Abstract:
Irreversible electroporation (IRE) is a nonthermal tumor/cell ablation technique in which a series of high-voltage short pulses are used. As a new approach, we aimed to investigate the rupture of giant unilamellar vesicles (GUVs) using the IRE technique under different osmotic pressures (Π), and estimated the membrane tension due to Π. Two categories of GUVs were used in this study. One was prepared with a mixture of dioleoylphosphatidylglycerol (DOPG), dioleoylphosphatidylcholine (DOPC) and cholesterol (chol) for obtaining more biological relevance while other with a mixture of DOPG and DOPC, with specific molar ratios. We determined the rate constant (kp) of rupture of DOPG/DOPC/chol (46/39/15)-GUVs and DOPG/DOPC (40/60)-GUVs induced by constant electric tension (σc) under different Π. The σc dependent kp values were fitted with a theoretical equation, and the corresponding membrane tension (σoseq) at swelling equilibrium under Π was estimated. The estimated membrane tension agreed well with the theoretical calculation within the experimental error. Interestingly, the values of σoseq were almost same for both types of synthesized GUVs under same osmotic pressure. We also examined the sucrose leakage, due to large osmotic pressure-induced pore formation, from the inside of DOPG/DOPC/chol(46/39/15)-GUVs. The estimated membrane tension due to large Π at which sucrose leaked out was very similar to the electric tension at which GUVs were ruptured without Π. We explained the σc and Π induced pore formation in the lipid membranes of GUVs.

Abstract:
The membrane translocation efficiency of cell penetrating peptides (CPPs) has been largely studied, and poly-arginines have been highlighted as particularly active CPPs, especially upon negatively charged membranes. Here we inquire about the influence of membrane mechanical properties in poly-arginine adsorption, penetration and translocation, as well as the subsequent effect on the host membrane. For this, we selected anionic membranes exhibiting different rigidity and fluidity, and exposed them to the nona-arginine KR9C. Three different membrane compositions were investigated, all of them having 50% of the anionic lipid 1,2-dioleoyl-sn-glycero-3-phospho-(1’-rac-glycerol) (DOPG), thus, ensuring a high affinity of the peptide for membrane surfaces. The remaining 50% was a saturated PC (1,2-dipalmitoyl-sn-glycero-3-phosphocholine, DPPC), an unsaturated PC (1,2-dioleoyl-sn-glycero-3-phosphocholine, DOPC) or a mixture of DOPC with cholesterol. Peptide-membrane interactions were studied using four complementary models for membranes: Langmuir monolayers, Large Unilamellar Vesicles, Black Lipid Membranes and Giant Unilamellar Vesicles. The patterns of interaction of KR9C varied within the different membrane compositions. The peptide strongly adsorbed on membranes with cholesterol, but did not incorporate or translocate them. KR9C stabilized phase segregation in DPPC/DOPG films and promoted vesicle rupture. DOPC/DOPG appeared like the better host for peptide translocation: KR9C adsorbed, inserted and translocated these membranes without breaking them, despite softening was observed.

Abstract:
Different molecular weight polysaccharides of Dendrobium officinale (DOPs) have gradually attracted attention because of their broad biological activities. They, however, remain poorly defined whether their antitumor activity is associated with molecular weight. In this study, the physicochemical, antioxidant, and antitumor properties of DOPs, including the crude polysaccharide (DOP) and its six degradation fractions (DOP1–DOP6) extracted from Dendrobium officinale, were determined. Consequently, DOPs were mainly composed of different ratios of mannose and glucose as follows: 5.15 : 1, 4.62 : 1, 4.19 : 1, 4.46 : 1, 4.32 : 1, 4.29 : 1, and 4.23 : 1, and their molecular weights were significantly different ranging from 652.29 kDa to 11.10 kDa. With the concentration increase of DOPs, the scavenging capacity against OH and DPPH free radicals increased. The antitumor ability of DOPs was different that DOP1–DOP5 (Mw: 176.29 kDa–28.48 kDa) exhibited the best antiproliferation activity than DOP (Mw: 652.29 kDa) and DOP6 (Mw: 11.10 kDa) in HeLa cells rather than PC9, A549, and HepG2 cells. Moreover, it is worth mentioning that DOP1 and DOP5 showed stronger capability on inducing apoptosis of HeLa cells than DOP and DOP6 via the mitochondrial pathway by upregulating the ratio of the Bax/Bal-2 mRNA expression. The results demonstrated that DOPs can be used as the potential natural antioxidant and antitumor products in pharmaceutical industries, and the molecular weight is a crucial influential factor of their antitumor activity that 28.48 kDa–176.29 kDa is a suitable range we may refer to.

