Full textHost cell proteins (HCPs) are unwanted by-products in the production of monoclonal antibodies (mAbs), and even at low levels, they can affect the safety, efficacy, and stability of biopharmaceuticals. While ELISA is widely used for HCP detection, it lacks full impurity coverage. This work explores complementary mass spectrometry-based methods to address these limitations. An immune-capture MS approach targets non-immunoreactive HCPs missed by ELISA, while advanced LC-MS/MS workflows using peptide standards enable more accurate and flexible quantification. These tools aim to improve impurity profiling and strengthen quality control in mAb manufacturing
