Ketatnya persaingan industri yang berada dikawasan Pekanbaru menyebabkan berkurangnya pangsa pasar dan pemasaran produk tidak merata Manfaat yang diharapkan dari penelitian ini adalah Perusahaan dapat mengetahui posisi strategi organisasinya saat ini yang dilihat dari faktor ekternal dan faktor internal serta mengetahui posisi strategi perusahaan. Penelitian ini menggunakan metode SWOT. Melalui penerapan metode SWOT ini, CV. Sekar Sari Roti dapat merancang strategi pemasaran dan posisi strategi perusahaan dengan tahapan penilaian faktor Internal dan eksternal serta diagram SWOT dan analisa diagram fishbone. Berdasarkan metode SWOT menunjukkan posisi strategi di CV. Sekar Sari Roti berada pada kuadran II Stability yaitu memanfaatkan peluang yang ada dengan cara meminimalkan kelemahan yang ada.

Abstract:
The article presents the results of the analysis of the reproductive ability of cows depending on the level of their milk productivity. The studies were conducted in the Republic of Mari El on cows of the Holstein black-and-white breed (n = 3828). There have been studied the influence of different levels of milk yield on the duration of the service period and period between calvings, the output of calves per 100 cows, the coefficient of reproductive ability. The studies used the comparison method, correlation, regression and one-factor analysis of variance. It has been established that the milk yield increases with the age of cows, but at the same time the duration of the service period and calving interval increases. In first-calf cows, the service period was 128.8 days (Cv = 75.1 %), the calving interval was 403.6 days (Cv = 23.5 %). By the third lactation, with an increase in milk yield by 1030 kg (15.4 %), the duration of the service period increased by 7.2 days (5.6 %), the calving interval -by 12.3 days (3.0 %). The share of the impact (ƞ2x) of milk yield on the duration of the service period was 5.4 %, the calving interval -4.7 %. The correlation coefficients between milk yield for 305 days of the first lactation and the duration of the service period and calving interval were 0.24 and 0.22 (p<0.05), with milk yield for the entire lactation 0.81 (p<0.05). The optimal indicators of the service period (89.4 days), the calving interval (369.4 days), the reproductive capacity (0.94) and the output of calves (1.0) were possessed by the first heifers with low milk productivity-milk yield less than 5000 kg of milk. In animals with the milk yield of 6000-9000 kg of milk or more, the duration of the service period was 12.4-249.6 % higher, the calving interval -by 1.9-32.5 % (p<0.05). According to the regression coefficients, on average, each increase in milk yield per 1000 kg increased the duration of the service period by 25.9 days, the calving interval -by 23.1 days, which worsened the output of calves by 0.24 %, and the coefficient of reproductive ability decreased by 16.0 %.

Abstract Determination of activities of phospholipase A2 (PLA2) in human sera was based on the hydrolysis of phospholipids from [1-14C]oleic acid-labeled Escherichia coli biomembranes. The E. coli membranes served as substrate specifically for the PLA2 of human serum and were essentially resistant to other lipases in human sera, i.e., lipoprotein lipases, hepatic triacylglycerolipase, or pancreatic lipase in acute pancreatitis. Exchange of phospholipids between the serum and the biomembrane compartment aggravates the determination of PLA2 activity in human serum, which is naturally rich in phospholipids. In our modified E. coli assay, which overcomes these difficulties, the main substrate components phosphatidylethanolamine (70%) and cardiolipin (25%) were &gt; 90% labeled in the sn-2 position. Fatty acids released by PLA2 activity were eluted from an aminopropyl solid-phase column directly into scintillation vials, where the radioactivity was counted. The ratio of [1-14C]oleic acid to released total fatty acids was used to calculate true enzymatic activity. The linear assay range extended from 0 to 3.6 U/L (0-60 nkat/L), with a detection limit of &lt; 0.03 U/L (&lt; 0.5 nkat/L). Within-assay imprecision (CV) was &lt; 6% and between-assay is &lt; 10% over the whole activity range. The normal range for men was 0-0.44 U/L (0-7.33 nkat/L) and for women 0.044-1.11 U/L (0.73-18.4 nkat/L). Patients with septicemia, pancreatitis, acute respiratory distress syndrome, or other severe diseases had PLA2 values up to 540 U/L (9000 nkat/L).

Abstract:
Greenhouse and nursery managers rely on testing laboratories with the expectations of accuracy and consistency. The Greenhouse and Nursery Media Analysis Proficiency (GNMAP) Testing program was initiated to provide laboratories servicing greenhouses and nurseries with inter-laboratory quality control. The GNMAP program operational guidelines are based on those outlined under ISO 9000, ISO/IEC Guide 43 and Draft ISO/IEC Guide 24, which describe the requirements for proficiency testing. Nine laboratories enrolled in the program in 2003 and submitted results for root zone media and fertilizer solutions. Data analysis provided the minimum, maximum and median values; median absolute deviation (MAD); overall reproducibility (Rd); individual reported lab values; repeatability (Rp) of lab value (CV for the individual lab); and mean lab value reported. The Rd was calculated from the median of all lab Rp values and is a measure of intra-lab variance. A measure of inter-lab variance was determined by calculating the relative median deviations (RMD = MAD/Median × 100). For one of the media distributed, results for the saturated media extract included median pH values from 4.3 to 6.9 with MAD values averaging 0.1 across the three samples. The electrical conductivity (EC) median values ranged from 0.36 to 4.57 dS/m with RMD averaging 31% of the median. The main variability between laboratories for the majority of the macro cations was closely aligned with measured EC. Cations (K, CA and Mg) concentrations ranged from 17 to 502 mg/L with Ca typically in the greatest concentration. Cation inter-lab precision, based on the RMD ranged from 9-32% across the three substrate samples. The greatest RMD was 31.8% for Ca and 9.2% for K. The Rd values for the cations averaged 5%.

