Several biomaterials and techniques for bone grafting have been described in the literature for atresic bone tissue replacement caused by edentulism, surgical resectioning, and traumas. A new technique involves tissue engineering, a promising option to replace bone tissue and solve problems associated with morbidity of autogenous grafting. This literature review aims to describe tissue-engineering techniques using ex vivo cell culture as an alternative to repair bone maxillary atresias and discuss the concepts and potentials of bone regeneration through cell culture techniques as an option for restorative maxillofacial surgery.

This paper presents a novel fabrication process for micro metal parts using biologic etching method. The sheet metal parts can be etched by metabolism and biochemical reaction of microorganism. Thiobacillus ferrooxidans (T. f.) was employed in the machining process of metal components. By this bio-etching method, several shims with a thickness of 0.1mm and a diameter of only 2mm with a thickness of 0.07 mm and a diameter of 15mm and 16mm were fabricated.

Abstract:
Purpose – The purpose of this paper is to investigate if a firm’s organizational culture affects the set of quality techniques it uses, and if these quality techniques affect the relationship between organizational culture and operational performance. Design/methodology/approach – Based on data collected from 250 firms in Brazil and Denmark, structural equation modeling is used to investigate the relationship between organizational culture and the use of quality techniques, and its impact on operational performance. Four quality technique groups, four cultural profiles adopted from the Competing Values Framework and a set of operational performance indicators are used to operationalize the study. Findings – Culture does not appear to be an unequivocal predictor of the use of quality techniques. Furthermore, while most quality technique groups contribute indirectly to the total effect on operational performance in the developmental, group and hierarchical cultures, the performance effects are insignificant for all four groups in the rational culture. Practical implications – Managers need to be actively aware of the cultural characteristics of their organization before adopting quality techniques, in order to benefit most from the use of these techniques. Originality/value – Most previous studies address the relationships between culture, quality management and performance at the level of quality practices. This study takes the unitarist-pluralist discussion to the level of quality techniques and extends that discussion to what should be its core, namely, the influence of quality techniques on the performance impact of culture.

Abstract:
This study examines the application of the krama inggil lexicon by young generation based on education level as a form of implementation of subasita culture in Kudus. This study aims to describe the application of the krama inggil lexicon by today's younger generations, and to interpret the subasita culture that is reflected through the application of the krama inggil lexicon. This research is a qualitative descriptive study with a case study model. This study used 14 informants as data sources. The data of this research are in the form of informants' utterances which are indicated to contain the krama inggil lexicon. The technique of collecting data used observation techniques, listening proficiently, interviewing techniques, recording techniques, note taking techniques and questionnaire techniques. The data analysis technique used a qualitative descriptive technique. The results showed that: 1) the krama inggil lexicon was produced more by the younger generation with middle and lower education level; and 2) subasita culture is applied by the younger generation in different ways.

Abstract:
Quantitative polymerase chain reaction (qPCR) offers a rapid, highly sensitive analytical alternative to the traditional culture-based techniques of microbial enumeration typically used in water quality monitoring. Before qPCR can be widely applied within surface water monitoring programs and stormwater assessment research, the relationships between microbial concentrations measured by qPCR and culture-based methods must be assessed across a range of water types. Previous studies investigating fecal indicator bacteria quantification using molecular and culture-based techniques have compared measures of total concentration, but have not examined particle-associated microorganisms, which may be more important from a transport perspective, particularly during the calibration of predictive water quality models for watershed management purposes. This study compared total, free-phase, and particle-associated Escherichia coli concentrations as determined by the Colilert defined substrate method and qPCR targeting the uidA gene in stream grab samples partitioned via a calibrated centrifugation technique. Free-phase concentrations detected through qPCR were significantly higher than those detected using Colilert although total concentrations were statistically equivalent, suggesting a source of analytical bias. Although a specimen processing complex was used to identify and correct for inhibition of the qPCR reaction, high particle concentrations may have resulted in underestimation of total cell counts, particularly at low concentrations. Regardless, qPCR-based techniques will likely have an important future role in stormwater assessment and management.

