Abstract:
Il contenuto della tesi riguarda le analisi numeriche e sperimentali effettuate su un campione di cavo superconduttivo del sistema magnetico del reattore sperimentale per la fusione nucleare “ITER”. In particolare, il campione di cavo denominato “Toroidal Field Insert” o “TFI”, appartiene al sistema magnetico toroidale della macchina e viene inserito in un solenoide esterno in modo da replicare le condizioni di campo magnetico tipiche del normale funzionamento di questo conduttore. Le analisi sperimentali effettuate sul campione sono mirate alla caratterizzazione del comportamento durante un ipotetico ciclo di vita del cavo. I parametri principalmente studiati risultano essere: la caratterizzazione dello stato superconduttivo prima e dopo le varie sollecitazioni imposte, l’efficacia idraulica del raffreddamento e la stabilità termica del magnete. In modo complementare alla analisi dei dati sperimentali, due modelli numerici a diverse scale sono stati sviluppati e testati: un primo modello, alla scala di sistema, si occupa dello studio termico e idraulico dell’intera porzione di cavo testata; il secondo, alla scala di componente, si occupa della simulazione elettromagnetica di un riscaldatore induttivo installato sul conduttore volto a misurarne la stabilità. Il confronto tra l’analisi numerica e quella sperimentale ha permesso la comprensione dei principali fenomeni in gioco e la caratterizzazione del conduttore testato.

Abstract:
La performance des réseaux sans fil où les utilisateurs partagent l'air comme moyen de communication est fortement limitée par le phénomène d'interférence électromagnétique. En effet, deux utilisateurs proches qui communiquent sur la même fréquence verront leurs ondes interférer, ce qui peut entraîner la perte de l'information transmise. Ainsi, il est indispensable de mettre en place des protocoles d'accès visant à limiter l'interférence en choisissant de manière efficace les utilisateurs autorisés à émettre à chaque instant. D'un point de vue scientifique, il s'agit d'un problème difficile qui a attiré l'attention de la communauté en informatique et probabilités appliquées depuis plus de 30 ans. Récemment, une nouvelle classe de protocoles d'accès - appelés protocoles CSMA adaptatifs - a émergé, et semble très prometteuse : par exemple, il a été montré que ces nouveaux protocoles possèdent une propriété très attrayante de stabilité maximale. Le but de ce projet est d'approfondir la connaissance que l'on a des protocoles CSMA adaptatifs dits QB (pour l'anglais "Queue-Based") qui à ce jour est encore extrêmement limitée. Concernant ces protocoles, le but de ce projet est de prouver des résultats théoriques permettant de comprendre le compromis réalisable entre débit et délai. Modèle probabiliste - d'un point de vue technique, il s'agit d'étudier le modèle suivant: chaque utilisateur du réseau est représenté par le nœud d'un graphe G, appelé graphe d'interférence, et tel que deux voisins du graphe ne peuvent être actifs simultanément. Des paquets à transmettre arrivent à chaque nœud au cours du temps, et le but est de choisir quels nœuds sont actifs à un moment donné. Le protocole CSMA-QB répond à cette question de la manière suivante : lorsqu'un nœud est actif, il se désactive à taux constant et lorsqu'il est inactif et qu'aucun de ses voisins ne le bloquent, alors il s'active à un taux qui dépend du nombre de paquets en attente de transmission via une fonction ψ appelée fonction d'activation. Le but général de la thèse est de comprendre l'influence de la topologie de G et du choix de ψ sur la performance du protocole. Pour cela, il s'agira d'étudier le temps de mélange de la dynamique de Glauber ainsi qu'un phénomène classique en théorie des probabilités, appelé phénomène de moyennisation stochastique, qui permettent une compréhension fine du comportement dynamique du réseau
Performance of wireless networks, in which users share the air as support for their communications is strongly limited by electromagnetic interference. That is, two users close to each other trying to send a message on the same frequency will experience interference between their messages, eventually leading to the loss of some information. It is then crucial to develop medium access protocols aiming to limit the occurrence of such a phenomena by choosing in an effective (and distributed) manner which station is allowed to transmit. From a scientific point of view, it is a difficult issue which has had some attention from the community in the field of computer science and applied probability in the past 30 years. Recently, a new class of medium access protocols - called adaptive CSMA - emerged and seem quite promising: for example, it has been shown that they exhibit a desirable property: throughput optimality (maximum stability). The goal of this project is to increase the knowledge we have the adaptive CSMA (or CSMA QB, for Queue Based) which is to this day quite limited (notably in the expected waiting time of a request arriving in the system, called delay). Our goal will be to prove theoric results to enhance our understanding of the throughput/delay trade-off