Abstract The antibiotic, daptomycin (brand name Cubicin, Cubist Pharmaceuticals) is approved in the US for treatment of complicated skin and skin structure infections caused by gram-positive pathogens (e.g., MRSA, methicillin-resistant Staphylococcus aureus). Recently, this antibiotic has been shown to interfere with prothrombin time (PT) assays, resulting in a prolongation of clotting times and artificial increases in the INR (International Normalized Ratio). The level of interference varies with Daptomycin concentration in the plasma, the brand of prothrombin time reagent as well as the type (normal or abnormal) of patient sample being analyzed. The bactericidal action of the antibiotic is known and is through a depolarization of bacterial membranes in the presence of physiologic calcium (Silverman, et al, Antimicrobial Agents and Chemotherapy, Aug 2003); therefore, daptomycin interference with the PT clotting assay is likely mediated through the phospholipid component of the thromboplastin formulations. To examine the dependence of daptomycin interference for a number of commercial PT formulations, testing was performed with daptomycin spiked samples of normal pooled plasma as well as abnormal samples from oral anticoagulant therapy (OAT) patients. For spiked normal samples, a small degree of prolongation in clotting times was observed with most PT reagents, with one reagent showing a significant prolongation. In OAT samples spiked with Daptomycin, the PT prolongation was greater for all formulations, with the one reagent again showing greater prolongation as for the normals. Ultimately, this variation in response was attributed to differences in phospholipid composition among the reagents, supported by previous studies of daptomycin with neutral and acidic phospholipids in the presence of calcium (D. Jung, et al., Chemistry and Biology, Jul 2004). A four-dimensional experimental design matrix was constructed using a mixture of four synthetic phospholipids (DOPC, DOPS, DOPE and DOPG), and the relationship of daptomycin-PT interference to percent composition of the phospholipids was examined. In both normal and OAT plasmas spiked with daptomycin, the major contributor to PT prolongation was the concentration of DOPG (phosphatidylglycerol) in the lipid mixture, with additional contributions correlated with DOPC (phosphatidylcholine). Because of the ubiquitous use of these phospholipids in commercial PT formulations, this work suggests that daptomycin interference should be assessed for each combination of PT reagent and INR range used.

The extracellular concentration of the Group II human non-pancreatic secreted phospholipase A2 (hnpsPLA2) is elevated in a variety of inflammatory disorders. This enzyme is remarkable because it demonstrates almost zero activity with egg phosphatidylcholine (PC) or synthetic dioleoyl-phosphatidylcholine (DOPC) as substrate, but expresses high activity with the anionic phospholipid dioleoyl-phosphatidylglycerol (DOPG), a feature shared with the Group II enzyme from rat liver. The presence of certain membrane-bound anions can enhance hydrolysis of PC by the mammalian secreted PLA2S. In this study the ability of various non-polar anions to stimulate DOPC hydrolysis by secreted PLA2S has been investigated. The naturally occurring membrane anion, cholesterol sulphate, was particularly effective in stimulating the hydrolysis of both DOPC and also 1-stearoyl-2-arachidonyl phosphatidylcholine by hnpsPLA2. Activation of DOPC hydrolysis was also achieved with dioleoyl-phosphatidylserine (DOPS); however, DOPS was less effective than cholesterol sulphate. In contrast, the dianion dioleoyl-phosphatidic acid, a known activator of pig pancreatic PLA2, failed to activate the human enzyme. It remains to be established whether cell plasma-membrane hydrolysis by extracellular hnpsPLA2 can be activated in vivo by the presence of suitable membrane anions such as cholesterol sulphate and thus promote an inflammatory response within the cell.

