Abstract:
Studies have revealed that Dioscorea hispida tubers contain a poisonous substance called alkaloid dioscorine. The method for removing alkaloid dioscorine in Dioscorea hispida is explored in this research through a soaking process. The tubers were peeled, washed, sliced, and soaked for 5 days in either 1.0 M sodium chloride (NaCl) or distilled water. The aim of this study was to firstly identify the amount of toxic dioscorine remaining after soaking for 5 days, and then determine the best solution for removing dioscorine compounds in the tubers that were obtained from a tropical area in Peninsular Malaysia. The liquid chromatography electrospray ionization mass spectrometric (LC-ESI-MS) systems were used to identify the presence of alkaloid dioscorine compounds within Dioscorea hispida tubers. The analysis showed that no dioscorine compounds were present in day 5 for samples soaked in the NaCl solution. However, the relative abundance in the distilled water solution at day 5 was 281000, indicating a 95% decrease of the relative abundance value of the dioscorine compounds. This work aimed to determine the minimum days needed to remove the poisonous element before Dioscorea hispida tubers could be used for food consumption or for any other applications.

Abstract:
Gadung tuber (Dioscoreae hispida Dennt), Indonesian called “umbi gadung”, is one of the potential sources of staple foods in supporting food diversification. Gadung tuber contains carbohydrateof 29.7g in every 100 g of material. It also contains 3.2 g of protein, vitamin C, vitamin B and other minerals. The purpose of this paper is to present how to eliminate dioscorine and cyanide acid and to process some food diversification of gadung tubers. The innovation technology of post-harvest handling to eliminate dioscorine and cyanide acid content can be done by immersing and washing them in salt water or blanching. Gadung tubers of 2 mm slice immersed in salt water of 8% and 7.5% decrease cyanide acid content of 5.45 ppm; while shreded gadung tubers immerse in salt water of 7.5% for 72 hours can reduce cyanide acid content of 18.75 ppm. Gadung tubers can be processed to be stapple food such as flour, flake. Flour of gadung tubers combined with wheat flour or starch can be processed to become snack food such as cake, sponge cakes. Research conclude that gadung tubers is safety forconsumption by immersing in salt solution of 7.5% to 8% for 72 hours to eliminate dioscorine and cyanide acid content. Combination of 60% and 50% of gadung flour can be used to make good cake and sponge cake.

The use of gadung (Discorea hispida dennst.) tubers as food source is not seen as an attractive option due to dioscorine and cyanide anti-nutritional compounds contained therein. The presence of dioscorine and cyanide compounds makes gadung tubers known as poisonous food. Although the community has known its poisonous nature, the use of gadung tubers as natural pesticides (biopesticides) is not commonly known by the community. Using gadung tubers as biopesticides provides solution to unanswered problems caused by chemical pesticides. This article seeks to describe the potential use of anti-nutritional compounds of gadung tubers as biopesticides based on literature reviews. Various literature related to anti-nutritional compounds contained in gadung tubers and their use as biopesticides have been analyzed in depth. The literature review results show that dioscorine and cyanide anti-nutritional compounds are toxic and can be used as biopesticides. The effectiveness of gadung tubers on various agricultural pests depends entirely on the concentration of gadung tubers used. The use of gadung tubers as biopesticides is needed to increase and maintain the productivity of agricultural land in addition to preserving natural resources.

The potential of machine vision application in the determination of alkaloid inDioscorea hispidarhizome was explored. A camera vision system used in this research is chroma meter CR-400 made by Konica Minolta. The first component is the hardware component that functions as an image acquisitioned for the system. The second component is the software part which converts reading obtained from the hardware. From the design review, the images from a variety of harvested ofDioscorea hispidawill be captured and the reading in L*a*b color space were converted. The prediction was also made on the statistical relationship between Hue and dioscorine level in the fruit. With the same rhizomes, 40 grams was selected for chemical analysis at a laboratory for the programme of East Coastal Economic Regions Kementerian Pengajian Tinggi, Universiti Sultan Zainal Abidin, Malaysia (ECER-KPT UniSZA). The samples were selected from 12 portions of 2 plants (I and B) collected from Kampung Kudat, Ajil, Terengganu, Malaysia. The development on this study is grouped as non destructive method to determine the dioscorine content which is one of the alkaloid components in the rhizome ofDioscorea hispida.

