Abstract:
Phosphorus-containing cyclic nitrones, such as DEPMPO, CYPMPO, and DPPMPO, were oxidized by hydrogen tetrachloroaurate(III) to DEPMPOX, CYPMPOX, and DPPMPOX with the precipitation of Au(0). The reaction was depressed by the addition of chloride or hydroxide ions. The peculiar pH dependency was observed in DEPMPOX, CYPMPOX, and DPPMPOX formation, which should be caused by the diethoxyphosphoryl group in DEPMPO, the 1,3-propoxy cyclophosphoryl group in CYPMPO, and the diphenylphosphinoyl group in DPPMPO. The oxidation of the nitrones proceeded through the ligand exchange of Cl– in AuCl4– with >N+–O– in nitrone and the nucleophilic addition of the water molecule to the C-2 position in the nitrones, the stepwise intra-molecular transfer of three electrons from the nitrones to Au(III), and the release of the resulting Au(0). The phosphoryl group in the nitrones suppressed the first ligand-exchange interaction by its electronegativity, while the group promoted the electron transfer from the nitrones to Au(III) by its inductive effect.

Abstract:
Unsaturated lipids are rapidly oxidized to toxic products such as lipid hydroperoxides, especially when transition metals such as iron or copper are present. In a Fenton-type reaction Fe2+ converts lipid hydroperoxides to the very short-lived lipid alkoxyl radicals. The reaction was started upon the addition of Fe2+ to an aqueous linoleic acid hydroperoxide (LOOH) emulsion and the spin trap in the absence of oxygen. Even when high concentrations of spin traps were added to the incubation mixture, only secondary radical adducts were detected, probably due to the rapid re-arrangement of the primary alkoxyl radicals. With the commercially available nitroso spin trap MNP we observed a slightly immobilized ESR spectrum with only one hydrogen splitting, indicating the trapping of a methinyl fragment of a lipid radical. With DMPO or 5-diethoxyphosphoryl-5-methyl-1-pyrroline N-oxide (DEPMPO) adducts were detected with carbon-centered lipid radical, with acyl radical, and with the hydroxyl radical. We also synthesized lipophilic derivatives of the spin trap DEPMPO in order to detect lipid radical species generated in the lipid phase. With all spin traps studied a lipid-derived carbon-centered radical was obtained in the anaerobic incubation system Fe2+/LOOH indicating the trapping of a lipid radical, possibly generated as a secondary reaction product of the primary lipid alkoxyl radical formed. Under aerobic conditions an SOD-insensitive oxygen-centered radical adduct was formed with DEPMPO and its lipophilic derivatives. The observed ESR parameters were similar to those of alkoxyl radical adducts, which were independently synthesized in model experiments using Fe3+-catalyzed nucleophilic addition of methanol or t-butanol to the respective spin trap.

Abstract:
Le premier objectif de ce travail était d'étudier l'effet de la substitution en position 4 de nitrones de type DEPMPO sur la géométrie des adduits superoxyde, sur leur persistance et sur l'allure de leurs spectres RPE. Le deuxième objectif était de fournir des précurseurs pour la synthèse de nitrones plus spécifiques. Mon travail de doctorat a consisté en la synthèse et l'étude RPE de deux séries de nitrones fonctionnalisées par un groupement phényl en position 4. Les deux diastéréoisomères : la 4 PhDEPMPOt et la 4 PhDEPMPOc furent préparés et utilisés pour le piégeage de plusieurs espèces radicalaires. L'adduit de spin formé par réaction de la 4 PhDEPMPOc avec le superoxyde a présenté un signal composé de 8 raies montrant seulement un léger phénomène de largeur de raie alternée. Les temps de demi-vie de la 4PhDEPMPOc-OOH sont similaire ( ~15mm). La deuxième série de molécules, ce sont les nitrones dérivées de la 4HMDEPMPO. Cette molécule fut obtenu en trois étapes et utilisée comme précurseur pour la préparation de nitrones vectorisées. L'adduit de spin formé par réaction de la 4HMDEPMPOc avec le superoxyde a présenté un signal composé de 8 raies montrant seulement un phénomène de largeur de raie alternée résiduel. Le temps de demi-vie de la 4HMDEPMPO-cOOH est plus grand (21mn) que celui de la DEPMPO-OOH. Un premier test de vectorisation par une molécule présentant une activité biologique a été concluant puisque la 4BIODEPMPOc fut synthétisée et l'étude RPE a démontré que l'introduction du motif biotine est compatible avec le piégeage de radicaux libres et a présenté un adduit du superoxyde dont le signal fut facilement attribuable

