Abstract:
SummaryThe characteristics of La Serena cheese produced using animal rennet in place of the traditional vegetable rennet were evaluated. Total solids, fat and total N were higher in whey from animal rennet than in whey from vegetable rennet. Cheese produced using animal rennet had a higher moisture content and a lower pH. Proteolysis was more rapid in the cheese produced with vegetable rennet than in the cheese produced with animal rennet, but there was less lipolysis. Enterobacteriaceae and coliform counts were respectively 1·7 and 1·4 log units higher in 60 d vegetable rennet cheese than in 60 d animal rennet cheese. Softening of cheese texture was considerably more pronounced in vegetable rennet cheese, which also showed significantly higher flavour quality and intensity.

Abstract:
SummaryThe influence of lactic starter inoculation on La Serena cheese characteristics was investigated in duplicate experiments, each consisting of a vat inoculated with 1%Lactococcus lactissubsp.lactisskim milk culture and a control vat without lactic starter.Lc. lactisinoculation achieved a significant pH decrease in the interior and on the surface of cheese, which ranged from 0·09 to 0·25 units throughout ripening. Breakdown of αs1- and β-casein and formation of soluble N fractions were retarded byLc. lactisinoculation. Levels of αs1-casein, β-casein and pH 4·6-soluble N after 60 d were respectively 30·4, 34·9 and 28·8% in cheese made with starter and 18·4, 23·0 and 34·2% in control cheese. More acidic conditions in the interior of cheese made with starter resulted in a reduction of Enterobacteriaceae, coliform and staphylococci counts, which after 60 d were respectively 2·55, 2·40 and 0·29 log units lower than in control cheese. The firmer texture obtained throughout maturation for cheese made with starter was attributed to retarded proteolysis and lower pH value. Cheese flavour quality and intensity were significantly impaired byLc. lactisinoculation.

Abstract:
ABSTRACT Several Shigella flexneri mutants with defects in aromatic amino acid and/or purine biosynthesis have been evaluated as vaccines in humans or in animal models. To be suitable as a vaccine, a mutant has to show virulence attenuation, minimal reactogenicity, and a good immunogenic potential in animal models. With this aim, we have constructed five S. flexneri 5 (wild-type strain M90T) mutants with inactivation of one or two of the loci purEK, purHD, and guaBA, governing early or late steps of purine biosynthesis. The mutants have been analyzed in vitro in cell cultures and in vivo in the Sereny test and in the murine pulmonary model of shigellosis. M90T guaBA, M90T guaBA purEK, M90T guaBA purHD, and M90T purHD purEK gave a negative result in the Sereny test. In contrast, in the murine pulmonary model all of the strains had the same 50% lethal dose as the wild type, except M90T guaBA purHD, which did not result in death of the animals. Nevertheless, bacterial counts in infected lungs, immunohistochemistry, and reverse transcription-PCR analysis of mRNAs for tumor necrosis factor alpha (TNF-α), gamma interferon (IFN-γ), interleukin-1β (IL-1β), IL-6, IL-12, and inducible nitric oxide synthase (iNOS) revealed significant differences among the strains. At 72 h postinfection, M90T guaBA purHD still induced proinflammatory cytokines and factors such as IL-1β, IL-6, TNF-α, and iNOS, along with cytokines such as IL-12 and IFN-γ. Moreover, in the absence of evident lesions in murine tissues, this mutant highly stimulated major histocompatibility complex class II expression, showing a significant ability to activate the innate immunity of the host.

Villages scattered along the steep slopes of north-west Yunnan present a serene picture of colourful prayer flags fluttering from rooftops and only the occasional vehicle raising dust from dirt roads. But for the past few years, summers have been a time of intense and often violent conflict. In late July, neighbouring villages prepare to fight once again over access to forests which produce wild matsutake mushrooms, a high-value Japanese luxury food that has been harvested and exported from the region for the past 12 years. The quiet summer nightlife in the nearby county seat has been transformed to a bustling mushroom market busiest between midnight and dawn. Why has this market come into existence, and what have been its effects on access to and control over forest resources?

