Full textThe genus Salmonella belongs to the Enterobacteriaceae family and comprises two species: S. bongori and S. enterica. Both species include more than 2500 serovars. Within S. enterica serovars, the most studied are S. Typhi, S. Typhimurium and S. Enteritidis, the latter being the main causative agent of salmonellosis worldwide. During its infective cycle, Salmonella needs to adapt to variations in different environmental conditions, such as changes in pH, osmolarity and oxygen availability. In addition, these conditions serve as environmental signals that modulate the expression of virulence factors. One of these factors is the lipopolysaccharide (LPS), the main component of the envelope of Gram negative bacteria. The LPS presents three structural domains: the O antigen (AgO), the core region and lipid A, also called endotoxin. The latter anchors the LPS to the outer membrane, and is a known virulence factor responsible for the induction of the immune response. The lipid A domain is capable of undergoing covalent modifications, which modulate its toxicity and helps bacteria to evade antimicrobial agents and the immune system of the host. One of the main environmental cues faced by Salmonella during its infectious cycle is the oxygen availability. During the aerobic-anaerobic transition participates the global regulators ArcA and Fnr. Results from our laboratory show that changes in the polymerization degree of the AgO in response to oxygen availability is modulated by these regulators. This observation led us to examine whether global regulators Fnr and ArcA are involved in the control of probable oxygen-dependent modifications of lipid A in response to oxygen availability. The main goal of this study was to determine the effect of oxygen availability on covalent modifications of lipid A in S. Enteritidis and the participation of transcriptional factors ArcA and Fnr in this process. First, we mounted a method for LPS extraction followed by a hydrolysis step to obtain a highly-purified lipid A that can be used to study the composition of this molecule by MALDI-TOF. Thus, in aerobiosis we observed four major signals corresponding to hexa-acylated and hepta-acylated lipid A molecules and their corresponding hydroxylated species. Under this environmental condition, hydroxylated species were more abundant than non-hydroxylated species. In the case of anaerobic cultures we detected the same four signal, but this relationship is reversed, being more abundant the nonhydroxylated species. To determine the participation of ArcA and Fnr in the modulation of covalent modifications of lipid A, we generated ΔarcA and Δfnr mutant strains and a strain overproducing ArcA (parcA). Our results indicate that both regulators are involved in controlling structural changes of lipid A in response to oxygen availability. Additionally, we observed that these regulators control the incorporation of other covalent modifications into lipid A. LpxO enzyme (encoded by lpxO gene) is responsible of the hydroxylation of lipid A in Salmonella. Therefore, we studied the relative expression (anaerobic/aerobic) of this gene in wild-type S. Enteritidis and their Δfnr and parcA derivatives via qRT-PCR. Our results indicate that the expression of lpxO depends on oxygen availability, an observation that is consistent with changes in the levels of lipid A hydroxylation described herein. Furthermore, ArcA and Fnr participate in the modulation of the expression of this gene. The results of this study reveal a change in the hydroxylation degree of S. Enteritidis lipid A in response to oxygen availability, in which global regulators ArcA and Fnr are involved. These findings contribute to understanding the molecular mechanisms of adaptation to the environment used by Salmonella during infection

Full textThe Uberabinha River, located in the region of Triângulo Mineiro, in the state of Minas Gerais, Brazil, is part of the Araguari River sub-basin. From its head in the city of Uberaba to its mouth in the Araguari River, it measures approximately 150 km, spanning about 135 km in the municipality of Uberlândia. The river drains a watershed of about 2.190 km2, from which water is collected to supply a population that approaches half a million inhabitants. Later, the sewage produced by the city is also discharged directly into the river. This research consisted in the analyses of pH, temperature, dissolved oxygen and biochemical oxygen demand of samples collected in five points of the Uberabinha River, from a section starting upstream of the urban area down to 20.5 km downstream the last point of sewage discharge, encompassing approximately 42 km. In this river section, the landforms go from low to medium hills with many rapids and waterfalls. The samples were collected from June 1999 to May 2000 and the water flow was measured on the two extremes of the studied river section. The results show that the dissolved oxygen values are reduced when receiving the sewage and it tends to slowly return to its normal saturation rate. On the other hand, the biochemical oxygen demand follows a reverse path, as a result of the natural phenomenon of self-restoration even though the organic material is not yet completely removed from the studied river section.

