Abstract:
The essay focuses on Irish author Dermot Healy’s involvement with memories of old people within two collaborative projects: the making of a film based on the documentary novel I Could Read the Sky by Timothy O’Grady and Steve Pyke (1997), and the development of a documentary drama with the clients of a day care centre in Co. Monaghan, entitled Men to the Right, Women to the Left (2001). It examines the methods used to record the material and its subsequent creative use, particularly in comparison with the technique of British verbatim theatre, and in the context of the imperfections of individual memory that are deftly explored in Healy’s memoir The Bend for Home (1996). The essay ultimately argues that notwithstanding problems concerning authenticity, Healy’s play, alongside O’Grady and Pyke’s book and Nichola Bruce’s film version of it, should be regarded as vital contributions to the formation of Ireland’s cultural memory, particularly as they powerfully reconstruct “the mundane everyday” that is so often lost.

Abstract:
This thesis provides a Marxist analysis of the effect of class on historical alienation in Dermot Bolger’s fiction. Therefore, this study examines the influence of Irish history on Bolger’s choice of content, form and technique in order to argue that historical interpretation and literary technique are mediated through class stratifications. Chapter One investigates how The Journey Home challenges received ideas of what constitutes ‘reality’ which has, consequently, led to elements of critical dismissal used to maintain antiquated gaps, silences and notions of ‘reality’. In Chapter Two I look at A Second Life in order to examine how historical ruptures cannot just be seen in the nonlinear structure of Bolger’s novels, but can also be used to expose the silences and gaps that comprise the previously censored personal histories of Bolger’s characters. In Chapter Three I identify structural confines such as definitions, family roles and nationalism as instigating factors that lead to the alienation of those who do not conform to prescribed frameworks and are therefore oppressed by them. I further investigate how oppression also provides the pressure to rupture the linear trajectory of such approved frameworks and produce the nonlinear structure that can be recognised in The Family on Paradise Pier.

La littérature irlandaise, de même que la littérature sur l’Irlande, est fortement imprégnée de pastorale. Fort de ce constat, nous avons cherché à évaluer la part que pouvait encore jouer cette thématique chez les plus jeunes représentants de la littérature irlandaise et notamment chez trois de ses romanciers : Roddy Doyle, Dermot Bolger et Patrick McCabe. Au cœur du discours de la pastorale, tradition et modernité s’opposent et se renforcent mutuellement. Or, dans l’écriture de ces romanciers irlandais, elles n’ont de cesse de rappeler au lecteur que leur relation ambiguë forme également l’un des fondements idéologiques majeurs qui ont présidé à l’écriture de l’histoire de l’île. En raison de cet échange permanent entre écriture et pastorale, le jeune roman insulaire reflète ici un débat typiquement irlandais dans lequel nationalisme et révisionnisme, écriture et ré-écriture de l’histoire, dominent et conditionnent tout autre thème. De telle sorte que le fait littéraire y rejoint le fait politique, tandis que le mythe et la réalité s’y confondent parfois dans le propos de leurs personnages romanesques à l’identité souvent déchirée. Prisonniers de ce cadre de production littéraire et idéologique qu’ils contribuent, finalement, à perpétuer, Doyle, Bolger et McCabe affichent cependant leurs différences les uns par rapport aux autres et illustrent, par leurs récits, une géographie culturelle irlandaise complexe où un nationalisme révisionniste peut aisément côtoyer un révisionnisme nationaliste. En partant du principe que l’écriture de l’histoire ou du simple récit suppose d’abord sa ré-écriture, ces romanciers resserrent un peu plus les liens qui unissent texte et contexte dans leur écriture, et dépeignent une Irlande qui échappe, par conséquent, à toute définition préconçue<br>The pastoral pervades irish literature, and so does most literature “about” Ireland. Taking this observation as a starting point our study, we have tried to assess how much this important theme could still influence the new irish writers and in particular three of them : Roddy Doyle, Dermot Bolger AND Patrick McCabe. The opposition between tradition and modernity is at the core of the pastoral ’s reasoning and forms the tension that feeds this reasoning. But in the writing of our three irish writers, tradition and modernity keep reminiding readers that their ambiguous relathinship also gives its ideological motivation to the writing of the island's history. Owing to this permanent dialogue between writing and pastoral, the new generation of irish writers tends to illustrate a typically irish debate in which nationalism and revisionism, the writing and re-writing of history, form key themes. So much so that literature and politics keep interwining in their novels while myth and reality get mixed up in the minds of their characters, thus affecting their own sense of identity. Conditionned by this literary and ideological framework which they themselves contribute to perpetuate, Doyle, Bolger and McCabe tend, however, to differ frome each other in such a way that their writings reflect the complexity of an irish cultural geography where revisionist nationalism and nationalist revisionsim unsurprisingly stand side by side. Considering that the writing of history or that of a simple story first supposes a principle of re-writing, these novelists bring text and context in tight connection in their own writing and depict an ireland that goes far beyond preconceived definitions

