Abstract:
The DMI1, DMI2, and DMI3 genes of Medicago truncatula, which are required for both nodulation and mycorrhization, control early steps of Nod factor signal transduction. Here, we have used diverse approaches to pave the way for the map-based cloning of these genes. Molecular amplification fragment length polymorphism markers linked to the three genes were identified by bulked segregant analysis. Integration of these markers into the general genetic map of M. truncatula revealed that DMI1, DMI2, and DMI3 are located on linkage groups 2, 5, and 8, respectively. Cytogenetic studies using fluorescent in situ hybridization (FISH) on mitotic and pachytene chromosomes confirmed the location of DMI1, DMI2, and DMI3 on chromosomes 2, 5, and 8. FISH-pachytene studies revealed that the three genes are in euchromatic regions of the genome, with a ratio of genetic to cytogenetic distances between 0.8 and 1.6 cM per μm in the DMI1, DMI2, and DMI3 regions. Through grafting experiments, we showed that the genetic control of the dmi1, dmi2, and dmi3 nodulation phenotypes is determined at the root level. This means that mutants can be transformed by Agrobacterium rhizogenes to accelerate the complementation step of map-based cloning projects for DMI1, DMI2, and DMI3.

Abstract:
This paper aims to propose predictive equations for the small-strain shear modulus (Gmax) and small-strain damping ratio (Dmin) of a granulated mixture with plastic and nonplastic materials to reduce the dynamic energy of the ground. Polyurethane bead (PB) and glass bead (GB) were used as the plastic and nonplastic materials, respectively. 180 resonant-column tests were conducted with various conditions affecting the dynamic properties, such as nonplastic particle content (PC), void ratio (e), particle-size ratio (sr), and mean effective confining pressure (σm′). The results showed that Gmax and Dmin, respectively, increased and decreased as e decreased with increasing σm′ of material mixtures. In addition, Gmax decreased with an increase in PC, whereas Dmin increased. It was also found that sr of materials affected the changes in Gmax and Dmin. With an increase in sr, Gmax increased while Dmin decreased because small particles do not hinder the behavior of large particles as the size of larger particles increases. Finally, based on the results, new equations for estimating Gmax and Dmin of a granulated mixture with PB and GB were proposed as functions of PC, e, median grain size (D50), and σm′.

Abstract:
Abstract Introduction: Double minutes (dmin) are small chromatin particles that lack a centromere and represent a form of extrachromosomal gene amplification. They have been found in a variety of solid tumors but are rare in hematological malignancies. C-MYC and MLL are the most common genes amplified on the dmin in AML. Here we report a case of AML in which FISH did not demonstrate C-MYC or MLL gene amplification on the dmin. Case Report: A seventy-seven-year-old man was diagnosed with acute leukemia. Histology and flow cytometry (blasts positive for CD13, CD33, CD34, HLA-DR, CD15, CD117 and cytoplasmic MPO) were consistent with M2 AML. Chromosome analysis revealed abnormal karyotype with hypodiploidy 42–44 and loss of chromosome 5, 7,13,17,18, and Y. Structural rearrangements involving chromosomes 2,7,11,19 and 20 in addition to 2 marker chromosomes and dmin. FISH did not reveal C-MYC gene amplification. FISH did reveal MLL gene amplification, but not on the dmin. The patient received induction chemotherapy, and expired on day 14 due to cardiac complications. Discussion: Double minutes are a mechanism for upregulated oncogene expression. After the development of molecular technology, FISH, it became possible to identify the amplified gene. A previous review (Thomas et al. Double minute chromosomes in monoblastic (M5) and myeloblastic (M2) acute myeloid leukemia: two case reports and a review of literature. Am J Hematol 2004 Sep;77(1):55–61), found C-MYC to be amplified on the dmin in 25 cases, MLL was found to be amplified on the dmin in 5 cases, and only 3 cases in which neither gene was found to be amplified on the dmin. Double minutes are associated with advanced age at presentation, a more aggressive clinical course, poor outcome with therapy, and shortened survival. The survival in most cases is reported to be less than 12 months.

