Abstract:
ABSTRACTThe main functions of the dynamic cantilever calibrator (DCC), which are related to characterization of AFM probes and instruments, are demonstrated on a variety of probes. The resonant frequency, Q-factor and spring constant of the rectangular and V-shaped probes were evaluated by thermal tune method. The inverse optical sensitivity and optical beam deflection noise, which define performance of AFM microscopes, were extracted from DCC data. Peculiarities of thermal tune studies and the use of DCC for advanced applications are discussed.

Abstract:
This study employs a sample survey to determine and analyse the corporate finance practices of South African listed companies in relation to cost of capital, capital structure and capital budgeting decisions.The results of the survey are mostly in line with financial theory and are generally consistent with a number of other studies. This study finds that companies always or almost always employ DCF methods such as NPV and IRR to evaluate projects. Companies almost always use CAPM to determine the cost of equity and most companies employ either a strict or flexible target debt‐equity ratio. Furthermore, most practices of the South African corporate sector are in line with practices employed by US companies. This reflects the relatively highly developed state of the South African economy which belies its status as an emerging market. However, the survey has also brought to the fore a number of puzzling results which may indicate some gaps in the application of finance theory. There is limited use of relatively new developments such as real options, APV, EVA and Monte Carlo simulation. Furthermore, the low target debt‐equity ratios reflected the exceptionally low use of debt by South African companies.

P. 34-39.The physico-chemical properties of suspensions of graphite-nitric acid were studied. The ratios of the components in suspension providing the possibility of their anodic oxidation with the formation of compounds of the implementation of graphite were determined. The values of electrical conductivity of suspensions of graphite-nitric acid with different ratio of the solid and liquid phases were determined .The maximum value of the electrical conductivity of suspensions is achieved at the mass ratio of graphite to HNO3» 1:0.66. The design of the electrochemical cell for carrying out chronovoltamperometric measurements on a suspension graphite electrode was developed. The nature and kinetics of electrode processes on dispersed graphite electrodes in 60% nitric acid were studied. It is shown that under anodic polarization of dispersion graphite electrode the complex of anodic processes occurs: the oxidation of surface of functional groups, the formation of compounds of implementation and evolution of oxygen. Maximum electrochemical reversibility of the anodic processes on the suspensions of graphite-HNO3 observed at Erev< 1.2 V, the rate of reactions of formation of compounds of the introduction of graphite and reactions involving surface functional groups on the electrode slurry increases in the range of potentials of 1.2-1.7 V. At that, the current is increase mainly causes by electrochemically reversible process of intercalation, since at the reverse cycle in the cathode area also recorded the growth of the current due to the reaction of deintercalation. An electrochemical oxidation of dispersed graphite electrode in HNO3 should be in the range of potentials from 1.2 V to 2.1 V, which will provide formation of compounds of introduction of graphite with the maximum rate and minimum power input.

Abstract:
511 Background: Penile cancer is rare and there are scant data on the optimal chemotherapy regimen. The majority of trials are single arm, non-randomized studies. Here we report on a systematic review aiming to compare taxane combination chemotherapy regimens with non-taxane regimens for APC. Methods: A systematic review was conducted in accordance with the PRISMA guidelines. Medline, Embase and Cochrane Central Register of Controlled Trials databases were searched using the following terms: penile cancer, penis, antineoplastic combined chemotherapy, taxane, docetaxel, paclitaxel, platinum, cisplatin, carboplatin. Studies were identified using preplanned eligibility criteria by 2 investigators (EA-E and JR). Data were extracted independently by EA-E and JR. Studies were weighted by study sample size and those comparing taxane-based chemotherapy were compared to non-taxane therapy using the Mann Whitney test. Results: The search identified 1929 publications and 40 were selected for further assessment. Of these, 8 met eligibility criteria (7 prospective and 1 retrospective). Three studies tested taxane combinations (docetaxel, cisplatin and 5FU [DCF] and paclitaxel, ifosfamide and cisplatin [TIP]). A total of 148 men with APC were treated with non-taxane regimens and 98 men received a taxane combination. Patient characteristics (age, ECOG status, stage, number of cycles) were comparable between the two groups. Partial response and overall response rates were significantly higher in the taxane versus the non-taxane group (35.7% vs. 24.2% p = 0.01 and 41.9% vs. 32.5% p = 0.007) respectively. Grade3/4 neutropenia was significantly higher in taxane group than non-taxane group (27.8% vs 19.4% p = 0.02). Median PFS and OS was numerically but not significantly higher in the taxane versus the non-taxane group (5.7 vs. 4.4 months, p = 0.45 and 12.1 vs 10 months, p = 0.48, respectively). Conclusions: Compared to non-taxane-based regimens, taxane combinations have higher response rates and may improve survival in APC. Hematologic toxicities are worse with taxane containing regimens. Taxane combinations should be the preferred regimen for suitable men with APC.

