Full textThe primary purpose of this research is to identify if a correlation exists between the U.S. Army selection procedure for enlisted soldiers in the ranks of E-1 through E-4 attending the Defense Language Institute and the academic outcome. This thesis focuses on two specific concepts: (1) how the U.S. Army currently identifies those soldiers to be trained as Russian linguists; and (2) whether the Army needs to incorporate changes to its current identification procedures currently used to select soldiers to attend the Russian linguist course at Defense Language Institute, Foreign Language Center (DLIFLC) are examined. A comparison is then made with the soldiers ability to successfully complete the courses in which enrolled. The purpose is to identify the causes that influence attrition. Secondly, this study analyzes the enrollment data produced by DLIFLC to determine if any reliable correlation exists between the current linguist identification procedures and the success or failure of soldiers enrolled in the Russian language course.

Esta dissertação apresenta uma análise da trajetória dos jovens participantes do Programa de Vocação Científica/Programa de Desenvolvimento Local Integrado e Sustentável /Escola Politécnica de Saúde Joaquim Venâncio/Fundação Oswaldo Cruz desde 2003 a 2008. Foi realizada uma revisão da literatura sobre políticas públicas e juventude objetivando discutir a construção social e política dos programas voltados para esta população, bem como a identificação de marcos históricos e conceituais que orientam as políticas sociais da Fundação Oswaldo Cruz. Como estratégia de pesquisa de campo foram realizadas entrevistas com gestores Fiocruz e com coordenadores de projetos e de programas institucionais voltados para a comunidade de Manguinhos, bem como, a realização de um grupo focal com egressos do PROVOC/DLIS com o objetivo de analisar as expectativas desses alunos ao ingressarem e ao final do programa, assim como, identificar o percurso institucional e pessoal do aluno e as possíveis repercussões sociais, familiares e profissionais dos jovens que participam do Programa. Conclui enfatizando posicionamentos e sugestões que contribuam para que novos caminhos possam ser pensados na questão da inserção de jovens de comunidades carentes em Programas de Vocação Científica mantidos por instituições de pesquisa.
This work presents an analysis of the trajectory of youth participants of Scientific Vocation Program - Integrated and Sustainable Local Development Programme of Joaquim Venancio Polytechnic Health School of Oswaldo Cruz Foundation (PROVOC DLIS / EPSJV / FIOCRUZ) program from 2003 to 2008. We performed a literature review on public policies and youth aimed to discuss the social and political construction of programs for this population, and the identification of historical and conceptual foundations that guide the social policies of FIOCRUZ. As a research strategy, interviews were conducted with managers of Fiocruz and with managers of projects and institutional programs aimed at the community of Manguinhos, as well as, conducting a focus group with alumnal of PROVOC / DLIS with the aim of analyzing the expectations of these students at entry and at the end of the program, as well as tracing institutional and student's personal and possible social, familiar and professionals repercussions of these who participate in the program. Concludes by emphasizing positioning and suggestions to help ensure that new ways can be thought on the issue of integration of young people from poor communities in Scientific Vocation Program maintained by research institutions.

