Abstract In this paper, we study the boundedness and compactness of the commutator of Calderón– Zygmund operators T on spaces of homogeneous type (X, d, µ) in the sense of Coifman and Weiss. More precisely, we show that the commutator [b, T] is bounded on the weighted Morrey space L ω p , k ( X ) L_\omega ^{p,k}\left( X \right) with κ ∈ (0, 1) and ω ∈ Ap (X), 1 < p < ∞, if and only if b is in the BMO space. We also prove that the commutator [b, T] is compact on the same weighted Morrey space if and only if b belongs to the VMO space. We note that there is no extra assumptions on the quasimetric d and the doubling measure µ.

We show that we have a ‘stronger’ spatial determinism forZ2-actions: to determine the completeZ2-name of a point, it is enough to know the name of a fraction of the orbit whose density can be made arbitrarily small. Fast transport of axonal vesicles and organelles is a microtubule-associated movement (Griffin, J. W., K. E. Fahnestock, L. Price, and P. N. Hoffman, 1983, J. Neuroscience, 3:557-566; Schnapp, B. J., R. D. Vale, M. P. Sheetz, and T. S. Reese, 1984, Cell, 40:455-462; Allen, R. D., D. G. Weiss, J. H. Hayden, D. T. Brown, H. Fujiwake, and M. Simpson, 1985, J. Cell Biol., 100:1736-1752). Proteins that mediate the interactions of axoplasmic vesicles and microtubules were studied using stable complexes of microtubules and vesicles (MtVC). These complexes formed spontaneously in vitro when taxol-stabilized microtubules were mixed with sonically disrupted axoplasm from the giant axon of the squid Loligo pealei. The isolated MtVCs contain a distinct subset of axoplasmic proteins, and are composed primarily of microtubules and attached membranous vesicles. The MtVC also contains nonmitochondrial ATPase activity. The binding of one high molecular mass polypeptide to the complex is significantly enhanced by ATP or adenyl imidodiphosphate. All of the axoplasmic proteins and ATPase activity that bind to microtubules are found in macromolecular complexes and appear to be vesicle-associated. These data allow the identification of several vesicle-associated proteins of the squid giant axon and suggest that one or more of these polypeptides mediates vesicle binding to microtubules.

ABSTRACT It is not clear how the host initially recognizes and responds to infection by gram-negative pathogenic Brucella spp. It was previously shown (D. S. Weiss, B. Raupach, K. Takeda, S. Akira, and A. Zychlinsky, J. Immunol. 172:4463-4469, 2004) that the early macrophage response against gram-negative bacteria is mediated by Toll-like receptor 4 (TLR4), which signals in response to lipopolysaccharide (LPS). Brucella, however, has a noncanonical LPS which does not have potent immunostimulatory activity. We evaluated the kinetics of TLR4 activation and the cytokine response in murine macrophages after Brucella infection. We found that during infection of macrophages, Brucella avoids activation of TLR4 at 6 h but activates TLR4, TLR2, and myeloid differentiation factor 88 (MyD88) at 24 h postinfection. Interestingly, even though its activation is delayed, MyD88 is important for host defense against Brucella infection in vivo, since MyD88−/− mice do not clear the bacteria as efficiently as wild-type, TLR4−/−, TLR2−/−, or TLR4/TLR2−/− mice.

The cDNA (termed FCa) comprised 3,239 base pairs, including an open reading frame encoding 994 amino acids which showed the highest degree of homology with the adult rabbit fast-twitch Ca2+-ATPase isoform (C. J. Brandl, S. de Leon, D. R. Martin, and D. H. MacLennan, J. Biol. Chem. 262:3768-3774, 1987). Radiolabeled FCa hybridized to a 3.2-kilobase transcript in chicken skeletal muscle RNA but not to cardiac muscle RNA, which confirmed its identity as encoding the fast Ca2+-ATPase isoenzyme. FCa was transfected into the mouse myogenic line C2C12, from which a protein of 100 kilodaltons was immunopurified by using a monoclonal antibody specific for the avian fast Ca2+-ATPase. Immunofluorescence microscopy of a line (designated C2FCa2) stably expressing the avian Ca2+-ATPase localized the protein to the nuclear envelope and a population of cytoplasmic vesicles. A similar pattern was observed when C2FCa2 cells were stained with DiOC6(3), a cyanine dye that labels endoplasmic reticulum and mitochondria (M. Terasaki, J. Song, J. R. Wong, M. J. Weiss, and L. B. Internal consistencies and convergent validities of (a) the Hopkins Symptom Checklist-25 (HSCL-25; Mollica, Wyshak, deMarneffe, Khuon, & Lavelle, 1987), (b) the Harvard Trauma Questionnaire (HTQ; Mollica et al., 1992), (c) the Impact of Event Scale (IES-R; Weiss & Marmar, 1997), (d) the Bradford Somatic Inventory (BSI; Mumford et al., 1991), (e) the Clinician Administered PTSD Scale (CAPS-1; Blake et al., 1990), and (f) the Social Adaptation Self-Evaluation Scale (SASS; Bosc, Dubini, & Polin, 1997), were determined in 150 asylum seekers from Chechnya, Afghanistan, and West Africa using diagnostic interviews as a criterion. For all the instruments, except the SASS, estimates of internal consistency were satisfactory. When used as a continuous measure of symptom frequency and intensity, the CAPS-1 had excellent convergent validity for all three ethnic groups, while formal PTSD criteria yielded high numbers of false negative diagnoses. Among the questionnaires, for Chechnyans the HTQ, and for Afghans and West Africans the IES-R achieved best results with respect to convergent validity.

