Abstract:
Este trabalho estuda o processo de trefilação combinada do aço SAE 1045, que consiste nas etapas de pré-endireitamento com múltiplos rolos, jateamento, trefilação, corte e endireitamento por rolos, tendo como produto final barras acabadas trefiladas. Este produto final é empregado na fabricação de hastes de amortecedores para indústria automotiva. O Objetivo do estudo é de entender como são geradas e como se comportam as principais heterogeneidades mecânicas do material processado e a influência dos parâmetros do processo e das características do material na formação e modificação dessas heterogeneidades, buscando como resultado parâmetros otimizados para o processo. Foram variados no processo três parâmetros e avaliadas a interação existente entre eles. Os parâmetros modificados foram ângulo da ferramenta de trefilação (fieira), o ângulo de endireitamento e polimento por rolos cruzados (PERC) e a colocação de revestimento de carbo-nitreto de titânio (TiCN) no núcleo da ferramenta de trefilação (fieira). Foi realizada caracterização do aço, como microestrutura, composição química e microdureza. As tensões residuais superficiais das amostras foram caracterizadas utilizando o método de difração de raios-X. Após avaliação das tensões residuais superficiais, foi analisada a variação das tensões com a profundidade através do método do furo cego (hole-drilling), A classificação das inclusões foi de acordo com o método microscópico A (“piores campos”) da norma ASTM E45-97, feita através da maior severidade encontrada analisando o material aleatoriamente através de varredura com microscópio em sua superfície. Os resultados obtidos permitiram conhecer a combinação de ângulo de fieira e de endireitamento por rolos cruzados (PERC) mais favoráveis para gerar níveis baixos de tensões residuais. Além do ângulo, foi possível verificar que a geometria das zonas da ferramenta de trefilação (fieira) influência os níveis de tensões residuais após a trefilação. O núcleo da fieira com revestimento em todos os ensaios realizados apresentou menores níveis de tensões residuais, mostrando ser uma condição importante para a otimização do processo principalmente para a redução das tensões trativas após a trefilação.
This paper studies the process of cold drawing combined SAE 1045 steel, which consists of the steps with multiple pre-straightening rolls, sandblasting, drawing, cutting and straightening by rollers, with the final product finished extruded bars. This final product is used in the manufacture of rods of shock absorbers for the automotive industry. The objective of the study is to understand how they are generated and how they behave the main mechanical heterogeneities of the processed material and the influence of process parameters and material characteristics of the formation and modification of these heterogeneities, seeking as a result the optimized parameters for the process. We varied three parameters in the process and evaluated the interaction between them. The parameters were modified angle of wire drawing tool (spinneret), the angle of straightening and polishing rolls (PERC) and the placement of coating titanium carbo-nitride (TiCN) in the core of the wire drawing tool (spinneret). We performed characterization of steel, such as microstructure, chemical composition and microhardness. The surface residual stresses of the samples were characterized using the method of X-ray diffraction. After evaluation of surface residual stresses, we analyzed the variation of stress with depth by the method of blind hole (hole-drilling), Classification of inclusions was according to the microscopic method ("worst fields") of ASTM E45 -97, made more severe by analyzing the material found randomly through scanning with microscope on its surface. The results obtained allow us to know the combination of angle stringer and straightening by rollers (PERC) more favorable to generate low levels of residual stresses. In addition to the angle, we found that the geometry of the areas of wire drawing tool (spinneret) influence the levels of residual stress after wire drawing. The core of the spinneret clad in all tests had lower levels of residual stress, proving to be an important condition for the optimization of the process mainly to the reduction of tensile stress after wire drawing.

År 1054 skedde en splittring mellan kristendomens två största inriktningar, den ortodoxa och den katolska kyrkan. Detta skedde efter århundraden av oenigheter, missförstånd, maktkamper med mera. Förutom maktkamperna berör konflikterna Kristi natur och Filioque tillägget. Uppsatsen berör även relationerna mellan den ortodoxa och den katolska kyrkan efter splittringen. Undersökningen visar att det finns skillnader i hur olika författare återger de historiska händelserna. Detta förstärker bilden av två kyrkor som inte är eniga om vems fel splittringen var och vilka händelser som var mest avgörande.

