Abstract:
Liver tissue samples were reviewed from 35 Doberman Pinschers with chronic active hepatitis in the precirrhotic stage. Thirty dogs had elevated hepatic copper concentrations, and five had normal liver copper concentrations. The earliest changes were inflammation and scar tissue deposition around the small hepatic vein branches. There was also apoptosis of scattered hepatocytes in zone 3. Inflammation consisted of macrophages, lymphocytes, and plasma cells. As the disease progressed, collagen deposition increased around the hepatic veins; in some liver specimens, thin scar tissue septa radiated from the hepatic vein branches, and inflammation spread to include the portal tracts. The sinusoids adjacent to the scar tissue were converted to endothelial-lined, thin-walled vessels. Chronic active hepatitis (commonly referred to as Doberman hepatitis or chronic active hepatitis of Dobermans) is a progressive fibrosis, inflammation and hepatocyte loss beginning among zone 3 hepatocytes around the terminal hepatic vein branches. The histomorphologic changes were the same among those Dobermans with elevated hepatic copper and those with normal hepatic copper. The cause was not determined, but these morphologic studies support the idea of immune-mediated disease.

Abstract:
Ligneous conjunctivitis (LC) was diagnosed in four unrelated Doberman pinschers. Thick, opaque membranes of the palpebral conjunctivae and nictitating membranes were present bilaterally. Three dogs had concurrent signs of multisystemic disease. A thick, amorphous, eosinophilic, hyaline-like material in the substantia propria of the conjunctiva--containing a moderate, mononuclear cell infiltrate--was evident on histological examination. A predominance of T lymphocytes, few macrophages, and weak positive staining for immunoglobulin G (IgG) and immunoglobulin A (IgA) were evident by immunohistochemical staining. The clinical and histological appearance of LC in Doberman pinschers and humans is similar.

Abstract:
von Willebrand disease (vWD) is the most common inherited coagulopathy in dogs, particularly in Doberman Pinschers. We developed a pyrosequencing-based assay to estimate the frequency of the c.7437G>A mutation associated with vWD type 1 in the Doberman Pinscher population of Buenos Aires, Argentina. We found a 0.41 frequency for the mutated allele, which varied significantly within families (family 1 = 0.43, family 2 = 0.58, unrelated animals = 0.35). The use of a popular founder male carrier of mutant allele A increased vWD incidence within a family and in the general population. The mode of inheritance was confirmed as autosomal dominant with incomplete penetrance. No differences were found between sexes and coat colors. Pyrosequencing was a good complement to clinical and coagulation tests for vWD type 1 diagnosis and a useful alternative for detecting the c.7437G>A mutation.

SummaryTwo dogs, both Doberman n pinschers, developed ischiatic nerve palsy following femoral head and neck excisions. Each had undergone surgery entailing the incorporation of a muscle flap (in one case deep gluteal, in the other biceps femoris) to reduce bone-on-bone contact after ostectomy. At later exploratory surgery, the ischiatic nerve was found to be entrapped by muscle in the ischiatic fossa, caudal to the greater trochanter. Partial excision of the ischium relieved the compression, allowing rapid recovery of function. The complication of ischiatic nerve entrapment may warrant consideration when contemplating femoral head and neck excision in cases in which the greater trochanter and ischial tuberosity are abnormally close.Ischiatic nerve palsy developed several weeks after femoral head and neck excision in two Dobermann pinschers. Both dogs had undergone surgery involving muscle sling placement. The ischiatic nerve was found to be entrapped in muscle in each case. Resection of portions of the ischium allowed rapid recovery of function.

Abstract:
This study tested the hypothesis that levothyroxine supplementation increases plasma von Willebrand factor (vWf) concentration and enhances vWf function. The effects of levothyroxine administration were evaluated in 8 euthyroid Doberman Pinschers with plasma vWf concentration <30%. Levothyroxine (0.04mg/kg PO q12hours) and placebo were administered for 30 days in a 2-period, 2-treatment, double-blinded, crossover design with a 30-day washout period between treatments. Buccal mucosal bleeding time (BMBT), vWf antigen concentration (vWf:Ag), vWf collagen binding activity (vWf:CBA), Factor VIII coagulant activity (FVIII:C), serum total thyroxine (T4), free thyroxine (fT4), 3,5,3â -triiodothyronine (T3), and thyroid stimulating hormone were measured on days 0, 2, and 30 of each treatment period. The dogs had markedly low plasma vWf:Ag concentrations (mean 8.9%; reference range 70-180%) and vWf:CBA (mean 11.1%; reference range >70%). All dogs had FVIII:C activity within reference range. The response to placebo versus active levothyroxine treatment revealed no significant differences between groups at any time for BMBT, vWf:Ag, vWf:CBA, and FVIII:C. Serum total thyroxine, fT4, and T3 were significantly higher in the levothyroxine-treated group compared to the placebo group at days 2 and 30. Thyroid stimulating hormone was significantly lower in the levothyroxine-treated group compared to the placebo group at days 2 and 30. Levothyroxine (0.04mg/kg) caused laboratory evidence of hyperthyroidism but did not affect plasma FVIII:C and vWf:Ag concentration or the vWf-dependent functional parameters of collagen binding and BMBT. The results of this study do not reveal a direct action of levothyroxine supplementation on plasma vWf concentration or activity in euthyroid Doberman Pinschers.