Ce travail analyse les rapports complexes qui se tissent entre traduction et culture, et plus particulièrement les problématiques qui émergent de la traduction de références culturelles étrangères. Il prend pour objet d’étude un corpus de romans historiques contemporains irlandais traduits en français. Ce corpus se compose de cinq romans écrits par Sebastian Barry, Dermot Bolger, Roddy Doyle et William Trevor, dont l’intrigue se déroule pendant une même période historique, la première moitié du vingtième siècle. Ce cadre historique partagé garantit la présence d’un socle de références culturelles communes. L’étude à la fois descriptive et contrastive des solutions adoptées pour traduire ces références permet donc d’une part de rendre compte des zones de résistance spécifiques de la culture irlandaise au transfert interculturel, et d’autre part de tenter de dégager certaines tendances, certains systématismes au sein des différentes traductions. En outre, les romans du corpus révisent tous un certain nombre de constructions historiques, identitaires et culturelles, notamment la vision homogène et exclusive d’une irlandité catholique, gaélique et rurale. Cette approche commune constitue une clé de compréhension importante et donc un enjeu non négligeable pour la traduction des références culturelles de ces romans. Cette thèse s’attache donc également à examiner les déformations que subissent ces représentations culturelles spécifiques au cours du processus de traduction. Les recherches effectuées dans les domaines de la traductologie, mais aussi de la littérature et de l’histoire irlandaises viennent appuyer et compléter l’étude<br>This study analyses the complex relationship between translation and culture, and more specifically the issues arising from the translation of cultural references into a different language. It focuses on a body of contemporary Irish historical novels translated into French. This corpus comprises five novels written by Sebastian Barry, Dermot Bolger, Roddy Doyle and William Trevor. All five novels take place during the same historical period, namely the first half of the twentieth century. This shared historical context guarantees the presence of a base of cultural references common to all novels. This study will therefore take on both a descriptive and comparative approach in order to analyse the range of solutions that were implemented to translate these references. It will aim at uncovering the areas of Irish culture that demonstrate a particular resistance to intercultural transfer, as well as foregrounding recurring translational trends within the translated texts. Additionally, the novels under study all revise a number of historical, cultural and identity constructs, in particular the idea of a homogeneous Irishness that is exclusively Gaelic, Catholic and rural. This approach constitutes an essential key to understanding the novels and therefore represents a significant issue and challenge for the translation of cultural references. Accordingly, the study also attempts to examine the modifications undergone by these specific cultural representations during the translation process. It is supported and completed by researches carried out in the fields of translation studies as well as Irish literature and history

