Although mycoplasmas possess a very limited genome, little is known about their virulence mechanisms and methods of persistence in the host. Examination of a wide range of mycoplasma species found considerable variation in their ability to form a biofilm. Mycoplasma putrefaciens, M. cottewii, M. yeatsii, M. agalactiae and M. bovis produced prolific biofilms. Conversely, the highly pathogenic mycoplasma and causative agent of contagious bovine pleuropneumonia, Mycoplasma mycoides subsp. mycoides SC, was unable to produce a biofilm. Biofilms were found to be considerably more resistant to stress, including heat and desiccation, than planktonic cells. A link between the biofilm phenotype and genotype as determined by molecular typing was found for M. bovis. Analysis of biofilms using fluorescent staining combined with confocal microscopy demonstrated that mycoplasma biofilms formed a highly differentiated structure with stacks and channels. Biofilm formation may indicate that mycoplasmas are capable of surviving in the environment.

Ligand-mediated approaches to gene transfer offer an alternative to viral vectors for both in vivo and in vitro applications. Although a significant percentage of the plasmid-based DNA complex is lost to lysosomal degradation following receptor-mediated endocytosis, simultaneous infection with adenovirus has been shown to increase the level of transgene expression [Curiel, Agarwal, Wagner and Cotten (1991) Proc. Natl. Acad. Sci. U.S.A. 88, 8850-8854; Wagner, Zatloukal, Cotten, Kirlappos, Mechtler, Curiel and Birnstiel (1992) Proc. Natl. Acad. Sci. U.S.A. 89, 6099-6103]. In this study we describe an adenovirus-based ligand complex where the plasmid DNA, polycation-ligand conjugate and adenovirus are contained within a single particle structure. At the core of the transfection particle is a replication-defective recombinant adenovirus encoding a cDNA minigene for human placenta alkaline phosphatase that was chemically modified with poly(L-lysine) (Ad-pLys). Electron microscopy of an adenovirus-based ligand complex formed by successively adding plasmid DNA and an asialo-orosomucoid-poly(L-lysine) conjugate to Ad-pLys revealed structures that appeared as intact viral particles coated with a dense biomolecular layer. Adenovirus-based ligand complexes containing either a luciferase or beta-galactosidase reporter plasmid were shown to efficiently deliver the plasmid transgene to cells that express the hepatic asialoglycoprotein receptor. Furthermore, the poly(L-lysine) modification greatly reduced the infectivity potential of the virus without causing a concomitant loss of augmented gene transfer. As an alternative to infectious virions, incomplete products of viral assembly were also considered as a source for endosomalytic activity. However, these defective virions were unable to significantly enhance plasmid transgene delivery.

One-hundred-five (105) clinical isolates of mycoplasma from caprine origin and one isolate from ovine were surveyed for plasmids, which were present in thirty-three (31%) of them. These mycoplasmas originated from 13 herds. Ten of them were symptomatic for mycoplasmal disease (mastitis, polyarthritis, septicemia) and three herds were asymptomatic, i.e., clinically normal. Twenty-eight isolates were Mycoplasma mycoides subspecies mycoides LC (large colony or caprine biotype), four were Mycoplasma capricolum subsp. capricolum and one was Mycoplasma cottewii. The isolated plasmids were linearized by EcoRI, EcoRV, EcoRI and EcoRV or BamHI and EcoRV, and were of five sizes (1.1, 1.6, 1.7, 1.8, and 1.9 Kbp). Based on restriction enzyme digestion and size of the linearized supercoiled extrachromosomal DNA, five plasmid types were recovered (p1II, p2III, p2V, p3I, and p4IV). The small size of these DNA elements probably exclude replicative forms of DNA virus, which are equal or larger than 8.0 Kbp.