Abstract:
Las señales mediadas por los Receptores Nucleares de Hormonas desempeñan un importante papel en el control de la diferenciación celular. El objetivo de este trabajo es analizar los mecanismos moleculares a través de los cuales se ejerce este control. Como sistema experimental utilizamos células de neuroblastoma humano SH-SY5Y, cuya diferenciación se induce mediante tratamiento con ácido retinoico (RA) que produce la parada en el crecimiento y desencadena la diferenciación celular. Estudiar el mecanismo molecular a través del cual, actúa el RA, también resulta interesante desde un punto de vista biomédico, puesto que los retinoides y sus derivados sintéticos son utilizados en la terapia de neuroblastoma y también de otros tipos de cáncer. Mediante Display Diferencial Ordenado hemos identificando 43 genes regulados (positiva o negativamente) por RA en células SH-SY5Y, entre los que se incluyen factores de transcripción y proteínas relacionadas con el ciclo celular, la transducción de señales y las funciones neuronales. Entre los genes regulados negativamente por RA se encuentra ID3, un factor de transcripción de la familia de los HLH, los cuales desempeñan un papel crucial en la diferenciación celular. ID3 actúa inhibiendo la unión al DNA de ciertos factores de la familia HLH. ID3 bloquea la diferenciación interfiriendo con la actividad de factores HLH neurogénicos. Cuando la expresión de ID3 se reduce por efecto de RA, se activa la cascada de bHLH proneurales, desencadenando la diferenciación celular. La expresión de otros genes bHLH cambió durante la diferenciación inducida por RA: la expresión del gen específico de neuroblastos en proliferación ASCL1 (HASH-1) disminuyó rápidamente tras 6 h de tratamiento con RA, mientras que la expresión de genes promotores de la diferenciación aumentó como es el caso de NEUROD6 y NEUROD1.Los niveles de otros miembros de la familia de los IDs (ID1, ID2) están regulados también negativamente durante la diferenciación inducida por RA en células SH-SY5Y.Mediante Western Blot estudiamos los niveles de proteínas ID1, ID2 e ID3 en células SH-SY5Y tratadas a diferentes tiempos con RA. Los niveles de proteínas disminuyen de manera notoria tras 24 h de tratamiento con RA, en paralelo a lo observado a nivel de ARN mensajero. Los resultados obtenidos indican que el RA produce una regulación negativa coordinada de los genes IDs. El tratamiento con otros inductores de la diferenciación como es el caso del TPA, también dio lugar a una regulación negativa y coordinada de los genes IDs. Esto resalta la importancia de estos genes en los procesos de diferenciación celular. Además hemos visto que la regulación negativa y coordinada de los genes IDs por tratamiento con RA es un mecanismo complejo que implica síntesis de nuevas proteínas y actividad fosfatidil-inositol-3-kinasa (PI3K). El tratamiento con RA activa la vía de señalización intracelular de la PI3K/AKT, resultando en un incremento en la actividad PI3K de extractos de células tratadas con RA y un rápido incremento en la fosforilación de la proteína diana AKT en Serina 473.La activación de la vía PI3K/AKT es necesaria para la diferenciación celular, y probablemente tiene una gran relevancia a nivel fisiológico, puesto que acopla procesos vitales como son la diferenciación y la supervivencia celular. Aunque el mecanismo molecular por el cual el RA activa la vía PI3K/AKT no está todavía definido, los resultados obtenidos indican que, probablemente, podría tratarse de una acción extragenómica del RAR, entre otras razones por la rapidez con la que se produce. Hemos tratado de caracterizar el mecanismo por el cual el RA activa la vía PI3K/AKT. En principio partimos de la hipótesis de que el RA pudiera inducir la expresión de una tirosina-kinasa que fuera capaz de activar la PI3K.Hemos encontrado un buen candidato para esta actividad: RET, el receptor de la familia de neurotrofinas del GDNF, es inducido directa- y fuertemente por RA. En las células SH-SY5Y se expresa uno de los co-receptores que actúa junto a RET, el GFRA2, pero no hemos logrado detectar la expresión de ninguno de los ligandos de la familia del GDNF, que pudiera activar RET actuando de un modo autocrino/paracrino. Por otro lado, no hemos podido observar fosforilación de RET en residuos tirosina, lo que probaría su activación. Estos resultados apoyan la hipótesis que planteamos en la cual el RA desempeñaría una acción de tipo no genómica.Los resultados obtenidos, nos llevan a postular, que el RA desempeña un papel muy importante en la regulación de la supervivencia de las células neurales. La regulación positiva del gen anti-apoptótico BCL2 durante la diferenciación inducida por RA en células de neuroblastoma es un hecho conocido desde hace tiempo, y la diferenciación incrementa la resistencia a la apoptosis inducida por drogas. También se han descrito efectos del RA sobre la supervivencia celular en cultivos primarios de neuroblastos de médula espinal y en neuronas derivadas de células madre neurales. Los resultados presentados aquí proporcionan un mecanismo molecular para explicar estos efectos. Además, pensamos que el fenómeno aquí descrito en células de neuroblastoma pudiera ser relevante en la diferenciación neural en general. La coordinación de las acciones genómicas y extragenómicas del RA da lugar al acoplamiento entre la diferenciación y la supervivencia celular. Por una parte el RA regula la transcripción de genes específicos implicados en diferenciación celular. Por otra parte, a través de una acción extragenómica, el RA activa la vía de PI3K/AKT, que está implicada en la supervivencia celular. Los dos tipos de acciones no se producen de un modo independiente, sino que están entrelazados. La activación extragenómica de la vía PI3K/AKT también está implicada en la regulación transcripcional de genes decisivos para la diferenciación, como hemos demostrado en el caso de la regulación negativa coordinada de los genes IDs. Recíprocamente, el RA puede contribuir a la supervivencia celular a través de la regulación transcripcional positiva de receptores de neurotrofinas, como RET y de trkB. Este acoplamiento entre diferenciación y supervivencia es un fenómeno novedoso.
Nuclear Hormone Receptors (NHR) are key regulators of cell differentiation. We have used the neuroblastoma cell line SH-SY5Y, that undergoes terminal differentiation after addition of Retinoic Acid (RA), as a model system to study the molecular mechanisms through which NHR control this process. By using Ordered Differential Display-PCR we have identified more than 50 genes differentially expressed in RA-treated SH-SY5Y cells. We have found that the expression of ID3, a member of the HLH family of transcription factors, is downregulated during RA-induced differentiation, and run-on transcription experiments demonstrated that this effect was transcriptional. Other ID's, like ID1 and ID2, that were expressed at lower levels in SH-SY5Y cells, were equally downregulated by RA. The levels of ID proteins decreased in parallel to the observed transcriptional repression. The expression of other bHLH genes changed during RA-induced differentiation: the expression of neuroblast-specific ASCL1 (HASH-1) gene was promptly reduced after RA treatment, whereas the expression of differentiation-promoting genes NEUROD6 (NEX-1, HATH-2) and NEUROD1 was increased. Our hypothesis is that ID's block cell differentiation by interfering the action of the neurogenic HLH transcription factors, and that RA released that block by downregulating ID's gene expression. Treatments with TPA, another inducer of neuroblastoma cell differentiation, also resulted in coordinated downregulation of the ID gene expression, underscoring the role of ID genes in differentiation. Downregulation of the ID gene expression by RA involves a complex mechanism, since full transcriptional repression required newly synthesized proteins and signaling by the Phosphatidylinositol-3 kinase. RA treatment activates the Phosphatidylinositol-3 kinase/Akt signaling pathway, resulting in increased Phosphatidylinositol-3 Kinase activity in extracts from RA-treated cells and a rapid increase in phosphorylation of Akt in Ser473. Inhibition of Phosphatidylinositol-3 kinase by LY294002 impaired RA-induced differentiation, as assessed by morphological and biochemical criteria. We propose that RA, by activating the Phosphatidylinositol-3 kinase/Akt signaling pathway, plays an important role in the regulation of the neuronal cell survival.

