Abstract:
The use of nanocarriers is an intriguing approach in the development of efficacious treatment for brain disorders. The aim of the conducted research was to evaluate and quantify the impact of a liposomal nanocarrier formulation on the brain drug delivery. A novel approach for investigating the blood-brain barrier transport of liposomal DAMGO is presented, including in vivo microdialysis in rat, a high quality LC-MS/MS bioanalytical method and pharmacokinetic model analysis of the data. Factors limiting the brain distribution of the free peptide DAMGO were also investigated. Microdialysis, in combination with plasma sampling, made it possible to separate the released drug from the encapsulated and to quantify the active substance in both blood and brain interstitial fluid over time. The opioid peptide DAMGO entered the brain to a limited extent, with a clearance out of the brain 13 times higher than the clearance into the brain. The brain to blood ratio of unbound drug was not affected when the efflux transporter inhibitors cyclosporine A and elacridar were co-administered with DAMGO. Nor was the transport affected in the in vitro Caco-2 assay using the same inhibitors. This indicates that DAMGO is not transported by P-glycoprotein (Pgp) or breast cancer resistant protein (Bcrp). The blood-brain barrier transport was significantly increased for DAMGO when formulated in liposomes, resulting in 2-3 fold higher brain to blood ratio of unbound DAMGO. The increased brain delivery was seen both for glutathione tagged PEGylated liposomes, as well as for PEGyalted liposomes without specific brain targeting. The improvement in brain delivery was observed only when DAMGO was encapsulated into the liposomes, thus excluding any effect of the liposomes themselves on the integrity of the blood-brain barrier. Modeling of the data provided additional mechanistic understanding of the brain uptake, showing that endocytosis or transcytosis of intact liposomes across the endothelial cell membranes were unlikely. A model describing fusion of the liposomes with the luminal membrane described the experimental data the best. In conclusion, the studies presented in this thesis all contribute to an increased understanding of how to evaluate and improve brain delivery of CNS active drugs and contribute with important insights to the nanocarrier field.

Abstract:
Chronic pain is a significant healthcare problem. It is disabling and diminishes quality of life. Opioids, such as morphine, remain a primary pharmacologic management for chronic pain. Opioids act at mu opioid receptors (MOPr) in the rostroventral medial medulla (RVM) to produce their analgesic effect. The RVM is a critical relay in pain inhibitory and facilitatory pathways of pain modulation. Furthermore, chronic inflammatory pain, produced by CFA hindpaw injection, leads to adaptive changes in the RVM that change the balance of these pathways and increase the potency of opioids. MOPr are known to produce their effects via Gi/o proteins. Pretreatment of several pain modulatory regions with pertussis toxin (PTX) effectively attenuates the antinociceptive effects of MOPr agonists, such as DAMGO. In the RVM, PTX effectively reduced DAMGO stimulated GTPãS binding in uninjured rats. However, despite their effective inactivation of Gi/o proteins, PTX did not diminish the antinociceptive effects of DAMGO in the RVM of uninjured rats. In contrast, in rats with a chronic inflammatory injury, PTX completely abolished the antinociceptive effects of DAMGO. These results suggest a transition from Gi/o independent to Gi/o dependent mechanisms following CFA treatment. In addition, the anti-hyperalgesic effects of DAMGO were not inhibited by PTX, suggesting that DAMGO produces anti-hyperalgesia and antinociception by different mechanisms. In the RVM, MOPr are present both postsynaptically and presynaptically. Postsynaptic MOPr are thought to produce antinociception by activating GIRK channels, resulting in hyperpolarization and inhibition of pain facilitatory neurons. Indeed, inhibition of GIRK channels in the RVM, via microinjection of tertiapin-Q, attenuated the antinociceptive effects of DAMGO in uninjured rats, providing the first behavioral evidence that MOPr agonists produce analgesia via this proposed mechanism. Interestingly, however, tertiapin-Q did not block the anti-hyperalgesic effects of DAMGO, nor did it diminish the antinociceptive effects of DAMGO in the contralateral hindpaw of CFA treated rats. Furthermore, these differential effects of tertiapin-Q in the uninjured and injured rats are not the result of transcriptional down regulation of GIRK channels in the RVM. Finally, tertiapin-Q alone in the RVM produced a modest antinociception in uninjured rats, providing the first evidence of constitutive GIRK channel activity in the RVM and demonstrating a role for these in pain modulation. Presynaptic MOPr are thought to produce antinociception by decreasing GABA release onto pain inhibitory neurons. Indeed, microdialysis studies demonstrated that levels of GABA release were decreased in response to DAMGO perfused into the RVM, as well as to high potassium after perfusion of DAMGO. However, they were not decreased in rats after CFA treatment. This suggests that chronic inflammatory injury alters the presynaptic actions of MOPr agonists in the RVM. Interestingly, levels of GLU release where not altered by DAMGO in uninjured or injured rats. Moreover, basal levels of GLU and GABA were also unaltered by CFA treatment. In conclusion, although MOPr mediate their antinociceptive effects in other pain modulatory regions via Gi/o proteins, this is not the case in the RVM during an uninjured state. However, MOPr-induced antinociception transitions from Gi/o independent to Gi/o dependent mechanisms after CFA treatment. Additionally, these results support both the presynaptic and the postsynaptic postulates by which MOPr agonists are thought to produce their analgesic effects. However, although CFA treatment alters the activity of neurons in the RVM and promotes changes that result in an enhanced anti-hyperalgesic and antinociceptive response to DAMGO in the RVM, neither the postsynaptic nor the presynaptic mechanism, in isolation, seem to account for this enhancement.