Abstract:
Background: CD3/CD19 cell depletion (Barfiled RC, et al, Cytotherapy, 2004), αβ T-cell/CD19 cell depletion (Locatelli F, et al, Blood, 2017), CD34+ positive selection (Aversa F, et al, NEJM, 1998) are designed to deplete T cells and reduce AGVHD following allogeneic stem cell transplantation (AlloSCT). These approaches achieved low rates of AGVHD, but the grafts had few T and B cells. To improve immune reconstitution we undertook an alternative approach to addback small numbers and percentages of immune cells in the final HSCT product. We previously reported a very low incidence of AGVHD in pediatric recipients receiving CD34 enriched HPC products with peripheral blood mononuclear cells (PBMNC) addback containing a fixed dose of 2 x 105 CD3/kg from MUD donors (Geyer/Cairo et al, BJH, 2012). Recently we demonstrated that despite a 5 log depletion of T cells, PBMNC addback (fixed at 2 x 105 CD3/kg) facilitated rapid hematopoietic engraftment, high levels of donor chimerism and immune reconstitution with a low probability of Grade II-IV AGVHD. Patients had a 1 yr OS of 90% following familial haploidentical (FHI) CD34 Enriched Stem Cell Transplantation in patients with SCD (Cairo, JAMA Pediatr, 2020). Objective: To determine the final immune cell concentration following CD34 enrichment and PBMNC (2 x 105 CD3/kg) addback and determine the effect on engraftment and T and NK cell immune reconstitution. Methods: Patients and/or their guardians signed written informed consents and/or assents (NCT NCT02675959). CD34+ enrichment was performed using a CD34+ reagent system (CliniMACS; Miltenyi Biotec). Mononuclear cells (2 × 105 CD3 cells/kg of recipient body weight) were removed from the leukapheresis collection prior to CD34+ enrichment and were cryopreserved as a source of MNC addback (T cells). The addback products were analyzed for CD3+CD56- T cells, CD3-CD56+ NK cells, CD3+CD56+ NKT cells, Lin-CD123+ HLA-DR+ DC cells and Lin-CD11c+ HLA-DR+ DC cells by multicolor flow cytometry analysis. Th1/Th2 cytokines were measured by multiplex assays. T cell activity was measured by viral T cells IFN-g and plasma cytokines. NK function was measured by NK receptor expression by flow cytometry analysis and in vitro cytotoxicity. Results: We identified in the PBMNC addback, mean+SEM white blood cell (WBC) percentage of: CD3+ CD56- T cells = 56.4±5%; CD3- CD56+ NK cells = 4.6±1%; CD3+ CD56+ NKT cells = 5.1±0.6%; CD19+ B cells = 29.9±3.5%. Lin- WBC consisted of: CD123+ HLA-DR+ DC cells = 18.4±8.2%; CD11c+ HLA-DR+ DC cells = 6.0±3.0%. There were 20.0+9.1e6 T cells, 1.1+0.3e6 NK cells, 1.6+0.7 e6 NKT cells, 8.6+2.5e6 B cells, 1.2+0.6e6 CD123+DC and 0.8+0.5e6 CD11c DC in the final infused products (Fig.1). We found that percentages of IFN-g+ in CD4 cells in response to CMV (pp65), ADV (hexon) and EBV (BZLF1), ranged from 0.2%+0.1% to 0.5%+0.1%, while percentages of IFN-g+ in CD8 cells in response to the antigens ranged from 0.7%+0.3% to 3.7%+1.8% when examined at days 180, 270 and 365. NK (CD3- CD56+) reconstitution was extremely rapid and occurred as early as day 30 (35.5±8.6%, 2710+1624.4 cells/ul total cells; p&lt;0.01 vs pre-t). There were no significant differences pre-HSCT vs day 365 in plasma cytokines (Th1 and Th2) and growth factors released including IFN-g, TNF-a, IL-18, IL-4, IL-5, IL-6, IL-10, G-CSF, MCP-1 and MIP1a. There was also robust expression of NK receptor expression including NK cytotoxicity receptors, NK KIR receptors, and C-type lectin-like receptors at day 30 as compared to pre-HSCT. NK cytotoxicity, as measured using PBMC cells from recipients at different time points against K562 (E:T=10:1), was also significantly increased at day 30 (26.2±2.8%) and day 180 (28.3±3%) vs pre-HSCT (16.1±2.1%) (p&lt;0.01). As a NK cell activation marker, CD107a expression and granzyme B levels in gated NK cells peaked at day 30. Conclusion: PBMNC addback to CD34 enriched HPC products, with a final dose of 2 × 105 CD3 cells/kg, led to stem cell products with a diverse mixture of T, NK, NKT, DC1, and DC2 cells. Immune reconstitution following PBMNC addback to CD34 enriched cells resulted in excellent CD4 and CD8 responses to CMV, ADV and EBV, and rapid functional NK cell reconstitution (Supported by FDA R01FD004090 (MSC)). Zur Lösung des komplexen Problems der Bewertung ganzer Unternehmen werden im wesentlichen drei Methoden eingesetzt: die Adjusted- Present-Vcdue-Methode (APV-Methode), die Equity-Methode und die Entity-Methode. Diese Methoden führen außer in seltenen Spezialfällen jedoch nicht zum gleichen Wert des Eigenkapitals. Die Adjusted-Present-Value-Methode kann aufgrund ihrer theoretischen Fundierung als Referenzmodell angesehen werden, das unter realistischen Rahmenbedingungen „richtige“ Bewertungsergebnisse produziert. Es werden die Abweichungen der Bewertungsergebnisse der übrigen Methoden gegenüber denen der APV-Methode quantifiziert und Anwendungsempfehlungen für die praktische Unternehmensbewertung abgeleitet.