Full textA região Nordeste vem sendo alvo de diversas políticas públicas apresentadas como soluções para dinamizar a economia regional e promover a melhoria da qualidade de vida da população. Nesse contexto, vem se destacando a execução da metodologia DLIS (Desenvolvimento Local Integrado e Sustentável) que define estratégias locais de desenvolvimento através da participação das comunidades. A partir do diagnóstico participativo no qual são destacados os problemas, as vocações e potencialidades do local é elaborado um plano de desenvolvimento do qual é extraída a agenda local com ações prioritárias que deverão ser executadas por vários parceiros das esferas federal, estadual, municipal e organizações da sociedade civil local. Implantada em todo país, em Sergipe, através da atuação do SEBRAE (Serviço Brasileiro de Apoio às Micro e Pequenas Empresas) a metodologia já foi implantada em 26 dos 75 municípios. Os planos de desenvolvimento, principalmente os que têm como escala de atuação o local, podem de fato contribuir para o desenvolvimento sustentável local, se forem consideradas as singularidades em termos de fatores geográficos, políticos, históricos e culturais e ainda, se for garantida a participação de todos os atores de desenvolvimento. Nosso trabalho está focado na avaliação da metodologia DLIS, sem, no entanto, entende-la como um modelo legitimador do desenvolvimento sustentável a ser perseguido por todos os municípios. Face nosso entendimento da importância dessa metodologia enquanto política de desenvolvimento e a necessidade da mesma ser avaliada sob a perspectiva substantiva de seus conteúdos, nosso trabalho procurou avaliar os impactos das ações implementadas pelo DLIS/SEBRAE no que diz respeito à sustentabilidade cultural e à participação local, preconizadas como indispensáveis ao alcance do desenvolvimento sustentável local, tomando-se como estudo de caso o município de Canindé do São Francisco/SE e, para tal, debruçamos nossas análises a partir de alguns vieses. Analisamos que o modo de vida, os valores culturais, a identidade dos atores sociais locais não foram fatores considerados no processo. Seus anseios em relação ao desenvolvimento sustentável local não foram contemplados. Desvendamos ainda o grau de inserção dos atores sociais locais no processo participativo e o tratamento conferido ao local pela metodologia. Entendemos esse estudo como contribuição à avaliação de metodologias de políticas sociais participativas que propõem uma nova relação entre Estado e sociedade, mas também, como tema fecundo que ressalta a inserção da dimensão cultural nos planos de desenvolvimento. Os resultados podem ser relevantes ainda para os formuladores de políticas públicas sociais, agentes de financiamento ao desenvolvimento local e organizações do terceiro setor interessados em conhecer os impactos dessas ações a partir da avaliação e percepção dos seus supostos beneficiários.

The Local Development Integrated and Sustainable - DLIS represented in the 1990s in Brazil, a strategic investment in the design and operation of public policies to combat poverty and social exclusion, sought to promote involvement and participation of civil society, building partnerships, establishing a vision of development more inclusive and integrated, surveillance and social control in the process of setting priorities in allocation of public resources in order to initiate a nationwide process of development more sustainable. This study focused on the theoretical debate, historical facts and conceptual categories relevant to understanding the new meaning that space takes place before an international context of economic globalization of capital and strong influence of neoliberalism in the planning of public policies of the Brazilian state. We attempted to ascertain the perceptions of implementing agents (governmental and non governmental organizations) the methodological strategy concerned about the efficiency of the process of community organization through the political role of civil society in local space. This was an analytical research of a qualitative approach, carried out through field study. As procedures, cite the literature, analysis of official documents, specific legislation. As a tool for data collection was used semi-structured interviews, participant observation and oral history. The interviews were conducted between November 2005 and January 2006, together with research subjects in the municipalities of Pedro Régis (forest area) and Nova Olinda (interior). The sample (n = 14) was intentional because it encompassed only professionals who have direct or indirect link with the municipalities that joined the expansion of the Active Community Program, the state of Paraíba. It was concluded that there was a dissociation between the principles and objectives of the strategy of the DLIS government with the intention of effecting an articulated process and sustainable development, which is useful in principle the rationalization of public resources and strengthening of visions of hegemonic power which showed a fragmented conception of development.