Abstract:

Today the utilization of all kind of multimedia services in networks increases and due to this demand for real-time communication gets higher for every day. One of the most common protocols used today is Ethernet. It is of interest to find real-time protocols that are useful together with already existing protocols since it is expensive to rebuild the network infrastructure. Several new protocols have been proposed to solve the upcoming problems.

The objective of this project is to make an inventory and a comparison between Ethernet and these new protocols. During this project four different protocols capability to fulfill the demands for real-time communication have been studied and compared with Ethernet. Three of the protocols are built on the same basic technique as Ethernet and these are PCSMA, CSMA-DCR and DOD-CSMA-CD. The fourth protocol is ATM and it transmits in a different way over communication networks.

The analysis of this project is pointing out advantages, similarities and differences between the protocols from a real-time perspective. All four protocols are more suitable for real-time requirements than Ethernet.

Abstract:
Initially, a systematic characterisation of all the possible ways in which two communicating pairs of nodes can interfere with each other is made. Using this as a building block and assuming independence of the stations, an estimate for the network throughput can be derived. The latter proves to be quite accurate for symmetric networks and manages to follow the performance trends in an arbitrary network. Following this, a more detailed Markovian-based mathematical model is proposed for the analysis of the hidden node case. This approach does not rely on common assumptions, such as renewal theory and node synchronisation, and is highly accurate, independently of the system parameters, unlike prior methods. Moreover, the usual decoupling approximation is not adopted; on the contrary, a joint view of the competing stations is taken into consideration. The model is firstly developed based on the assumption that the network stations employ a constant contention window for their backoff process. However, later in the thesis this assumption is relaxed, and performance curves are derived for the case when the stations employ the Binary Exponential Backoff Scheme, as is the case in practice. The Markovian state space is kept relatively small by employing an iterative technique that computes the unknown distributions. The adoption of this technique makes the analysis computationally efficient.

L’elaborato è dedicato all’esame della forma di emanazione e del sistema d’impugnazione degli atti del CSM circa la carriera giuridica dei magistrati, con particolare attenzione alle numerose implicazioni di livello costituzionale ed amministrativo sollevate dalla problematica. I principali obiettivi perseguiti sono: a) la verifica della costituzionalità del sistema predisposto dalla legge 24 marzo 1958, n. 195, la quale stabilisce che i provvedimenti consiliari siano emanati con decreto presidenziale o ministeriale e che possano essere sindacati dai giudici amministrativi; b) l’individuazione e la risoluzione delle ambiguità interpretative e dei dubbi applicativi che sono emersi fin dall’introduzione della legge in parola e che tutt’ora continuano costantemente a riproporsi, non avendo trovato compiuta sistemazione né in dottrina né in giurisprudenza; c) la ricognizione e l’esame critico della prassi giurisprudenziale, specialmente in ordine alle decisioni più recenti, tenuto conto della mancanza di studi aggiornati in merito pur a fronte dell’ampio numero di pronunce. Sulla base dell’ipotesi di partenza dell’irrinunciabilità, per la piena comprensione della portata precettiva della legge sopra citata, di un’esaustiva analisi delle premesse teoriche necessarie per una piena comprensione dello stessa, lo svolgimento è sostanzialmente articolato in due parti fondamentali. La prima incentrata sulla ricostruzione del perimetro costituzionale dell’indagine, con specifico riferimento all’assetto costituzionale della Magistratura. La seconda, riconducibile nell’alveo proprio del “diritto amministrativo”, concernente a) il significato e la funzione da attribuire alla forma di emanazione degli atti consiliari; b) i poteri ministeriali e presidenziali nella fase di esternazione; c) la tipologia degli atti impugnabili e dei vizi sindacabili in sede giurisdizionale. Con la consapevolezza che lo sviluppo dell’argomento prescelto rappresenta un’angolazione di visuale privilegiata relativamente allo svolgersi dei rapporti tra Potere Esecutivo ed Ordine giudiziario, la tesi affronta complesse questioni di carattere generale quali il principio di separazione dei poteri, il concetto di organo costituzionale e quello di autodichìa, la nozione di autonomia pubblica, i princìpi di imparzialità ed uguaglianza. Il vaglio della giurisprudenza costituzionale ed amministrativa, ampiamente richiamata, è condotto trattando a fondo, sia aspetti connotati da un elevato tasso di tecnicità, come la disciplina del conferimento d’incarichi direttivi, sia interrogativi concernenti diversi aspetti problematici. Segnatamente, l’indagine ha riguardato – tra l’altro – l’ammissibilità del sindacato del giudice amministrativo sugli atti “amministrativi” delle autorità non incardinate nella P.A.; la compatibilità dell’interpretazione evolutiva con l’art. 138 Cost.; la definizione di atto amministrativo; la stessa nozione costituzionale di pubblica amministrazione. La presa d’atto della tendenza giurisprudenziale ad estendere il sindacato al vizio di eccesso di potere conduce infine ad alcune riflessioni conclusive in merito alla conciliabilità del ruolo assunto dal giudice amministrativo col quadro normativo delineato dai Costituenti.

