Full textIn vitro diagnostic is now a major actor in the domain of healthcare. There is a need for quick assays able to detect low concentrations of biomarker. Magnetic agglutination assay have been developed in that purpose. Antibodies grafted on superparamagnetic particles allow the formation of aggregates in a few minutes, thanks to the application of a magnetic field. Aggregates are generally observed by a turbidimetric method. First part of that work consisted in understanding the phenomena observed by turbidimetry. An agglutination model was developed, allowing predicting the number of aggregates formed, depending on the biomarker concentration. The model was confronted to experimental results. Experiments showed the effect of the way magnetic field was applied on aggregation, especially when important quantities of analyte were used. Concentrations around one picomolar were detected in less than one minute. In order to detect lower concentrations, an individual detection method, relying on the principles of flow cytometry, was then used. Decreasing of the background noise allowed reaching limits of detection of tens to hundreds of femtomolars. Parameters important to the assay, especially the size of the particles and the intensity of the magnetic field, were studied. This work make possible to detect a large variety of proteins, using the magnetic agglutination technique

Full textHuman plasma contains low molecwar weight growth factors potentiating certain in vitro biological effects of insulin-like growth factors. Recently three peptides were identified with HWESAS, HWE and WGHE as respective structures. Although it is difficult to product monoclonal antibodies directed against peptides characterized by a structure consisting of an amino acid number below 8, this study describes the production of such antibodies for peptides having 6, 4, or 3 residus. The mice immunizations were performed with homologous synthetic peptides coupled to a maleimide activated keyhole limpet hemocyanin and the hybridomas were prepared by standard techniques. By using an inhibition test, the specificity of monoclonal antibodies was deterrninated : they cross-reacted with original peptides as well as derivated peptides only when C terminus was not modified. Concerning the affinity, measured by non competitive enzyme immunoassay, the values range from 0,89 to 3,89 108 M-1. In addition, an ELISA was developped with antibodies directed against HWESAS to measure the serie concentration of this peptide in humans. The levels of peptide observed in healthy subjects were about 2µg/ml, they were dramatically decreased in chronic renal failure but norrnalized after a successful renal transplantation. These findings suggest that the kidney may play an important role in the production and/or metabolism of this hexapeptide. Such monoclonal antibodies will be useful tools for elucidation of low molecular weight growth factor production leading to a better understanding of their physiological and pathological significances

Full textMy work was focused on the monocyte and macrophage lineages. We have shown that Monocyte/macrophage derived from ES cells are cells extremely specialized in tissue remodeling, pro-angiogenesis and immune suppression but with low inflammatory potential. I have investigated the characteristics of monocytes/macrophages from iPS. Those monocyte/macrophage were quite similar to hESC derived, but exhibited more inflammatory potential suggesting some incomplete reprogrammation during derivation of IPS. We hypothesize that the decrease or absence of inflammatory potential of the monocyte/macrophages could be related to a decrease activation of the JAK2/STAT pathway induced by IFN-y and GM-CSF, two cytokines implicated in MI polarization. In this purpose we derive iPS from patient harboring the JAK2V617F mutation, a gain of fonction mutation associated with myéloproliférative neoplasm. In preliminary results, no significant differences were observed in the polarization of JAK2V617F or JAK2WT monocyte/Macrophages derived from IPS. Furthermore we found that IFN-y was capable to normally induce STAT1 activation in these cells suggesting that the blockage in inflammatory response is downstream STAT1 and may be related to epigenetic regulation of inflammatory gens. Finally I have obtained preliminary results showing that JAK2V617F may induce independence to bFGF of the pluripotent iPSC, a result very similar to those reported by Griffiths and al on JAK2V617F mESC who could maintain their pluripotent phenotype without addition of LIF. This was not related to the induction of the canonical STAT pathway but to an effect of nuclear JAK2 on the epigenetic regulation of Nanog

