ABSTRACT The CpxAR (Cpx) two-component regulator controls the expression of genes in response to a variety of environmental cues. The Cpx regulator has been implicated in the virulence of several gram-negative pathogens, although a role for Cpx in vivo has not been demonstrated directly. Here we investigate whether positive or negative control of gene expression by Cpx is important for the pathogenesis of Salmonella enterica serotype Typhimurium. The Cpx signal pathway in serotype Typhimurium was disrupted by insertional inactivation of the cpxA and cpxR genes. We also constitutively activated

ABSTRACT In Escherichia coli, the Cpx two-component regulatory system activates expression of protein folding and degrading factors in response to misfolded proteins in the bacterial envelope (inner membrane, periplasm, and outer membrane). It is comprised of the histidine kinase CpxA and the response regulator CpxR. This response plays a role in protection from stresses, such as elevated pH, as well as in the biogenesis of virulence factors. Here, we show that the Cpx periplasmic stress response is subject to amplification and repression through positive and negative autofeedback mechanisms.

ABSTRACT The Cpx pathway is a two-component signal transduction system that senses a variety of envelope stresses, including misfolded proteins, and responds by upregulating periplasmic folding and trafficking factors. CpxA resides in the inner membrane and has both kinase and phosphatase activities. CpxR, the response regulator, mediates a response by activating transcription of stress-combative genes. Signal transduction is subject to feedback inhibition via regulon member CpxP and autoamplification. Recently, it was shown that the Cpx pathway is also upregulated when cells adhere to hydroph

ABSTRACT In Escherichia coli, envelope stress can be overcome by three different envelope stress responses: the σE stress response and the Bae and Cpx two-component systems. The Cpx envelope stress response is controlled by the sensor kinase CpxA, the response regulator CpxR, and the novel periplasmic protein CpxP. CpxP mediates feedback inhibition of the Cpx pathway through a hypothetical interaction with the sensing domain of CpxA. No informative homologues of CpxP are known, and thus it is unclear how CpxP exerts this inhibition. Here, we identified six cpxP loss-of-function mutations using

In an effort to identify these remaining factors, we screened a collection of random lacZ operon fusions for those fusions whose transcription is regulated by CpxA/R. Abstract:
In cell/tissue cryopreservation, cryoprotective agents (CPAs) should be added before freezing and removed after thawing. Nowadays people mainly apply centrifugation for CPA removal, which may cause many problems. Meanwhile, a simple and cheap method for real-time monitoring of the residual CPA concentration during processing remains an unfilled need. In this work, a “dilution-filtration” system with hollow fiber dialyzer was implemented and approved to remove dimethyl sulfoxide (Me2SO) effectively. Compared to other methods (centrifugation and microfluidic approach), this one holds advantages

Abstract:
Previous research suggests that bisexual women’s rate of cannabis use is 2 to 7 times higher than their heterosexual peers; however, factors contributing to this are unclear. Trait impulsivity (i.e., tendency to act without forethought) and history of childhood physical abuse (CPA) are two risk factors that may be relevant for bisexual women’s cannabis use. Specifically, bisexual women indicate high levels of risk-taking and commonly report histories of CPA. While both impulsivity and CPA have been identified as predictors of cannabis use in heterosexual women, research has yet to explore thes