Abstract:
La glucoamilasa (GA) es uno de los enzimas producidos en mayor cantidad por la industria biotecnológica. Se emplea en el procesado del almidón degradándolo y liberando residuos de glucosa al medio. Saccharomyces cerevisiae (var. diastaticus) posee genes denominados STA que codifican GAs (Yamashita et al 1987). Este enzima presenta una estructura atípica ya que posee un dominio N-terminal, rico en residuos de serina y treonina (STRD o "Ser and Thr rich-domain") ausente en las otras GAs fúngicas y carece de un dominio de unión al almidón o (SBD o "Starch Binding domain) presente en la mayoría enzimas. La ausencia de este dominio hace que la GA de S. cerevisiae sea ineficaz en la degradación de almidón insoluble, necesaria para el procesado de las formas de almidón utilizadas en la industria. La presente tesis doctoral tiene como objetivo el estudio de las propiedades estructurales y funcionales de la glucoamilasa codificada por el gen STA1 de Saccharomyces cerevisiae así como la modificación del enzima, utilizando técnicas de ingeniería de proteínas, para obtener variantes con propiedades mejoradas y con mayor eficiencia catalítica. La mejora de sus propiedades abre interesantes posibilidades en el desarrollo de nuevos procedimientos fermentativos que permitirían la transformación directa de almidón en etanol.Se han obtenido estirpes de S. cerevisiae, de laboratorio y de origen industrial, capaces de sobreproducir el enzima ya que la levadura produce glucoamilasa en baja proporción. La mejora en la producción ha permitido la caracterización bioquímica del enzima, en concreto, se han analizado sus propiedades fisico-químicas y catalíticas. Especialmente se han estudiado las propiedades derivadas de la extrema glicosilación que presenta esta proteína.Por otro lado, se ha caracterizado estructuralmente la GA de S. cerevisiae obteniendo y analizando modelos tridimensionales de los dominios que conforman el enzima. En base a los estudios de los modelos obtenidos se ha construido un enzima híbrido, por adición de un dominio SBD, para conferir al enzima de mayor afinidad por el substrato y en consecuencia mayor capacidad catalítica. Por último se han mejorado las propiedades intrínsecas del enzima, mediante técnicas de mutagénesis aleatoria obteniendo una estirpe de levadura que secreta una variante enzimática más eficaz.
The glucoamylases (GAs) are important enzymes in the biotechnological industry. They are widely used for processing starch to simple sugars and ethanol. Saccharomyces cerevisiae (var diastaticus) carries genes designated STA that encode secreted glucoamylase (Yamashita et al., 1987). These enzymes present an atypical structure with two well defined parts, an amino-terminal domain, rich in residues of serine and threonine and a carboxy-terminal domain that contains catalytic domain. These enzymes in yeast lack a starch binding domain (SBD) present in others fungi glucoamylases that represent an important shortcoming for the digestion of insoluble starch. In this doctoral thesis we have studied the structural and functional properties of the Saccharomyces STA1- encoded glucoamylase. Furthermore we have modificated the enzyme using directed evolution to obtain mutant variants with improved properties. The improvement of activity opens interesting possibilities for the development of new fermentatives procedures that would allow the direct transformation of starch in ethanol. We have obtained S. cerevisiae strains, of laboratory and industrial origin, capable of overproducing this enzyme. The improvement of the production has allowed the biochemical characterisation of the enzyme, focusing on the study of the properties derivated from the extreme glycosilation that the protein presents. On the other hand, S. cerevisiae GA has been characterized structurally, obtaining and analyzing three-dimensional models of the enzyme domains. This model allowed the construccion and analysis of an engineered version of S. cerevisiae GA, which contains the starch binding domain from Aspergillus niger. The resulting hybrid enzyme has substrate binding capability and hydrolyses insoluble starch. Last but not least, we have improved the catalytic efficiency of the enzyme using random mutagenesis and recombination in yeast. Thus, we have obtained a yeast strain that secretes a more effective enzymatic variant.

In this paper, we identify sufficient conditions for Lyapunov Mean Square Stability (LMSS) of a contention-based network of first-order systems, with state-based schedulers. The stability analysis helps us to choose policies for adapting the scheduler threshold to the delay from the network and scheduler. We show that three scheduling laws can result in LMSS: constant-probability laws and additively increasing or decreasing probability laws. Our results counter the notions that increasing probability scheduling laws alone can guarantee stability of the closed-loop system, or that decreasing probability scheduling laws are required to mitigate congestion in the network.

Priority schemes employing the inherent properties of carrier-sense multiple-access (CSMA) schemes are investigated and then applied to the integrated transmission of voice and data. A priority scheme composed of 1-persistent and non-persistent CSMA protocols is proposed. The throughput and delay characteristics of this protocol are evaluated by mathematical analysis and simulation, respectively. The approach of throughput analysis is further extended to another more general case, p-persistent CSMA with two persistency factors, the throughput performance of which had not been analyzed before. Simulations are carried out to study the delay characteristics of this protocol. After careful consideration of the features of the priority schemes studied, two protocols are proposed for integrated voice and data transmission. While their ultimate purpose is for integrated services, they have different application. One of them is applied to local area network; the other is suitable for packet radio network. The distinctive features of the former are simplicity and flexibility. The latter is different from other studies in that collision detection is not required, and that it has small mean and variance of voice packet delay. Performance characteristics of both of these protocols are examined by simulations under various system parameter values.

