ZusammenfassungHausaufgaben erscheinen im Fremdsprachenunterricht weiterhin als zwar stetig eingesetztes, aber eher ungeliebtes Anhängsel, um das sich keiner wirklich kümmert. Der Artikel fasst – vor allem auf der Grundlage allgemeinpädagogischer und -didaktischer Literatur – wichtige Ergebnisse zum Phänomen Hausaufgaben zusammen und formuliert Forschungsfragen und -desiderate für den Fremdsprachenunterricht. Anhand von drei Schwerpunktbereichen des DaF-/DaZ-Unterrichts zeigt er, wie veränderte Formen von Hausaufgaben in einem sich verändernden DaF-/DaZ-Unterricht aussehen bzw. aussehen könnten. Indem er Konzepte aus anderen Bereichen betrachtet (und etwas gegen den Strich denkt), zeigt er schließlich Perspektiven auf, wie die Zukunft der Hausaufgabenpraxis aussehen und wie (digital basierte) Hausaufgabenkonzepte den DaF/DaZ-Unterricht insgesamt verändern könnten.

Abstract:

DAZ (Deleted in Azoospermia), the putative azoospermia factor gene in human, encodes a ribonucleoprotein-type RNA-binding protein required for spermatogenesis. A Drosophila homologue of DAZ, called boule, is also essential for spermatogenesis. A mouse homologue, Dazla, is implicated in both spermatogenesis and oogenesis. Here, we report the identification and characterization of daz-1, the single DAZ homologue in the nematode Caenorhabditis elegans. Loss of daz-1 function caused sterility in hermaphrodites, by blocking oogenesis at the pachytene stage of meiosis I. Epistasis analysis suggested that this gene executes its function succeeding gld-1, which governs the early pachytene stage in the oogenic pathway. Spermatogenesis did not appear to be affected in daz-1 hermaphrodites. Males defective in daz-1 produced sperm fully competent in fertilization. Analysis employing sex-determination mutants indicated that the daz-1 function was required for meiosis of female germline regardless of the sex of the soma. Transcription of daz-1 was restricted to the germline, starting prior to the onset of meiosis and was most conspicuous in cells undergoing oogenesis. Thus, daz-1 in C. elegans is an essential factor for female meiosis but, unlike other DAZ family members so far reported, it is dispensable for male meiosis.

Abstract:

Integrating regional linguistic variation more heavily into classes of German as a foreign language (DaF) and German as a second language (DaZ) has been subject to discussion for quite some time in the research literature. Surveys have shown that students of DaF and DaZ are interested in learning about regional varieties of German, but that this often fails for two reasons: the lack of appropriate materials as well as reluctance on the part of teachers (cf. Baßler/Spiekermann 2001, Spiekermann 2007). Consequently, there are numerous non-standard variants of which students are not aware. It has also been observed that non-standard linguistic structures can lead to miscomprehension (cf. Negele 2012: 94). This con-tribution calls attention to two desiderata: 1. Areal linguistic variation is covered only sel-dom in DaF and DaZ classes. 2. There is insufficient suitable teaching material for treating and imparting knowledge on regional linguistic variants in these classes. The goal of the present contribution is then twofold: to provide teachers and textbook authors with information about the resources available over the research platform Regionalsprache.de (REDE) and then to give some impulses for its use in DaF and DaZ courses.

Abstract:

In diesem Beitrag werden in einem ersten Schritt die Inhalte und Ziele eines Seminars an der Universität Wien zum Aufbau von „Curriculumkompetenz“ bei angehenden DaF/DaZ-Lehrkräften erläutert. Zu Beginn, in der Mitte und am Ende des Wintersemesters 2020/21 reflektierten die SeminarteilnehmerInnen über die Entwicklung ihrer persönlichen Curriculumkompetenz im Rahmen von Online-Befragungen, die das Erhebungsinstrument für die Daten der in diesem Beitrag fokussierten Längsschnittstudie sind. Anschließend werden die Ergebnisse der drei Online-Befragungen präsentiert und diskutiert. Es zeigt sich, dass Studierende in ihrer universitären Ausbildung kaum je mit Fragen der Curriculumtheorie, Curriculumforschung sowie Curriculumentwicklung in Berührung kommen. Im Zuge der Leistungsbeurteilung mussten die Studierenden in sogenannten „Teacher Design Teams“ selbst DaF/DaZ-Curricula erstellen, was sich ihnen als eine ertragreiche Aufgabe darstellte. Als schwieriger hingegen gestaltete sich für sie die Curriculumforschung, die sie in ihren Seminararbeiten durchführen mussten.