NaNiO2 was prepared through oxidation of a Na2NiO2/ NiO mixture (1:1) in dried oxygen at 500 °C. Single crystals have been grown by annealing of NaNiO2 powder at 600 °C for 83 d in a flow of dried oxygen. According to the X-ray analysis of the crystal structure (C2/m, Z = 2, a = 5.3177(2), b = 2.8458(1), c = 5.5819(3) Å , β = 110.409(2)°, R1(all) = 3.4%, 185 independent reflections), the Jahn-Teller distorted NiO6 octahedra, sharing edges, build up layers lying parallel to the ab plane. A phase transition (associated with an energy of 2.9 kJ/mole) to a high temperature rhombohedral form (R3̅m, Z = 3, a = 2.958(1), c = 15.748(2) Å , at 300 °C) was observed by Guinier and DSC measurements at 195 °C. The magnetic susceptibility of the monoclinic phase can be described by the Curie-Weiss law between 100 and 330 K: μ = 2.01μB (g = 2.32), Θ = 37 K, indicating the dominance of ferromagnetic interactions of S = 1/2 spins within the NiO2 layers. Antiferromagnetic interlayer interactions produce an overall antiferromagnetically ordered structure below 18 K.

Abstract Background Hematemesis is one of the most eventful complications in patients with reflux esophagitis (RE). But bleeding is not observed in all patients who have RE with esophageal ulcer. We retrospectively studied the association of bleeding with background factors in patients with RE with esophageal ulcer. Methods Between January 2013 and January 2018, 30 patients were endoscopically diagnosed as RE with Los Angeles classification D (LA-D). These 30 patients were enrolled in our study. Fifteen of them were suddenly admitted to our hospital because the RE exhibited bleeding (B-group). Patients with Mallory-Weiss syndrome, peptic ulcers, and any malignancies were excluded as possible sources of the bleeding. Bleeding was not observed in the remaining 15 RE patients (NB-group). In these 30 patients, we retrospectively studied the relations between the bleeding and the following background factors: age, gender, body mass index (BMI), esophageal hiatal hernia, endoscopical atrophy of the gastric mucosa which is related to the secretion of gastric juice, cognitive decline (so called dementia), diabetes mellitus, and drugs (NSAIDs, PPI). Results The B-group included 5 males and 10 females. The NB-group also included 5 males and 10 females. The mean age of the B-group was 81.7 ± 8.2 (M ± SD) yrs, which was statistically (P < 0.05) greater than that of the NB-group, which was 71.5 ± 11.0 yrs. Older age, larger esophageal hiatal hernia, and cognitive decline (so-called dementia) were significantly (P < 0.05) associated with bleeding on logistic regression analysis. Multiple logistic regression analysis showed that older age and dementia were significantly (P < 0.05) associated with bleeding. Conclusion It is well known that esophageal hiatal hernia is a major factor in erosive RE, such as RE with LA-D. On the other hand, it was speculated that older RE patients and those with cognitive decline could not easily explain their RE complaints, and their condition might be left unattended for long periods. This lack of attention may result in sudden bleeding in patients with non-treated RE. Further studies are needed. Disclosure All authors have declared no conflicts of interest.

A new three dimensional hydrogen bonded cobalt frame work from [Co(nicotinamide)2(thiocyanate)2(H2O)2] was synthesized and characterized by X-ray diffraction, magnetism ,TGA and IR spectroscopy. The compound crystallizes in Triclinic space group P-1 with a = 7.5475(19), b = 8.054(2), c =8.932(2). Alpha=73.347(4), beta=70.067(4), gamma=66.559(4) with space group P-1 Z = 1, 'C14 H16 N6 Co O4 S2', Mr =455.38, F(000) = 233 and μ(MoKα) =0.71073 mm-1. The final R = 0.0497 and wR = 0.1461 for 4185 observed reflections with I > 2σ(I) and R = 0.0721 and wR = 0.1619 for all data. X-ray diffraction analyses revealed that Co(1) is linked by the nicotinamide ligands to form the ladder shape along the c axis, which is further extended into two-dimensional networks via the joint of Co(2) along the a axis. Moreover, these two dimensional motifs are interconnected by the thiocyanate S...H bridges to form a complicated 3-D polymeric framework. The magnetic susceptibility data at 1000Oe external field in the temperature range 2-300 K obeys the Curie-Weiss law, giving θ = -24.12 K and C =2.43 thus indicating a dominant strong antiferromagnetic interaction and/or spin orbital coupling between the Co (II) ions.

No valid animal model has been established to evaluate the risks associated with PERV transmission to human patients by pig tissue xenotransplantation or to study the potential pathogenesis associated with PERV infection. In previous work we isolated two genes encoding functional human PERV receptors and proved that introduction of these into mouse fibroblasts allowed the normally nonpermissive mouse cells to become productively infected (T. A. Ericsson, Y. Takeuchi, C. Templin, G. Quinn, S. F. Farhadian, J. C. Wood, B. A. Oldmixon, K. M. Suling, J. K. Ishii, Y. Kitagawa, T. Miyazawa, D. R. Salomon, R. A. Weiss, and C. Patience, Proc. Natl. Acad. Sci. USA 100:6759-6764, 2003). In the present study we created mice transgenic for human PERV-A receptor 2 (HuPAR-2). After inoculation of transgenic animals with infectious PERV supernatants, viral DNA and RNA were detected at multiple time points, indicating productive replication. This establishes the role of HuPAR-2 in PERV infection in vivo; in addition, these transgenic mice represent a new model for determining the risk of PERV transmission and potential pathogenesis. These mice also create a unique opportunity to study the immune response to PERV infection and test potential therapeutic or preventative modalities.