Abstract:
Abstract Homogeneous staining region (hsr) and double minutes (dmin), cytogenetic indicators of gene amplification, are rare findings in hematologic malignancies, and most frequently associated with acute myeloid leukemias (AML). Herein, we report a 74-year-old female presenting AML with maturation (AML-M2) with c-MYC amplification in hsr and dmin. Conventional cytogenetic techniques showed 46, XX, hsr(4)(q25)[15]/46, XX,~20dmin[5]. The c-MYC amplification in hsr was suspected by multicolor fluorescence in situ hybridization (mFISH) using 24XCyte Human mFISH Painting Kit. Amplification of c-MYC in hsr and dmin were confirmed by fluorescence in situ hybridization (FISH) using LSI MYC(8q24) Dual color, Break Apart Rearrangement Probe. The presentation of both dmin and hsr was rare in AML cases. Majority of AML cases with c-MYC or MLL amplification in dmin or hsr were women older than 50 years, had de novo AML with complex karyotypes and had a short survival. But, this case had no chromosomal aberration besides c-MYC amplification and showed a good response to chemotherapy with a complete remission for 21 months.

Abstract:
The filling of channels in porous media with particles of a material can be interpreted in a first approximation as a packing of spheres in cylindrical recipients. Numerous studies on micro- and nanoscopic scales show that they are, as a rule, not ideal cylinders. In this paper, the channels, which have an irregular shape and a circular cross-section, as well as the packing algorithms are investigated. Five patterns of channel shapes are detected to represent any irregular porous structures. A novel heuristic packing algorithm for monosized spheres and different irregularities is proposed. It begins with an initial configuration based on an fcc unit cell and the subsequent densification of the obtained structure by shaking and gravity procedures. A verification of the algorithm was carried out for nine sinusoidal axisymmetric channels with different Dmin/Dmax ratio by MATLAB® simulations, reaching a packing fraction of at least 0.67 (for sphere diameters of 5%Dmin or less), superior to a random close packing density. The maximum packing fraction was 73.01% for a channel with a ratio of Dmin/Dmax = 0.1 and a sphere size of 5%Dmin. For sphere diameters of 50%Dmin or larger, it was possible to increase the packing factor after applying shaking and gravity movements.

The Doctor of Ministry is a professional degree accredited by the Association of Theological Schools. As delineated by ATS, the theological program requires to meet specific learning outcomes in a minimum of 30 credit hours with a culminating project that contributes to the understanding of ministry practice. Practical theology is a discipline that seeks to take “both practice and theology seriously”. As a consequence, the DMin can be generally conceptualized as practical theology. However, this paper demonstrates a number of the specific ways in which this general claim can be substantiated. It does this with reference to a number of theoretical discussions within practical theology as to the discipline’s nature. It then examines the implication of this for the status of the DMin, student learning, program design, and the nature of the DMin project.

Abstract:
Within the ultrarelativistic quantum molecular dynamics (UrQMD) model, the effect of initial density fluctuations on cumulants of the net-proton multiplicity distribution in Au + Au Collisions at sNN = 7.7 GeV was investigated by varying the minimum distance dmin between two nucleons in the initialization. It was found that the initial density fluctuations increased with the decrease of dmin from 1.6 fm to 1.0 fm, and the influence of dmin on the magnitude of the net-proton number fluctuation in a narrow pseudorapidity window (Δη≤ 4) was negligible even if it indeed affected the density evolution during the collision. At a broad pseudorapidity window (Δη≥ 4), the cumulant ratios were enlarged when the initial density fluctuations were increased with the smaller value of dmin, and this enhancement was comparable to that observed in the presence of the nuclear mean-field potential. Moreover, the enhanced cumulants were more evident in collisions with a larger impact parameter. The present work demonstrates that the fingerprint of the initial density fluctuations on the cumulants in a broad pseudorapidity window is clearly visible, while it is not obvious as the pseudorapidity window becomes narrow.

