Abstract:
We have initiated a phylogenetic study using restriction fragment length polymorphisms to examine nuclear DNA variation in a number of Rosa species. Random genomic clones were isolated from the cultivar `Confection'. To generate these clones, genomic DNA was digested with the restriction enzymes Hind III and Eco RI and the resulting fragments cloned into a pUC8 plasmid and transformed into the E. coli bacterial strain JM83. Individual clones from the DNA library were screened for polymorphism by Southern hybridization methods. Those clones displaying polymorphisms were used in combination with one, two, or three restriction enzymes to identify different size restriction fragments. Each fragment was treated as a unit character and was used to generate a phylogenetic tree using the computer program “Phylogenetic Analysis Using Parsimony” (PAUP version 3.0). Results of the studies on the amount of genetic diversity and phylogenetic affinities of Rosa species among the different sections of the subgenus Rosa will be presented.

Abstract:
Advances in cancer treatment have led to significant improvements in long-term survival in many types of cancer, but heart dysfunction and heart failure, associated with cancer treatment, have also increased. Anthracyclines are the main cause of this type of cardiotoxicity. In this study, we describe a combined experimental and cell morphology analysis approach for the high-throughput measurement and analysis of a cardiomyocyte cell profile, using partial least square linear discriminant analysis (PLS-LDA) as the pattern recognition algorithm. When screening a small-scale natural compound library, rosmarinic acid (RosA), as a candidate drug, showed the same cardioprotective effect as the positive control. We investigated the protective mechanism of RosA on a human cardiomyocyte cell line (AC16) and human induced pluripotent stem-cell-derived cardiomyocytes (hiPSC-CMs). We showed that RosA pretreatment suppressed doxorubicin (Dox)-induced cell apoptosis and decreased the activity of caspase-9. RosA promotes the expression of Heme oxygenase-1 (HO-1) and reduces the production of reactive oxygen species (Ros), which is induced by Dox. Meanwhile, it can also promote the expression of cardiac-development-related protein, including histone deacetylase 1 (HDAC1), GATA binding protein 4 (GATA4) and troponin I3, cardiac type (CTnI). Collectively, our data support the notion that RosA is a protective agent in hiPSC-CMs and has the potential for therapeutic use in the treatment of cancer therapy-related cardiac dysfunction and heart failure.

We know there are authors that Pessoa read and that influenced his work, but not all the books he read were in his private library. Abstract:
The collections of Meise Botanic Garden comprise extensive and varied documentation on the genus Rosa. The library collection contains valuable and often rare 19th century publications about the genus Rosa, frequently with annotations by François Crépin. In addition to this, the archives hold the unpublished manuscripts and thousands of letters from Crépin’s correspondents reflecting the extraordinary scientific network he built up to facilitate the exchange of herbarium collections together with erudite discussions on the taxonomy of the genus Rosa. Crépin exchanged letters with all the important rhodologists of his time, such as E. Burnat, H. Christ, P.A. Déséglise, P.E. Parmentier, H. Takashima, and W. Wirtgen, and also with horticulturists, such as J. Gravereaux. This unique network and diversified patrimony of historical collections took shape over a period of several decades during which Crépin prepared a monograph of the genus Rosa that unfortunately was never finished. Crépin assembled a 19th century herbarium with ca. 40 000 specimens of the genus Rosa. In contrast to other important herbaria, the Crépin collection covers the whole natural area of distribution of Rosa and documents an unparalleled range of Rose taxa. The combination of documentary sources of several different types makes the collections of Meise Botanic Garden extremely valuable. They offer great opportunities for multidisciplinary studies by taxonomists and historians with the aim of solving the still insufficiently understood relations between taxa in the genus Rosa. Linking this unique heritage of Meise Botanic Garden with present-day molecular techniques promises rewarding research in the near future. We can conclude that the collections related to Roses are major masterpieces of the Garden. In 2016 the Piaget Foundation, the World Federation of Rose Societies and ‘De Vrienden van de Roos’, supported the Botanic Garden with a grant which allowed us to image 13 000 out of the 40 000 herbarium sheets. The public is invited to help us generate the label data on the Garden’s crowdsourcing platform (https://www.doedat.be/). In this project we tested also the quality of extracted DNA of 96 specimens of the Crépin Herbarium. 69% of the samples yielded with the Nanodrop technology good quality and quantity of DNA and for 29% of the samples it was even possible to amplify the rDNA gene marker ITS (with 650bp). These results are encouraging and illustrate that this historic herbarium could be a useful tool for research on roses.