Abstract:
Glycosylation may be a general strategy for the transport of biologically active neuro(glyco)peptides into the brain. With that in mind, a series of modified DAMGO analogues were synthesized and subjected to conformational analysis, and in vitro and in vivo studies related to opioid analgesia. Those studies will help to determine the balance of carbohydrate and peptide, to reach maximum BBB transport; in other words this is a study to test the biousian hypothesis. 1) The μ-agonist DAMGO was altered by incorporating moieties of increasing water solubility into the C-terminus, including carboxamide and simple glycosides. The hydrophilic C-terminal moieties were varied from glycinol in DAMGO to L-serine amide (LYM100), L-serine amide β-D-xyloside (LYM50), L-serine amide β-Dglucoside (LYM110), L-serine amide β-lactoside (LYM147). Two phosphopeptides LYM1311 and LYM1312 were synthesized with the phosphate group attached to Lserine amide at the C-terminus. Conformational analysis experiments included: 1HNMR, diffusion, variable temperature experiments to find the temperature coefficient, circular dichroism, 2DNMR noesy and tocsy, and molecular modeling. The peptides associate with SDS micelles with a strong electrostatic component. The SDS micelles stabilized the β-turn that is nascent in water. CSI (chemical shift indexes), temperature coefficients and circular dichroism do not give much insight into the structural conformation. 2D NMR analysis followed by molecular modeling confirmed a β-turn preferred conformation. No specific type of β-turn could be assigned to the DAMGO analogs. 2) Antinociceptive mouse tail-flick studies were performed, and opioid binding was determined. Analgesic potency (i.v.) increased, passing through a maximum (A₅₀ ≈ 0.2 μmol/Kg) for LYM100 & LYM50 as membrane affinity vs. water solubility became optimal, and then dropped off (A₅₀ ≈ 1.0 μmol/Kg) for LYM110 & LYM147 as water-solubility dominated the molecular behavior. Correlation of i.v. A₅₀ values with estimated hydrodynamic values (glucose units) for the glycoside moieties, or the hydrophilic/hydrophobic Connolly surface areas (A₅₀ vs e^(-Awater/Alipid)), provided U-shaped or V-shaped curves, as predicted by the “biousian hypothesis.” The μ-selective opioid agonism was maintained upon modifications at the C-terminus. The optimal “degree of glycosylation” that achieves the maximum degree of transport for the DAMGO peptide message seems to be between the peptide with the carboxamide C-terminal group and the xyloside.

Abstract:
This dissertation work is focused on the analytical and numerical examination of the mechanical response of polypropylene (PP) under scratch deformation by a semispherical indenter. The finite element (FE) method is employed as the analysis technique and ABAQUS??, a commercial FE package is adopted to perform the analysis. Important physical and computational considerations on the implementation of FE analyses for the scratch problem are reviewed. It is shown through the discussion of the generated results that a good understanding can be gained on how different scratch conditions can affect scratch behavior of PP. A phenomenological deduction of the scratch damage process and mechanisms is also established. Considering the two main damage modes of polymers, shear yielding and crazing, it is shown that the two damage modes not only exist in the scratch deformation, and moreover, that they may compete against each other for dominance. A parametric study is also performed to assess the influence of material and surface properties on scratch response of material. A secondary research effort is also made to investigate the material constitutive modeling of polymers. Focusing on elastomeric or rubbery materials, a new mixed network model between the Gaussian and eight-chain non-Gaussian models is proposed. This mixed model inherently preserves the good predictive power of these two models and yields better predictions over a wider range of deformation than that of the rubber model adopted by ABAQUS??.