The socio-cultural development of the community is a sign that the community in a certain area has experienced a change in the process of thinking. Changes in lifestyle caused by the inclusion of external culture will influence the pattern of thinking and perspective on health, because culture and health have a significant correlation. It is important to straighten out the beliefs or culture that the relationship adheres to with health, so that there are no mistakes in people's thinking patterns or perspectives that can adversely affect their health. For this reason, a therapeutic communication technique is needed by health workers. Based on the theory of S.O.R (Stimulus-Organism-Response) and four basic techniques of therapeutic communication; Listening techniques, questioning techniques, concluding techniques and techniques change the perspective as testers in data analysis. The conclusion from the results of this study shows that the technique used by health workers at the Bumi Sehat clinic in dealing with patient fanaticism is to combine the use of verbal and non-verbal language actively and effectively in each of the basic techniques of applied therapeutic communication.

The research method used was qualitative research. The researcher was the primer instrument, but the researcher needed the other's instruments. Therefore, additional data collection techniques used were interviewing and observing. The kind of interviewing technique was used in this research was the in-depth interview. Participant observation passively was selected to observe the subject and object of the research as additional data collection technique. The results suggest that sex education can be applied early and realized through the concept of school implanted is the existence of separate classes. Thus, separation of classes by gender based on the aspect of religiosity is very influential to sex education and characters education. Sex education can be applied to an early age and manifested through the concept of a school planted that is the existence of separate classes.

Since Vietnam is integrating internationally in all aspects of social life, more and more foreigners are learning Vietnamese. However, there are still very few quantitative studies on teaching and learning Vietnamese idioms effectively. This study aims to fill the theoretical gap and provide more evidence on the factors affecting Vietnamese idiom learning effectiveness. This study was conducted through a cross-sectional survey using a targeted sampling technique (n=100). The multiple linear regression models is applied to prove the hypotheses. The R language is used to analyze research data. The research results show that the willingness to learn Vietnamese and the strategy to learn Vietnamese have a positive and significant impact on the effectiveness of learning Vietnamese idioms. The results of this study continue to confirm the role of cognitive techniques in learning idioms as the results of previous studies.

Various tissue engineering techniques have been created in research spanning two centuries, resulting in new opportunities for growing cells in culture and the creation of 3-D tissue-like constructs. These techniques are classified as scaffold-based and scaffold-free techniques. Cell sheet, as a scaffold-free technique, has attracted research interest in the context of drug discovery and tissue repair, because it provides more predictive data for in vivo testing. It is one of the most promising techniques and has the potential to treat degenerative tissues such as heart, kidneys, and liver. In this paper, we argue the advantages of cell sheets as a scaffold-free approach, compared to other techniques, including scaffold-based and scaffold-free techniques such as the classic systemic injection of cell suspension.

The culturing of cells in the laboratory under controlled conditions has always been crucial for the advancement of scientific research. Cell-based assays have played an important role in providing simple, fast, accurate, and cost-effective methods in drug discovery, disease modeling, and tissue engineering while mitigating reliance on cost-intensive and ethically challenging animal studies. The techniques involved in culturing cells are critical as results are based on cellular response to drugs, cellular cues, external stimuli, and human physiology. In order to establish in vitro cultures, cells are either isolated from normal or diseased tissue and allowed to grow in two or three dimensions. Two-dimensional (2D) cell culture methods involve the proliferation of cells on flat rigid surfaces resulting in a monolayer culture, while in three-dimensional (3D) cell cultures, the additional dimension provides a more accurate representation of the tissue milieu. In this review, we discuss the various methods involved in the development of 3D cell culture systems emphasizing the differences between 2D and 3D systems and methods involved in the recapitulation of the organ-specific 3D microenvironment. In addition, we discuss the latest developments in 3D tissue model fabrication techniques, microfluidics-based organ-on-a-chip, and imaging as a characterization technique for 3D tissue models.

Brain abscess is a severe infectious disease with high mortality and mobility. Although culture-based techniques have been widely used for the investigation of microbial composition of brain abscess, these approaches are inherent biased. Recent studies using 16S ribosomal sequencing approaches revealed high complexity of the bacterial community involved in brain abscess but fail to detect fungal and viral composition. In the study, both culture-independent nanopore metagenomic sequencing and culture-based whole-genome sequencing using both the Illumina and the Nanopore platforms were conducted to investigate the microbial composition and genomic characterization in brain abscess. Culture-independent metagenomic sequencing revealed not only a larger taxonomic diversity of bacteria but also the presence of fungi and virus communities. The culture-based whole-genome sequencing identified a novel species in Prevotella and reconstructs a Streptococcus constellatus with a high GC-skew genome. Antibiotic-resistance genes CfxA and ErmF associated with resistance to penicillin and clindamycin were also identified in culture-based and culture-free sequencing. This study implies current understanding of brain abscess need to consider the broader diversity of microorganisms.

