Abstract:
Internet of Things (IoT) technologies are undeniably already all around us, as we stand at the cusp of the next generation of IoT technologies. Indeed, the next-generation of IoT technologies are evolving before IoT technologies have been fully adopted, and smart dust IoT technology is one such example. The concept of smart dust IoT technology, which features very small devices with low computing power, is a revolutionary and innovative concept that enables many things that were previously unimaginable, but at the same time creates unresolved problems. One of the biggest problems is the bottlenecks in data transmission that can be caused by this large number of devices. The bottleneck problem was solved with the Dual Plane Development Kit (DPDK) architecture. However, the DPDK solution created an unexpected new problem, which is called the mixed packet problem. The mixed packet problem, which occurs when a large number of data packets and control packets mix and change at a rapid rate, can slow a system significantly. In this paper, we propose a dynamic partitioning algorithm that solves the mixed packet problem by physically separating the planes and using a learning algorithm to determine the ratio of separated planes. In addition, we propose a training data model eXtended Permuted Frame (XPF) that innovatively increases the number of training data to reflect the packet characteristics of the system. By solving the mixed packet problem in this way, it was found that the proposed dynamic partitioning algorithm performed about 72% better than the general DPDK environment, and 88% closer to the ideal environment.

Abstract:
The injection of an antagonistic anti-murine c-kit monoclonal antibody ACK2 during mouse embryonic development produced three distinctive pigmentation patterns on the coat of the offspring. Pattern 1 consisted of pigmentation in craniofacial and caudal regions and was induced by an ACK2 injection between 9.5 and 11.5 days post coitum (dpc). In pattern 2, the entire coat was unpigmented and was induced by the injection at around 13.0 dpc. Pattern 3 consisted of pigmented patches spreading ventrolaterally from the dorsoanterior trunk regions towards the anterior and posterior directions and it was induced by ACK2 administered at 14.5-15.0 dpc. We investigated the embryological basis of these nonuniform pigmentation patterns to elucidate the process of melanoblast differentiation between lineage commitment and colonization into developing hair follicles. The results showed the following. (1) Melanocyte differentiation at the embryonic stage from 10.5 to 12.5 dpc progresses in a spatially nonuniform fashion, being faster in the craniofacial and caudal regions than in the trunk; pattern 1 reflects this. (2) Melanoblasts are activated to proliferate synchronously upon entering into the epidermis; pattern 2 correlates with this process. (3) c-kit functions as a survival signal for proliferating melanoblasts in the epidermis. (4) The melanoblasts that enter developing hair follicles can survive without a c-kit signal; pattern 3 essentially represents the hair follicles colonized by these cells. Analysis of the melanoblast distribution of ls/ls embryos that bear a loss-of-function mutation in the endothelin 3 gene suggested that endothelin 3 is required for early melanoblast differentiation before entering into the epidermis, whereas proliferation in the epidermis takes place without this molecule. Based on these data, we propose 4 distinct steps of embryonic melanocyte differentiation: (1) migration in the dermis, which requires both c-kit and endothelin 3; (2) a state before epidermal entry that is resistant to anti-c-kit mAb; (3) cell proliferation after entering the epidermal layer, which requires c-kit and endothelin receptor B but not endothelin 3 and (4) integration into developing hair follicles, which renders melanoblasts resistant to anti-c-kit mAb. Thus, melanoblast differentiation proceeds by alternately repeating c-kit -dependent and c-kit-independent stages and c-kit functions as a survival factor for the proliferating melanoblasts.

Background/Objectives: For data center-based enterprises, the use of data traffic continues to increase. As a result, the increasingly efficient processing of increasing data traffic is becoming a major issue. Effective processing of traffic can be performed through the addition of physical network equipment, but this requires considerable costs.Methods/Statistical analysis: To solve this problem, researches on server virtualization and network virtualization technology are being actively conducted. However, while network virtualization is a cost-effective solution, open-source virtual switches used in network virtualization technologies have the performance penalty because they handle packets at the kernel of server's operating system. As a result, the Intel has developed the Data Plane Development Kit (DPDK) technology to address the performance degradation of the operating system.Findings: In this paper, we propose the DPDK performance test environment based on Open Virtual Switch (OVS) to verify the stability and performance of the DPDK-oriented virtualization technology, in association with a new KREONET-S project which aims tothe softwarization of the Korea Research Environment Open Network (KREONET) infrastructure.Improvements/Applications: We perform conducted experiments on Software-Defined Local Area Network (LAN) and Wide Area Network (WAN) environment using KREONET-S show the result.

Abstract:
In implementing the 2013 curriculum, Higher Order Thinking Skills (HOTS) must be integrated into learning activities. Similarity are sub material of similarity and congruence that students must learn. In fact, the higher order thinking skills of junior high school students in similarity material is low. To overcome this problem, it is necessary to develop a learning materials for learning in the form of plane similarity according to the level of student thinking in geometry, which is based on Van Hiele's theory to improve students' higher-order thinking skills. Van Hiele's theory was chosen because it suggests the development of students' thinking in learning of geometry occurs through five levels sequentially so that learning devices can be developed according to the level of van Hiele students. The purpose of this research is to obtain a description of the learning materials of plane similarity based on van Hiele theory based on van Hiele theory to improve students' high-level thinking skills that are valid, practical, and effective. This type of research is a research development using the 4-D model proposed by Thiagarajan then simplified to a 3-D model. Validation of learning devices, participant observation, questionnaires, and tests are the data collection techniques used. Abstract:
MicroRNAs (miRNAs) belong to the class of small non-coding RNAs which have important roles throughout development as well as in plant response to diverse environmental stresses. Some of plant miRNAs are essential for regulation and maintenance of nutritive homeostasis when nutrients are in excess or shortage comparing to optimal concentration for certain plant species. Better understanding of miRNAs functions implies development of efficient technology for profiling their gene expression. We set out to establish validate the methodology for miRNA gene expression analysis in cucumber grown under suboptimal mineral nutrient regimes, including iron deficiency. Reverse transcription by ?stem-loop? primers in combination with Real time PCR method is one of potential approaches for quantification of miRNA gene expression. In this paper we presented a method for ?stem loop? primer design specific for miR398, as well as reaction optimization and determination of Real time PCR efficiency. Proving the accuracy of this method was imperative as ?stem loop? RT which consider separate transcription of target and endogenous control. The method was verified by comparison of the obtained data with results of miR398 expression achieved using a commercial kit based on simultaneous conversion of all RNAs in cDNAs.