Abstract:

The Deleted in Azoospermia (DAZ) gene family encodes putative translational activators that are required for meiosis and other aspects of gametogenesis in animals. The single Caenorhabditis elegans homologue of DAZ, daz-1, is an essential factor for female meiosis. Here, we show that daz-1 is important for the switch from spermatogenesis to oogenesis (the sperm/oocyte switch), which is an essential step for the hermaphrodite germline to produce oocytes. RNA interference of the daz-1 orthologue in a related nematode, Caenorhabditis briggsae, resulted in a complete loss of the sperm/oocyte switch. The C. elegans hermaphrodite deficient in daz-1 also revealed a failure in the sperm/oocyte switch if the genetic background was conditional masculinization of germline. DAZ-1 could bind specifically to mRNAs encoding the FBF proteins, which are translational regulators for the sperm/oocyte switch and germ stem cell proliferation. Expression of the FBF proteins seemed to be lowered in the daz-1 mutant at the stage for the sperm/oocyte switch. Conversely, a mutation in gld-3, a gene that functionally counteracts FBF, could partially restore oogenesis in the daz-1 mutant. Together, we propose that daz-1 plays a role upstream of the pathway for germ cell sex determination.

§ Die in Buchrezensionen vertretenen Ansichten und Meinungen sind die der jeweiligen Rezensentinnen und Rezensenten und reflektieren nicht notwendigerweise die Position von SCENARIO.§ Das Potenzial ästhetischen Lernens könnte im Fremd- und Zweitsprachenunterricht noch viel stärker genützt werden. Dies ist der Grundtenor des von Nils Bernstein und Charlotte Lerchner herausgegebenen Sammelbandes „Ästhetisches Lernen im DaF-/DaZ-Unterricht“, in dem auch gleich viele Möglichkeiten aufgezeigt werden, wie dieses Potenzial ausgeschöpft werden könnte. Der Band geht auf einen DaF-Kongress im März 2013 an der Universidad Nacional Autónoma de México (UNAM) in Mexiko-Stadt zurück. Die Buchpublikation wurde um die Dimension DaZ erweitert und versammelt 25 Beiträge mit theoretischen Überlegungen, empirischen Forschungsergebnissen und vor allem vielen praktischen Anregungen zum ästhetischen Lernen im DaF/DaZ-Unterricht. Dabei wird Ästhetik nicht nur im engen Sinne einer „allgemeinen Theorie des Schönen“, sondern auch in der ursprünglich viel weiter gefassten Bedeutung des altgriechischen Begriffs aisthesis verstanden, der „erst einmal nur wertfrei Wahrnehmung“ meint (vgl. u.a. den Beitrag von Susanne Even, S. 192). So können sinnliche Wahrnehmung und die damit einhergehenden Emotionen, Gedanken, Eindrücke als Voraussetzung für die Auseinandersetzung mit künstlerischen Qualitäten analysiert und fruchtbar gemacht werden. Abstract:

The azoospermia factor c (AZFc) region of the Y chromosome consists of repetitive amplicons and is therefore highly susceptible to structural rearrangements, such as deletions and duplications. The b2/b3 deletion is a partial AZFc deletion that is conventionally determined by the selective absence of sY1191 in sequence-tagged site polymerase chain reaction (PCR) and is generally believed to retain two of the four deleted in azoospermia (DAZ) genes on the Y chromosome. In the present study we determined the copy number and expression of DAZ genes in sY1191-negative individuals. Using a DAZ dosage PCR assay and Southern blot analysis we evaluated the expression of four DAZ genes in five of six sY1191-negative individuals. Furthermore, cloning and immunoblot analyses revealed that three or more DAZ genes are expressed in sY1191-negative testes with germ cells. The results indicate that the selective absence of sY1191 not only means b2/b3 deletion with two DAZ genes, but also includes another AZFc configuration with four DAZ genes. These results exemplify the prevalence of variations in the AZFc region of the human Y chromosome.