L’objectif de cette étude est d’analyser des recherches portant sur des interventions fondées sur le sport en milieu de réhabilitation afin de comprendre comment et pourquoi de telles initiatives peuvent avoir un impact sur la réhabilitation des jeunes participants. Une revue de la littérature à été effectuée à partir de quatre bases de données : ERIC, Criminal Justice Abstracts, Psycinfo et Sport Discuss. Toutes les études répondant aux critères d’inclusion (N = 10) ont d’abord été analysées de manière descriptive, puis compréhensive à la lumière de la Théorie du changement (Weiss, 1998). Ces analyses ont permis de mettre en lumière les facteurs associés au fonctionnement des interventions basées sur le sport auprès des jeunes contrevenants. Les grands thèmes sous lesquels se regroupent les principaux facteurs sont (a) les jeunes participants (participation et non participation), (b) l’intervention basée sur le sport (objectifs et mécanismes), (c) l’expérience sportive des participants (entraîneurs et discipline sportive), (d) les effets de l’intervention sur les participants (attitude face à la délinquance, comportements délinquants, physique, psychologique, social et psychoéducative), et finalement, (e) les facteurs influençant la relation entre la participation sportive et les impacts sur les jeunes participants (intra personnels, interpersonnels et programmation). La présente étude mène à des recommandations sur l’implantation d’une intervention basée sur le sport en milieu de réhabilitation.

Abstract:
The aftermath of the global financial crisis that began in 2007 in the United States and has yet to end has demonstrated the durability of policies and analytical frameworks that have characterised the neoliberal era. Among these has been a strong rhetorical commitment to market-based institutions as synonymous with individual freedom, in contradistinction to the inefficient, bureaucratic state. This has disguised what amounts to a reconfiguration of state power during the last 40 years, such that it is arguably more pervasive than during the preceding Keynesian era. The books under review here each reveal the ideological character of laissez-faire while in various ways hinting at its mythological properties. This mythical aspect is most prominently displayed by Edmund Phelps’ treatment of ‘corporatism’ as responsible for our current ills, when it is in fact integral to contemporary political economy, and cannot be otherwise, absent truly revolutionary change. Abstract:
The comprehensive study of low temperature magnetoresistance (MR) has been carried out on high quality PrB6 and NdB6 single crystals at temperatures 2–20K and in magnetic fields up to 8T. The negative MR (Δρ/ρ<0) was found in the paramagnetic state at T>TN for both compounds. The analysis of the experimental data allows us to establish three contributions to MR. In addition to the main Brillouin type negative component –Δρ/ρ~Mloc2~H2 which may be interpreted in terms of Yosida theory [Phys. Rev. 107 (1957) 396] both the linear and nonlinear magnetic contributions were also deduced. According to detailed analysis in the framework of spin–polaron approach proposed by A.V. Bogach et al. [Physica B 378–380 (1957) 769] the last component should be ascribed to the ferromagnetic (FM) nanodomains embedded in the metallic RB6 matrix. The estimation of local magnetic susceptibility χloc=(–1/H•(d(Δρ/ρ)/dH))1/2 is deduced directly from the quadratic part of negative MR and reveals Curie–Weiss type behaviour in paramagnetic (PM) phase with paramagnetic Curie temperatures Θp~– 22K and –31K for PrB6 and NdB6 correspondingly. The results of undertaken analysis allow to conclude in favour of the concurrence between AF and FM iteractions as the main reason of incommensurate (IC) magnetic structure formation in RB6.

Abstract:
Представлены методики экспериментов для изучения переохлажденной воды с использованием микроволновой спектроскопии. Одна методика связана с получением глубокого переохлаждения воды в порах силикатного материала, другая основана на получении аморфного состояния в образце пресного льда при его пластической деформации. Показаны возможности методик при изучении свойств переохлажденной воды. При атмосферном давлении и температуре –45 °С (на линии Видома) был определен интервал температур, в котором наблюдаются аномалии микроволновых потерь переохлажденной воды, находящейся в порах силикагеля. ABSTRACT Using isogenic recombinant murine coronaviruses expressing wild-type murine hepatitis virus strain 4 (MHV-4) or MHV-A59 spike glycoproteins or chimeric MHV-4/MHV-A59 spike glycoproteins, we have demonstrated the biological functionality of the N-terminus of the spike, encompassing the receptor binding domain (RBD). We have used two assays, one an in vitro liposome binding assay and the other a tissue culture replication assay. The liposome binding assay shows that interaction of the receptor with spikes on virions at 37°C causes a conformational change that makes the virions hydrophobic so that they bind to liposomes (B. D. Zelus, J. H. Schickli, D. M. Blau, S. R. Weiss, and K. V. Holmes, J. Virol. 77: 830-840, 2003). Recombinant viruses with spikes containing the RBD of either MHV-A59 or MHV-4 readily associated with liposomes at 37°C in the presence of soluble mCEACAM1a, except for S4R, which expresses the entire wild-type MHV-4 spike and associated only inefficiently with liposomes following incubation with soluble mCEACAM1a. In contrast, soluble mCEACAM1b allowed viruses with the MHV-A59 RBD to associate with liposomes more efficiently than did viruses with the MHV-4 RBD. In the second assay, which requires virus entry and replication, all recombinant viruses replicated efficiently in BHK cells expressing mCEACAM1a. In BHK cells expressing mCEACAM1b, only viruses expressing chimeric spikes with the MHV-A59 RBD could replicate, while replication of viruses expressing chimeric spikes with the MHV-4 RBD was undetectable. Despite having the MHV-4 RBD, S4R replicated in BHK cells expressing mCEACAM1b; this is most probably due to spread via CEACAM1 receptor-independent cell-to-cell fusion, an activity displayed only by S4R among the recombinant viruses studied here. These data suggest that the RBD domain and the rest of the spike must coevolve to optimize function in viral entry and spread.

