Abstract:
Diazoxide (Diaz), an activator of mitochondrial ATP-sensitive K+ (mitoKATP) channels, is neuroprotective, but the mechanism of action is unclear. We tested whether Diaz preserves endothelium-dependent (hypercapnia) or -independent [iloprost (Ilo)] cerebrovascular dilator responses after ischemia-reperfusion (I/R) in newborn pigs and whether the effect of Diaz is sensitive to 5-hydroxydecanoate (5-HD), an inhibitor of mitoKATP channels. Anesthetized, ventilated piglets ( n = 48) were equipped with closed cranial windows. Changes in diameter of pial arterioles were determined with intravital microscopy in response to graded hypercapnia (5–10% CO2-21% O2-balance N2, n = 25) or Ilo (0.1–1 μg/ml, n = 18) before and 1 h after 10 min of global I/R. Experimental groups were pretreated with vehicle, NS-398 (a selective cyclooxygenase-2 inhibitor, 1 mg/kg), Diaz (3 mg/kg), or 5-HD (20 mg/kg) + Diaz. Potential direct effects of Diaz and 5-HD on hypercapnic vasodilation were also tested in the absence of I/R ( n = 5). To confirm the direct effect of Diaz on mitochondria, mitochondrial membrane potential in cultured piglet cerebrovascular endothelial cells was monitored using Mito Tracker Red. Hypercapnia resulted in dose-dependent pial arteriolar vasodilation, which was attenuated by ∼70% after I/R in vehicle- and NS-398-treated animals. Diaz and 5-HD did not affect the CO2 response. Diaz significantly preserved the postischemic vasodilation response to hypercapnia, but not to Ilo. Diaz depolarized mitochondria in cultured piglet cerebrovascular endothelial cells, and 5-HD completely abolished the protective effect of Diaz, both findings indicate a role for mitoKATP channels. In summary, preservation of arteriolar dilator responsiveness by Diaz may contribute to neuroprotection.

Abstract:
Synopsis The family matriarch dies without a written succession plan, leaving her children to determine how to cope with the continuity of the family’s expanding food empire. This becomes increasingly difficult when one of the siblings wants to incur expensive, yet required, renovations to the family’s original restaurant. The situation is further complicated by the fact that the two older siblings are focused on corporate expansion efforts, while the youngest is trying to demonstrate her competence in running the family’s historical restaurant. A central focus of the case is to understand and identify effective strategies that should guide the firm-related choices each sibling makes. Research methodology This case, which was developed from field interviews and personal experience, highlights the array of competing financial and personal objectives and tensions involved in a family business. An interactive tool allows users to conduct multiple scenario analyses to determine if the company’s manufacturing expansion goals can be achieved while simultaneously honoring the family’s restaurant roots. Relevant courses and levels This case was designed specifically for the undergraduate junior or senior business or economics student who has already taken basic finance, economics, strategy, entrepreneurship, or psychology courses. Typically, by the third or fourth year of study in a traditional undergraduate program, virtually all of the core themes, concepts, theses, and theories associated with the case have been addressed in previous business or economics coursework. Theoretical bases The case provides an intentional opportunity for students to demonstrate their emerging financial analysis competencies, while concurrently synthesizing the so-called “soft” skills associated with rational decision making, organizational behavior analysis, business strategy, entrepreneurship, and negotiations.

Abstract:
The presidency of Porfirio Diaz is often remembered as a period of social degradation and oppression of freedoms in Mexico. His extensive thirty-five year virtual dictatorship brought about a great deal of hardship for the lower classes and an inequitable system of wealth distribution. This image of Porfirio Diaz as a ruthless despot has become an almost impenetrable one in the post-revolutionary Mexican tradition. However, what is often overlooked or unaddressed are the immense economic and social benefits that were still brought about under his rule. Through massive infrastructural improvement programs, superb financial management, and an unrelenting desire to see the implementation of law and order, Porfirio Diaz brought much prosperity and stability to Mexico when it needed it most. Stability, an expression rarely uttered in discussions of Mexican history, was implemented at all costs under Diaz—no small feat considering the near century of political and judicial disorder that preceded his rule. This essay in no way intends to justify the oppressive policies of Diaz, nor excuse the suffering of the lower classes he brought about. Rather, it intends to provide a more balanced view of his rule; bringing to light the more beneficial economic and social policies he implemented that would bring Mexico into the modern, twentieth century world.