This collective speaks a Jê language and inhabits a transition area between the Cerrado and Amazon biomes. I seek here to apply a method which, based on the assumptions of the Lévi-Strauss's kinship theory, articulated with the basic notions of graph theory, performs a thorough scan of empirical network alliance. This method also postulates which categories, rules and practices, analytical plans traditionally used in kinship studies, are taken as impact levels of tying kinship term, the exchange.<br>O presente trabalho centra sua atenção na aliança de casamento entre os Xavante residentes na Terra Indígena Marãiwatsédé, localizada no leste mato-grossense, distante cerca de 1.200 km de Cuiabá. Esse coletivo fala uma língua Jê e habita uma área de transição entre os biomas do Cerrado e Amazônia. Busco aqui aplicar um método que, partindo de pressupostos da teoria do parentesco de Lévi-Strauss, articulado a noções elementares da teoria dos grafos, realiza uma varredura exaustiva da rede empírica de aliança. O método também postula que categorias, normas e práticas, planos analíticos tradicionalmente usados nos estudos de parentesco, sejam tomados como níveis de repercussão do termo subordinante do parentesco, a troca.

Doutoramento em Música<br>O presente trabalho inscreve-se no domínio da etnomusicologia, resulta da realização de trabalho de campo multissituado e propõe-se compreender a comunidade católica damanense residente em Damão (Índia) e no Reino Unido partindo do estudo da música adotada e praticada pelos seus membros. O repertório performativo analisado neste trabalho inclui música (religiosa e secular) e dança entendidas pelos damanenses como herdadas do antigo colonizador, música e dança anglo-saxónica e de Bollywood transmitida pelos meios de comunicação de massa atuais e, ainda, aquela que é veicularmente considerada pelos damanenses católicos como “a música damanense” ou “a música original de Damão” e à qual é aqui dado um enfoque especial: o mandó. Argumento que a compreensão deste repertório e dos diferentes significados de que se reveste nos permite também compreender a própria damanidade, cuja performance é caracterizada pelos mesmos princípios que definem a da música. Permite-nos, igualmente, entender o modo como os damanenses católicos vivem a sua condição de integração na Índia enquanto comunidade pós-colonial não-independente. Este trabalho procura contribuir, portanto, para a inscrição da realidade dos territórios poscoloniais integrados no quadro da teoria do poscolonialismo e reflete sobre o protagonismo da música na construção de lugares de memória e de imaginação tanto no território de origem (Damão) como na diáspora (Reino Unido).<br>The present work, inserted into the domain of ethnomusicology, results from multi-situated fieldwork and aims to understand, from the study of the music adopted and performed by its members, the Catholic Damanese community living in Daman (India) and in the United Kingdom. The performative repertoire analised in this work includes music (religious and secular) and dance saw by the Damanese as inherited from the former colonizer, Anglo-Saxon and Bollywood music and dance broadcasted by mass media, and that consensually perceived by the Catholic Damanese as “the Damanese music” or “the original music of Daman” which is a main focus of this work: the mandó. I hold that understanding that repertoire and the various meanings it encompasses allows us to understand damanity itself, whose performance is characterised by the same principles defining that of music. It allows us, also, to comprehend the way the Catholic Damanese live their condition of integration into India as a post-colonial non-independent community. Therefore, this work is intended to contribute to the rise of the postcolonial integrated territories’ reality into the framework of the postcolonialism theory, reflecting on the role of music in the construction of lieux de mémoire and places of imagination in the homeland (Daman) as well as in the diaspora (United Kingdom).