The article is dedicated to the conceptual and specific analysis of the emergence of the culture of legal techniques under the conditions of a new technological form. The identification and analysis of key types of culture of legal techniques, allows to show their specificity following typological groups. Its systemic unity, which has its specificities, can be considered as the second dominant of the culture of legal techniques. The article further offers a primary doctrinal definition of the concept of culture of legal techniques based on the identified dominant characteristics and manifestations of the culture of legal techniques, studied in the context of the search for ways of effective functioning of the system of power and powerless principles in the Russian legal system. This phenomenon is in the formation stage. The authors have used dialectical, historical-political, formal-legal, and comparative-legal methods. It is concluded that a promising systematic understanding of the essence and meaning of the culture of legal techniques will help to improve the legal culture as a whole and thus increase the effectiveness of the law in modern society.

The social change dynamic in globalization era impacts on the students’ reduced love to local wisdom culture so that they understand poorly the meaning of local wisdom culture existence as cultural heritage rich of life prudence values. This research aimed to analyse the internalization of local wisdom culture values through I-Spring technology-based art and culture learning. The unit of analysis used in this qualitative research with case study approach was the stakeholders of SMA Batik 2 Surakarta selected using purposive sampling technique. Techniques of collecting data used were observation, in-depth interview, and documentation. Data validation was carried out using source triangulation, while data analysis was carried out using an interactive model. The result of research showed that I-Spring technology-based art and culture learning could be a learning media facilitating the students to internalize local wisdom culture values. Through I-Spring learning media, students could internalize, externalize, and objectify local wisdom values in order to achieve the balance of students’ conscience, supra-ritual, and intellectuality related to God through the content of local wisdom values.

Abstract:
In cancer research and drug screening, multicellular tumor spheroids (MCTSs) are a popular model to bridge the gap between in vitro and in vivo. However, the current techniques to culture mixed co-culture MCTSs do not mimic the structural architecture and cellular spatial distribution in solid tumors. In this study we present an acoustic trapping-based core-shell MCTSs culture method using sequential seeding of the core and shell cells into microwells coated with a protein repellent coating. Scaffold-free core-shell ovarian cancer OVCAR-8 cell line MCTSs were cultured, stained, cleared and confocally imaged on-chip. Image analysis techniques were used to quantify the shell thickness (23.2 ± 1.8 µm) and shell coverage percentage (91.2 ± 2.8%). We also show that the shell thickness was evenly distributed over the MCTS cores with the exception of being slightly thinner close to the microwell bottom. This scaffold-free core-shell MCTSs formation technique and the analysis tools presented herein could be used as an internal migration assay within the MCTS or to form core-shell MCTS co-cultures to study therapy response or the interaction between tumor and stromal cells.

ABSTRACTCulture of periprosthetic tissue specimens in blood culture bottles is more sensitive than conventional techniques, but the impact on laboratory workflow has yet to be addressed. Herein, we examined the impact of culture of periprosthetic tissues in blood culture bottles on laboratory workflow and cost. The workflow was process mapped, decision tree models were constructed using probabilities of positive and negative cultures drawn from our published study (T. N. Peel, B. L. Dylla, J. G. Hughes, D. T. Lynch, K. E. Greenwood-Quaintance, A. C. Cheng, J. N. Mandrekar, and R. Patel, mBio 7:e01776-15, 2016,https://doi.org/10.1128/mBio.01776-15), and the processing times and resource costs from the laboratory staff time viewpoint were used to compare periprosthetic tissues culture processes using conventional techniques with culture in blood culture bottles. Sensitivity analysis was performed using various rates of positive cultures. Annualized labor savings were estimated based on salary costs from the U.S. Labor Bureau for Laboratory staff. The model demonstrated a 60.1% reduction in mean total staff time with the adoption of tissue inoculation into blood culture bottles compared to conventional techniques (mean ± standard deviation, 30.7 ± 27.6 versus 77.0 ± 35.3 h per month, respectively;P< 0.001). The estimated annualized labor cost savings of culture using blood culture bottles was $10,876.83 (±$337.16). Sensitivity analysis was performed using various rates of culture positivity (5 to 50%). Culture in blood culture bottles was cost-effective, based on the estimated labor cost savings of $2,132.71 for each percent increase in test accuracy. In conclusion, culture of periprosthetic tissue in blood culture bottles is not only more accurate than but is also cost-saving compared to conventional culture methods.