O Desenvolvimento Local Integrado e Sustentável DLIS, representou no Brasil dos anos 90 uma inversão estratégica na concepção e operacionalização de Políticas Públicas voltadas ao enfrentamento da pobreza e exclusão social. Buscou promover envolvimento e participação da sociedade civil, constituir parcerias, instituir uma visão de desenvolvimento mais abrangente e integradora e promover a fiscalização e controle social no processo de definição de prioridades na alocação de recursos públicos desencadeando em âmbito nacional um processo de desenvolvimento mais sustentável. O foco do presente estudo foi o debate teórico, fatos históricos e categorias conceituais relevantes para a compreensão da ressignificação que o espaço local adquire diante de um contexto internacional de globalização econômica do capital e forte influência do neoliberalismo no planejamento das políticas públicas do Estado brasileiro. Buscou conhecer as percepções dos agentes implementadores (governamentais e não governamentais) da metodologia em questão sobre sua eficácia no processo de organização comunitária, por meio da atuação política da sociedade civil, no espaço local. Tratou-se de uma pesquisa analítica, de cunho qualitativo, realizada através de estudo de campo. Como procedimentos aplicados citam-se a pesquisa bibliográfica, análise de documentos oficiais e legislação específica. Como instrumento de coleta de dados foram utilizadas entrevista semi-estruturada, observação de participante e história oral. As entrevistas foram realizadas entre os meses de novembro de 2005 a janeiro de 2006, junto aos sujeitos da pesquisa nos municípios de Pedro Régis (zona da mata) e Nova Olinda (sertão). A amostra (n=14) foi intencional porque englobou apenas profissionais que tiveram vinculação direta ou indireta com os municípios que aderiram à expansão do Programa Comunidade Ativa, no Estado da Paraíba. Concluiu-se que existiu uma dissociação entre os princípios e objetivos da metodologia do DLIS com a intenção governamental de efetivar um processo articulado e sustentável de desenvolvimento, sendo útil a princípio a racionalização dos recursos públicos e reforço de visões hegemônicas de poder, o que evidenciou uma concepção fragmentária de desenvolvimento.

A new trend in terms of actions for the local development has been configured. These actions consider the territory and its actors. It is in this context that the inductions methodology of the Integrated and Sustainable Local Development (DLIS, from the portuguese Desenvolvimento Local Integrado e Sustentável) arises. This Program is a strategy of participatory induction for the development, which mobilizes communities resources, that in partnership with the State (in its three levels) and the market, reaches diagnostics and identify potential vocations, prepare integrated development plans, in the perspective of the subjects involvement as proponents and protagonists of a social action in their territories. With this study it was aimed to understand the multidimensionality of the DLIS process in the state of Roraima, and at the same time, looking for the causal link between the socio-economic trajectories (results) of DLIS and social capital. The survey is characterized as qualitative research. The case study method was applied, involving the municipalities of Rorainópolis, Uiramutã, Baliza and Pacaraima. It was adopted three dimensions analytical contextual, which are: conceptual, implementation and of impact. In the evaluation of social capital were considered as variables the participation, trust, cooperation and networks. During the data interpretation, the following approaches were employed: contextual analysis, descriptive analysis, differences in proportions analysis and correspondence analysis. The survey results showed that some concepts and categories employed by the Program, while important, have some failures. Two municipalities, Rorainópolis and Uiramutã, were considered as good results. This, because they combine a number of issues that expressed desirable conditions to turn the DLIS effective with its multiple dimensions. Moreover, the municipalities of Baliza and Pacaraima were classified as bad results municipalities. The results of the DLIS in Rorainópolis and Uiramutã are linked to the social capital (positive relation). Despite of the extensive literature that assigns the relevance of the social capital, it does not have, yet, satisfactory instruments to measure the social capital. In this context and in fact, the understanding of these results represents merely an approximation.