Abstract:
La premiere partie de ce travail est consacree a la mise au point de la synthese du 2-diethylphosphono-2-methyl-3,4-dihydro-2h-pyrrole 1-oxyde (depmpo), une molecule particulierement interessante pour son application au piegeage de spin (spin trapping) en milieu biologique et en milieu organique. La synthese de cette nitrone -phosphoree est realisee, en deux etapes, par addition de diethylphosphite sur la 2-methylpyrroline (rendement >95%) suivie de l'oxydation de l'aminophosphonate obtenu. Le controle de cette reaction d'oxydation est de premiere importance pour obtenir la depmpo. Parmi les oxydants testes, le dimethyldioxirane et le reactif de davis ont donne les meilleurs resultats (rendement >80%). Les produits secondaires formes dans ces reactions sont identifies. Une bibliographie detaillee sur la formation des nitrones a partir de l'oxydation des amines secondaires complete cette premiere partie. La seconde partie de ce travail concerne l'application de la depmo au piegeage de divers radicaux centres sur le carbone, le soufre et l'oxygene. Chacun des adduits obtenus est identifie et caracterise par rpe. Une analyse de la geometrie des cycles pyrrolidinoxyle est egalement abordee ici

La 5-diéthoxyphosphoryl-5-méthyl-pyrroline-N-oxyde (DEPMPO) est une nitrone très performante pour le piégeage des radicaux libres oxygénés, notamment le radical superoxyde. Elle présente des capacités de piégeage supérieure à celle de la 5,5-diméthyl-pyrroline-N-oxyde (DMPO) (meilleure persistance de l'adduit DEPMPO-OOH, absence de décomposition en adduit DEPMPO-OH), avantages liés à l'introduction d'un groupement diéthoxyphosphoryle P(O) (OEt)2 sur le cycle pyrrolidinique. Le premier objectif de ce travail est de comprendre pourquoi l'introduction du groupe P(O) (OEt)2 contribue la stabilisation de l'adduit DEPMPO-OOH. Dans ce but, nous avons synthétisé la 5-diéthoxyphosphoryl-5-phényl-pyrroline-N-oxyde (DEPPPO), la 5-méthyl-5-trifluorométhyl-N-oxyde (TFMPO), la méthyl(5-méthylpyrrolidin-5-yl-N-oxyde)phosphinate d'éthyle (EMPMPO) et l'oxyde de dipropyl(5-méthylpyrrolidin-5-yl-N-oxyde) phosphine (DPPMPO). L'analyse des spectres RPE et des calculs de mécanique moléculaire ont montré qu'il existe une stabilisation par effet anomère des adduits du radical superoxyde. La seconde partie de ce travail est consacrée à l'étude du contrôle de l'échange conformationnel observé dans le cas des adduits DEPMPO-OOR (R = H, alkyl). Pour cela, nous avons synthétisé deux nitrones dérivées de l'EMPO et de la DEPMPO substituées par un groupe phényle en position 3 : la 5-diéthoxyphosphono-5-méthyl-3-phényl-1-pyrroline N-oxyde (DEPMPPO) et la 5-éthoxycarbonyl-5-méthyl-3-phényl-1-pyrroline N-oxyde (EMPPO). L'introduction d'un groupement phényle en position "cis" par rapport au groupe -P(O) (OEt)2 supprime le phénomène d'alternance de largeur de raie lié à l'échange chimique dans les adduits OOR, simplifiant ainsi l'allure du spectre RPE.

Le spin trapping in vitro des radicaux hydroxyle et superoxyde a ete effectue en solution tampon phosphate 0,1 m et les constantes de couplage hyperfin des adduits de spin obtenus ont ete determinees. Les constantes de vitesse de piegeage des radicaux hydroxyle et superoxyde avec la depmpo sont similaires a celles trouvees avec la 5,5-dimethyl-1-pyrroline n-oxide (dmpo). Cependant nous avons trouve que l'adduit de spin depmpo-superoxyde etait plus persistant (15 fois a ph 7) que l'adduit dmpo-superoxyde. Pour la premiere fois, en utilisant la depmpo, la production de radicaux superoxyde pendant la reperfusion de curs de rats isoles au cours d'une ischemie myocardique provoquee a ete clairement caracterisee