At this time the case of azoospermia is quite common in infertile men. Azoospermia is a condition where the semen does not contain sperm. Many causes azoospermia, including the deletion of a gene at the locus that is located on the Y chromosome long arm (YQ) known as AZF gene (Azoospermia Factor). One of the genes in the AZF region are genes that AZFc DAZ (Deleted in Azoospermia). The purpose of this study was to detect the presence of the DAZ gene deletions in men with azoospermia cases using PCR (Polymerase Chain Reaction). The study design was descriptive. Venous blood samples with EDTA anticoagulant taken from 10 men azoospeermia then extracted to obtain DNA. DNA samples were then carried out PCR with primers DAZ. The PCR products were separated by electrophoresis in 2% agarose gel and visualized using UV translluminator. Of the 10 samples, four patients including DAZ gene deletion was detected experience while the other six do not experience DAZ gene deletions. It concluded that found their DAZ gene deletions in men with azoospermia using the PCR method.

Zusammenfassung Ziel des Beitrags ist es, auf für den Kontext Deutsch als Fremdsprache (DaF) und Deutsch als Zweitsprache (DaZ) relevante Aspekte der medizinischen Kommunikation von Ärztinnen und Ärzten hinzuweisen, die einerseits durch die Neuentwicklung von Versorgungsstrukturen (z. B. Palliativversorgung) und andererseits durch die Reformbemühungen um den Stellenwert der klinischen Kommunikationsausbildung (z. B. „Masterplan Medizinstudium 2020“) bedingt sind. Dazu werden Überlegungen zu einer Neuzuschneidung, Erweiterung und Ausdifferenzierung der kommunikativen Kompetenz im Kontext von medizinischer Aus- und Weiterbildung angestellt. Abschließend werden die sich hieraus ergebenden Implikationen für die Vermittlung von DaF und DaZ für Ärztinnen und Ärzte dargestellt.

Abstract:

Deletions in distal Yq interval 6 represent the cause of 10–15% of idiopathic severe male infertility and map to a region defined AZFc (azoospermia factor c). The testis-specific gene DAZ is considered a major AZFc candidate, and its deletion has been associated with a severe disruption in spermatogenesis. However, DAZ is actually a multicopy gene family consisting of seven clustered copies spanning about 1 megabase. Only deletions removing the entire DAZ gene cluster together with other genes have been reported in infertile males. Because no case of spermatogenic failure has been traced to intragenic deletions, point mutations, or even deletions not involving all the DAZ copies, the definitive proof for a requirement of DAZ for spermatogenesis is still debatable. Here we report the first case of a partial deletion of the DAZ cluster removing all but one of the copies. This deletion is present in a patient affected with severe oligozoospermia who had a testicular phenotype characterized by a great quantitative reduction of germ cells (severe hypospermatogenesis). The absence of this deletion in the fertile brother of the patient suggests that this de novo mutation indeed caused the spermatogenic failure.

In today's society, we are witnessing an ever-increasing diversity, which is becoming more and more present through different channels and especially in the media. Language teaching is usually also teaching in perspective and behaviour and should consequently also reflect the current debates in society. Therefore, the question arises as to whether social discourses on the topic of diversity are finding their way into textbooks for German as a foreign and second language in a frequent and differentiated way. The article examines various aspects of diversity in six current textbooks and finally formulates proposals for an open approach to the topic.