El presente trabajo de investigación tuvo como objetivo comparar cómo se ha realizado la adaptación del lenguaje gráfico del manga Hana Yori Dango al lenguaje audiovisual del drama coreano Boys Over Flowers. Esta investigación se centra mayormente en la comparación de un lenguaje a otro, es decir, identificar y comparar la manera en qué ciertos elementos del manga se adaptan al lenguaje audiovisual, y a su vez interpretar la manera cómo se representa el discurso del amor, poder y violencia en la historia. La investigación fue de tipo analítica, comparativa e interpretativa, con un enfoque cualitativo. Tiene como unidad de análisis el manga Hana Yori Dango y el drama coreano Boys Over Flowers. La metodología empleada fue el análisis del discurso de los ejes temáticos del poder, amor y violencia de la trama de la historia del manga y el drama basado en la observación de doce escenas del drama Boys Over Flowers, considerando como criterios de selección, la similitud del discurso narrativo y la importancia de las escenas en la curva dramática del drama. También se tiene presente la similitud de la representación icónica y estética del manga dentro del drama, es decir similitud en las viñetas e imágenes. En conclusión, la adaptación del drama se va a ver condicionado por el manga. Esto se puede apreciar a través de las acciones de los personajes, de lo que dicen entre ellos; así como también algunas equivalencias estéticas del drama a partir de la imagen que es representada en el manga. A pesar de los cambios y modificaciones que se realizaron en varias escenas, la línea dramática de la historia se mantiene. En cuanto a los temas de amor, poder y violencia. Se puede apreciar que la historia toma como tema principal el amor que tienen los protagonistas, el cual va ser puesto a prueba a través de diversas situaciones, en donde el poder y la violencia serán obstáculos de separación de los protagonistas.<br>Tesis

Abstract:
<p> Dambos are seasonally saturated grassy valleys mainly found on the central African plateau. They are usually sub-divided into three catenary units - gleyed and frequently inundated bottoms, flat and rarely inundated floors, and sloping sandy margins - fringed at the interfluve by the well-drained uplands. Since dambos constitute &sim;11% of Africa's arable land, soil information is required to guide sustainable use of the land. Further, it is important to determine the extent of the wetland environment in these landscapes so as to avoid miss-use of the land, which could arise because of varying definitions of the wetland in these landscapes. In addition, lack of knowledge about the true nature of dambo wetlands limits our understanding of their greenhouse gas (GHG) source and sinks strengths, which prevents projection of future GHG scenarios accompanying dambo use. This study was conducted so as to address these inadequacies, and is guided by the following specific objectives: (i) delineate dambo soil-landscape elements using aerial gamma-ray and terrain data; (ii) characterize a dambo wetland; and (iii) determine CH<sub>4</sub> and CO<sub>2</sub> sources and sinks in a dambo landscape.</p><p> The area Hansen et al. (2009) studied was revisited. For objective 1, their model training and validation data were used. For objectives 2 and 3, data (e.g., soil, water table and gas samples) were collected from experimental plots in the four landscape positions, geographically constrained around dambo bottom pixels. Data used were collected during the main (March to July 2008) and short rainfall (October and November 2009) seasons in the area.</p><p> An ANOVA analysis showed landscape position to have a proportionate influence on the variability of eU (46%), K% (28%) and eTh (27%); owing to the differences in soil properties along dambo cross-profiles. The results based on random forests (RF) and multinomial-ISODATA modeling, where gamma-ray and derivatives of a Shuttle Radar Topography Mission (SRTM) digital elevation model (DEM) were used to classify dambo catenary units, were accurate, but only slightly better than the method which made no use of gamma-ray (e.g., conditional inference tree) . It was concluded that dambo landscape elements can be mapped by using these two data sources; although terrain data provides more information. Based upon a combination of hydrology and soil properties, dambo bottoms were the only element shown to constitute the dambo wetland. This zone is inundated for at least three-quarters of the main rainfall season and soils are hydric. Using the landscape map created by Hansen et al. (2009), the wetland was found to constitute only &sim;15% of the dambo. This is smaller than what was mapped by FAO-Africover and the Department of Survey and Mapping, Uganda (DSM). The wetland was also found to be the main source of CH<sub> 4</sub> and sink of CO<sub>2</sub>, with additional strengths attributed to the neighboring floor. Given that these constitute less than 20% of the landscape, dambo net contribution to the regional CH<sub>4</sub> budget is insignificant because 80% of the landscape is a sink. The worry, though, is the ongoing degradation, with the impact this has on the release of CO<sub>2</sub>.</p>