Abstract: Three days after assuming a position at the Brazilian Academy of Letters, João Guimarães Rosa, the latest immortal of the House founded by Machado de Assis, passed away, filling with dismay not only his peers, but the whole country. Abstract:
Microsatellite or simple sequence repeat (SSR) markers were developed from Rosa wichurana Crépin to combine two previously constructed tetraploid rose (Rosa hybrida L.) genetic maps. To isolate SSR-containing sequences from rose a small-insert genomic library was constructed from diploid Rosa wichurana and screened with several SSR probes. Specific primers were designed for 43 unique SSR regions, of which 30 primer pairs gave rise to clear PCR products. Seventeen SSR primer pairs (57%) produced polymorphism in the tetraploid rose 90-69 mapping family. These markers were incorporated into existing maps of the parents 86-7 and 82-1134, which were constructed primarily with AFLP markers. The current map of the male parent, amphidiploid 86-7, consists of 286 markers assigned to 14 linkage groups and covering 770 cm. The map of the female tetraploid parent, 82-1134, consists of 256 markers assigned to 20 linkage groups and covering 920 cm. Nineteen rose SSR loci were mapped on the 86-7 map and 11 on the 82-1134 map. Several homeologous linkage groups within maps were identified based on SSR markers. In addition, some of the SSR markers provided anchoring points between the two parental maps. SSR markers were also useful for joining small linkage groups. Based on shared SSR markers, consensus orders for four rose linkage groups between parental maps were generated. Microsatellite markers developed in this study will provide valuable tools for many aspects of rose research including future consolidation of diploid and tetraploid rose genetic linkage maps, genetic, phylogenetic and population analyses, cultivar identification, and marker-assisted selection.

Abstract:
Although simple sequence repeat (SSR) markers have been developed for species in the closely related genera Fragaria L. (strawberry) and Rubus L. (raspberry and blackberry), the number of SSRs available is insufficient for genetic mapping. Our objective was to use and compare multiple approaches for developing additional SSRs for Fragaria and Rubus. The approaches included: the development of SSRs from GenBank sequences from species of varied relatedness to Fragaria and Rubus and identified with two different data-mining methods (BLAST and SSRIT); the evaluation of some previously published SSRs designed from related species; and the development of SSRs from a genomic library made from F. ×ananassa Duschene ex Rozier `Earliglow'. When an SSR was developed from a known gene sequence, the location of the repeat in the gene was determined to evaluate the effect on amplification and polymorphism detection. Cross-generic amplification between closely related Fragaria and Rubus as well as transference from species of varied relatedness to Fragaria and Rubus also was evaluated and indicated limited transference within the subfamily Rosoideae. However, development of SSRs for Fragaria and Rubus from Rosa L. (rose) and Rosaceae genera outside Rosoideae was not efficient enough to be practical for new map development. SSRIT was superior to BLAST for identifying GenBank sequences containing repeats. SSRs developed from repeats found in either the 5′UTR (80% polymorphic) or 3′UTR (85% polymorphic) were most likely to detect polymorphisms, compared with those developed from coding regions (30%). SSRs developed from the genomic library were only slightly superior to GenBank-derived SSRs in their ability to detect polymorphisms.

Abstract:
PurposeThis paper examines the socio-political affordances of metrics in research evaluation and the consequences of epistemic injustice in research practices and recorded knowledge.Design/methodology/approachFirst, the use of metrics is examined as a mechanism that promotes competition and social acceleration. Second, it is argued that the use of metrics in a competitive research culture reproduces systemic inequalities and leads to epistemic injustice. The conceptual analysis draws on works of Hartmut Rosa and Miranda Fricker, amongst others.FindingsThe use of metrics is largely driven by competition such as university rankings and league tables. Not only that metrics are not designed to enrich academic and research culture, they also suppress the visibility and credibility of works by minorities. As such, metrics perpetuate epistemic injustice in knowledge practices; at the same time, the reliability of metrics for bibliometric and scientometric studies is put into question.Social implicationsAs metrics leverage who can speak and who will be heard, epistemic injustice is reflected in recorded knowledge and what we consider to be information.Originality/valueThis paper contributes to the discussion of metrics beyond bibliometric studies and research evaluation. It argues that metrics-induced competition is antithetical to equality and diversity in research practices.