Diacyl phosphatidylglycerols (PG) are the major anionic lipids in theEscherichia colimembrane. Short‒chain dihexanoyl phosphatidylglycerol (DHPG) was previously utilized for the structural determination, by NMR spectroscopy, of the peptide corresponding to the N-terminal membrane anchor of the glucose‒specific enzyme IIA (IIAGlc) fromE. coli. This study explores the possible use of lipid micelles of dioctanoyl phosphatidylglycerol (DOPG) and didecanoyl phosphatidylglycerol (DDPG) as alternatives to DHPG. At a peptide concentration of 1 mM, the minimum peptide/lipid molar ratios required for the formation of the lipid‒binding amphipathic helix are approximately 1 :40, 1 :5, and 1 :5 for DHPG, DOPG, and DDPG, respectively. Based on the lipid titration, the critical micelle concentration (CMC) of DHPG was estimated to be ~50 mM. The1H spectral linewidths of the peptide bound to a variety of lipid micelles decrease in the following order: DDPG > DOPG > DHPG. The helical regions of the peptide in different anionic lipids were elucidated based on chemical shift indexes (CSI). Residues Leu2‒Leu9, Leu2‒Val10, and Leu2‒Val10 were found to be helical in DHPG, DOPG, and DDPG, respectively, indicating that the lipid chain length had only a subtle effect on the amphipathic helix of the peptide. In light of the minimum peptide/lipid ratio and the spectral linewidth, and the CSI‒derived peptide structure, DOPG is proposed as a good compromise for structural studies of this membrane‒associated peptide by solution NMR spectroscopy.

Prunus spinosa fruits (PSF) contain different phenolic compounds showing antioxidant and anti-inflammatory activities. Innovative drug delivery systems such as biomimetic nanoparticles could improve the activity of PSF extract by promoting (i) the protection of payload into the lipidic bilayer, (ii) increased accumulation to the diseased tissue due to specific targeting properties, (iii) improved biocompatibility, (iv) low toxicity and increased bioavailability. Using membrane proteins extracted from human monocyte cell line THP-1 cells and a mixture of phospholipids, we formulated two types of PSF-extract-loaded biomimetic vesicles differing from each other for the presence of either 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) or 1,2-dioleoyl-sn-glycero-3-phospho-(1′-rac-glycerol) (DOPG). The biological activity of free extract (PSF), compared to both types of extract-loaded vesicles (PSF-DOPCs and PSF-DOPGs) and empty vesicles (DOPCs and DOPGs), was evaluated in vitro on HUVEC cells. PSF-DOPCs showed preferential incorporation of the extract. When enriched into the nanovesicles, the extract showed a significantly increased anti-inflammatory activity, and a pronounced wound-healing effect (with PSF-DOPCs more efficient than PSF-DOPGs) compared to free PSF. This innovative drug delivery system, combining nutraceutical active ingredients into a biomimetic formulation, represents a possible adjuvant therapy for the treatment of wound healing. This nanoplatform could be useful for the encapsulation/enrichment of other nutraceutical products with short stability and low bioavailability.