L’objectif du présent article est d’initier un dialogue entre les domaines de la psychologie, de la psychoéducation et de la criminologie avec celui des sciences du sport dans le but de synthétiser les récents progrès réalisés en sciences du sport et d’en faire profiter l’intervention basée sur le sport auprès des jeunes contrevenants. Une revue de la littérature a d’abord été effectuée à partir de quatre bases de données : ERIC, Criminal Justice Abstracts, Psycinfo et Sport Discuss. Toutes les études répondant aux critères d’inclusion (n = 8) ont ensuite été analysées de manière descriptive, puis compréhensive à la lumière de la Théorie du changement (Weiss, 1998). Ces analyses ont permis de mettre en lumière les facteurs associés au rôle potentiel de l’entraîneur dans le développement positif des jeunes participants. Ces facteurs concernent le savoir-être et le savoir-faire de l’entraîneur. De plus, cette étude a permis de dégager l’effet que la participation sportive peut avoir sur le bien-être psychologique des jeunes, ainsi que ses conséquences développementales. Finalement des recommandations pratiques concernant la mise en place d’une intervention basée sur le sport en milieu de réhabilitation sont présentées. Ces interventions devraient être dispensées par un intervenant qui : (a) connait bien les besoins des jeunes contrevenants et adapte ses interventions à ceux-ci, (b) possède une philosophie de développement holistique, (c) tend à établir des relations horizontales avec les participants et (d) leur donne une rétroaction positive.

Still, the risk of endogenous retrovirus transmission represents a major obstacle, since two human-tropic porcine endogenous retroviruses (PERVs) had been characterized in vitro (P. Le Tissier, J. P. Stoye, Y. Takeuchi, C. Patience, and R. A. Weiss, Nature 389:681–682, 1997). Here we addressed the question of PERV distribution in a French Large White SPF pig herd in vivo. First, PCR screening for previously described PERV envelope genes envA, envB, and envC(D. E. Akiyoshi, M. Denaro, H. Zhu, J. L. Greenstein, P. Banerjee, and J. A. Fishman, J. Virol. 72:4503–4507, 1998; Le Tissier et al., op. cit.). demonstrated ubiquity of envAand envB sequences, whereas envC genes were absent in some animals. On this basis, selective out-breeding of pigs of remote origin might be a means to reduce proviral load in organ donors. Second, we investigated PERV genome carriage inenvC negative swine. Eleven distinct full-length PERV transcripts were isolated. The sequence of the complete envelope open reading frame was determined. The deduced amino acid sequences revealed the existence of four clones with functional and five clones with defective PERV PK-15 A- and B-like envelope sequences. The occurrence of easily detectable levels of PERV variants in different pig tissues in vivo heightens the need to assess PERV transmission in xenotransplantation animal models.