Full textEl objetivo más importante de la investigación fue lograr sustituir el cambio del agente oxidante utilizado en la técnica de la determinación de la demanda química de oxígeno (DQO), la cual nos indica la cantidad de materia orgánica oxidable en el medio. Al aplicar la química verde, se sustituye al cromo (VI), un agente tóxico y cancerígeno utilizado en la técnica estandarizada (NMX-AA-030-SCFI-2001), por uno más amigable para el medio ambiente como para el ser humano, como lo es el peróxido de hidrógeno (H2O2). Se realizaron determinaciones de DQO utilizando diferentes tipos de agua para ambos métodos, haciendo una comparación de ellos con un análisis estadístico, obteniendo como resultado favorable que dichos resultados son comparables estadísticamente, logrando obtener una técnica más verde. Además de lograr sustituir el agente oxidante, se logra eliminar el proceso de digestión de los tubos (calentamiento a 150º C por 2 h), dejando reaccionar los dos tubos por 2 h sin calentamiento. Otro cambio que se realizó fue la disminución de la concentración del sulfato de plata a la mitad de su concentración original, sin que interfiera éste cambio en los valores de la DQO. Para mejorar aún más la técnica, se aplicó luz ultravioleta para acelerar la reacción, logrando disminuir el tiempo de reacción a 1 h. Finalmente los residuos que se generan de dicha determinación son tóxicos debido a que contienen mercurio, cromo y plata en medio ácido. Por ello se su tratamiento electroquímico, logrando separar la plata del resto de los residuos, mientras que el mercurio queda en la solución. Al final de todo, se obtiene una técnica menos tóxica, menor generación de residuos tóxicos, disminución en el consumo de energía eléctrica, que puede ser utilizada con confianza al ser comparada con la estandarizada por medio de análisis estadístico.

Pesquisas sobre o efeito do tempo e da temperatura de incubação de amostras líquidas, na estimativa da matéria orgânica biodegradável contida nestas, atualmente são raras, tendo em vista que têm sido aceitos, como padrão, os resultados apresentados nos trabalhos clássicos de Streeter e Phelps e de Theriault, divulgados em 1925 e 1927, respectivamente. Entretanto, esses estudos são passíveis de vários questionamentos, principalmente no que se refere às estimativas feitas em amostras de águas residuárias que não sejam o esgoto sanitário ou soluções sintéticas. Dentre as questões pendentes mais instigantes estão a de que seria possível a redução do tempo de incubação das amostras para obtenção da demanda bioquímica de oxigênio (DBO) e que haveria influência da temperatura na demanda bioquímica de oxigênio última (DBOu ) da amostra. Para conhecer o efeito da temperatura nas estimativas de DBO e de DBOu , é necessário, também, entender quais os efeitos da mesma variável na obtenção dos coeficientes de desoxigenação (k) e de temperatura (θ); qual o modelo cinético que melhor descreve a progressão da DBO e o efeito de interferentes nessas análises. Para tanto, foram incubadas amostras de esgotos sanitários tratados em fossa séptica (ESF), águas residuárias de laticínios (ARL) e da suinocultura (ARS), para obtenção do coeficiente de desoxigenação à 20 oC (k20). As mesmas amostras também foram armazenadas sob temperaturas de 30 e 35 oC, por tempo equivalente ao período de incubação de 5 dias e à 20 oC, tempos esses calculados usando a equação de Arrhenius modificada por Streeter e Phelps, obtendo-se, assim, os coeficientes θ30 e θ35. Em outro teste, quantificações de DBO foram feitas em vários dias, usando o esgoto sanitário bruto (ESB), a fim de obter θ a partir dos valores de k20 e k35, e verificando-se se a demanda última varia com a temperatura, com a obtenção do coeficiente de temperatura 20 35 da demanda última (φ) a partir da DBOu e da DBOu . Avaliou-se, também, o efeito da inibição da nitrificação nas amostras sobre os valores estimados de k e, usando os valores de k20 típicos de literatura, também foram estimados os valores de θ, verificando se erros de estimativa de k, implicam em erros significativos do coeficiente de temperatura. Os experimentos foram conduzidos no Laboratório de Qualidade da Água (LQA) do Departamento de Engenharia Agrícola (DEA) da Universidade Federal de Viçosa (UFV). Os valores de θ calculados a partir de k20 obtidos experimentalmente foram de 1,048 para ESF, 1,046 para ARL e 1,020 para ARS, sob a temperatura de 35 o C, e de 1,041, 1,070, 1,028, para as mesmas águas residuárias, quando incubadas à 30 oC. Os valores de θ estimados utilizando-se valores de k20 obtidos na literatura não foram estatisticamente diferentes, em nível de 5% de probabilidade, pelo teste "t". Igual conclusão foi obtida ao se estimar o valor do coeficiente de temperatura, usando o valor de k obtido após novo ajuste do modelo de primeira ordem, utilizando