Full textHighly active antiretroviral therapy (HAART) dramatically improves rates of mortality and morbility due to HIV infections. Unfortunately, clinical failures are observed partly due to inefficient cellular concentration of drug enable to suppress the viral load. The determination of intracellular contents of drugs in targets cells should be contributed to optimize the antiviral treatment by a therapeutic drug monitoring (TDM). In this order we developed highly specific and sensitive immunological techniques directed against the Saquinavir (SQV) and against the active metabolite of ddI, the ddATP. The anti-SQV antibodies were characterized using two sensitive immunological methods (ELISA and RIA) allowing the quantification of plasma and cellular contents of HIV patients. For the quantification of ddATP, we developed two immunological approaches. The first one named “indirect” method, use the ddA as hapten. Then, the determination of the ddI phosphorylated metabolites contents are performed after purification and dephosphorylation of biological samples. The second one, named “direct” approach, use the ddATP molecule and an isostère of ddATP as haptens? The isostère molecule was selected on structural and electronic analogies basis between the ddATP and the isostère molecules. The anti-ddATP, the anti-isostere and anti-ddA antibodies were characterized using the same radioactive probe. The specificity study of anti-isostere antibodies allows us to conclude that the substitution of pyrophosphate by citrate moiety is a useful alternative to produce antibodies against phosphorylated molecules. The ddATP radioimmunoassay was used to determine in an in vitro model (peripheral blood mononuclear cells) the intracellular contents of all the phosphorylated forms of ddA (ddAMP, ddADP and ddATP). The ddI plasma content and the dd AMP, ddATP and ddATP intracellular contents were measured in five patients infected by HIV and treated by polythérapie

Full textIn this study, a concept of microfluidic chip with embedded planar coils is designed and fabricated for the aim of trapping effectively functionalized magnetic nanobeads and immobilizing antibody (IgG type). The planar coils as a heart of microfluidic chip is designed with criterion parameters which are optimized from simulation parameters of the maximum magnetic field, low power consumption and high power efficiency by FE method. The characterization of microcoils such as effectively nanobeads (300 nm) at low temperature (<37oC) is performed and confirmed. The channel network in PDMS material is designed for matching with entire process (including mixing and trapping beads) in microfluidic chip. A process of PDMS’s surface modification is also carried out in the assemble step of chip in order to limit the non-specific adsorption of many bio substances on PDMS surface. The microfluidic chip assemble is performed by using some developed techniques of reversible packaging PDMS microfluidic chip (such as stamping technique, using non-adhesive layer, oxygen plasma combining with solvent treatment). These packaging methods are important to reused microchip (specially the bottom substrate) in many times. The immobilization of antibody IgG-type is performed inside microfluidic chip following the standard protocol of bead-based ELISA in micro test tube. The result showed that IgG antibodies are well grafted on the surface of carboxyl-beads (comparing to result of standard protocol); these grafted antibodies are confirmed by coupling them with labeled second antibody (Fab-FITC conjugation)

Full textThis thesis was devoted to the design of fluorescence immunoassays aiming at the detection of biological warfare agents. We developed a new concept of competitive immunoassay for continuous flow detection involving a fluorescence resonance energy transfer (FRET). This procedure is based on the use of an heterotrifunctional reagent (tripod) bearing i) a fluorophore donor (D), ii) a molecule structurally close to the target and iii) a linker to the solid phase. The binding of an anti-target antibody labeled with an acceptor molecule (A) on the tripod generates the decrease of the donor emission via the FRET phenomenon (quenching). The presence of the target competiting with the tripod for the binding to the antibody leads to an increase of the fluorescence signal. The solid phase can be easily further reactivated by adding the labeled antibody. This method was evaluated in microtiter plates using small molecules (substance P, atrazine, aflatoxine) and larger proteins (β-lactoglobuline, botulinum toxin, ricin). Spectral properties of the dyes, A/D ratio and A-D distance were demonstrated to have largely influenced FRET efficiency and hereby the sensitivity of the assay. The sensitivity also strongly depends on the binding characteristics of the antibody. The main difficulty lies on the relative affinity characteristics of the antibody towards the tripod and the analyte, since the antibody should exhibit ideally a strong affinity for the tripod and even stronger affinity for the analyte. Assays performed in glass microcells using a CCD camera and a fluorescence microscope allowed continuous-flow detection and are promising for the build-up of a biosensor. We also present a simple method for performing rapid assays in microtiter plates thanks to the difference in fluorescence signal observed between free and immobilized dyes which proved to be efficient for the design of a simultaneous multiplexed immunoassay