Background: Early intervention can reduce the rate of progression and improve overall survival (OS) in smouldering multiple myeloma (SMM) patients (pts) (1, 2). The most powerful predictor of prognosis in active MM is whether pts achieve minimal residual disease (MRD) negativity in bone marrow with treatment (3), however treating MRD relapse has not been evaluated in a randomized fashion. The REMNANT study will evaluate whether treating MRD relapse after first line (1.L) treatment prolongs progression free survival (PFS) and OS for MM pts versus treating relapse after 1.L. treatment at progressive disease (PD) (4). To establish a homogenous group of MRD negative pts after 1.L treatment including autologous stem cell transplantation (ASCT), pts are enrolled at diagnosis and treated with Norwegian standard of care (SOC) 1.L treatment. MRD negative pts will move on to the randomized part. Methods: The REMNANT study is an academic, multicenter, open-label, randomized phase II/III study of NDMM pts eligible for ASCT (see Figure 1). 391 pts across Norway will be included in the phase II part of the study and receive SOC 1.L treatment according to Norwegian national guidelines; VRd (V: 1,3 mg/m2 SC Days 1, 4, 8, 11; R: 25 mg PO Days 1-14; d: 20 mg PO Days 1, 2, 4, 5, 8, 9, 11, 12) for 4 pre-transplant induction and 4 post-transplant consolidation cycles (all 21-d cycles). After induction pts will undergo tandem or single ASCT, depending on toxicity and response to first ASCT. The primary endpoint of the phase 2 part of the study is the number of pts who achieve MRD negative (Euroflow NGF 10 -5) complete response (CR) 30-45 days post consolidation. Safety evaluations and pts-reported outcome assessment will be measured. Pts (176) achieving MRD negative CR will be randomly assigned in a 1:1 ratio to receive 2.L treatment at MRD reappearance (arm A) or at progressive disease (PD) as defined by the IMWG criteria (4) (arm B). Randomization will be stratified by R-ISS stage at diagnosis and single vs tandem ASCT. Pts in arm A will be followed with MRD assessment every 4 month and start 2.L treatment at loss of MRD negative CR. Pts in arm B will be followed up by standard criteria and start 2.L treatment at PD. Both arms will receive the same 2.L treatment; KdD (all 28-d cycles) (K: 70mg/m2 iv Days 1,8,15 d: 40 mg Days 1, 8, 15, 22 D: 1800 mg SC Days 1, 8, 15 during C 1-2, Days 1, 8 during cycle 3-6, Day 1 from cycle 7,). 2.L treatment will continue until disease progression, unacceptable AEs or patient withdrawal. In arm A MRD Euroflow will be assessed after 6 and 18 months of 2L therapy. In arm B MRD Euroflow will be assessed if &gt;CR is achieved but not before 6 months of 2 L therapy, and again after 12 consecutive months. The co-primary endpoint is progression and death by any cause (PFS) and death by any cause alone (OS). Parental Involvement in their children's schooling has long been recognized as a critical component of good education. This study aims to find out the relationship between socioeconomic status, interpersonal communication, and school climate with parental involvement in early childhood education. Using survey and correlational research design, data collection was carried out through accumulation techniques with tests and questionnaires. The data analysis technique used statistical analysis and multiple regressions. The findings in the socio-economic context of parents show that the measure of power is an indicator in the very high category with a total score of 5, while the measures of wealth, honour and knowledge are included in the high category with a total score of 4 in relation to parental involvement. The form of interpersonal communication, the openness of parents in responding happily to information / news received from schools about children is a finding of a significant relationship with parental involvement in early childhood education. The school climate describes the responsibility for their respective duties and roles, work support provided, and interpersonal communication relationships, parents at home and teachers at school. Abstract:
A partir de análises que constataram que falantes de Língua de Herança (LH), ao estudarem a língua na fase adulta, produziam elementos fonéticos/fonológicos relacionados à variante da região de origem dos pais, que foram aprendidos na infância, apresentamos nesse artigo repercussões desse resultado de pesquisa para as famílias e para os professores de línguas de herança. Para isso, utilizamos Cummins (1983), com o conceito de LH, Gontijo e Silva (2016) e Souza (2016), que estudam o ensino de LH em diferentes contextos. Também nos baseamos em Izquierdo et al (2013) e Xavier (2013), para compreendermos aspectos da memória humana e em Fonseca, Weiss e Dutra (2018), que observaram os indícios da atuação da memória fonológica no registro da fonética das línguas aprendidas na primeira infância. O aluno de língua de herança tem nessa memória fonológica uma vantagem - uma marca que lhe confere uma identidade cultural, e uma habilidade valorizada na proficiência em L2. Abstract:
BackgroundEngineered immune cells hold tremendous promise for the treatment of advanced cancers. As the scale and complexity of engineered cell therapies increase, reliance on viral vectors for clinical production limits translation of promising new therapies. Here, we present an optimized platform for CRISPR/Cas9-targeted, non-viral engineering of primary human T cells that overcomes key limitations of previous approaches, namely DNA-induced toxicity and low efficiency integration of large genetic cargos.MethodsA systematic optimization of nucleic acid delivery, editing reagent composition, and culture protocol was performed to overcome DNA toxicity. Targeted knockin (KI) at AAVS1 and TRAC was compared across multiple vector configurations with genetic cargos ranging from 1 to 3 kilobases (kb) in size. Integration efficiency was measured by flow cytometry and sequencing. Off-target editing and integration were evaluated using GUIDE-seq and targeted locus amplification (TLA), respectively. Phenotype and function of non-virally and lentivirus engineered CAR-T cells was compared using flow cytometry, cytokine profiling and cytotoxicity assays.ResultsWe identified a temporal window following T cell activation where transfection efficiency, cell-cycle-status, and cytosolic DNA sensor expression were optimal for targeted DNA integration and reduced toxicity. Within this window, we targeted a 1kb GFP reporter to the AAVS1 locus with an efficiency of ~45% using homologous recombination (HR). Efficiency was reduced to ~11% with a larger ~3kb TCR cassette targeted to the TRAC locus, consistent with previous reports.1–3 To improve large cargo integration we employed homology mediated end-joining (HMEJ) and short homology design (48bp vs. ~1kb for traditional HR).4 Using HMEJ, knockin of the 1kb GFP cassette at AAVS1 reached ~70%. Strikingly, integration of the 3kb TCR at TRAC reached ~50% using HMEJ. Additional optimization of the culture protocol doubled post-engineering survival and proliferation (up to ~35-fold expansion in 7 days). Non-virally engineered TRAC KI CAR-T cells were phenotypically and functionally equivalent to lentivirally engineered T cells in vitro. In vivo assays in xenograft models are underway and results will be presented.ConclusionsComprehensive, orthogonal optimization of parameters impacting nucleic acid delivery and DNA-toxicity in combination with novel modalities for integration achieved knockin of TCR and CAR cargo at efficiencies equivalent to that of current viral vector platforms without compromising expansion or function. Background:Osteoarthritis (OA) is the commonest disease affecting hip joints and has high prevalence across various age groups [1,2]. Effusion is a hallmark of OA and could represent a potential target for therapy [3–5]. Positive correlations of effusion to clinical outcomes are not well established, partly due to variability in manual assessment. Voxel-based volume quantification could reduce this variability [6].Objectives:We examine the inter-observer agreement of manual assessment of voxel-based effusion volume from coronal STIR MRI sequences at two time points and examine the feasibility of using Artificial Intelligence (AI) for standalone volume assessment.Methods:Our algorithm is based on Mask R-CNN [7] and was trained on labeled effusion regions in MRI slices from 68 patients with hip osteoarthritis. For validation, 2 human readers measured effusion from MRI STIR sequences of 25 patients at baseline and at 8 weeks follow-up. AI was used to measure effusion volume as an independent reader. Agreement between human readers and AI was assessed using absolute difference in volume (DV), Coefficients of Variation (CoV) and intraclass correlation coefficient (ICC).Results:Effusion regions detected by AI closely correlated with manual segmentation (Figure 1) for all samples. Differences in volumes measured by each pair of readers are summarized in Table 1. Abstract:
Adrenocortical carcinoma (ACC) is a rare and in most cases steroid hormone-producing tumor with variable prognosis. The purpose of these guidelines is to provide clinicians with best possible evidence-based recommendations for clinical management of patients with ACC based on the GRADE (Grading of Recommendations Assessment, Development and Evaluation) system. We predefined four main clinical questions, which we judged as particularly important for the management of ACC patients and performed systematic literature searches: (A) What is needed to diagnose an ACC by histopathology? (B) Which are the best prognostic markers in ACC? (C) Is adjuvant therapy able to prevent recurrent disease or reduce mortality after radical resection? (D) What is the best treatment option for macroscopically incompletely resected, recurrent or metastatic disease? Other relevant questions were discussed within the group. Selected Recommendations: (i) We recommend that all patients with suspected and proven ACC are discussed in a multidisciplinary expert team meeting. (ii) We recommend that every patient with (suspected) ACC should undergo careful clinical assessment, detailed endocrine work-up to identify autonomous hormone excess and adrenal-focused imaging. (iii) We recommend that adrenal surgery for (suspected) ACC should be performed only by surgeons experienced in adrenal and oncological surgery aiming at a completeen blocresection (including resection of oligo-metastatic disease). (iv) We suggest that all suspected ACC should be reviewed by an expert adrenal pathologist using the Weiss score and providing Ki67 index. (v) We suggest adjuvant mitotane treatment in patients after radical surgery that have a perceived high risk of recurrence (ENSAT stage III, or R1 resection, or Ki67 >10%). (vi) For advanced ACC not amenable to complete surgical resection, local therapeutic measures (e.g. radiation therapy, radiofrequency ablation, chemoembolization) are of particular value. However, we suggest against the routine use of adrenal surgery in case of widespread metastatic disease. In these patients, we recommend either mitotane monotherapy or mitotane, etoposide, doxorubicin and cisplatin depending on prognostic parameters. In selected patients with a good response, surgery may be subsequently considered. (vii) In patients with recurrent disease and a disease-free interval of at least 12 months, in whom a complete resection/ablation seems feasible, we recommend surgery or alternatively other local therapies. Furthermore, we offer detailed recommendations about the management of mitotane treatment and other supportive therapies. Finally, we suggest directions for future research.