o conjunto de dados depois de expurgo dos dados tardios (não houve diferença significativa). Usando o mesmo teste, verificou-se que o uso do valor de θ de 1,047, recomendado na literatura, não implica em erros significativos na estimativa de k. No segundo experimento, foram obtidos k20 e k35, respectivamente, de 0,1593 e 0,2262 d-1, tendo valor de 20 θ igual a 1,024. Os valores estimados de DBOu 35 e DBOu foram, respectivamente, de 579 mg L-1 e 661 mg L-1. O modelo de segunda ordem apresentou desempenho ligeiramente superior sobre o modelo de primeira ordem, na descrição dos dados. Porém, dada à pequena diferença nos resultados e a simplicidade em sua utilização, o modelo de primeira ordem é, ainda, preferível. Os coeficientes φ ajustados foram de 0,00948 e de 0,00967, respectivamente usando os modelos de primeira e segunda ordens, valores suficientemente pequenos para indicar desprezível ou inexistente variação do valor de DBOu com a temperatura. O aumento na temperatura da amostra proporcionou, no entanto, diferentes valores de θ para as águas residuárias.
Research into the effect of time and temperature incubation of liquid samples, in the estimation of biodegradable organic matter contained in these nowadays are rare, considering that they have been accepted as standard the results presented in classical studies of Streeter and Phelps, and Theriault, released in 1925 and 1927, respectively. However, these studies are subject to various questions, mainly in relation in the estimations made on samples of wastewater other than the sewage or synthetic solutions. Among the outstanding questions, the most intriguing that it would be possible to reduce the incubation time of the samples to obtaining the biochemical oxygen demand (BOD) and that there is effect of temperature on biochemical oxygen demand ultimate (UBOD) of the sample. To determine the effect of temperature on estimates of BOD and UBOD, it is also necessary to understand what the effects of the same variable in obtaining the deoxygenation (k) and temperature coefficients (θ); which the kinetic model that best describes the progression of the BOD and the effect of interferents in these analyzes. Thus, samples of sewage treated in septic tank (ESF), dairy wastewater (ARL) and swine wastewater (ARS) were incubated for obtaining the deoxygenation coefficient at 20 oC (k20). The same samples were also stored at temperatures of 30 and 35 o C, for time equivalent to an incubation period of 5 days and at 20 oC, these times were calculated using the Arrhenius equation modified by Streeter and Phelps, obtaining thereby the coefficients and θ30 θ35. In another test, measurements of BOD were made for several days, using the raw sewage (ESB) to obtain θ from values of k20 and k35, and checking whether the ultimate demand varies with temperature, obtaining the temperature coefficient of the ultimate demand (φ) from UBOD20 and UBOD35 . It was also evaluated the effect of inhibiting the nitrification in the samples on the estimated values of k and, using typical values of k20 literature were estimated values of θ too, verifying if errors in the estimation of k, implying in significant errors in the temperature coefficient. The experiments were conducted at the Laboratory of Water Quality (LQA), Department of Agricultural Engineering (DEA), Federal University of Viçosa (UFV). The values of θ calculated from experimental data of k20 were 1.048 to ESF, 1.046 to ARL and 1.020 to ARS, under a temperature of 35 oC, and 1.041, 1.070, 1.028, for the same wastewater when incubated at 30 oC. The values of θ estimated using values of k20 obtained from the literature were not statistically different at 5% probability, using the "t" test. Same conclusion was obtained when estimating the value of temperature coefficient, using the value of k obtained after new adjust from first order model using the data after the purge of late data (no difference significant). Using the same test, it was found that the use of the value θ of 1.047 recommended in the literature, does not imply significant errors in the estimation of k. In the second experiment were obtained k35 and k20, respectively, 0.1593 and 0.2262 d-1, with θ value of 1.024. The estimated values of UBOD20 and UBOD35 were, respectively, 579 mg L-1 and 661 mg L-1. The second order model performance was slightly higher on the first-order model, in describing the data. However, given the small difference in the results and the simplicity to use, the first order model is also preferred. The results of coefficients φ adjusted were 0.009484 and 0.00967, respectively using the first and second models, values sufficiently small to show no or negligible value UBOD variation with temperature. The increase in temperature of the sample provided, however, different values of θ for the wastewater.

