Abstract:
The binding data for oxygenation of human hemoglobin, Hb, at various temperatures and in the absence and presence of 2,3-diphosphoglycerate, DPG, and inositol hexakis phosphate, IHP, were analyzed for extraction of mean intrinsic Gibbs free energy, DeltaGo, enthalpy, DeltaHo, and entropy, DeltaSo, of binding at various partial oxygen pressures. This method of analysis considers all the protein species present such as dimer and tetramer forms which were not considered by Imai et al. (Imai K et al., 1970, Biochim Biophys Acta 200: 189-196), in their analysis which was based on Adair equation. In this regard, the values of Hill equation parameters were estimated with high precision at all points of the binding curve and used for calculation of DeltaGo, DeltaHo and DeltaSo were also calculated by analysis of DeltaGo values at various temperatures using van't Hoff equation. The results represent the enthalpic nature of the cooperativity in Hb oxygenation and the compensation effect of intrinsic entropy. The interpretation of results also to be, into account the decrease of the binding affinity of sites for oxygen in the presence of DPG and IHP without any considerable changes in the site-site interaction (extent of cooperativity). In other words, the interactions between bound ligands, organic phosphates and oxygen, are more due to a decreasing binding affinity and not to the reduction of the cooperative interaction between sites. The results also document the more heterotropic effect of IHP compared to DPG.

Abstract:
Background To identify fluid responsiveness, a correlation between respiratory variations in pulse pressure (DeltaPP) and respiratory variations in pulse oximetry photoplethysmographic waveform amplitude (DeltaPOP) in mechanically ventilated patients has been demonstrated. To evaluate the agreement between the two methods, knowledge about the repeatability of the methods is imperative. However, no such data exist. Based on knowledge of slow oscillation in skin blood flow, the authors hypothesized that the variability of DeltaPOP would be larger than that of DeltaPP when calculations were performed continuously over a long recording period. Methods Respiration, continuous invasive blood pressure, pulse oximetry, and skin microcirculation were recorded in 14 mechanically ventilated intensive care unit patients. No intravenous fluid challenges were given, and no other interventions were performed during the measurements. Seventy consecutive comparisons between DeltaPP and DeltaPOP were calculated for each of the 14 patients. Results For all patients, DeltaPOP was 13.7 +/- 5.8% and DeltaPP was 5.8 +/- 2.6% (P < 0.001). There was a larger intraindividual (8.94 vs. 1.29; P < 0.001) and interindividual (26.01 vs. 5.57; P < 0.001) variance of DeltaPOP than of DeltaPP. In six patients, there was no significant correlation between DeltaPP and DeltaPOP. A Bland-Altman plot showed poor agreement between the two methods. Conclusion A large variability of DeltaPOP and a poor agreement between DeltaPP and DeltaPOP limits DeltaPOP as a tool for evaluation of fluid responsiveness in intensive care unit patients. This is in contrast to DeltaPP, which shows a small variability.

Abstract:
Background Respiratory variations in pulse oximetry plethysmographic waveform amplitude (DeltaPOP) are related to respiratory variations in pulse pressure (DeltaPP) and are sensitive to changes in preload. The authors hypothesized that DeltaPOP can predict fluid responsiveness in mechanically ventilated patients during general anesthesia. Methods Twenty-five patients referred for cardiac surgery were studied after induction of general anesthesia. Hemodynamic data (cardiac index, central venous pressure, pulmonary capillary wedge pressure, DeltaPP, and DeltaPOP) were recorded before and after volume expansion (500 ml hetastarch, 6%). Fluid responsiveness was defined as an increase in cardiac index of 15% or greater. Results Volume expansion induced changes in cardiac index (2.0+/-0.4 to 2.3+/-0.5 mmHg; P<0.05), DeltaPP (11+/-7 to 6+/-5%; P<0.05), and DeltaPOP (12+/-9 to 7+/-5%; P<0.05). DeltaPOP and DeltaPP were higher in responders than in nonresponders (17+/-8 vs. 6+/-4 and 14+/-7 vs. 6+/-4%, respectively; P<0.05 for both). A DeltaPOP greater than 13% before volume expansion allowed discrimination between responders and nonresponders with 80% sensitivity and 90% specificity. There was a significant relation between DeltaPOP before volume expansion and percent change in cardiac index after volume expansion (r=0.62; P<0.05). Conclusions DeltaPOP can predict fluid responsiveness noninvasively in mechanically ventilated patients during general anesthesia. This index has potential clinical applications.