People from rural areas, who are less accessible to or are reluctant about medical services, sometimes prefer local traditional medicine practitioner for treatment of common ailments. But because they are not aware of the fact that such practitioners lack scientific knowledge, they suffer the serious consequences of such primitive treatments. These people are often served some kind of a plant root preparation and may lead to a fatal event from an overdose of such medication. One such case is reported here who after the advice of such a traditional medicine practitioner ended in a fatal event as a result of overdose of a root preparation from bitter yam. This article also describes the clinico- pathological aspects of bitter yam poisoning.

(Dioscorea cayennensis) is part of the Brazilian food habit, it is necessary to study the proteins present in this plant. The objective of this work was to isolate and characterize biochemically the dioscorin proteic fraction (DPF) present in the tubers of D. cayennensis, characterizing the presence of antinutritional compounds and antioxidant, antibacterial and antifungal biological activities. Isolation of DPF was performed with protein extraction in Tris-HCl 0.05M pH 8.3, followed by precipitation in Ammonium Sulfate (F45-75). The molecular weight of DPF was determined by SDS-PAGE and MALDI-ToF, characterized by the presence of a major component with a mass of approximately 28 kDa corresponding to dioscorin. When tested for the presence of phytochemicals and antinutritional compounds (lectins and trypsin inhibitors), the presence of these compounds was not detected in the DPF. The amino acid composition of the fraction was determined and the antioxidant activity estimated by the ABTS and DPPH methods. Glutamic acid, Aspartic acid and Arginine were the amino acids that showed higher levels in DPF, which explains the antioxidant activity, suggesting that the interaction of these amino acids seems to act on reactive molecules making them stable. Finally, in the evaluation of the antibacterial and antifungal activity of the total extract and DPF against different strains of microorganisms, an antibacterial potential was detected between 50-500 μg/mL of protein for inhibition of the growth of strain of L. monocytogenes.<br>Sementes, rizomas e tubérculos são geralmente boas fontes de moléculas bioativas, as quais estão relacionadas à promoção da saúde, as proteínas e outros compostos oriundos de fontes vegetais, vem ganhando relevância em pesquisas científicas. Considerando que o consumo do tubérculo de inhame (Dioscorea cayennensis) faz parte do hábito alimentar brasileiro, faz-se necessário o estudo das proteínas presentes neste vegetal. O objetivo deste trabalho foi isolar e caracterizar bioquimicamente a fração proteica dioscorina (FPD) presente nos tubérculos de D. cayennensis, detectando a presença de compostos antinutricionais e atividades biológicas antioxidante, antibacteriana e antifúngica. O isolamento do FPD foi realizado com a extração proteica em Tris-HCl 0,05M pH 8,3, seguido de precipitação em Sulfato de Amônio (F45-75). A massa molecular da FPD foi determinada por SDS-PAGE e MALDI-ToF. Caracterizado pela presença de um componente majoritário com massa de aproximadamente 28 kDa, correspondente a dioscorina. Quando submetida a testes de avaliação da presença de fitoquímicos e compostos antinutricionais (lectinas e inibidores de tripsina), não foi detectada na FPD a presença destes compostos avaliados. A composição aminoacídica da fração foi determinada e a atividade antioxidante estimada pelos métodos de ABTS e DPPH. Ácido glutâmico, Ácido Aspártico e Arginina foram os aminoácidos que apresentaram maiores teores na FPD, o que explica a atividade antioxidante verificada, sugerindo que a interação destes aminoácidos parece atuar em moléculas reativas tornando-as estáveis. Por fim, na avaliação da atividade antibacteriana e antifúngica do extrato total e FPD frente a diferentes cepas de micro-organismos, foi detectado potencial antibacteriano entre 50-500 μg/mL de proteína para inibição do crescimento da cepa L. monocytogenes.