Abstract:
Psalm 104 paints a picture of Yahweh dressed in the royal robes of both Aten and Baal. The variety of references to Ancient Near Eastern gods points to a rich and complicated history of the origins of Psalm 104, Israel and Israelite theology. The origins of Psalm 104 in all likelyhood include oral traditions, cultic contexts and years of redactional rearrangement and rewriting. Akhenaten used a centralised cult and monolatrism to achieve political stability in Egypt. Years later Hammurabi used Marduk as chief deity in Babylonia to centralise power and create political and religious stability in his kingdom. It is within this Ancient Near Eastern tradition of using religion and one specific deity to achieve political stability that Psalm 104 finds its origins and cultic context. The early Israelite monarchy finds itself amidst uncertain political circumstances. The early kings used a centralised monarchy and monolatrism to achieve economic and political stability. The early Israelite government also used other Egyptian influences including governing methods, policies and the use of Egyptian scribes. The monarchy used cultic personal to communicate politically sanctioned theology and contact between cultic personal and Egyptian scribes intertwined with a Canaanite heritage has a cosmopolitan and culturally diverse Psalm 104 at its end. The psalm is used as a war liturgy, with Chaoskampf-motifs to legitimise the Israelite king s military action and kingship. This is done by ritual re-enactment on cultic level connecting Yahweh s mythic action to that of the king s present conflict.

Abstract:
This thesis attempts to examine and define the degree of influence which the Turks exerted on Islamic art of the Seljuqid period (1040--1194 AD) specifically, and on Islamic art of the medieval period generally. As this thesis represents a first investigation of the topic, it was necessary to retrace Turkish history from its beginnings to fully understand its dynamic, but also to analyze the art historical and cultural past of the Turkish peoples in order to assess the degree of probability of Turkish influence on Islamic art as well as the means of its penetration. The vaster arena of this research is the field of Central Asian history and the growing awareness of the important cultural ramifications of its widespread Indo-Buddhist culture.
Due to the complexity of the thesis topic, a simple method has been followed to present the material. The thesis is divided into three chapters, each addressing a major issue. The first chapter introduces the four major Turkish steppe dynasties and their art in so far as archaeology permits. The second chapter deals with the process of Islamicization of the Turks, while the third chapter broaches the issue of Turkish influence on Islamic art of the Seljuqid period under four headings: architecture, architectural decoration, animal imagery, and figurative iconography. The basic premise of this paper is the assumption that, if the Turks played such a major role in the political developments of medieval dar al-islam, they must have also contributed, consciously or not, to the formation of medieval Islamic art.

The German Committee for Reinforced Concrete's guideline on concrete with recycled aggregates has regulated the use of recycled concrete for many years. With increasing discussion on the topics of sustainability and resource efficiency, the demand for recycled aggregates is growing. This development has been taken into account with the adoption and continuation of the regulations of the DAfStb guideline in the revised DIN 1045. This topic also plays a role in other areas such as cement production. In future, the fine components of recycled aggregates can also be used as a further main component of cement. In addition to the sometimes restrictive specifications of the regulations, innovative ideas can of course always be realised via approvals, for example concrete components that contain exclusively recycled aggregates.

Abstract Thermal spraying technologies take part in the current worldwide scene as serviceable and profitable technologies for a wide range of intents. The growing adoption of those techniques has driven companies to progressive investments in efforts to reach innovative designs and market needs. Wear resistance is among the desired features which certain components must be able to provide while performing. In this context tribology has an outstanding importance, where the adoption of suitable materials and operational parameters jointly contribute for a longer lifetime of parts. This contribution presents a comparison of AISI 1045 (DIN C45) steel surfaces coated by means of arc spray process, employing cored wires as feedstock. Corresponding light microscopy, SEM-EDX, 3Dprofilometry, hardness and ball-on-disk analyses were conducted to understand the tribological behaviour of two unlubricated iron-based layers with and without carbides reinforcement. The results point out equivalent thickness, porosity and as-sprayed roughness for both coatings. Higher hardness and lower friction coefficients for the carbide free coating as well as a downward trend considering the coefficients of friction for coated and uncoated surfaces under heavier loads.