Uma nova tendência em termos de ações para o desenvolvimento vem se configurando. Trata-se de ações que levam em conta o território e seus atores. Nesse âmbito, insere-se a metodologia de indução do Desenvolvimento Local Integrado e Sustentável (DLIS). Esse Programa de política pública se constitui uma estratégia participativa de indução do desenvolvimento, pela qual se mobilizam recursos das comunidades, que em parceria com o Estado (em seus três níveis) e o mercado, realizam diagnósticos, identificam potencialidades e vocações, elaboram planos integrados de desenvolvimento, na perspectiva de envolvimento dos sujeitos como proponentes e protagonistas da ação social em seus territórios. Com este trabalho, buscou-se compreender a multidimensionalidade do processo de DLIS, no estado de Roraima, procurando, ao mesmo tempo, o estabelecimento de um nexo causal entre as trajetórias sócio-econômicas (resultados) do DLIS e o capital social. A pesquisa se enquadrou na modalidade qualitativa. Adotou-se como estratégia metodológica o estudo de caso, envolvendo os municípios de Rorainópolis, Uiramutã, Baliza e Pacaraima. Três dimensões analítico-contextuais foram adotadas, quais sejam: conceitual, de implementação e de impacto. Na avaliação do capital social, as variáveis participação, confiança, cooperação e redes foram consideradas. Na interpretação dos dados, as seguintes abordagens foram utilizadas: análise contextual, análise descritiva, análise das diferenças de proporções e análise de correspondência. Os resultados da pesquisa revelaram que alguns conceitos e categorias adotados pelo Programa, embora relevantes, apresentam alguma ordem de problema. Dois municípios, Rorainópolis e Uiramutã, foram considerados municípios de bons resultados. Isso porque atenderam a um conjunto de questões que expressavam condições desejáveis para que o desenvolvimento local integrado e sustentável, em suas múltiplas dimensões, se efetivasse. Por seu turno, os municípios de Baliza e Pacaraima foram classificados como municípios de maus resultados. Os resultados do DLIS em Rorainópolis e Uiramutã estão associados ao capital social (relação positiva). A despeito da extensa literatura que trata da relevância desse tema, não se têm, ainda, instrumentos satisfatórios para medir capital social. Nesse contexto, entendese que estes resultados representam, de fato, uma aproximação.

Full textA promoção do desenvolvimento é um compromisso assumido por decisores políticos, organizações sociais e cidadãos em geral. A criação de mecanismos de monitorização e avaliação do desenvolvimento local assumem particular importância para a concretização operacional dos princípios subjacentes à integração e sustentabilidade das comunidades. Neste trabalho é efetuado a abordagem ao conceito de Desenvolvimento Local Integrado e Sustentável – DLIS – como uma estratégia de transformação e evolução da sociedade e o objetivo pretendido foi contribuir para a construção do índice que permita quantificar e percecionar o nível de DLIS dos concelhos portugueses. O índice proporciona aos decisores políticos avaliar as áreas que carecem de atuação de políticas públicas e auxilia na perceção das boas práticas de outros municípios. Foram definidas seis dimensões que explicam, no seu conjunto, o DLIS e realizaram-se uma série de testes para aferir os indicadores com potencial de contribuição para a construção do índice. Posteriormente foram calculadas as dimensões apoiado em aplicações de análise fatorial de componentes principais. As regiões de Lisboa e do Norte apresentam os melhores valores nas várias dimensões do DLIS, sendo que, as restantes têm na sua maioria, resultados nas dimensões do DLIS inferiores à média
Promoting development is a commitment made by policy makers, social organizations and citizens in general. The creation of mechanisms for monitoring and evaluation of local development are of particular importance to the achievement of operational principles underlying the integration and sustainability of communities. In this work it is approached the concept of Integrated and Sustainable Local Development – DLIS – as a strategy of transformation and evolution of society. The intended goal was to contribute to the construction of the index that allows to quantify and perceive the level of DLIS of Portuguese municipalities. The index allows policy makers to assess areas needing action of public policies and helps in perception of good practices from other municipalities. Six dimensions were defined that explain, as a whole, the DLIS and a series of tests were conducted to measure the indicators with the potential to contribute to the construction of the index. Subsequently, it was calculated the dimensions supported by analysis applications factorial. The regions of Lisbon and the North have the best values in the various dimensions of DLIS, and the rest have mostly results in the dimensions of DLIS below average.