Le travail présenté dans le manuscrit concerne la conception d'un dispositif médical de classe III pour des applications en ingénierie tissulaire du derme et du disque intervertébral.Il s'agit d'une solution aqueuse de chitosane, stérilisée par autoclave, et dont le pH et l'osmolarité ont été ajustés selon un procédé de dialyse classique. Cette solution possède des propriétés de gélification in situ innovantes, ce qui lui confère de très bonnes propriétés mécaniques quelques minutes après l'injection, sans l'utilisation d'agents de réticulation mais de façon modulable en fonction de la concentration en polymère. De plus, cette solution possède une bonne injectabilité favorisant le développement d'une technique d'implantation mini-invasive pour la régénération du derme et du disque intervertébral. Un critère de performance rhéologique a notamment été mis au point afin de relier l'injectabilité aux propriétés mécaniques de l'injectât (dermal filler).Les propriétés mécaniques de l'injectât gélifié doivent être comparables au tissu natif en particulier pour le dermal filler mais aussi pour le disque intervertébral. Le comportement mécanique viscoélastique du disque intervertébral a été évalué par des essais de relaxation de contraintes et modélisé avec un modèle de Maxwell solide à trois branches. Ces essais ont été conduits sur des disques sains, ayant subi une altération de structure (fenestration) et après l'injection de biopolymères.La solution de chitosane a été injectée (i) dans le tissu cutané de deux modèles animaux : le porc et le rat et (ii) dans le disque intervertébral de deux modèles animaux : le porc et le lapin. La biocompatibilité ainsi que la réponse biologique de solutions/gels physique de chitosane in vivo ont été validées pour tous ces modèles animaux<br>This work deals with the conception of a class III medical device for applications in tissue engineering of dermis and intervertebral disc.This device consists in an aqueous chitosan solution, sterilized by autoclaving, with pH and osmolality adjusted by a dialysis process. This chitosan solution shows in situ gelation ability with a post-injection increase of mechanical properties. This feature is related to polymer concentration, gelation time, and is performed without external cross-linking agent. In addition, the solution exhibits a good injectability allowing the development of minimally invasive techniques to treat dermis and intervertebral disc diseases. A rheological performance criterion was defined linking injectability to mechanical properties of the implant (dermal filler).Mechanical properties of gel implant formed in contact with body fluids, in situ, have to be similar to that of native tissues. The viscoelastic behavior characterization of intervertebral disc was performed using stress relaxation and was modeled using a generalized solid Maxwell model (composed of three Maxwell elements). The tests were performed on healthy disc, fenestrated discs and after biopolymers injection.Chitosan solutions were injected into (i) porcine and rat cutaneous tissue and (ii) porcine and rabbit intervertebral discs. The biocompatibility and biofunctionality of chitosan solutions and physical hydrogels was evidenced in vivo for all animals

Des cellules dermiques dissociees ont ete cultivees dans un lattis tridimensionnel de collagene bovin. Le derme artificiel ainsi constitue a ete utilise comme substrat pour la culture de fragments d'epiderme embryonnaire de peau dorsale. Dans un premier temps, divers parametres du comportement des cellules dermiques ont ete analyses: leur capacite a retracter le lattis de collagene, a proliferer et a synthetiser des constituants matriciels (fibronectine, collagene de types i, iii et v) ainsi que leur repartition et leur morphologie. Dans un deuxieme temps, de l'epiderme embryonnaire a ete cultive a la surface du derme artificiel. Le comportement de l'epiderme (etaliement, histogenese et differenciation) varie en fonction de la qualite du substrat. L'etalement est modere et l'integrite histologique est maintenue a condition que le substrat contient des cellules dermiques vivantes ou tuees. Seul un derme artificiel comportant des cellules dermiques vivantes permet une stratification et une keratinisation quasi normales ainsi que la reconstitution d'une membrane basale presque continue. En conclusion, les resultats obtenus montrent que les performances histogenetiques de l'epiderme sont influencees par la constitution du substrat sur lequel il est cultive. La technique du derme artificiel embryonnaire, par la simplification qu'elle apporte par rapport a l'utilisation d'un derme veritable et par la possibilite qu'elle offre d'en varier la composition, peut servir a l'analyse de certains aspects des interactions dermo-epidermiques au cours du developpement de la peau et des phaneres