Abstract:
Fine blanking is a near net shape process that is used for manufacturing of final product. In the fine blanking process, V-ring indentation is applied to create hydrostatic pressure and prevent premature fracture in an undesired direction. Furthermore, a small clearance between the punch and die is employed along with a counterforce punch that causes concentrated strain in the sheared band region. This process generally applies for sheet metal up to 6 mm. Application of this process for blanking of sheets thicker than 6 mm require some precautions for sound production. In the present research work, finite element analyses have been performed to investigate the effects of thickness variations on the shear band localization and shear surface quality in the fine blanking process. Simulations have been performed for two thicknesses of 6 and 8 mm. As blank materials, steel sheets of AISI 1045 have been used for numerical investigations.

Abstract In this paper, the impact of die manufacturing errors on the drawbead restraining force is studied for straight and curved drawbead systems. The effect of manufacturing errors, including machining and assembly errors, is studied through a finite element analysis (FEA) model. The modeling procedure is validated for a single case by experiments, and compared to the existing drawbead data in the literature. The results indicate that for the curved drawbead system, the restraining force increases with curvature. The restraining force of the curved system is close to the sum of the contributions from an identical straight drawbead system and the metal flow-in deformation of the curved flange. Also, the sensitivity of the restraining force for the curved drawbeads to the machining errors, is similar to that of the straight drawbeads.

Face milling tests on AISI 1045 steel were performed to study the flank wear of multilayered coated carbide tools. The cutting tools were dual (TiN/TiAlN) and triple (TiN/Al2O3/TiCN) layered, coated carbide inserts processed by PVD and CVD respectively. As expected, the depth of cut (DOC) did not play an important role on the development of flank wear while the cutting speed had a significant role in the development of flank wear. Using confocal laser scanning microscopy (CLSM) and wavelet transform, the flank wear evolution was analyzed and abrasive wear was found to be a dominant tool wear mechanism. Adhesion of the work material was also observed after the carbide substrate was exposed. Edge chipping and micro-fracture were additional tool failure modes. After comparing the performance of the two inserts, we concluded that the dual layer coating was superior to the triple layer coating under various cutting conditions mainly due to the benefit coming from the coating processes themselves. It was claimed that the superior performance of the multilayer coating came from preventing the gross crack-induced removal of coating materials by propagating the fracture along the coating interfaces. However, no such observations were found in our milling experiment. Therefore, the hardness of the coating materials is the most important criteria for the development of flank wear.

The use of gas mixtures is widely spread in today’s industry. The fields of application go from cooling within refrigerators to the deep freezing of meat balls. One particular application is within the industry of gas heat treating of metal pieces. Such heat treatment involves the cooling of metal pieces, called quenching. The present work investigates the dependence of the performance of a given gas mixture on the geometry of the metal pieces to quench. The heat transfer coefficient and the convection Nusselt number across a short cylinder (D = 150 mm, L = 300 mm) in axial flow for mixtures of hydrogen and nitrogen were computed for six Reynolds numbers in the range 2.0 104 to 2.0 105. The Nusselt number decreased continually with increasing hydrogen content. Since the existing correlations did not adequately represent the calculated data, modified correlations were developed to describe the influence of transport properties and flow parameters on the convective heat transfer over the front and lateral surfaces of the cylinder. These correlations are valid for the range of Reynolds numbers 2.0 104 to 2.0 105 and in the range of Prandtl numbers 0.4 to 0.73. A relation giving the heat transfer uniformity as a function of the Reynolds number and Prandtl number of the gas mixture flowing around the cylinder in axial flow has been developed. This correlation is valid within the same range of Reynolds and Prandtl numbers previously given.