Full textThe manufacturing of waveguides from insulating C fibers reinforced polymer composites necessitates to render internal surfaces electrically conductive. This can be achieved by metallization but technological bottlenecks arise due to the complex geometry and the chemical inertia of the surface. The CFRP matrix is composed of RTM6 epoxy whose glass-transition temperature equals 216 °C, limiting the choice of metallization techniques. Additionally, the surface energy of the polymer (constituting the surface of the CFRP) is low (20-40 mJ/m2) and non-polar, i.e. a poor adherence is expected for metallic coatings. A DLI-MOCVD process is used to form the metallic coating because it combines a good control of reactive fluxes, high deposition rates, and because it is relevant for the coating of three-dimensional substrates. The choice of the metal (Cu) is dictated by the anterior know-how of the research group, by the possibility to deposit at a temperature well below 216 °C, and because Cu shows an excellent electrical conductivity and a good resistance to ambient ageing. The Cu film is formed from the organometallic precursor (hfac)Cu(MHY), developed par P. Doppelt at CNRS (Paris). The precursor is diluted in anhydrous octane to make it injectable by DLI. After the mounting and implementation of the DLI-MOCVD reactor dedicated to the PhD work, a study of the chemical kinetics of the deposition from this precursor helped in the definition of the optimal conditions; noticeably the deposition temperature fixed at 195 °C. The coatings are partially covering and interfacial adherence is poor, but they are conductive. Consecutively, we identify, develop and test several pretreatments in order to modify the topography and reactivity of the CFRP surface. Based on 6 recipes, we show that mechanical anchorage is preponderant over the chemical reactivity to improve the adherence. The two best pretreatments are the solution chemistry treatment CircupositTM, and the in situ treatment with O3. At this stage of the work, Cu films are conductive and adherent but they do not wet the entire composite surface, even after long deposition duration. We then demonstrate that the injection of a Co precursor (Co2(CO)5) alternating with the Cu deposition leads to the formation of a pure metallic Cu film (> 98%) which is covering, conductive and adherent. The role of the Co precursor is not yet established. Nevertheless, we start the preliminary process industrialization studies consisting in the improvement of the growth rate and the yield. Noticeably, costs are decreased by replacing the octane solvent. With it, the Cu coating is adherent without any pretreatment of the substrate surface! The Co precursor is still necessary to ensure the proper covering of the entire surface, though. In these conditions, and after adjustments of some parameters, the first 60 mmlong waveguides are coated. The thermal cycling resistance is excellent. Therefore, waveguides length is increased to 300 mm, the minimum dimension for the radiofrequency transmission tests (final property) to be correct. The increase of length causes a complex adaptation of the process in order to guarantee the deposition of a uniform coating over the length, in terms of thickness and electrical properties. Radiofrequency tests result in a stationary waves rate < 30 dB, which is acceptable at the industrial level. Yet, losses are too important in comparison with current waveguides, but many proposals are formulated to improve our solution

Full textThe presence of bacteria and biofilms is permanent concern in many fields. Their presences are at the origin of many events which have high costs for the health system. In this objective, this work aimed to elaborate transparent nanocomposite thin films which are composed of nanometric metallic particles of antibacterial element (Ag or Cu) embedded in an oxide matrix (TiO2). The DLI-MOCVD process (Direct Liquid Injection-Metal Organic Chemical Vapour Deposition) was used to elaborate these thin films. This process allows the quantity of precursors injected into the CVD reactor to be controlled and porous body to be coated. The growth mechanisms and the structure of the Titanium dioxide (TiO2) are influenced by the presence of the organic precursor which contains the antibacterial element. The mole fraction of precursor (Ag or Cu) modifies the physico-chemical and structural properties of films. The antibacterial activity was tested according to the standard JIS Z 2801: 2000 with S. aureus and E. coli without light Specific tests were optimised in order to evaluate their activity in environments more representative. The composition of coatings impacts strongly the antibacterial activities from inactive to bactericidal properties. Correlations between the microstructure and composition of films and their antibacterial properties are discussed

