Full textThis research was done to develop a new portographic technique in guinea pigs (Cavia porcellus) using sodium fluorescein as dye. 20 male healthy guinea pigs, between 10 to 16 weeks old and with a minimal weight of 700 g were used. Those animals were distributed in four experimental groups (A,B,C and D), and they received as treatment Sodium Chloride 0,9% for control group (A), Sodium fluorescein 0,1% (B), 0,5% (C) and 1% (D), respectively. The guinea pigs had abdominal surgeries in order to expose the liver and the small bowel. The distal jejunum and ileon were exposed to search an adequate jejunal vein to insert the intravenous catheter. The anatomical point of inoculation was selected based in shape, diameter, and accessibility of the vein, as next to the mesenteric border of the bowel as possible. A catheter size of 24 USP was used to inject the solution treatment according to the experimental group using doses of 2 ml/kg, and the hour of inoculation was registered. Direct observation of the Surface of the liver was evaluated for each animal under the ultraviolet light of a Wood’s lamp. The intensity of the coloration of the liver, the area of distribution of the tincture in the liver parenchyma, the time to obtain the largest area of coloration of the liver and the time to excrete the entire fluorescein dye in urine were evaluated. The statistic shows the difference that exists among the three groups that received any of the fluorescein solutions (B, C, and D groups) against the control group (A) for each of the four variables of the study. The comparison among the three groups treated using fluorescein shows no difference among B, C and D groups for the variables area of distribution of the tincture in the liver parenchyma and time to obtain the greatest area of coloration of the liver. When the intensity of the coloration of the liver was studied, we found there were no differences between C and D groups and there were difference between those groups against B group (whom received the less concentrated solution of fluorescein). The time of total excretion of fluorescein dye in urine was analyzed; we found that all guinea pigs visibly excreted the fluorescein dye in urine under the ultraviolet light of Wood’s lamp in the course of 16 hours post inoculation, without definitive differences among B, C and D groups. Only the comparison between B and C groups showed significative difference. We conclude that the injection of sodium fluorescein solutions through a jejunal vein as tributary of portal venous system has a high tinctional distribution in the liver, independently of the dye dilution used; dilutions of 0,5% and 1% of sodium fluorescein are equally useful to obtain a high intensity of coloration of the liver. The time to obtain the major area of coloration of the liver parenchyma was the same independent of the dilution of the dye used, and every guinea pig of this study excreted the fluorescein dye in urine in the course of 16 hours post inoculation. Aditionally, none of guinea pigs of this study died during the experimental research; it was suggested that sodium fluorescein dye is secure to be used by intravenous injection in guinea pigs. Full text--- The guinea pig production is an activity that becomes increasingly more important in our country due to increased demand for the meat of this species in domestic and international markets. However, health knowledge in guinea pigs are still scarce, making it vulnerable to various diseases. In this context stands salmonellosis, disease that causes high mortality and morbidity rates of production of guinea pigs. This study identified the most common pathologic lesions predominated in organs of guinea pigs infected with Salmonella spp. This retrospective study was conducted on necropsy of 125 guinea pigs, obtained from the Laboratory of Histology, Embryology and Veterinary Pathology, College of Veterinary Medicine of the Universidad Nacional Mayor de San Marcos, in a period between 2001 to 2007, the diagnosis of salmonellosis was confirmed by the Veterinary Microbiology Laboratory of the University, resulting in 81 guinea pigs positive for Salmonella spp. and 44 negatives. The results show that most organs were injured, with a median of 5 organs per animal, the liver being the organ that pathological lesions often presented with a 87.7% ± 0.07 (71/81) being the prevailing pathomorphological image necrotic hepatitis in 44.4% (36/81). Likewise, it was made the classification of pathological lesions in inflammatory processes, circulatory disorders, degenerative and adaptive, inflammation being the most common pathological disorder, accounting for 43.3% ± 4.8 (177/408) of total injured organs found in the 81 guinea pigs infected with Salmonella spp. Key Words: Guinea pigs, Salmonella spp., anatomopathological lesions.

Full text--- The objetive of this study was to isolate and identify bacterial strains by molecular techniques belonging to genera with potential for probiotics in the intestine of guinea pigs (Cavia porcellus) as possible future alternative to antibiotics. To do this, we used 13 guinea pigs, ten infants from 2 to 6 days old and three adults from 2 months old, the intestine was extracted from each sample were taken by scraping the intestinal mucosa, they were planted in different culture media in order to achieve isolated colonies with phenotypic characteristics of bacterial species of known potential probiotic (Lactobacillus, Enterococcus, Streptococcus, Bacillus, Bifidobacterium). We identified 27 representative strains by amplification, sequencing and bioinformatic analysis of 16S rRNA gene. DNA sequences were compared with three different databases: Blast server for bacterial identification, Ribosomal Database Project, BIBI database to identify the genus. It turned out that 85.18% (23) obtain Lactobacillus, Enterococcus, Streptococcus and Bacillus, the 14.81% (4) remaining bacterial species gram positive identified that are not objective of this study, but that however, contribute to the identification of guinea pig intestinal microbiota, was found Staphylococcus. Finally, we reached the target in question, identification of probable bacterial genera with probiotic potential. Is suggested to evaluate, in subsequent research, the probiotic potential of these bacterial genera. Keywords: Guinea pig, probiotic potential, PCR, identification.

Realiza la identificación molecular de bacterias presentes en el ciego del cuy mediante técnicas moleculares. ADN genómico bacteriano fue obtenido a partir de200 uL de contenido cecal de cuy. Fragmentos de 728 pares de bases del gen 16S rDNA fueron amplificados a partir de un pool de ADN genómico bacteriano mediante PCR. Los productos de PCR fueron clonados al azar en PGEM–T vector plasmid (Promega), transformados en Escherichia coli JM109 y 50 colonias fueron seleccionadas al azar y secuenciadas en ambos sentidos en un secuenciador automático ABI 3130 Genetic Analyzer. Dieciséis secuencias únicas del gen 16S rDNA fueron obtenidas mostrando altos grados de similaridad (> 95%) y un valor de e (e value -140) con secuencias de bacterias previamente descritas en bases de datos del GeneBank, Blast server for bacterial identification, Ribosomal database project y BiBi database indicando la presencia de Escherichia coli, Escherichia albertii, Shigella sp., Shigella sonnei, Shigella boydii y Hafnia alveien contenido cecal de cuy. El presente trabajo ha demostrado la presencia de bacterias del genero Escherichia, Shigella y Hafnia en el contenido cecal del cuy mediante el secuenciamiento del gen 16S rDNA.
