The first, explored with the discussion of Ubi Caritas and the (2010) String Quartet, concerns the interpretation of harmony; that is harmony, plainly being the vertical component in music but having an inbuilt propensity for horizontal movement, including line and counterpoint. In echoes of Poems & Prose, there is a disregard for any horizontal reasoning, harmony is constrained to the point of isolation and focus fundamentally shifts to the chord as 'object'. I consider this 'objective' sense in detail, in subsequent music in this portfolio. A second theme hinges on a discussion of 'musical material' (the term devised by Theodor Adorno); this considered alongside Samuel Beckett's description of a relationship, between 'mess and confusion' (Beckett's terms for material) and the 'form' that contains it. In Echoes of Poems & Prose, I consider material explicitly, in particular the singular sound. With Fountains and Stato di Cambiamento control of the sounds and their overall architecture become increasingly obscure, with issues around form, substantively re-defining the compositional process. A third theme is the consideration of aspects of structure, which become of particular significance in the final pieces Triptych and Divergenza (the term 'structure' being as defined by John Cage). In Triptych, exploration is made of a confining form into which structural material grows; material that yields intensely colourful musical moments. In the final piece Divergenza, the Fibonacci sequence applies a vice-like grip on the material, but as I remove the conceptual dependence on this sequence, the music's intrinsic characteristics of rhythm and character grow to become of central importance.

Three antimicrobial peptides, maculatin 1.1, uperin 3.6 and caerin 4.1 have been isolated from the respective skin glands of the Australian amphibians Litoria genimaculata, Uperoleia mjobergii and Litoria caerulea. To gain a deeper insight into their mechanisms of action, three dimensional structural studies have been conducted using circular dichroism, two-dimensional nuclear resonance and computer modelling techniques. The role of central flexibility within antibiotic peptides in their interaction with bacterial membranes is also discussed.

Abstract:

There has been a significant increase in the numbers of students undertaking doctoral study over the last 20-30 years. This means that the PhD is no longer solely an elite degree designed to prepare graduates for an academic career. Instead, emphasis has increasingly been placed on the role of the PhD in producing 'advanced knowledge workers' who are expected to make a contribution to the production of knowledge in a knowledge-driven economy. This has led to an increased focus on the educational dimension of the PhD and the educational processes involved in developing students to become researchers in a range of contexts. However, the educational processes involved in the PhD are complex and differ across higher education systems, institutions and disciplines. They include formal and informal activities and involve a large number of actors with different expectations about the aims and outcomes. This study aims to gain an in-depth understanding of the educational processes of the PhD by exploring the complexity underlying these processes. The research was based on case studies in six departments. The case studies focused on the PhD processes of the six departments from three disciplines in four higher education institutions in England. Interviews with PhD Programme Directors, supervisors and students were complemented by analysis of institutional and departmental documents. The research was guided by a multi-level framework to examine the institutional, departmental, interpersonal and individual levels, and the inter-relatedness between levels. As such rich narratives provide insight into factors such as the PhD thesis and its influence on the supervisory relationship, formal initiatives such as assessment and coursework, and the Skills Training Programme and its underlying notion of employability. Three forms of complexity were identified. The first relates to the fact that the educational processes are individualistic in nature, and there is a need to understand the influences of the personal, social, educational and professional domains of the individual students and supervisors independently and collectively in shaping these complex processes. The second underlines the tensions and potential contradictions within and between actors as a result of the interpretation and implementation of these processes across the four levels. The third concerns a tension between the need for these processes to remain individualistic and the pressure for departments and institutions to provide standardised processes for all students. By identifying and gaining a greater understanding of these complexities, this research contributes to a more comprehensive understanding of the educational processes of the PhD based on grounded empirical evidence. This understanding is important in developments for enhancing the quality of PhD education, and in developing programmes which support students to become researchers in a range of different employment contexts.

Abstract:

The focus of this research is on the development of ontology and on a more effective way to measure corporate reputation that takes into consideration the orientations of a company’s various stakeholders. The focus by researchers and by practitioners on corporate reputation and on its management attests to an expanding interest. Yet there remains disparate knowledge about how corporate reputation should be defined; about what are its key components; about the relationships between those components and about how corporate reputation should be measured. This point to a need for clarification: to develop a methodology based on ontology of corporate reputation that has relevance for a company’s various stakeholder groups. This research builds on a review of the academic literature and employs text analysis, the nominal group technique and a quantitative survey among stakeholders about the reputation of a high profile company. Theory-driven analysis provides insights into the corporate reputation construct and into a tool for measurement that takes into consideration stakeholder perceptions of a company’s reputation. The results of the study indicate that, in the eyes of its stakeholders, a company’s reputation is driven by nine factors: image, identity, management leadership, performance, corporate brand, products and services, financial performance, ethical management and leadership, and corporate leadership. Not all nine components share the same degree of relevance for stakeholders: different stakeholder groups rank the importance of the components of corporate reputation differently; they evaluate the reputation of the same company differently. The drivers of stakeholders’ overall evaluations of a company’s reputation vary by stakeholder segment. Stakeholder groups are seen to display the characteristics of segments.