Kegiatan kreativitas seni warna anak usia dini ini dapat menjadi solusinya. Tujuan penelitian ini adalah agar dapat mengembangkan aspek kreativitas anak usia dini di era pandemi covid 19 ini. Metode yang digunakan adalah penelitian kualitatif deskripstif yang menganalisis persistiwa dari suatu tindakan yang dilakukan. Sampel yang digunakan dalam pengimplementasian kreativitas seni warna ini dapat berupa anak usia 3-5 tahun yang berada disekitar kompleks perumahan Sumber Mukti Indah Rembang. Adapun orangtua anak dapat dijadikan acuan dalam proses kegiatan kreativitas seni warna. Hasil penelitian yang didapat adalah adanya antusias anak dalam melakukan aktivitas kreativitas seni warna dengan menggunakan cat air. Anak merasa senang dan dapat menghasilkan hasil karya yang estetik. Adapun kegiatan yang diterapkan yaitu melukis menggunakan cotten bad, fingger painting, serta bercerita dengan menggunakan hasil karya masing-masing. Tujuan dari adanya kegiatan ini adalah agar dapat menumbuhkan jiwa kreativitas seni anak, emosional, bahasa, dan motorik anak. Abstract The Covid 19 that has hit the world has become the main problem of the emergence of online learning held at home. In this situation children can feel boredom due to less creative learning factors in its implementation. This early childhood color art creativity activity can be a solution. The purpose of this research is to develop the creative aspects of early childhood in the era of the Covid 19 pandemic. The method used is descriptive qualitative research that analyzes the events of an action taken. The sample used in the implementation of color art creativity can be in the form of 3-5 year old children around the Sumber Mukti Indah Rembang housing complex. The child's parents can be used as a reference in the process of color art creativity activities. The results obtained were the children's enthusiasm in doing color art creativity activities using watercolors. Children feel happy and can produce aesthetic works. The activities implemented were painting using cotten bad, fingger painting, and telling stories using their respective works. The purpose of this activity is to foster children's artistic creativity, emotional, language and motor skills.

Abstract It has been seen from the above survey that a few of the treadwear predictor correlations have the required accuracy to provide a satisfactory alternative to road testing of tires for the wear performance of carbon blacks, depending on requirements and circumstances. Correlations based on laboratory rubber testing of rubber mixes and tread compounds are to be preferred over those based solely on carbon black colloidal and morphological measurements. In the author's opinion, Cotten and Dannenberg's correlation of treadwear index with Angle Abrasion measurements of 30 phr compounds and heated bound rubber, and Westlinning and Wolff's correlation with rebound resilience, and the Monsanto Rheometer determination of αF are satisfactory in most instances for the prediction of the treadwear index behavior of carbon blacks.

Abstract Salix is a genus of considerable taxonomic complexity, and accurate identification of its species and hybrids is not always possible. Quantification of ovules was used in this study to verify the parentage of a few hybrids of Salix. It has been shown that ovule numbers in willow hybrids are the mean of the ovule numbers of their parents. The ovule index of a prostrate specimen of S. ×cottetii affirmed that this was a hybrid of S. myrsinifolia Salisb. and S. retusa L., and the ovule index of the ornamental cultivar ‘The Hague’ affirmed that this was a hybrid of S. caprea L. and S. gracilistyla Miq. Finally, we also examined a confusing group, previously identified in North America as S. pentandra. The ovule indexes and other morphological characters indicated that there were four taxa among the studied specimens: S. pentandra, S. ×meyeriana, S. serissima Fernald, and a hybrid of S. serissima and S. fragilis that has not previously been described. It was concluded that quantification of ovules in willows is a reliable tool that can be used in willow taxonomy, genetics and population studies.

Amongst the rich and fairly diverse echinoid faunas from the Campanian marl/marly limestone facies as exposed at the large quarries of the cement industry near Höver and Misburg (east of Hannover, northern Germany), there are a few rare taxa, most of which have not been recorded previously from the Lower Saxony Basin. These include <i>Echinogalerus peltiformis</i> (Wahlenberg, 1821), <i>Conulus (C.) matesovi</i> Poslavskaia & Moskvin in Moskvin 1959, <i>Hagenowia blackmorei</i> Wright & Wright, 1949, <i>Diplodetus</i> sp., and <i>Peroniaster cotteaui</i> Gauthier in Peron 1887. New material of another rare species, <i>Hemiaster</i> gr. <i>aquisgranensis</i> Schlüter, 1899, is here described. The palaeoecological significance of the rarity of these is briefly discussed. Of particular note, in a palaeobiogeographic context, are <i>E. peltiformis</i> and <i>C. matesovi</i>. The former is well known from Lower and Upper Campanian arenitic facies in southern Sweden, while the latter was held to be confined to the Lower Campanian of the Caucasus and Kazakhstan. These two species may have immigrated into the Lower Saxony Basin following transgressive pulses or within transgressive systems tracts. <br><br> Die reiche und vergleichsweise diverse Echinidenfauna der Mergel/Mergelkalk-Rhythmite des Campans von Misburg und Höver bei Hannover enthält einige seltene Taxa, die bisher aus dem Niedersächsischen Becken nicht bekannt waren. Abstract:
A phylogenetic tree of the Mycoplasma mycoides cluster was inferred from a set of concatenated sequences from five housekeeping genes (fusA, glpQ, gyrB, lepA and rpoB). The relevance of this phylogeny was reinforced by detailed analysis of the congruence of the phylogenies derived from each of the five individual gene sequences. Two subclusters were distinguished. The M. mycoides subcluster comprised M. mycoides subsp. mycoides biotypes Small Colony (SC) and Large Colony (LC) and M. mycoides subsp. capri. The latter two groups could not be clearly separated, which supports previous proposals that they be united into a single taxonomic entity. The Mycoplasma capricolum subcluster included M. capricolum subsp. capricolum, M. capricolum subsp. capripneumoniae and Mycoplasma sp. bovine group 7 of Leach, a group of strains that remains unassigned. This group constituted a distinct branch within this cluster, supporting its classification as a subspecies of M. capricolum. Mycoplasma cottewii and Mycoplasma yeatsii clustered in a group that was distinct from Mycoplasma putrefaciens and they were all clearly separated from the M. mycoides cluster. In conclusion, this approach has allowed us to assign phylogenetic positions to all members of the M. mycoides cluster and related species and has proved the need to adjust the existing taxonomy. Furthermore, this method may be used as a reference technique to assign an unequivocal position to any particular strain related to this cluster and may lead to the development of new techniques for rapid species identification.