I decided to write this master thesis in relation with my work experience at CSC within the service desk. Working at the service desk is a very complete mission that has to be well understood from analysts in order to reach high performance and thus satisfy users. Because it is the goal of CSC, satisfying customers, I decided to focus on this principle and with the results of my research and work experiences I would like to bring in this thesis some findings and recommendations I came up with in order to optimize the service desk quality and to improve the customer satisfaction. In the first chapter, I will mainly describe the work of an analyst at the service desk, and I will define the incident management process. It will help understanding how a service desk works. The incident management process is monitored by many metrics. Some of these metrics are documented and some of them are not and their impacts are different. We will see on what we should focus our efforts. We will see in the second chapter the impacts of these metrics on performance. Measuring the performance of the service desk is important in order to be aware of what should be improved to reach the highest standard of services provided. We will see in the end of this chapter that the most important performance indicator is the customer satisfaction. From this fact, I will bring some findings from seven months of experience within the service desk in order to improve the quality of the services and the customer satisfactions. I will focus on what can be implemented from the service desk analysts not from the management. I will also focus on the effects of the steps already taken to improve customer satisfaction.

The main goal of this Master's thesis is to estimate the value of the CSC Computer Sciences s.r.o. company to the date of 31st March 2015. To achieve this goal a strategic analysis, financial analysis, analysis of value generators and financial plan has been carried out. In this thesis the assets and the items of income statement have been divided as neccessary and unnecessary for company's operations to achieve more accurate statement of value, these two parts are separately evaluated and then sumed. The evaluation itself has been accomplished using DCF profit two stage method, which is divided into two parts that consist of a 5 year prediction phase and a residual value phase. The company has currently no long-term foreign capital that an interest has to be paid for, so the free cash flow to the firm equals the free cash flow to the equity. The final value of the company is then compared with the book value of assets.

The Collaborative Synchronization Management Tool (CSMT) is an analysis tool that enables morphological and statistical analysis of plans. Input to the CSMT consists of a Cross Impact Matrix (CIM) where the relationships between the different activities of a plan are reflected as its values. This thesis proposes Modeling and Simulation as an alternative method of generating the CIM-values. The usage of this method will hopefully increase traceability and limit subjectivity, and it will also be less time-consuming. Determining the level of detail of the models was shown to be a challenge, as well as finding a suitable case scenario to validate the generation method more thoroughly. The results have provided positive indicators to the usefulness of the generation method of input to the CSMT but the evaluation remains somewhat inconclusive.

In a packet radio network, multiple terminals broadcast information over a shared communications medium. Messages are transmitted from a source terminal to a destination terminal over multiple relays or hops. The actions of one terminal in the network directly affect the actions of other terminals within its range.

The analysis of packet radio networks is complex because system performance depends on the topology of the terminals in the network along with the random access protocol used. Researchers have been unable to model a packet radio network in its most general form. In this project, a model is developed for specific topologies of a packet radio network with a given random access protocol.

The sponsor of this project has developed a packet radio network for an indoor wireless alarm system. At the start of the project, the effect of message collisions on system performance was unknown. As a result of this research, a collision model for the network has been developed for topologies of four, five, and six alarm units. The model has been validated with actual system testing. Furthermore, the effects of indoor propagation on system performance have been studied. The results of this work have led to the development of an installation plan for the indoor wireless alarm system.

In this thesis the performance of the ALOHA and CSMA MAC protocols are analyzed in spatially distributed wireless networks. The main measurement metric used is probability of outage, a metric that is referred to in most of the related research done in this field, but has not been treated in detail thus far. Some of the research done on other performance metrics such as transmission capacity, throughput, bit error rate, spatial reuse and delay are also noted and described briefly in this report. In our system model, users/packets arrive randomly in space and time according to a Poisson point process, and are thereby transmitted to their intended destinations using a fully distributed MAC protocol (either ALOHA or CSMA). Our model allows simultaneous transmissions between many transmitter-receiver pairs in the network. An SINR-based model is considered, and a packet transmission is encountered as successful if the received SINR is above a predetermined threshold value for the entire duration of the packet. Accurate bounds on the probability of outage, which is a function of the density of transmissions, are developed for both MAC protocols. The methods used to reach the obtained analytical results are presented in detail, and the analytical results are shown to follow the simulation results tightly. We also present the methods that were used to find expressions for the outage probability, which turned out to not be as successful as desired in terms of following the simulation results tightly for all densities. Furthermore, the derived bounds for the probability of outage are used to determine the performance advantage that CSMA provides over ALOHA and also to gain insight into the design of general MAC protocols for ad hoc networks. Our final comparison results show that CSMA performs better than unslotted ALOHA, and worse than slotted ALOHA in terms of probability of outage.