Abstract:
The concept of immunosurveillance suggests that the innate and adaptative immune system eliminate developing tumors. However, tumor development is associated with important modifications of the stroma which, by multiple mechanisms, restrain the immune response notably by affecting dendritic cells (DC) recruitment and functions. My thesis project aims at deciphering how the tumor environment alters the mechanisms and pathways of DC recruitment and impairs their functions. First, we determine if the site of tumor transplantation affects tumor immunogenicity. We show that tumors transplanted in the dermis (i.d.), an environment containing multiple DC subsets, induce a protective anti-tumoral immune response and tumor rejection. By contrast, the same tumor implanted in the subcutaneous tissue (s.c), mainly containing monocytes, is not rejected. Rejection of i.d. tumor is associated with a rapid (within 2 days) recruitment of DC within the tumor and rapid migration of DC towards tumor draining lymph nodes (dLN) where they present the tumor antigens (TAA) to CD4 and CD8 T lymphocyte. These events also occur for s.c. tumors but with a delayed kinetic. Thus the kinetic of DC mobilisation is decisive for tumor immunogenicity. Analysis of the DC subpopulations (TIDC) infiltrating the i.d. or s.c. tumors at 4 days (D4) and 8 days (D8) post-tumor transplantation, revealed that the different DC subpopulations are present at similar frequencies. Based on these findings, we proposed that i.d. tumor are rapidly infiltrated by dermal DC (DDC), whereas in s.c. tumor, the absence of inflammatory signals would limit DDC recruitment. In this latter case, DC would mainly come from local differentiation of blood-born precursors of DC (pre-cDC). Local differentiation of pre-cDC within the immunosuppressor tumor environment may affect their differentiation program and functions. We found that pre-cDC infiltrate i.d. and s.c. tumors starting at D4 and their frequency increases at D8. To determine the DC origin in tumors, we use CD11c-DTR-GFP mice in which CD11c+ cells express a fusion protein constituted by the diphtheria toxin receptor (DTR) and GFP. To track DDC trafficking within tumors, we injected anti-MHCII antibody before tumor implantation and analysed MHCII+ CD11c+ DDC infiltration in tumor by biphotonic microscopy. At D3 post-tumor transplantation, the DDC infiltration was higher in i.d. than s.c. tumors. To analyse the impact of this early DDC recruitment on anti-tumor immunity, we inhibit early recruitment of DC by injection of pertussis toxin (PTX), a chemokine receptor protein G inhibitor, during the three first days of tumor development. For i.d. tumors, PTX treatment induced 60% reduction of DC recruitment starting at D4. In s.c. tumor, while this effect was observable at D3 (60% reduction) and increased to 80% at D4. These results suggest that early recruitment of DDC to i.d. tumors may be be chemokine independent. PTX treatment, which inhibits DDC migration from tumors to dLN inhibits the early TAA presentation to CD4 and CD8 T cells but did not impaired i.d. tumor rejection. Collectively, these results suggest that a first wave of DDC may infiltrate i.d. tumor. The initial wave of DDC may rapidly activate the adaptive immune system and induce protective anti-tumoral immune response. For s.c. tumor, this first wave in delayed or limited. Tumor neoangioenesis would permit an input of pre-cDC which would differentiate locally into cDC1 and cDC2. To consolidate this model we are developing new protocols to efficiently inhibit early recruitment of DDC in i.d. tumor. Moreover, to determine the DC origin and pathways of DC recruitment in tumors, we exploit several experimental approaches to directly analyse DDC migration toward i.d. and s.c. tumor<br>Le concept d’immunosurveillance postule que le système immunitaire inné et adaptatif élimine les tumeurs naissantes. Cependant, le développement de tumeurs est associé à des modifications importantes du stroma qui, par des mécanismes multiples, inhibent la réponse immunitaire en affectant notamment le recrutement et la fonction des cellules dendritiques (DC). Mon projet de thèse vise à préciser comment l’environnement tumoral affecte les mécanismes et les voies de recrutement des DC dans les tumeurs et leurs fonctions. Tout d’abord, nous avons déterminé si la composition en DC du tissu d’implantation affecte l’immunogénicité tumorale. Nous avons montré que les tumeurs implantées dans le derme (i.d.), un environnement riche en DC dermales (DDC), induisent une réponse anti-tumorale protectrice. En revanche, une même tumeur transplantée dans le tissu sous-cutané (s.c.), contenant principalement des monocytes, n’est pas rejetée. Le rejet des tumeurs i.d. est associé à un recrutement précoce et rapide des DC dans la tumeur (dès 2 jours post-injection) et une migration, dans les ganglions drainants (dLN), de DC qui présentent les antigènes tumoraux (TAA) aux lymphocytes T (LT) CD4+ et CD8+. Dans les tumeurs s.c. ces événements sont présents mais retardés. Ceci indique que la cinétique de mobilisation des DC est déterminante pour l’immunogénicité tumorale

