no<br>Cryptolepine, a naturally occurring indoloquinoline alkaloid used as an antimalarial drug in Central and Western Africa, has been found to bind to DNA in a formerly unknown intercalation mode. Evidence from competition dialysis assays demonstrates that cryptolepine is able to bind CG-rich sequences containing nonalternating CC sites. Here we show that cryptolepine interacts with the CC sites of the DNA fragment d(CCTAGG)2 in a base-stacking intercalation mode.

Environmental signals such as photoperiod and temperature provide plants with seasonal information, allowing them to time flowering to occur in favourable conditions. Most ecotypes of the model plant Arabidopsis thaliana flower earlier in long photoperiods and after prolonged exposure to cold (vernalization). The vernalized state is stable through mitosis, but is not transmitted to progeny, suggesting that the vernalization signal may be transmitted via a modification of DNA such as cytosine methylation. The role of methylation in the vernalization response is investigated in this thesis.

To test whether araC interfered with NGF signalling, I examined the effect of araC treatment on signalling pathways both before and during apoptosis. Using a solid-phase kinase assay and antibodies to active forms of signalling proteins, I found that treatment of neurons with araC induced apoptosis without affecting NGF-maintained signalling. AraC caused no reduction in the activity of either mitogen activated protein kinase/extracellular signal-regulated kinase (MAPK/ERK) or protein kinase B/Akt, a kinase implicated in NGF-mediated survival. Two kinds of affinity chromatography, CDP-choline- and CTP-Sepharose 4B, were investigated for purification of the cytosolic CTP:phosphocholine cytidylyltransferase from rat liver. The enzyme did not show strong affinity for the CDP-choline Sepharose resin, but bound to the CTP-Sepharose column in the presence of 14 mM magnesium acetate.