We carried out a study on throughput estimation based on analysis and modeling of the medium access control in HomePlug 1.0 standard based PLC networks. The data communication using electrical energy wires has advantages such as presenting characteristics of ubiquity due to the existent infrastructure, but faces significant obstacles as fading and noise. The main standard of this kind of home network is the HomePlug, which defines a protocol based on the method of multiple access with collision avoidance (CSMA/CA). The HomePlug adds to the CSMA/CA a technique called deferral counter (DC) that adapts the contention of the nodes in accessing the medium according to network load. The objective of this work is todo a comparative study of the throughput, which is the ratio of the packet payload, i.e., the amount of data that is inserted into the body of the datagram, and the frame transmission time. To this end, we evaluate what is the theoretical maximum throughput of the PLC channel, we developed a simulator for the CSMA/CA and we propose a simple probabilistic model to describe the throughput on the network PLC. Finally, we make a comparison between the results obtained with the simulator and the probabilistic model to those observed from a real PLC network, proving that the results of the theoretical maximum throughput and the simulation results are close and that the probability model becomes a tool for calculating throughput in PLC networks.
Neste trabalho, realizou-se um estudo sobre estimação da vazão baseada na análise e modelagem do controle de acesso ao meio em redes PLC padrão HomePlug 1.0. A comunicação de dados através do canal elétrico tem vantagens como apresentar as características de ubiquidade e da infra-estrutura pré-existente, porém enfrenta obstáculos importantes como a atenuação e o ruído. O principal padrão desse tipo de rede domiciliar é o HomePlug, que define um protocolo baseado no método de acesso múltiplo com prevenção de colisões (CSMA/CA). O HomePlug associa ao CSMA/CA uma técnica chamada de contador de adiamentos (DC) que adapta a contenção dos nós no acesso ao meio de acordo com a carga da rede. O objetivo deste trabalho é realizar um estudo comparativo da vazão, que é a razão entre carga útil do pacote, ou seja, a quantidade de dados que preenchem o corpo do datagrama, e o tempo de transmissão do quadro. Para isso, avaliou-se qual é a vazão máxima teórica do canal PLC, desenvolveu-se um simulador para o protocolo CSMA/CA e foi proposto um modelo probabilístico simples para descrever a vazão na rede PLC. Foi realizada uma comparação entre os resultados obtidos com o simulador e o modelo probabilístico com os dados observados em uma rede PLC real, comprovando que os resultados da vazão máxima teórica e os resultados da simulação são próximos e que o modelo probabilístico passa a ser uma ferramenta para cálculo de vazão em redes PLC.

The new concept of Multi-Level Current Reinjection (MLCR) combines the advantages of DC ripple reinjection, multi-level conversion and soft-switching technique. Taking advantage of the soft-switching technique which uses zero current switching for the main bridge switches, thyristor based MLCR current source converter (CSC) is proposed. This concept adds self-commutation capability to thyristors and produces high quality line current waveforms. Various thyristor based MLCR CSC topologies have been simulated extensively using PSCAD/EMTDC in this thesis and their performance characteristics investigated. Questions have been raised about the ability to force the main thyristors off using the reinjection bridge in a real-world implementation, where there are inevitable stray capacitances and inductances which may influence the thyristor turn-off; and simulation switching models may not represent the switching characteristics fully or accurately. For this proof of concept, a small scale prototype has been built in the laboratory. The 3-level MLCR CSC, which increases the pulse number from 12 to 24, is chosen to verify the concept. The experimental investigation of the 3-level MLCR CSC, under steady-state conditions, verified the following: • The reinjection current allows the main bridge thyristors to be switched at negative firing angles. • This current reinjection technique allows self-commutation capability in a practical system despite the finite turn-off times of the thyristor. • This current reinjection technique improves the harmonic characteristics of the thyristor based converter. • It is observed that the deviation of the actual waveforms from the theoretical waveforms is mainly due to the snubber across the reinjection switch, and a trade-off in the choice of snubber components is required.

This paper examines the competence and independence of Chinese auditors. I find significant positive correlation between the modified audit opinions (MAOs) of China A-share companies from 2002 to 2011 and parameters of the clients' company-level control risks, auditor changes, and relative client importance. The evidence indicates that Chinese auditors generally demonstrate competence and independence. I argue that with the "Special Treatment" (ST) regulation, the audit quality of Chinese auditors remains satisfactory; without the ST regulation, the audit quality of Chinese auditors deteriorates. I use the regression results to substantiate that the audit failures of cross-listed Chinese companies occur because of the unavailability of the ST regulation, since the ST regulation has helped to improve the competence of auditors in China. Finally, I analyze the audit quality of "Big Four" affiliate companies in China, and find that the "Big Four" in China do not exhibit higher audit quality compared to local Chinese auditing firms. In fact, the quality of the "Big Four" auditing firms equals that of their non-"Big Four" peers..