Abstract:
L’interaction épithélium/mésenchyme est cruciale pour de nombreux processus physiopathologiques. Lors de ma thèse, je me suis intéressée aux signaux mésenchymateux régulant les cellules épithéliales en utilisant comme modèle la peau, qui est composée de 2 compartiments : l’épiderme (épithélium) et le derme (mésenchyme). Les intégrines sont impliquées ces interactions. CD98hc est une protéine transmembranaire à double fonction qui chaperonne des transporteurs d’acides aminés et régule la signalisation des ß intégrines. Elle est exprimée dans les cellules prolifératives telle les cellules épithéliales. Dans la peau, CD98hc est exprimée l’épiderme mais également dans les fibroblastes, cellules post-mitotiques. Mon hypothèse a été que CD98hc participe aux régulations des interactions derme/épiderme. Grâce à un modèle de KO conditionnel de CD98hc dans les fibroblastes dermiques, j’ai mis en évidence que CD98hc permet le maintient des propriétés mécaniques et biologiques du derme, et, de ce fait, régule l’épiderme en conditions d’homéostasie, de perturbation de la barrière et lors de la formation de cancer. De plus, le rôle de CD98hc dans cette interaction apparait comme étant lié à l’âge. En conclusion, mes travaux de thèse montrent le rôle central de l’expression dermique de CD98hc dans le maintien de l’homéostasie cutanée au cours du vieillissement ainsi que lors de la tumorigenèse<br>The epithelial/mesenchymal interaction is crucial for many physiopathological processes. During my PhD, I focused on mesenchymal signals that regulate epithelial cells behavior using the skin as model. The skin is composed of 2 main compartments: the epidermis (epithelium) and the dermis (mesenchyme). While this crosstalk involves integrins, its regulations are poorly understood. The transmembrane protein CD98hc interacts with amino acid transporter and regulates integrin signaling. CD98hc which is expressed at the cell membrane of proliferative cells, specifically epithelial cells, is required for tissue homeostasis. We found that besides its expression in keratinocytes, CD98hc is also expressed in post-mitotic dermal fibroblast. Hence, I hypothesized that CD98hc is involved in epidermis/dermis crosstalk. Using a conditional KO mouse model that harbor a CD98hc deletion in dermal fibroblast, I have shown that dermal CD98hc is required to maintain mechanical and biochemical properties of the dermis. Moreover, I have shown that those CD98hc-dependent dermal properties are implicated in the regulation of the epidermal cell behavior during homeostasis, cutaneous barrier disruption and tumorigenesis. Moreover, the role of CD98hc in those processes seems to be age-related. To conclude, during my PhD, I have revealed a major role of CD98hc in the maintenance of skin homeostasis during aging and tumorigenesis

Abstract:
Well-known characteristics of aging skin are the development of fine lines and wrinkles, but also changes in skin tone, skin texture, thickness and moisture content [1, 2]. Rejuvenation of the skin aims at reversing the signs of aging and can be established in the epidermis as well as in the dermis. Aged dermis for that matter has a degenerated collagen matrix. To regenerate this matrix, fibroblasts need to be stimulated into synthesizing new collagen. Among the rejuvenation methods used the non-ablative techniques are gaining popularity. These methods produce dermal remodeling without obvious epidermal injury using a thermal approach. The generation of heat would cause collagen injury and contraction followed by collagen synthesis [2, 3]. However, little research on physiological evidence can be found in contemporary literature. In this study, the effects of heat shocks of different temperatures on the expression of procollagen type 1and type 3 of human dermal fibroblasts in ex-vivo skin are investigated.