Abstract:
Despite modern advances used to estimate the discharge coefficient ( C d ), it is still a major challenge for hydraulic engineers to accurately determine C d for side weirs. In this study, extra tree regression (ETR) was used to predict the C d of rectangular sharp-crested side weirs depending on hydraulic and geometrical parameters. The prediction capacity of the ETR model was validated with two predictive models, namely, extreme learning machine (ELM) and random forest (RF). The quantitative assessment revealed that the ETR model achieved the highest accuracy in the predictions compared to other applied models, and also, it exhibited excellent agreement between measured and predicted C d (correlation coefficient is 0.9603). Moreover, the ETR achieved 6.73% and 22.96% higher prediction accuracy in terms of root mean square error in comparison to ELM and RF, respectively. Furthermore, the performed sensitivity analysis shows that the geometrical parameter such as b/B has the most influence on C d . Overall, the proposed model (ETR) is found to be a suitable, practical, and qualified computer-aid technology for C d modeling that may contribute to enhance the basic knowledge of hydraulic considerations.

Abstract Abstract SCI-32 MicroRNAs (miRs) inhibit stability and/or translation of mRNAs, usually by binding to specific sites in the 32′UTRs of their target mRNAs. Due to imperfect (i.e. partially complementary) miR:mRNA base-pairing, miRs can block translation of many mRNAs and serve as powerful master switches to regulate cell functions. Therefore, we profiled miR expression in human CD34+ hematopoietic stem-progenitor cells (HSPCs) and combined human HSPC miR expression, mRNA expression, and miR-mRNA target predictions to hypothesize that certain HSPC-expressed miRs (HE-miRs) target several mRNAs critical to hematopoiesis. On this informatic basis, we formulated a model of hematopoietic differentation in which many genes specifying hematopoietic differentiation are expressed by early HSPCs, but held in check by miRs [1]. In addition, we noted that the miR-23a cluster (i.e. adjacent, co-transcribed miR-23a, miR-27a, and miR-24-2) is not expressed or is expressed at levels >2-fold lower in 50% of acute myeloid leukemias and 80% of acute lymphoid leukemias tested compared to normal human HSPCs. ‘Re-expressing’ 1 or more of these miR-23a cluster members in leukemia cells promotes their apoptosis and reduces their proliferation, thus suggesting that these miRs have a tumor suppressive role. We have identified YWHAQ (14-3-3q) and several other 14-3-3 isoforms, which are anti-apoptotic and have established roles as oncogenes, as miR-23a cluster target molecules. Artificial manipulation of these HE-miRs and their target genes may lead to novel strategies for leukemia treatment and/or for expansion of normal HSPCs. Since the CD34+ HSPCs that we studied initially include rare stem cells and various stages of progenitors, we have expanded our miR profiling to more highly purified subsets of mouse HSPCs. Several previously described (e.g. miR-155 [1], miR-451 [2], miR-146 [3]) and novel HE-miRs are expressed differentially in lineages/stages of HSPCs, and their selective expression has been confirmed in human HSPC subsets. We are using cellular gain- and loss-of-function approaches with hematopoietic functional assays to determine whether these HE-miRs control human hematopoiesis. The prevalence of ADHD children increases every year. Some researchers have shown that psychosocial behavior therapy (antecedents-consequences modification) was effective to decrease hyperactivity and increase attention to ADHD children. This study aims to find out the effectiveness of antecedents-consequences modification by parents and teachers to decrease hyperactivity and increase attention to a 6 years old boy with ADHD. The study was a single case experimental design. Psychosocial behavior therapy has been used with antecedents-consequences modification. The antecedents-consequences modification was applied by teacher at school and parents at home. Data were analyzed using Wilcoxon Signed Rank Test. Results showed that there’s a significant decrease of hyperactivity behavior and significant increase of doing his assignment both at school and also at home. Not only about the content of behavior therapy itself, but how to give the therapy is important. Parents and teacher should do the therapy consistently, immediately, specifically and saliency to reach the target of intervention. The Late Preclassic to Classic period (400 B. C.-A. D. 900) Maya community of La Milpa, Belize, has recently revealed an ancient water and land-use system. As demonstrated at other southern Maya Lowland sites, the Maya created a microwatershed to store and convey water during the four months of seasonal drought. In addition to water conservation measures associated with reservoirs, deliberate channelization, diversion weirs, and postulated fields, the importance of rainy-season erosion control is indicated. Given the ancient population densities identified in the Maya area, coupled with the seasonal scarcity of water, we posit a “skill-oriented” economy.