Abstract:
Undergraduate college “science partners” provided content knowledge and a supportive atmosphere for K–5 teachers in a university–school professional development partnership program in science instruction. The Elementary Science Education Partners program, a Local Systemic Change initiative supported by the National Science Foundation, was composed of four major elements: 1) a cadre of mentor teachers trained to provide district-wide teacher professional development; 2) a recruitment and training effort to place college students in classrooms as science partners in semester-long partnerships with teachers; 3) a teacher empowerment effort termed “participatory reform”; and 4) an inquiry-based curriculum with a kit distribution and refurbishment center. The main goals of the program were to provide college science students with an intensive teaching experience and to enhance teachers' skills in inquiry-based science instruction. Here, we describe some of the program's successes and challenges, focusing primarily on the impact on the classroom teachers and their science partners. Qualitative analyses of data collected from participants indicate that 1) teachers expressed greater self-confidence about teaching science than before the program and they spent more class time on the subject; and 2) the college students modified deficit-model negative assumptions about the children's science learning abilities to express more mature, positive views.

Abstract:
The Internet of Things (IoT) combines many devices with various platforms, computing capabilities and functions. The heterogeneity of the network and the ubiquity of IoT devices place increased demands on security and privacy protection. Therefore, cryptographic mechanisms must be strong enough to meet these increased requirements, but at the same time they must be effective enough to be implemented on devices with disabilities. One of the limited devices are microcontrollers and smart cards. This paper presents the performance and memory limitations of modern cryptographic primitives and schemes on various types of devices that can be used in IoT. In this article, we provide a detailed assessment of the performance of the most commonly used cryptographic algorithms on devices with disabilities that often appear on IoT networks. We relied on the most popular open source microcontroller development platform, on the mbed platform. To provide a data protection function, we use cryptography asymmetric fully homomorphic encryption in the binary ring and symmetric cryptography AES 128 bit. In addition, we compared run-time encryption and decryption on a personal computer (PC) with Windows 7, the Bluetooth Low Energy (BLE) Nano Kit microcontroller, the BLE Nano 1.5, and the smartcard ML3-36k-R1.

Abstract:
Abstract Background: A large, urban community hospital developed an insertion bundle to support the safe implementation of a policy of extended dwell time (clinical indication) for inpatient peripheral intravenous lines (PIVs). Methods: Internal evaluation of practices through direct observations as well as evidence-based guidelines and historic data on PIV-related bloodstream infections helped drive the bundle elements. A surveillance plan was in place to continue measurement of these outcomes during the postimplementation period. Results: At 12 months following implementation, the organization documented a 37% reduction (P = .03) in primary bacteremias (combining PIV and central line-associated bloodstream [CLABSI] infections) and a 19% percent reduction in PIV bloodstream infections. CLABSI rates were also reviewed, as 20% of CLABSI were noted to also have peripheral access present during the year prior to implementation. CLABSI standardized infection ratios for the publicly reported intensive care units decreased from 1.3 to 0.32 (P = .02). In addition, intravenous line start kit use decreased 48% during the year following bundle implementation. Conclusions: Careful planning and development of an education bundle and an insertion bundle in a community hospital setting allowed for longer dwell times and a trend of decreased bloodstream infections.

Abstract:
Background Maternity services have a unique opportunity to support women and families to build resilience and mitigate against the harmful impact of parental exposure to adverse childhood experiences (ACEs) but, most importantly, to prevent exposure to ACEs in future generations. Aim To identify ACEs in families who use maternity services in order to improve the professional response to risk, build parental resilience and strengthen parenting capacity. Methods A quality improvement project piloted an ACEs screening tool with 44 women and their partners when booking for maternity services. Implementation was supported by the development of a range of bespoke tools. Evaluation took place through quantitative data analysis and qualitative feedback from professionals and parents. Findings The use of the ACEs screening tool successfully identified ACEs which would otherwise not have been known using the previous antenatal booking questions. The bespoke tool kit was well-received by women, their partners and professionals. Identification and discussion of ACEs enabled appropriate support to be offered. Conclusion Identifying ACEs in maternity services and offering additional support requires further work before wider implementation. These interventions have the potential to reduce risk, build resilience and strengthen parenting capacity which could protect infants from experiencing a cycle of adversity.