Introdução: Historicamente as plantas medicinais foram utilizadas com diversas aplicações ressaltando-se o potencial alimentício, medicinal e tóxico. Na contemporaneidade com a pandemia da COVID-19 o aspecto medicinal de muitas espécies tem sido aplicado. Dentre os sintomas ocasionados pela doença é pertinente enfatizar os prejuízos ao sistema nervoso. Objetivo: O presente estudo tem o intuito de evidenciar o efeito benéfico da utilização de plantas medicinais frente aos sintomas de enfermidades do sistema nervoso através de dados científicos. Método: Trata-se de uma revisão da literatura com caráter descritivo; utilizando as plataformas SciELO, BVS e PubMed e como descritores: plantas medicinais, terapias complementares, doenças do sistema nervoso e COVID-19 nos idiomas português e inglês. A seleção das obras ocorreu após leitura do título e resumo, com data de publicação entre 2016-2021. Resultados: O processo de infecção ocasionado pelo SARS-CoV-2 pode gerar nos indivíduos sintomas que acometem o sistema nervoso como: transtornos de ansiedade e insônia, prejudicando a recuperação do paciente. Estudos descrevem que plantas medicinais que atuam sobre esse sistema a exemplo da Matricaria chamomilla L. (Camomila), do Cymbopogon citratus D.C. (Capim cidreira), da Passiflora incarnata L. (Maracujá) e da Melissa officinalis L. (Melissa) apresentam resultados satisfatórios. Ressalta-se que essas espécies poder ser preparadas utilizando a infusão e tendo como indicação terapêutica a ingesta de 150mL três vezes ao dia para alcance do efeito calmante nos transtornos de ansiedade e distúrbios do sono. É válido ressaltar que elas são constituídas de metabólitos secundários responsáveis pela atividade terapêutica e que podem ocasionar efeitos colaterais ao indivíduo, devendo o seu uso ser feito sob indicação e monitoramento de um profissional qualificado. Conclusão: O chá dessas ervas atua sobre o sistema nervoso auxiliando no enfrentamento dos sintomas psíquicos ocasionados pela COVID-19, tornando o seu consumo uma ferramenta complementar no auxílio de enfrentamento a essa doença.

IBTS trålexpedition i Västerhavet genomförs två gånger årligen, i kvartal 1 och 3, och täcker Skagerrak, Kattegatt och delar av östra Nordsjön. 2024 års expedition under kvartal 1 drabbades av flera stormar och vilket gjorde det generellt svårt att planera provtagningen. Dagtid används bottentrålen GOV. Under expeditionen genomfördes totalt 45 godkända tråldrag med GOV-trålen, 4 i Nordsjön, 22 i Skagerrak och 19 i Kattegatt. Den totala fångsten uppgick till knappt 10,5 ton (ca 53 % utgjordes av sillfiskar) och inkluderade 65 fiskarter. Den biologiska provtagningen, som också innefattar insamling av otoliter för åldersbestämning, gjordes på de kommersiellt betydelsefulla arterna. Totalt togs 4692 otoliter från 10 olika arter. Preliminära abundansindex för 1-gruppen hos målarterna redovisas i rapporten. Larvtrålningen nattetid resulterade i endast 33 godkända tråldrag pga vädret och det fångades bl.a. 323 sillarver, 52 sardinlarver och 6 glasålar men inga skarpsillslarver.