Pada penelitian terdahulu dihasilkan SOP audit internal berdasarkan integrasi standar ISO 9001:2008 dan 14001:2004 sehingga perlu diperbaharui karena perubahan standar ISO dan agar sesuai dengan kondisi dan kebutuhan perusahaan saat ini. Hal ini mendasari perancangan SOP audit internal berdasarkan ISO 9001:2015 Klausul 9.2 dan ISO 14001:2015 Klausul 9.2 yang telah mempertimbangkan risiko melalui metode benchmarking terhadap perusahaan yang telah mengimplementasikan proses audit internal secara continue. Hasil penelitian ini telah terverifikasi memenuhi requirement ISO 9001:2015 dan 14001:2015, sesuai kebutuhan CV XYZ serta telah mengantisipasi risiko kegagalan proses audit internal di CV XYZ. CV XYZ merupakan perusahaan manufaktur yang telah menerapkan Sistem Manajemen Mutu (SMM) sesuai dengan ISO 9001:2008. CV XYZ memiliki beberapa SOP hasil penelitian sebelumnya, salah satunya adalah SOP management review yang telah sesuai dengan integrasi ISO 9001:2008 dan ISO 14001:2004. Perubahan standar pada ISO 9001:2015 dan ISO 14001:2015 menuntut perbaikan SOP sehingga dapat memenuhi requirement baru dan sesuai dengan kebutuhan maupun kondisi perusahaan. Berdasarkan hal tersebut dilakukan perbaikan terhadap SOP hasil penelitian sebelumnya sehingga didapatkan SOP yang memenuhi requirement standar baru. Perancangan SOP dilakukan dengan mengintegrasikan requirement ISO 9001:2015 dan ISO 14001:2015 serta menyusun proses bisnis dengan melakukan benchmark. Proses bisnis yang didapat dibandingkan dengan hasil requirement integrasi sehingga didapatkan proses bisnis sesuai dengan requirement. Berdasarkan ISO 9001:2015 dan ISO 14001:2015 mengenai penerapan risk based thinking, disusun risk register dengan menggunakan risk assessment sebagai bentuk pertimbangan risiko dalam memenuhi requirement tersebut. Penanganan risiko pada risk register menjadi masukan dalam penyusunan SOP management review. Hasil dari penelitian ini adalah SOP management review sesuai dengan requirement integrasi ISO 9001:2015 dan ISO 14001:2015 yang sudah mempertimbangkan risiko. Manfaat dari SOP hasil penelitian ini yaitu CV XYZ dapat menjamin pelaksanaan proses management review karena penyusunan prosesnya berdasarkan hasil benchmark dengan perusahaan yang telah menerapkan proses tersebut secara berkelanjutan. CV Citra Dragon is a small and medium-sized industry that produces agricultural machine tools (alsintan) located in Padang Pariaman, West Sumatra. CV Citra Dragon has been active in production since 1976 and has the potential to increase the production of alsintan in order to compete in and outside the country. Therefore, CV Citra Dragon needs to improve the quality of the products produced. The best way to meet product quality is implementing Quality Management System (QMS). QMS of CV Citra Dragon necessary to expand to other products which is thresher machine. The research based on ISO 9001-2015 standards. The design of QMS on CV Citra Dragon was done through field survey. Documents made on thresher production units are two compulsory and mandatory records, working procedures on thresher production, as well as work instructions and recording forms required. Once a quality document has been created, the analysis was performed by comparing the pre-existing QMS with the most recent QMS tailored to ISO 9001-2015. Therefore, some documents must be obtained by ISO 9001-2015 in accordance with the QMS used CV Citra Dragon was based on the main process approach. The results of documents that have been verified by the company are the scope of the QMS, quality policy, quality objectives, criteria for evaluation and selection of suppliers, product realization planning records, design and development records, product review, and monitoring records and measurement results. The existence of a good quality document is very important for companies to implement QMS.

Hal tersebut terjadi karena kurangnya sosialisasi yang dilakukan oleh perushaan, sehingga mengakibatkan karyawan tidak dapat memahami prosedur-prosedur yang sesuai dengan sistem manajemen mutu yang ada.Oleh sebab itu, untuk mengatasi permasalahan mengenai penerapan sistem manajemen mutu ISO 9001:2015 di perusahaan, maka akan diindentifikasi mengenai permasalahan apa saja yang menyebabkan penerapan tidak dapat berjalan secara penuh. Kemudian faktor-fator tersebut akan dianalisis dengan menggunakan metode Gap Analysis dan Fault Tree Analysis (FTA).Berdasarkan hasil analisis yang telah dilakukan dengan menggunakan dua metode yaitu Gap Analysis dan FTA, maka diketahui bahwa perusahaan telah dapat meningkatkan presentase penerapan Sistem Manajemen Mutu ISO 9001:2015 berdasarkan range presentase peniliaian sebesar 73%-97% dari sebelumnya sebesar 58%-90%.

