Abstract:

Denna uppsats handlar om normbrytande karaktärer i serierna Bron, The bridge och The tunnel och deras kulturella kontext. Genom karaktärsanalys av de kvinnliga huvudkaraktärerna i respektive serie undersöks vad deras normbrytande beteenden och tankesätt kan spegla av dess kulturella kontext. Genom att analysera hur karaktärerna bemöts av bikaraktärer, analyseras bilden av hur accepterande den kulturen som karaktären befinner sig i är. Uppsatsen behandlar ämnen som den ”problematiska detektiven” och normativa beteenden i det syfte att påvisa hur adaptioner påverkas av kulturell kontext.

Abstract:

True crime has established itself as a genre with a huge amount of dedicated viewers worldwide,and has gone from ”trash culture” to big, Emmy award-winning productions. Consumers of thegenre has evolved from passively watching the series on the couch, to actively getting engaged in thecase over internet forums as ”digital detectives”. In combination with the technological developmentand the growth of the media, including the shift in power this has contributed to, we now face newchallenges that society has not yet been able to address. The genres influence on its consumers iswidely debated, as is the influence of today’s media climate on our perception of the world, andstudies that address why we expose ourself to this type of content have been written about anddocumented in numerous ways. However, there is no account of the consumers’ own understandingof the relationship between the media, increased opportunities of communication and the genres'growing popularity. This study aims to investigate how today’s media climate and the digital culturewe live in have contributed to the phenomenon that true crime is, and what factors causethe consumers to evolve from passive to active. The documentary Making a Murderer serves asbasis for the study, it is available in two seasons and depicts the murder of Teresa Halbach.To fulfill the purpose of the study, a qualitative method approach was used. As part of the pilotstudy a focus group was conducted, which was then supplemented with personal interviews. Thedata that emerged in these studies were processed and analyzed thematically with the support of thedocumented theories. From the themes and patterns that were developed, conclusions could bewritten, the sub-questions could be answered and the purpose of the study was fulfilled. I came tothe conclusion that mediatization and convergence culture can be assumed to play a major part inthe emergence of digital detectives, as well as in the development of a relatively new form of spoilerculture in which collective intelligence is used to reform society. Furthermore, I found a concernamong the consumers about how dangerous it can be if media and civilians act as judges insociety, and how the possible lack of source criticism can affect the public’s perception of the world.In order for consumers to evolve from passive to active, and for them to be affected on an emotionallevel, a real and detailed picture of the case needs to be portrayed, with socio-economic factors aswell as an insight into the protagonist’s life.

Abstract:

I dagens samhälle är barnen mer stillasittande, det kommer larmrapporter som varnar för att detta kan leda till hälsoproblem hos många unga. Vi har valt att titta på hur pedagoger arbetar med rörelse och motorisk träning i skolan, och om alla barn får möjlighet till rörelse under hela skoldagen. Vi vill även se om rörelse och motorik kan ha en koppling till inlärning. Undersökningen vi har gjort bygger på intervjuer med sju informanter med olika professioner. Studien visar att rörelse och motorik har en indirekt koppling till inlärning, då detta påverkar barnens koncentration. Resultatet visar att många pedagoger är medvetna om rörelsens betydelse, men att man arbetar med rörelse och motorik i olika utsträckning på de olika skolorna.

Abstract:

Syftet i föreliggande uppsats är att undersöka hur tre kvinnliga detektiver i tre samtida kriminalromaner gestaltas utifrån ett intersektionellt perspektiv och ett genusperspektiv. Vid analysen använde jag mig av Paulina de los Reyes och Diana Mulinaris samt Nina Lykkes intersektionalitetsbegrepp. Jag använde mig även av Maria Nikolajevas schema över hur manligt och kvinnligt gestaltas inom litteraturen. Utifrån analysen kan det konstateras att samtliga av de undersökta kvinnliga detektiverna till viss del eller helt bröt mot de typiska skildringarna i Nikolajevas schema, vilket går i linje med den tidigare forskningen inom området. Det framkommer även att det är fler faktorer än kön som är av betydelse för detektiverna och deras maktpositioner, både inom arbete och privatliv. Vidare problematiserar jag de framkomna resultaten i analysen ur ett genreteoretiskt perspektiv, för att i förlängningen även diskutera och argumentera för kriminallitteraturens plats i skolan.