Abstract:
The culture environment affects both the maternal and the embryonic expression of genes, and is likely to alter both oocyte and embryo developmental competence. Search for better and less variable culture conditions simulating the in vivo environment has led to the development of defined culture media, with lower negative impact on the molecular reprogramming of oocytes and embryos. This study aimed to evaluate the embryo development and relative expression of Hsp-70 and Bax transcripts in bovine blastocysts produced from oocytes matured in chemically defined IVM medium supplemented with insulin-like growth factor (IGF)-I, insulin, and polyvinyl alcohol (PVA) or polyvinylpyrrolidone 40 (PVP) without FSH and LH. Immature cumulus–oocyte complexes (COCs) from slaughtered cows were matured for 24 h in α-MEM plus IGF-1 and insulin and supplemented with 0.1% PVA (treatment 1; n = 410) or 0.1% PVP (treatment 2; n = 350). Neither FSH nor LH was used in both treatments. The control group consisted of COCs matured in TCM plus FSH and 10% estrous cow serum (n = 400). In vitro fertilization took place in Fert-Talp droplets for 22 h. Presumptive zygotes were co-cultured with cumulus cells in CR2aa supplemented with 10% fetal calf serum under mineral oil in a humidified atmosphere of 5% CO2 at 38.5C. Preimplantation embryo development was determined by cleavage, blastocyst, and hatching rates. For RNA extraction, blastocysts at Day 8 post-fertilization were frozen in liquid nitrogen and subsequently thawed. Total RNA extraction was performed using Rneasy Micro kit (Qiagen, Valencia, CA, USA) and first strand synthesized using Superscript III First Strand Synthesis kit (Invitrogen, Chicago, IL, USA). Relative quantification was performed in duplicate using real-time PCR (ABI Prism 7000 Applied Biosystem, Foster City, CA, USA); reactions consisted of a mixture of iTaq SYBR Green Supermix with ROX (Bio-Rad, Waltham, MA, USA) with cDNA equivalent to 0.8 embryos and specific primers for Hsp-70 and Bax genes, using as endogenous reference the expression of H2a gene. Data of embryo development were analyzed by chi-square test and calculations of relative quantification were performed by REST (Relative Expression Software Tool; Pfaffl et al. 2002 Nucl. Acids Res. 30, e36), using the value found in the control group as calibrator. Cleavage rate was higher (P < 0.05) for the control group (67%) than for treatments 1 (54%) and 2 (59%). Nevertheless, there was no difference (P > 0.05) in blastocyst (40.2%, 45.0%, and 43.5%) and hatching (78%, 77%, and 75%) rates among treatments 1 and 2 and the control group, respectively. Similarly, no difference (P > 0.05) in relative expression of Hsp-70 and Bax transcripts (0.69 0.69 and 0.53 0.32 for treatment 1, and 1.08 0.75 and 0.70 0.47 for treatment 2) in comparison to the calibrator group was detected. These results show that bovine oocytes can be matured in serum-free and gonadotrophin-free medium supplemented with PVA or PVP without altering post-cleavage development and relative expression of some genes associated with stress and apoptosis.

Abstract:
Abstract The MDS1-EVI1 locus is of considerable interest due to its role in myeloid malignancies and dysplasias. It is well established now that the locus has two different transcription start sites (TSS) located 0.5 Mb apart, and these have the capacity to encode different isoforms, which variably contain zinc finger DNA binding domains and a SET-like domain that may have histone modifying ability. In order to better understand the biological role of this locus, we knocked in a lacZ allele into the Mds1 (upstream) TSS by homologous recombination in ES cells and created mice harboring this modified allele (K. Lezon-Geyda, S. Lin, G. Steele-Perkins et al, in preparation). By staining for beta-galactosidase activity, we documented the distribution of Mds1 activity during embryonic development and in the adult. During development, five major organ systems showed expression: musculoskeletal, renal, cardiac, neural, and hematopoietic, and in the latter three, there was a striking and highly specific spatiotemporal pattern of expression suggesting that Mds1-Evi1 plays important regulatory roles. In the developing heart, staining was seen in the anterior heart field specifically during the formation of the cardiac outflow tract, with significant spatiotemporal overlap with Mef2c, which encodes an important cardiac transcriptional regulatory protein. Thereafter, expression in the heart is very low. Beta-gal staining in the hematopoietic system in the embryo is limited to the clusters of nascent hematopoietic progenitors that develop at day 9.5 p.c. in the ventrolateral wall of the dorsal aorta and bud into the vascular lumen. Strikingly, we see no staining in other endothelium, nor in the fetal liver of 12.5–14.5 day embryos, wherein the majority of fetal hematopoiesis takes place. In adult bone marrow, there is beta-gal activity exclusively in the lin− c-kit+ Sca1+ progenitor population, with all of the beta-gal-positive cells being in the progenitor pool, and nearly all of the progenitor cells staining. While homozygous mice are viable, they are small, kyphotic, and have a shortened lifespan. Morphologic and quantitative analysis of the peripheral blood failed to reveal any significant abnormality. To assess the function of the hematopoietic system more rigorously, competitive repopulations of homozygous Mds1-deficient marrow progenitors with wildtype progenitors were performed. Within several weeks after transplant, the Mds1−/− cells were undetectable in the recipients, revealing that the homozygous Mds1-null bone marrow progenitors are deficient in their repopulating ability. To identify what function Mds1-Evi1 plays in hematopoetic cells, we used shRNA to suppress its expression in the myeloid cell lines 32Dcl3 and DA-1. This revealed an increase in steady state levels of cell death, as documented by histone release, TUNEL staining, and caspase activation. These data suggest that a primary role for the Mds1-Evi1 locus in hematopoietic cells is to promote their survival, thus allowing normal expansion at the progenitor stage.