Les protocoles de communications à accès aléatoires sont des candidats prometteurs pour les futurs systèmes de communications sans fil dédiés aux applications machine à machine (M2M). Ces méthodes d’accès sont généralement basées sur des techniques d'accès aléatoires mettant en œuvre des concepts simples de sondage de canal et de report de la transmission pour réduire les collisions, tout en évitant l'utilisation d'ordonnanceurs complexes. Parmi les différents protocoles, Carrier sense multiple access/collision avoidance with a Request-To-Send/Clear-To-Send (CSMA/CA-RTS/CTS) est un protocole qui pourrait être adopté pour les scénarios de M2M. Cette approche est efficace pour éviter les collisions entre les paquets de données. Cependant dans le cas d’un réseau très dense, les performances sont dégradées à cause de la forte probabilité de collisions. Pour atténuer cet effet, les collisions entre les messages de contrôles RTS doivent être réduites. Cette thèse propose de résoudre ce problème en divisant le canal commun en sous-canaux pour transmettre les messages de contrôle de demande d’accès au canal ; le canal commun est utilisé dans son ensemble pour la transmission de données. L’ajout d’un degré de liberté pour le message de demande d’accès permet de réduire la probabilité de collision, et donc d’améliorer les performances du système notamment dans des scénarios avec des nombres importants de nœuds souhaitant communiquer. Dans ce travail, nous dérivons ainsi une solution complète de méthode d’accès en s'appuyant sur le CSMA / CA - RTS / CTS et en multiplexant une configuration multi-canal pour les messages RTS et un canal unique pour la transmission de données. Une version améliorée, basée sur l'ordonnancement des utilisateurs, est également étudiée. Un modèle analytique a été développé, analysé et validé par simulations. Celui-ci est une extension du modèle Bianchi. Les performances en termes de débit saturé, de temps de transmission et de la probabilité de rejet de paquets sont discutées. Opportunistic protocols are promising candidates for future wireless systems dedicated to machine to machine (M2M) communication. Such protocols are usually based on a random access with simple techniques of medium sensing and deferring to reduce collisions while avoiding the use of complex schedulers. Among different protocols, Carrier sense multiple access/collision avoidance with a Request-To-Send/Clear-To-Send (CSMA/CA-RTS/CTS) is an opportunistic protocol which could be adopted for M2M scenarios. Such approach is efficient to avoid collisions between data packets but in a very dense network, the random access used to send the RTS suffers itself from a high probability of collision which degrades the performance. In order to mitigate this effect, RTS collisions should be reduced. This thesis proposes to address this issue by splitting the common channel in sub-channels for transmitting the RTS messages. While the common channel is used as a whole for data transmission. Multiple nodes can then contend in time and frequency for these RTS sub-channels, thereby reducing RTS collisions and increasing overall efficiency. In this work, we thus derive a complete protocol solution relying on CSMA/CA - RTS/CTS multiplexing a multi-channel configuration for RTS messages and a unique channel for data transmission. An enhanced version based on users scheduling is integrated as well. In this thesis, the proposed protocol is investigated from a joint PHY-MAC point of view. This strategy is shown to provide better system performance particularly for loaded networks. An accurate analytical model derived as a straightforward extension of the Bianchi model is analyzed and validated by simulations. Performance in terms of saturation throughput, transmission delay and packet drop probability is discussed

The purpose of the study is to, by using a specific model called Communication Systems/Situations Model CSM, identify the social context of a given organization and examine how it relates to the dimensions of communicational behaviour which are presented in CSM. We would also like to see how the different parts of CSM affects the choice of communication channels. The thesis is based on interviews with eight employees in the specific organization, whom we have asked a number of questions regarding how they communicate with each other and through which channels. The results show that the social context is very informal within the organization and the relationships between the employees are very open and personal. The dimensions of communicational behaviour relate to the context in many ways and are clearly a result of what kind of social context that exists in the organization. The conclusions we have drawn from the results are that a hierarchical pattern can bee seen between social context, dimensions of communicational behaviour and choice of communication channels in the studied organization. The hierarchical pattern is that the social context in a way surrounds the dimensions and in turn the dimensions affect the choice of communication channels.

Abstract:
The aim of this study was to examine how commercial sexual exploitation of children (CSEC) was constructed as a social problem in the Philippines. More specifically its aim was to find out how seven respondents from four different organisations in the Philippines constructed CSEC as a social problem and how they looked upon different solutions of the issue. The study was based on qualitative semi-structured interviews made in Manila the capital city of the Philippines and a city four hours from Manila called Olongapo. The earlier research as well as the result and analysis were sorted by using the themes: character, causes, solutions and actors which are parts of an analysis scheme constructed by Jönsson (2010). The empirical material was explained by using analysis based on three different kinds of perspective of approaching a social problem as well as earlier research. The result shows that CSEC is constructed as a social problem in the country and there can be different explanations of the issue. It is for example explained as a human rights issue in the Philippines and the problem also exists on a global level. CSEC is described to be normalised in the country because of the need of an income. The problem is also explained to be a cause of the existing traditional values and the lack of implementation of the laws that should protect the children. The conclusion of the study is that depending on how CSEC is constructed as a social problem, its risk factors and who are the perpetrators will affect which solutions that are seen as suitable.

Abstract:

In this thesis the optimal transmission rate in ad hoc wireless networks is analyzed. The performance metric used in the analysis is probability of outage. In our system model, users/packets arrive randomly in space and time according to a Poisson point process, and are thereby transmitted to their intended destinations using either ALOHA or CSMA as the MAC protocol. Our model is based on an SINR requirement, i.e., the received SINR must be above some predetermined threshold value, for the whole duration of a packet, in order for the transmission to be considered successful. If this is not the case an outage has occurred. In order to analyze how the transmission rate affects the probability of outage, we assume packets of K bits, and let the packet duration, T, vary. The nodes in the network then transmit packets with a requested transmission rate of Rreq=K/T bits per second. We incorporate transmission rate into already existing lower bounds on the probability of outage of ALOHA and CSMA, and use these expressions to find the optimal packet duration that minimizes the probability of outage. For the ALOHA protocol, we derive an analytic expression for the optimal spectral efficiency of the network as a function of path loss, which is used to find the optimal packet duration Topt . For the CSMA protocol, the optimal packet duration is observed through simulations. We find that in order to minimize the probability of outage in our network, we should choose our system parameters such that our requested transmission rate divided by system bandwidth is equal to the optimal spectral efficiency of our network.