Abstract:
Expression of the proapoptotic protein Bax under the control of a GAL10 promoter in Saccharomyces cerevisiae resulted in galactose-inducible cell death. Immunofluorescence studies suggested that Bax is principally associated with mitochondria in yeast cells. Removal of the carboxyl-terminal transmembrane (TM) domain from Bax [creating Bax (deltaTM)] prevented targeting to mitochondrial and completely abolished cytotoxic function in yeast cells, suggesting that membrane targeting is crucial for Bax-mediated lethality. Fusing a TM domain from Mas70p, a yeast mitochondrial outer membrane protein, to Bax (deltaTM) restored targeting to mitochondria and cytotoxic function in yeast cells. Deletion of four well-conserved amino acids (IGDE) from the BH3 domain of Bax ablated its ability to homodimerize and completely abrogated lethality in yeast cells. In contrast, several Bax mutants which retained ability to homodimerize (deltaBH1, deltaBH2, and delta1-58) also retained at least partial lethal function in yeast cells. In coimmunoprecipitation experiments, expression of the wild-type Bax protein in Rat-1 fibroblasts and 293 epithelial cells induced apoptosis, whereas the Bax (deltaIGDE) mutant failed to induce apoptosis and did not associate with endogenous wild-type Bax protein. In contrast to yeast cells, Bax (deltaTM) protein retained cytotoxic function in Rat-1 and 293 cells, was targeted largely to mitochondria, and dimerized with endogenous Bax in mammalian cells. Thus, the dimerization-mediating BH3 domain and targeting to mitochondrial membranes appear to be essential for the cytotoxic function of Bax in both yeast and mammalian cells.

Abstract:
Notch-mediated cellular interactions are known to regulate cell fate decisions in various developmental systems. A previous report indicated that monocytes express relatively high amounts of Notch-1 and Notch-2 and that the immobilized extracellular domain of the Notch ligand, Delta-1 (Deltaext-myc), induces apoptosis in peripheral blood monocytes cultured with macrophage colony-stimulating factor (M-CSF), but not granulocyte-macrophage CSF (GM-CSF). The present study determined the effect of Notch signaling on monocyte differentiation into macrophages and dendritic cells. Results showed that immobilized Deltaext-myc inhibited differentiation of monocytes into mature macrophages (CD1a+/−CD14+/− CD64+) with GM-CSF. However, Deltaext-myc permitted differentiation into immature dendritic cells (CD1a+CD14−CD64−) with GM-CSF and interleukin 4 (IL-4), and further differentiation into mature dendritic cells (CD1a+CD83+) with GM-CSF, IL-4, and tumor necrosis factor-α (TNF-α). Notch signaling affected the differentiation of CD1a−CD14+macrophage/dendritic cell precursors derived in vitro from CD34+ cells. With GM-CSF and TNF-α, exposure to Deltaext-myc increased the proportion of precursors that differentiated into CD1a+CD14− dendritic cells (51% in the presence of Deltaext-myc versus 10% in control cultures), whereas a decreased proportion differentiated into CD1a−CD14+ macrophages (6% versus 65%). These data indicate a role for Notch signaling in regulating cell fate decisions by bipotent macrophage/dendritic precursors.