Abstract:
COUNTESS SERENI AND THE TOWN OF BLATNA The thesis is not only a contribution to the regional history of Blatná nad Blatná district, but also a probe to the life of a baroque noble marital pair. The first part of the study (the 1st and 2nd chapters) takes heed of the research survey up to now, mainly of the personality of J.P.Hill, and the summary of sources. The reader is also acquainted with the life milestone of Marie Alžběty (Mary Elizabeth), countess Sereni, née Waldstein (1698 - 1787), and her husband, count Josef (Joseph) Sereni. The Blatná demesne was under their rule in the period of 1709 to 1787 (part 3.1). Part 3.2 deals with the official career of count Josef Sereni. Chapters 4 and 5 present an insight to the everyday life of barogue nobility, mainly through the medium of preserved accounts of the count household; the author deals with the question of banqueting, their residences, domestic stuff and wardrobe, primarily taking account of noble self-presentation. Its specific form - the problems of family legends - is analysed in chapter 6. The most extensive chapter (No 7) deals mainly with widowed Marie Alžběta's activities at Blatná demesne, namely through baroque religiosity. Tyler, John. "A Pragmatic Standard of Legal Validity." Thesis, 2012. http://hdl.handle.net/1969.1/ETD-TAMU-2012-05-10885.

Full text
Abstract:
American jurisprudence currently applies two incompatible validity standards to determine which laws are enforceable. The natural law tradition evaluates validity by an uncertain standard of divine law, and its methodology relies on contradictory views of human reason. Legal positivism, on the other hand, relies on a methodology that commits the analytic fallacy, separates law from its application, and produces an incomplete model of law. These incompatible standards have created a schism in American jurisprudence that impairs the delivery of justice. This dissertation therefore formulates a new standard for legal validity. This new standard rejects the uncertainties and inconsistencies inherent in natural law theory. It also rejects the narrow linguistic methodology of legal positivism. In their stead, this dissertation adopts a pragmatic methodology that develops a standard for legal validity based on actual legal experience. This approach focuses on the operations of law and its effects upon ongoing human activities, and it evaluates legal principles by applying the experimental method to the social consequences they produce. Because legal history provides a long record of past experimentation with legal principles, legal history is an essential feature of this method. This new validity standard contains three principles. The principle of reason requires legal systems to respect every subject as a rational creature with a free will. The principle of reason also requires procedural due process to protect against the punishment of the innocent and the tyranny of the majority. Legal systems that respect their subjects' status as rational creatures with free wills permit their subjects to orient their own behavior. The principle of reason therefore requires substantive due process to ensure that laws provide dependable guideposts to individuals in orienting their behavior. The principle of consent recognizes that the legitimacy of law derives from the consent of those subject to its power. Common law custom, the doctrine of stare decisis, and legislation sanctioned by the subjects' legitimate representatives all evidence consent. The principle of autonomy establishes the authority of law. Laws must wield supremacy over political rulers, and political rulers must be subject to the same laws as other citizens. Political rulers may not arbitrarily alter the law to accord to their will. Legal history demonstrates that, in the absence of a validity standard based on these principles, legal systems will not treat their subjects as ends in themselves. They will inevitably treat their subjects as mere means to other ends. Once laws do this, men have no rest from evil.

Abstract:

Abstract:
Political violence is everywhere. But how does it emerge and what can be done about it? This book addresses the diversity of violence in South Asia, South East Asia and Western Europe. It examines the various forms of ideological backgrounds, structural conditions, relations and aims of non-state actors who are involved in violence in these regions and certain countries. Thereby, this book presents a similar diversity of theoretical and disciplinary approaches towards explaining the same phenomenon: violence. The rationale behind this collection of approaches and case studies is to identify communalities on the one hand, and to counter simple, unidimensional explanations of why non-state actors resort to violence on the other. Finally, it provides policy recommendations on how to counter violence. With contributions by Greg Barton, D. SubaChandran, Aurel Croissant, Rohan Gunaratna, Kevin McDonald, Subrata K. In Chapter 3, the author begins with labour colonies for the ‘idle poor’ in Europe (Holland, France, Britain, and Germany). In each case the volume analyses the colonies themselves and the population targeted but also the colonialism used to justify their existence. To this end, the writings of Jan van den Bosch of Holland, Alexis de Tocqueville, and Gustave de Beaumont of France; William Booth and Beatrice Webb of Britain; and Max Weber and Max Sering of Germany are examined as they weave together, in each case, domestic colonialism with various other schools of thought (Protestant Christianity, republicanism, paternalism, liberalism, socialism, and nationalism, respectively) to justify particular kinds of colonies for specific kinds of populations in each country.

Abstract:
Chapter 4 returns to John Higgins, who edited a selection of extant Mirror complaints for publication in 1586–7. Beginning with the reprinting of Thomas Norton and Thomas Sackville’s Inns of Court tragedy Gorboduc in 1590 alongside John Lydgate’s Serpent of Division, a publication which similarly pulled together ancient British and Roman narratives of assassination and civil conflict, the chapter interprets Higgins’s revisions and additions to his own First Part of the Mirror and suggests that they point to a change of emphasis. Higgins seems to have updated the work to sharpen its political focus at a moment of heightened national and international tension. The chapter then turns to his engagement with the use of moral exempla in the collection of Roman tragedies with which he enlarged the edition, and argues for a growing sense of disillusionment in their efficacy.

Abstract:
Chapter 6 explores the dramatic events spurring Melania and her entourage to leave Rome in advance of the Gothic sack of the city in August 410. After the death of Melania’s father in perhaps 405 or 406, Melania and Pinian went to a suburban property to begin their renunciations. Goths advanced into Italy starting in 401; from 407 onward, Alaric demanded ransom money and goods to prevent his attack on the city. Serena, wife of Stilicho and a member of the imperial court, had arranged with the emperor for court officials to assist the couple with the divestment of their property. When both Stilicho and Serena were killed, Melania and Pinian lost a major source of protection. The emperor Honorius dawdled in meeting Alaric’s requests, and Roman senators resisted parting with their wealth to pay the ransom. The Roman city prefect perhaps attempted to confiscate the couple’s property in order to pay the ransom but was killed in a riot over the bread supply. The extreme difficulty of divesting themselves of their possessions is clearly on display (Serena herself had claimed that she could not afford to buy their Roman mansion), as is the fierce opposition of their relatives, especially Pinian’s brother. The sack of Rome elicited various responses from Christian and pagan writers.

This chapter discusses Texas's growing presence in national affairs. It explains the economic, demographic, and political contexts in which religion emerged from World War II and contributed to the further shaping of race relations, faith convictions, and power. Churches benefited from the population growth driven by the postwar baby boom as well as from prosperous economic conditions. In many ways, the late 1940s and 1950s were a time of calm serenity in which religious leaders could focus on church growth and family formation. That was certainly an image that made sense to later observers who viewed the period from the perspective of the more turbulent civil rights era that followed. And yet these were years of remarkable developments in religion, politics, and business—years of inequality, discrimination, and conflict. These as well as new discussions about the separation of church and state set the stage for the civil rights movement and shaped the response to it.

Abstract:
This chapter explores the “Official Nationality,” an ideology created by Tsar Nicholas I. Nicholas sought to develop an alternative ideology that would counter Western European liberalism and emphasize Russia's distinctiveness, thereby forestalling revolution. To this end, in 1832 he appointed Count Sergei Uvarov as minister of popular enlightenment to reform the educational system and instill within it a new “national” spirit. Official Nationality was thus the moral counterpart of more practical reforms instituted by Uvarov. It consisted of three principles—“Orthodoxy, Autocracy, Nationality.” According to Uvarov, these were “the principles which form the distinctive character of Russia, and which belong only to Russia.” They were to be the basis of the moral education of Russia's youth, but they soon extended beyond the educational system to become the ideological principles of Nicholas's Russia as a whole.