Abstract:

Abstract Our purpose was to understand how social workers experience the possible complexities that working with economical aid can entail. As an example we chose to focus on long-term economical aid work. We aimed to address the question of which factors could affect their work. In a prior study, done by Dellgran & Höjer (2003) the conclusion was drawn that the combination of economical aid, healthcare and schools makeup a third of practical social work in the terms of the proportion of the amount of social workers, but only 8 percent of the research is done in those areas. Other studies (Stranz, 2007) demonstrate that the research that is done on working with economical aid is deficient. We interviewed six social workers in two smaller municipalities in the southern of Sweden. Our result indicated that the social workers felt pressured by their work-load and the fact that the amount of new cases had increased. They thought that a possible reason for this could be the weak economic climate and changes in the labour market policies. They also felt that alterations and long office turnaround times at other administrative authorities also contributed. The results showed that the majority of the social workers in our study used a motivational approach in their meetings with clients and they also had the best interest in mind for each and every client. We chose to use the theoretical perspectives of Human service organizations by Yeheskel Hasenfeld (1983) and Demokratins väktare (our translation: the guardians of democracy) by Lennart Lundquist (1998) when we analyzed our results. Abstract:

This master's thesis is a study of how librarians are depicted in crime fiction. 12 American and English mysterynovels featuring librarians in a central role were studied and analysed using character theory. Recurring traitswere identified and organised into themes. A number of prominent traits and themes emerged that show librariansas orderly and organised bibliophiles, but with a taste for adventure and excitement. They are keen problem solverswho enjoy a challenge, at work, or in the form of crime detection. These traits show fairly different sides ofthe characters, and hint of librarians having something of a dubble nature. Some of the traits resonate with findings in earlier studies in the field, but the old stereotype was only parti -ally confirmed. In general the image of librarians presented in the mystery novels is a positive one, which inmany respects also rings true against the background of actual librarians and library work. This is a two years master's thesis in Archive, Library and Museum studies.

Abstract:

Environmental storytelling är något som kan verka enkelt men är mycket komplext. Det är något som låter spelaren, likt en detektiv, undersöka ledtrådar och dra slutsatser ifrån dessa. För att förstå detta i grunden krävdes kunskap inom berättande och teori om uppbyggande av narrativ. Detta följdes sedan av kunskap inom mise-en-scéne, environmental storytelling och människors tolkning och deras sammankopplande av de bitar de har framför sig. Det sista var information om hur objekt uppfattas genom inbjudande till handling tillsammans med form. Dessa teorier appliceras sedan på objekt för att skapa ett visuellt berättat narrativ i en spelmiljö. Tillsammans med en kvalitativ intervju som kvantifierades för resultat fick undersökningen resultat. Den visade på att korrekt designade objekt med specifika fysiska tecken var relativt framgångsrika på att förmedla ett specifikt, skrivet narrativ, bildat av fyra separata händelseenheter.

Abstract:

The aim of this paper is to examine the British television series Sherlock with a genre and narration analysis and to examine how the classic detective genre has developed in terms of narration and to connect it with changes in the contemporary society. The main questions are how the material coincide and differentiate with the original genre the classic detective story as well as the postmodern genre the conspiracy thriller. The methods used are genre and narrative analysis in an attempt to position the narrative of the material within the detective story genres traditional narrative. The analysis is based on functionalist theory about the functions of media texts, genre and narrative theory as well as postmodern theory. The premise is the view on texts as symbolic actions, and the hermeneutic task to disclose that symbolism. The findings of the paper is that Sherlock contain a high amount of intertextuality and join both the classic detective and the conspiracy thriller. The evolution of the genre can be accredited to changes in society’s conflicts and ideals, which in the modern era of the classic detective were questions of class society, a new judicial system and the decline of organized religion and in contemporary society are questions of globalization, infinite networks and conspiracies.