Cette étude se propose de reconstituer la manière dont le De materia medica de Dioscoride, le plus important traité de pharmacie de la fin de l'Antiquité et de tout le Moyen Age méditerranéen, fut transmis, recopié, lu et étudié depuis sa rédaction (en grec), dans la seconde moitié du Ier siècle de notre ère, jusqu'aux premières éditions imprimées, à la Renaissance. Pour cela, l'ensemble de la tradition manuscrite directe est examinée: à chacun des représentants (papyrus et manuscrits) est consacrée une étude particulière détaillée, portant sur ses aspects codicologiques, historiques et philologiques. Il s'agit, à chaque fois de faire apparaître d'une part l'intérêt de ce témoin pour la reconstitution de l'histoire du texte de ce traité, d'autre part l'importance qu'il peut avoir pour l'établissement du texte. Ce faisant, il est possible de mettre en évidence l'existence de plusieurs grandes familles textuelles mais aussi d'un certain nombre de témoins isolés. Outre la tradition directe, sont examinées les citations de Dioscoride par Oribase. Enfin, les traductions latines de la fin de l'Antiquité et du Moyen Age ainsi que les traductions arabes médiévales font également l'objet d'une étude qui n'a pas tant pour but d'analyser leurs développement que d'étudier leurs rapports avec la tradition grecque. Une des principales conclusions de ces recherches est que l'édition de référence du De materia medica, publiée par Max Wellmann au début du XXe siècle, ne repose pas sur des critères satisfaisants, notamment parce qu'un certain nombre de manuscrits importants y ont été sous-estimés ou même laissés de côté. Il conviendrait donc d'en réaliser une nouvelle édition<br>The purpose of this work is to examine how Dioscorides' De materia medica, the most famous pharmalogical treatise in the mediterranean word during late antiquity and the Middle Ages was passed on, copied, read and studied from the time it was written (in Greek) in the second half of the first century A. D. , until the first printed editions, at the time of the Renaissance. In order to do this, the whole direct manuscript tradition is examined: a specific and detailed study is devoted to each papyrus or manuscript, from a codicological, historical and philological point of view, firstly to bring to light the information this particular manuscript can offer us concerning the history of the text and, secondly, to determine its interest for someone intending to edit this work with a view to subsequent publication. This method enables us to divide the various manuscripts into different textual groups but it also shows that several do not belong to any group. Not only is the direct greek manuscript tradition examined but also the chapters from De materia medica handed down by Oribasius in his collectiones medicae. Lastly, translations fromt both late antiquity and the middle ages, both latin and arabic, of De materia medica are analysed, mainly to determine their textual relations to the greek tradition. One of the most important results of this research is the demonstration that the last edition of De materia medica, published by Max Wellmann at the beginning of the 20th century, is no longer valid, chiefly because many important manuscripts were either undervalued or merely omitted. A new edition of this treatise is now therefore necessary

Mon travail a consisté essentiellement à proposer une traduction moderne et scientifique de l'herbier du Ps.-Apulée, un texte du IVe siècle qui contient des recettes médicales associées à diverses plantes médicinales. Cet ouvrage a connu un grand succès au Moyen Âge et il en existe de nombreux manuscrits. Néanmoins, il a longtemps été méprisé et considéré comme une simple compilation d'ouvrages grecs. L'édition commentée la plus récente est celle d'Ackermann en 1788 et il n'y a pas de traduction française qui tienne compte l'édition d'E. Howald et H. Sigerist en 1927 et des progrès accomplis depuis dans la connaissance de ce texte. Nous avons donc essayé de traduire le texte de l'Herbier édité par ces deux savants tout en expliquant, critiquant et parfois contestant certains de leurs choix. Nous avons aussi cherché dans l'introduction et dans les notes à situer l'oeuvre dans la tradition médicale en travaillant à la fois sur les sources de l'oeuvre et sur sa postérité et nous avons joint à la traduction en annexes un certain nombre de nos outils de travail. Nous espérons que ce travail permettra de mieux comprendre ce que représentaient les plantes médicinales, " les simples ", dans l'Antiquité tardive.