Abstract:
Lipid-protein nanodiscs (LPNs) are nanoscaled fragments of a lipid bilayer stabilized in solution by the apolipoprotein or a special membrane scaffold protein (MSP). In this work, the applicability of LPN-based membrane mimetics in the investigation of water-soluble membrane-active peptides was studied. It was shown that a pore-forming antimicrobial peptide arenicin-2 from marine lugworm (charge of +6) disintegrates LPNs containing both zwitterionic phosphatidylcholine (PC) and anionic phosphatidylglycerol (PG) lipids. In contrast, the spider toxin VSTx1 (charge of +3), a modifier of Kv channel gating, effectively binds to the LPNs containing anionic lipids (POPC/DOPG, 3 : 1) and does not cause their disruption. VSTx1 has a lower affinity to LPNs containing zwitterionic lipids (POPC), and it weakly interacts with the protein component of nanodiscs, MSP (charge of -6). The neurotoxin II (NTII, charge of +4) from cobra venom, an inhibitor of the nicotinic acetylcholine receptor, shows a comparatively low affinity to LPNs containing anionic lipids (POPC/DOPG, 3 : 1 or POPC/DOPS, 4 : 1), and it does not bind to LPNs/POPC. The obtained data show that NTII interacts with the LPN/POPC/DOPS surface in several orientations, and that the exchange process among complexes with different topologies proceeds fast on the NMR timescale. Only one of the possible NTII orientations allows for the previously proposed specific interaction between the toxin and the polar head group of phosphatidylserine from the receptor environment (Lesovoy et al., Biophys. J. 2009. V. 97. № 7. P. 2089-2097). These results indicate that LPNs can be used in structural and functional studies of water-soluble membrane-active peptides (probably except pore-forming ones) and in studies of the molecular mechanisms of peptide-membrane interaction.

Abstract:
As a direct consequence of the misuse of antibiotics since its first discovery, bacteria have developed extensive resistance mechanisms making them once again potential lethal threats. This eventuality has triggered a vast research effort from scientists worldwide to find solutions to mitigate antimicrobial resistance. One such option is the identification of new potential antimicrobial agents, like for example antimicrobial peptides (AMPs). Various methods have been applied to evaluate the properties and determine the complex mechanism of AMPs. However, the details of the mechanism remain unknown and hence the work in this project seeks to examine the suitability of using TIR Raman, a vibrational spectroscopy technique which is sufficiently surface sensitive to study the interaction of AMPs in contact with lipid bilayers, which are just a few nanometres thick. In order to evaluate the information that could be extracted from TIR Raman, measurement of solid supported lipid bilayers in the absence of peptides were first carried out. In particular, the lipid DMPC with a phase transition close to room temperature, was measured at various temperatures to determine spectral changes associated with the transition. For the peptide-membrane interactions, the AMP LL37 was put into contact with solid supported lipid bilayers modelling the cell membranes of bacteria (DOPE, DOPG) or humans (DOPC) respectively. The data clearly indicates that the membrane composition, and specifically the lipid head group charge, play an important role in the peptide-membrane interactions. In the bilayers mimicking bacteria cell membranes, the peptide either absorbed onto or inserted into the bilayer. In contrast, for the bilayer modelling a human cell membrane, no significant variations were detected, indicating no interaction with LL37. The findings presented in this work generally coincide with similar research of LL37 using other techniques. At hand of the herein presented data, TIR Raman has proven suitable and effective in detecting effects of antimicrobial peptides in contact with model lipid bilayers, and hence can be recommended for further studies.
Som en direkt följd av missbruket av antibiotika sedan det först upptäcktes, har bakterier utvecklat omfattande resistensmekanismer vilket föranlett dem att återigen utgöra potentiellt dödlig hot. Denna situation har manat fram en väsentlig forskningsinsats från forskare världen över att hitta lösningar för att minska antimikrobiell resistens. Ett sådant alternativ har varit identifieringen av nya potentiella antimikrobiella substanser, så som till exempel antimikrobiella peptider. Ett flertal metoder har använts för att både evaluera peptiders egenskaper och fastställa deras komplexa mekanism. Detta till trots förblir de exakta detaljerna i mekanismen okända, vilket föranlett arbetet i detta projekt att undersöka lämpligheten i att använda TIR Raman, en vibrational-spektroskopisk metod som är tillräckligt ytkänslig för att studera interaktionen hos antimikrobiella peptider i kontakt med lipidmembran, vilka endast är några få nanometer tjocka. För att evaluera informationen som kan utvinnas med TIR Raman, utfördes först mätningar av lipidmembran, skapade på ett solitt underlag, utan tillägg av peptider. Mer noggrant, har lipiden DMPC med en fasövergång nära vid rumstemperatur, mätts vid olika temperaturer för att fastställa spektrala variationer associerade till övergången. För peptid-membran interaktionerna, sattes den antimikrobiella peptiden LL37 i kontakt med lipidmembran som modellerar cellmembranet hos bakterier (DOPE, DOPG) respektive människor (DOPC). Mätdatan indikerar tydligt att membrankompositionen, och där specifikt laddningen av lipidens huvudgrupp, spelar en viktig roll i membran-peptid interaktionerna. För lipidmembranen som imiterar bakteriella cellmembran, adsorberade peptiden till membranet eller integrerades in i det. Till skillnad från detta, kunde inga signifikanta variationer detekteras för lipidmembranet som modellerade ett mänskligt membran vilket indikerar att det inte interagerar med peptiden LL37. Upptäckterna som presenteras i detta arbete sammanfaller generellt med andra, liknande studier där andra instrument använts för att undersöka LL37. Det kan ur materialet som presenterats här utläsas att TIR Raman visat sig lämpligt och effektivt i detekteringen av antimikrobiella peptider i kontakt med modeller av lipidmembran, och kan därav rekommenderas för fortsatta studier.