Abstract:
Background Digital phenotyping and machine learning are currently being used to augment or even replace traditional analytic procedures in many domains, including health care. Given the heavy reliance on smartphones and mobile devices around the world, this readily available source of data is an important and highly underutilized source that has the potential to improve mental health risk prediction and prevention and advance mental health globally. Objective This study aimed to apply machine learning in an acute mental health setting for suicide risk prediction. This study uses a nascent approach, adding to existing knowledge by using data collected through a smartphone in place of clinical data, which have typically been collected from health care records. Methods We created a smartphone app called Strength Within Me, which was linked to Fitbit, Apple Health kit, and Facebook, to collect salient clinical information such as sleep behavior and mood, step frequency and count, and engagement patterns with the phone from a cohort of inpatients with acute mental health (n=66). In addition, clinical research interviews were used to assess mood, sleep, and suicide risk. Multiple machine learning algorithms were tested to determine the best fit. Results K-nearest neighbors (KNN; k=2) with uniform weighting and the Euclidean distance metric emerged as the most promising algorithm, with 68% mean accuracy (averaged over 10,000 simulations of splitting the training and testing data via 10-fold cross-validation) and an average area under the curve of 0.65. We applied a combined 5×2 F test to test the model performance of KNN against the baseline classifier that guesses training majority, random forest, support vector machine and logistic regression, and achieved F statistics of 10.7 (P=.009) and 17.6 (P=.003) for training majority and random forest, respectively, rejecting the null of performance being the same. Therefore, we have taken the first steps in prototyping a system that could continuously and accurately assess the risk of suicide via mobile devices. Conclusions Predicting for suicidality is an underaddressed area of research to which this paper makes a useful contribution. This is part of the first generation of studies to suggest that it is feasible to utilize smartphone-generated user input and passive sensor data to generate a risk algorithm among inpatients at suicide risk. The model reveals fair concordance between phone-derived and research-generated clinical data, and with iterative development, it has the potential for accurate discriminant risk prediction. However, although full automation and independence of clinical judgment or input would be a worthy development for those individuals who are less likely to access specialist mental health services, and for providing a timely response in a crisis situation, the ethical and legal implications of such advances in the field of psychiatry need to be acknowledged.

Abstract:
Abstract Abstract 81 Hematopoietic stem cells (HSC) have the ability to self-renew and give rise to all hematopoietic lineage cells. Understanding signals that regulate the balance between self-renewal and differentiation of HSCs is an important issue in stem cell biology as well as regenerative medicine. Notch signals are critical regulators of the lymphoid lineage fate, but their role in adult HSC function is currently under debate. We explored the role of the LRF (Leukemia/Lymphoma Related Factor), a Notch repressor (also known as Zbtb7a, pokemon, OCZF and FBI-1) in HSC function, as it plays key roles in embryonic development, oncogenesis, and hematopoiesis. Conditional inactivation of the LRF gene in mouse HSCs (LRFF/FMx1-Cre mice) led to the development of CD4/CD8 DP (double positive) T-cells at the expense of B-cell development in the bone marrow (BM) in a Notch-dependent manner. Absolute numbers of the most primitive HSCs (LT-HSCs), defined as CD150+CD48−Flt3−Vcam-1+IL7Rα−LSK (Lin−Sca1+c-Kit+), were significantly reduced, while lymphoid-biased multi-potential progenitors (LMPPs: CD150−CD48+Flt3+Vcam-1+/−IL7Rα−LSK) and common lymphoid progenitors (CLPs: Lin−CD150−CD48+Flt3+Vcam-1−IL7Rα+) were barely detectable in LRFF/FMx1-Cre mice one month after pIpC injection. Enhanced T cell development and concomitant loss of B cell development was also seen in LRF−/− fetal liver (FL). Lin−IL7Rα+c-Kit+PIR+ (Paired Immunoglobulin-like receptors) T cell precursors were significantly increased in LRF−/− FL, indicating that Notch-mediated aberrant lymphoid fate determination also takes place during fetal hematopoiesis. To address which Notch gene(s) are targeted by LRF, we studied the HSC/progenitor population of conditional LRF knockout (LRFF/FMx1-Cre) as well as LRF/Notch1 double conditional knockout mice (LRFF/FNotch1F/FMx1-Cre). In the absence of Notch1, normal B cell development was restored in LRFF/FMx1-Cre mice. Reduction of LT-HSCs in LRFF/FMx1-Cre resulted from high Notch1 activity, as loss of Notch1 rescued LT-HSC numbers, suggesting that LRF functions to maintain HSCs and normal lymphoid fate by blocking Notch1. HSCs in active cell cycle are sensitive to 5-fluoro-uracil (5-FU) treatment, which causes remaining dormant HSCs to be recruited into the cell cycle to rapidly produce new cells and to quickly re-establish the hematopoietic system. To examine the self-renewal capacity of LRF deficient LT-HSC, LRFF/FMx1-Cre mice were treated with 5-FU after pIpC injection and the recovery of LT-HSC numbers examined. While control LT-HSC numbers recovered to pretreatment levels 3 wk after 5-FU treatment, levels in LRFF/FMx1-Cre mice remained low, accompanied by DP T cell development in the BM. Furthermore, after 5-FU treatment, LT-HSC numbers of LRFF/FNotch1F/FMx1-Cre were compatible to those of control and LRFF/FMx1-Cre mice, indicating that lack of self-renewal capacity in LRF deficient LT-HSCs was due to excessive differentiation toward T cells caused by Notch1. In support of this idea, when mice were given 5-FU weekly as a challenge to assess their HSC function in vivo, the survival percentage in LRFF/FMx1-Cre mice was much lower than in controls (0% versus 50% in 1 month, P <0.0001) and that of LRFF/FNotch1F/FMx1-Cre mice was compatible to controls. Serial bone marrow transplant experiments further demonstrated functional defects of LRF deficient HSCs, as they failed to reconstitute the hematopoietic system in secondary recipients. Microarray analysis and subsequent Gene Set Enrichment Analysis demonstrated upregulation of genes that were enriched in progenitor compartments. Since LRF can act as a transcriptional repressor, mRNA levels of Notch receptors and Notch ligands were examined using the same data set. A Notch target gene Hes1, but not Notch1 itself, was upregulated, and increased levels of Hes1 was also confirmed by real-time q-PCR in FACS-sorted LT-HSCs, as well as in 10.5 d.p.c whole embryos. These data suggest that LRF does not transcriptionally regulate Notch1, as LRF loss led to Notch1 target gene activation at the LT-HSC level without affecting Notch1 mRNA. Our genetic studies clearly indicate that LRF is indispensable for the maintenance of the HSC pool by repressing T cell-instructive signals mediated by Notch1 in the BM niche. Our findings shed new light on the regulatory mechanisms underlying the balance between HSC self-renewal and differentiation. Disclosures: No relevant conflicts of interest to declare.