Abstract:
We had a surprise in January 2012 when a practising QC was parachuted straight into the Supreme Court (as the Judicial Committee of the House of Lords became in 2009). But if there were expressions of discontent they were definitely sub rosa since the man in question, Jonathan Sumption QC, was then, and has since proved himself to be, pre-eminently qualified for the position. He is one of our foremost medieval historians, a Fellow of Magdalen College who taught History, before leaving to pursue a career at the Bar, where he rose to dizzy heights. His as yet uncompleted history of the Hundred Years’ War has received the highest praise. So it perhaps was not surprising that in this octocentenial year he should have been asked by the Friends of the British Library (an audience I warrant as challenging as this) to speak to them about Magna Carta on 9 March 2015. If I may say so, his address ‘Magna Carta then and now’ is a masterpiece. It completes the destruction of the hermeneutical myth originated by Sir Edward Coke which had been commenced by William McKechnie in his landmark essay published in 1905.

Abstract:
Purpose The aim of this paper is to describe the Rosarium Project which is currently curating nonfiction materials about the genus Rosa written at the turn of the twentieth century and published in popular American periodicals. This is achieved by encoding the texts following the guidelines set forth by the Text Encoding Initiative Consortium (TEI). Design/methodology/approach This paper explains what text encoding is and why following the guidelines set forth by the TEI Consortium was the best choice for the Rosarium Project. It then goes on to outline the workflow and choices made by the principal researcher which are needed to move the project steadily forward. Findings The principal researcher on the Rosarium Project has found that encoding with the TEI was easy to learn and fun to do, as well as intellectually stimulating. Librarians should find text-encoding projects of their own specialist subjects equally doable. Originality/value The Rosarium Project is unique, in that it is curating early twentieth century articles on the subject of roses that appeared in popular magazines. These materials are hidden away in online repositories and libraries worldwide. This project is of value, in that it provides primary sources to researchers in areas of popular culture, horticulture and garden history and also acts as an example of what librarians can contribute to the Digital Humanities.

Summary The aim of the article was to obtain maximum information about plant mucilage, its sources and applications in the pharmaceutical industry. This study focuses on the scientific articles and books available in Internet resources and college library that deal with the sources, applications, extraction and isolation of plant mucilage. Mucilage is obtained mainly from plant sources and can be isolated easily. Due to the low cost, easy availability, non-toxicity, non-irritancy, and biocompatibility, mucilage is of great demand in the field of pharmaceuticals. Hibiscus rosa-sinensis L., Trigonella foenum-graecum L., Abelmoschus esculentus L, Plantago ovata Forssk. and Aloe barbadensis L. are some common sources of mucilage. The isolation methods vary depending on the part of the plant where mucilage is present. It is commonly used as gelling agent, suspending agent, binder, and disintegrant. Since it is hydrophilic in nature, chances of deterioration are higher. In this review, different mucilage sources and their isolation methods are discussed in detail. Mucilage is used as excipient in many formulations of tablets, suspensions, gels, etc. The study explores the potential of plant mucilage as an excipient in pharmaceutical formulations. The biodegradable and biocompatible properties of this inexpensive excipient make it more favourable for the newer formulation development.

Abstract:
Recognizing and authenticating wheat varieties is critical for quality evaluation in the grain supply chain, particularly for methods for seed inspection. Recognition and verification of grains are carried out manually through direct visual examination. Automatic categorization techniques based on machine learning and computer vision offered fast and high-throughput solutions. Even yet, categorization remains a complicated process at the varietal level. The paper utilized machine learning approaches for classifying wheat seeds. The seed classification is performed based on 7 physical features: area of wheat, perimeter of wheat, compactness, length of the kernel, width of the kernel, asymmetry coefficient, and kernel groove length. The dataset is collected from the UCI library and has 210 occurrences of wheat kernels. The dataset contains kernels from three wheat varieties Kama, Rosa, and Canadian, with 70 components chosen at random for the experiment. In the first phase, K-nearest neighbor, classification and regression tree, and Gaussian Naïve Bayes algorithms are implemented for classification. The results of these algorithms are compared with the ensemble approach of machine learning. The results reveal that accuracies calculated for KNN, decision, and Naïve Bayes classifiers are 92%, 94%, and 92%, respectively. The highest accuracy of 95% is achieved through the ensemble classifier in which decision is made based on hard voting.

