa, a qual foi realizada por meio de uma pesquisa-ação, e dividida
em duas etapas diferentes, sendo uma destinada ao estudo piloto da PEO na empresa A, e a
outra à implementação da PEO na empresa B. Deste modo, a aplicação do método foi
desenvolvida a partir de um sistema de reuniões em três fases intituladas: lançamento da PEO;
desenvolvimento da PEO e finalização da fase intensiva de PEO, as quais foram realizadas em
um período entre dois e três meses. A análise de dados foi estruturada a partir de cinco
aspectos principais, os quais foram avaliados durante a aplicação do método nas empresas
estudadas, sendo eles: quanto às ações desenvolvidas na fase de PEO, quanto ao
desenvolvimento das reuniões e a participação dos agentes; quanto à estrutura organizacional
e departamental das empresas; quanto à gestão e a coordenação de projetos; e quanto ao
formato de contratação dos projetistas e subempreiteiros. Os resultados obtidos com a
realização deste trabalho comprovaram a eficácia do método da PEO e a sua viabilidade
enquanto potencializador na melhoria da gestão da interface projeto-execução de obras.
Portanto, pode-se concluir que a PEO é viável de ser aplicada em empresas de construção e
incorporação de edifícios, uma vez que este método pode contribuir para a integração das
etapas de projeto e execução, bem como na melhoria da comunicação e na interação entre os
agentes envolvidos na produção de edifícios.

The work describes the study of the main stages of technological, financial and construction schedule, itemized budget and technological prescription for performance building envelope. A substantial part of the work is to perform a technological prescription ceiling cladding structure above 1.NP, which is supplemented by technical-economic balance for the implementation of this ceiling construction, along with formwork drawings describes the optimal design of monolithic ceiling structure above 1.NP. Suitable design of the implementation process of the ceiling structure is achieved by reducing the cost of implementation of nearly 10% and shorten the length of the lease formwork system of almost 34% compared to the original assumption.