Namun, standarisasi tersebut masih mencakup secara keseluruhan dan belum ada standarisasi khusus untuk bagian IT. Maka peneliti tertarik untuk menganalisis manajemen risiko pada bagian IT CV. XY. Manajemen risiko adalah suatu proses mengidentifikasi, mengukur risiko, serta membentuk strategi untuk mengelolanya melalui sumber daya yang tersedia.Dalam penilaian risiko, ada beberapa tahapan meliputi identifikasi asset, identifikasi risiko, dan analisis risiko. Dari hasil penelitian, didapatkan hasil bahwa ada kemungkinan – kemungkinan risiko dengan berbagai tingkatan. Maka peneliti menyimpulkan bahwa IT CV. XY belum memenuhi syarat standar ISO 31000 : 2018, dikarenakan dari beberapa tahapan pengamatan, wawancara, serta penilaian terhadap risiko itu sendiri masih banyak temuan risiko yang belum bisa terpecahkan oleh perusahaan.

Meskipun sistem CRM sudah terlaksana, namun belum semua terlaksana dengan baik sesuai dengan konseptual yang ada. Pada penelitian ini, digunakan pertimbangan persyaratan ISO 9001:2015 klausul 8.2.1 komunikasi pelanggan dan konsep CRM untuk mendapatkan hasil analisis Gap yang baik dan lengkap. Metode yang digunakan yaitu business process improvement dengan menggunakan langkah apply techniques improvement. Abstract:
Water stress treatments on some morphological, biomass, physiological, biochemical characteristics and budding success of apricot and plum cultivars/Myrobolan 29 C plum rootstock have been studied in a pot-experiment trial. Three different water stress levels (control-100 %, medium-75 %, and severe-50 % of the pot capacity) were applied. The results showed that measured morphological, biomass, physiological and biochemical characteristics as well as budding success of cultivars on the rootstocks were affected by water stress. Water stress, both moderate and severe, significantly decreased the relative shoot length, diameter and shoot stem cross sectional area, plant total fresh and dry weights. Increases in water stress led to increases in peroxidase activities, phenol and proline contents in plum rootstock. Increases in water stress also resulted in reduction in budding success in both Formosa (plum cv)/Myrobolan 29 C (90.00 % and 48.33 %) and San Castrese (apricot cv)/Myrobolan 29 C (87.50 % and 42.83 %).

Abstract Sodium hydroxide digestion of unhomogenlzed kidney and skeletal muscle for 20 min at 70°C was a superior method for extracting gentamicin from tissues, compared with simple homogenlzatlon, trichloroacetic acid precipitation of homogenized tissue, and sodium hydroxide digestion of homogenized tissue. Fluorescence polarization Immunoassay was used to quantltate gentamicin. Sodium hydroxide digestion of unhomogenlzed tissue allowed for the recovery of 90.0 ± 5.9% (Ẋ ± SD) from renal cortex and 79.9 ± 3.5% from skeletal muscle. The limit of sensitivity was 17.4 ng/g kidney tissue, 15.8 ng/g digested muscle, and 39.0 ng/g digested heart. The wlthin-assay coefficient of variation (CV) at 100 ng/g kidney was 9.2%; at 500 ng/g kidney, the CV was 2.5%; and at 2000 ng/g kidney, the CV was 1.5%. The between-assay coefficient of variation was &lt;7.5% for all concentrations from kidney, and the 99% confidence Interval at 100 ng/g kidney was 71.7-112.4 ng gentamlcin/g kidney. The wlthln-assay coefficient of variation (CV) at 100 ng/g muscle was 15%; at 500 ng/g muscle, the CV was 2.6%; and at 2000 ng/g muscle, the CV was 2.3%. The between-assay coefficient of variation was &lt;15% for all concentrations from muscle, and the 99% confidence interval at 100 ng/g muscle was 72.5-136.8 ng gentamlcin/g muscle. Gentamlcln-free milk could be distinguished from milk containing gentamicin concentrations of 10 ng/mL milk with 95 % confidence, and from milk containing concentrations of 30 ng gentamicin/mL milk with 99% confidence. Quantitative results at or below the tolerance level can be obtained within 90 min of sample acquisition using these extraction and assay methods

The bulb scale explants were treated with 0.1 % HgCl2 (3 min, 4min, 5min, 6 min, 7min, 8min and 9 min) and control (without treatment) were cultured on MS media, among the treatments, 5 minutes timing resulted in minimum contamination [fungal % (6.67), bacterial % (6.67)] and maximum survival % (83.33%). The best surface sterilization time was further taken into consideration for treatment of explants, sterilization and cultured in the MS Basal media supplemented with BAP (0.5, 1.0 mg/l) in combination with 2,4-D (0.5,1.0,1.5,2.0, and 2.5 mg/l) and 2,4-D (0.5,1.0,1.5,2.0, and 2.5 mg/l) alone along with control. Basal media supplemented with BAP (1.0 mg/l) and 2,4-D (1.50 mg/l) produced maximum callus % (90.00%) and spread, profuse green callus was also recorded in similar combination which opened prospects for developing an indirect means of in vitro regeneration of hybrid Lilium Cv. Tresor there by strengthening the way biotechnology which could be used for improvement and satiate the national and international demands of this cut flowers.