La présente étude porte sur la tradition de l'œuvre botanique et pharmacologique de Dioscoride, l'auteur du De materia medica, au cours des six premiers siècles de notre ère. Apres des remarques de méthode sur les problèmes particuliers qu'offre l'interprétation des textes anciens dans ce domaine, en particulier pour l'identification des espèces végétales citées (chap. 1), nous étudions le contexte de la composition de l'œuvre : qui fut Dioscoride, ce qu'il a écrit, etc. ; nous confirmons en particulier qu'il est sans doute bien l'auteur du traité des remèdes simples que l'on connait sous son nom (2) ; nous étudions ensuite la structure interne du De materia medica (3). Le problème des fraudes sur les remèdes et des succédanés, enfin, chez Dioscoride et ses contemporains, retient notre attention (4). Ces travaux préliminaires nous permettent de mieux comprendre la réception de l'œuvre, d'abord chez les médecins du 1er siècle (5), puis chez Galien, dont l'œuvre complexe mérite qu'on s'y attarde ; nous pensons que la pharmacologie de Galien (qui s'intéressait aussi au problème des fraudes) s'est construite à la fois avec l'appui de l'acquis dioscoridien et en réaction contre lui (6 et 7). Le principal témoin de la transmission dioscoridienne, après Galien, est l'herbarius dioscorideus, archétype perdu de deux célèbres manuscrits illustrés (8). La sélection qui y est opérée, à l'intérieur du corpus des notices dioscoridiennes, montre que son "auteur" était attentif à ce qu'on peut appeler l'accessibilité des remèdes, l'euporia (9). Cette tendance est également illustrée par un groupe de textes tardifs qui traitent des euporista et des succédanés (10). Dioscoride, devenu un classique de la matière médicale, est aussi à la racine de ce développement d'une littérature pseudépigraphe. A travers la transmission de son œuvre, nous voyons se dessiner l'histoire d'une science ; prescience ou pseudoscience, peut-être ; littérature scientifique, certainement (11)<br>The present work takes a closer look at the tradition of Dioscorides' botanical and pharmacological works during the first six centuries of our era. After some general remarks about the special problems that await the student of ancient texts dealing with pharmacology or botany, mainly that of identifying plant species mentioned by ancient writers (chap. 1), we study the context of Dioscorides' writing: who was Dioscorides? What works did he write? Among other questions, we give arguments for supporting the opinion that he is the author of the treatise about simple medicines, that goes under his name (chap. 2). We also take a look at the internal structure of de materia medica (chap. 3). Furthermore, we study the problem of adulterations and substitute medicines in Dioscorides' time and in his work (chap. 4). All these preparatory discussions enable us to understand the way Dioscorides was read by later writers, first those of his own century (chap. 5), then by Galen, whose huge and complex work in the realm of pharmacology, needed closer scrutiny: we consider that galen's pharmacology is built with the help of Dioscorides' legacy, but also as a reaction against it (chap. 6 and 7). The main witness of Dioscorides'transmission, after Galen, is the herbarius dioscorideus, in other terms the lost archetype of two well-known illustrated manuscripts (chap. 8). The way it carefully chooses to reproduce only some of Dioscorides chapters shows that its "author" had an eye for what we may call the accessibility of remedies, the euporia (chap. 9). This tendency is also exemplified by a group of later texts dealing with euporista and substitute medicines (chap. 10). Dioscorides, by then a classic of materia medica, is also at the root of that whole growth in pseudepigraphic literature. Through the transmission of his work, we can follow the history of a science; a pre-science or a pseudo-science maybe, but a scientific literature in its own right