Abstract:
Abstract mRNA degradation is an important cellular mechanism involved in the control of gene expression. Several genome-wide profiling methods have been developed for detecting mRNA degradation in plants and animals. However, because many of these techniques use poly (A) mRNA for library preparation, degradation intermediates are often only detected near the 3′-ends of transcripts. Previously, we developed the Truncated RNA End Sequencing (TREseq) method using Arabidopsis thaliana, and demonstrated that this method ameliorates 3′-end bias. In analyses using TREseq, we observed G-rich sequences near the 5′-ends of degradation intermediates. However, this finding remained to be confirmed in other plant species. Hence, in this study, we conducted TREseq analyses in Lactuca sativa (lettuce), Oryza sativa (rice) and Rosa hybrida (rose). These species including A. thaliana were selected to encompass a diverse range in the angiosperm phylogeny. The results revealed similar sequence features near the 5′-ends of degradation intermediates, and involvement of translation process in all four species. In addition, homologous genes have similar efficiencies of mRNA degradation in different plants, suggesting that similar mechanisms of mRNA degradation are conserved across plant species. These strong sequence features were not observed in previous degradome analyses among different species in plants.

Abstract:
A number of viruses in the genera Ilarvirus and Nepovirus have been shown to be associated with specific diseases in rose, but many graft-transmissible rose diseases still have unknown etiologies. One of these diseases originally was detected by grafting from nonsymptomatic roses to Rosa multiflora indicator plants. Double-stranded RNAs (dsRNAs) were recovered and used as templates for cDNA synthesis and generating a cDNA library. Analysis of deduced amino acid sequences clearly positioned this virus as a member of the family Luteoviridae. The name rose spring dwarf associated virus (RSDaV) is tentatively proposed for the novel virus because the symptoms of this virus on R. multiflora are consistent with previous descriptions of rose spring dwarf disease (RSD). Phylogenetic analysis revealed a close relationship of RSDaV with members of the genus Luteovirus. Aphid transmission studies identified the rose-grass aphid (Metapolophium dirhodum) and yellow rose aphid (Rhodobium porosum) as vectors for this new virus. Host range data showed that RSDaV has a host range including both monocots and dicots. A specific reverse-transcription polymerase chain reaction assay was developed and revealed the presence of the RSDaV in several rose cultivars. RSDaV-inoculated rose plants developed RSD symptoms, confirming its role in the etiology of the disease.

Abstract:
When assessing the quality of the grain supply chain's quality, it is essential to identify and authenticate wheat types, as this is where the process begins with the examination of seeds. Manual inspection by eye is used for both grain identification and confirmation. High-speed, low-effort options became available thanks to automatic classification methods based on machine learning and computer vision. To this day, classifying at the varietal level is still challenging. Classification of wheat seeds was performed using machine learning techniques in this work. Wheat area, wheat perimeter, compactness, kernel length, kernel width, asymmetry coefficient, and kernel groove length are the 7 physical parameters used to categorize the seeds. The dataset includes 210 separate instances of wheat kernels, and was compiled from the UCI library. The 70 components of the dataset were selected randomly and included wheat kernels from three different varieties: Kama, Rosa, and Canadian. In the first stage, we use single machine learning models for classification, including multilayer neural networks, decision trees, and support vector machines. Each algorithm's output is measured against that of the machine learning ensemble method, which is optimized using the whale optimization and stochastic fractal search algorithms. In the end, the findings show that the proposed optimized ensemble is achieving promising results when compared to single machine learning models.

Abstract:
A Review of:
Shachaf, Pnina, and Sarah M. Horowitz. “Virtual Reference Service Evaluation: Adherence to RUSA Behavioral Guidelines and IFLA Digital Reference Guidelines.” Library & Information Science Research 30.2 (2008): 122-37.

Objectives – This study evaluates the level to which virtual (asynchronous e-mail) reference services adhere to professional guidelines. Specifically, it addresses the following research questions:
 1) To what extent do virtual reference services adhere to the American Library Association (ALA) Reference and User Services Association (RUSA) and the International Federation of Library Associations (IFLA) guidelines?
 2) How does the level of adherence to RUSA or IFLA guidelines vary based on request type, user name, and institution?
 3) Is there a correlation between outcome measures of reference transactions (accuracy, completeness, and satisfaction) and the level of adherence to RUSA or IFLA guidelines?