O artigo tem por objetivo examinar os contextos de produção dos termos Alemão como Língua Estrangeira, Alemão como Segunda Língua, Alemão como Terceira Língua e Língua Adicional empregados na literatura científica para denotar uma L≠1. Mobiliza-se como base teórica desse estudo a abordagem discursiva SKAD (Sociology of Knowledge Approach to Discourse; KELLER 2011a, 2011b), que investiga regimes de saber em diferentes áreas e contextos e explora as condições de possibilidade de determinadas ordens do conhecimento. Argumenta-se que os termos Alemão como Língua Estrangeira e Alemão como Segunda Língua, cunhados nos países de língua alemã, atribuem aos aprendizes de alemão como não-L1 posições de sujeito que não são mais considerados apropriados e admissíveis por estes. Como contraposição, propõe-se o termo de língua adicional, a partir da perspectiva de quem não fala a língua como L1, com o objetivo de neutralizar certos efeitos de sentido hierárquicos que os outros dois termos implicam. Nesse ponto de vista, o fato de que a noção ampla de língua adicional não permite diferenciações conceituais em termos do local de aprendizagem e da qualidade do processo de apropriação da L≠1, não é visto como um problema e até mesmo considerado uma vantagem.

Abstract:

Mutations in Deleted in Azoospermia (DAZ), a Y chromosome gene, are an important cause of human male infertility. DAZ is found exclusively in primates, limiting functional studies of this gene to its homologs: boule, required for meiotic progression of germ cells in invertebrate model systems, and Daz-like (Dazl), required for early germ cell maintenance in vertebrates. Dazl is believed to have acquired its premeiotic role in a vertebrate ancestor following the duplication and functional divergence of the single-copy gene boule. However, multiple homologs of boule have been identified in some invertebrates, raising the possibility that some of these genes may play other roles, including a premeiotic function. Here we identify two boule paralogs in the freshwater planarian Schmidtea mediterranea. Smed-boule1 is necessary for meiotic progression of male germ cells, similar to the known function of boule in invertebrates. By contrast, Smed-boule2 is required for the maintenance of early male germ cells, similar to vertebrate Dazl. To examine if Boule2 may be functionally similar to vertebrate Dazl, we identify and functionally characterize planarian homologs of human DAZL/DAZ-interacting partners and DAZ family mRNA targets. Finally, our phylogenetic analyses indicate that premeiotic functions of planarian boule2 and vertebrate Dazl evolved independently. Our study uncovers a premeiotic role for an invertebrate boule homolog and offers a tractable invertebrate model system for studying the premeiotic functions of the DAZ protein family.

SummaryThe Deleted in AZoospermia (DAZ) gene family regulates the development, maturation and maintenance of germ cells and spermatogenesis in mammals. The DAZ family consists of two autosomal genes, Boule and Dazl (Daz-like), and the Daz gene on chromosome Y. The aim of this study was to analyze the localization of DAZL and BOULE during testicular ontogeny of the seasonal-breeding Syrian hamster, Mesocricetus auratus. We also evaluated the testicular expression of DAZ family genes under short- or long-photoperiod conditions. In the pre-pubertal and adult testis, DAZL protein was found mainly in spermatogonia. BOULE was found in the spermatogonia from 20 days of age and during the pre-pubertal and adult period it was also detected in spermatocytes and round spermatids. DAZL and BOULE expression in spermatogonia was strictly nuclear only in 20-day-old hamsters. We also detected the novel mRNA and protein expression of BOULE in Leydig cells. In adult hamsters, Dazl expression was increased in regressed testis compared with non-regressed testis and DAZL protein expression was restricted to primary spermatocytes in regressed testis. These results show that DAZL and BOULE are expressed in spermatogonia at early stages in the Syrian hamster, then both proteins translocate to the cytoplasm when meiosis starts. In the adult regressed testis, the absence of DAZL in spermatogonia might be related to the decrease in germ cell number, suggesting that DAZ gene family expression is involved in changes in seminiferous epithelium during photoregression.