Full text
Abstract:
Background The process of triaging is performed as an effective solution to balance limited resources against high patient volumes, based on an assessment of the patient’s medical condition and the application of an established patient categorization protocol. In Princess Marina Hospital (PMH), a national referral government hospital in the capital city of Botswana, the Princess Marina Hospital Accident and Emergency Centre Triage Scale (PATS) has been in use since 2010. Because the rules of these triage scales are very well defined, these protocols have been shown to be amenable to translation into computer algorithms. Thus, clinical decision support systems (CDSSs) that can assist with information management to support clinicians’ decision-making abilities can be developed, leading to improved healthcare quality and patient safety. Objective This study aims to determine the feasibility of development of a mobile triage app based on the adult PATS using Open Data kit (ODK) open source software to be used as a CDSS on smartphones and tablet computers for correct patient categorization. Methods A user-centered design approach was used in designing the app, with participants recruited from the staff at the Accident and Emergency Department (A&E) at PMH. Forty clinical vignettes were used in the evaluation of the performance of the app as compared to the paper-based system currently in use with the emergency physician at PMH providing the gold standard categorization of these vignettes. Usability testing was also performed. Results The app scored 90% (n=36) of the vignettes correctly, as compared to the paper-based system which scored 82.5% (n=33) of the vignettes correctly. Both systems achieved an over-triage score of 7.5% with an equal number of vignettes over-triaged (n=3). The results of the chi-square test indicate that the difference in triage scores between the paper-based system and the mobile app is statistically significant at P=.001 in favour of the ODK app. An overall positive outcome was also achieved in the usability test with ease of use and speed of triage determined to be the most recurring themes in the user feedback survey. While the app does not require an internet connection for triaging patients, a reliable wireless internet connection is required to upload data to the server for viewing by medical officers and physicians in real time, and this can be provided by the hospital as part of the Botswana government eHealth strategy. Additionally, the app developed in this research allows for data collection up to the point of triage categorization, meaning that a separate form would be required for capturing the rest of the information on the PMH A&E triage form. Conclusions The triage app developed in this research was found to determine the triage category of patient vignettes more accurately than the traditional paper-based system based on PATS triage guidelines with good results obtained in usability testing. Future work includes use of the app developed in this research in a live setting involving real patients in the A&E in PMH.

Abstract:
Mast cells are thought to participate in a variety of immune responses, such as parasite resistance and the allergic reaction. Mast cell development depends on stem cell factor (Kit ligand) and its receptor, c-Kit. Gab2 is an adaptor molecule containing a pleckstrin homology domain and potential binding sites for SH2 and SH3 domains. Gab2 is phosphorylated on tyrosine after stimulation with cytokines and growth factors, including KitL. Gab2-deficient mice were created to define the physiological requirement for Gab2 in KitL/c-Kit signaling and mast cell development. In Gab2-deficient mice, the number of mast cells was reduced markedly in the stomach and less severely in the skin. Bone marrow–derived mast cells (BMMCs) from the Gab2-deficient mice grew poorly in response to KitL. KitL-induced ERK MAP kinase and Akt activation were impaired in Gab2-deficient BMMCs. These data indicate that Gab2 is required for mast cell development and KitL/c-Kit signaling.

Abstract:
The thymus in mice lacking both the receptor tyrosine kinase c-kit and the common cytokine receptor γ chain (γc) is alymphoid because these receptors provide essential signals at the earliest stages of thymocyte development. The signals transduced by these receptors potentially regulate proliferation, survival, or differentiation, but the contribution of each receptor to distinct intracellular signaling cascades is only poorly defined. Here, we have examined whether enforced expression of Bcl-2 can rescue thymocyte development in c-kit and γc single or double mutant mice. A bcl-2 transgene (Eμ-bcl-2-25; expressed in the T cell lineage) was introduced into (a) c-kit and γc wild-type (c-kit+γc+bcl+), (b) c-kit–deficient (c-kit−γc+bcl+), (c) γc-deficient (c-kit+γc−bcl+), or (d) c-kit and γc double-deficient mice (c-kit−γc−bcl+). The bcl-2 transgene was functionally active in wild-type and c-kit or γc single mutants, as it promoted survival of ex vivo isolated thymocytes, including pro-T cells. In vivo, however, transgenic Bcl-2 did not release T cell precursors from their phenotypic block and failed to increase progenitor or total thymocyte cellularity in c-kit or γc single or double mutants. These data argue strongly against a role for Bcl-2 as a key mediator in signaling pathways linked to cytokine and growth factor receptors driving early thymocyte development.