Full textThis work is focused on development of growth protocols by direct liquid injection chemical vapor deposition (DLI-CVD) of catalytic porous films which could be used in proton exchange membrane fuel cells (PEMFC). The aim of this work was to reduce or even proscribe platinum in catalysts having large specific surface area i.e. being very porous. Besides, the aim is also to use mainly low cost precursors.Cerium oxide, which is a material widely used as catalyst, has been chosen to partially substitute platinum. Porous CeO2 layers were obtained by the optimization of processing parameters such as deposition temperature or precursors flow rates. Controlled platinum doping of cerium oxide surface has been obtained from deposition of ceria and platinum at the same time. The platinum precursor flow rate has allowed adjusting the load and the oxidation state of Pt in films without changing their porosity. For example, it was possible to synthesize thin cerium oxide films composed of surface Pt doped nanoparticles. In such a case, the Pt content is 5% in top most layers and its oxidation state is +II. The effect of precursor flow rate on the platinum oxidation state was confirmed from the study of the Pt / TiO2 system. Finally, platinum has been fully replaced by tungsten oxycarbide. Full textThe industrial development of the next generations of telecommunications will have to go through the adoption of new piezoelectric materials in the form of thin films. In that regard, the direct liquid injection metalorganic chemical vapor deposition (DLI-MOCVD) of thin films of lithium niobate (LiNbO3), a ferroelectric material presenting exceptional properties in the field of acoustics and optics was studied in this work. Fine control of both the microstructure and the composition of these layers is a critical parameter in obtaining functional thin films, especially for radiofrequency filter applications. For this reason, in this work, we first focused on the deposition of epitaxial thin films of controlled composition at high temperature for the realization of surface acoustic wave (SAW) devices. Then, in a second phase, on the deposition of highly oriented thin films on silicon-based heterostructures at a lower temperature for the realization of bulk acoustic wave (BAW) components. The optimization process presented in this manuscript demonstrates the impact of several deposition parameters: total pressure, injection flow rate, precursor molar ratio, on the thin films, and more particularly on its lithium composition, decisive parameter in the definition of material properties. The relative arrangement of the lattice of the film with that of the substrates are presented as well, for all epitaxial growth orientations. Finally, surface acoustic wave and bulk acoustic wave devices based on lithium niobate thin films deposited by DLI-MOCVD were fabricated and characterized

Full textAtmospheric MOCVD processes assisted by Direct Liquid Injection, attractive for on-line strip coating of metal pieces, were developped for the growth of advanced metallurgical coatings. Two chromium metal organic sources have been employed under various ambience : Cr (CO)6 for coatings in the Cr-O-N-C system, and bisbenzenechromium for metal chromium, carbide (Cr-C) and nitride chromium (CrN) monolayer and CrN/Cr-C multilayer coatings with a typical 25 nm biperiod. A thermodynamic approach has permited the calculation of a new ternary phase diagram in the Cr-C-N system and the study of the solvent impact on the nature of the coatings in comparison with chemical systems without solvent. Preliminary mechanical properties from scratch test, nanoindentation and residual stress measurements as well as tribological and corrosion behavior of the mono and multilayer Cr-based coatings are discussed in relation with their microstructure. Nanostructured CrN/Cr-C multilayer coatings exhibit promising hardness and wear resistance

Dementia with Lewy bodies (DLB) is the second most common neurodegenerative dementia only to Alzheimer''s disease (AD). In fact, DLB shares common pathological features with AD and Parkinson''s disease (PD), that simultaneously accompany α-synuclein and Aβ. These misfolded proteins accumulate in the brain and result in a large number of neuronal damage and brain atrophy, eventually leading to of the damages of the corresponding neurological function. However, there is currently no clinical treatment of drugs for the improvement of DLB. It is in urgent need for developing new therapeutic strategies of DLB drugs. According to previous studies, the risk of dementia in diabetics is 60% higher than that of the normal control. In addition, many researches have pointed out that the relationship between metabolism and cognitive impairment is bidirectional. Particularly, the resistance of insulin signals appears to be the common feature of such senile neurodegenerative diseases. Since glucagon-like peptide-1 (GLP-1) is known to improve insulin signaling, we