The dry matter yields of cultivar trials (from 1976 to 1998 at 15 sites in Finland) of perennial grass sward (meadow fescue (Festuca pratensis) cv. Boris), annual grass sward (Italian ryegrass (Lolium multiflorum), cv. Barmultra and Mitos), spring barley (Hordeum vulgare cv. Otra, Arra, Arve) and oat (Avena sativa cv. Veli) were used to estimate metabolisable energy yields (MEY) by using the feeds metabolisable energy concentration values (MJ/kg DM) from ruminant feed tables. Harvest index (HI) of barley and oat was set to 50%, and straw yields and whole crop cereal silage (WCCS) yields were generated from grain yields accordingly. The MEY in the third year of perennial grass (81.4 GJ/ha) was significantly lower than that in the first (90.0 GJ/ha) and second years (90.7 GJ/ha). However, on average, the one to three year old perennial grass-swards had significantly higher MEY than the annual grass swards (87.7 vs. 83.3 GJ/ha, respectively). The MEYs of perennial and annual grass swards were substantially higher than the MEY of barley grain (52.7 GJ/ha) and oat grain (47. 8 GJ/ha). When the total herbage of cereals, i.e. straw and grain, was used in the calculations, at a ME value of 6.0 MJ/ kg dry matter (DM) for straw, the MEY of barley rose to 75.8 GJ/ha and that of oat to 72.6 GJ/ha. Additionally, the MEY of barley was estimated in the WCCS production situation by converting total herbage to MEY by using ME value 9.9 MJ/kg DM. The MEY of barley in the WCCS calculations was 77.4 GJ/ha, which was significantly lower than the MEY of annual and perennial grass swards. The MEY of barley was a) 60%, b) 86%, and c) 88% of the average MEY of one to three year old perennial grass sward when the MEY of barley was calculated according to a) grain, b) grain + straw, and c) whole crop cereal silage. Perennial grass sward was the most productive of the studied crops in metabolisable energy production for ruminants.

This study established a fully automated computer-aided diagnosis (CAD) system for the classification of malignant and benign masses via breast magnetic resonance imaging (BMRI). A breast segmentation method consisting of a preprocessing step to identify the air-breast interfacing boundary and curve fitting for chest wall line (CWL) segmentation was included in the proposed CAD system. The Chan-Vese (CV) model level set (LS) segmentation method was adopted to segment breast mass and demonstrated sufficiently good segmentation performance. The support vector machine (SVM) classifier with ReliefF feature selection was used to merge the extracted morphological and texture features into a classification score. The accuracy, sensitivity, and specificity measurements for the leave-half-case-out resampling method were 92.3%, 98.2%, and 76.2%, respectively. For the leave-one-case-out resampling method, the measurements were 90.0%, 98.7%, and 73.8%, respectively.

Electrochemical characterization of ascorbic acid oxidation on a graphite screen printed electrode (SPE) modified with La3+/Co3O4nanocubes is performed by applying cyclic voltammetry (CV), chronoamperometry (CHA), and differential pulse voltammetry (DPV) techniques. Synthesized La3+/Co3O4nanocubes for SPE modification, La3+/Co3O4/SPE, enhance the ascorbic acid electrooxidation kinetics by reducing the anodic overpotential. Excellent La3+/Co3O4/SPE electrochemical properties provide sensitive ascorbic acid voltammetric determination with low detection limit, good stability and quick response towards electrooxidation of ascorbic acid as compared to bare SPE. Under optimized conditions, DPV current demonstrates a linear response for ascorbic acid over a concentration range of 1.0 to 900.0 mM with a correlation coefficient of 0.9997, and detection limit (LOD) (S/N = 3) = 0.3 mM. The proposed procedure offers a potential application for producing the sensor with good repeatability. Also, fast response of fabricated sensor can allow a real-time analysis of real samples.

Seventeen strength and power athletes (n = 11 males, 6 females; height: 177.5 ± 7.0 cm, 165.8 ± 11.4 cm; body mass: 90.0 ± 14.1 kg, 66.4 ± 13.9 kg; age: 30.6 ± 10.4 years, 30.8 ± 8.7 years), who regularly performed weightlifting movements during their resistance training programs, were recruited to examine the effect of altering body posture and barbell position on the between-session reliability of force-time characteristics generated in the isometric mid-thigh pull (IMTP). After participants were familiarised with the testing protocol, they undertook two testing sessions which were separated by seven days. In each session, the participants performed three maximal IMTP trials in each of the four testing positions examined, with the order of testing randomized. In each position, no significant differences were found between sessions for all force-time characteristics (p = >0.05). Peak force (PF), time-specific force (F50, F90, F150, F200, F250) and IMP time-bands (0–50, 0–90, 0–150, 0–200, 0–250 ms) were reliable across each of the four testing positions (ICC ≥ 0.7, CV ≤ 15%). Time to peak force, peak RFD, RFD time-bands (0–50, 0–90, 0–150, 0–200, 0–250 ms) and peak IMP were unreliable regardless of the testing position used (ICC = <0.7, CV = >15%). Overall, the use of body postures and barbell positions during the IMTP that do not correspond to the second pull of the clean have no adverse effect on the reliability of the force-time characteristics generated.

Foi utilizado um delineamento inteiramente casualizado em fatorial 4 x 2 (quatro doses de adubação de RRB e duas condições de matéria seca do RRB) com três repetições. A aplicação de doses crescentes de RRB seco ou úmido aumentou de forma linear (p < 0,05), os teores de N, P, K+ e matéria orgânica do solo. As diferentes doses de RRB seco ou úmido aumentaram linearmente (p < 0,05) a BIO seca, os teores de N e PB do capim Xaraés após 60 dias de cultivo, embora se tenha observado comportamento quadrático (p < 0,05) para os teores de N e PB do capim Xaraés após 120 dias de cultivo. Desta maneira, a Urochloa brizantha cv. Xaraés apresenta maior produção de BIO seca, independente da época de corte e da condição de MS do RRB no momento da aplicação.

