Full textA epilepsia é uma doença que se manifesta por crises epilépticas recorrentes, não provocadas. Evidências sugerem que a inflamação desempenha um papel na patofisiologia destas crises. Embora os leucotrienos cisteínicos (CysLTs) tenham sido implicados no desenvolvimento de crises convulsivas, nenhum estudo investigou se o bloqueio dos receptores CysLT1 potencializa a ação anticonvulsivante de antiepilépticos clássicos, assim como se a expressão dos receptores de CysLT é alterada por inflamação. Neste estudo mostramos que o agonista inverso de CysLT1, montelucaste, sinergicamente aumenta a ação anticonvulsivante do fenobarbital contra crises convulsivas induzidas em um modelo de injeção aguda de pentilenotetrazol (PTZ). Além disso, é mostrado que o LTD4 (leucotrieno D4) previne o efeito do montelucaste. A análise isobolográfica revelou que o valor de DE50 mix, calculado experimentalmente para uma combinação de proporção 1: 1 de montelucaste e fenobarbital foi de 0,06 ± 0,02 umol, ao passo que o valor de DE50 add, calculado foi de 0,49 ± 0,03 umol. O índice de interação encontrado foi de 0,12, indicando uma interação sinérgica. A associação dos fármacos diminuiu significativamente o DE50 para o efeito anticonvulsivante do fenobarbital de 0,74 para 0,04 umol (na ausência e na presença de montelucaste, respectivamente) e, consequentemente, a sedação induzida por fenobarbital em doses equieficazes. Posteriormente foi avaliado se o montelucaste e o fenobarbital diminuem as crises convulsivas em animais previamente abrasados, assim como se o tratamento farmacológico ou o abrasamento alteram a expressão de receptores CysLTs. O montelucaste (10 mg/kg; s.c.) e o fenobarbital (20 mg/kg, s.c.) aumentaram a latência para crises convulsivas generalizadas em camundongos abrasados. O montelucaste aumentou a imunorreatividade do receptor CysLT1 em camundongos não abrasados e que foram desafiados por PTZ que não foram abrasados. Entretanto, o desafio de PTZ diminuiu a imunorreatividade do receptor CysLT2 apenas em camundongos abrasados. Antagonistas do receptor CysLT1 parecem emergir como agentes terapêuticos adjuntos promissores no tratamento de crises refratárias. Não obstante, estudos adicionais são necessários para avaliar as implicações clínicas deste trabalho.

Full textCysteines 172 and 192 from the large subunit of the photosynthetic enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) are a pair of vicinal residues evolutively conserved among cyanobacteria, algae and higher plants. In the present study, it has been analyzed the effect of the substitution of Cys172 and Cys192 by serine on the catalytic properties, thermostability, three-dimensional structure of the mutant enzymes, and the in vivo turnover and inactivation of Rubisco enzymes in stressed cells.1. Site-directed mutagenesis of conserved cysteine residues (Cys172 and Cys192) of Rubisco in Chlamydomonas reinhardtii and Rubisco mutants characterization.The most remarkable effect of the C172S mutant was a 15% increase in the value of the specificity factor compared to the wild-type enzyme, while the specificity factor of C192S Rubisco was identical to wild-type.The C192S enzyme was more sensitive to inactivation through oxidation of cysteines than the wild type and the rest of the enzymes. The results suggest that the Cys192 protects the critical modification of the Cys172. The results of thermal stability, proteolytic susceptibility assays and holoenzyme mobility in native electrophoresis show that the substitution of Cys172 and/or Cys192 affect to the enzyme structure. 2. Crystallization and Structure determination of C172S and C192S Rubisco mutants.X-ray structures of the mutant enzymes reveal that the substitution at position 172 causes a significant shift of the main chain backbone atoms of β-strand1 of the α/β-barrel.3. In Vivo turnover and inactivation of the C172S, C192S, C172S/C192S and wild-type Rubisco Enzymes in stressed Cells.The absence of Cys172 protects Rubisco from degradation under saline stress conditions. The C172S mutant enzyme is more resistant to inactivation under oxidative (diamide induced) stress.In a oxidative conditions (diamide stress) Cys172-Cys192 disulfide bond formation could act as a redox signaling pathway.