Zusammenfassung Einleitung Qualitäts- und Risikomanagement (QRM) in Krankenhäusern sind nicht nur gesetzlich gefordert, sondern führen maßgeblich zu einer optimierten patienten- und ablauforientierten Krankenversorgung. Zur Evaluation der Reifegrade des QRM-Systems am Universitätsklinikum Schleswig-Holstein (UKSH) wurde ein strukturiertes Analysetool entwickelt, das einfach und effizient anzuwenden ist. Methode Vier Qualitätskriterien – Qualitätssicherung (QS), Critical Incident Reporting System (CIRS), Beschwerdemanagement (BM) und Prozessmanagement (PM) – wurden mittels eines strukturierten Erhebungsbogens zur Eigen- und Fremdbewertung an den beiden Standorten des UKSH in Kiel und Lübeck analysiert. Jedes Qualitätskriterium wiederum wurde in vier Kategorien von A (voll implementiert) bis D (nicht implementiert) eingeteilt. Zunächst fand eine Fremdbewertung durch das zentrale QRM und im Anschluss die Eigenbewertung der insgesamt 46 analysierten Kliniken zu den jeweiligen vier Qualitätskriterien statt. Unstimmigkeiten zwischen Eigen- und Fremdbewertungen wurden in einem kollegialen Dialog gelöst. Letztendlich resultierte aus den erhobenen Daten eine Gesamtreifegradbeurteilung pro Klinik. Ergebnisse Der Gesamtreifegrad A wurde von 3 der 46 Kliniken (6,5%) erzielt. Keine Klinik erhielt die Gesamtreifegradbeurteilung D. 50% aller Kliniken erreichten Reifegrad B und 43,5% Reifegrad C. Die Verteilung der unterschiedlichen vier Qualitätskriterien zeigte insgesamt eine gute Implementierung des Beschwerdemanagements (Reifegrade A und B mit 78,3%) auf, während bei CIRS ein hoher Anteil der Reifegrade C und D (73,9%) vorlag. QS und PM wiesen eine sehr ähnliche Verteilung auf mit den Reifegradbeurteilungen B und C (87% QS; 91% PM). Diskussion Durch das strukturierte Analysetool sind sowohl die Gesamtreifegrade des QRM-Systems an 46 Kliniken des UKSH als auch detailliert die Reifegrade 4 wesentlicher Qualitätskriterien (QS, CIRS, BM, PM) erhoben worden. In der Konsequenz wurde ein umfangreiches Maßnahmenpaket zur Verbesserung des QRM-Systems eingeleitet. Zukünftig werden die Reifegrade im 2 Jahresrhythmus re-evaluiert. Dieses qualitative Reifegradmodell ermöglicht auf einfache und effiziente Weise, präzise Aussagen bezüglich des Vorhandenseins, der Ausprägung und der Weiterentwicklung des Qualitäts- und Risikomanagements zu generieren.

Kontrazeptive Compliance bedeutet, dass eine Verhütungsmethode korrekt, d.h. in genau der Weise angewendet wird, wie dies medizinisch von der Methode her erforderlich bzw. vom Arzt her vorgeschrieben ist. Non-Compliance beschreibt das Missverhältnis zwischen korrektem und realem Anwendungsverhalten. Verschiedene Indikatoren weisen darauf hin, dass die kontrazeptive Compliance ungenügend ist. In den meisten Studien wird eine Rate von 40–50% ungeplanter Schwangerschaften angegeben; knapp die Hälfte dieser Schwangerschaften enden in Schwangerschaftsabbrüchen, deren Zahl weiterhin hoch ist. Bei hormonalen und Barrieremethoden besteht eine deutliche Diskrepanz zwischen der Effizienz bei korrekter Anwendung versus der Effizienz bei typischer Anwendung. Die Abbruchraten der kontrazeptiven Therapie nach einem Jahr sind bis zu 50%. Gründe für Non-Compliance sind a) Mangelnde oder fehlende Motivation durch Ambivalenz bezüglich Kinderwunsch, sowie sexuelle und partnerschaftliche Konflikte; (b) Kognitive Faktoren (Fehlinformationen, irrationale Ängste); (c) Erschwerter Zugang zu kontrazeptiven Methoden; (d) Verhaltensfehler (Vergessen, Verlieren) und (e) Nebenwirkungen der Methoden. Drei Strategien zur Verbesserung der Compliance werden eingesetzt: a) Verbesserung der Beratung und Betreuung; b) Entwicklung von Methoden, die keine oder nur geringe Compliance erfordern; c) Verbesserung der Lebensqualität bei Anwendung kontrazeptiver Methoden durch Minderung negativer Nebenwirkungen und Steigerung des nicht kontrazeptiven Nutzens.

Lomaya and Pilohayanga Weirs are located in Limboto-Bone- Bolango River, Bone Bolango District, and Gorontalo Province. Pilohayanga Weir is located at 1,500 meters downstream of Lomaya Weir, it has 1,045 hectares of irrigation services area while also serving as a supplier of raw water requirements. The damage that occurred in Pilohayanga Weir resulted in its decreasing function, and also resulted in unmet irrigation and raw water needs. Meanwhile, Lomaya Weir has 3,148 hectares of irrigation services area. The aim of this research is to find the optimal solution among several alternative design concepts to meet Pilohayanga Weir’s water needs by supplying water from Lomaya Weir, considering both the technical aspects of hydraulic and the cost of implementation which must be within the economical boundary. In conclusion, the exact solution required is by the addition of several designs, these include: a) creating a new intake area on the right side of Pilohayanga Weir; b) creating an additional channel, flushing out sediment from Lomaya Weir; c) creating sand trap; and d) creating a carrier which will be channelled to Pilohayanga Irrigation Area.