Abstract:
Gastrointestinal stromal tumors (GISTs) are the commonest mesenchymal tumors of the gastroenteric tract, and are generally believed to originate from the neoplastic transformation of the interstitial cells of Cajal, the pacemaker structures of the stomach and intestine. Exon and genetic mutations (point/deletions) are fundamental for the development of GISTs: the constitutional characteristic of this neoplasm is the presence of the cell surface Kit receptor. Kit is the product of the proto-oncogene cKit, situated in chromosome 4. Ninety-eight percent of GISTs express mutated isoforms of Kit or of PDGFRA (Platelet growth factor receptor a). Kit mutation is the basic condition for autophosphorylation of tyrosine kinase residues in proteins. Autophosphorylation initiates pathogenetic processes in Cajal cells, toward a neoplastic transformation. Imatinib mesilate and, more recently, sunitinib are tyrosine kinase inhibitors, specific antagonists for Kit and PDGFRA, with good activity against GISTs. Most molecular and clinical data currently available concern imatinib. Exon mutations are strategic as prognostic and as predictive factors. In recent years, much evidence suggests that survival, response to therapy and resistance to imatinib are related to different mutations. In the near future, GIST patients will receive treatment differentiated by expressed Kit and PDGFRA mutations, thus truly individualized therapy.

<p>Student achievement in chemistry is relatively low because chemistry topics are not integrated with students’ expertise programs. The lack of variation in teaching material and learning model also reduces the motivation to study chemistry. One of the science-chemistry materials in vocational high schools is separation of mixture, taught in the agribusiness and agro technology specialization programs. Many business sectors such as cafes are starting to stand in the community, requiring coffee serving skills and also knowledge about coffee that can be viewed from the chemistry side. Chemical changes due to coffee cultivation and the processes through which coffee can be consumed can be learned through interactive and communicative teaching materials</p><p>One way to present and integrate coffee topics in learning is to develop an Edu-Kit which contains problems in coffee that are integrated with separation of mixture topic. Separation of mixture is contained in the 2013 Curriculum. The choice of problem-based learning models is assessed according to the characteristics of vocational learning, which is to present real problems and solve them while learning the concepts needed. The Edu-Kit development uses the research and development (R&D) method of Borg and Gall. E-book was chosen as an application in the development of Edu-Kit which includes text, sound, picture and video. Validation data were collected from 2 validators and readability tests by students. Data were analyzed using quantitative and qualitative techniques. The effectiveness of the Edu-Kit uses quasi experimental, pretest and posttest designs. There were 62 samples determined by the saturation sampling technique. The sample is divided into two classes, namely the experimental and control classes. Abstract:
Communication between mammalian oocytes and their surrounding granulosa cells through the Kit–Kit ligand (KL, or stem cell factor, SCF) system has been shown to be crucial for follicular development. Our previous studies (Reddy et al. 2005, Liu et al. 2006) have indicated that the intra-oocyte KL–Kit–PI3 kinase (PI3K)–Akt–Foxo3a cascade may play an important role in follicular activation and early development. In the present study, using in situ hybridization and in vitro culture of growing oocytes from 8-day-old postnatal mice, we have demonstrated that another Akt substrate, glycogen synthase kinase-3 (GSK-3), is expressed in growing oocytes. Also, treatment of cultured mouse oocytes with soluble KL not only leads to increased Akt kinase activity in the oocytes, which can phosphorylate recombinant GSK-3 in vitro, but also leads to phosphorylation of oocyte GSK-3α and GSK-3β, which can result in the inactivation of GSK-3 function in oocytes. In addition, we have shown that the regulation of GSK-3α and GSK-3β in cultured oocytes by soluble KL is accomplished through PI3K, since the PI3K-specific inhibitor LY294002 completely abolished the KL-induced phosphorylation of GSK-3α and GSK-3β. Moreover, blockage of the Kit signaling pathway by a Kit function-blocking antibody, ACK2, resulted in reduced phosphorylation of GSK-3. Taken together, our data suggest that the cascade from granulosa cell-derived KL to Kit–PI3K–Akt–GSK-3 in oocytes may take part in regulation of oocyte growth and early ovarian follicular development.

Nowadays, Software Defined Network (SDN) develops rapidly for its novel structure which separates the control plane and the data plane of network devices. Many researchers devoted themselves to the study of such a special network. However, some limitations restrict the development of SDN. On the one hand, the single controller in the conventional model bears all threats, and the corruption of it will result in network paralysis. On the other hand, the data will be increasing more in SDN switches in the data plane, while the storage space of these switches is limited. In order to solve the mentioned issues, we propose two corresponding protocols in this paper. Specifically, one is an anonymous protocol in the control plane, and the other is a verifiable outsourcing protocol in the data plane. The evaluation indicates that our protocol is correct, secure, and efficient.