The objectives of the research were: to compile teaching materials for the art of music and dance based on interactive multimedia in the Class VIII Cultural Arts learning in the form of a Book Creator. The target achieved in this research is to produce interactive multimedia teaching materials which are integrated into a single unit in the form of a Book Creator. The results of the study resulted in Cultural Arts teaching materials in the form of Book Creator consisting of e-books (materials for the art of music and dance), Malay music learning videos on Sri Langkat songs and techniques for playing Malay musical instruments, and Nias dance learning videos on Giri-giri dance and Nias dance techniques. It is hoped that the existence of Cultural Arts teaching materials using interactive multimedia (Book Creator) can increase students' creativity in learning Cultural Arts.

Abstract:
The aim of this research is to explain the significance of digital literacy-based learning in the study of SKI at Madrasah Ibtidaiyah. In fact, few teachers have introduced digital literacy-based learning in the classroom. This is due to teachers' lack of understanding of how to use digital technology. This thesis employs qualitative descriptive analysis techniques in conjunction with field research. Techniques for gathering data included observation, interviews, and documentation. Descriptive analysis methods were used to analyze the data. Furthermore, comparing, checking the data's validity, and decrypting the data are all tasks that must be completed. The study's findings show that SKI's digital literacy-based learning is divided into four stages: (1) learning preparation, which includes deciding digital content based on the syllabus and lesson plans that will be provided to students; (2) digital material selection, which includes choosing digital files that will be shown during learning, such as images, movies, animated videos, or songs by examining whether or not the digital file is authorized. The learning material presented becomes more relevant and students become more engaged in learning as a result of digital literacy. This study serves as a resource for teachers interested in implementing digital literacy-based SKI learning, especially at Madrasah Ibtidaiyah

Background: The Sanger sequencing technique has been questioned and challenged by advanced high-throughput sequencing approaches. Sanger sequencing seems to be an obsolete technology. However, there are still research problems that could be answered using the Sanger sequencing technology. Fastidious obligate anaerobic bacteria are mostly associated with abscesses in animals. These bacteria are difficult to isolate from abscesses and are frequently excluded due to the bias of conventional bacterial culturing. Aim: This study demonstrated the usefulness of a broad-range polymerase chain reaction (PCR) with Sanger sequencing to identify the majority population of bacteria in abscesses from exotic pet animals. Materials and Methods: This study performed a pilot investigation of abscesses from 20 clinical cases (17 rabbits, 2 hedgehogs, and 1 sugar glider) using standard culture methods for both aerobes and anaerobes and broad-range nested PCR targeting the 16S rRNA gene followed by the Sanger sequencing technique. Results: The standard culture and PCR techniques detected bacteria in 9 and 17 of 20 samples, respectively. From the 17 sequencings of the 16S rRNA, 10 PCR products were found to be closely related with obligate anaerobes including Bacteroides spp., Fusobacterium spp., Prevotella spp. Phylogenetic analysis using the rpoB gene revealed that the species for the Bacteroides was thetaiotaomicron and for the Fusobacterium was varium and nucleatum. However, the amplification of the rpoB gene for the Prevotella spp. was unsuccessful. Correlations between the standard culture and PCR techniques were found in 9 (6 positive and 3 negative samples) of 20 samples. Eleven samples were discordant between the standard culture and PCR techniques which were composed of eight samples negative by culture but positive by PCR and three samples had different bacteria by the culture and PCR techniques. Conclusion: According to this study, broad-range PCR combined with Sanger sequencing might be useful for the detection of dominant anaerobic bacteria in abscesses that were overlooked based on conventional bacterial culture.