Zusammenfassung Im Beitrag werden mehr als 180 Gegenstände aus Eisen ausgewertet, die zwischen dem 3. Jahrtausend v. Chr. und dem Anfang des 1. Jahrtausends v. Chr. im Raum zwischen den Karpaten und dem Ural gefunden wurden. Die Objekte stammen von verschiedenen Fundorten und aus 15 archäologischen Kulturen. Der zeitliche Rahmen der Studie entspricht der Bronzezeit und dem Übergang zur frühen Eisenzeit auf Grundlage der aktuellen chronologischen Vorstellungen in Osteuropa. Der Auswertung liegt eine umfangreiche Datenbank zugrunde. Vorgestellt und analysiert werden: a) die Anzahl der Eisenobjekte und ihre Verbreitung; b) die Kategorien der Gegenstände, die aus Eisen gefertigt wurden; c) die Dynamik und die Art und Weise wie Eisen in Gebrauch kam; d) forschungsgeschichtliche Aspekte. Die Auswertung des Materialkorpus mit den aktualisierten Daten und der frühen Funde aus Eisen sowie Schlacken ergibt ein konkretes Bild ihrer quantitativen und qualitativen Verteilung über zwei Jahrtausende in dem Arbeitsgebiet. Die Untersuchung wirft ein neues Licht auf die Tendenzen in der frühen Eisennutzung in Osteuropa. Das Fazit der Autoren, dass Osteuropa als ein eigenständiges Zentrum bei der Herausbildung der Eisenmetallurgie in der Alten Welt anzusehen ist, muss noch weiter überprüft und durch moderne naturwissenschaftliche Analysen abgesichert werden.

This article aims to present a research result on meanings of formative practice in the context of Institutional Program of Initiation to Teaching (PIBID). As a research method, adopted the sociological phenomenology that allowed, among other aspects, to understand the symbolic dimension of formative practice for each students. One of the principles of that method consists in the perspective that senses emerge from the relation with the other, bringing to surface the idea of intersubjectivity, thus configuring a phenomenology of social relations. In order to access the senses produced in the world of life, in encounter of consciousness with the world, in specific case of this research, in the context of formative practice, semistructured interview has been used with five undergraduate students invited to give a break in the flow of life for think about teacher training. The senses revealed in this study are close to the concern with student learning, having the teacher as the main driver of this process, as well as the student and his way of learning. There are also senses about difficulties that each student presents in application of knowledge and draws attention to the relation between knowledge and reality. Sense about the teacher-student relationship reveals concern with professional and human dimension as well as authority of teacher's knowledge in the classroom with students.Resumo Este artigo tem como objetivo apresentar resultado de pesquisa sobre sentidos de prática formativa no contexto do Programa Institucional de Bolsa de Iniciação à Docência (PIBID). Como método de pesquisa adotou-se a Fenomenologia Sociológica que permitiu, entre outros aspectos, compreender a dimensão simbólica da prática formativa para cada um dos estudantes. Um dos princípios desse método consiste na perspectiva de que os sentidos nascem da relação com o outro trazendo à tona a ideia de intersubjetividade configurando, assim, uma Fenomenologia das relações sociais. Para acessar os sentidos produzidos no mundo da vida, no encontro da consciência com o mundo, no caso especifico desta pesquisa, no contexto da prática formativa, foi utilizada a entrevista semiestruturada com cinco estudantes de licenciaturas diversas, convidados a suspender o fluxo da vida para pensar sobre sua formação docente. Os sentidos revelados neste estudo se aproximam na preocupação com a aprendizagem do estudante tendo o professor como principal condutor desse processo como, também, o estudante e seu modo de aprender. Aparecem, também, sentidos a respeito das dificuldades que cada aluno apresenta na aplicação do conhecimento e chama à atenção para a relação entre conhecimento e realidade. Sentidos sobre a relação professor-estudante revelam preocupação com a dimensão profissional e humana, bem como a autoridade do conhecimento do professor, em sala de aula, com os estudantes.ResumenEste artículo tiene como objetivo presentar resultados de investigación sobre sentidos de práctica formativa en el contexto del Programa Institucional de Beca de Iniciación a la Docencia (PIBID). Background In many plant species, pollination syndromes predict the most effective pollinator. However, other floral visitors may also offer effective pollination services and promote mixed pollination systems. Several species of the species-rich Penstemon (Plantaginaceae) exhibit a suite of floral traits that suggest adaptation for pollination by both hymenopterans and hummingbirds. Transitions from the ancestral hymenopteran pollination syndrome to more derived hummingbird pollination syndrome may be promoted if the quantity or quality of visits by hummingbirds is increased and if the ancestral pollinator group performs less efficiently. The quantification of such shifts in pollination systems in the group is still limited. We aimed to investigate floral traits linked to this pollination syndrome in Penstemon gentianoides with flowers visited by bumblebees and hummingbirds. Methods We investigated the floral biology, pollinator assemblages, breeding system and nectar production patterns ofP. gentianoides inhabiting a temperate montane forest in central Mexico. Pollination experiments were also conducted to assess the pollinator effectiveness of bumblebees and hummingbirds. Results P. gentianoides flowers are protandrous, with 8-d male phase (staminate) flowers, followed by the ∼1–7 d female phase (pistillate phase). Flowers display traits associated with hymenopteran pollination, including purple flowers abruptly ampliate-ventricose to a broad throat with anthers and stigmas included, and long lifespans. However, the nectar available in the morning hours was abundant and dilute, traits linked to flowers with a hummingbird pollination syndrome. Two hummingbird species made most of the visits to flowers, Selasphorus platycercus (30.3% of all visits), followed by Archilochus colubris (11.3%). Bumblebees (Bombus ephippiatus, B. huntii and B. weisi) accounted for 51.8% of all recorded visits, but their foraging activity was restricted to the warmer hours. Hummingbirds made more foraging bouts and visited more flowers than hymenopteran species. Flowers experimentally pollinated by B. ephippiatus produced significantly more fruits than those pollinated by S. platycercus. However, there was no statistical difference in the number of seeds produced per fruit when a bumblebee or a hummingbird was the pollinator. Conclusions We have shown that bumblebees and hummingbirds visit and pollinate P. gentianoides flowers. Despite floral traits resembling the hymenoptera pollination syndrome, flowers of P. gentianoides offer characteristic nectar rewards to flowers with a hummingbird pollination syndrome. Although pollination efficiency is higher among flowers visited by hymenoptera, the noteworthy percentage of fruit production and number of seeds per fruit derived from hummingbird pollination highlights the importance of hummingbirds as a functional group of pollinators that might have potential evolutionary consequences to the plants.