Malgré le regain d'intérêt qu'ont connu ces dernières années les archives de Dioscore d’Aphroditê (Moyenne-Égypte, VIe s. après J.-C.), les documents coptes qu'elles contenaient sont restés curieusement sous-étudiés : en apportant des données nouvelles par rapport à la composante grecque, mieux étudiée, ils concourent à une meilleure compréhension historique de ces archives bilingues. Par ailleurs, faisant partie d'un ensemble majoritairement grec, ils posent le problème de l'usage et de la fonction du copte face au grec, langue de l'administration, autrement dit des rapports entre la langue nationale des Égyptiens et celle du pouvoir byzantin. Cette thèse constitue l’édition commentée d’un corpus de lettres coptes en grande partie inédit des archives de Dioscore. Il s’agit en effet du genre documentaire le mieux représenté du versant copte de cet ensemble archivistique. Le premier volume est constitué d’une synthèse qui pose le problème du rapport entre grec et copte dans un milieu villageois du VIe s. comme celui d'Aphroditê, qui traite des traits dialectaux, paléographiques et formulaires du copte en usage dans cette région et qui étudie les données historiques susceptibles de compléter celles livrées par les archives grecques. Le volume II contient les éditions commentées de ces vingt-et-une lettres, alors que le volume III contient les annexes (textes complémentaires, index, bibliographie et planches)<br>Despite the renewed interest in recent years in Dioscorus’ archives from Aphrodito (Middle Egypt, sixth century AD), the Coptic documents they contained have remained curiously understudied: by providing new data to that provided by the Greek texts, the best studied part of this archive, they contribute to a better historical understanding of these bilingual archives. Moreover, as part of a predominantly Greek dossier, they raise the issue of the use and function of Coptic in relation to Greek, the language of the administration, i.e., the relationship between the national language of the Egyptians and the one of Byzantine power. This thesis is the annotated edition of a corpus of Coptic letters, most of which have not previously been published, from Dioscorus’ archives. This text type is the documentary genre best represented in the Coptic component of this archival dossier. The first volume consists of a synthesis, which emphasizes the problem of the relationship between Greek and Coptic in a village like Aphrodito in the middle of the sixth century. It also deals with dialectal features, palaeographic and Coptic grammatical forms in use in the area, and studies historical data that complement those delivered by the Greek archives. Volume II contains the editions of twenty-one letters, with commentary, while volume III contains the appendices (complementary texts, indices, bibliography and plates)

Cette thèse est une édition commentée des papyrus grecs ayant appartenu à Dioscore d'Aphrodité (Egypte VIe s. Après J. -C. ). Une première partie regroupe des textes provenant de sa bibliothèque : un codex de l'Iliade d'Homère, un codex de Scholies à l'Iliade et un ensemble de tables de conjugaison des verbes contractes. La seconde partie est consacrée à l'oeuvre de Dioscore : 48 textes, majoritairement en vers, sont édités avec une traduction et un commentaire, accompagnés d'une présentation paléographique, littéraire, historique et grammaticale. Un chapitre conclusif tente de brosser le profil culturel de Dioscore en s'appuyant sur sa bibliothèque, son oeuvre et sur les textes documentaires qu'il rédigea en tant que notaire. La combinaison de ces trois sources révèle quelques constantes caractéristiques de la vie culturelle de la société protobyzantine.

La thèse consiste en l’édition des lettres grecques des archives de Dioscore d’Aphrodité (Égypte, VIe s. apr. J.-C.), qui constituent le plus important ensemble papyrologique d'époque byzantine connu de nos jours. Quatre-vingt-cinq documents y sont présentés sous la forme d'une nouvelle édition – ou, dans le cas de textes inédits, d'une première édition –, qui s'accompagne d'une traduction et d'un commentaire. La mise en parallèle des textes souvent fragmentaires du corpus, dont les pièces demeurent dispersées dans les collections du monde entier suite à la découverte clandestine des archives, a en outre permis de raccorder certains fragments encore inédits à des pièces déjà répertoriées, et de contribuer ainsi à la restitution d'un texte plus complet. Au-delà de l'aspect philologique, l’apport de cette documentation est aussi bien culturel qu’historique : elle permet d’éclairer l’arrière-plan multilingue en comparant la correspondance officielle écrite en grec avec la correspondance privée des mêmes archives, qui était de préférence rédigée en copte, ainsi qu'en analysant les pratiques d'écritures de certains scribes bilingues ; elle révèle les modalités épistolaires et les usages propres à la pratique écrite de l’administration ; elle dévoile les différents problèmes, matériels ou financiers, auxquels un village de Moyenne-Égypte devait faire face dans son quotidien ; enfin, elle contribue à la connaissance des institutions administratives et des rapports entre instances centrales et locales un siècle avant la conquête arabo-musulmane de l'Égypte<br>The dissertation concerns the edition and the study of the Greek letters belonging to the archive of Dioscorus of Aphrodite (Egypt, VI CE), the largest papyrological ensemble of the Byzantine age. Eighty-five documents are analyzed and highlighted by virtue of a new edition - or, when it comes to unpublished texts, of a first edition - followed by a translation and a commentary. Moreover, the study of the corpus, which is scattered across different collections around the world as a consequence of the clandestine discovery of the archive, allowed to join some unpublished fragments to some already known texts and to reconstruct, in this way, a more complete text. The contribution of this documentation is not only philological, but also cultural and historical: by comparing the official correspondence written in Greek with the private correspondence from the same archive, which was preferably written in Coptic, it helps to illuminate the multilingual background; it unfolds the modalities of epistolary exchanges and the written practices of the administration; it reveals the issues encountered by an Upper Egypt village in its daily life; finally, it contributes to the knowledge of the administrative institutions, as well as to the comprehension of the relations between central and local authorities a century before the Arab-Muslim conquest of Egypt