Abstract:
AbstractSilver(I) complexes of thioureas and thiocyanate, [(Tu)AgSCN], [(Metu)AgSCN], [(Dmtu)AgSCN], [(Tmtu)(AgSCN)1.5], [(Imt)AgSCN], and [(Diaz)AgSCN] (where Tu = thiourea, Metu = N-methylthiourea, Dmtu = N,N′-dimethylthiourea, Tmtu = N,N,N′,N′-tetramethylthiourea, Imt = 1,3-imidazolidine-2-thione, and Diaz = 1,3-diazinane-2-thione), have been prepared and characterized by elemental analysis, IR and NMR spectroscopy, and thermal analysis. The crystal structure of one of them, [(Diaz)Ag(SCN)] (1), was determined by X-ray crystallography. The crystal structure of 1 shows that the complex exists in the form of a chain-like polymer comprising [Ag(μ2-Diaz)(μ2-SCN)] units. The silver atoms are bridged by μ2-thione sulfur atoms of Diaz and μ2-thiocyanate sulfur atoms. Thereby each silver atom adopts a distorted tetrahedral coordination environment comprising four sulfur atoms, two from thione and two from thiocyanate ligands. An upfield shift in the >C=S resonance of thiones in 13C NMR and a downfield shift in the N–H resonance in 1H NMR are consistent with the sulfur coordination to silver(I). The appearance of a band around 2100 cm–1 in the IR and a resonance around 125 ppm in the 13C NMR spectrum indicates the binding of thiocyanate to silver(I).

Abstract:
Abstract Sulfonylureas increase the incidence of severe hypoglycemia in people with type 2 diabetes and might increase the risk of sudden cardiac death. Sulfonylureas stimulate insulin secretion by closing pancreatic ATP-sensitive potassium ion (KATP) channels. To investigate the role of KATP channel modulators on cardiac arrhythmias and mortality in the setting of severe hypoglycemia, adult Sprague-Dawley rats underwent hyperinsulinemic (0.2 U/kg/min) severe hypoglycemic (10 to 15 mg/dL) clamps with continuous electrocardiography. The rats were randomized for treatment with intravenous vehicle (VEH), the sulfonylurea glibenclamide (GLIB; KATP channel blocker; 5 mg/kg/h), or diazoxide (DIAZ; KATP channel opener; 5 mg/kg/h). The results demonstrated that GLIB completely prevented first-degree heart block compared with VEH (0.18 ± 0.09/min) and DIAZ (0.2 ± 0.05/min). Second-degree heart block was significantly reduced with GLIB (0.12 ± 0.1/min) compared with VEH (0.6 ± 0.2/min) and DIAZ (6.9 ± 3/min). The incidence of third-degree heart block was completely prevented by GLIB compared with VEH (67%) and DIAZ (87.5%). Hypoglycemia-induced mortality was completely prevented by GLIB compared with VEH (60%) and DIAZ (82%). In conclusion, although GLIB increases the risk of hypoglycemia by increasing insulin secretion, these results have demonstrated a paradoxical protective role of GLIB against severe hypoglycemia-induced fatal cardiac arrhythmias.

Abstract:
Diaz-Guerrero hypothesized in 1967 that Anglo-Americans differed from Mexicans along a dimension of active versus passive coping styles. Specifically, he thought Anglo-Americans preferred to engage in other-modifications (active coping) whereas Mexicans preferred to engage in self-modifications (passive coping) in response to stressful events. In a test of Diaz-Guerrero's proposal the Coping Orientations to Problems Experienced was given to 108 Anglo- and Hispanic-American female inmates; no differences between the groups were found on measures of active and passive coping. The Hispanic group did not differ on active or passive coping as a function of measured acculturation; however, for both groups, education and active coping mildly correlated, suggesting that socioeconomic status may be more related to active coping than culture.