Rice husk (RH) was used as the precursor for porous carbon preparation applying as electrode materials. Porous carbon was synthesized by pyrolysis of RH under inert gas atmosphere followed by hydrothermal treatment in NaOH to remove the silica fraction and K2CO3 activation to increase porosity of archived carbon. The temperature of RH decomposition was determined by the thermal gravimetric analysis (TGA) method in the nitrogen atmosphere. The performance of porous carbon and C-SiOx were examined using Brunauer-Emmett-Teller (BET) measurement and Scanning electron microscopy (SEM). The electrochemical characterizations of the synthesized materials were analyzed using cyclic voltammetry (CV) and chronopotentiometry techniques. The effect of parameters of hydrothermal and activation steps on the capacity of these porous carbons were researched. The activated porous carbon which obtained by K2CO3 activation at 9000C in 2 hours has high specific surface area (SBET = 769.3 m2/g). Specific capacitance of the as-prepare electrode is 116.2 F.g-1 at scan rate of 5 mV/s. Therefore, as-prepared porous carbon from RH would be proper candidate for application as electrode material.

Background Thyroid nodules diagnosed as 'atypia of undetermined significance/follicular lesion of undetermined significance' (AUS/FLUS) or 'follicular neoplasm/suspected follicular neoplasm' (FN/SFN), according to Bethesda’s classification, represent a challenge in clinical practice. Computerized analysis of nuclear images (CANI) could be a useful tool for these cases. Our aim was to evaluate the ability of CANI to correctly classify AUS/FLUS and FN/SFN thyroid nodules for malignancy. Methods We studied 101 nodules cytologically classified as AUS/FLUS (n = 68) or FN/SFN (n = 33) from 97 thyroidectomy patients. Slides with cytological material were submitted for manual selection and analysis of the follicular cell nuclei for morphometric and texture parameters using ImageJ software. The histologically benign and malignant lesions were compared for such parameters which were then evaluated for the capacity to predict malignancy using the classification and regression trees gini model. The intraclass coefficient of correlation was used to evaluate method reproducibility. Results In AUS/FLUS nodule analysis, the benign and malignant nodules differed for entropy (P < 0.05), while the FN/SFN nodules differed for fractal analysis, coefficient of variation (CV) of roughness, and CV-entropy (P < 0.05). Considering the AUS/FLUS and FN/SFN nodules separately, it correctly classified 90.0 and 100.0% malignant nodules, with a correct global classification of 94.1 and 97%, respectively. We observed that reproducibility was substantially or nearly complete (0.61–0.93) in 10 of the 12 nuclear parameters evaluated. Conclusion CANI demonstrated a high capacity for correctly classifying AUS/FLUS and FN/SFN thyroid nodules for malignancy. This could be a useful method to help increase diagnostic accuracy in the indeterminate thyroid cytology.

Solid polymer blend electrolyte membranes (SPBEM) composed of chitosan and dextran with the incorporation of various amounts of lithium perchlorate (LiClO4) were synthesized. The complexation of the polymer blend electrolytes with the salt was examined using FTIR spectroscopy and X-ray diffraction (XRD). The morphology of the SPBEs was also investigated using field emission scanning electron microscopy (FESEM). The ion transport behavior of the membrane films was measured using impedance spectroscopy. The membrane with highest LiClO4 content was found to exhibit the highest conductivity of 5.16 × 10−3 S/cm. Ionic (ti) and electronic (te) transference numbers for the highest conducting electrolyte were found to be 0.98 and 0.02, respectively. Electrochemical stability was estimated from linear sweep voltammetry and found to be up to ~2.3V for the Li+ ion conducting electrolyte. The only existence of electrical double charging at the surface of electrodes was evidenced from the absence of peaks in cyclic voltammetry (CV) plot. The discharge slope was observed to be almost linear, confirming the capacitive behavior of the EDLC. The performance of synthesized EDLC was studied using CV and charge–discharge techniques. The highest specific capacitance was achieved to be 8.7 F·g−1 at 20th cycle. The efficiency (η) was observed to be at 92.8% and remained constant at 92.0% up to 100 cycles. The EDLC was considered to have a reasonable electrode-electrolyte contact, in which η exceeds 90.0%. It was determined that equivalent series resistance (Resr) is quite low and varies from 150 to 180 Ω over the 100 cycles. Energy density (Ed) was found to be 1.21 Wh·kg−1 at the 1st cycle and then remained stable at 0.86 Wh·kg−1 up to 100 cycles. The interesting observation is that the value of Pd increases back to 685 W·kg−1 up to 80 cycles.