Abstract:
Abstract Abstract 909 Hematopoietic stem cells (HSCs) are able to self-renew and differentiate into all blood lineages. Lineage commitment can take place at any stage from daughter cells of HSCs to multipotent progenitors. We have previously suggested that lineage commitment takes place at the level of HSCs (Takano et al. 2004). According to our asymmetric model, one HSC gives rise to two daughter cells consisting of one HSC and one lineage-committed progenitor. To verify this model, we performed a series of single-cell transplantation, followed by paired daughter cell experiments. The Ly5 congenic system has been commonly used to distinguish test donor cells from recipient cells and competitor cells. Since the Ly5 antigen is only expressed in leukocytes, platelets and erythrocytes have not been examined for evaluation of multi-lineage reconstitution. To assess lineage contribution more precisely, we have recently generated a transgenic mouse line in which Kusabira Orange (KuO) is expressed in all blood lineages including platelets and erythrocytes. CD150+CD41−CD34−/lowc-Kit+Sca-1+Lin− (CD150+CD41−CD34−KSL) cells, in which HSCs are highly enriched (Kiel et al. 2005, Morita et al. 2010), and CD150−CD41−CD34−KSL cells were individually isolated from KuO transgenic mice (B6-Ly5.1), and were transplanted into lethally irradiated mice (B6-Ly5.2), along with 2×105 bone marrow cells (B6-Ly5.1/Ly5.2). For secondary transplantation, 1×107 bone marrow cells from recipient mice were transplanted to lethally irradiated mice. Peripheral blood cells of primary and secondary recipient mice were periodically analyzed after transplantation. In some mice, full-lineage reconstitution was detected after transplantation with single CD150+CD41−CD34−KSL cells. In others, only common myeloid progenitor-type reconstitution was detected whereas common lymphoid progenitor-type reconstitution was not detected at all. Interestingly, megakaryocyte lineage (Meg)-restricted reconstitution and Meg/erythrocyte lineage-restricted (ME) reconstitution were observed after transplantation with single CD150+CD41−CD34−KSL cells, but not after transplantation with single CD150−CD41−CD34−KSL cells. These results suggest that Meg and ME progenitors are closely related to HSCs in their development. Furthermore, we performed paired daughter cell assays combined with single cell transplantation. Following division of CD150+CD41−CD34−KSL cells in culture, individual two daughter cells were separated by micromanipulation techniques and were transplanted into lethally irradiated mice with 2 × 105 competitor cells. Preliminary data showed in effect that the recipient mice of each single daughter cell derived from one CD150+CD41−CD34−KSL cell exhibited asymmetric differentiation; one with full-lineage reconstitution and the other with Meg-restricted reconstitution. Our findings offer a new perspective of HSC differentiation pathways. Disclosures: No relevant conflicts of interest to declare.

Abstract:
In many robotics studies, deep neural networks (DNNs) are being actively studied due to their good performance. However, existing robotic techniques and DNNs have not been systematically integrated, and packages for beginners are yet to be developed. In this study, we proposed a basic educational kit for robotic system development with DNNs. Our goal was to educate beginners in both robotics and machine learning, especially the use of DNNs. Initially, we required the kit to (1) be easy to understand, (2) employ experience-based learning, and (3) be applicable in many areas. To clarify the learning objectives and important parts of the basic educational kit, we analyzed the research and development (R&D) of DNNs and divided the process into three steps of data collection (DC), machine learning (ML), and task execution (TE). These steps were configured under a hierarchical system flow with the ability to be executed individually at the development stage. To evaluate the practicality of the proposed system flow, we implemented it for a physical robotic grasping system using robotics middleware. We also demonstrated that the proposed system can be effectively applied to other hardware, sensor inputs, and robot tasks.

Abstract:
Background and Aim: Newcastle disease (ND) is a viral infection that causes high mortality and economic loss in the poultry industry. The Office International des Epizooties (OIE) recommends several diagnostic methods for the detection of ND, including isolation and molecular tests. However, these detection methods are time-consuming and highly expensive. Therefore, this study was conducted to develop a coagglutination kit as a novel diagnostic tool for ND in the poultry industry. Materials and Methods: Two adult male New Zealand White rabbits weighing 2.5 kg were vaccinated using ND life vaccine intraperitoneally. The vaccination was conducted once a week for 4 weeks with multilevel doses. Rabbits' serum was collected at week 6 and inactivated at 56°C for 30 min. The serum was precipitated using ammonium sulfate and reacted with protein A of Staphylococcus aureus to produce the agglutination kit for detecting ND virus. A total of 25 chickens suspected with ND infection from a local poultry farm in Yogyakarta were used as the test samples. The chickens were necropsied, and the brain, spleen, lung, intestine, and feces were collected. Half of these organs were subjected to tests using the coagglutination kit and reverse transcription-polymerase chain reaction (RT-PCR). The other half was processed for histopathology. Data were analyzed qualitatively. Results: Of the 25 samples, 13 (52%) were positive for ND infection when tested using both the ND coagglutination kit and RT-PCR. The positive samples also exhibited several histopathological changes, including perivascular cuffing surrounding the cerebral blood-brain barrier, hemorrhagic pneumonia, splenitis, and necrotic hemorrhage enteritis. Conclusion: This study confirmed that the ND coagglutination kit could be used as a novel diagnostic tool for the detection of ND virus infection in the poultry industry.

Abstract:
Objective Development of a novel pediatric airway kit and implementation with simulation to improve resident response to emergencies with the goal of improving patient safety. Methods Prospective study with 9 otolaryngology residents (postgraduate years 1-5) from our tertiary care institution. Nine simulated pediatric emergency airway drills were carried out with the existing system and a novel portable airway kit. Response times and time to successful airway control were noted with both the extant airway system and the new handheld kit. Results were analyzed to ensure parametric data and compared with t tests. A Bonferroni adjustment indicated that an alpha of 0.025 was needed for significance. Results Use of the airway kit significantly reduced the mean time of resident arrival by 47% ( P = .013) and mean time of successful intubation by 50% ( P = .007). Survey data indicated 100% improved resident comfort with emergent airway scenarios with use of the kit. Discussion Times to response and meaningful intervention were significantly reduced with implementation of the handheld airway kit. Use of simulation training to implement the new kit improved residents’ comfort and airway skills. This study describes an affordable novel mobile airway kit and demonstrates its ability to improve response times. Implications for Practice The low cost of this airway kit makes it a tenable option even for smaller hospitals. Simulation provides a safe and effective way to familiarize oneself with novel equipment, and, when possible, realistic emergent airway simulations should be used to improve provider performance.