Based on these results, sterile physiological saline flushing with or without PULL THRU brush was selected for evaluation on 40 flexible endoscopes by adenosine triphosphate (ATP) measurement and bacterial culture. Acceptance criteria from the French National guideline (<25 colony-forming units [CFU] per endoscope and absence of indicator microorganisms) were used as part of the evaluation.RESULTSOn biofilm-coated PTFE tubes, physiological saline in combination with a PULL THRU brush generated higher mean ATP values (2,579 relative light units [RLU]) compared with saline alone (1,436 RLU; P=.047). In the endoscope samples, culture yield using saline plus the PULL THRU (mean, 43 CFU; range, 1–400 CFU) was significantly higher than that of saline alone (mean, 17 CFU; range, 0–500 CFU; P<.001). In samples obtained using the saline+PULL THRU brush method, ATP values of samples classified as unacceptable were significantly higher than those of samples classified as acceptable (P=.001).CONCLUSIONPhysiological saline flushing combined with PULL THRU brush to sample endoscopes generated higher ATP values and increased the yield of microbial surveillance culture. Consequently, the acceptance rate of endoscopes based on a defined CFU limit was significantly lower when the saline+PULL THRU method was used instead of saline alone.Infect Control Hosp Epidemiol 2017;38:1062–1069

<p><em>The aim of this study is to identify and describe the cultural elements translated using adaptation technique and linguistic elements translated using variation technique in Batman movie subtitled into Mataraman Javanese. Then explain the translator’s reason of using the adaptation and variation technique, and describe the effect of both techniques towards the translation quality. This is a prescriptive qualitative study and categorized as an embedded research. The sources of data are 1) script of Batman movie and its translation in Mataraman Javanese, 2) the raters as informant who evaluate the translation in terms of accuracy and acceptability, and 3) the target audiences to rate the comprehension.</em></p><em>Based on the research result, the researcher collected 110 data which consist of 48 adaptation data and 62 variation data. The adaptation data in this study are ecology, material culture, social culture, procedure/ activity/ artistic term, economic system and language. Meanwhile, the variation data contain the personal pronouns for first person, second person, and third person translated into speech level of ngoko (showing low politeness), madya (showing middle politeness) and krama (showing high politeness). The use of adaptation and variation techniques by the translator is in line with the purpose of Mataraman Javanese program held by local TV station to accommodate the local wisdom. For that reason, the translation ideology of this subtitle is domestication by employing those translation techniques which are closes to the target language culture.</em> <em>Generally, the use of adaptation and variation techniques in this study is quite accurate, acceptable in the target culture and comprehensible to the target audience.</em>

The soybean is important to the economy of Brazil, so the estimation of the planted area and the production with higher antecedence and reliability becomes essential. Techniques related to Remote Sensing may help to obtain this information at lower cost and less subjectivity in relation to traditional surveys. The aim of this study is to estimate the planted area with soybean culture in the crop of 2008/2009 in cities in the west of the state of Paraná, in Brazil, based on the spectral dynamics of the culture and through the use of the specific system of analysis for images of Landsat 5/TM satellite. The obtained results were satisfactory, because the classification supervised by Maximum Verisimilitude - MaxVer along with the techniques of the specific system of analysis for satellite images has allowed an estimate of soybean planted area (soybean mask), obtaining values of the metrics of Global Accuracy with an average of 79.05% and Kappa Index over 63.50% in all cities. The monitoring of a reference area was of great importance for determining the vegetative phase in which the culture is more different from the other targets, facilitating the choice of training samples (ROIs) and avoiding misclassifications.

Based on the preliminary research, the tenth-grade students lacked ability in mathematical problem solving of sine rule. One of the factors that influenced this problem was the teacher's learning model, which was still mechanistic. The teacher did not relate between learning and environment or local culture. Based on this, this study aims to develop HLT on the topic of a valid ethnomathematical PBL-based sine rule. The development model used is the Plomp model and the Gravemeijer and Cobb model, which consists of the initial investigation stage, product development stage, and assessment stage. In this article, the discussion is focused on the HLT development stage. The data analysis technique used is descriptive statistics and descriptive techniques. Data collection instruments used: validation sheets, observation sheets, interview sheets, questionnaires, and field notes. This study resulted in the HLT of the sine rule based on PBL nuances of Minangkabau Culture, which met the validity criteria of 0.771 and the characteristics of validity in terms of content and constructs.

Techniques to provide in vitro tissue culture have undergone significant changes during the last decades, and current applications involve interactions of cells and organoids, three-dimensional cell co-cultures, and organ/body-on-chip tools. Efficient computer-aided and mathematical model-based methods are required for efficient and knowledge-driven characterization, optimization, and routine manufacturing of tissue culture systems. As an alternative to purely experimental-driven research, the usage of comprehensive mathematical models as a virtual in silico representation of the tissue culture, namely a digital twin, can be advantageous. Digital twins include the mechanistic of the biological system in the form of diverse mathematical models, which describe the interaction between tissue culture techniques and cell growth, metabolism, and the quality of the tissue. In this review, current concepts, expectations, and the state of the art of digital twins for tissue culture concepts will be highlighted. In general, DT’s can be applied along the full process chain and along the product life cycle. Due to the complexity, the focus of this review will be especially on the design, characterization, and operation of the tissue culture techniques.