Abstract:
Atualmente, a pressão de preconsolidação (sigmap) tem sido utilizada como uma ferramenta no estudo do processo de compactação do solo. O objetivo deste trabalho foi determinar a configuração e o número adequado de amostras para determinação da sigmap em um Latossolo Vermelho distroférrico, bem como caracterizar sua variabilidade e distribuição espacial, utilizando métodos da Estatística Clássica e da Geoestatística. O estudo foi realizado em uma área experimental, no município de Lavras-MG, sob plantio convencional nos últimos 30 anos, no qual são realizados anualmente uma aração e duas gradagens. A área experimental possui dimensões de 32 x 160 m (5.120 m²), com 68 pontos distanciados a 10 m no eixo X e a 8 m no eixo Y, mais seis transectos, sendo quatro com pontos distanciados de 2 m e dois com pontos distanciados de 1 m, visando detectar variações a pequenas distâncias. Foram coletadas 98 amostras indeformadas na profundidade de 0-3 cm, as quais foram submetidas ao ensaio de compressão uniaxial, para obtenção das sigmap. Por meio dos parâmetros da Estatística Clássica, estabeleceu-se o número adequado de amostras para determinar a sigmap, que foi igual a 10 pontos. A maior variabilidade foi obtida para a sigmap (CV 14,8 %), e a menor, para a umidade do solo (CV 12,0 %). Ambas as variáveis apresentaram distribuição normal, com modelo esférico ajustado, e moderada estrutura de dependência espacial, com alcance de 19,5 e 90,0 m, respectivamente. Em futuras amostragens para determinação da sigmap, em condições similares às da área estudada, sugere-se dispor os pontos de coleta com intervalo igual ao alcance de dependência espacial, visando associar menor esforço de amostragem com maior representatividade da área. Apesar de a razão de dependência espacial encontrada para as variáveis estudadas ter sido moderada, seu alcance deve ser considerado no planejamento de novas amostragens. A declividade do solo influenciou indiretamente os valores de sigmap. O mapeamento da área permitiu observar zonas de maior e menor suscetibilidade à compactação, possibilitando a tomada de decisão sobre onde começar a trafegar para evitar danos à estrutura do solo.

Background. Momentary lapses in concentration contribute to workplace accidents. Given that blood glucose (BG) and hydration levels have been shown to affect vigilance, this study proposed to investigate these parameters and functional movement patterns of ski-resort workers and to determine whether an educational program to stabilize BG and hydration and encourage joint stability had an effect in decreasing occupational injuries.Methods. Seventy-five instructors, patrollers and, lift-operators at five snowsport resorts were evaluated for BG, vigilance, workload, dietary/hydration practices, and functional-movement patterns. Injury rates were tabulated before and after an educational program for nutrition and functional-movement awareness and compared to other resorts.Results. Workers showed poor stability at the lumbar spine, knee, and shoulder. BG levels were normal but variable (%CV = 14±6). Diets were high in sugar and fat and low in water and many nutrients. Medical Aid and Lost Time claims declined significantly by65.1±20.0% (confidence interval −90.0% ≤μ≤−40.2%) in resorts that used the educational program whereas four control resorts not using the program experienced increases of33.4±42.9% (confidence interval −19.7% ≤μ≤−86.7%;F[2,12] = 21.35,P<0.0001) over the same season.Conclusion. Provision of snowsport resort workers with educational programs encouraging hydration, diet to stabilize BG, and functional-movement awareness was effective in reducing worksite injuries in this population.

All the five isolates of Alternaria alternata isolated from different agro climate zone of Rajasthan were tested for their variability in terms of cultural, conidial, pathogenic characteristics and toxin production. All the five isolates differed in cultural characters i.e. dark black colored and very fast mycelial growth with smooth margins (90.00 mm), light black with white at centre and fast growing (80.00 mm), dark brown and medium mycelium growth with smooth margins (75.00 mm), black colored, medium flat mycelial growth with smooth margins (68.00 mm) and white with slightly black in colour with slow mycelial growth (65.00 mm) were observed in Aa-1, Aa-2, Aa-3, Aa-4 and Aa-5 respectively. The variability in conidial morphology of five different isolates was simple, septate, pale to dark brown in colour, often geniculate with one conidial scar. In respect of pathogenic variability, showed significant variations in terms of disease intensity and incubation periods. The isolates Aa-1 was highly pathogenic on Isabgol cv. RI-89 under artificial inoculation conditions showing 52.12% disease intensity followed by Aa- 3 ,Aa-2, Aa-4 and Aa-5 isolates. The variability in toxin production was reflected in terms of time taken in inducing wilting symptoms of Isabgol cuttings. Abstract In vitro and screen house experiments were conducted to investigate the effectiveness of thirteen phytochemicals from Artemisia elegantissimia and A. incisa on root knot nematode, Meloidogyne incognita in tomato (Lycopersicon esculentum L.) cv. Rio Grande. A positive control (Carbofuran) and negative control (H2O) were also used for comparison. Effectiveness of phytochemicals against juveniles (J2s) mortality and egg hatch inhibition were evaluated after 24, 48 and 72 hours of incubation at three concentrations viz; 0.1, 0.2 and 0.3 mg/mL in vitro conditions. Amongst thirteen phytochemicals, Isoscopletin (Coumarin), Carbofuran and Apigenin (Flavonoid) showed the highest mortality and egg hatch inhibition of M. incognita at all intervals. Inhibition of eggs and J2s mortality were the greatest (90.0%) and (96.0%) at 0.3 mg/mL concentration. Application of phytochemicals caused reduction in number of galls, galling index, and egg masses on tomato plant and enhanced plant growth parameters under screen house conditions. Gall numbers (1.50), galling index (1.00), number of juveniles (4.83) and egg masses (4.00) were greatly reduced and plant growth parameters such as; plant height (28.48 cm), fresh (72.13 g) and dry shoot weights (35.99 g), and root fresh (6.58 g) and dry weights (1.43 g) were increased significantly by using Isoscopletin. In structure activity relationship, juveniles of M. incognita, exhibited variations in their shape and postures upon death when exposed to different concentrations of phytochemicals of Artemisia spp. The present study suggests that Artemisia based phytochemicals possess strong nematicidal effects and can be used effectively in an integrated disease management program against root knot nematodes.