postulate that GLP-1 may be potentially used as a treatment strategy in DLB. In. the present study, we developed a conditional approach to stimulate α-synuclein overexpression, and then treated Aβ in this cell model to mimic the pathological condition of DLB. By using this in vitro model, we explored the putative molecular mechanisms that may cause DLB. In addition, we also established a rat''s DLB animal model, and investigated whether Liraglutide could improve behavioral symptoms of DLB. The results showed that Aβ significantly induced cytotoxicity of α-synuclein, whereas administration of Liraglutide significantly increased cell viability and inhibited the expression of pSer307-Insulin receptor substrate 1 (IRS-1) to activate AKT signaling. This indicates Liraglutide can alleviate insulin resistance caused by Aβ-induced α-synuclein neurotoxicity. Moreover, Liraglutide can increase autophagy-related protein expression by activating the AMP-activated protein kinase (AMPK)/ mammalian target of rapamycin (mTOR) pathway, which stimulate nuclear factor erythroid 2-relaated factor2 (Nrf2), superoxide dismutase 1 (SOD1) and silent information regulator-mammalian ortholog 1 (Sirt1) to reduce the oxidative stress induced by Aβ and α-synuclein. The results of animal experiments also showed that compared with the disease group, DLB rats injected with Liraglutide showed significant improvements of working memory, cognition function and depression by T-maze, object recognition and depression behavior test. In conclusion, our study demonstrated that Liraglutide can attenuate Aβ-induced α-synuclein neurotoxicity in both in in vitro and in vivo models, which may provide a possible treatment strategy for DLB in the future.

Full textThis thesis, evaluates the cold conditions to which workers are exposed, particularly in the agro-industrial sector, evaluating two cases, a food distribution and a dairy company. Both companies are big consumers of cold, and are dependent on it to ensure the quality of the sold product. Thus, since both evaluated cases are cold consumers, the workers of these companies will be exposed to these conditions during the work day. In order to evaluate the thermal comfort of workers international standards and procedures were followed. Measurements of environmental and individual nature were conducted in order to characterize the areas and conditions where workers are inserted, as well as levels of activity of each worker. The type of activity that each worker performs, allowed to identify different types of exposure, cases of continuous and cases of intermittent cold exposure. The standards used, allowed to determine the thermal comfort indices, like IREQ index (ISO 11079, 2007) and the PMV and PPD index (ISO 7730, 2008).To determine these indices was necessary to determine other parameters, such as metabolic levels (ISO 8996, 2004), clothing insulation (ISO 9920, 2007), thermal sensations expressed by workers during cold exposure and the use of measurement procedures (ISO 7726, 1998). The obtained results allowed to identify cases that require attention, due to poor insulation level used for conditions that workers in general are exposed. As the level of thermal insulation is not adequate in most cases, the time limit recommended for cold exposure is lower than the time that workers are exposed to it. Finally, taking into account the type of exposure to which each worker was subject, optimal levels of clothing thermal insulation that give them the minimum thermal conditions to withstand the environmental conditions in each area assessed were determined.

Find full textParkinson’s disease (PD) is a devastating neurodegenerative disorder that affects 10 million people worldwide and is characterized by pronounced motor symptoms. Dementia with Lewy Bodies (DLB) involves both cognitive and motor deficits and affects ~1 million people in the United States. To date there is no cure for PD or DLB, and current treatments address only a subset of the symptoms that define these diseases. PD and DLB are ‘synucleinopathies’, defined as disorders involving the accumulation in patients’ brains of Lewy bodies. Lewy bodies are cellular inclusions that consist largely of aggregated species of alpha-synuclein (aSyn), a presynaptic protein that exists as both cytosolic and membrane-bound forms. Pathophysiological findings suggest that aggregated aSyn is involved in neurodegeneration in PD and DLB. However, mechanisms by which aSyn forms neurotoxic aggregates, and neurotoxic processes that distinguish different synucleinopathies such as PD and DLB, are poorly understood. To address these gaps, we have (i) designed a protocol to establish a primary cell culture model that can recapitulate key neuropathological features of PD, (ii) examined effects of expressing aSyn variants in a rat model of PD, and (iii) examined the expression profiles of neuroprotective genes in PD and DLB brain specimens.