Abstract:
Protective immunity against larval Taenia crassiceps has been shown to rely on T cells; however, the roles of the specific subsets of T cells during infection are not known. To investigate a possible role for γδT cells, this study investigated larval infection in δ-chain knock-out C57BL/6 (deltaKO) and wild-type C57BL/6 mice. It was found that deltaKO mice and C57BL/6 mice were equally susceptible to infection suggesting γδT cells do not play a major role in protective immunity. Cytokine production by concanavalin A (ConA)-stimulated spleen cells from infected deltaKO mice and C57BL/6 mice were determined. All infected mice demonstrated an increased IL-10 production suggesting a Th1-inhibitory function. Cells from infected deltaKO mice and C57BL/6 mice did not show increases in IL-4 production. Heavily-infected C57BL/6 mice showed a decrease in IFN-γ production compared to deltaKO mice. These observations suggest that an increase in IL-10 production best correlates with a non-protective immune response. To make comparisons between in vitro cytokine production and systemic immune responses, cytokine levels in serum were determined. C57BL/6 mice and deltaKO mice showed increases in serum levels of IL-4 and IFN-γ at 52 days post-infection. The systemic immune response of these mice, therefore, is a mixed Th1/Th2-type response and γδT cells are apparently not responsible for the systemic increases in these cytokines.

Abstract:
Abstract This study involves the experiences of children and adolescents with regard to their participation in the Golden Life martial arts project of the Golden Leaf Sports Association. I have conducted qualitative research, in which I have randomly selected children and adolescents who have then participated in semi-structured interviews. The purpose of this study is to understand how children and adolescents experienced the martial arts project in the fall of 2017, and what happened to the participants after the project was over. The question of this paper thus becomes how the martial arts project served as an arena of socialization. The theory used is situational learning from Lave and Wenger (1991) in which social learning used in athletic contexts can be applied to people embracing instructions. Futhermore this theory relates to how physical activity in a social context facilitates learning. But also legitimate peripheral participation that deals with how to acquire knowledge where knowledge building is secondary to the ability to learn and acquiring knowledge in a group already established. In such a way, legitimacy is created and it is within this framework that one learns what is going to be performed. The result of this study is based on the respondents own stories, where an analysis is made in relation to the theory and, to a certain extent, the previous research relating to sport and integration in relation to the sociological process. The result from the gathering of the data also shows how there is a connection between what the interviewees described in relation to the theory of legitimate peripheral participation. The resemblance between how the excercise is described and the actual training is easily identified. The martial arts environment and the environment's structure of coaches and other members of the association can be clearly linked to the theory. Due to the association being open to other people, even if they do not belong to the regular business they do invite others for participation. This means that the participants feel welcome, which leads to the participation and learning of martial arts.

Abstract:
BACKGROUND: Breastfeeding provides health benefits for children. In order tosucceed with breastfeeding, women need support from family and healthcareprofessionals, but their own coping strategies are crucial. Earlier studies highlight theneed for further development of breastfeeding education.DESIGN: A retrospective cross-sectional study with qualitative approach using openquestions to process the answers.METHOD: Between September 2012 and March 2014, 303 women responded toAmningshjälpen’s survey. All women had participated in breastfeeding education. The data was analysed with qualitative content analysis with an inductive approach.RESULTS: The result is presented as two themes and four subthemes withassociated categories. The first theme, “Strengthened self-esteem when breastfeedingworks, despite problems during breastfeeding”, with subthemes “Feeling pride fromsucceeding with breastfeeding” and “Solving breastfeeding problems”, described theexperiences of women who succeeded with breastfeeding. The second theme, “Lostself-esteem when breastfeeding is a constant struggle” with subthemes “Strugglingwith breastfeeding, leading to decommissioning” and “Experiencing crushedexpectations” described the experiences of women who were unsuccessful withbreastfeeding.CONCLUSION: Participating in the breastfeeding education helped many women.Recurrent breastfeeding problems lower the woman’s self-esteem. Nurses have animportant role in supporting women who breastfeed. Resources should be put intomaking sure that pediatric nurses’ education in breastfeeding is up to date, and thatthey have the ability to support women with breastfeeding problems