Full textThe purpose of this work was to investigate the effect of high concentration oxygen inhalation over the recovery kinetic of physiological variables modified by physical activity to fatigue, in young healthy men that regularly exercise at least three times a week. With that goal, 12 volunteers followed an standardized incremental exercise protocol on a treadmill, in which they were administered at once, immediately after fatigue, the content of either two 90% oxygen or placebo (21% oxygen) containers, using an oral-nasal mask, with a two to three weeks interval. The oxygen and placebo containers were provided by INDURA S.A., labeled “A” and “B”, in order that neither the researchers nor the volunteers knew which letter corresponded to the 90% oxygen containers (double-blind). At rest and after 0, 4, 14, 21 and 35 minutes post fatigue, cardiac frequency and arterial blood pressure were measured, and blood samples were taken to determine glycemia, lactatemia, hematocrit and total plasmatic protein concentration. This was a comparative, transversal study with a quasi-experimental design. The hypothesis suggests that 90% oxygen administration after fatigue accelerates the recovery kinetic of physiological variables modified by physical activity to fatigue. The data obtained was statistically analyzed with the SPSS v.18.0 computer program, where the Student’s t-test was applied to each one of the related variables. Significantly lower values were observed when administering 90% oxygen (p < 0,05) on systolic arterial pressure (0, 4 and 21 minutes post fatigue), glycemia (21 minutes post fatigue) and lactatemia (14 and 21 minutes post fatigue). Also, hematocrit (0, 21 and 35 minutes post fatigue) and total plasmatic protein concentration (immediately post fatigue) values were significantly higher when 90% oxygen was administered. These results provide evidence with respect to the acceleration of the recovery kinetic of some physiological variables modified by physical activity on the evaluated volunteers, when supplemental oxygen is supplied post fatigue on this standardized exercise protocol.

Full textLa enfermedad de Alzheimer (EA) es un desorden neurodegenerativo caracterizado por la pérdida progresiva de memoria. Los oligómeros solubles sinaptotóxicos del péptido β-amiloide (AβOs) son reconocidos como los agentes causantes de la EA. Los AβOs se unen a las neuronas hipocampales e inducen la generación de señales de Ca2+intracelulares persistentes, que se deben a su entrada por el receptor de N-metil-D-aspartato (NMDA) y la subsecuente amplificación a través dela activación de los receptores de ryanodina (RyR). Los RyR residen en el retículo endoplasmático (RE) y son activados por Ca2+, liberando Ca2+ desde el lumen de este compartimiento al citoplasma. Las señales de Ca2+ persistentes y de baja intensidad generadas por los AβOs y mediadas por los RyR inducen la producción de especies reactivas de oxígeno (ROS) intracelulares por un mecanismo no del todo dilucidado. Por su parte, los ROS disminuyen el umbral de activación de los RyR por Ca2+, debido a que la actividad de estos receptores está estrechamente regulada por su estado redox. Antecedentes del laboratorio muestran que los AβOs, adicionados en concentraciones subletales a cultivos primarios de neuronas de hipocampo, causan la fragmentación de la red mitocondrial y la disminución de los niveles transcripcionales de la isoforma 2 del RyR (RyR2). Adicionalmente, la incubación previa de las neuronas con el antioxidante general N-acetil-cisteína (NAC) previene la generación de señales de Ca2+ aberrantes y la fragmentación de la red mitocondrial inducida por los AβOs. En la presente tesis, se estudiaron los efectos delos antioxidantes NAC y EUK-134 sobre la disfunción mitocondrial y la disminución de la expresión de RyR2 inducidos por los AβOs. Para cumplir con este objetivo, se trataron cultivos hipocampales de ratas con 0,5 μM AβOs y se evaluó: 1) la generación de ROS en la mitocondria utilizando la sonda MitoSox (que detecta superóxido - O2−·) o la transfección de las neuronas con HyPerMito (que detecta peróxido de hidrógeno -H2O2) por microscopia confocal; 2) la morfología de la red mitocondrial en neuronas cargadas con el marcador mitocondrial MitoTracker, por microscopía