ABSTRACT Men with severe infertility suffer a high risk of Y chromosome deletion, hence screening for these cases is recommended prior to treatment with assisted reproduction. Our study aimed to investigate and detect the azoospermia factor (AZF) region deletion, rearrangement and deleted azoospermia (DAZ) gene copy number variations in Egyptian azoospermic infertile men. This was tested on 54 Egyptian nonobstructive azoospermic (NOA) infertile men, with age ranged from 21 to 45 years (mean: 31.4 ± 6.1 years), by STS ± multiplex PCR using a set of 14 sequence tagged sites (STSs) from three different regions of the Y chromosome: AZFa, AZFb, AZFc and sY587/DraI PCRRFLP assay to determine DAZ copy number variations. The results revealed a significant prevalence of AZFc subtypes deletion and reduced DAZ gene dosage in Egyptian azoospermic cases affecting Y chromosome deletions. To our knowledge, this study is the first one to investigate AZFc subtypes deletion and DAZ gene dosage in Egyptian infertile men. Abstract:

The superalloy FGH98 was successfully diffusion bonded (DB) with medium-entropy alloy (MEA) Al3Ti3(CrCoNi)94 using pure Ni as the interlayer at a temperature range of 1050–1170 °C for 1 h under 5 MPa. The microstructure and mechanical properties of joints were investigated. The diffusion bonding seam was composed of an interlayer zone (IZ) and two diffusion-affected zones (DAZ). The IZ and DAZ beside the FGH98 consisted of cubic Ni3(TiAl)-type γ′ phases due to the diffusion of Ti and Al atoms. Meanwhile, the DAZ adjacent to the MEA consisted of spherical γ′ phases. Both of the γ′ phases with different morphology kept the coherent relationship with the matrix. Moreover, increase of bonding temperature led to the morphology of interlayer γ′ phase to transform from sphere to cube. Due to the strengthening effect of a mass of γ′ phase distributed evenly in IZ and the DAZ beside the FGH98, the microhardness and Young’s modulus of these two zones were higher than that of DAZ near the MEA. The maximum shear strength of DB joint, 592 MPa, was achieved in the joint bonded by 1150 °C, which was the typical ductile fracture feature confirmed by the shear dimples.

Abstract:

The amplification outcomes of two hearing aid prescriptions, NAL-NL1 and Digital Perception Processing (DPP), of nine moderate to moderately severe hearing-impaired adults were compared in the same digital hearing instrument. NAL-NL1 aims at optimizing speech intelligibility while amplifying the speech signal to a normal overall loudness level (Dillon, 1999). DPP focuses on restoring loudness based on normal and impaired cochlear excitation models (Launer and Moore, 2003). In this comparison, DPP resulted in better sentence recognition performance than the NAL-NL1 algorithm in the signal-front/noise-side condition, and the two prescriptions gave similar performance in the signal-front/noise-front condition. Subjective evaluations by the participants using the Abbreviated Profile for Hearing Aid Benefit and sound quality comparisons did not give conclusive results between the two prescriptions.With each hearing aid prescription, the ability of the hearing aid circuitry to reduce the effects of noise was evaluated by a sentence-in-noise test in three conditions: (1) adaptive directional microphone (DAZ), (2) multichannel noise reduction system (FNC), and (3) a combination of FNC and DAZ (FNC + DAZ). In the signal-front/noise-side condition, DAZ and FNC + DAZ gave better performance than FNC in nearly all participants, whereas in the signal-front and noise-front evaluation, the conditions revealed no significant differences.

Vielerorts finden in den Schulferien Theatercamps statt, die vor allem sprachschwachen Kindern eine Chance bieten, ihre Sprachkenntnisse spielerisch und handlungsgebunden zu erweitern. Anschlussprojekte, die die Nachhaltigkeit der Fördereffekte sichern und auf diesen aufbauen, gibt es hingegen kaum. Es fehlt an bereichsübergreifenden Konzepten, die im Camp gewonnenen positiven Einstellungen gegenüber Sprache und Schrift in den Schulalltag zu transferieren. Diesem Desiderat begegnend wird in Tübingen derzeit ein durchgängiges Förderkonzept für Grundschulkinder mit Deutsch als Zweitsprache (DaZ) und Sprachförderbedarf erprobt: Im Rahmen von Theater-AGs werden die Kinder, die zur Zeit des Feriencamps die Klasse 2 besuchten, zwei weitere Jahre gefördert, die Kinder der Klasse 3 mindestens ein Jahr. In einem solchen Förderzeitraum ist es möglich, systematisch die Sprachkompetenz aufzubauen und die GrundschülerInnen auf die bildungssprachlichen Anforderungen der Sekundarstufe I vorzubereiten. Während sich die thematisch-inhaltlichen Vorgaben für die Sprachförderung im Camp aus dem gemeinsam zu entwickelnden und aufzuführenden Theaterstück ergeben, werden in den Theater-AGs Inhalte aus dem Schulfach MeNuK (Mensch, Natur, Kultur) dramapädagogisch erarbeitet. Im folgenden Beitrag wird die Entwicklung vom außerschulischen Ferien-Theatercamp hin zum Anschlussprojekt der fachsensiblen Sprachförderung in schulischen Theater-AGs noch einmal im Detail nachvollzogen, wobei exemplarische Fördereinheiten die bereichsübergreifende methodische Kontinuität illustrieren.