Full textThe radicular cysts (RCs) and dentigerous (DCs), despite having different etiologies, form a pathological cavity lined by epithelium, which grows due to the buildup of fluid inside, as the surrounding bone is reabsorbed and the epithelium will being induced to proliferate. The epithelial proliferation, which has been identified as one of the key processes in the growth of odontogenic cystic lesions, is influenced by growth factors such as EGFR (epidermal growth receptor factor) and podoplanin (PDPN), many of which may have its production stimulated mainly during inflammatory processes. The objective of this research was to evaluate and compare the immunohistochemical expression of EGFR and PDPN in 30 cases of RCs and 30 cases of DCs, semiquantitatively, in light microscopy, associating it with the degree of inflammation, cellular localization of immunostaining and with the immunostained epithelial layers. Data were statistically analyzed by Chi-square test and Fisher exact test, considering a significance level of 5 %. The results showed high immunoreactivity of both proteins in the lesions studied, only statistically significant difference was observed in immunostaining of PDPN (p=0.033), which proved higher in RCs. The other analyzed parameters showed no relevant significant differences. We conclude that, as EGFR and PDPN showed high immunoreactivity in cystic lesions analyzed, these proteins participate the pathogenesis of these lesions through the epithelial stimulation process, despite having different etiologies. Furthermore, it can infer that the higher immunostaining of PDNP in RCs that DCs showed no distinction indicator between the two lesions, regarding their etiologies, once this protein also showed a considerable expression in DCs, independent of the intensity of the inflammatory infiltrate
Os cistos radiculares (CRs) e dent?geros (CDs), apesar de possu?rem etiologias diferentes, formam uma cavidade patol?gica revestida por epit?lio, a qual cresce em fun??o do ac?mulo de l?quido em seu interior, ? medida que o osso ao redor ? reabsorvido e o epit?lio vai sendo induzido a proliferar. A prolifera??o epitelial, que tem sido apontada como um dos processos determinantes no crescimento das les?es c?sticas odontog?nicas, ? influenciada por fatores de crescimento como o EGFR (receptor do fator de crescimento epid?rmico) e a podoplanina (PDPN), muitos dos quais podem ter sua produ??o estimulada principalmente durante processos inflamat?rios. O objetivo desta pesquisa foi avaliar e comparar a express?o imunoistoqu?mica do EGFR e da PDPN em 30 casos de CRs e 30 casos de CDs, de forma semiquantitativa, em microscopia de luz, associando-a com o grau de inflama??o, localiza??o celular da imunocolora??o e com as camadas epiteliais imunomarcadas. Os dados foram avaliados estatisticamente por meio de testes do Qui-quadrado e Exato de Fisher, considerando-se um n?vel de signific?ncia de 5%. Os resultados mostraram que houve elevada imunorreatividade das duas prote?nas nas les?es estudadas, sendo observada apenas diferen?a estat?stica significativa na imunoexpress?o da PDPN (p=0,033), que se mostrou mais elevada nos CRs. Os demais par?metros analisados n?o demonstraram diferen?as significativas relevantes. Conclui-se que, como o EGFR e a PDPN apresentaram elevada imunoexpress?o nas les?es c?sticas analisadas, essas prote?nas participam da patog?nese dessas les?es atrav?s da estimula??o epitelial, apesar de apresentarem etiologias diferentes. Al?m disso, pode-se inferir que a maior imunomarca??o da PDPN em CRs do que em CDs n?o se mostrou indicador de distin??o entre as duas les?es, com rela??o ?s suas etiologias, uma vez que nestes ?ltimos essa prote?na tamb?m apresentou express?o consider?vel, independente da intensidade do infiltrado inflamat?rio

Full textAneurysmal Bone Cysts (ABC) and Solitary Bone Cysts (SBC), both non-epithelial cysts of the jaws are defined as benign lesions of an unclear aetiology. There is limited literature available on these two primary non-epithelial cysts of the jaws, especially in African populations. This retrospective study focused on the clinical and radiographic features, as well as management of the non-epithelial cysts of the jaws presenting at the University of the Western Cape Oral Health Centre from 1970-2018. The aim of this study was to describe the clinical and radiological features of non-epithelial cysts of the jaws that presented at the Departments of Maxillo-Facial and Oral Surgery and Diagnostics and Radiology at the University of the Western Cape Oral Health Centre as well as their management and recurrence patterns.