confocal; y 3) los niveles transcripcionales de RyR2, por PCR en tiempo real. Los resultados muestran que la incubación de las neuronas con 0,5 μM AβOs generó un aumento lento pero progresivo de la concentración de O2−· y deH2O2mitocondrial. El tratamiento previo con NAC o con EUK-134 previno tanto el aumento de los niveles de O2−· como deH2O2 inducidos por los AβOs. De forma interesante, el tratamiento con ryanodina, un inhibidor de los RyR, también previno el aumento en los niveles de O2−· yH2O2 inducidos por los AβOs, sugiriendo que este efecto es mediado por los RyR. Además, la incubación previa de las neuronas con EUK-134 previno la fragmentación de la red mitocondrial inducida por los AβOs, tal como se demostró con NAC. Por último, la incubación con NAC también previno de forma significativa la disminución de la expresión de RyR2 inducida por los AβOs, sugiriendo que los ROS podrían modular la transcripción de RyR2. Como conclusión, los datos obtenidos en esta tesis apoyan la idea de que los efectos deletéreos de los AβOs sobre la mitocondria y sobre los niveles transcripcionales de RyR2 son mediados por el RyR, cuya actividad es, a su vez, modulada por los ROS. Estos efectos deletéreos de los AβOs son prevenidos farmacológicamente por el uso de antioxidantes como el NAC y el EUK-134
Alzheimer's disease (AD) is a neurodegenerative disorder characterized by progressive memory loss. Synaptotoxic soluble oligomers of the β-amyloid peptide (AβOs) are recognized as the causative agents of the disease. AβOs bind to hippocampal neurons and elicit persistent intracellular Ca2+ signals, which are due to its entry by the N-methyl-D-aspartate (NMDA) receptor and subsequent amplification by the activation of ryanodine receptors (RyR). RyRare resident in endoplasmic reticulum (ER) and are activated by Ca2+, releasing Ca2+ from the lumen of this organelle to the cytoplasm. The persisted RyR-mediated Ca2+ signals generated in neurons in response to AβOs induce the production of reactive oxygen species (ROS), by a mechanism not yet fully elucidated. In turn, ROS decrease the threshold of RyR activation by Ca2+, since RyR activity is tightly regulated by its redox state. Previous works from the laboratory showed that AβOs, added at sublethal concentration to primary cultures of hippocampal neurons, induces mitochondrial network fragmentation, increases the concentration of reactive oxygen species (ROS) and causes deregulation of transcriptional levels of RyR2.Interestingly, previous incubation of neurons with the general antioxidant NAC prevented the generation of aberrant Ca2+ signals and mitochondrial fragmentation induced by AβOs. In this thesis, we studied the protective effects of the antioxidants N-acetyl-cysteine (NAC) and EUK-134 on the mitochondrial dysfunction and the decrease in RyR2 expression induced by AβOs. To this, rat hipocampal neurons were treated with 0,5 μM AβOs to evaluate: 1) Mitochondrial ROS generation, using the MitoSox dye (detects superoxide - O2−·) or the transfection of the neurons with HyPerMito (detects hydrogen peroxide -H2O2) by confocal microscopy; 2) Mitochondrial morphology in neurons loaded with the dye MitoTracker, by confocal microscopy; and 3) RyR2 trascriptional levels, by real time PCR. The results showed that the incubation of neurons with 0.5 μM AβOs generated a slow but progressive increase in the mitochondrial concentration of both O2−· and H2O2. Pretreatment with NAC or EUK-134 prevented this effect. Interestingly, treatment with ryanodine, a RyR inhibitor, also prevented the increase in H2O2 andO2−·in response to AβOs, indicating that RyR mediates this effect. Moreover, preincubation of the neurons with EUK-134 prevented mitochondrial network fragmentation induced AβOs, as previously shown with NAC. Finally, the incubation with NAC also significantly prevented the decrease in RyR2 expression induced by AβOs, indicating that ROS might modulate RyR2 transcription. In conclusion, the data obtained in this study support the idea that the deleterious effects of AβOs on mitochondrial morphology and function and on RyR2expression are mediated by RyR, whose activity is modulated by ROS. These deleterious effects of AβOs are prevented pharmacologically by using antioxidants like NAC and EUK-134
