Os autores que participam do dossiê, brasileiros e estrangeiros, discutem aspectos teóricos e metodológicos da produção de Ruy Mauro e também apropriam-se de suas formulações para pensar a América Latina nos dias de hoje. O dossiê se encerra com a tradução de um texto do próprio Marini, que discute o movimento estudantil na América Latina e que foi originalmente publicado em 1970, na revista Les temps modernes. A organização do dossiê foi feita por Maíra Machado Bichir, Marina Machado Gouvea e Mirella Rocha, a quem agradecemos pelo trabalho.

This magister’s thesis is about modern lighting of vehicles, the visual range and afterglow in poor visibility. The first part of the thesis provides a detailed analysis of the current situation and problems, including a comprehensive description of the sources of light radiation, lamps, structures, and describe the systems used in modern lighting technology of vehicles. In the second part of the thesis there are the scientific methods to determine the issue of visual range and afterglow of the headlights. The second part is about the application of acquired techniques to detect an afterglow and the visual range of vehicles with modern headlights with a light source with xenon lamp, compared with a vehicle with a halogen lamp and a vehicle with an older halogen headlight.

This thesis is focused on comparison of parameters of light sources (halogen, xenon, LED) installed in Skoda Octavia 3rd generation. The opening part solves analysis of the current state of lighting technology and its impact on traffic safety. The next part is focused on general parameters of light, construction of light sources, optical systems and adaptation of the human eye to various light conditions, especially contrast. The practical part is focused on comparing the contrast values of individual light sources in relation to the dark and lightly dressed pedestrian in the area without artificial lighting.

Abstract:
This master’s thesis deals with the possibilities of illumination of motor vehicles. Thesis is divided into two sections, from which the first discusses precisely the possibilities of lighting of motor vehicles at the present time. What are the modern lighting techniques, that can be found on the market and what is their impact on safety enhancement in road traffic. Second section is about the practical measurement of low and high beam afterglow. This measurement is being compared between two types of headlamps, namely halogen headlights and xenon headlights. Part of the measurement is as well measurement of illumination intensity precisely for these headlights. The output of measurement is the creation of own illumination intensity diagrams and afterglow diagrams of particular headlights, in comparison to isolux diagrams provided by the headlights manufacturer. Consequently are solved the issues about utilization of the isolux diagrams in expert praxis.