Dependence of the binding energy of carbon atoms in nanotubes on the tube diameter is studied. The full-electron orbital free modeling method, developed by us in the framework of the density functional theory, was used for calculation of the binding energy. Nanotubes of limited lengths with the armchair ends were investigated. The tube diameter D, was varied from 0,68 nm up to 1,50 nm; numbers of included atoms were changed from 80 up to 320. Three sets of tubes were studied: the tube length was 0,87 nm in the first set, 1,36 nm in second set, and 1,86 nm in the third set. For the first set the energy minimum (-7.50 eV) was found at Dmin = 1,22 nm, for the second set (-7.62 eV) at Dmin = 1.00 nm, and for the third set (-8.01 eV) at Dmin = 1.06 eV.

Abstract:
Double minute chromosomes (dmins) are a form of gene amplification presenting as small spherical paired chromatin bodies. Dmins are rare in hematologic malignancies and are generally associated with a poor prognosis. Some case reports identified MYC or MLL gene amplification performing as dmin in myeloid neoplasms. FLT3 (FMS-related tyrosine kinase 3) acts as an oncogene in myeloid neoplasms which is associated with several signal transduction pathways. Genomic amplification of FLT3 has not been reported in hematological disease. The current study attempts to demonstrate the existence of double minute chromosomes via FLT3 gene amplification in a patient diagnosed with chronic myelomonocytic leukemia (CMML). Routine G-banded karyotype, array-based comparative genomic hybridization, and fluorescence in situ hybridization analyses were used to characterize the cytogenetic abnormality in the patient’s bone marrow. FLT3 amplification as dmins in a patient with CMML was revealed. This case study reports a rare double minute chromosome via FLT3 amplification in CMML by using array-based comparative genomic hybridization and fluorescence in situ hybridization analyses. The study also proposed another possible mechanism of FLT3 genes in leukemogenesis.

Abstract:
One of the major goals of this thesis is to investigate the extent to which correspondence noise, (i.e., the false pairing of dots in adjacent frames) limits motion detection performance in random dot kinematograms (RDKs). The performance measures of interest are Dmax and Dmin i.e., the largest and smallest inter-frame dot displacement, respectively, for which motion can be reliably detected. Dmax and threshold coherence (i.e., the smallest proportion of dots that must be moved between frames for motion to be reliably detected) in RDKs are known to be affected by false pairing or correspondence noise. Here the roles of correspondence noise and receptive field geometry in limiting performance are investigated. The range of Dmax observed in the literature is consistent with the current information-limit based interpretation. Dmin is interpreted in the light of correspondence noise and under-sampling. Based on the psychophysical experiments performed in the early parts of the dissertation, a model for correspondence noise based on the principle of receptive field scaling is developed for Dmax. Model simulations provide a good account of psychophysically estimated Dmax over a range of stimulus parameters, showing that correspondence noise and receptive field geometry have a major influence on displacement thresholds.