Design – Unobtrusive evaluation of researcher-generated queries.

Setting – Fifty-four academic libraries in North America.
Subjects – A total of 324 queries were sent to the 54 libraries, with each library receiving six different types of requests from six different user names.

Methods – Researchers developed two coding schemes for the guidelines (34 codes and 12 attributes for the RUSA guidelines and 33 codes and 10 attributes for the IFLA guidelines). Each of the six user names used represented an ethnic and/or religious group identity: Mary Anderson (Caucasian, Christian), Moshe Cohen (Caucasian, Jewish), Ahmed Ibrahim (Arab), Latoya Johnson (African American), Rosa Manuz (Hispanic), and Chang Su (Asian). The six request types were designed so that three would be answered (questions 1-3) and three would be out of scope and not answered (questions 4-6). The following queries were sent, individualized for each institution: 1) Dissertation query; 2) Sports team query; 3) Population query; 4) Subject query; 5) Article query; 6) Request for a PDF copy. The 324 queries were uploaded into NVivo 2 software, and all e-mail transactions were coded and analyzed.
Main Results – Analysis of the 324 transactions from 54 libraries showed the following results:
 1) Low levels of adherence to both sets of guidelines;
 2) Varied levels of adherence based on request types and user names on
 both sets of guidelines;
 3) Variation in institutional rank according to different sets of guidelines;
 4) No correlation between user satisfaction and adherence to either set of guidelines.

Conclusion – This study suggests that higher levels of virtual reference service effectiveness could be achieved by automatically integrating some less observed behaviours (e.g., thank you notes) into replies sent to users and by increasing librarians’ awareness of professional guidelines through training and detailed institutional policies. The authors also suggest that librarians should be aware of their tendencies to react differently to different user groups, and that administrators can facilitate this by providing diversity workshops.

Abstract:
Purpose This paper aims to describe the Rosarium Project, a digital humanities project being undertaken at the Phillips Memorial Library + Commons of Providence College in Providence, Rhode Island. The project focuses on a collection of English language non-fiction writings about the genus Rosa. The collection will comprise books, pamphlets, catalogs and articles from popular magazines, scholarly journals and newspapers written on the rose published before 1923. The source material is being encoded using the Text Encoding Initiative (TEI) Consortium’s P5 guidelines and the extensible markup language (XML) editor software <oXygen/>. Design/methodology/approach This paper outlines the Rosarium Project and describes its workflow. This paper demonstrates how to create TEI-encoded files for digital curation using the XML editing software <oXygen/> and the TEI Archiving Publishing and Access Service (TAPAS) Project. The paper provides information on the purpose, scope, audience and phases of the project. It also identifies the resources – hardware, software and membership – needed for undertaking such a project. Findings This paper shows how straightforward it is to encode transcriptions of primary sources using the TEI and XML editing software and to make the resulting digital resources available on the Web. Originality/value This paper presents a case study of how a research project transitioned from traditional printed bibliography to a web-accessible resource by capitalizing on the tools in the TEI toolkit using specialized XML editing software. The details of the project can be a guide for librarians and researchers contemplating digitally curating primary resources and making them available on the Web.

Abstract:
A review of:

Shachaf, Pnina, and Sarah Horowitz. "Are Virtual Reference Services Color Blind?" Library & Information Science Research 28.4 (Sept. 2006): 501-20.

 Abstract

Objective – To examine whether librarians provide equitable virtual reference services to diverse user groups.

Design – Unobtrusive method of defined scenarios submitted via e-mail.

Setting – Twenty-three Association of Research Libraries (ARL) member libraries from across the United States. All ARL member libraries were invited to participate, with the 23 acceptances providing 19% participation.

Subjects – Anonymous librarians from the 23 participating libraries’ virtual e-mail reference services. Up to 6 librarians from each library may have been involved.

Six fictitious personas were developed to represent particular ethnic or religious groups, whereby the ethnic or religious affiliation was only indicated by the name chosen for each user and the corresponding e-mail address. Names were selected from lists of names or baby names available online: Latoya Johnson (African-American), Rosa Manuz (Hispanic), Chang Su (Asian - Chinese), Mary Anderson (Caucasian/Christian), Ahmed Ibrahim (Muslim), and Moshe Cohen (Caucasian/Jewish). These personas were used to submit reference queries via e-mail to the virtual reference services taking part in the study.