In 2012, Katmai National Park & Preserve joined the stranded seabird survey efforts spearheaded by the University of Washington?s Coastal Observation and Seabird Survey Team (COASST). COASST is a citizen science network started in 1999 with the goal of establishing a baseline dataset of seabird mortality to identify unusual die-offs. During the winter of 2015 and spring of 2016, the Gulf of Alaska experienced a large die-off of common murres (Uria aalge; COMU) due to a severe marine heatwave in the North Pacific. Katmai National Park & Preserve (KATM) staff contributed to the identification and documentation of seabird carcasses washed ashore from this die-off; in 2016 alone, 90 murres (likely COMU; only counting new birds) were documented along a 1 km stretch of Swikshak beach. KATM staff have conducted a total of 105 COASST surveys documenting 203 seabird carcasses on four 1 km section of beaches in KATM and Bristol Bay: Swikshak, Hallo, North Hallo, and Naknek. During the peak of the die-off in 2016, Swikshak beach contained an average of 20.6 carcasses per survey. During a typical year (using 2014 as an example), the average was 0.33 carcasses, highlighting the severe impact that die-off events like this have on seabird colonies. These events have occurred throughout Alaska annually since 2015 (USFWS 2021) and have affected some of our beaches every year, particularly Naknek Beach in 2019.

Explosibility measurements on coal dusts from the Cape Breton Development Corporation's Lingan Mine, TransAlta's Highvale Mine and the Quintette Mine in B.C. have been carried out along with some tests on Pittsburgh Standard coal dust. The Quintette coal dust would not explode in the classical Hartmann apparatus, but did explode in the new 20-L vessel using a more powerful ignition source. The minimum explosible concentrations of the Lingan, Highvale and Pittsburgh coal dusts were all about the same (40 - 45 mg/L), that of the Quintette was higher (140 mg/L). The difference may be attributed to the much greater mean particle size of the Quintette dust. The explosion pressures (in kPa) were: Highvale, 600, Pittsburgh, 520, Lingan, 510, and Quintette, 440. The minimum oxygen concentrations required for explosions were (in % oxygen): Highvale 10.4, Lingan 10.5, and Quintette 14. The minimum ignition temperatures of dust clouds were (in °C): Highvale 510, Lingan 600, Quintette 620 and Pittsburgh 620. Further work is required to reconcile limit values.

Abstract:
The main objective of this study was to develop data that would facilitate a consistently effective method of biological control of fire blight disease to be developed and to enable its implementation for disease control by ensuring its compatibility with variations in the biological, environmental, and chemical conditions present in pear orchards. As considerable information on the pathogen and biological control of fire blight was already gathered from studies in California and elsewhere, an emphasis was placed on investigating the genetics and ecology of Erwinia amylovora, the causal agent of fire blight in Israel. Studies of plasmid profile, virulence on several host, serological characteristics, as well as DNA fingerprints with selected primers all revealed E. amylovora strains in Israel to be homogeneous. Strains did vary in their resistance to streptomycin, with those from more northern locations being resistant while those in the southern costal plain were all sensitive to streptomycin. Resistance appeared to be conferred by chromosomal mutations as in streptomycin-resistant strains in California. The biological control agent Pseudomonas fluorescens strain A506 colonized flowers of both the Costia and Spodona pear cultivars in Israel as well as Bartlett pear in California. Flowers that were open at the time of spray inoculation of trees subsequently harbored from 105 to 107 cells of strain A506 per flower, while those that opened subsequent to spraying developed population sizes of about 105 cells/flower within 5 days. The incidence of fire blight infections were reduced about 3-fold in several trials in which moderate amounts of disease occurred in the plot areas; this degree of biological control is similar to that observed in California and elsewhere. On two occasions warm and moist weather that favored disease led to epidemics in which nearly all flowers became infected and which was so severe that neither P. fluorescens strain A506 nor chemical bactericides reduced disease incidence. A novel method for identifying antagonistic microorganisms for biological control of fire blight and other diseases was developed. A bacterial ice nucleation gene was introduced into E. amylovora to confer an Ice+ phenotype and the population sizes of this modified pathogen on flowers that had been pre-treated with potential control agents was estimated by measuring the freezing temperature of colonized flowers. Antagonistic strains that prevented the growth of E. amylovora in flowers were readily detected as those in which flowers froze at a low temperature. The method is both rapid and unbiased and several bacterial strains with substantial biological control potential have been identified using this method.