Abstract:
The purpose of this study is to examine how students with reading and writing disabilities experienced participation in intensive instruction in reading. Such an achievement means that students every day for 4-6 weeks is working about 20 minutes with structured teaching with the aim of developing their reading skills. The research about intensive instruction in reading coherently show that similar achievements are producing good results. In contrast, this research does not include how students experience intensive instruction. Therefore, I have chosen to examine some students' experiences of this and focused on the experience of working, the experience of their own reading development and how it is to leave the classroom to participate in intensive instruction.   The study is based on qualitative interviews with six students in grade 4 who have participated in intensive instruction in reading. Each student was interviewed individually and their stories were then analysed based on a life-world phenomenological approach from a student perspective.   The result shows that these students are positive towards working with intensive instruction in reading and experience themselves having developed their reading skills through it. They describe their own development in relation to the material they have worked with and what it has meant to them both in school and at home. The students mention both positive and negative experiences of leaving the classroom to participate in intensive instruction.

Abstract:
Introduction: Being a relative of a person with an addiction disease involves a strong mental and physical stress. An important protection factor for the relatives is breaking the silence and shame and seek help. Ersta Vändpunkten is a clinic for these relatives and offers educational groups with a therapeutic objective. This study focuses on program for parents/relatives to children in the so-called little child group (4-6 years). Focus in the program is to support the parents in how they can help the child to understand what is happening in the family and how to take the child’s perspective. The aim of the study is to share the parent’s experiences of participating in this group. Questions: How has participation in the parenting program affected parenting ability? How has participation in the parent group affected the family? Methods: This is a web-based survey, the questionnaire has been analyzed with descriptive statistics. Results: 30 parents/relatives have been asked to participate of whom 17 responded (57%). The survey reveals that the parent/relative now has an improved relationship with the child and that communication in the family has changed for the better. Other relationships, to relatives, friends and to the addictive part have not changed noticeably. All respondents feel the child’s and their own self-esteem have become much better/better after participating. There is also a consensus among respondents that their ability to set boundaries towards the addicted part has become much better/better. Discussion: While the high non-response rate endangers firm conclusions, the findings are nevertheless promising.

Fem stycken sjuksköterskor intervjuades under november 2017.

Abstract:

Abstract:

Abstract:

Abstract:
The growing needs for high speed positioning devices in the automated manufacturing industry have been challenged by robotic science for more than two decades. Parallel manipulators have been widely used for this purpose due to their advantage of lower moving inertia over the conventional serial manipulators. Cable actuated parallel robots were introduced in 1980’s to reduce the moving inertia even further. In this work, a new cable-based parallel robot is introduced. For this robot, the cables are used not only to actuate the end-effector but also to apply the necessary kinematic constraints to provide three pure translational degrees of freedom. In order to maintain tension in the cables, a passive air cylinder is used to push the end-effector against the stationary platform. In addition to low moving inertia, the new design benefits from simplicity and low manufacturing cost by eliminating joints from the robot’s mechanism. The design procedure and the results of experiments will be discussed in the following.

Abstract:
Artificial Intelligence has seen several breakthroughs in two-player perfect information game. Nevertheless, Doudizhu, a three-player imperfect information game, is still quite challenging. In this paper, we present a Doudizhu AI by applying deep reinforcement learning from games of self-play. The algorithm combines an asymmetric MCTS on nodes of information set of each player, a policy-value network that approximates the policy and value on each decision node, and inference on unobserved hands of other players by given policy. Our results show that self-play can significantly improve the performance of our agent in this multi-agent imperfect information game. Even starting with a weak AI, our agent can achieve human expert level after days of self-play and training.

Abstract:
ACME facility (Advanced Core-cooling Mechanism Experiment) is a large-scale test facility used to validate the performance of passive core-cooling system under SBLOCA (Small Break Lost of Coolant Accident) for the CAP1400, an upgraded passive safety nuclear power plant of AP1000. To simulate the features of passive safety system properly, DELTABAR, a kind of differential pressure flow meter, is designed to measure different mass flow of ACME. Because of the low pressure loss of DELTABAR, Zero-Drift problem of differential pressure flow meters in ACME is amplified, and some of the measured values are distorted seriously. To minimize the influence of Zero-Drift, analysis on zero-drift phenomenon is made, and a compensation method is proposed. The method is applying to PBL flow meters, and the result shows that the method is applicable.
