Retinal diseases such as age-related macular degeneration (AMD) are the major cause of blindness in the developed world. Early diagnosis of these diseases is difficult as symptoms appear only at advanced stages. Nevertheless, several studies suggest that impairment of dark adaptation (the ability of the retina to adapt to low lighting) is a cue to AMD. Age-related macular degeneration (AMD) is the major cause of blindness in older people in developed countries. Although the early stages of the disease are thought to be symptomless, it is known that rod function systematically deteriorates as the disease progresses. The rod-mediated sensitivity recovery, called dark adaptation (DA), is abnormal in early AMD and it is clear this alteration precedes changes in visual function such as visual acuity (VA). Light adaptation optimises visual responses to illumination changes. Although it is well established that adaptation is faster for cone than rod photopigments, these dynamics are unknown for melanopsin photopigments in human eyes. This thesis reports the development of a novel method for characterising the melanopsin photobleach regeneration characteristic and its effect on the cone-driven pupil light reflex. The first chapter explains the motivation for measuring rod function, in particular the rod’s dynamic recovery from a substantial bleach which results in so-called ‘rate limited’ recovery of sensitivity. The physiological processes that underpin the replenishment of the rod photopigment are described and discussed, and explain the way in which rod function can act as a marker for retinal health. Overall, this chapter explains why rod function is worthy of further investigation.Then follows a description of the experimental methods used in the study of rod function, presented in later chapters.

Here we measure the contrast discrimination thresholds for light and dark bars individually, and find that the contrast discrimination thresholds for the whole sinusoid can be explained as ideal summation of the light and dark bar thresholds. We propose a model for light bar, dark bar, and sinusoidal contrast discrimination which involves local light adaptation and multiplicative noise. The model accounts for the data very well, and also accounts for contrast discrimination of light and dark edges.

Vision requires the photoreceptors in the eye to rapidly respond to changes in light intensity. These processes are accomplished within rod photoreceptors by the visual pigment rhodopsin that initiates a downstream signaling cascade called phototransduction. Rhodopsin is composed of an apoprotein opsin that is covalently bonded with light sensitive 11-cis retinal. Rhodopsin is activated when 11-cis retinal is photoisomerized into all-trans retinal.