Abstract Introduction Traditionally, tissue engineering techniques have largely focused on 2D cell culture models—monolayers of immortalized or primary cells growing on tissue culture plastic. Although these techniques have proven useful in research, they often lack physiological validity, because of the absence of fundamental tissue properties, such as multicellular organization, specialized extracellular matrix structures, and molecular or force gradients essential to proper physiological function. More recent advances in 3D cell culture methods have facilitated the development of more complex physiological models and tissue constructs; however, these often rely on self-organization of cells (bottom-up design), and the range of tissue construct size and complexity generated by these methods remains relatively limited. By borrowing from advances in the additive manufacturing field, 3D bioprinting techniques are enabling top-down design and fabrication of cellular constructs with controlled sizing, spacing, and chemical functionality. The high degree of control over engineered tissue architecture, previously unavailable to researchers, enables the generation of more complex, physiologically relevant 3D tissue constructs. Three main 3D bioprinting techniques are reviewed—extrusion, droplet-based, and laser-assisted bioprinting techniques are among the more robust 3D bioprinting techniques, each with its own strengths and weaknesses. High complexity tissue constructs created through 3D bioprinting are opening up new avenues in tissue engineering, regenerative medicine, and physiological model systems for researchers in the military medicine community. Materials and Methods Recent primary literature and reviews were selected to provide a broad overview of the field of 3D bioprinting and illustrate techniques and examples of 3D bioprinting relevant to military medicine. References were selected to illustrate specific examples of advances and potential military medicine applications in the 3D bioprinting field, rather than to serve as a comprehensive review. Results Three classes of 3D bioprinting techniques were reviewed: extrusion, droplet-based, and laser-assisted bioprinting. Advantages, disadvantages, important considerations, and constraints of each technique were discussed. Examples from the primary literature were given to illustrate the techniques. Relevant applications of 3D bioprinting to military medicine, namely tissue engineering/regenerative medicine and new models of physiological systems, are discussed in the context of advancing military medicine. Conclusions 3D bioprinting is a rapidly evolving field that provides researchers the ability to build tissue constructs that are more complex and physiologically relevant than traditional 2D culture methods. Advances in bioprinting techniques, bioink formulation, and cell culture methods are being translated into new paradigms in tissue engineering and physiological system modeling, advancing the state of the art, and increasing construct availability to the military medicine research community.

At present, the earth-dwelling caves of thousands of years are rapidly disappearing and the techniques of construction are facing the danger of being lost as well, which is bound to bring about the loss of an architectural culture. This paper tries to approach the folk construction techniques of these traditional earth-dwelling caves, which will have a special significance to the passing-down and protection of such a cave housing culture.

Although stated differently, source and target text are conceptually and logically comparable. The concepts or meanings of the source and target text are not always the same. The process requires the translators’ adaptability skills to select necessary translation methods and techniques considered appropriate to the target readership. Therefore, the present study would like to examine translation methods and techniques employed in the treatment of cultural terms in the novel. The English translation of ‘Sukreni’ and the original Indonesian novel are the foci of this current study. This is a qualitative study that makes use of the content analysis approach. The data analysis technique categorizes the translation results based on cultural concepts, techniques, and methods. The fourteen translation techniques of Molina & Albir and the six translation methods of Hervey & Higgins are the frameworks of analysis in this study. Espindola & Vasconcellos’s concept of cultural elements distinguishes the types of cultural concepts portrayed in the original novel. The results show that the translation methods adopted to accommodate the cultural terms are adaptation and communicative methods. At the same time, the commonly used translation techniques are modulation, transposition, and adaptation. It can be concluded that the translator’s ideology also came to play alongside the choice of the translation methods and techniques when it comes to the readership of the target language. Methods and techniques of translations that are more inclined to the target language positively impact the translation results. Anthroponymy is the dominant cultural term in the original novel in the cultural category.