Methods – Five different types of reference queries were developed for use in this study. Three were based on prior published research as they were deemed to be answerable by the majority of libraries. They included a dissertation query, a sports team query, and a population query all designed to be tailored to the target institution. The other 2 queries were developed with participating institutions’ virtual reference guidelines in mind, and were thought to not be answered by the target institutions when submitted by unaffiliated users. They consisted of a subject query on a special collection topic that asked for copies of relevant articles to be sent out, and an article query requesting that a copy of a specific article be e-mailed to the patron.

The study was conducted over a 6 week period beginning the second week of September, 2005. Each week, 1 fictitious persona was used to e-mail a reference query to the virtual reference service of each of the 23 participating institutions. Five of each type of query were sent by each persona. During September and October 2005, a total of 138 queries were sent. Each institution received a different query for each of the first 5 weeks, and in the sixth week they received a repeat of a previous request with details of title or years altered. All other text in every request sent was kept consistent. Each institution only received 1 request from each persona during the study.

In order to eliminate any study bias caused by an informed decision regarding the order in which personas were used, they were randomly arranged (alphabetically by surname). Furthermore, to avoid suspicions from responding librarians, queries were e-mailed on different days of the week at different times. This created some limitations in interpretating response times as some queries were submitted on weekends.

All queries were analysed by Nvivo software in order to identify attributes and patterns to aid qualitative analysis. Each transaction (a single query and any related responses) was classified according to 12 attributes and 59 categories based on various associations’ digital reference guidelines. Transactions were coded and then 10% re-coded by a different coder. This led to the clarification and refinement of the coding scheme, resulting in the number of categories used being reduced to 23. Coding was then performed in 3 iterations until 90% agreement between the 2 coders was reached. The final inter-coder reliability was 92%. The study did not support cross tabulation among user groups on most content categories due to the small sample size.

Main results – Response times varied greatly between users. Moshe (Caucasian/Jewish) received an average turn-around of less than a day. At the other end of the spectrum, Ahmed’s (Muslim) responses took an average of 3.5 days. Both Ahmed and Latoya (African-American) sent queries which took over 18 days to receive a response. The length (number of words) of replies also indicated a differing level of service with Mary (Caucasian/Christian) and Moshe receiving far lengthier responses than the other 4 personas. Number of replies (including automatic replies) was examined in comparison with the number of replies which answered the question, and again indicated Mary and Moshe were receiving a better level of service.

The way in which the user was addressed by the librarian was examined as another measure of service, i.e. first name, full name, honorific. This again mirrored the low level of service received by Ahmed. The professional endings used by librarians in their replies also reinforced the high quality of service received by Moshe across other categories.

Results for Rosa (Latino) and Chang (Asian - Chinese) were average for most categories presented.

Conclusion – In this study, a discriminatory pattern was clearly evident, with the African-American and Muslim users receiving poor levels of service from virtual reference librarians across all dimensions of quality evaluated. The Caucasian (Christian and Jewish) users also noticeably received the best level of service. It is noted, however, that the sample size of the study is not large enough for generalisations to be drawn and that future, more statistically significant studies are warranted. Many other questions are raised by the study for possible future research into racism exhibited by library staff and services.