Abstract:
Introduction: oligodendrocyte injury after ischemic stroke influences the integrity of white matter, which ultimately leads to neurological deficits. Netrin-1 (NT-1) plays a crucial role in axon guidance during neural development. Also, it promotes oligodendrocyte progenitor cell proliferation. Our previous study demonstrates that netrin-1 overexpression improves neurobehavioral outcomes after ischemia by promoting focal angiogenesis. In this study, we investigate whether netrin-1 facilitates white matter recovery during focal ischemia and further to explore which specific receptor involves in the white matter reconstruction. Methods: sixty adult male ICR mice underwent adeno-associated virus (AAV) mediated AAV-Netrin-1 or AAV-GFPgene transfer. These mice received one hour transient middle artery occlusion (MCAO) and 7, 14, 28 days reperfusion at one week after the gene transfer. Western blot and immunohischemistry were used to determine the location and quantification of exogenous NT-1 expression. Neuronalbehavior test were performed to eveluate the neuralbehavioral outcomes. Oligodendrocyte progenietor cell proliferation, maturation and myelination were examined by immunohischemistry. Results: NT-1 was highly expressed in the mouse brain after two weeks of AAV-NT-1 gene transfer, which mainly expressed in neurons and astrocytes. Neurobehavioral outcomes were greatly improved at 7, 14 and 28 days after reperfusion in AAV-NT-1 treated mice compared to the GFP control mice ( p <0.05), The number of proliferated oligodendrocyte progenietor cells, mature oligodendrocytes and MBP positive neurofilaments in the corpus callosum and the striatum in the ipsilateral hemisphere at 7, 14 and 28 days after reperfusion were increased in the NT-1 treated group compared to GFP control mice ( p <0.01). NT-1 receptor DCC and Unc5H2 are involved in the proliferation of oligodendrocyte progenitor cells,But only Unc5h2 was involved in the re-myelination process. Conclusion: NT-1 overexpression improves neurobehavioral outcomes and promotes oligodendrocyte progenietor cell proliferation and maturation. Oue results suggest that netrin-1 not only promotes angiogenesis but also enchances remyelination.