Background: Glycyrrhizic Acid (GRA), a potent antioxidant triterpene saponin glycoside and neuroprotective properties exhibits an important role in the treatment of neurological disorders i.e. cerebral ischemia. GRA is water soluble, therefore it’s have low bioavailability in the brain. Objective: To enhance brain bioavailability for intranasally administered Glycyrrhizic Acidencapsulated- chitosan-coated-PCL-Nanoparticles (CS-GRA-PCL-NPs). Methods: Chitosan-coated-PCL-Nanoparticles (CS-PCL-NPs) were developed through double emulsification- solvent evaporation technique and further characterized for particle size, zeta potential, size distribution, encapsulation efficiency as well as in vitro drug release. UPLC triple quadrupole Qtrap MS/MS method was developed to evaluate brain-drug uptake for optimized CS-GRA-PCL-NPs and to determine its pharmacokinetic in rat’s brain as well as plasma. Results: Mean particles size (231.47±7.82), polydispersity index (PDI) i.e. (0.216±0.030) and entrapment efficiency (65.69±5.68) was determined for developed NPs. UPLC triple quadrupole Qtrap MS/MS method study showed a significantly high mucoadhesive potential of CS-GRA-PCL-NPs and least for conventional and homogenized nanoformulation; elution time for GRA and internal standard (IS) Hydrocortisone as 0.37 and 1.94 min at m/z 821.49/113.41 and 363.45/121.40 were observed, respectively. Furthermore, intra and inter-assay (%CV) of 0.49-5.48, %accuracy (90.00-99.09%) as well as a linear dynamic range (10.00 ng/mL -2000.0 ng/mL), was observed. Pharmacokinetic studies in Wistar rat brain exhibited a high AUC0-24 alongwith an amplified Cmax (p** < 0.01) as compared to i.v. treated group. Conclusion: Intranasal administration of developed CS-coated-GRA-loaded-PCL-NPs enhanced the drug bioavailability in rat brain along with successfully UPLC-MS/MS method and thus preparation of GRA-NPs may help treat cerebral ischemia effectively. The toxicity studies performed at the end revealed safe nature of optimized nanoformulation.

Abstract:
A rapid and selective UPLC-DAD method was developed and validated for simultaneous analysis of the novel two-drug combination Darvoni® for the treatment of HCV: Sofosbuvir (SF)/Daclatasvir (DC) in human plasma using Ledipasvir as internal standard (IS) where the extraction process was conducted using automated SPE. Although the analysis of the combination after concomitant oral intake of two tablets of SF and DC individually was reported in literature, yet simultaneous analysis of this new combination in human plasma after a single oral dose was not previously reported. The adopted chromatographic separation was achieved on Waters® Acquity UPLC BEH C18 column (2.1 × 50 mm, 1.7 µm) as a stationary phase using isocratic elution using a mobile phase system of ammonium formate (pH 3.5; 5 mM) and acetonitrile (60:40 v/v) pumped at a flow rate of 0.2 mL.min−1. The UV detection was carried out at 261 nm for SF and 318 nm for DC and IS. SF was eluted at 1.123 min while DC was eluted at 3.179 min. The proposed chromatographic method was validated in accordance with guidelines of FDA for bioanalytical method validation. A linear range was achieved in the range of 25-6400 and 50-12800 ng.mL−1 for SF and DC, respectively. The proposed UPLC-DAD method was found to be accurate with % bias ranging between -10.0-7.2 for SF and -6.9-8.0 for DC. Also it was proved to be precise with % CV for intraday precision ranging between 3.8-9.6 for SF and 2.8-9.2 for DC whereas interday precision ranged between 5.1-9.3 for SF and 3.7-9.1 for DC. Moreover, % extraction recovery ranged between 90.0-107.2 for SF and 93.1-108.0 for DC using the suggested method. The adopted chromatographic method was successfully applied to the therapeutic drug monitoring of SF and DC in healthy volunteers after the oral intake of one Darvoni® tablet.

Appropriate monitoring and control of modifiable risk factors, such as the level of low-density lipoprotein cholesterol (LDL-C) and other types of dyslipidemia, have an important role in the prevention of cardiovascular diseases (CVD). Recently, various nutraceuticals with lipid-lowering effects have gained attention. In addition to the plant-derived bioactive compounds, recent studies suggested that plant cells are able to release small lipoproteic structures named extracellular vesicles (EVs). The interaction between EVs and mammalian cells could lead to beneficial effects through anti-inflammatory and antioxidant activities. The present study aimed to assess the safety of the new patented plant-based product citraVes™, containing extracellular vesicles (EVs) from Citrus limon (L.) Osbeck juice, and to investigate its ability to modulate different CV risk factors in healthy subjects. A cohort of 20 healthy volunteers was recruited in a prospective open-label study. All participants received the supplement in a spray-dried formulation at a stable dose of 1000 mg/day for 3 months. Anthropometric and hematobiochemical parameters were analyzed at the baseline and after the follow-up period of 1 and 3 months. We observed that the supplement has an effect on two key factors of cardiometabolic risk in healthy subjects. A significant change in waist circumference was found in women after 4 (85.4 [79.9, 91.0] cm, p < 0.005) and 12 (85.0 [80.0, 90.0] cm, p < 0.0005) weeks, when compared to the baseline value (87.6 [81.7, 93.6] cm). No difference was found in men (baseline: 100.3 [95.4, 105.2] cm; 4 weeks: 102.0 [95.7, 108.3] cm; 12 weeks: 100.0 [95.3, 104.7] cm). The level of LDL-C was significantly lower at 12 weeks versus 4 weeks (p = 0.0064). Our study evaluated, for the first time, the effects of a natural product containing plant-derived EVs on modifiable risk factors in healthy volunteers. The results support the use of EV extracts to manage cardiometabolic risk factors successfully.