Abstract:
Myelodysplastic syndromes (MDS) ultimately will progress to a higher-risk form or acute myeloid leukemia. This evolution is accompanied by acquisition of genetic hits following ancestral mutations, with further subclonal diversification of the clonal architecture. Conceptually, therapeutic approaches targeting ancestral hits have the potential to eradicate MDS at early stages of ontogenesis. Founder SF3B1 mutations are frequent in MDS and therefore represent rational targets for drug development. During our drug discovery efforts we identified an existing drug that selectively inhibits the growth of SF3B1 mutant (SF3B1MT) cells. We consequently examined the effects of known compounds possibly arresting clonal expansion of SF3B1MT MDS cells. For our studies, we generated CRISPR/Cas9 knock-in human cells stably expressing the recurrent SF3B1 K700E mutation and a chromophore-tagged reporter (mRFP). Because of the high frequency of SF3B1 mutations and their effects on the splicing of a multitude of genes, we hypothesized that some of the corresponding downstream effects could be effectively targeted. Using luminescent cell viability assays and flow cytometry analysis, we screened a 3,000 compound library for selective sensitivity against isogenic SF3B1MT cells. A subset of this library contained 23 calcium2+ channel blockers (CCBs) of which one specific dihydropirimidine (DHP) showed the highest inhibitory activity against SF3B1MT cells. Several studies point towards a role of calcium signaling in MDS. Proteins active in calcium metabolism (GPR68, calpain, calpastatin) are involved in modulating sensitivity to drugs used in MDS (e.g., lenalidomide). Divalent substrates including Fe2+ may use CCs and it's plausible that our DHP can inhibit iron overload by blocking Fe2+ trafficking through L-type CCs. In this regard, CCBs have been shown to stimulate the activity of adenosine 5'-triphosphate (ATP)-binding cassette (ABC) transporters. Furthermore, it is known that the SF3B1 mutations reduce the expression of the iron transporter ABCB7, leading to increased iron accumulation. All of this led us to further evaluate the effects of DHP in vitro and in vivo. DHP had higher growth inhibitory activity against SF3B1MT cells when tested in vitro using 8 serial concentrations in half-log dilutions for a period of 3 days (20% and 60% growth inhibition at 1 and 3μM). Mixed competitive experiment of K700EmRFP and WTGFP cells treated with increased doses of DHP (1, 3, 5, 10, 20, 50 μM) reduced the competitiveness of K700EmRFP over the time inducing a 3-fold reduction at 50μM. K700EmRFP cells expressed half the amount of ABCB7 mRNA compared to WTGFP cells by RT-PCR. Therapeutic index provided by DHP was determined in vivo. Bone marrow cells of B6. Gt(ROSA)26Sortm1Sor/J (CD45.2) donors were transplanted in pre-lethally irradiated B6.SJL-PtprcaPepcb/BoyJ (CD45.1) recipients. Two weeks after transplantation, average engraftment (measured as percentage of CD45.2(+) donor cells) was 96% ± 0.02. DHP (10 mg/Kg) was orally administered. No decrease in the proportion of CD45.2(+) donor cells was seen post-treatment compared to pre-treatment (96±0.01 % vs. 96%±0.009). Similarly no change in the proportion of CD45.2(+) donor cells was observed in competitive repopulation experiments when B6. Gt(ROSA)26Sortm1Sor/J cells were mixed 1:1 with transgenic Sf3b1+/K700E cells (40%±0.13 vs. 44%±0.02). In contrast, preliminary experiments showed that DHP had an effect on reducing Sf3b1+/K700E allele burden in two chimeric mice (to 21% and 24%) compared to pre-treatment. DHP is structurally unique in comparison to other DHP-based CCBs; in that it possess a -CH20-CH2CH2NH2 moiety linked directly to the DHP scaffold that may in itself provide opportunities or modes for Fe2+ chelation as well as its L-type CCB activity. Our observations and structural analyses therefore provide impetus to explore this feature to possibly improve the drug's efficacy. In sum, we have demonstrated that the clonal growth of cells carrying SF3B1 mutations might be suppressed using the known L-type CCB DHP. This might represent a novel modulator of ATPase activity of ABC transporters in SF3B1MT expressing low ABCB7 levels. Aquatic Larvae of Stenopsychid caddisfly (Stenopsychie marmorata) survive by attaching its catching nets at the bottom of the rocks in the flowing water. It was hypothesized that S. marmorata larva connects small pebbles by producing both silk-like protein and strong adhesive protein simultaneously. A 98 kDa protein(Smap-98k)was identified as an adhesive component by constructing a silk gland –specific cDNA library of S. marmorata. The cDNA sequence of Smap-98k was 2,679 bp long and encoding a 893 amino acids–long open reading frame (ORF) in which the first 19 residues are predict to be the signal peptide. The alignment of the Cys residues indicated the primary structure of this protein to consist of 15 degenerated repeats, each about 50 residues long and contains 6 conserved Cys residues. The Smap-98k was characterized by an abundance of Cys residues and charged amino acids with epidermal growth factor-like (EGF-like) structure. The most common amino acid of this protein was Cys (11.98%), with Pro (9.91%) and Glu (9.26%) following order of magnitude. Cys was assumed to play a role in maintaining the topology of charged amino acids on the molecular surface by intramolecular disulphide-bond formation. The gene was expressed specially in the silk gland similarity to the major silk proteins such like heavy fibroin (H-fibroin) and Light fibroin (L-fibroin) of S. marmorata larvae. The sequence of the protein showed certain homology to the silk-185 kDa of Chironomus pallidivittatus (Midge) which also spin silk underwater. The characterizations of abundance of Cys residues and charged amino acids also shared by Megabalanus rosa cement protein (Mrcp-20k ) and Mytilus galloprovincialis foot protein 2 (Mgfp 2) which both were produced in the marine environment. Although the similarity among Smap-98k, Mrcp-20k and Mgfp 2 sequences were very low, the functional relationship in underwater adhesion of these proteins should be noted.

