These changes often lead to the formation of aberrant transcripts that can induce nonsense-mediated decay, and a subsequent lack of functional protein. For some genetic disorders, including inherited retinal diseases (IRDs), reproducing splicing dynamics in vitro is a challenge due to the specific environment provided by, e.g. the retinal tissue, cells of which cannot be easily obtained and/or cultured. Here, we describe how to engineer splicing vectors, validate the reliability and reproducibility of alternative cellular systems, assess pre-mRNA splicing defects involved in IRD, and finally correct those by using antisense oligonucleotide-based strategies.

AbstractIn this chapter, we present the various categories of Face Manipulation and their use within different remote operational systems. We then use the example of remote identity document onboarding systems to illustrate how each category can be used in practice to compromise such a system. After a definition of the different Face Manipulation categories and the common algorithms used to produce them, we go through the various manipulation detection algorithms and common image and video forgery datasets. We then introduce some known counter-forensics methods that can be used by an attacker to avoid detection. Knowing the detection methods and the counter-forensic, we present how we can build up a safer system by using the correct methods at the correct time. But also how knowledge about the tampering process could be used to design the user experience to make the systems harder to compromise. We complete this review by the standardisation effort and legal aspect on the matter. And we conclude by discussing the remaining challenges and perspectives for better use of nowadays detection methods in practical usage.

AbstractWe show how to define forward- and reverse-mode automatic differentiation source-code transformations or on a standard higher-order functional language. The transformations generate purely functional code, and they are principled in the sense that their definition arises from a categorical universal property. We give a semantic proof of correctness of the transformations. In their most elegant formulation, the transformations generate code with linear types. However, we demonstrate how the transformations can be implemented in a standard functional language without sacrificing correctness. To do so, we make use of abstract data types to represent the required linear types, e.g. through the use of a basic module system.

Overconfidence leads to risky behavior, including when people are around guns. Does overconfidence also shape attitudes about gun ownership and use? We evaluate this possibility by conducting nationally representative surveys in six countries in the Americas, including the United States. Results show that overconfident individuals are more willing to accept the use of guns and more likely to declare their willingness to use guns. This work shows that some demographic groups end up underrepresented in household inflation expectations surveys due to item nonresponse. Nonresponses can lead to misleading inferences about inflation bias, and this paper shows how to correct for such nonresponse bias in average inflation expectations. These findings have important implications for how central banks use household inflation expectations measures, and how policymakers communicate with the population. One main implication is that policy outreach can be improved with more targeted communications.