Abstract:

Das Ziel der vorliegenden Arbeit bestand in der molekularzytogenetischen Charakterisierung von invasiv duktalen und invasiv lobulären Mammakarzinomen sowie der Identifizierung unterschiedlicher genetischer Alterationsmuster in Tumorsubgruppen der invasiv duktalen Mammakarzinome. Mit Hilfe der komparativen genomischen Hybridisierung (CGH) wurden 30 invasiv duktale und 20 invasiv lobuläre Mammakarzinome auf genetische Veränderungen untersucht. In beiden Tumorarten überwiegen DNA-Deletionen gegenüber DNA-Gewinnen, daher scheint die Inaktivierung tumorsupprimierender Gene einen größeren Einfluß auf das maligne Wachstumsverhalten auszuüben. Invasiv duktale Mammakarzinome zeichnen sich durch eine im Mittel größere Anzahl an Alterationen pro Tumorfall aus als invasiv lobuläre Karzinome (14,9 vs. 8,9). Neben einer größeren Anzahl an DNA-Verlusten ist auch die Anzahl der DNA-Überrepräsentierungen deutlich höher verglichen mit invasiv lobulären Mammakarzinomen (6,2 pro Tumorfall vs. 2,7 pro Tumorfall). DNA-Gewinne auf Chromosom 1q und DNA-Verluste auf den Chromosomen 6q, 11q22- qter und 13q fanden sich in >=40% der Fälle in beiden histologischen Tumortypen. In invasiv duktalen Mammakarzinomen traten DNA-Gewinne mit größerer Frequenz auf den Chromosomen 6p, 8q, 11q13, 16p, 17q, 19p/q und 20q auf. Ebenfalls häufiger waren DNA-Deletionen auf den Chromosomen 2q, 3p, 4p/q, 5q, 7p, 8p, 9q, 10q und 15q zu finden. DNA-Verluste auf den Chromosomen 16q, 17p, 18q und 22q wurden dagegen vermehrt in invasiv lobulären Karzinomen detektiert. Gut (G1) und schlecht (G3) differenzierte invasiv duktale Mammakarzinome zeichnen sich durch ein unterschiedliches genetisches Muster aus. Während gut differenzierte Tumore durch DNA-Gewinne auf 1q, 11q11-13, 16p und 20q gekennzeichnet sind, weisen die schlecht differenzierten Tumore zusätzlich DNA-Deletionen im Bereich der Chromosomen 5q, 18q und 21q21 auf. Außerdem läßt sich ein unterschiedliches genetisches Muster bei Östrogenrezeptor- positiven und Östrogenrezeptor-negativen invasiv duktalen Mammakarzinomen feststellen. Die Östrogenrezeptor-negativen Tumore zeigen eine größere Anzahl an Alterationen, dazu gehören zusätzliche DNA-Überrepräsentierungen auf 1p, 6p und 22q und DNA-Verluste der Chromosomen 5q, 7p, 8p und 12q. Somit läßt sich feststellen, daß sich invasiv duktale und invasiv lobuläre Mammakarzinome durch ein wiederkehrendes Muster chromosomaler Veränderungen charakterisieren lassen.
The aim of the study was to analyze invasive ductal and invasive lobular breast carcinomas regarding to molecular DNA imbalances as well as different DNA imbalances in tumor subgroups of the invasive ductal carcinomas. Comparative genomic hybridization (CGH) was applied to analyze 30 invasive ductal and 20 invasive lobular breast carcinomas to carry out genetic alterations. In both tumor subgroups, DNA losses showed a higher incidence compared to DNA gains, suggesting a higher influence of inactivation of tumor suppressor genes in tumor progression. Invasive ductal carcinomas showed a higher incidence of alterations per case compared to invasive lobular carcinomas (14,9 vs. 8,9). Besides a higher incidence of DNA losses, particularly DNA gains were statistically significant in invasive ductal carcinomas (6,2 per tumorcase vs. 2,7 per tumorcase). DNA gains on chromosome 1q and DNA losses on chromosomes 6q, 11q22-qter and 13q were investigated in >=40% of all tumor cases. DNA gains were observed with a higher frequency in invasive ductal carcinomas on 6p, 8q, 11q13, 16p, 17q, 19p/q and 20q. Additionally, DNA losses showed a higher incidence on 2q, 3p, 4p/q, 5q, 7p, 8p, 9q, 10q and 15q compared to invasive lobular carcinomas. In contrast, DNA losses on 16q, 17p, 18q and 22q reached statistical significance in invasive lubular carcinomas. Well-differentiated (G1) and poorly differentiated (G3) invasive ductal carcinomas reflected different genetic imbalances. Well-differentiated carcinomas are associated with DNA gains on 1q, 11q11-13, 16p and 20q, whereas poorly differentiated tumors showed additional DNA losses on 5q, 18q and 21q21. Furthermore, the investigation indicated that the average number of alterations is correlated also to the estrogen receptor content of the invasive ductal carcinomas. Tumors with estrogen receptor negative content showed a higher incidence of alterations on the following regions, i.e., DNA gains on 1p, 6p and 22q and DNA losses on 5q, 7p, 8p and 12q. Consequently, invasive ductal and invasive lobular carcinomas are characterized by distinct patterns of chromosomal alterations.