Abstract:
This thesis presents a novel optical communication technique that provides a second, low data rate channel on an existing high-speed fiber optic link. The second channel is derived using an acousto optic fiber phase modulator and interferometeric receiver. This method modulates the optical phase of the primary high speed optical signal with a low frequency sine wave. At the receiving end of the low speed path, an interferometer and band pass fiber are used to recover the low-speed signal. Information is carried on the low frequency sine wave by use of FSK modulation. The method is noninvasive in that the low-speed channel is derived without electrically, optically or physically affecting the performance of the high-speed optical path. The method is ideal for overlaying network management channels on a fiber network. The thesis includes both analysis and experimental verification of the technique.

Doug and Mike Starn: Evolution from Photography to Public Art investigates the pioneering installations and public art by Doug and Mike Starn, establishes their position within the complete oeuvre and examines the confluence of media they have worked in, while situating the artists and their work within the contemporary art historic context. Intrinsic characteristics of the Starn brothers’ work are the principles of interconnectedness, continuity, duality and change, a continuous evolution combined with a stunning ability to reinvent their work, redefining entire art genres in the process. Identical twins Doug and Mike Starn, born in 1961 and included in the 1987 Whitney Biennial at the age of 26, work collaboratively. The primary medium of photography characterizes their early work, evolving in the 1990s to include artist books, large-scale video projections, and installations. Incidentally, their first permanent public art commission is tied to the reconstruction of New York’s infrastructure following the tragedy of September 11, 2001. The goal is to delineate context, process, and significance of the artists’ foray into public art. An analysis and interpretation of artistic production, context, partnerships, process, scale and reception will reveal the transition from stellar gallery production to installation art and exceptional public art. Highlights of the pair’s stellar career include the completion of two important public art projects in New York City in 2008 and 2010, one permanent and one temporary: the permanent public art environment See it split, see it change on the concourse of South Ferry Subway Station at the tip of Manhattan and the temporary, monumental Big Bambú installation on the Metropolitan Museum’s roof garden. These outstanding works were preceded by years of artistic inquiry and development in the genre of photography, whose path the study explores.
Das Œuvre von Doug und Mike Starn spannt den Bogen von der Fotografie zur Kunst am Bau und zur Kunst im öffentlichen Raum. Zwischen diesen Polen bewegt sich die Entwicklung der Künstlerzwillinge, die sich in ihrer Zusammenarbeit mit den Themen Licht, Kontinuität und Vergänglichkeit beschäftigen. Als junge Künstler entwickelten sie groß-formatige, mit Klebeband zusammengesetzte Foto-Collagen, die bereits 1987 zur Beteiligung am Whitney Biennial führten. Zu den Höhepunkten ihres künstlerischen Schaffens gehören zwei wichtige Projekte im öffentlichen Raum mit Standort New