Die vorliegende Längsschnittstudie beschäftigt sich mit der Frage, ob sich Ergebnisse hinsichtlich vorschulischer Prädiktoren der Lesekompetenz einsprachig deutscher Kinder auf Kinder mit Deutsch als Zweitsprache (DaZ) übertragen lassen. Ein Pfadmodell zur Vorhersage der Lesekompetenz zum Ende der ersten bzw. zweiten Klassenstufe wurde dabei sowohl auf einsprachige (n = 111) als auch auf DaZ-Kinder (n = 282) angewendet. Zunächst wurde die Pfadstruktur der am Ende der Kindergartenzeit bzw. zu Beginn der Schulzeit erfassten Vorläuferfertigkeiten nonverbale Intelligenz, phonologisches Arbeitsgedächtnis, sprachliche Kompetenz, Schnelles Benennen, Buchstabenkenntnis und phonologische Bewusstheit untereinander betrachtet. Hier unterschieden sich die Effektmuster bei einsprachigen und DaZ-Kindern nicht signifikant. Ebenso zeigt sich in beiden Sprachgruppen ein vergleichbares Bild hinsichtlich der Einflussfaktoren des Leseverständnisses. Bedeutsame Unterschiede zwischen den Sprachgruppen ergaben sich jedoch bezüglich der Vorhersage der Lesegeschwindigkeit: In der Gruppe der einsprachigen Kinder war die phonologische Bewusstheit der bedeutsamste Prädiktor. Bei den DaZ-Kindern hingegen war der Einfluss der phonologischen Bewusstheit auf die Lesegeschwindigkeit nicht von Bedeutung, dafür erfolgte der größte Teil der Varianzaufklärung durch die nonverbale Intelligenz.

Abstract:

A protein in RAW 264.7 macrophages, which became phosphorylated in response to LPS (lipopolysaccharide), was identified as the RNA-binding protein called DAZAP1 [DAZ (deleted in azoospermia)-associated protein 1]. The phosphorylation of this protein was prevented by specific inhibition of MKK1 [MAPK (mitogen-activated protein kinase) kinase 1], indicating that it was phosphorylated via the classical MAPK cascade. Further experiments showed that DAZAP1 was phosphorylated stoichiometrically in vitro by ERK2 (extracellular-signal-regulated protein kinase 2) at two Thr-Pro sequences (Thr269 and Thr315), and that both sites became phosphorylated in HEK-293 (human embryonic kidney 293) cells in response to PMA or EGF (epidermal growth factor), or RAW 264.7 macrophages in response to LPS. Phosphorylation induced by each stimulus was prevented by two structurally distinct inhibitors of MKK1 (PD184352 and U0126), demonstrating that DAZAP1 is a physiological substrate for ERK1/ERK2. The mutation of Thr269 and Thr315 to aspartate or the phosphorylation of these residues caused DAZAP1 to dissociate from its binding partner DAZ. DAZ interacts with PABP [poly(A)-binding protein] and thereby stimulates the translation of mRNAs containing short poly(A) tails [Collier, Gorgoni, Loveridge, Cooke and Gray (2005) EMBO J. 24, 2656–2666]. In the present study we have shown that DAZ cannot bind simultaneously to DAZAP1 and PABP, and suggest that the phosphorylation-induced dissociation of DAZ and DAZAP1 may allow the former to stimulate translation by interacting with PABP.