Las necesidades de las personas son muy dinámicas y cambiantes; cada vez requieren de productos y/o servicios más integrales, personalizados y que satisfagan los requerimientos del cliente. Además, las personas tienen una vida cada vez más activa: hoy en día, por lo general, ambos padres de familia trabajan y realizan múltiples actividades, por lo que el tiempo que disponen para realizar y controlar los quehaceres domésticos diarios es reducido. Considerando los niveles de inseguridad y delincuencia que hay en Lima, donde hay robos en todas las zonas y en cualquier momento, la búsqueda de personal de limpieza seguro y confiable para hogares es una tarea bastante exigente; este personal extraño al hogar no solo debe realizar bien su trabajo, sino tener un buen comportamiento durante las horas que pasa en los distintos ambientes de la casa de su empleador. El mercado ofrece distintas opciones como agencias de empleo, que muchas veces no son seguras ni adecuadas ya que no han hecho una evaluación previa de los trabajadores que promueven; además de las referencias o recomendaciones que las persona pueden recibir de su entorno social. Como consecuencia de este análisis se ha identificado que existe la necesidad de un servicio de limpieza para hogares que sea más seguro, dinámico y que facilite a los potenciales clientes la seguridad de contar con personal de servicio seguro y eficiente para que realice estas labores domésticas en horarios flexibles, según las necesidades del cliente. Debido a ello se propone el desarrollo de un negocio en línea, por medio de una plataforma digital (aplicativo móvil), en donde el cliente pueda hacer el pedido y pago de un servicio cómodo, de fácil uso y acceso inmediato en cualquier momento y lugar; lo que permitirá el ahorro de tiempo y tener mayor seguridad de las personas que ingresan al hogar del cliente.

Abstract:
Applications are increasingly hosted on shared, heterogeneous distributed systems. Web applications and “cloud” computing are broad examples of this, but even relatively isolated servers often depend on shared, network-accessible resources. This standardization around service layers abstracts aspects of service delivery, but often entails a large collection of component systems. The complexity that is abstracted from a functional perspective can give rise to runtime issues. Critical issues arising out of this model include resource utilization (sizing, capacity testing, resource monitoring) and dependency management (identifying and monitoring dependencies between components and potential for impact to an application). This thesis presents an automated solution called DepMap for identifying and monitoring file, network and other communication dependencies in applications by analysis of low-level operating system activity. This work is applicable to resource utilization, diagnosis of performance issues, characterization of workloads, and systems management. This thesis discusses DepMap’s requirements, design and implementation, and evaluates the effectiveness and performance on simulated and actual applications. DepMap can be used to create models of system behavior, based both on observations over time and on injection of delays into selected system operations to understand impact to the larger system. When applied to two commercial systems DepMap was able to identify changing dependencies, and to characterize the behavior of network and storage dependencies. On a cluster of systems hosting a web application, DepMap showed unexpected variation in network transmission time between peers in the cluster, and helped to uncover large jumps in system clock times arising from unreliable Network Time Protocol (NTP) services. On a database server that processes hundreds of gigabytes of data each day, DepMap was able to characterize I/O workload to storage connected by Fiber Channel and iSCSI. This work showed that serious performance limitations existed in the storage server due to fragmentation, design assumptions poorly suited to a data warehouse workload, and competition from other storage consumers. This information was used to design and validate a new server/storage platform specifically for this workload. The insight gained by using DepMap in this case has provided dramatically improved performance (10-15x faster throughput for some relevant workloads) and large cost savings relative to other options.

Functionally Graded Piezoceramics (FGP) offer performance similar to conventional piezoceramic actuators while reducing the problems associated with their bonded construction (high stress levels, large stress discontinuities, delamination, etc.). This paper presents the Dual Electro/Piezo Property (DEPP) gradient method and the tools necessary for designing, modeling, and producing DEPP FGP actuators including: material property gradient maps, a Micro-Fabrication by Co-eXtrusion (MFCX) process, and experimentally validated analytic and numeric performance and stress modeling methodologies that account for continuous and layered material gradients and complex electric field profiles. These models predict a dramatic internal stress reduction achieved by the DEPP method. Preliminary reliability testing confirm this with an increase in piezoelectric actuator lifetimes over 1010 cycles, improvement of almost four orders of magnitude compared to conventional piezoceramic actuation.

Abstract Advanced classical Probabilistic Risk Assessment (PRA) effectively combines various methods for quantitative risk evaluation, such as event trees, fault trees, and Bayesian networks. PRA methods and tools provide the means for the qualitative reliability evaluation (e.g., cut sets) and the computation of quantitative reliability metrics (e.g., end states probabilities). Modern safety-critical systems from various industrial domains tend toward a high level of autonomy and demand not only reliability but also resilience, the ability to recover from degraded or failed states. The numerical resilience analysis of such dynamic systems requires more flexible methods. These methods shall enable the analysis of the systems with sophisticated software parts and dynamic feedback loops. A suitable candidate is the Dual-graph Error Propagation Model (DEPM) that can capture nontrivial failure scenarios and dynamic fault-tolerance mechanisms. The DEPM exploits the method for the automatic generation of Markov chain models and the application of probabilistic model checking techniques. Moreover, the DEPM enables the analysis of highly-customizable system resilience metrics, e.g., “the probability of system recovery to a particular state after a specified system failure during a defined time interval.” In this paper, we show how DEPM-based resilience analysis can be integrated with the general PRA methodology for resilience evaluations. The proposed methodology is demonstrated on a safety-critical autonomous UAV system.