The technique of data processing is done is composed of six variables. The sixth variable is adopted from instruments that are formulated using scoring techniques. Scoring techniques in this study is "Likert scale" of scale associated with a statement about a person's attitude towards something (Kinner in Umar, 2008). Through the Likert scale, measured variables are translated into indikatot variables, hereinafter used as a starting point in preparing the items that can be a statement or a question.There is a significant influence of variable X to variable Y of 0.617 or 61.7%. It was concluded that if the variable X is high then lead to higher performance, and vice versa if the variable X is getting low, the resulting performance of employees will also be low. Based on the analysis of data obtained about the characteristics of organizational culture on employee performance in PDAM Merauke, it can be concluded that: Variable X1, X2, X3, and X4 positive and significant impact on the variable Y, and variable X5 but not significant positive effect on the variable Y, this can be seen that the value X5 by 0.264> 0.05

Abstract:
This study aims to describe the implementation of literacy culture programs of elementary school. This study used phenomenology of a qualitative approach. The subjects were the headmaster, teachers, students who were selected by using purposive sampling technique. The validity data test used triangulation techniques by comparing the results of observations, interviews, and documentation. The results showed that: 1) The policy for implementing a literacy culture at elementary school by using the School Literacy Movement program of the government. It is based on Permendikbud No. 23, 2015 focusing on character development. (2) The implementation of literacy culture of elementary school. It is based on the habituation stage that carried out 10 minutes of reading Al Qur’an and storybooks before lessons. The development stage of the activities carried out the respond to the contents of reading books by reading independently, giving appreciation for student literacy achievements, and having a school literacy team. The learning stage is by getting used to reading textbooks and using various strategies that support learning.

Abstract:
Coconut cultivation faces a number of acute problems that reduce its productivity and competitiveness. These problems include various biotic and abiotic challenges as well as an unstable market for its traditional oil-based products. Around 10 million small-holder farmers cultivate coconut palms worldwide on c. 12 million hectares of land, and many more people own a few coconut palms that contribute to their livelihoods. Inefficiency in the production and supply of appropriate seedlings for replanting a generally ageing resource remains an issue, especially where there is uncertainty about the ability of local populations to resist potential disease threats. However, tissue culture in such cases is expected to provide pragmatic solutions. Over the past 60 years much research has been directed towards developing and improving protocols for (i) embryo culture; (ii) clonal propagation via somatic embryogenesis and (iii) germplasm conservation via cryopreservation. Recent advances have provided new ways to improve these protocols, especially cryopreservation. Although effective embryo culture and cryopreservation are now possible, the low efficiency of conversion of somatic embryos to ex vitro seedlings still restrains the large-scale clonal propagation of coconut. Although tissue culture in coconut has developed over the recent decades, further improvement of protocols and their application to a wider range of germplasm will boost their adoption for the breeding, conservation and propagation of coconut.

The results showed that tourists’ perceptions of local tourism culture in Wuxi are mainly focused on religious culture, landscape and garden culture, and historical culture. Further analysis revealed an obvious structural mismatch between the tourists’ perceptions and the supply of local cultural tourism resources. The findings suggest that the focus should be on the clustered development of tourism culture in Wuxi, the branding of Wu Culture, and the strengthening of the association between scenic spots in the future.

ABSTRACT The genetic diversity of Trichodesmium spp. from natural populations (off Bermuda in the Sargasso Sea and off North Australia in the Arafura and Coral Seas) and of culture isolates from two regions (Sargasso Sea and Indian Ocean) was investigated. Three independent techniques were used, including a DNA fingerprinting method based on a highly iterated palindrome (HIP1), denaturing gradient gel electrophoresis of a hetR fragment, and sequencing of the internal transcribed spacer (ITS) of the 16S-23S rDNA region. Low genetic diversity was observed in natural populations of Trichodesmium spp. from the two hemispheres. Culture isolates of Trichodesmium thiebautii, Trichodesmium hildebrandtii, Trichodesmium tenue, and Katagnymene spiralis displayed remarkable similarity when these techniques were used, suggesting that K. spiralis is very closely related to the genus Trichodesmium. The largest genetic variation was found between Trichodesmium erythraeum and all other species of Trichodesmium, including a species of Katagnymene. Our data obtained with all three techniques suggest that there are two major clades of Trichodesmium spp. The HIP1 fingerprinting and ITS sequence analyses allowed the closely related species to be distinguished. This is the first report of the presence of HIP1 in marine cyanobacteria.