Abstract:
AbstractThey never met. But the ‚friendship by correspondence‘ (1628–1634) between the traveller Pietro Della Valle and the desk-bound librarian of the imperial library in Vienna, Sebastian Tengnagel, was built on a solid foundation: their shared passion for the Christian and Muslim East, its languages and its books was an „iron bond“ that gave rise to a close and immediate understanding between two men, despite their huge differences of temperament and experience. However, these letters (kept at the Vatican Archives and the Austrian National Library) do not just bear witness to the growing knowledge of the East in the first half of the 17th century. They also show that early Catholic Orientalism had its roots in a highly stratified terrain, in which military conflict, irenic tensions, missionary propaganda, philological investigation, religious disputes, a rejection of book censorship, and theories on sovereignty were layered and interwoven. But how, and why, would anyone choose to become an orientalist, and what did it entail? Rome and Vienna, the cities from which the letters were written, were both normative centres with universalist ambitions; both were engaged in a profound rethinking and redefinition of secular and religious power. The pages written by Tengnagel and Della Valle reflect the writers’ love of and interest in books. But reading carefully between the lines you can also hear the ‚noises off‘ of the cities in which they were penned.

Abstract:
Role of the library in documenting the scholarship of Lublin University of Technology academicsThe paper presents new functions and roles performed by Lublin University of Technology Library in context of the university parametric marks. A library has always played an important role in the scientific environment of universities. The primary aim of its activity is supporting the academics’ scientific performance. It has been done by providing the documentation of their scholarship. The composites of information on publications from various sources not only register the items but also positively influence the scholarly communications by enhancing discoverability. These databases, additionally equipped with tools necessary to analyse citations, start to play a more and more important role. Our bibliographic database has been enriched recently by new parametric indicators: journal mark indicators, citations, for example, according to the Web of Science. It allows the library to support the system of evaluation of scientific institutions conducted by the Ministry of Education and Higher Education in Poland. The paper presents a description of the important role played by the Scientific and Technical Information Department at the University as the main coordinator concerning documentation and information including parametric evaluation of the university 2013–2016.

Abstract:
The article presents the tasks, activities and role of the National Library in Sofia in the period since Bulgaria regained its independence in 1878 to taking over power by the communists in 1944. The introductory part shows the cultural development of the country. Further considerations considered circumstances of establishment of the national library and the first legal regulations. The biggest difficulty was finding a suitable building for the library. Its regular work was stopped by the World War II. A few headmasters merited for the development of significant scientific and cultural institutions of Bulgaria. The following are presented in detail in the text: Konstantin Ireczek,Penczo Slawejkow, Nikola Michow, and Stilian Czilingirow. Aside from regular library processes, protection of the Balkan cultural heritage belonged to the most important task of the National Library. Both this function and collection were the prime and foremost confirmation of distinctness and independence of the people after nearly 500 years of Turkish captivity.

Abstract:
The first significant exhibition devoted to Roma/ Gypsy history and culture was organised in the Regional Museum in Tarnów in 1979. After its success, collections connected with the history of this ethnic group were initiated. The Ethnographic Museum in Tarnów (the branch of the Regional Museum) opened a permanent exhibition entitled “Gypsies. History and culture” as the first permanent museum exhibition devoted to Roma matters in 1990. The Gypsy collections of the museum amount to almost 1000 exhibits; moreover, it gathers professional photographic, cinematographic and phonographic documentation and archives, and it runs a specialised library. Based on this permanent Roma exhibition and collections, the museum in Tarnów organises numerous cultural and educational projects, as well as those that promote Roma culture and history. The International Roma Caravan Memorial is a regular event which has been organised since 1996. It is a project which consists in a kind of a reconstruction of a Gypsy wandering caravan, during which the participants visit places connected with Roma martyrdom during the Second World War in Małopolska. In addition, the museum publishes a scientific annual entitled “Studia Romologica”.