York. See it split, see it change (2008) wurde für die South Ferry Transitstation geschaffen und die monumentale, temporäre Installation Big Bambú (2010) war auf dem Dach des Metropolitan Museum of Art installiert. Ersteres Werk ist die umfangreichste Glasinstallation der New Yorker öffentlichen Verkehrsbetriebe, letzteres die größte Außenausstellung in der Geschichte des Metropolitan Museum. Den bahnbrechenden Arbeiten der Starns gingen zwei Jahrzehnte künstlerischer Entwicklung voraus, deren Zusammenhang mit dem späteren Werk in dieser Studie aufgezeigt wird. Fluxus è stato definito il più radicale e sperimentale movimento artistico degli anni Sessanta. Dalla prima comparsa ad oggi è stato osannato, analizzato, dimenticato e riscoperto molte volte, tuttora però rimane una delle più grandi incognite critiche della storia dell’arte del Novecento. La ricerca si sviluppa secondo uno schema tripartito: indagare origini, ascendenze e ispirazioni; collocare e contestualizzare il periodo di nascita e sviluppo; esaminare influenze e lasciti. Attraverso un confronto di manifesti, scritti autografi e opere si è cercato di verificare punti di contatto e di continuità tra Fluxus e le Avanguardie Storiche, con particolare riferimento a Futurismo e Dadaismo. Successivamente si è cercato di ricostruire le dinamiche che hanno portato, alla fine degli anni Cinquanta, al definirsi di un terreno fertile dal quale sono germinate esperienze strettamente legate quali Happening, Performance Art e lo stesso Fluxus, del quale si sono ripercorsi i cosiddetti “anni eroici” per evidenziarne le caratteristiche salienti. Nella terza sezione sono state individuate diverse ipotesi di continuazione dell’attitudine Fluxus, dal percorso storico-filologico dei precoci tentativi di musealizzazione, alle eredità dirette e indirette sulle generazioni successive di artisti, fino alla individuazione di idee e concetti la cui attualità rende Fluxus un elemento imprescindibile per la comprensione della cultura contemporanea.
Fluxus has been called the most radical and experimental art movement of the sixties. From the first appearance it has been praised, analyzed, forgotten and rediscovered many times, but still remains one of the great critical unknowns of the Art History of the twentieth century. The research develops according to a tripartite scheme: to investigate the origins and the inspirations; to place and contextualize the period of the birth and development; to examine influences and legacies. Through a comparison of manifestos, autograph texts and works has tried to verify points of contact and continuity between Fluxus and the Avant-garde, with particular reference to Futurism and Dadaism. Then the research tried to reconstruct the dynamics that led to the end of the fifties, to be defined as a fertile ground from which germinated experiences - closely related - such as Happenings, Performance Art and Fluxus as well, which it retraced the so-called "heroic years" to highlight the salient features. In the third section, several hypotheses of the Fluxus-attitude have been identified, from the historical-philological path of the early attempts of "musealization", the direct and indirect inheritance on later generations of artists, up to the identification of ideas and concepts whose actuality makes Fluxus an essential element for the understanding of contemporary culture.