Full textGrowing evidence has shown that leukotrienes are important contributors in the pathophysiology of several SNC inflammatory diseases where excitotoxicity is involved, including traumatic brain injury, encephalitis, Parkinson's disease, ischemia, epilepsy and neuropathic pain. However little is known about the molecular mechanism by which leukotrienes facilitate excitatory activity in the brain. Thus, in this study we investigated the effect of antagonists for cysteinyl leukotrienes receptors (CysLT) on PTZ-induced seizure in mice. Bay-u9973 (3 and 30 nmol), montelukast (0.03 and 0.3 μmol) and pranlukast (1 and 3 μmol), increased the latency to generalized seizures and decreased the mean amplitude of EEG recordings during seizures. LTD4 (0.2 and 2 pmol) reverted the anticonvulsant effect of montelukast (0.3 μmol). Montelukast (0.03 and 0.3 μmol) prevented PTZ-induced BBB disruption, an effect that was reversed by LTD4 (6 pmol). In addition, doses of LTD4 (0.2 and 2 pmol) which reversed the effect of montelukast in crisis did not alter the protective effect of montelukast on the barrier, dissociating the anticonvulsant of protective effect on BBB. The confocal microscopy analysis revealed that 1. PTZ increased the number of CD45+ and IgG cells in cerebral cortex, indicating BBB leakage with leukocyte infiltration; 2. while LTD4 (6 pmol) potentiated, montelukast decreased the effect of PTZ on leukocyte migration. Considering that increase levels of leukotrienes and decrease in Na+,K+-ATPase are common findings in several excitotoxic conditions, including epileptic seizures, we also investigated the effects of LTD4 on the activity of Na+,K+-ATPase activity in mice hippocampal slices. LTD4 10 and 100 nM decreases Na+,K+-ATPase activity alpha 2, 3 and alpha 1 subunits, respectively, in mice hippocampal slices. The inhibitory effect of LTD4 on Na+,K+-ATPase activity was not observed in hippocampal homogenates, indicating that it requires intact cells. Moreover, we showed that LTD4-induced decrease Na+,K+-ATPase activity was reversed by CysLT1R inverse agonis, montelukast (1 μM). In addition, we also showed that possibly the PKC activation pathway is involved in LTD4-induced decrease of Na+,K+-ATPase activity in mice hippocampal slices, since PKC inhibitor, GF 109203X (0,3 μM), prevent this effect. Finally, but not least important, we have demonstrated that animals injected with LTD4 (2 pmol/3 μL icv), there also occurs a decrease in Na+,K+-ATPase activity, corroborating our in vitro findings and confirming the biological importance of this work. In summary, we showed that CysLT1 receptor activation modulates hippocampal Na+,K+-ATPase activity in mice, suggesting a possible mechanism for the involvement of leukotrienes in several dosorders related with brain inflammation and hyperexcitability.
Evidências crescentes têm mostrado que os leucotrienos são importantes contribuintes na patofisiologia de diversas doenças inflamatórias do sistema nervoso central nas quais a excitotoxicidade esteja envolvida, incluindo trauma crânio-encefálico, encefalite, doença de Parkinson, isquemia, dor neuropática e epilepsia. Entretanto pouco se sabe sobre o mecanismo molecular pelo qual os leucotrienos facilitam a atividade excitatória no encéfalo. Assim, neste trabalho investigamos o efeito de antagonistas para receptores de leucotrienos cisteínicos (CysLT) sobre as convulsões induzidas por PTZ em camundongos. Bay-u9973 (3 and 30 nmol), montelucaste (0.03 and 0.3 μmol) e pranlucaste (1 and 3 μmol), aumentaram a latência para as crises e diminuíram a amplitude média do EEG durante as crises. Montelucaste (0.03 and 0.3 μmol) preveniu a ruptura da BHE induzida pelo PTZ, e o efeito foi revertido pelo LTD4 (6 pmol). Além disso, as doses de LTD4 (0.2 and 2 pmol) que reverteram o efeito do montelucaste nas crises não alteraram o efeito protetor do montelucaste sobre a barreira, dissociando o efeito anticonvulsivante do efeito protetor sobre a BHE. As análises de microscopia confocal revelaram: 1) PTZ aumentou o número de células CD45+ e IgG no córtex, evidenciando a ruptura da BHE; 2) enquanto o LTD4 (6 pmol) potencializou, o montelucaste diminuiu o efeito do PTZ sobre a migração leucocitária. Considerando que níveis aumentados de leucotrienos e diminuição na atividade da Na+,K+-ATPase são achados comuns em diversas condições excitotóxicas, incluindo crises epilépticas, também investigamos o efeito do LTD4 sobre a atividade da Na+,K+- ATPase em fatias de hipocampo de camundongos. LTD4 nas doses de 10 e 100 nM, diminuiu a atividade das subunidades alfa 2/3 e alfa 1, respectivamente. O efeito inibitório do LTD4 na atividade da Na+,K+-ATPase não foi reproduzido quando realizado com homogeneizado de hipocampo, indicando que esse efeito requer a célula intacta. A fim de nos certificarmos de que o LTD4 (10 nM) estava se ligando ao receptor CysLT1, incubamos as fatias com anticorpo anti- CysLT1, e verificamos que, na presença do anticorpo, o LTD4 perde o efeito. Além disso, observamos que a diminuição na atividade da Na+,K+-ATPase induzida pelo LTD4 foi revertida pelo montelucaste (1 μM), agonista inverso dos receptores CysLT1. Neste trabalho mostramos ainda que a ativação da PKC possivelmente esteja envolvida no efeito do LTD4 sobre a atividade da Na+,K+-ATPase em fatias de hipocampo de camundongos, uma vez que o GF 109203X (0,3 μM), inibidor da PKC, preveniu esse efeito. Por fim, mas não menos importante, também demonstramos que em animais injetados i.c.v. com LTD4 (2 pmol/3 μL, i.c.v.), também ocorre uma diminuição na atividade da Na+,K+-ATPase, corroborando com nossos achados in vitro e confirmando a importância biológica deste trabalho. Assim, este trabalho mostrou evidências do envolvimento dos receptores CysLT1 nas crises induzidas por PTZ bem como na permeabilidade da BHE, sendo a modulação da enzima Na+,K+-ATPase um possível mecanismo para a implicação dos leucotrienos em diversas doenças do SNC relacionadas com inflamação e hiperexcitabilidade.