Abstract:
Brucella Mutagenesis (TAMU) The working hypothesis for this study was that survival of Brucella vaccines was directly related to their persistence in the host. This premise is based on previously published work detailing the survival of the currently employed vaccine strains S19 and Rev 1. The approach employed signature-tagged mutagenesis to construct mutants interrupted in individual genes, and the mouse model to identify mutants with attenuated virulence/survival. Intracellular survival in macrophages is the key to both reproductive disease in ruminants and reticuloendothelial disease observed in most other species. Therefore, the mouse model permitted selection of mutants of reduced intracellular survival that would limit their ability to cause reproductive disease in ruminants. Several classes of mutants were expected. Colonization/invasion requires gene products that enhance host-agent interaction or increase resistance to antibacterial activity in macrophages. The establishment of chronic infection requires gene products necessary for intracellular bacterial growth. Maintenance of chronic infection requires gene products that sustain a low-level metabolism during periods characterized little or no growth (1, 2). Of these mutants, the latter group was of greatest interest with regard to our originally stated premise. However, the results obtained do not necessarily support a simplistic model of vaccine efficacy, i.e., long-survival of vaccine strains provides better immunity. Our conclusion can only be that optimal vaccines will only be developed with a thorough understanding of host agent interaction, and will be preferable to the use of fortuitous isolates of unknown genetic background. Each mutant could be distinguished from among a group of mutants by PCR amplification of the signature tag (5). This approach permitted infection of mice with pools of different mutants (including the parental wild-type as a control) and identified 40 mutants with apparently defective survival characteristics that were tentatively assigned to three distinct classes or groups. Group I (n=13) contained organisms that exhibited reduced survival at two weeks post-infection. Organisms in this group were recovered at normal levels by eight weeks and were not studied further, since they may persist in the host. Group II (n=11) contained organisms that were reduced by 2 weeks post infection and remained at reduced levels at eight weeks post-infection. Group III (n=16) contained mutants that were normal at two weeks, but recovered at reduced levels at eight weeks. A subset of these mutants (n= 15) was confirmed to be attenuated in mixed infections (1:1) with the parental wild-type. One of these mutants was eliminated from consideration due to a reduced growth rate in vitro that may account for its apparent growth defect in the mouse model. Although the original plan involved construction of the mutant bank in B. melitensis Rev 1 the low transformability of this strain, prevented accumulation of the necessary number of mutants. In addition, the probability that Rev 1 already carries one genetic defect increases the likelihood that a second defect will severely compromise the survival of this organism. Once key genes have been identified, it is relatively easy to prepare the appropriate genetic constructs (knockouts) lacking these genes in B. melitensis Rev 1 or any other genetic background. The construction of "designer" vaccines is expected to improve immune protection resulting from minor sequence variation corresponding to geographically distinct isolates or to design vaccines for use in specific hosts. A.2 Mouse Model of Brucella Infection (UWISC) Interferon regulatory factor-1-deficient (IRF-1-/- mice have diverse immunodeficient phenotypes that are necessary for conferring proper immune protection to intracellular bacterial infection, such as a 90% reduction of CD8+ T cells, functionally impaired NK cells, as well as a deficiency in iNOS and IL-12p40 induction. Interestingly, IRF-1-/- mice infected with diverse Brucella abortus strains reacted differently in a death and survival manner depending on the dose of injection and the level of virulence. Notably, 50% of IRF-1-/- mice intraperitoneally infected with a sublethal dose in C57BL/6 mice, i.e., 5 x 105 CFU of virulent S2308 or the attenuated vaccine S19, died at 10 and 20 days post-infection, respectively. Interestingly, the same dose of RB51, an attenuated new vaccine strain, did not induce the death of IRF-1-/- mice for the 4 weeks of infection. IRF-1-/- mice infected with four more other genetically manipulated S2308 mutants at 5 x 105 CFU also reacted in a death or survival manner depending on the level of virulence. Splenic CFU from C57BL/6 mice infected with 5 x 105 CFU of S2308, S19, or RB51, as well as four different S2308 mutants supports the finding that reduced virulence correlates with survival Of IRF-1-/- mice. Therefore, these results suggest that IRF-1 regulation of multi-gene transcription plays a crucial role in controlling B. abortus infection, and IRF-1 mice could be used as an animal model to determine the degree of B. abortus virulence by examining death or survival. A3 Diagnostic Tests for Detection of B. melitensis Rev 1 (Kimron) In this project we developed an effective PCR tool that can distinguish between Rev1 field isolates and B. melitensis virulent field strains. This has allowed, for the first time, to monitor epidemiological outbreaks of Rev1 infection in vaccinated flocks and to clearly demonstrate horizontal transfer of the strain from vaccinated ewes to unvaccinated ones. Moreover, two human isolates were characterized as Rev1 isolates implying the risk of use of improperly controlled lots of the vaccine in the national campaign. Since atypical B. melitensis biotype 1 strains have been characterized in Israel, the PCR technique has unequivocally demonstrated that strain Rev1 has not diverted into a virulent mutant. In addition, we could demonstrate that very likely a new prototype biotype 1 strain has evolved in the Middle East compared to the classical strain 16M. All the Israeli field strains have been shown to differ from strain 16M in the PstI digestion profile of the omp2a gene sequence suggesting that the local strains were possibly developed as a separate branch of B. melitensis. Should this be confirmed these data suggest that the Rev1 vaccine may not be an optimal vaccine strain for the Israeli flocks as it shares the same omp2 PstI digestion profile as strain 16M.