In the first part of my thesis, I describe the development of an optimized protocol to prepare primary midbrain and cortical cultures from rat embryonic brains for the study of PD and other synucleinopathies. The establishment of cellular models that simulate specific aspects of neuropathology can enable the characterization of molecular perturbations that lead to dopaminergic (DA) neuronal death. Our primary midbrain mixed culture model provides an outstanding opportunity to explore therapeutic strategies to rescue DA neurons from toxicity elicited by a range of PD-related insults. In addition, our primary cortical mixed cultures can be used to model cortical neuropathology in various CNS disorders including synucleinopathies.
A number of mutations in the gene that codes for aSyn are associated with familial, early-onset forms of PD. A major goal of my thesis research is to characterize neurotoxic effects of a recently discovered familial substitution, A53E. This mutant was chosen based on the rationale that the introduction of a negatively charged residue at position 53 could potentially interfere with aSyn-membrane interactions and favor A53E aggregation, as we described for other familial aSyn mutants. For the first time, we have reproduced the neurotoxicity of A53E seen in human patients by expressing the mutant protein in rat midbrain. Rats injected unilaterally in the substantia nigra (SN) with rAAV encoding A53E and another familial mutant, A53T, but not rAAV encoding WT aSyn or a vector-control (‘stuffer’) virus, exhibited a significant motor impairment. Immunohistochemical analysis at 14 weeks after the viral injection revealed that brain sections from aSyn-expressing rats exhibit key features reminiscent of neuropathology in human PD, including nigral dopaminergic neuron loss (confirmed by unbiased stereology), striatal terminal depletion, and aSyn inclusion formation. In addition, it was determined that WT aSyn and the A53E and A53T mutants invaded the non-injected substantia nigra, implying that expressed aSyn protein can spread throughout the brain in the rat rAAV-aSyn model. These results yield insights into the molecular basis for the neurotoxicity of A53E and shed light on a potential role for membrane-induced aSyn aggregation in PD pathogenesis in vivo, thus setting the stage for developing therapies to slow neurodegeneration in the brains of familial and idiopathic PD patients.
aSyn neurotoxicity varies with the expression of neuroprotective proteins, and misfolded aSyn affects cellular functions and gene expression. These observations suggest that differential gene expression patterns can inform us about similarities and differences in pathogenic mechanisms of different synucleinopathy disorders. A third phase of my thesis research was aimed at determining the expression levels of a panel of candidate neuroprotective genes in post-mortem brain samples from DLB and PD patients and age-matched controls (5 individuals in each group). mRNAs encoding the following proteins were quantified via qRT-PCR in homogenates prepared from the frontal cortex and the BA24 region encompassing the cingulate gyrus: DJ-1, a protein with antioxidant and chaperone activities; PGC1α, a master regulator of mitochondrial biogenesis and oxidative metabolism; MsrA, an antioxidant enzyme responsible for repairing oxidatively damaged proteins; and ATP13A2, a lysosomal protein involved in autophagy. In addition to yielding new insights into differential gene expression patterns in cortex versus cingulate gyrus, the data revealed differences in mRNA expression levels in DLB versus non-DLB cortical tissue. Although levels of all four neuroprotective mRNAs were increased (or showed a trend towards being increased) in DLB cortex, Western blot analysis revealed that only the DJ-1 and PGC1α proteins showed a trend towards being up-regulated, whereas levels of ATP13A2 and MsrA were unchanged. These findings suggest that there is a failure to induce cellular antioxidant responses and lysosomal autophagy at the protein level in DLB cortex, and in turn this failure could contribute to neuropathology. Interestingly, analysis of the same panel of neuroprotective genes in PD cortical samples did not show significant differences in mRNA or protein levels compared to control samples, suggesting that different neuroprotective mechanisms are induced in DLB versus PD cortex. These studies shed light on brain-region specific changes in gene expression associated with different synucleinopathy disorders, and they set the stage for developing new diagnostic tests and therapeutic strategies.