Abstract:

Bakgrund: Idag lever människor allt längre och den äldre populationen ökar. En allt större andel av personer som flyttar in på särskilda boenden för äldre är i behov av palliativ vård redan från den dag de flyttar in. Smärta är vanligt förekommande i livets slutskede. Syfte: Att beskriva sjuksköterskors erfarenheter av att bedöma smärta hos äldre personer med sänkt medvetandegrad vid palliativ vård vid livets slutskede. Metod: En kvalitativ intervjustudie genomfördes med elva sjuksköterskor verksamma inom särskilt boende för äldre. En kvalitativ innehållsanalys användes för analysen. Resultat:Den kvalitativa innehållsanalysen ledde fram till att två kategorier utkristalliserades. Komplext att bedöma smärta samt Professionellt förhållningsätt. Sjuksköterskorna såg smärtbedömning som en komplex uppgift och en av de svåraste uppgifterna i yrket. Smärtskattningen kunde underlättas om de hade erfarenhet att luta sig mot och om sjuksköterskan hade kännedom om personen sedan tidigare. Strukturerade bedömningar med bedömningsinstrument användes som komplement till klinisk bedömning av smärta enligt sjuksköterskorna. Slutsats: För att smärtskattningsinstrument ska användas bör de vara användarvänliga, lättillgängliga och passa den målgrupp som avses att bedömas. Ytterligare forskning behövs för att komma fram till hur sjuksköterskors smärtbedömning hos äldre personer med sänkt medvetandegrad vid palliativ vård i livets slutskede kan förbättras och kvalitetssäkras. Background: Today, people are living longer, and the older population is increasing. An increasing proportion of people moving into special housing for the elderly need palliative care from the day they move in. Pain is common in the end of life. Aim: To describe nurses' experiences of assessing pain in older people with lowered consciousness in palliative care at the end of life. Method: A qualitative interview study was conducted with 11 nurses active in nursing homes Results: The qualitative content analysis led to two categories being crystallized. Complex to assess pain and professional approach. The nurses saw pain assessment as a complex task and one of the most difficult tasks in the profession. Pain assessment could be facilitated if the nurses had experience to lean on and if the nurse had knowledge of the older person since before. Abstract:

Syftet med denna läromedelsstudie var att undersöka samt bidra med ökad kunskap om hur arbetet med läsförståelse behandlas i två läromedel i svenska för årskurs 3. De två läromedlen som analyserades var Livet i Bokstavslandet och Nyckeln till skatten. Analysen av data skedde genom en innehållsanalys, med hjälp av två kodningsscheman och en kodningsmanual. Studiens första delfokus var kvantitativt och behandlade lässtrategier i arbetsböcker, där analysen av det insamlade materialet gjordes utifrån åtta på förhand utarbetade kategorier. Dessa kategorier utgick från tidigare forskning om lässtrategier. Studiens andra delfokus var kvalitativt och behandlade lärarens roll i arbetet med läsförståelse i lärarhandledningar, där analysen gjordes genom ett sökande efter bakomliggande teman. Studiens tredje och sista fokus behandlade likheter och skillnader mellan läromedlen. Studien har genomsyrats av det sociokulturella perspektivet, inom vilket lärande sker i interaktion med andra människor. Resultaten av studien visade att eleverna får möta en variation av lässtrategier genom arbete med arbetsböckerna, och att de båda läromedlen dessutom följde ett liknande mönster i hur frekvent lässtrategierna förekom. Resultaten visade fortsättningsvis att lärarhandledningarna skiljde sig åt desto mer gällande graden av social aktivitet. Där lärande i socialt sammanhang förekom i Livet i Bokstavslandet var det i form av gruppuppgifter, vilka påbjöd socialt samspel och elevernas aktiva deltagande. Där lärande i socialt sammanhang förekom i Nyckeln till skatten var det däremot i form av samtal i helklass, där en av riskerna är ett passivt deltagande från elevernas sida. Livet i Bokstavslandet hade dessutom mer utförliga instruktioner för hur läraren skulle arbeta med läsförståelse. Utifrån detta drogs slutsatsen att arbete med Nyckeln till skatten ställer högre krav på lärarens kompetens inom läsförståelse och lässtrategier.

Detta är en kvalitativ och beskrivande studie kring hur elever uppfattar att arbeta med historiska källor. Studien beskriver även hur förutsättningarna för och uppfattningarna om en god historieundervisning i skolan har förändrats och hur dagens förutsättningar ser ut. Metoden med vilket denna studie gör detta är genom att intervjua en fokusgrupp med sex elever och med kvalitativa analyser av 23 elevsvar från ett skriftligt kunskapstest. Dessutom genomfördes en enkät med 40 elever i år 8 och texter och styrdokument analyserade utifrån ett sociokulturellt perspektiv. Internationell och nationell forskning visar att en förutsättning för att eleverna ska nå historisk empati, är att de får arbeta med och ha tillgång till rikligt med historiska källor och att de får möjlighet att öva i perspektivtagande. Studien visar på att eleverna uppfattar samtal och diskussion kring källor som en viktig stödstruktur för ett historiskt lärande. Däremot upplevde de mindre nytta med att läraren föreläser om källorna. Att ha en referensram att placera källan i samt att ha en tydlig frågeställning uppfattade eleverna också som viktigt. Den undersökta elevgruppen hade svårt att visa sin förmåga till källanvändning vid kunskapstestet, men en djupare analys av testets konstruktion visade att testet hade vissa oklarheter. Eleverna uttryckte källanvändning som betydelsefullt i avsikt att utveckla kunskap om det förflutna och för att förstå olika aktörers handlingsval. Även att utveckla källkritiska färdigheter ansågs som viktigt. En metakognitiv analys kring betydelsen av att arbeta med källor kan stärka elevernas förmågor att använda källor.