In collaboration with HappyTail, a Swedish company developing a mobile application for dog owners, this qualitative research study examines and identifies important factors, for developers or other stakeholders, to have in mind when developing mobile applications that aim to support dog health. According to behavioural scientists, there needs to be a bridge between health themed mobile applications and behavioural change theories in order to achieve desirable results. Therefore, literature on dog health as well as on behavioural and persuasive technologies were reviewed. To fully understand dog owners’ behaviours, empirical data was collected from five in-depth interviews with dog owners recruited from groups on Facebook. The data collected, in combination with the reviewed literature, results in increased understanding of dog owners’ behaviours in relation to their dogs’ health, their sources of motivation and inspiration, and how persuasive technologies can affect their behaviours towards their dogs. It turned out that psychological aspects, such as the relationship between the dog and dog owner, was crucial among the interviewees, and that they prefer to use dog themed mobile applications with features such as social support and tailored content as parts of the design. Finally, ethical aspects that need to be considered when including dogs as stakeholders in new technologies were discussed. The result is valuable for researchers investigating dog owners’ behaviour as well as for those developing new technology, especially mobile applications, with focus on dog health.
I samarbete med HappyTail, ett svenskt företag som utvecklar en mobilapplikation för hundägare, syftar den här kvalitativa undersökningen till att identifiera viktiga faktorer som personer verksamma inom applikationsutveckling med fokus på hundhälsa bör ha i åtanke under utvecklingsskedet. Enligt forskare inom beteendevetenskap är det viktigt att det finns en bro mellan mobilapplikationer med hälsotema och befintliga teorier inom området beteendeförändring för att kunna uppnå önskvärda resultat. Därför har litteratur som berör hundens hälsa samt viktiga faktorer för beteendeförändring i samband med utveckling av ny teknik beaktats. För att få ökad förståelse för hundägares beteende utfördes fem djupgående intervjuer med deltagare rekryterade från hundrelaterade grupper på Facebook. Tillsammans med den granskade litteraturen resulterade den insamlade datan i en djupare inblick i hundägares liv, vilka faktorer som motiverar och inspirerar dem till att ta hand om sin egen såväl som sin hunds hälsa, och hur teknik bör vara utformad för att påverka dem till att ändra beteende gentemot sin hund. Det visade sig bland annat att relationen mellan hund och hundägare var extra viktig för deltagarnas syn på sin hunds hälsa, och att social support och personalisering är grundstenar för en intressant mobilapplikation med hundrelaterat innehåll. Avslutningsvis diskuteras även vilka etiska aspekter som är nödvändiga att beakta när hundar inkluderas i utvecklingen av ny teknik. Undersökningens resultat har ett värde för forskare som specialiserar sig på hundägares beteende samt för utvecklare av ny teknik som strävar efter att förbättra hundars hälsa.

Educa Dog es un proyecto que busca educar mascotas con problemas de conducta, que busca diferenciarse empleando métodos innovadores de obtener la atención del can, además de ofrecer el servicio a domicilio. La idea de negocio se presenta a raíz de la inquietud por encontrar un lugar donde se eduque un cachorro sin que este sea maltratado o sufra estrés, de esta forma se pudo identificar una necesidad que no es cubierta por el mercado actual. Informes realizados por empresas de investigación, demostraron mediante encuestas que solo en Lima el 56% de los hogares tiene mascotas en casa y de este porcentaje el 79% cuenta por lo menos con un can, traduciendo esta cifra a números, se puede decir que 1 millón de hogares en Lima tienen canes, lo que supone una alta demanda en servicios relacionados a mascotas, en este caso en particular para canes. A su vez, el creciente respeto por los animales, la comunidad animalista y los “Dog Lover” propicia el ambiente adecuado para establecer una idea diferenciada, con un potencial mercado interesado en invertir en el aprendizaje de una conducta adecuada de su mascota. Por todo lo mencionado anteriormente, creemos que es necesario cubrir esta serie de requerimientos que han surgido estos últimos años por los criadores de perros, en especial a aquellos que han desarrollado un gran nivel de afinidad por su mascota.
Educa Dog is a project that seeks to educate pets with behavioral problems, which seeks to differentiate using innovative methods of getting the attention of the dog, in addition to offering home delivery. The business idea arises from the concern to find a place where you can educate a puppy without it being mistreated or suffer stress, this way you could identify a need that is not covered by a very substantial market. It was possible to identify that only in Lima 56% of households have pets at home and of this percentage 79%, translating this figure into numbers, it can be said that 1 million households in Lima have dogs, which supposes a high demand in services related to pets, in this case in particular for dogs. In turn, the growing respect for animals, the animal community and the "Dog Lover" propitiates the right environment to establish a differentiated idea, with a potential market interested in investing in the learning of appropriate behavior of your pet. For all the above, we believe it is necessary to cover this series of needs that have emerged in recent years by dog breeders, especially those who have developed a high level of affinity for their pet.