Abstract:
La producció de proteïnes recombinants ha esdevingut la opció predilecta per la producció de productes biofarmacèutics i enzims industrials. Com a factories cel·lulars, els llevats presenten nombroses avantatges enfront d’altres microorganismes, com la seva capacitat per realitzar modificacions post-traduccionals a les proteïnes, l’ampli ventall d’eines genètiques i la facilitat per créixer a altes densitats, entre d’altres. En aquest context, la idoneïtat del llevat metilotròfic Pichia pastoris en la producció de proteïnes recombinants ha destacat per sobre del llevat clàssic Saccharomyces cerevisiae, presentant una rellevància creixent en la producció de productes comercials recombinants. Entre d’altres factors, la presència del promotor AOX1 (PAOX1), d’expressió elevada i fortament regulat pel metanol, ha contribuït a la popularitat d’aquest sistema d’expressió. Per tal de millorar la producció de proteïna recombinant, s’han provat diverses estratègies, sent l’increment de la dosi gènica una de les més populars. Tot i que es pot obtenir una millora de la productivitat heteròloga amb aquesta metodologia, elevades dosis gèniques normalment comporten una sobrecàrrega metabòlica que deriva en productivitats baixes. Durant el transcurs d’aquesta tesi, s’ha utilitzat la lipasa de Rhizopus oryzae (Rol) com a proteïna model, expressada sota el control de PAOX1. Primer, es va construir una col·lecció de soques de P. pastoris amb un nombre creixent de còpies de ROL, i es va descriure per primer cop l’ús de la ddPCR (de l’anglès Droplet Digital PCR) per la determinació acurada de la dosi gènica a P. pastoris, demostrant una precisió més elevada que amb l’ús de la qPCR (de l’anglès quantitative real-time PCR) convencional. Segon, es va realitzar un estudi transcriptòmic mitjançant microarrays d’un grup de soques multicòpia, per tal d’entendre millor l’impacte de la dosi gènica de ROL a les rutes metabòliques. Concretament, les dades van revelar una forta regulació negativa de les rutes relacionades amb el metanol (entre d’altres), en consonància amb les dades fisiològiques obtingudes, que mostraven que les taxes de consum de la font de carboni i el creixement cel·lular eren dependents del nombre de còpies. A partir d’aquestes observacions, es va modificar una soca amb 4 còpies de ROL per tal que expressés una variant desregulada del factor de transcripció Mxr1, principal regulador de la ruta del metanol. Les dades fisiològiques i els nivells transcripcionals dels principals gens implicats en el metabolisme del metanol van confirmar la millora en la producció de Rol i l’increment en l’expressió dels gens relacionats amb el metanol i ROL. Finalment, es va dur a terme un estudi comparatiu a nivell transcriptòmic de la resposta de P. pastoris a l’expressió de tres proteïnes recombinants diferents, per tal d’estudiar l’impacte de la complexitat proteica i l’ús de sistemes d’expressió constitutius o induïbles. En resum, el present estudi descriu per primera vegada una aproximació a nivell de sistema de l’impacte d’una dosi gènica creixent en el llevat P. pastoris, permetent així el disseny de noves estratègies d’enginyeria cel·lular per tal de generar soques més productores, mitjançant la biologia sintètica i l’enginyeria metabòlica.
Recombinant protein production is becoming the preferred option to synthesized biopharmaceutical and industrial enzymes. As a host system, yeast factories provide numerous advantages over other microorganisms, such as their capacity to perform post-translational protein modifications, the wide genetic available tools, and the facility to growth at high densities, among others. In this context, the suitability of methylotrophic yeast Pichia pastoris for recombinant protein production has stand out over the classical yeast Saccharomyces cerevisiae, presenting an increasing relevance for the production of commercial recombinant products. Among several factors, the presence of the strong and tightly regulated AOX1 promoter (PAOX1) has contributed to the popularity of this expression system. To further increase the recombinant protein production, several strategies have been attempted, being the increase of the heterologous gene dosage one of the most widely reported. Although a heterologous productivity improvement can be achieved with this methodology, high gene dosages normally entail a metabolic burden in the cell that results in lower productivities. In the course of this thesis, it has been used the lipase of Rhizopus oryzae (Rol) as a protein model expressed under the control of the PAOX1. Firstly, we constructed a series of P. pastoris strains harbouring an increase gene dosage of ROL, and we reported for the first time the use of Droplet Digital PCR (ddPCR) for an accurate copy number determination in P. pastoris, allowing a higher precision than using conventional quantitative real-time PCR (qPCR). Secondly, a transcriptomic study of a set of multicopy strains was carried out using microarrays, to better understand the impact of ROL gene dosage on the metabolic pathways. Concretely, transcriptomic data revealed a high downregulation of methanol-related pathways (among others), in agreement with the physiological data that had been showed that C-source uptake rate and growth yields were gene dosage dependents. Based on these observations, a strain carrying 4 copies of ROL was remodified to express a deregulated variant of the Mxr1 transcription factor, the main regulator of methanol metabolism. Physiological data and transcriptional levels of the main genes involved in methanol metabolism confirmed the improvement of Rol production and the increasing expression levels of methanol-related genes and ROL. Finally, it was performed a comparative transcriptomic analysis of the response of P. pastoris to the expression of three different recombinant proteins, to study the impact of protein complexity and the use of constitutive or inducible expression systems. Overall, in the present study we describe the first systems-level approach to determine the impact of increasing gene dosage in the yeast P. pastoris, hence allowing the design of novel cell engineering strategies to generate high-producing strains by means of the synthetic biology and metabolic engineering.