Abstract:
Building on the melancholia of the films analysed thus far, a comparison between Alfonso Cuarón’s 2006 Children of Men, based in a world in mourning for its future, and the space opera Serenity (Joss Whedon, 2005), which concludes the story arc of Whedon’s TV series Firefly, gives an opportunity to further the discussion of history through an analysis of two mythic formations of hope, the redeemer and the frontier respectively. The chapter rounds off the theme of personal redemption versus the redemption of the collective, and the theme of obligation to the past necessitating a politics not of a deferred future but of redeeming the present in the name of the numberless dead, by an analysis of what does and does not count as human in the two films.

Abstract:
The image of the pre-revolutionary country estate in the Russian émigré magazines “Firebird” (1921–1926) and “Chimes” (1925–1929) is polyphonic and intermedial, being created with copies of artistic paintings related to the estate theme (by Sergei Sudeikin and Konstantin Somov in “The Firebird”, and by Sergei Zhukovsky and Sergei Vinogradov in “Chimes”), articles on characteristic features of the estate way of life, and on unique estates of the past (“Old Estates” by Nikolai Misheev in the “Chimes”), stories and poems related to the “estate topos” (“Spring on Krestovsky” by Sasha Chorny, “Youth” by Ivan Bunin, “On a Wolf Fur Coat” by Konstantin Balmont, etc., in “Firebird”). One can call the Sasha Chorny’s poem “The House over the Velicaia River” (“Pictures from Russian Life”) published in “Chimes” the real encyclopedia of the Russian estate life. The image of the estate in the émigré magazines is enshrined in myths of an idyllic image of the lost homeland, and is rooted in the idyllic reminiscences of the lost homeland. Life in harmony with nature, life and everyday activities arranged as works of art, one’s cultural and spiritual development, on one hand, becomes an unattainable nostalgic ideal for emigrants, and, on the other, it is a life-giving memory uniting Russians abroad.

Abstract:
This chapter picks up the narrative with the second marriage of the second Romanov tsar, Aleksei Mikhailovich, in 1671, and considers how changing political structures at court, a rising wall of Orthodox confessionalism, and the pious personality of the “most serene” tsar combined to produce further changes to the wedding ritual. The chapter also explores how changes in the way tsars wed reflected in aspects of their own biographies — their religiosity, their physical health, their mental health, and their ages. It elaborates the role that weddings played in this dynamic time when the ritual ground on which all politics rested was shifting tumultuously beneath them. Ultimately, the chapter ends by exploring the vastly reduced weddings of Aleksei's sons in the 1680s, concluding with Peter I's first wedding in 1689.

Abstract:
This chapter shows how Russian conservativism in the reign of Alexander I was multifaceted, often in opposition to state policy, and far from united. But there was a common core among its various strands. Men such as Aleksandr Shishkov, Fyodor Rostopchin, Sergei Glinka, Aleksandr Sturdza, and Nikolai Karamzin fused reason and faith to support a form of enlightened autocracy unlimited by law but firmly bound by morality. They also shared a concern with creating a modern Russian culture and a strong state that would be able to unite the country and provide stability in the face of internal and external pressures. In these ways they set the tone for Russian conservatives of future generations.

Abstract:
Susan Sered, author of the seminal work Uninsured in America: Life and Death in the Land of Opportunity (2005), returned to the same communities to learn how the people she originally interviewed were faring after the implementation of the ACA. Not a single person she interviewed had remained in the same coverage status for more than a few years at a time. Even with insurance, health care was hardly affordable for many. Most important, geographically driven health disparities had been exacerbated by the 2012 U.S. Supreme Court ruling, leaving large numbers of people to fall into the “coverage gap.” The existence of these gaps, together with the inconsistent nature of coverage and the absence of a human rights ethos, created barriers and resentment, with many people feeling that other categories of people received greater benefits.