Abstract:
Dermal tissue ablation and layer-separation with minimal thermal damage to the surrounding tissue via an ultra-short pulsed (USP) laser has been investigated. At first, a parametric study for wet tissue surface line ablation was investigated. The line features were measured by both microscope and scanning electron microscopy. The dermis ablation threshold and the incubation factor were determined through the line ablation results. It is found that a general microscope may give inflated ablation line width. Following the ablation experiment, histological view of ablated wet tissue samples with different combinations of laser parameters has been undertaken to obtain proper laser parameters for tissue separation with minimal thermal damage. The separation of a whole piece of wet dermal tissue into two thin layers was presented. A thin dermal tissue layer pushing to 220 μm thickness was obtained.

Aging of the skin is a universal process. Well-known characteristics are the development of fine lines and wrinkles, but also changes in skin tone, skin texture, thickness and moisture content are aspects of aging [1,2]. The aging process can be divided into intrinsic aging, such as changes in e.g. dermal components like collagen, elastic fibers, glucosaminogycans and fibroblasts, and extrinsic aging, which is caused by external factors e.g. damage by UV-radiation [3]. Rejuvenation of the skin aims at reversing the intrinsic and extrinsic signs of skin aging and can be established in the epidermis as well as in the dermis.

Abstract:
Treatment of vascular skin lesions, such as port wine stain (PWS) birthmarks, uses laser energy to thermally damage the hypervascular regions in the dermis. Delivery of sufficient laser energy to the dermis can be confounded by the more superficial epidermal melanin layer, a broadband optical absorber. Laser pulses directed at skin lesions may result in excessive heating and scarring of the epidermis, resulting also in limiting the energy available to treat the dermal lesion. Cryogen spray cooling is used to selectively cool the epidermis, allowing the use of higher laser fluences which cause thermal damage in the deeper lesion, while preventing hypertrophic scarring of the epidermis. In order to optimize cryogen cooling and laser pulse parameter for treatment, the depth profile of the epidermal melanin layer and vascular lesion must be known. We use a photoacoustic probe to determine this depth profile. A Q-switched Nd:YAG laser emitting 532 nm light pulses of 4 ns duration is used to excite acoustic waves in epidermal melanin and hemoglobin in the vascular lesions via a miniature probe incorporating a piezoelectric detector. We used acoustic propagation time to determine the spatial relationship of the absorbing structures, thereby giving the necessary means to determine cryogen spurt and laser pulse timing. We present a finite difference thermal model along with data from tissue phantoms showing the effectiveness of the photoacoustic method.

Abstract:
Dermoid cyst of ovary is the second most common type of ovarian germ cell tumor which constitutes 30 to 40% among ovarian tumors. It occurs mostly in women of reproductive age group between 20 and 40 years and very rarely in postmenopausal women. Postmenopause has its own set of symptoms and risks. One such risk is the possibility of malignancy of ovarian cyst with an incidence of 0.5 to 2%. We present an unusual and rare case of an 82 year old woman, who presented with complaints of pain abdomen and constipation for one year duration. Colonoscopy revealed diverticulitis. Despite being treated for diverticulitis, her symptoms persisted. CT was done which showed a right ovarian mass. Diagnostic laparoscopy was done and pus seen in the abdominal cavity was collected, bowel was distended, and dermoid cyst of ovary of 12 × 10 cm size which had undergone torsion three and a half times. Detorsion of ovary with right oophorectomy was done. Histopathology confirmed features of dermoid cyst with torsional changes in the wall and focal gangrene with no evidence of malignancy. Dermoid cyst occurs very rarely in postmenopausal women and treatment of choice is oophorectomy. Authors with this case highlight the proper management of ovarian dermoid cyst in symptomatic postmenopausal women.