Malgré la large diffusion de ses œuvres, Eusèbe d’Alexandrie constitue une véritable énigme historique. Dans trois manuscrits grecs, son corpus supposé est bien accompagné de trois discours offrant davantage de détails biographiques, mais le récit contrefactuel qu’ils donnent leur a tôt valu d’être considérés comme fictifs. Quoique signée par un homme, Jean, se présentant comme le notaire d’Eusèbe, cette vie se heurte pourtant aux données historiques et fait de ce dernier le successeur de Cyrille. Or, si fiction il y a, force est de remarquer qu’elle réécrit une période tourmentée et substitue à l’hérétique Dioscore le pieux Eusèbe. Pour tâcher de mieux saisir l’identité d’Eusèbe, il faudrait donc commencer par s’intéresser à celle de Jean, signataire supposé d’une vie impossible. Cette thèse se propose de revoir à nouveaux frais l’édition des trois textes dits biographiques et d’en proposer traduction et commentaire afin d’estimer qui les a composés, quand, pourquoi et comment<br>In spite of the vast diffusion of his production, little is known about Eusebius of Alexandria. Three Greek codices do compile his collection alongside three texts displaying more biographical details, but their counterfactual recollection of events quickly led historians to view them as fictional. Although it is signed by a man, John, asserting he was Eusebius’s notary, this biography turns the enigmatic writer into Cyril’s successor, colliding thus with the many historical sources that remembered Dioscorus, the so-called Heresiarch, as Cyril’s actual successor. No matter how fictional this biography can appear, it is clear that it tried to erase out a troubled period of time. If one wants to understand who Eusebius really was, one should then first start to consider John, the alleged writer of an impossible tale. This essay contains therefore a new critical edition of the three biographical texts, with a French translation and a commentary aiming at figuring out who wrote them, when, why and how

Gargilius Martialis’ (3rd century A. D.) Medicinae ex holeribus et pomis : a pharmacopoeia for domestic use in Roman Africa – All components of Gargilius Martialis’ short handbook on The Remedies extracted from Vegetables and Fruits show that the aforenamed work has been written by a retired officer who was anxious to offer his Afro-Roman countrymen – on the fringe of the scientific pharmacology – a form of medication resolutely empiric and usual which stands at the level of individual or collective experience. Since by drawing up this collection of therapeutic formulae Quintus Gargilius Martialis was adding to the sensible remedies – stemmed from the compilation of a main source (Pliny’s Natural History) and of two secondary Greek sources (Dioscorides’ Materia Medica as well as Galen’s monumental works) – a whole series of incantations, spells and all manner of treatments conveyed through popular traditions obviously in order to produce a manual of family medicine. Thus it’s certainly not a matter of chance if about 570 A. D. the wealthy senator Cassiodorus – who enjoined the monks of the monastery he had founded at Vivarium (in the South of Italy) to assist the patients in giving them appropriate remedies – left at the library of the religious institution some medical texts of the highest importance : among other books featured precisely Gargilius Martialis’ treatise.