The aim of this study was to test whether cholinergic mechanisms regulate dopamine (DA) release from the carotid body (CB) and interact with DA D2 autoreceptors. One hundred forty-two CBs from adult rabbits were infused in vitro in a surviving medium bubbled with O2 (Bairam A, Marchal F, Cottet-Emard JM, Basson H, Pequignot JM, Hascoet JM, and Lahiri S. J Appl Physiol 80: 20–24, 1996 ). CB DA content and release were measured after 1 h of exposure to various treatments: control, cholinergic agonist (0.1–50 μM carbachol), full muscarinic antagonist (1 and 10 μM atropine), antagonists of M1 and M2 muscarinic receptors (1 and 10 μM pirenzepine and 10 μM AFDX-116, respectively), and the DA D2 receptor antagonist domperidone (1 μM), alone and with carbachol (1 μM). Compared with control, the release of DA was significantly increased by carbachol (1–50 μM), AFDX-116, and domperidone and decreased by atropine (10 μM) and pirenzepine (10 μM). The effects of domperidone and carbachol were not significantly different but were clearly additive. It is concluded that, in the rabbit CB, M1 and M2 muscarinic receptor subtypes may be involved in the control of DA release, in addition to the DA D2 autoreceptors.

Abstract:
Most geophysical applications in North American coal exploration have centered around the conventional surface seismic reflection method to provide continuous subsurface coverage for evaluating both good and anomalous coal reserve areas (Ruskey, 1981; Dobecki and Bartel, 1982; Greaves, 1984; Lawton, 1985; Lyatsky and Lawton, 1988; Gochioco and Cotten, 1989; Lawton and Lyatsky, 1989; Gochioco and Kelly, 1990; Gochioco, 1991; Henson and Sexton, 1991). The surface seismic reflection method, however, has inherent resolution limitations because the seismic wavelet must propagate substantial distances through the weathered layer, resulting in rapid attenuation of the desired higher frequencies. Since the depths and thicknesses of coal seams are usually known before‐hand, it is imperative that the seismic reflection associated with the target coal seam is absolutely identified in the seismic section to avoid misinterpretations. However, it is common that checkshot data and sonic and density logs are not available to generate synthetic seismograms to assist in the interpretation of coal seismic data. To overcome some of these limitations, the vertical seismic profiling (VSP) technique was tested in a coal exploration program to provide additional information for correlation with surface seismic reflection [or common‐depth‐point (CDP)] data and a synthetic seismogram generated from density and sonic logs.