Sixties was a special period in the history of Czech literature, because during these years popular literature (especially the detective stories) was written as an artistic literature. During the sixties a lot of detective novels with artistic value were written. In the past (especially during the fifties), vacation literature was considered bad. In the first half of the sixties mainly socialist detective stories were written. There were important political opinions of the author. Socialist authors celebrated police work. In the late sixties detective stories under the Anglo-Saxon tradition were written. For authors, it was important to respect the rules of writing crime fiction. The thesis presented here with deals with an analysis and genre definition of the Nordic detective story, which is currently very popular in the Czech Republic. It is namely an interpretation of the works done by two contemporary Nordic authors in question, i.e. Stieg Larsson and Jo Nesbø. With the first of the two authors I will be looking into the complete trilogy The Millenium containing volumes of Män som hatar kvinnor (2005), Flickan som lekte med elden (2006) and Luftslottet som sprängdes (2007). In this thesis I analyze detective Sherlock Holmes method based on the semiotics of Ch. S. Peirce. I would like to show that although Holmes detective about his method claim that it is deductive, in-depth analysis shows that this method is far more complex and involves induction and abduction, and that each of these methods has its own importance in the detective's reasoning. The work is divided into three parts. The first part focuses on the Sherlock Holmes detective in terms of logical methods. The second part is focused on methods of reasoning by Ch. S. (anglicky): In this thesis, two representative works of two post-war schools of Japanese detective fiction are analysed based on the knowledge of modern narratology. Two mentioned schools are so called social school of detective fiction (shakaiha 社会派), which appeared at the beginning of 60ties, and new authentic school of detective fiction (shin honkakuha 新本格派), which appeared at the beginning of 80ties. This thesis focuses on a theoretical understanding of the term "detective fiction", it describes the development of the detective genre in post-war Japan while focusing on the debates on "authentic" and "inauthentic" detective fiction and describes typical features of two previously mentioned schools. The by using a theoretical apparat suitable for analysing works of very schematic detective genre, two works - Points and lines (Ten to sen 点と線, 1958) written by Matsumoto Seichō 松本清張 (1909-1992) and Tokyo Zodiac Murders (Senseijutu satsujin jiken 占星術殺人事件, 1981) written by Shimada Sōji 島田荘司 (1948 - ) - are analysed. Analyses focus on composition schemes of both works and on the example translated from original works, existence or absence of elements typical for both schools are demonstrated while a different usage of these elements is showed.

The abstract Introducing part of the work is focused on development of literary genre noir in Italian context, especially on ca. last twenty years, introduces its authors in the area of Emilia- Romagna and Bologna as a city of crime. In the analytic part, the two of Lucarelli's noir series with commissioner Coliandro and inspector Negro are comparatively analised from following chosen aspects: the character of protagonist, the character of criminal, the type of a crime, investigative methods, settings, and in the end language, expression means and role of the narrator. In conclusion, the specifics of literary genre noir in settings of Bologna are described, including the relation between fiction and reality in Lucarelli's writings and the constants of the author's poetic are outlined on the basis of comparative analysis of the series.

Abstract:

Trotz erheblicher Fortschritte auf dem Gebiet der Strahlentherapie ist es bis heute noch nicht möglich, die Strahlenempfindlichkeit eines Individuums bereits vor Therapiebeginn vorherzusagen. Diese Tatsache führt dazu, dass es einerseits bei einem Teil der Patienten zu starken Nebenwirkungen infolge einer Bestrahlung kommt und andererseits die Therapie oftmals nicht in ausreichendem Maße anspricht. Die Entwicklung eines verlässlichen prädiktiven Tests stellt daher ein wichtiges Ziel der strahlentherapeutischen Forschung dar und stand auch im Zentrum dieser Arbeit. Methodisch kam dabei der Koloniebildungstest sowie die fluoreszenzmikroskopische Detektion und Bildanalyse des Histons gamma-H2AX, einem relativ neuen Marker für DNA-Doppelstrangbrüche, zum Einsatz. Untersucht wurde eine sehr heterogene Gruppe aus 5 Fibroblasten- sowie 5 Tumorzelllinien. Unter den Fibroblastenzelllinien befanden sich 2 normale Hautfibroblasten, 2 Hautfibroblasten von Brustkrebspatientinnen mit überdurchschnittlich starken Hautreaktionen nach der Bestrahlung sowie eine Zelllinie mit bekannter AT-Mutation. An Tumorzelllinien kam ein Adenokarzinom der Brust, ein Malignes Melanom, ein Fibrosarkom und zwei isogene aber unterschiedlich strahlensensible Glioblastomzelllinien, die sich in Hinblick auf ihre Proteinkinasenaktivitäten unterscheiden, zum Einsatz. Durch den Koloniebildungstest konnte eine große Bandbreite der klonogenen Überlebensraten erkannt werden, wobei Zelllinien mit Proteinkinasedefekten die größte Empfindlichkeit gegenüber ionisierender Strahlung aufwiesen. Der Verlauf des Histons gamma-H2AX in Hinblick auf die Induktion, die Abbaukinetiken, die verbliebenen Reste nach 18 Stunden Reparaturdauer sowie die dosisabhängigen Kurvensteigungen zeigten jeweils einen charakteristischen Verlauf für jede untersuchte Zelllinie. Interessanterweise war die Hintergrundfluoreszenz bei Tumorzelllinien signifikant höher als diejenige bei Fibroblastenzelllinien. Die strahlensensible Glioblastomzelllinie mit Proteinkinasedefekten zeigte eine deutlich protrahierte Phosphorylierung des Histons H2AX. Zwischen den Überlebensraten der Koloniebildungstests und den Ergebnissen der gamma-H2AX-Detektion wurden keine Korrelationen gefunden. Wie in dieser Arbeit gezeigt werden konnte, stellt der Verlauf des Histons gamma-H2AX einen stark zelllinienabhängigen Parameter dar. Das Histon gamma-H2AX besitzt dadurch ein hohes Potential um individuelle Mechanismen einer Zelllinie nach Einwirkung äußerer Noxen, wie beispielsweise ionisierende Strahlung, zu untersuchen. Es bietet interessante Ansatzpunkte zur Beurteilung neuer Therapieregimes als auch zur Entwicklung und Bewertung strahlenmodulierender Chemotherapeutika
Despite of the efforts of modern radiotherapy, about 5-10% of tumor patients develop severe side effects of normal tissue after radiotherapy treatment. Therefore, there is a growing interest to establish a reliable method to predict a normal tissue’s radiosensitivity. On the other hand, some tumors did not respond adequately to the standard irradiation protocols. The development of a predictive test for radiotherapy is therefore one of the important goals of radiation research. The present study used two different methods to evaluate cellular reactions after irradiation – the colony forming test and digital image analysis of histone gamma-H2AX, a marker of DNA double strand breaks. Ten different cell lines derived from normal and malignant tissues were examined. Among them were 2 normal skin fibroblast lines, 2 fibroblast cell lines derived from the skin biopsies of tumor patients with adverse early skin-reactions to radiotherapy, and one fibroblast cell line with a known mutation of the AT gene. The five examined tumor cell lines included a fibrosarcoma (HT 1080), a breast carcinoma (MCF7), a melanoma (Colo-800) and two isogenic glioblastoma (MO59J and MO59K) cell lines. The results of the colony forming test for the 10 cell lines studied showed a wide range of the SF2-values (surviving fraction at 2 Gray). Cell lines with defects in the protein kinases, MO59J and AT, showed the lowest surviving fractions, 0.06 and 0.17, respectively. Interestingly, the background level of gamma-H2AX was significantly higher in malignant cell lines compared with non-malignant ones. We found that the glioblastoma cell line MO59J which is deficient in the protein kinases DNA-PK and ATM showed a delayed phosphorylation of H2AX. Comparison between the parameters of the colony-forming test and of histone gamma-H2AX revealed no correlation between the SF2-values and the induction and disappearance of histone gamma-H2AX for the cell sample tested. However, the induction, the kinetics of disappearance, residual and background parameters of histone gamma-H2AX showed a strong cell line specific behaviour. Our results suggest that histone gamma-H2AX seems to be a very useful cell-type-specific marker for DNA double-strand breaks which could be used as a patient specific molecular marker to assess the effect of radiosensitizers or different radiotherapy schedules in order to optimize the tumor treatment