The master thesis aims to compare the DCF Entity method and the DCF APV method. In the practical part of the thesis, both of these methods are applied to evaluate the company KOVOVÝROBA HOFFMANN, s.r.o. When applied correctly and using the same input data, DCF Entity as well as DCF APV results are supposed to equal. However, both methods dispose of its pros and cons. Despite its imperfections, the DCF Entity method is commonly used in practise, whereas the DCF APV method lacks of interest. This thesis, however, points out that the DCF APV could be not merely an equal alternative to the DCF Entity method, but could also avoid some of its imperfections or give extra information compared to the DCF Entity method. As a consequence, we can expect increasing importance of the DCF APV method.

The diploma thesis focuses on corporate valuation of Paramo, a. s., estimating value-in-use of equity by 1st January 2009. The thesis is divided into 8 parts. In the first two chapters, the prerequisites for valuation and the introduction of the company are presented. The next part comprises the financial analysis. Afterwards, the strategic analysis follows, which results in the prediction of the total revenues of the examined company. The prediction of value generators is based on the previous chapters and consequently is transformed in the financial plan. In the concluding part the company is evaluated by using a revenue method called DCF APV. The method of market comparison, in the form of direct valuation method and capital market data, was selected in order to confirm the final valuation outcome. After these calculations was applied liquidation value to make better decision about the final value.

The thesis consists of a business valuation of BERNARD Family Brewery, a brewery headquartered in the town of Humpolec and belonging to a group of several smaller breweries (related to its size). In 2001, top management decided to finance its company growth through venture capital. The main objective of this thesis is to indicate the market value as of 1/1/2009 by utilizing appraisement measures in accordance with International Valuation Standards (IVS). The business valuation is primarily determined using two well-known valuation approaches - adjusted present value method (based on income analysis) and comparable companies analysis (based on market analysis). In addition, the use of advanced statistics is applied when needed. The purpose of this assessment is to find an appropriate value for owners and other third-party stakeholders. In order to provide a full range of understanding, the paper consists of six main parts that involve a company profile, strategic analysis, financial analysis, value drivers, financial plan and a final valuation. Further, all these parts are split up into few sections.

The aim of this diploma thesis is to estimate the market value of the company Racio Ltd. as of the date 07/01/2014. Market value represents the amount of money, which is an average market customer willing to pay. The thesis is divided into two parts. The first, theoretical part deals with the definition of related basic terms, describes the process of business valuation including used formulas and methods. The second part is practical; business valuation theoretical framework is applied to specific company -- Racio, s.r.o. Starting point of business valuation process is strategic analysis; the outcome is prediction of company sales forecast. The aim of my thesis is to estimate market value of the company Cestovni kancelar Fischer, a.s. as of 31st December, 2012. The thesis is divided into five main chapters. It starts with an overview which is followed by the strategic analysis. At the end of this chapter, the sales are predited for the second phase of the valuation. In the next part, the financial analysis have been performed to confirm the financial health of the company. In the third part generators of value have been analysed and predicted. On the basis of the generators of value the complete financial plan has been set up. In the last chapter, the company value was appraised using DCF APV and market comparison methods.