Abstract:

Firstly, the effect of pectin on acid milk gels in concentrated, quiescent systems was investigated by passive microrheology using two complementary techniques: diffusive wave spectroscopy (DWS) and multiple particle tracking (MPT). DWS, by allowing probing the mechanical properties of the network at high frequency, gave information on its microstructure. The addition of high methoxyl pectins was shown to change the network structure which has been explained by bridging of the casein micelles by the polymer as the system was undergoing acidification. On the other hand, the presence of low methoxyl pectin in the acid milk gel was shown to have no effect on the microstructure of the network at low concentration of polymer (0.1%w/w) which has been attributed to the sensitivity of this low DM pectin to calcium: LM pectin are trapped by calcium and not able to interact with casein micelles anymore. Multiple particle tracking was used to probe the effect of pectin on the heterogeneity of the system by following the distribution of the displacements of added micro beads at a given time lag during the gelation using the Van Hove distribution. Furthermore, the surface chemistry of the probes was modified in an attempt to control their location in the system. Finally, the mean square displacements of the casein micelles obtained by DWS and, of k-casein coated particles obtained by MPT were shown to give good agreement for the same acid milk system. Having established that the interaction between casein micelles and low methoxyl pectin is prevented by the pectin sensitivity to calcium, the effect of the pectin fine structure was investigated on the interaction between k-casein and pectin by surface plasmon resonance (SPR). The amount of pectin binding on a k-casein coated gold surface was shown to be strongly dependant on the pectin fine structure. It was concluded that small negative patches on the pectin backbone, likely to comprise of around two consecutive unmethylesterified galacturonic acid, are the most effective for pectin binding to k-casein. The effect of the direct interaction between pectin and k- casein on ‘calcium-free casein micelle mimics’ in pectin solution was then investigated using coated latex beads. A pectin structure with a limited number of negative patches on its backbone was also shown to limit the potential for destabilization via bridging.

Abstract:

This thesis concerns the metaphysics of scale. It investigates the implications of a physical determinable being dimensionful. In particular, it considers the case study of mass, as it features within Newtonian Gravity. Nevertheless, most of the terminology, methodology and arguments developed should be relatively straightforwardly applicable to other determinables and theories. Weak Absolutism about mass holds that mass ratios obtain in virtue of absolute masses. Weak Comparativism denies this. In the first five chapters I argue in favour of Weak Absolutism over Weak Comparativism. The sixth chapter argues against reducing mass to other non-mass facts. The overall conclusion is Strong Absolutism about mass within Newtonian Gravity: mass ratios obtain in virtue of absolute masses, which themselves are fundamental (i.e. they do not require anything further in order to obtain). Comparativism promises to recover all the virtues of absolutism, in particular its empirical adequacy, but at a lower 'metaphysical cost'. Special attention is given to Dasgupta's recent comparativist proposal. Dasgupta interprets the requirement of empirical adequacy in terms of the undetectability of the absolute mass scale. I argue that undetectability is an unsuitable way of understanding empirical adequacy and that we would do better to understand it in terms of a theory's ability to correctly generate the set of empirically possible worlds (or at least the actual world). I refute Dasgupta's comparativism both on my terms and on his own terms. I subsequently develop and strongly criticise alternative versions of comparativism. Chapter five sheds doubt on the supposed 'metaphysical parsimony' of comparativism. This debate should be of particular interest to readers who engage with the substantivalism-relationalism debate. These debates are much more entwined than previously acknowledged, which provides a significant source of mutual inspiration, although I do also draw out some important disanalogies.

Abstract:

The increasing availability of molecular data means that phylogenetic studies nowadays often use datasets which combine a large number of loci for many different species. This leads to a trade-off. On the one hand more complex models are preferred to account for heterogeneity in evolutionary processes. On the other hand simple models that can answer biological questions of interest that are easy to interpret and can be computed in reasonable time are favoured. This thesis focuses on four cases of phylogenetic analysis which arise from this conflict. - It is shown that edge weight estimates can be non-identifiable if the data are simulated under a mixture model. Even if the underlying process is known the estimation and interpretation may be difficult due to the high variance of the parameters of interest. - Partition models are commonly used to account for heterogeneity in data sets. Novel methods are presented here which allow grouping of genes under similar evolutionary constraints. A data set, containing 14 genes of the chloroplast from 19 anciently diverged species is used to find groups of co-evolving genes. The prospects and limitations of such methods are discussed. - Penalised likelihood estimation is a useful tool for improving the performance of models and allowing for variable selection. A novel approach is presented that uses pairwise dissimilarities to visualise the data as a network. It is further shown how penalised likelihood can be used to decrease the variance of parameter estimates for mixture and partition models, allowing a more reliable analysis. Estimates for the variance and the expected number of parameters of penalised likelihood estimates are derived. - Tree shape statistics are used to describe speciation events in macroevolution. A new tree shape statistic is introduced and the biases of different cluster methods on tree shape statistics are discussed.