O choque frio em bactérias causa a indução de proteínas de choque frio de baixo peso molecular (CSPs), que desestabilizam estruturas secundárias do mRNA, permitindo sua tradução. Caulobacter crescentus possui quatro genes codificando CSPs: cspA e cspB são induzidos sob choque frio, e cspC e cspD, na fase estacionária. Neste trabalho, foi determinada uma nova seqüência para o gene cspC, revelando que a proteína CspC possui dois domínios CSD, como CspD. O mutante nulo para cspC apresentou sensibilidade em baixa temperatura e menor viabilidade em fase estacionária, com alterações na morfologia. A região regulatória foi mapeada por fusões de transcrição, e uma região ativadora da expressão foi identificada, mostrando uma regulação transcricional. Algumas condições nutricionais que disparam a indução do gene foram determinadas, indicando que sua expressão é influenciada pela ausência de glicose no meio, mas não pela ausência de nitrogênio. Este perfil de indução não depende da região ativadora, que, por sua vez, é necessária para os máximos níveis de expressão.
The cold shock response in bacteria involves the expression of cold shock proteins (CSPs), which destabilize secondary structures on mRNAs, allowing their translation. Caulobacter crescentus possesses four genes encoding CSPs: cspA and cspB are induced upon cold shock, while cspC and cspD are induced at stationary phase. In this work, a new sequence for the coding region of the cspC gene was determined, revealing that CspC contains two cold shock domains, like CspD. A null cspC mutant was sensitive to low temperature, presented reduced viability at stationary phase, and altered morphology. The regulatory region of cspC was mapped by transcriptional fusions, identifying a region responsible for activation of cspC expression, suggesting a transcriptional regulation. Some nutritional conditions triggering cspC induction were determined, indicating that its expression is influenced by glucose starvation, but not by nitrogen starvation. This expression profile was not dependent on the activation region, which, in turn, was required for maximum levels of expression.

Long dismissed as a third world issue, the commercial sexual exploitation of children (CSEC) has been a significant problem across the United States over the past two decades, causing major concerns for child welfare stakeholders. The literature has shown a broad range of adverse consequences associated with CSEC. These include—but are not limited to—malnutrition, severe trauma, physical and sexual assault, sexually transmitted diseases, unwanted pregnancy, and medical neglect. The purpose of the study is to assess the readiness level of a large county in Southern California with regards to the well-being of children with a past or current record of victimization from commercial sexual exploitation. Embracing the Community Readiness Model, this case study aimed to evaluate (a) the county’s level of knowledge about programs and services to address CSEC, (b) the county leadership’s attitude toward addressing CSEC, (c) the county’s attitude toward addressing CSEC, (d) the county’s knowledge about CSEC, and (e) explore the county’s range of resources available to address CSEC. Results indicated that the county’s overall readiness score was 4.1 out of a possible 9. That is, the county had a low readiness level with respect to addressing the challenges associated with CSEC within its boundaries. In recent years, the brand issues have been considered as one of very important management strategies and measurements by the firms and academic research. Therefore, how to establish and implement an unconventional marketing approach to Strategic Brand Management has become an important worldwide topic for business management. However, as we review the existing literature on Strategic Brand Management, we found that most of the current literature take on the perspectives of the sellers’ marketing strategies, only a very few from the buyers’ perspectives, such as Strategic Supply Management. Since machinery is an important industry, with B2B business model, supplier management is a critical management issue. Therefore, this study will use a heavy machinery corporation as a case study company, analyzing its 100 procurement cases with 211 suppliers (each case has two assessments) as samples for a case study to investigate what are key factors in supplier's strategic brand management which mostly influence buyers' procurement qualification decision? Thereby affecting whether it obtain the qualification of entering the procurement process. The study found that Suppliers' strategic brand management have a positive impact relations to strategic partner levels, and the suppliers conform to the strategic partner levels has a significant chance of ascending into the procurement process. Abstract:
Recent works show that, by incorporating queue length information, CSMA/CA multiple access protocols can achieve maximum throughput in general ad-hoc wireless networks. In all of these protocols, the aggressiveness with which a link attempts to grab the channel is governed solely by its own queue, and is independent of the queues of other interfering links. While this independence allows for minimal control signaling, it results in schedules that change very slowly. This causes starvation and delays - especially at moderate to high loads. In this work we add a very small amount of signaling - an occasional few bits between interfering links. These bits allow us a new functionality: switching - a link can now turn off its interfering links with a certain probability. The challenge is ensuring maximum throughput and lower delay via the use of this new functionality. We develop a new protocol - Switch-enabled Queue-based CSMA (SQ-CSMA) - that uses switching to achieve both of these objectives. This simple additional functionality, and our protocol to leverage it, can be “added on'' to every existing CSMA/CA protocol that uses queue lengths. Interestingly, we see that in every case it has a significant positive impact on delay, universally furthering the performance of existing protocols.

This thesis investigates the stability of the random multiaccess protocols, slotted CSMA and slotted CSMA/CD, utilizing one power level and two power levels to create beneficial power capture effect. Use of more than two equally spaced power levels provides no significant improvement in the throughput achievable when realistic capture thresholds are considered. The investigation centers on a technique known as pseudo-Bayesian stability. Another task of this thesis is to stabilize multichannel slotted CSMA and slotted CSMA/CD with pseudo-Bayesian technique. The multichannel slotted CSMA and slotted CSMA/CD show a large improvement in throughput over a traditional single channel with a combined bit rate.

Demographic changes like ageing workforce raised questions about work–life balance and productivity among older workers. This study proposes a research in constructing Work-Life Balance as a major link between Work-Life Conflict and Job Performance of older workers. Specifically, it uses Personality as a moderator, because the attitude toward work or life is dependent on one’s personality or his or her point of view about job or personal life. Employee with positive affectivity may pay no regard to stress created by conflict between work and personal life. A survey was conducted within 2 weeks. Main respondents are 55-64 age blue-collar workers and their supervisors from two Taiwan industries: China Steel Corporation and China Shipbuilding Corporation. The sample size is 153. Results demonstrate that Work-Life Balance has a significant effect on Job Performance of older workers, and two types of Work-Life Conflict (time-based and strain-based) have a negative effect on Work-Life Balance of older employees. It is interesting that time-based conflict and strain-based conflict have no direct impact on Job Performance. However, Work-Life Conflict is negatively related to Job Performance through reduced Work-Life Balance. Also, there was no moderating effect of Personality on the relationship between Work-Life Conflict and Work-Life Balance. Despite research limitations, this study still provides some significant results and future research suggestions.