The goal of the study was to improve the methodology of HIV genotyping for analysis of HIV drug resistance and HIV clustering. Using the protocol of Gall et al. (A. Gall, B. Ferns, C. Morris, S. Watson, M. Cotten, M. Robinson, N. Berry, D. Pillay, and P. Kellam, J Clin Microbiol 50:3838–3844, 2012, doi:10.1128/JCM.01516-12), we developed a robust methodology for amplification of two large fragments of viral genome covering about 80% of the unique HIV-1 genome sequence. Importantly, this method can be applied to both viral RNA and proviral DNA amplification templates, allowing genotyping in HIV-infected subjects with suppressed viral loads (e.g., subjects on antiretroviral therapy [ART]). The two amplicons cover critical regions across the HIV-1 genome (includingpolandenv), allowing analysis of mutations associated with resistance to protease inhibitors, reverse transcriptase inhibitors (nucleoside reverse transcriptase inhibitors [NRTIs] and nonnucleoside reverse transcriptase inhibitors [NNRTIs]), integrase strand transfer inhibitors, and virus entry inhibitors. The two amplicons generated span 7,124 bp, providing substantial sequence length and numbers of informative sites for comprehensive phylogenic analysis and greater refinement of viral linkage analyses in HIV prevention studies. The long-range HIV genotyping from proviral DNA was successful in about 90% of 212 targeted blood specimens collected in a cohort where the majority of patients had suppressed viral loads, including 65% of patients with undetectable levels of HIV-1 RNA loads. The generated amplicons could be sequenced by different methods, such as population Sanger sequencing, single-genome sequencing, or next-generation ultradeep sequencing. The developed method is cost-effective—the cost of the long-range HIV genotyping is under $140 per subject (by Sanger sequencing)—and has the potential to enable the scale up of public health HIV prevention interventions.

Physiol. 83(2): 376–382, 1997.—To determine the effect of coca chewing on heart rate (HR), mean arterial blood pressure (MAP), and plasma volume and their relationship with the hormones regulating cardiovascular and body fluid homeostasis, 16 male volunteers were examined at rest and during 1 h of cycle exercise at ∼75% of their peak oxygen uptake in two trials separated by 1 mo. One trial was performed after the subjects chewed a sugar-free chewing gum (Coca− trial), whereas the other was done after the subjects chewed 15 g of coca leaves (Coca+), with the order of the Coca− and Coca+ trials being randomized. Blood samples were taken at rest, before (R1) and after 1-h chewing (R2), and during the 5th, 15th, 30th, and 60th min of exercise. They were analyzed for hematocrit, hemoglobin concentration, red blood cell count, plasma proteins, and for the fluid regulatory hormones, including plasma catecholamines [norepinephrine (NE) and epinephrine], renin, arginine vasopressin, and the atrial natriuretic peptide (ANP). During the control trial (Coca−), from R1 to R2, there was no significant change in hematologic, hormonal, and cardiovascular status except for a small increase in plasma NE. In contrast, it can be calculated that coca chewing at rest induced a significant hemoconcentration (−3.8 ± 1.3% in blood and −7.0 ± 0.7% in plasma volume), increased NE and MAP, and reduced plasma ANP. Chewing coca before exercise reduced the body fluid shifts but enhanced HR response during exercise. These effects were not accompanied by changes in NE, epinephrine, renin, and arginine vasopressin plasma levels. In contrast, plasma ANP response to exercise was lower during the Coca+ trial, suggesting that central cardiac filling was reduced by coca use. It is likely that the reduction in body fluid volumes is a major contributing factor to the higher HR at any given time of exercise after coca chewing.

The effect of glucose (Glc) supplementation and route of administration on these relationships was studied in 26 GD athletes who were randomly assigned to receive 1.3 g/kg Glc by slow intravenous infusion (IV; n = 9), oral administration (PO; n = 9), or artificially sweetened placebo in 1 liter of water (Asp; n = 8) in the 2 h preceding a graded maximal exercise. Performance and La were similar among the three groups in normal glycogen (NG) or GD conditions. However, slightly improved performances were observed in GD compared with NG and were associated with a shift to the right in La curves. Blood Glc concentrations were higher in IV and PO before exercise, but they rapidly decreased to lowest levels in IV, gradually decreased over time in PO, and remained stable in Asp or NG. Insulin concentrations were highest in IV and lowest in Asp and NG at onset of exercise, rapidly decreasing in IV and PO although remaining at higher levels than in Asp or NG. In contrast, higher serum levels of free fatty acids were measured during exercise in Asp with no significant differences in glucagon or glycerol among the three groups. Free and sulfated NE increases were smaller in IV than in PO and Asp on exhaustion. In contrast, free and conjugated Epi were most increased in IV, with smallest increases in Asp. Dopamine levels were most increased in IV at exhaustion. We conclude that the changes of Epi and NE concentrations, associated with the activation of glucoregulatory mechanisms, including hyperinsulinemia, display different magnitude and time courses during exercise in GD subjects who receive oral vs. intravenous load of Glc before exercise. We speculate that the magnitude of insulin surge after acutely increased Glc before exercise in GD subjects may exert dissociative effects on adrenal-dependent glycogenolysis and on sympathetic responses.