Abstract:
Gram-negative septicemia is known to be associated with activation of the contact system and disseminated intravascular coagulation. Alpha-l-antitryosin-Pittsburgh (AT-P), a naturally occurring lethal nutation (358Met →> Arg) is a potent inhibitor of all the contact enzymes as well as the coagulation enzyme, thrombin. We hypothesized that the administration of AT-P might modulate the course of gram-negative sepsis. The pig was selected as the animal model of choice since its fibrinolytic, kinin-forming, and coagulation systems are similar to man as well as its response to endotoxin. Young pigs were infused with viable pseudomona aeruginosa (2x108 CFU), and were either untreated (Group I) or treated (Group II) for 6h, with recombinant AT-P (rAT-P), produced in yeast. Coagulation and hemodynamic parameters as well as survival rates were evaluated. The mean pulmonary artery pressure, mean systemic blood pressure, white blood count and platelet count were the same in both groups during the entire experiment. rAT-P was removed by immunoadsorp-tion prior to assay of coagulation parameters. At lh, the changes for all coagulation descriptors, AT-I II, Factor XI, fibrinogen, and fibrinogen degradation products were statistically significant (pπ.05). Prekal1ikrein levels, however, were not statistically different between groups. Survival rates in Group I were significantly less than Group II (pπ.05), during hours 2-5, but did not differ at 6h, when the functional plasma rAT-P levels were lowest. A discrepancy was observed between antigenic and functional levels of rAT-P that increased linearly during the course of the experiment. This suggested that activation of proteases was occurring with subsequent inactivation by rAT-P. This study indicates that rAT-P affords protection in experimental gram-negative septicemia by modulation of the activity of the serine proteases resulting from activation of the contact phase of blood coagulation, fibrinolysis, kallikrein and thrombin formation.

Abstract:

Wilhelm, S., and A. Henschen. "ON THE IDENTIFICATION OF POLYMORPHIC SITES IN HUMAN FIBRINOGEN PEPTIDE CHAINS." In XIth International Congress on Thrombosis and Haemostasis. Schattauer GmbH, 1987. http://dx.doi.org/10.1055/s-0038-1643327.

Abstract:
Human fibrinogen has repeatedly been shown to occur in a great number of different molecular forms. Some types of heterogeneity are evident already from variations in solubility properties and in ion-exchange-chromatographic as well as gel-electrophoretic behaviour of the total molecule and of its peptide chain components. The reason for these variations are partly known. Thus, degradation of the C-terminal parts and phosphorylation of two serine residues gives rise to heterogeneity in the Aα-chain. Differences in the sialylation of the carbohydrate side chain causes heterogeneity both in the Bβ- and the γ-chain. Differences in chain length at the C-terminus of the γ-chain are responsible for additional variation. All these variants are expected to exist in each human being. An other category of human fibrinogen variants may be due to genetic polymorphism within the population, i.e. the presence of inherited, common, normal variants. Seven sites of microheterogeneity have so far been tentatively identified, mainly by disagreements between protein and DNA sequence analyses. Three of the sites are located in the Aα-chain (positions 47, 296 and 312), three in the Bβ-chain (positions 162, 296 and 44-8) and one in the γ-chain (position 88). The aim of the present study was to identify these sites on the proteinchemical level in pooled plasma as well as in plasma from single individuals, especially various members of the same family. For this purpose suitable fibrinogen fragments containing the tentatively microheterogeneous sites were isolated after cleavage of fibrinogen with cyanogen bromide, trypsin and/or chymotrypsin by repeated fractionations by means of conventional and reversed-phase high-performance liquid chromatography and counter-current distribution. The components were characterized by N-terminal sequence and amino acid composition. Polymorphism in human fibrinogen has previously only once been identified by restriction fragment length analysis in a non-transcribed region of the Aα-chain locus but never in the transcribed regions, i.e. the peptide chains. The present investigation will allow the estimation of the number of peptide chain haplotypes and their possible correlation to other genetic variants of fibrinogen.