The first nociceptive synapse in the spinal cord dorsal horn represents an important site, where nociceptive synaptic transmission can be modulated under pathological conditions. One of the modulatory mechanism involves activation of the transient receptor potential vanilloid 1 (TRPV1) that is expressed on central terminals of primary nociceptive neurons, where it regulates release of neurotransmitters and neuromodulators. Previous studies suggested that changes in TRPV1 activity may be related to effects of chemokine CCL2 (C-C motif ligand 2) and may be also involved in synaptic transmission modulation after µ-opioid receptors (MOP-R) activation. Because CCL2 receptors CCR2 often co-localize with TRPV1 and MOP-R, the goal of this work was to studypossible interactions of these receptors on the pre-synaptic endings of primaryafferents in the spinal cord dorsal horn and their role in nociceptive signalling under pathological conditions. The presented thesis focused on the effect of CCL2 during peripheral neuropathy and its interference with µ-opioid receptor activation. -5- ABSTRACT A huge effort has been put in determining the mechanism of the development of tolerance and dependence in context of clinical use of morphine for treatment of severe pain. Understanding of this mechanism would help to design new and more efficient pharmaceuticals. This diploma paper discus the opiate receptors with a special focus on long-term effect of chronic morphine treatment, which was determined using a radioligand binding assays with a non-selective antagonist [3 H]Diprenorphine. One of the goals of this work was to create and optimise a method for preparation of pure plasma membranes from rat cortex using percoll gradient. There were five groups, which differed in the length of morphine treatment: ten days (M-10), twenty-eight days (M-28), ten days with seven days of regression (RM-10 twenty-eight days with seven days of regression (RM-28) and a control group (K). The loss of total opioid receptor number was noticeable after ten days and grew slightly during continuous morphine treatment and kept lowering in the period of regression. The total loss was approximately 30% of the control binding. The equilibrium dissociation constant (Kd), thus the affinity of [3 H]Diprenorphine wasn't significantly different among the groups. Drug addiction, opiates respectively, is a social problem which seriousness is currently on the rise. One of key elements causing addiction is tolerance to increasing doses of drug causing abstinence syndrome during withdrawal and craving. Opioid receptors are members of a large group of receptors coupled with heterotrimeric G-proteins (GPCR), whose properties can be investigated using agonist- stimulated binding [35 S] GTPγS. Many extracellular signals are transferred into a cell through GPCR. Opioid receptor agonists inhibit the activity of adenylyl cyclase and are coupled with G-protein group Gi/Go. This work is devoted to the study of changes in isolated plasma membranes of rat forebrain containing opioid receptors of healthy subjects with membranes acquired from morphine addicted subjects. The rats were long-term morphine treated in increasing doses, to develop the dependency. The comparison is done firstly by binding of [3 H]ouabain to Na,K-ATPase, which proves to be a negative standard of changes, secondly by binding [35 S]GTPγS to G-proteins, thereby providing the functional activity of G-protein in stimulating the binding by the agonist of δ-opioid receptors DADLE or agonist of µ-opioid receptors DAMGO. Este trabalho corresponde à dissertação do Mestrado Forense, com especialidade em Direito e Processo Civil, e destina-se a analisar a figura de direito civil da compensatio lucri cum damno e a sua expressão na jurisprudência portuguesa ao longo das últimas duas décadas. Para tal, é importante que seja feita, em primeiro lugar, uma breve menção à teoria da indemnização, com apelo à fórmula da diferença consagrada no artigo 566.º, n.º 2 do Código Civil, de modo a situarmos juridicamente a figura que serviu de base a esta dissertação e também uma menção ao próprio conceito de dano, elemento essencial da responsabilidade civil e também elemento essencial da figura do nosso estudo. De seguida, passaremos a fazer uma exposição da compensatio lucri cum damno, fazendo um percurso pelos seus pressupostos de aplicação e a sua presença no plano legal. Antes de passarmos à parte fulcral da nossa dissertação, faremos ainda referência à distinção entre este instituto e algumas figuras afins presentes no direito civil, de modo a evitar possíveis confusões por parte do leitor. Chegados à análise jurisprudencial em torno deste instituto, o método a seguir poderá não ser o mais convencional. A pesquisa que antecedeu este trabalho baseou-se na leitura aprofundada de 60 acórdãos dos tribunais superiores portugueses, judiciais e administrativos. Através desta pesquisa, conseguimos encontrar uma espécie de padrão no que diz respeito à compensatio lucri cum damno. Assim, para tornar a nossa exposição mais clara, iremos apresentar transversalmente os grupos de casos em que a compensatio lucri cum damno surge e é (ou não, como veremos) objeto de aplicação, ao invés de especificarmos a jurisprudência de cada instância, digase distinguir, a jurisprudência neste tema entre Supremos e Relações/ Tribunais Centrais. Será relevante também destacar alguns acórdãos, que por motivos mais adiante expostos, se mostraram interessante ou mereceram destaque neste nosso estudo. Concluindo esta dissertação, apresentaremos as nossas conclusões quanto à aplicação desta figura pelos nossos tribunais na esperança de termos feito um pequeno contributo para o estudo da compensatio lucri cum damno.