The present research project, called "Dog Care Web Platform", seeks to satisfy the problem of the deficient number of hotels, lodgings and day care for dogs that exist in modern Lima, due to the exponential growth of the number of families that have one or more dogs at home. The service connects via web to a dog owner who wants to host and care for his pet with a host. The target market of the Dog Care web platform is the person residing in modern Lima who have a can and leave on weekends or travel for work or vacation reasons and want to leave their pet under good care. A job title usually implies the responsibility and the rank of a job position. While traditional job title analysis has been focused on studying the responsibilities of job titles, this paper attempts to reveal the rank of job titles. Specifically, we propose to extract job title hierarchy from employees' career trajectories. Along this line, we first quantify the Difficulty of Promotion (DOP) from one job title to another by a monotonic transformation of the length of tenure based on the assumption that a longer tenure usually implies a greater difficulty to be promoted. Then, the difference of two job title ranks is defined as a mapping of the DOP observed from job transitions. A Gaussian Bayesian Network (GBN) is adopted to model the joint distribution of the job title ranks and the DOPs in a career trajectory. Furthermore, a stochastic algorithm is developed for inferring the posterior job title rank by a given collection of DOPs in the GBN. Finally, experiments on more than 20 million job trajectories show that the job title hierarchy can be extracted precisely by the proposed method.

Abstract Shaped charge jet (SCJ) research has long been an active area for industrial, academic, and defense organizations. Traditionally, the depth of penetration (DOP) has been one of the most important metrics for the evaluation of shaped charge jet performance, and early 1D analytical penetration models based on hydrodynamic penetration were created with this metric in mind [1]. As the standoff of a shaped charge jet increases, the DOP reaches a maximum and then begins to decrease. A simple 1D hydrodynamic penetration model must account for the totality of the jet material on axis penetrating, and as a result experimental DOP at longer standoffs is lower than the analytical models predicted. Some analytical models reasoned that since a velocity gradient evolves as a SCJ forms, contributions to penetration from jet material below a minimum jet or penetration velocity should be eliminated. These were better able to account for the difference between analytical hydrodynamic and experimental DOPs [2]. The actual difference between analytical hydrodynamic penetration theory and experimentally recorded values is now regarded to be a result of 3D phenomena including particle tumbling and motion transverse to the jet axis known as lateral drift [2]. The origins of these 3D phenomena have been attributed to sources including variability in the uniformity of the explosive charge or the microstructure of the liner [2,3].

Abstract:
This study was conducted in selected provinces of Bicol Region from April 2015 to May 2015 to describe the knowledge, attitudes and practices (KAP) of dog owners regarding rabies and dog bites. A purposive sampling was used in selecting the respondents of the study. Frequencies were tabulated for all variables. Of the 1,200 respondents, 2,193 dogs were recorded making a 2:1 ratio of dogs to householders in this particular study. Among these dogs 58% were vaccinated against rabies. The majority of the ones taking care of the dogs were female (57.3%) but the primary owner (62.9%) was the head of the family. Only 34.7% of the respondents knew that it is their duty to get their pets vaccinated against rabies. Around one-fourth (20.7%) admitted that someone in their household had been bitten by a dog but most respondents (62.5%) did nothing to the dog. The majority (57.7%) of the bite victims were youths (1-14 years old) and almost all (82.7%) of the wound bites were washed with soap and water. Television (44.9%) was the primary source of knowledge about rabies. The majority of participants (67.3%) said that humans are the main end-hosts that can be infected with rabies. Salivation or drooling (42.7%) and craziness (34.2%) were the main signs cited as behaviour of rabid dogs while craziness (40.2%) and hydrophobia or fear of water (25.4%) were cited for rabid humans. Most (33.9%) do not know the source of rabies but the majority (61.8%) believe that vaccination is the main preventive measure against rabies. The majority of participants (63%) reported that the local ordinances regarding rabies in their locality is about the Local Anti-Rabies Act and almost all (93.2%) admitted that vaccination is the most common anti-rabies program of the government.