In our randomized controlled trial, we investigated whether Wu-style Tai Chi (Tai Chi combined with Daoyin) as a potential exercise prescription is more effective than simplified Tai Chi in the prevention and treatment of hypertension and hyperlipidemia in the middle-aged and elderly. We randomly assigned 66 patients with hypertension and hyperlipidemia to one of the two groups: the Wu-style Tai Chi group or the simplified Tai Chi group; the simplified Tai Chi group only exercised simplified Tai Chi three times a week for 6 weeks. The Wu-style Tai Chi group participated in 60 min of Wu-style Tai Chi three times a week for 6 weeks. Serum biochemical tests were conducted at baseline and at the end of the study. Measurements of blood pressure were performed at the same time. Primary outcomes were compared within and between groups at baseline and at 6 weeks. The participants in the Wu-style Tai Chi group performed, at 6 weeks, significantly better than baseline on all of the primary outcomes (p value ≤ 0.05). The results also show significant difference within the simplified Tai Chi group from baseline to 6 weeks in TCHO (mmol/L), SBP (mmHg), and LDL-C (mmol/L) (p value < 0.05). From baseline to 6 weeks, the Wu-style Tai Chi group had significant differences at more test indexes in serum and blood pressure than the simplified Tai Chi group. At 6 weeks, the Wu-style Tai Chi group had a significantly greater mean improvement in the SBP (mmHg) than did the simplified Tai Chi group (mean between-group difference, −5.80 (mmHg) [95% CI, −14.01 to 2.41]; p = 0.007). The results showed that, compared with simplified Tai Chi, Wu-style Tai Chi had a better effect on hypertension in the middle-aged and elderly. At 6 weeks in LDL-C (mmol/L), the Wu-style Tai Chi group had significantly greater improvement between the two groups (means between-group difference, −0.45 (mmol/L) [95% CI, −0.89 to −0.17]; p = 0.03). The results showed that Wu-style Tai Chi protected the cardiovascular system of the middle-aged and elderly in improving LDL-C (mmol/L), and was more significant than simplified Tai Chi. After 6 weeks of exercise, Wu-style Tai Chi could effectively improve hyperlipidemia and hypertension. The total effective rate of cardiovascular disease was 90.00%. There was significant difference in the treatment effect of hypertension and hyperlipidemia between the two groups during 6 weeks (p = 0.039), showing that, in a small population of middle-aged and elderly subjects, Wu style Tai Chi could be useful in managing important CV risk factors, such as hypertension and hyperlipidemia.

Abstract:
In May of 2006, samples from tomato plants (Solanum lycopersicum cv. Known-you 301) exhibiting necrotic symptoms on stems, petioles, and leaves were collected from Chiayi County, Taiwan. Double-antibody sandwich-ELISAs were performed using Cucumber mosaic virus, Tomato mosaic virus, Potato virus Y, Watermelon silver mottle virus, and Chilli veinal mottle virus (ChiVMV) polyclonal antibodies. Three of eight samples reacted with antibodies against ChiVMV but not with the others. Using the potyvirus degenerate primers (Hrp 5/Pot 1) (2), an expected 1.5-kb DNA fragment including the 3′-end of the NIb gene, the complete coat protein (CP) gene, and the 3′-nontranslatable region of the virus was amplified from total RNA isolated from these three samples by reverse transcription (RT)-PCR. A homology search in GenBank indicated that the new tomato-infecting virus in Taiwan belongs to Pepper veinal mottle virus (PVMV) since they shared >90% amino acid identity in the CP gene. A virus culture (Tom1) isolated from one of the diseased tomatoes was then established in Chenopodium quinoa and Nicotiana benthamiana and the CP gene was amplified and sequenced (GenBank Accession No. EU719647). Comparisons of the 807-nt CP gene with those of five PVMV isolates available in GenBank showed 81.5 to 93.1% nucleotide and 90.0 to 97.8% amino acid identity. Tom1 induced irregular necrotic lesions on stems, petioles, and leaves of tomato while inducing only mild mottle symptoms on pepper. Serological cross reaction between ChiVMV and PVMV has been observed previously (1,3) and also found in this study. To differentiate these two potyviruses by RT-PCR, primer pair CPVMVup/dw (5′-TATTC(T/C)TCAGTGTGG(A/T/C)T(T/C)CCACCAT and 5′-(T/C)C(A/T)C(A/T)(A/T/G)(A/T)AA(A/G)CCATAA(A/C)(A/C)ATA(A/G)T(T/C)T) was designed on the basis of the comparison of the CP gene and the 3′-nontranslatable region of the PVMV and ChiVMV. DNA fragments of 171 and 259 bp are expected to be amplified from ChiVMV and PVMV, respectively, by RT-PCR with primers CPVMVup/dw. In a field survey done in 2006, samples from diseased peppers (Capsicum annuum) that reacted with the polyclonal antibodies against ChiVMV were further identified by RT-PCR with primers CPVMVup/dw, indicating that both ChiVMV and PVMV infected pepper crops (Capsicum spp.) in Taiwan. A pepper isolate (Pep1) of PVMV was obtained from Nantou County through three times of single lesion passages on C. quinoa and then propagated on N. benthamiana. The CP gene of Pep1 was amplified and sequenced (GenBank Accession No. EU719646) and found to share 99.1% nucleotide and 100% amino acid identity with that of Tom1. Pep1 caused mild mottle symptoms on leaves of both tomato and pepper. To our knowledge, this is the first report of the presence of PVMV in Taiwan as well as in East Asia. References: (1) B.
