Full textIn this work, we examine the reactivity of unsaturated alcohols and acids with bis (collidine) iodo (i) and bromo (i) hexafluorophosphate to obtain 8-membered ethers by halo cyclizations. In the first chapter, we study the influence resulting of the introduction of a cyclopropane ring in the carbon-chain of alcohols, if an ethylenic bond was present in terminal position. We then examine the haloetherification reaction using the iodo and bromo reagents. These reactions were carried out in ch2cl2 at rt. Substrats were added to the halo reagents. Products were purified by silica gel liquid chromatography, and their structures were established from their nmr and ir spectra. Usually, oxocanes were obtained with yields in the range 11-80%. In the particular case of the vinyl cyclopropane alcohol, the reaction leads to the formation of a tetrahydropyran derivative. Higher yields were obtained using the iodo reagent compare to the bromo reagent. In the second chapter, results were applied to the synthesis of natural products such as arenarans a and b. Our results show the limite of electrophilic cyclizations. In deed, the key step of our synthesis, i-e the formation of the oxocane failed, propably for steric hindrance. We examine different accesses to aplyparvanine. Aplyparvanine possessing interesting biological properties. We prepared different mono or di substituted alcohols and acids and study their reaction with hbi and hbb. We found than yields in oxocanes were low if a substituent was present in of the carbon-carbon double bond. However, substitution of the double bond in terminal position does not hinder the cyclization

Full textThe aim of this thesis is the synthetic applications of two peculiar cyclopropanols : l) The l-ethoxycyclopropanol, is converted into propargylic cyclopropanols upon treatment with acetylenic magnesium bromide or lithium. Hydride reduction and O-silylation lead to l-siloxy l-vinylcyclopropanes which undergo thermal C3 ---> C5 ring enlargement into l-siloxycyclopentenes ; then , these enol ethers are regiospecifically alkylated into 2, 3-disubstituted cyclopentanones. This scheme is illustrated by the total synthesis of ± ll-deoxyprostaglandin E₂ methyl ester. 2) The l-hydroxycyclopropanecarboxaldehyde tetrahydropyranyl and silylated ethers provide l-siloxy-l-vinylcyclopropanes which, after thermal rearrangement lead directly to 2,3-disubstituted cyclopentanones and cyclopentenones upon hydrolysis or dehydrosilylation, so avoiding the quite delicate enol ether alkylation. Dicranenones, new fatty acids structurally similar to prostanoids and jasmanoids, having antimicrobial activity, are prepared from this new synthon

Full textThis thesis is devoted to inter- and intramolecular Kulinkovich cyclopropanation on esters with secondary Grignard reagents or terminal alkenes in the presence of titanium(IV) tetraisopropoxide. In the first part, we studied the reactivity of various cycloalkylmagnesium bromides (from three to seven-membered ring sizes) with ethyl 3-chloropropionate promoted by Ti(Oi-Pr)4. We obtained 1,2,3-trisubstituted bicyclic cyclopropanols in moderate yields, and showed these Grignard reagents (except for cyclopropyl- and 3-cyclohexenyl bromides) could be cyclopropanated. The interest of this strategy was exemplified by the synthesis of the unknown exo-8-aminobicyclo[5. 1. 0]octane-8-carboxylic acid. In the second part, we developed a general method providing an easy access to medium-ring oxacycloalkenones. The synthetic protocol consists of the intramolecular Kulinkovich cyclopropanation on oxa-esters bearing a terminal carbon-carbon double bond, followed by Saegusa oxidation and base-induced dehydrochlorination. We obtained 7-, 8- and 9-membered oxacycloalkenones in good yields. In the third part, we applied previous methodology to the formal synthesis of (±)-heliannuols A, K and L. The later compounds were obtained in 8 and 10 steps in 14% and 12% overall yields, respectively. The extension to the enantioselective synthesis of these allelopathic compounds is under current investigation