Cette thèse s’intéresse à ce nouvel impératif de fabrication et à ses effets sur la réorganisation de l’acte de construire, la reconfiguration des pratiques de coordination et de conception collective entre les professionnels de la construction ainsi qu’à la redéfinition des frontières théoriques du projet architectural. Ce travail s’appuie sur une enquête ethnographique menée au sein d’un Grand Projet d’́Ecole reconnu pour sa forte implication dans la démarche BIM. C’est au cœur de la phase chantier de ce projet, lors des réunions de synthèse d’exécution, entre les acteurs et les objets, que s’ancre cette investigation. L’analyse du matériel empirique constitue d’observations et d’entretiens menés avec les acteurs du projet nous permet de comprendre les différentes réorganisations pratiques liées ̀a la fabrication collective d’une maquette numérique. Cette recherche nous permet aussi d’esquisser les perspectives de nouvelles pratiques numériques de projet et à préfigurer une ́évolution théorique des nouvelles limites du projet architectural<br>The BIM approach has become a necessity for the building sector involving the realization of a new technical object : the digital model. The research is focused on the new manufacturing of this object and its praticals effects on the way to build, the reconfiguration of coordination practices and collective design between construction professionals. We also attempted to draw a new definition of theoretical boundaries of the architectural project. Our work is based on an ethnographic investigation carried out within a Major School Project recognized for its strong involvement in the BIM process. The investigation takes place on the heart of the construction phase of this project, during the execution summary meetings, between actors and objects. The analysis of this material mainly composed by observations and interviews carried out with the project stakeholders allows us to understand the different practical reorganizations linked to the collective production of this new digital model. On the given topic are processed and all necessary supporting documents that enhances the clarity and orientation in matters large building for the preparation of the construction process. Given the scale, complexity and location of the building is structurally location technology project addresses the only earthworks and construction site of the main building object SO-01 only. Furthermore, nine small objects that are closely tied this main object within the premises of a residential building. Construction hors site (CHS), méthodes de construction modernes (MCM), fabrication hors site (FHS) et production hors site (PHS) sont tous des termes cités dans la littérature comme étant des solutions possibles pour résoudre les problèmes des délais, de qualité et de coût associés aux méthodes de construction traditionnelles. La mise en œuvre réussie de ces méthodes dans le cadre d'un projet de construction nécessite une analyse systématique et une prise de décision anticipée fondée sur des facteurs spécifiques au projet. Il n'existe pas actuellement une approche systémique efficace capable de répondre à l'évolution des besoins, de faire face à la complexité croissante des projets de construction et de prendre en compte les innovations, les technologies et les réglementations récemment mises au point. À cet égard, ces sujets ont été abordés dans la présente thèse de doctorat. L'objectif de ce travail de recherche est le développement d'un outil d'aide à la décision pour le choix des modes constructifs. Nous proposons un modèle de prise de décision multi-niveaux. Le premier niveau concerne l’étude de faisabilité et évalue l'applicabilité de la construction hors site. Le deuxième niveau propose une approche intégrée de prise de décision qui combine la méthode AHP (Analytic Hierarchy Process) et la méthode PROMETHEE. La combinaison des deux méthodes permet une évaluation minutieuse des différents modes constructifs et des scénarios pour un projet de construction donné. La méthodologie de recherche adoptée est une approche mixte, combinant des méthodes de collecte de données quantitatives et qualitatives, incluant trois études de cas, dix entretiens semi-structurés, des questionnaires avec différents acteurs de la construction, et un focus group avec neuf ingénieurs en construction. L'étude identifie d'abord les avantages et les limites de la construction hors site. Elle met ensuite en exergue les facteurs qui influencent les décisions relatives à l'utilisation de la construction hors site. Ensuite, elle présente et décrit un outil d’aide à la décision informatisé pensé pour aider les équipes projet dans leur processus décisionnel. Enfin, l'outil d'aide à la décision proposé a été testé auprès des professionnels de la construction sous la forme de deux études de cas. Ces dernières constituent une validation du modèle et l’outil informatisé, ainsi qu’une preuve de leur utilité et de son efficacité d’usage dans le processus décisionnel en construction<br>Offsite Construction (OSC), Modern Methods of Construction (MMC), Offsite Manufacturing (OSM), and Offsite Production (OSP) are all terms cited as being possible solutions for addressing time, quality and cost concerns associated with traditional construction method. Successful implementation of these methods on a particular project requires systematic analysis and early decision making based on specific factors of the project. There is a lack of an efficient systematic approach that is able to match the changing needs, deal with the growing complexity of building projects and take into consideration recently developed innovations, technologies and regulations. In this respect these issues have been addressed in this research study. The objective of this dissertation is the development of an early- phase decision tool to support the general contractor in selecting construction methods. We propose a multi-level decision framework. The first level conducts a feasibility study and evaluates the applicability of offsite construction. The second level proposes an integrated approach for the decision-making problem that combines the Analytical Hierarchy Process (AHP) and the Preference Ranking Organization Method for Enrichment Evaluations (PROMETHEE). The combination of both approaches enables a careful evaluation of different construction methods and scenarios for the same project. The research methodology adopted is a mixed approach, combining quantitative and qualitative data collection methods, including three case studies, 10 semi-structured interviews, questionnaires with different construction stakeholders and focus groups with 9 construction engineers. The study first identifies the advantages and limitation of offsite construction. Second, it highlights the factors influencing decisions on the use of prework. Then, it presents and describes a computerized tool to aid project teams in the decision-making process. Finally, the decision-making tool was tested and with construction practitioners on three different case studies. They validate that the developed framework and tool are useful and effective in the decision-making process

The master's thesis is elaborated on the basis of the project of the Sport and Health Center Olomouc. The project consists of two main buildings – the object of NH Hotel and the Sports Center Omega. The hotel also includes a conference center with three conference halls and covered parking. The object of Omega Sports Center is composed of sports halls for squash, badminton and tennis, along with changing rooms, wellness area and operational background of the object itself. The sports center also incorporates outdoor tennis courts and an underground tunnel connecting the Omega center and the NH Hotel. This thesis deals with the civil technological project of the building site and involves mainly the technical report of civil technological project, the study of realization of the main technological stages of the building object, the project of organization of construction, time schedule of aforementioned two main buildings, the technological standard with test and execution plans for the execution of underground construction by the impermeable concrete technology and for execution of variant solution of tennis hall roofing. Further, a comparison of the alternative roofing technology with current used roofing technology is performed, including the drawing documentation of new roofing alternative.