Abstract:
The tyrosine kinase KIT receptor, the protooncogene CD117, plays a key role in growth and maturation of oocytes and follicles. Relevant data are sparse for the corpus luteum (CL). We first confirmed the presence ofKITmRNA and KIT protein in bovine CL homogenates. We then localized KIT-positive (KIT+) cells in CL sections by immunohistochemistry. At the CL stage of early development, the former theca transforming into capsule/septa showed a strong band-like KIT+ immunoresponse. In addition, CD45+ leukocytes in septa included subpopulations of CD45+/KIT+ and CD14+/KIT+ leukocytes as validated by double immunofluorescence localization. At the early secretory stage, KIT+ cells appeared within the septa/capsule region and in the periphery of the CL parenchyma, there forming a complex network. This was separate from the capillary bed as determined by double staining for CD117 and FVIII-related endothelial cell antigen (FVIIIr). The KIT+ network coincided with cells positive for cytochrome P450 17α-hydroxylase, a thecal cell-specific enzyme. The late secretory stage was defined by an advanced manifestation of the KIT+ network in the CL periphery. At the stage of regression, the KIT+ network was absent. The CL of pregnancy expressed high levels ofKITmRNA and KIT protein uniformly throughout pregnancy. The KIT+ immunolocalization revealed small fibroblast-like cells, luteal cells with granules, and clusters of large luteal cells with staining of the cell membrane. We conclude that a majority of KIT+ cells in the bovine CL are primarily theca-derived small luteal cells, and that a minority represent KIT+ leukocytes, in some cases KIT+ monocytes.

Abstract:
Abstract INTRODUCTION. Germinal centers (GC) are unique sites in peripheral lymphoid tissue where clonal selection of B cells takes place. This occurs as a response to stimulation by various antigens originating, sometimes, follicular hyperplasia (FH). GC have been known as a major source of B-cell lymphomas including follicular (FL) and diffuse large cell (DLCL). DNA methylation of tumor-suppressor genes is a mechanism of gene silencing involved in the pathogenesis of FL and DLCL. Much less is known about the role of methylation in FH. We determined the methylation status of 6 tumor-suppressor genes in 43 patients with FH, 18 patients with FL and 49 patients with DLCL in order to see the differential implication of this epigenetic mechanism in the pathological and the physiological development of GC. MATERIAL AND METHODS. Genomic DNA extracted from paraffin-embedded samples of 43 FH, 18 FL and 49 DLCL after being treated with EZ DNA-Methylation Kit (Zymo Research) with the manufacturer’s instructions, were analyzed by methylation-specific polymerase chain-reaction to determine promoter hypermethylation of DAP-k, SHP1, Rarβ, p14, SHP1, MGMT and PDRM1. All samples were obtained mostly from lymph nodes and tonsils. Diagnosis was based on morphology and immunohistochemistry analysis. All cases were matched for age, sex and ethnic origin. RESULTS: DAP-k promoter methylation occurred with higher frequency in FL(89%) than in DLCL(78%) and FH(40%). SHP1 was methylated in 61% of FL, 58% of FH and 23% of DLCL. RARb was methylated in 67% of FL patients, 30% of DLCL and only 12% of FH. Eight (44%) FL, seventeen (35%) DLCL and four (10%) BFH patients showed MGMT methylation. Promoter hypermethylation of p14 was detected only in 5 (12%) FH, 2 (4%) DLCL and none FL patients. Methylation of PRMD1 was present only in 1 (6%) FL, 2 (6%) DLCL and 1 (4%) FH samples. CONCLUSIONS. Inactivacion of DAP-K and SHP1 is present in B-cell malignancies, DLCL and FL, and BFH. Therefore, it may represent a physiologic event conferring a temporal survival advantage necessary for a GC hyperplastic response. Inactivation of the retinoic acid response through the methylation of Rarâ is significantly more frequent in lymphomas than in FH. As reported in other tumors methylation of MGMT is more frequent in lymphomas than in FH. With our data methylation of Cyclin dependent kinase inhibitors p14 is not a differential pathogenic event in lymphomas of GC origin, in fact it is more frequent in FH. Promoter Methylation of PDRM1 is not the mechanism involved in lymphomagenesis in FL and DLCL, the two FH positive deserve further follow-up to determine its significance.

Abstract:
Reflector antennas have been widely used in many areas. In the implementation of parabolic reflector antenna for broadcasting satellite applications, it is essential for the spacecraft antenna to provide precise contoured beam to effectively serve the required region. For this purpose, combinations of more than one beam are required. Therefore, a tool utilizing ray tracing method is developed to calculate precise off-axis beams for multibeam antenna system. In the multibeam system, each beam will be fed from different feed positions to allow the main beam to be radiated at the exact direction on the coverage area. Thus, detailed study on caustics of a parabolic reflector antenna is performed and presented in this paper, which is to investigate the behaviour of the rays and its relation to various antenna parameters. In order to produce accurate data for the analysis, the caustic behaviours are investigated in two distinctive modes: scanning plane and transverse plane. This paper presents the detailed discussions on the derivation of the ray tracing algorithms, the establishment of the equations of caustic loci, and the verification of the method through calculation of radiation pattern.

The subject of the article is collective memory of the concept of work among the inhabitants of the Upper Silesian village of Bojszowy. The author of the study employs the classic understanding of collective memory, while simultaneously pointing to the importance of the communication process and the diversity of information sources shaping it. Empirical data was collected during research conducted in 2018 using in-depth interview techniques. The research sample consisted of 30 rural residents. Analysis of the functioning of collective memory about work revealed the development processes – especially industrialization and deagrarianization – taking place in Bojszów. The analysis clearly shows the specificity of the Upper Silesian village, where these processes began relatively early, and work was associated not only with agriculture, but also with industry and trade.
