Abstract:
Drosophila imaginal discs undergo extensive pattern formation during larval development, resulting in each cell acquiring a specific adult fate. The final manifestation of this pattern into adult structures is dependent on pulses of the steroid hormone ecdysone during metamorphosis, which trigger disc eversion, elongation and differentiation. We have defined genetic criteria that allow us to screen for ecdysone-inducible regulatory genes that are required for this transformation from patterned disc to adult structure. We describe here the first genetic locus isolated using these criteria: crooked legs (crol). crol mutants die during pupal development with defects in adult head eversion and leg morphogenesis. The crol gene is induced by ecdysone during the onset of metamorphosis and encodes at least three protein isoforms that contain 12–18 C2H2 zinc fingers. Consistent with this sequence motif, crol mutations have stage-specific effects on ecdysone-regulated gene expression. The EcR ecdysone receptor, and the BR-C, E74 and E75 early regulatory genes, are submaximally induced in crol mutants in response to the prepupal ecdysone pulse. These changes in gene activity are consistent with the crol lethal phenotypes and provide a basis for understanding the molecular mechanisms of crol action. The genetic criteria described here provide a new direction for identifying regulators of adult tissue development during insect metamorphosis.

Abstract:

Abstract. Context: Many aspects related to the teaching-learning process in swimming have not been subject of study. Objective: The purpose of the present investigation was to verify which methodological approach is more effective in improving the crawl technique: Exclusive practice of crawl, or practicing the four swimming strokes. Method: 26 male university students were randomly allocated into two groups: Crawl Group (GC): They exclusively performed crawl sessions. Swimming strokes group (GE): They underwent sessions of all the four swimming strokes. Results: After completing an eight-week intervention program, both groups enhanced their crawl technique. However, improvements observed in the GE were significantly greater than those achieved by the GC in the 25-metres crawl test (p <.001), stroke length (p=.006), average swimming speed (p=.000), and stroke index (p=.002). Conclusion: Practicing all four swimming strokes is more effective than crawl swimming only when it comes to improving the crawl technique in college-aged males.

Abstract:
ABSTRACT The white rot basidiomycete Phanerochaete chrysosporium produces an array of nonspecific extracellular enzymes thought to be involved in lignin degradation, including lignin peroxidases, manganese peroxidases, and the H2O2-generating copper radical oxidase, glyoxal oxidase (GLX). Preliminary analysis of the P. chrysosporium draft genome had identified six sequences with significant similarity to GLX and designated cro1 through cro6. The predicted mature protein sequences diverge substantially from one another, but the residues coordinating copper and constituting the radical redox site are conserved. Transcript profiles, microscopic examination, and lignin analysis of inoculated thin wood sections are consistent with differential regulation as decay advances. The cro2-encoded protein was detected by liquid chromatography-tandem mass spectrometry in defined medium. The cro2 cDNA was successfully expressed in Aspergillus nidulans under the control of the A. niger glucoamylase promoter and secretion signal. The recombinant CRO2 protein had a substantially different substrate preference than GLX. The role of structurally and functionally diverse cro genes in lignocellulose degradation remains to be established.

Abstract:

Abstract:
The aim of the present dissertation was to compare and characterize the cardiorespiratory, metabolic and biomechanical response between recreation swimmers and competitive swimmers. Twenty Portuguese male swimmers (eight masters and twelve elite) were evaluated. An incremental set of 7 x 200 m swims Crawl was applied to assess the cardio-respiratory, metabolic and biomechanical data. All energetic and biomechanical variables were related at velocity of the 4 mmol·l-1 to of lactate concentrations to compare all swimmers at the same intensity. Differences between both groups were testes with one away Anova (swimmer: recreation versus competition). The competitive swimmers presented a velocity at the 4 mmol·l-1 (F(1,18)= 73,541, p= 0,001), maximal oxygen consumption (F(1,18)= 13,152, p= 0,002), minimum swimming velocity where the maximal oxygen consumption is reached (F(1,18)= 29,364, p= 0,001), maximal total metabolic power at velocity at the 4 mmol·l-1 (F(1,18)= 11,717, p= 0,003), stroke frequency at velocity at the 4 mmol·l-1 (F(1,18)= 45,584, p= 0,001), mean swimming velocity (F(1,18)= 52,690, p= 0,001) and stroke index at velocity at the 4 mmol·l-1 (F(1,18)= 25,424, p= 0,001) significantly higher than recreational swimmers. There were no significant differences between the groups in the highest value of peak lactic acid after exercise (F(1,18)= 1,832, p= 0,193); energy cost at velocity at the 4 mmol·l-1 (F(1,18)= 1,50, p= 0,703); stroke length at velocity at the 4 mmol·l-1 (F(1,18)= 0,044, p= 0,836), and propelling efficiency at velocity at the 4 mmol·l-1 (F(1,18)= 0,315, p= 0,582). The competitive swimmers presented higher values than recreational swimmers in almost all variables assessed. This superiority allows them to achieve better performances when compared to recreational swimmers by the presence of a physiological profile more accurate.

Abstract:
Due to their potential for significant fuel consumption savings, Counter-Rotating Open Rotors (CRORs) are currently being considered as an alternative to high-bypass turbofans. When CRORs are mounted on an aircraft, several ‘installation effects’ arise which are not present when the engine is operated in isolation. This paper investigates how flow features arising from one such effect — the angle-of-attack of the engine centre-line relative to the oncoming flow — can influence the design of CROR engines. Three-dimensional full-annulus unsteady CFD simulations are used to predict the time-varying flow field experienced by each rotor and emphasis is put on the interaction of the front-rotor wake and tip vortex with the rear-rotor. A parametric study is presented that quantifies the rotor-rotor interaction as a function of the angle-of-attack. It is shown that angle-of-attack operation significantly changes the flow field and the unsteady lift on both rotors. In particular, a frequency analysis shows that the unsteady lift exhibits sidebands around the rotor-rotor interaction frequencies. Further, a non-linear increase in the total rear-rotor tip unsteadiness is observed for moderate and high angles-of-attack. The results presented in this paper demonstrate that common techniques used to mitigate CROR noise, such as modifying the rotor-rotor axial spacing and rear-rotor crop, can not be applied correctly unless angle-of-attack effects are taken into account.