In open channel hydraulics, Froude number, which contains two variables: water depth and water velocity, is extensively applied to describe flow type. To utilize single hydraulic variable as Froude number to simplify two variables, water depth and water velocity to describe the difference of flow types. The study sites were located in Dago Stream, Neihu district, Taipei City, Taiwan, with aim to find the confident intervals of fish habitat occurrence proportion and Froude number distribution in regular flow. After fieldwork in Dago stream, there are eight aboriginal fishes, however the Scaphesthes barbatulus and is excluded from this statistic due to the small sample sizes. The Hemibarbus labeo habitat was most likely to occur at lower Froude number class from 0.3 to 0.4. But the Monopterus albus was most likely to occur at higher Froude number class from 0.4 to 0.8. Zacco pachycephalus, Acrossocheilus paradoxus, Rhinogobius brunneus and Candidia barbata were extensively existed the range of Froude number between 0.3 to 1, but the optimum habitats of Rhinogobius brunneus and Candidia barbata appear from 0.8 to 1.1. These information can be applied to reconstructe an Environment of habitat heterogeneity and biodiversity which depends on changes of channel morphology when torrent control.

Esta investigação insere-se no campo da Etnologia e tem por objectivo o estudo comparativo das actividades lúdicas tradicionais, praticadas, ainda hoje, em Portugal e em Goa, Damão e Diu; enquadrou-se em contextos geográficos situados em continentes distintos, mas sofrendo na Índia, uma forte influência socio-cultural portuguesa, durante cerca de 450 anos. A metodologia utilizada baseou-se na recolha etnográfica servindo-se da observação participante para o registo e recolha de dados e do método comparativo para a sua análise. Foram valorizados os registos, áudio, vídeo e fotográfico assim como a recolha de materiais, como meios de preservar o património lúdico. Dos resultados obtidos verificou-se haver, ainda hoje, práticas lúdicas comuns - jogos, danças, rondas, jogos cantados infantis, assim como festividades e tradições populares dos adultos. São analisadas as semelhanças e as influências encontradas. Do estudo dos objectos lúdicos observaram-se semelhanças nas improvisações feitas com os materiais utilizados. Da abordagem que fizemos aos espaços, ressaltou a importância do sítio do jogo. Desta investigação resulta a necessidade de, em futuros estudos, aprofundar fontes ligadas às artes plásticas, à música, ao cinema, à literatura e de prestar especial atenção às novas formas de criação e comunicação. A preservação deste património lúdico poderá ajudar a manter a identidade cultural, assim como contribuir para a continuidade da existência dos laços que ligam Portugal e Goa, Damão e Diu.<br>The main purpose of this research, in the field of Ethnology, is the comparative study of the traditional ludic activities which are still performed today, both in Portugal and in Goa, Daman and Diu (India). The study took place in geographical contexts situated in different continents, but undergoing in India a strong Portuguese socio-cultural influence for about 450 years. The research methodology was based on an ethnographic approach. Participant observation was used to collect and record data which were analysed using the comparative method. Audio and video recordings, photographs, as well as the collection of materials, were chosen as a privileged way of preserving the ludic heritage. The results of the study indicate that nowdays there are still common ludic activities - games, dances, round dances, children's singing games, as well as festivities and popular traditions in general. Similarities and influences are analysed. The study of ludic props evidences sirnilar ways of improvising with the materials used. The approach to the study of space showed the importance of the sense ofplace. This research points out to the fact that future studies should further investigate sources connected to the Visual Arts, Music, Cinema, Literature, and also pay special attention to new forms of creativity and communication. The preservation of this ludic heritage may help to maintain the cultural identity as well as the bonds that link Portugal to Goa, Daman and Diu.