Abstract:
While spatial determinants of emmetropization have been examined extensively in animal models and spatial processing of human myopes has also been studied, there have been few studies investigating temporal aspects of emmetropization and temporal processing in human myopia. The influence of temporal light modulation on eye growth and refractive compensation has been observed in animal models and there is evidence of temporal visual processing deficits in individuals with high myopia or other pathologies. Given this, the aims of this work were to examine the relationships between myopia (i.e. degree of myopia and progression status) and temporal visual performance and to consider any temporal processing deficits in terms of the parallel retinocortical pathways. Three psychophysical studies investigating temporal processing performance were conducted in young adult myopes and non-myopes: (1) backward visual masking, (2) dot motion perception and (3) phantom contour. For each experiment there were approximately 30 young emmetropes, 30 low myopes (myopia less than 5 D) and 30 high myopes (5 to 12 D). In the backward visual masking experiment, myopes were also classified according to their progression status (30 stable myopes and 30 progressing myopes). The first study was based on the observation that the visibility of a target is reduced by a second target, termed the mask, presented quickly after the first target. Myopes were more affected by the mask when the task was biased towards the magnocellular pathway; myopes had a 25% mean reduction in performance compared with emmetropes. However, there was no difference in the effect of the mask when the task was biased towards the parvocellular system. For all test conditions, there was no significant correlation between backward visual masking task performance and either the degree of myopia or myopia progression status. The dot motion perception study measured detection thresholds for the minimum displacement of moving dots, the maximum displacement of moving dots and degree of motion coherence required to correctly determine the direction of motion. The visual processing of these tasks is dominated by the magnocellular pathway. Compared with emmetropes, high myopes had reduced ability to detect the minimum displacement of moving dots for stimuli presented at the fovea (20% higher mean threshold) and possibly at the inferior nasal retina. The minimum displacement threshold was significantly and positively correlated to myopia magnitude and axial length, and significantly and negatively correlated with retinal thickness for the inferior nasal retina. The performance of emmetropes and myopes for all the other dot motion perception tasks were similar. In the phantom contour study, the highest temporal frequency of the flickering phantom pattern at which the contour was visible was determined. Myopes had significantly lower flicker detection limits (21.8 ± 7.1 Hz) than emmetropes (25.6 ± 8.8 Hz) for tasks biased towards the magnocellular pathway for both high (99%) and low (5%) contrast stimuli. There was no difference in flicker limits for a phantom contour task biased towards the parvocellular pathway. For all phantom contour tasks, there was no significant correlation between flicker detection thresholds and magnitude of myopia. Of the psychophysical temporal tasks studied here those primarily involving processing by the magnocellular pathway revealed differences in performance of the refractive error groups. While there are a number of interpretations for this data, this suggests that there may be a temporal processing deficit in some myopes that is selective for the magnocellular system. The minimum displacement dot motion perception task appears the most sensitive test, of those studied, for investigating changes in visual temporal processing in myopia. Data from the visual masking and phantom contour tasks suggest that the alterations to temporal processing occur at an early stage of myopia development. In addition, the link between increased minimum displacement threshold and decreasing retinal thickness suggests that there is a retinal component to the observed modifications in temporal processing.

Abstract:

Soils have the capacity to retain metals due to the occurrence of the phenomenon of sorption, which is the accumulation of ions or molecules on the surface of organic and mineral colloids. For their high organic matter content, therefore large amount of organic colloids, the Histosols have high adsorptive capacity. Sorption can be described by means of different models, which consist in equations that seek to describe the phenomenon with minimal loss of information. Because each model is based on different assumptions, it is essential to select the best suited to the conditions to be analyzed, thus avoiding possible contamination of the environment caused by errors in predicting the adsorption capacity of the soil. This study aimed to select the models that best describe the sorption of Zn, Cd, Cu and Pb on Histosols. The study was conducted by an adaptation of the Batch of Lab Batch, with increasing concentrations of ZnCl2, CdCl2, Pb(NO3)2 and CuCl2. After determination of the concentration of adsorbed metals, the metal levels involved in complex internal and external sphere were evalueted through by desorption studies. The parameters of the sorption isotherms were determined by the ISOFIT program. To compare the fit of the models, the correlation coefficient, determination coefficient, the Akaike Information Criterion Corrected (AICc), AICc Range (?AICc) and the Weighting of AICc (AICcw) were analyzed. It was observed that the linear model was the best to describe the sorption of Zn, while Langmuir was the best to describe the sorption of Cd and Pb and the Freundlich showed better adjustment to the sorption of Cu. In general, the concentrations of N and fulvic acids were the most important factors that influences the sorption of metals. Most of Zn, Cd and Pb is sorbed in electrostatics bonds, easily reversible. However, Cu in strong and almost irreversible bonds.

