Abstract:
Clathrin coated vesicles were the first type of coated vesicle to be characterised. The coat consists of two components, clathrin and adaptor (or AP) complexes, the AP-1 complex is associated with the clathrin coated vesicles that bud from the TGN and the AP-2 complex is associated with the clathrin coated vesicles that bud from the plasma membrane. A new type of adaptor-like complex was discovered in our laboratory and was published in 1996. The putative CACHD1 gene was identified following a systematic search for ligand-binding proteins as novel drug targets. The CACHD1 gene encodes CACHD1 and has a predicted protein structure similar to that of the [alpha]2[delta] family of voltage-gated Ca2+ channel (VGCC) auxiliary subunits, which modulates the biophysical properties and plasma membrane expression of VGCCs. On this basis, the hypothesis that CACHD1 represents a novel [alpha]2[delta]-like VGCC auxiliary subunit was tested.

The initiation of transcription by the Bacillus subtilis sigma-M3 RNA polymerase at two Bacillus phage ɸ29 promoters and the effect of the delta subunit on initiation have been investigated by an in vitro kinetic analysis. The templates for the analysis were plasmids which carried the ɸ29 A2 or G2 promoter. The cloning and localization of the A2 promoter are reported here. Ca2+ entry through voltage-gated calcium channels (CaVs) triggers a range of physiological events, including synaptic neurotransmission and muscular excito-contraction coupling.
