External stimuli affect PGC-1α-regulation which in turn adapts cellular signals to meet them. For example, conditions like fasting and diabetes mellitus (DM) are known to activate PGC-1α expression in the liver, resulting in enhanced de novo glucose production, gluconeogenesis. Moreover, the hypercalcaemia associated with an elevated dose of calcitriol does not allow the use of vitamin D at a high concentration. Abstract:
The relevant literature concerning nonalcoholic steatohepatitis (NASH), the morphologically similar condition of alcoholic liver disease (ALD), and the hepatic cytochromes P450 (P450 or CYP) was reviewed with particular emphasis on CYP2E1. CYP2E1 is induced in ALD. It plays an important role in the pathogenesis of this condition by generating reactive oxygen species (ROS). In turn, ROS produce lipid peroxidation, which contributes to the cellular injury in ALD. CYP2E1 is constitutively expressed in acinar zone 3. The early lesions observed in both ALD and NASH are most pronounced in the

Abstract:
Changes in extracellular Ca2+ (Ca2+o) differentially modulate 25-hydroxyvitamin D3 1α-hydroxylase (1αOHase; encoded by CYP27B1) mRNA and protein levels in cell types including the renal proximal tubule (inhibitory), parathyroid, and skeletal osteoblasts (stimulatory) to control 1,25-dihydroxyvitamin D3 synthesis. We hypothesised that the calcium-sensing receptor (CaSR) mediated Ca2+o concentration-dependent control of 1αOHase, either directly through Ca2+o, or through the local production of parathyroid hormone related peptide (PTHrP).