This report presents the data collected in 2020 as part of the long-term terrestrial vegetation monitoring program at Channel Islands National Park. The purpose of this monitoring program is to document long-term trends in the park’s vegetation communities. Data are collected from 30-m-long transects using a point-line intercept method. In the past, each transect was sampled annually. However, beginning in 2012 the program began adding randomly located transects to improve the representativeness of the sampling, and transitioned to a rotating panel design. Now only a core subset of the transects are surveyed annually. Non-core transects are assigned to one of four panels, each of which is surveyed once every four years. Coronavirus disease (COVID-19) caused a global pandemic that prevented field data collection from most of the transects scheduled for 2020. As a result, just 29 of the 127 transects scheduled were visited, distributed as follows: Santa Barbara Island (n = 18), Santa Cruz Island (n = 11). The small size of this 2020 dataset offers limited comparisons to previous years. The program also planned to install 19 new transects on Santa Rosa and San Miguel islands to complete the probabilistic component of the study design (Tiszler et al. 2016a), but the safety response to COVID-19 did not permit these installations. Key 2020 results and interpretation: For the transects visited at both islands in 2020, a long-term decline in mean absolute cover, measured as the mean number of taxa intersecting each point along a transect, was apparent. Annual rainfall correlated positively with mean absolute cover of vegetation. Six vegetation communities were visited at Santa Barbara Island compared to nine at Santa Cruz Island, despite the smaller number of transects surveyed at the latter. Mean absolute cover at both islands together averaged 87%, with native plants covering 44% and non-natives covering 39%. Among vegetation communities, mean absolute cover ranged from a high of 137% in a Lyonothamnus grove to a low of 32% in a riparian community where few plants were encountered along one transect. Mean absolute cover was higher at Santa Cruz Island (105%) than Santa Barbara Island (77%), and mean absolute native cover followed the same pattern (57% vs 36%). For both islands combined, mean richness per transect was 11 taxa. Compared to Santa Barbara Island, Santa Cruz Island supported higher overall mean species richness (14 vs 9 taxa) and higher species evenness (0.70 vs 0.55). Santa Cruz Island’s higher absolute cover, higher native species cover, higher species richness, and higher species evenness indicate that its vegetation is in better overall condition compared to Santa Barbara Island, and that the rate of recovery after overgrazing is faster at Santa Cruz Island than Santa Barbara Island.