Full textDenna studie handlar om biodiversiteten och den biologiska affiniteten av mikrofossil från den neoproterozoiska eran, tonianperioden (1000-720 Ma). De har extraherats från övre formationen av Visingsögruppen i södra Sverige.Mikrofossilen har organiska cellväggar, är encelliga och har förmodats representera algcystor (resistenta reproduktiva strukturer), cyanobakterier, och andra organismer av okänd tillhörighet. Neoproterozoikum har den högsta graden av biologisk diversitet under prekambrium. Det är därför viktigt att studera diversiteten för att förstå utvecklingen av biosfären under denna period i samband med utvecklingen av miljöer. Den studerade samlingen härrör från ett borrhål på Visingsö i Vättern, och visar på större diversitet än från tidigare studier.Denna nya studie syftar till att bestämma biodiversiteten i den övre formationen av Visingsögruppen och att känna igen affiniteten av mikrofossilen med organiska väggar och deras ekologi. Vissa av de undersökta mikrofossilen hör sannolikt till grönalgerna. Kluster och fiber av cyanobakterier är rikligt förekommande, och några prover är ej biologiskt igenkännbara. Med hjälp av moderna analoger och sedimentologiska data är ekologin hos dessa mikrofossil utläs

Full textO desenvolvimento de qualquer investigação pressupõe a existência de algo que nos inquieta e nos induz curiosidade e este trabalho não constitui uma exceção. Assim sendo, formulou-se como objetivo de investigação, a análise de alguns artigos científicos sobre o tema das “lesões quisticas da mandíbula e da maxila”. O presente estudo reporta-se a uma revisão sistemática de literatura, como metodologia de investigação. Teve como objetivo descrever o conhecimento produzido acerca das lesões quisticas da mandíbula e da maxila. Foi utilizado o método de PICOD para a análise, avaliação e síntese da evidência empírica a incluir neste estudo. Os achados finais resultam da análise de 22 artigos, quer de perfil qualitativo, quer do perfil quantitativo, dos quais são todos estudos primários. Foram encontrados 4 temas centrais: lesões quisticas, quistos odontogénicos da mandibula e maxila, quistos de desenvolvimento, etiopatogenia. As principais conclusões do estudo evidenciaram no seu conjunto que existe um conjunto de lesões, tanto ao nível da mandíbula como a maxila que diariamente os profissionais de medicina dentária se confrontam. Cada uma destas patologias apresenta a sua própria complexidade e sinais clínicos, radiológicos e histológicos. The development of any research presupposes the existence of something that disturbs us and induces us curiosity and this work is no exception. Thus was formulated as objective research, analysis of some scientific papers on the topic of "cystic lesions of the mandible and maxilla" articles. The present study refers to a systematic literature review as a research method. Aimed to describe the knowledge produced about cystic lesions of the mandible and maxilla. PICOD the method for the analysis, evaluation and synthesis of empirical evidence to include in this study was used. The final findings resulting from analysis of 22 articles, either qualitative profile of either the quantitative profile of which is all primary studies. Four central themes were identified: cystic lesions, odontogenic cysts of jaw and jaw cysts development, pathogenesis. The main findings of the study showed that there is a whole set of injuries, both at the jaw the daily dental professionals are confronted level of the jaw. Each of these conditions has its own complexity and clinical, radiological and histological signs.