Abstract:
Compounds of the form R2Sn(dmit) (H2dmit = dimercaptoisotrithione) (R = Me, Et, Bu, Oct, Ph, o-MeOC6H4. MeO2CCH2CH2 and PriO2CCH2CH2) and RR'Sn(dmit) (R = Ph, R' = Me) were synthesised and characterised. Five of the compounds were further investigated using solid state 13C and 119Sn nmr and Mossbauer spectroscopy. The crystal structure of the compounds Et2Sn(dmit), (MeO2CCH2CH2)2Sn(dmit) and PhMeSn(dmit) were determined, and attempts were made to correlate these structures with the solid state nmr data. The addition of a halide ion to the tin atom in the compounds R2Sn(dmit) to give the anion [R2Sn(dmit)X]- (X = halogen) by reaction with a suitable onium halide was investigated. Substitution of the halide, X, in these [Y][R2Sn(dmit)X] (Y = onium cation) compounds was investigated using alkali metal halides and pseudohalides. The crystal structures of the compounds [1,4-dimethylpyridinium][Ph2Sn(dmit)NCS] and [1,4-dimethylpyridinium][Ph2Sn(dmit)I] were determined. The monoestertin bis-dmit complex [NEt4][MeO2CCH2CH2Sn(dmit)2], and the tin tris-dmit complexes. [NR4]2[Sn(dmit)3] (NR4 = NEt4 and 1,4- dimethylpyridinium), were prepared.. The crystal structures of [NEt4][MeO2CCH2CH2Sn(dmit)2] and [NEt4]2[Sn(dmit)3] were determined. A range of lead-dmit complexes, namely PbII(dmit), Ph2Pb(dmit), (Ph3Pb)2dmit and [NR4][Ph2(dmit)I] (NR4 = NEt4 and 1,4-dimethylpyridinium) was also synthesised. The crystal structure of (Ph3Pb)2dmit was determined, and cyclic voltammetry studies were carried out on Ph2Pb(dmit) and (Ph3Pb)2dmit.

Abstract:

Abstract:
During the process of petroleum production and transportation, equipment such as pumps and chokes will cause shear effects which break the dispersed droplets into smaller size. The smaller droplets will influence the separator process significantly and the droplet size distribution has become a critical criterion for separator design. In order to have a better understanding of the separation efficiency, estimation of the dispersed-phase droplet size distribution is very important. The objective of this paper is to qualitatively and quantitatively investigate the effect of shear imparted on oil-water flow by centrifugal pump. This paper presents available published models for the calculation of droplet size distribution caused by different production equipment. Also detailed experimental data for droplet size distribution downstream of a centrifugal pump are presented. Rosin-Rammler and Log-Normal Distributions utilizing dmax Pereyra (2011) model as well as dmin Kouba (2003) model are used in order to evaluate the best fit distribution function to simulate the cumulative droplet size distribution. The results confirm that applying dmax Pereyra (2011) model leads to Rosin-Rammler distribution is much closer to the experimental data for low shear conditions, while the Log-Normal distribution shows better performance for higher shear rates. Furthermore, the predictions of Modified Kouba (2003) dmin model show good results for predicting the droplet distribution in centrifugal pump, and even better predictions under various ranges of experiments are achieved with manipulating cumulative percentage at minimum droplet diameter F(Dmin).

Abstract:
Abstract Formation of voids is inevitable in plasma sprayed coatings, and the role of voids on coating properties has long been established. In fact, the void content within coatings is often adjusted by manipulating the process parameters to obtain coatings with desirable performance. Quantification of voids via image analysis allows the determination of not only the void content within a coating, but also the spatial distribution of the voids. Void content in plasma sprayed neodymium iron boron (Nd-Fe-B) coatings was varied from 1.8 to 8.2% by changing the standoff distance (SOD). Spatial distribution parameters, including near-neighbor distance (dmin), nearest-neighbor distance (dmean), and nearest-neighbor angle (θn), were determined via the Dirichlet tessellation method. Coefficient of variation (COV) values of dmin and dmean allow the determination of inhomogeneity and degree of clustering of the voids within a coating. The θn values show the anisotropy behavior of the voids within plasma sprayed coatings. The influence of void content and its spatial distribution within the coatings on the microhardness and elastic modulus of the coatings was determined.