<p>5-azacytidine and its derivatives are cytidine analogs used for leukemia chemotherapy. The primary effect of 5-azacytidine is the prohibition of cytosine methylation, which results in covalent DNA-methyltransferase crosslinks at cytosine methylation sites. These DNA-protein crosslinks have been suggested to cause chromosomal rearrangements and contribute to cytotoxicity, but the detailed mechanisms of DNA damage and the repair pathways of DNA-protein crosslinks have not been elucidated. </p><p>We used 2-dimensional agarose gel electrophoresis and electron microscopy to analyze plasmid pBR322 replication dynamics in Escherichia coli cells grown in the presence of 5-azacytidine. 2-dimensional gel analysis revealed the accumulation of specific bubble- and Y-molecules, dependent on overproduction of the cytosine methyltransferase EcoRII and treatment with 5-azacytidine. Furthermore, a point mutation that eliminates a particular EcoRII methylation site resulted in disappearance of the corresponding bubble- and Y-molecules. These results imply that 5-azacytidine-induced DNA-protein crosslinks block DNA replication in vivo. RecA-dependent X-structures were also observed after 5-azacytidine treatment. These molecules may be generated from blocked forks by recombinational repair and/or replication fork regression. In addition, electron microscopy analysis revealed both bubbles and rolling circles after 5-azacytidine treatment. These results suggest that replication can switch from theta to rolling circle mode after a replication fork is stalled by a DNA-methyltransferase crosslink. The simplest model for the conversion of theta to rolling-circle mode is that the blocked replication fork is cleaved by a branch-specific endonuclease. Such replication-dependent DNA breaks may represent an important pathway that contributes to genome rearrangement and/or cytotoxicity. </p><p>In addition, we performed a transposon mutagenesis screen and found that mutants defective in the tmRNA translational quality control system are hypersensitive to 5-azacytidine. The hypersensitivity of these mutants requires expression of active methyltransferase, indicating that hypersensitivity is dependent on DNA-methyltransferase crosslink formation. Furthermore, the tmRNA pathway is activated upon 5-azacytidine treatment in cells expressing methyltransferase, resulting in increased SsrA tagging of cellular proteins. These results support a "chain-reaction" model, in which transcription complexes blocked by 5-azacytidine-induced DNA-protein crosslinks result in ribosomes stalling on the attached nascent transcripts, and the tmRNA pathway is invoked for cleaning up the resulting pile-ups. In support of this model, an ssrA mutant is also hypersensitive to antibiotic streptolydigin, which blocks RNA polymerase elongation. These results reveal a novel role for the tmRNA system in clearance of coupled transcription/translation complexes in which RNA polymerase has become blocked.</p><br>Dissertation

Tese de mestrado, História Contemporânea, Universidade de Lisboa, Faculdade de Letras, 2012<br>O presente estudo debruça-se sobre a experiência dos prisioneiros de guerra portugueses após a queda do Estado da Índia Portuguesa em 1961 e 1962. Este é um caso ainda pouco estudado pela historiografica portuguesa, pelo que permanece uma história pouco desenvolvida e com muitos silêncios que aqui pretendemos clarificar. Cinquenta anos passados, pretendemos estudar o ocorrido para tentar compreender de forma mais completa os acontecimentos e as circunstâncias em que ocorreram. Através de um método objectivo e descritivo, o processo utilizado centrou-se na recolha arquivística e na reunião e análise de memórias dos intervenientes, relatos pessoais dispersos e distintos nas visões apresentadas das experiências. Os antecedentes que levaram à situação, os 5 meses de cativeiro, as experiências nos diferentes campos de prisioneiros e o complexo processo de negociações tiveram efeitos que influenciaram as transformações que o regime político vigente sofreu, nomeadamente através da consciencialização e ruptura do poder militar com o poder político, que culminou num conjunto de processos que resultaram no fim do regime do Estado Novo e na transição para um novo regime político.<br>Abstract: The present study focuses on the experience of the portuguese prisoners of war after the fall of the Portuguese State of India Portuguesa in 1961. This case is still little studied by the portuguese historiography. The story remains undeveloped with many silences that we want to clarify here. Fifty years after the events, we intend to study the incident to try to understand more fully the events and circumstances in which they occurred. Through an objective and descriptive method, the process used focused on the collection and archival collection and analysis of the memories of the people involved, dispersed and that represent diverses views of their experiences. The background that led to the situation, the five months of captivity, the experiences in the various prison camps and the complex negotiation process had effects that were influential in the transformations on the awareness and rupture of the military power in the support of the regime, that resulted in the end of the portuguese empire and the transition to a new political regime.