One of the emerging technologies is the use of microbial agents for the control of postharvest diseases of fruits and vegetables. A number of antagonistic microorganisms have been discovered which have the potential to effectively control postharvest diseases. Some of this technology has been patented and commercial products such as AspireTM (Ecogen Corporatin, Langhorne, PA, USA), Biosave 10TM and Biosave 11TM (Ecoscience Inc., Worchester, MA, USA) have been registered for commercial use. The principal investigator of this project was involved in developing the yeast-based biofungicide-AspireTM and testing its efficacy under commercial conditions. This research project was initiated to fill the gap between the knowledge available on development and commercial implementation of yeast biocontrol agents and basic understanding of various aspects related to introducing yeast antagonists to fruit surfaces, along with verification of population genetics. The main objectives of this study were: Study ecology, population dynamics and genetic diversity of the yeast antagonists Candida guilliermondii, C. oleophila, and Debaryomyces hansenii, and study the effect of preharvest application of the yeast antagonist C. oleophila naturally occurring epiphytic microbial population and on the development of postharvest diseases of citrus fruit during storage. Our findings, which were detailed in several publications, have shown that an epiphytic yeast population of grapefruit able to grow under high osmotic conditions and a wide range of temperatures was isolated and characterized for its biocontrol activity against green mold decay caused by Penicillium digitatum. Techniques based on random amplified polymorphic DNA (RAPD) and arbitrary primed polymerase chain reaction (ap-PCR), as well as homologies between sequences of the rDNA internal transcribed spacers (ITS) and 5.8S gene, were used to characterize the composition of the yeast population and to determine the genetic relationship among predominant yeast species. Epiphytic yeasts exhibiting the highest biocontrol activity against P. digitatum on grapefruit were identified as Candida guilliermondii, C. oleophila, C. sake, and Debaryomyces hansenii, while C. guilliermondii was the most predominant species. RAPD and ap-PCR analysis of the osmotolerant yeast population showed two different, major groups. The sequences of the ITS regions and the 5.8S gene of the yeast isolates, previously identified as belonging to different species, were found to be identical. Following the need to develop a genetically marked strain of the yeast C. oleophila, to be used in population dynamics studies, a transformation system for the yeast was developed. Histidine auxotrophy of C. oloephila produced using ethyl methanesulfonate were transformed with plasmids containing HIS3, HIS4 and HIS5 genes from Saccharomyces cerevisiae. In one mutant histidin auxotrophy was complemented by the HIS5 gene of S. cerevisiae is functionally homologous to the HIS5 gene in V. oleophila. Southern blot analysis showed that the plasmid containing the S. cerevisiae HIS5 gene was integrated at a different location every C. oleophila HIS+ transformant. There were no detectable physiological differences between C. oleophila strain I-182 and the transformants. The biological control ability of C. oleophila was not affected by the transformation. A genetically marked (with b-glucuronidase gene) transformant of C. oleophila colonized wounds on orange fruits and its population increased under field conditions. Effect of preharvest application of the yeast C. oleophila on population dynamics of epiphytic microbial population on wounded and unwounded grapefruit surface in the orchard and after harvest was also studied. In addition, the effect of preharvest application of the yeast C. oleophila on the development of postharvest decay was evaluated. Population studies conducted in the orchard showed that in control, non-treated fruit, colonization of wounded and unwounded grapefruit surface by naturally occurring filamentous fungi did not vary throughout the incubation period on the tree. On the other hand, colonization of intact and wounded fruit surface by naturally occurring yeasts was different. Yeasts colonized wounded surface rapidly and increased in numbers to about two orders of magnitude as compared to unwounded surface. On fruit treated with the yeast and kept on the tree, a different picture of fungal and yeast population had emerged. The detected fungal population on the yeast-treated intact surface was dramatically reduced and in treated wounds no fungi was detected. Yeast population on intact surface was relatively high immediately after the application of AspireTM and decreased to than 70% of that detected initially. In wounds, yeast population increased from 2.5 x 104 to about 4x106 after 72 hours of incubation at 20oC. Results of tests conducted to evaluate the effect of preharvest application of AspireTM on the development of postharvest decay indicated the validity of the approach.