Benzyl esters are easily deprotected by hydrogenolysis. It is often observed, however, as exemplified by the conversion of 1 to 2 reported (Adv. Synth. Catal. 2008, 350, 406) by Hironao Sajiki of Gifu Pharmacutical University, that alkene hydrogenation can be carried out selectively. Fernando Albericio of the University of Barcelona has developed (Tetrahedron Lett. 2008, 49, 3304) a family of thiophene-based esters 3 that can be removed with acid in the presence of t-butyl esters, and that are stable to the removal of FMOC groups. Vassiliki Theodorou of the University of Ioannina has found (Tetrahedron Lett. 2008, 49, 8230) that esters were rapidly saponified by methanolic NaOH in solvent CH2Cl2. Specific oligosaccharide synthesis depends heavily on the use of orthogonal methods for alcohol protection and deprotection. This is illustrated by the work (J. Org. Chem. 2008, 73, 1008) of Carolyn R. Abstract The dynamic behavior of large complex structures is often achieved by substructuring methods where the substructures are modeled separately. One of advantages of these methods is that one component may be changed without remodeling the others. This is particularly useful when components are designed and analyzed by different organizations. Althougth these methods use a variety of reduction techniques to couple the substructure models, the size of the final condensed system remains important due to large number of the junction degrees of freedom (Abv: dof) required for assembly. In this paper, we propose a method using a modal projection on the Ritz basis which, when correctly chosen, reduces the number of these dof before the assembly of substructures. This method is applied to certain substructuring methods: dynamic condensation, modal synthesis using free or constrained interfaces.

Abstract:
Using autonomous underwater vehicles (AUVs) for mapping the underwater topography of sea-ice and icebergs, or detecting keels of ice ridges, is foreseen as an enabling technology in future Arctic offshore operations. This paper presents a method for online iceberg drift estimation using a Simultaneous Localization and Mapping (SLAM) approach using an AUV with a multi-beam echosounder (MBE) during such survey/monitoring operations. Iceberg drift is affected by wind, current, and Coriolis forces. This can be hard to predict, making automated mapping of icebergs difficult. The method proposed in this paper estimates the iceberg’s pose using a particle filter, where each particle uses extended information filters to estimate the topography of the iceberg. A grid map is used to store the iceberg topography, and distributed particle mapping is used to avoid expensive copy operations during particle resampling. The proposed method is verified through a simulation study, using a 6 DOF AUV model, an MBE sensor model, and an iceberg topography taken from the PERD iceberg sightings database. The method is able to provide a georeferenced iceberg position, thus, estimating the iceberg’s drift trajectory. A topography estimate of the iceberg, corrected for iceberg drift, is also generated. Furthermore, the algorithm estimates the iceberg drift velocity, as well as the relative iceberg-AUV pose, for use in future iceberg mapping guidance algorithms. The simulation study illustrates the performance of the method, and a short execution time analysis is presented to illustrate the method’s real-time potential.

Heerema Marine Contractors (HMC) is entering a new era of pipe laying using the reel-lay method for deep and shallow water pipeline installation projects. The new Deep water Construction Vessel (DCV) Aegir is designed to be able to reel/J-lay pipelines for a range of pipe dimension and water depth combinations which on the one side is governed by the equipment limitations (reel drum size, top tension capacity) and on the other side is limited by the code (DNV, API) acceptance criteria for reel/J-lay installation. The paper describes how the code standard acceptance criteria, which have been used to estimate the reel-ability of DCV Aegir and are generally known as conservative approach, can be relaxed in a reliable manner in order to safely extend the DCV Aegir reeling capability. This approach will permit a reduction of minimum reelable pipe wall thickness and result in a wider envelope of water depth applications for reeling from the DCV Aegir. The methodology of relaxation for local buckling and residual ovality criteria has been developed with and approved by DNV specifically for the spooling-on with the DCV Aegir configuration. A series of bend rig tests have been performed at Heriot-Watt University to confirm and validate the new proposed minimum reelable pipe wall thickness. In line with the development of the extended reeling capability, the required tighter specifications for seamless pipe purchased for reeling has been discussed.