Abstract:

This master's thesis will explore the ways in which the fictional and real world blends with detective stories readers. In the methodological part, the author will focus on theoretical problems associated with perception of cult literary works, readers' activity and further expansion of fictional worlds. The phenomena the author will explore are, for example, fan clubs, fan fiction, fan art, naming real entities according to fictional characters and places, and searching for fictional entities in the real world. The practical part of this thesis will be devoted to three detective stories authors - A. C. Doyle, A. Christie and M. Jennings, and their functioning as cultural phenomena both in the Czech and world context.

Abstract:

The detective genre originated in the first half of the 19th century and it does not seem to be loosing its pace. On the contrary, new authors have been occurring, bringing new rules and attitudes to the classical formo Detective novel is usually c10sely connected to the settings it describes. The aim ofthis thesis is to find out specific features ofthe Sicilian detective novel. We have chosen two authors and their works for a detailed analysis: Leonardo Sciascia and his novels II giorno della civefta and A ciascuno il suo and Andrea Cami11eri and his La forma dell' acqua and II cane di terracofta. The thesis has two parts. Part one has three chapters: in first chapter we deal with the history of detective novel, the European detective style and the Anglo - Saxon style, or the classical and American detective stories. ln chapter two, we focus on the isle of Sicily, its history and the unique character of its inhabitants and their culture. ln the final chapter, we pay attention to the works of the most famous Sicilian writer Luigi Pirandello e. g.Ilfu Maftia Pascal or Cosi e (se vi pare) , his search for reality, human identity and the relativity ofthe truth, all ofwhich occur in Sicilian detective stories.

Abstract:

Synopsis The aim of the thesis is to analyse the category of the narrator in a detective story. This category can be considered firstly as an independent textual element and secondly in its connection to the plotline. In the structure of detective stories, apart from fabula and sujet, we can discern two mutually influential plotlines. One of them is the story of the crime (past), the other is the story of the reconstruction (investigation of the crime). The way they approach each other on different levels (the setting, characters) helps us to understand the structure of the whole story. The reader can actualize the narrative also thanks to the mediator, the mediator between the text and the reader being the narrator. In the detective story Distant Star the narrator is unreliable, which expresses itself in different ways. The unreliability is supported by the fact that it is a subjectivized narration of a character who can be identified as the narrator. 'The narrator - character' does not have a single focalization in this story. He comments (e.g. by use of brackets) upon his previous stands. The point of view of the narration changes along with the character. 'The narrator - character' can at the same time be considered as the author's alter ego. We may encounter the author himself thematized in the text...

Abstract:

The aim of this work is to describe the development of literary works of well-known Spanish writer Antonio Muñoz Molina on the basis of an analysis of his selected novels, which were published in the period up to the year 2000. They are Beatus Ille, El invierno en Lisboa, Beltenebros, El jinete polaco, Ardor guerrero and Plenilunio. In the introductory part of this thesis we briefly summarize the development of the Spanish novel, which occurred between the sixties and the eighties of the 20th century. This theoretical part will be followed up with the analysis of the author's works. Selected novels will be divided into three groups, which also correspond to the three main creative periods through which Muñoz Molina's work has passed. Within each chapter, we first describe the specifics of the set creative periods and then analyze all the novels we have included in that chapter. When distributing the novels to individual units, the chronological order in which they were published will be preserved in order to obtain the most accurate picture of the development that the author's creation has undergone. In the end, we summarize the defined creative periods and set the trends that Antonio Muñoz Molina is constantly returning in his novels.