Resumo: O trabalho analisa O Ateneu baseado na acepção tradicional do Bildungsroman ( romance de formação), cujo paradigma é Os anos de aprendizado de WilhelmMeister (Goethe). Este tipo de romance ressalta o aperfeiçoamento humano e tem sido considerado como gênero literário. O termo Bildungsroman foi incorporado ao léxico brasileiro por Massaud Moisés no Dicionário de Termos Literários, que o define como uma narrativa que lida com a experiência dos personagens vivida durante a educação ou os anos de formação. Fábio Lucas reconhece O Ateneu como um romance de fonnação de grande expressão nacional.Como um programa narrativo, o Bildungsroman considera o desejo do herói pela fonnação, a substituição da casa paterna pelo universo formativo, o encontro com tutores, e a trajetória formativa do protagonista como uma maneira de entender e enfatizar sua fonnação e desenvolvimento. Assim este trabalho tem o objetivo de entender o caráter de Sérgio criança e, ao mesmo tempo o de Sérgio adulto. Nesta tentativa, também se enfatiza a idéia de O Ateneu incorporar intrinsecamente formação e deformação
Abstract: This work analyses o Ateneu based on the traditional aception of Bildungs roman (formation romance), whose paradigm is The Years of Apprenticeship of Wilhelm Meister (Goethe). This type of romance stands out the human improvement and has been considered as a titerary geme. The term Bildungsroman was incorporated into brazilian lexis by massaud Moisés in Dicionário de Termos Literários, which defines it as a narrative which deals with the characters experiences tived during the education or the formation yaers. Fábio Lucas recognizes O Ateneu as formation romance with a great national expression. As a narrativa program, the Bildungsroman considers the hero longing for teh formation, the substitution of partenal home into the formative universe, the meeting with tutors and the formative path of the protagonist as a way of understanding and emphasiging its formation and development. Thus, this work has the objective to understand the character of child Sérgio and, at the same lime, to ty to construct adult Sérgio character. In this attempt, it also emphasizes the idea of the Ateneu incorporates intrinsicallyformation and deformation

Dosis® är en digital doseringsask från Victrix AB som med hjälp av en inbyggd datorenhet kan ställas in för att påminna patienter om doseringstillfällen genom alarm, röstmeddelande och vibration. För att vidareanpassa Dosis® till patienter med kognitiva nedsättningar vill Victrix AB implementera en låsfunktion till doseringsfacken. Facken skall endast kunna öppnas under doseringstillfällen för att undvika felmedicinering. Detta arbete fokuserar på att skapa underlag för utvecklingen av denna låsfunktion till Dosis®. Genom att bryta ned låsets funktion i delfunktioner har koncept strategiskt kunnat utvecklas. Genom ett morfologiskt schema har utvalda konceptfragment från alla låsets delfunktioner slutligen kombinerats till sex potentiella lösningar.
Dosis® is a digital pill box from Victrix AB which has a built in computer unit that can be set to remind patients of when it’s time to take their medicine through alarm, voice messages and vibrations. To adapt Dosis® for patients with cognitive dysfunctions Victrix AB wants to implement an automated locking system. The locking system is supposed to prevent patients from opening pill compartments outside of the pre-set time for medication. By doing this Victrix AB hopes to decrease the risk of these patience overdosing. This work is focused on producing a basis from which an automated locking system could be designed. The function of the locking system was broken down into sub functions, making it possible to strategically develop new solutions. Through a morphological schedule chosen concept fragments where combined from all sub functions to finally produce six potential solutions.

Abstract:
Ketersediaan tanaman pangan terutama kacang tanah diupayakan dapat memenuhi kebutuhan masyarakat yang terus meningkat. Penelitian ini dilakukan dengan harapan dapat meningkatkan hasil panen serta kualitas benih kacang tanah dengan pengaplikasi paclobutrazol dan penggunaan dosis SP36 yang efektif. Rancangan Acak Kelompok (RAK) (2 faktor dan 3 ulangan). Faktor pertama yaitu paclobutrazol (4 taraf) dan faktor kedua adalah dosis pupuk SP36 (3 taraf). Hasil menunjukan bahwa perlakuan dengan penerapan paclobutrazol berpengaruh terhadap peningkatan hasil panen dan kualitas benih kacang tanah. Pada perlakuan aplikasi paclobutrazol terbaik untuk meningkatkan hasil panen yaitu tanpa menggunakan paclobutrazol dan pada penggunaan dosis SP36 125 kg/ha merupakan hasil yang paling tinggi yaitu 1.05 ton/ha benih bernas. Penggunaan dosis SP36 terbaik untuk meningkatkan kualitas benih yaitu 125 kg/ha dengan nilai 98.78% untuk daya perkecambahan dan 98,78% unutk jumlah kecepatan perkecambahan benih. Interaksi antara konsentrasi paclobutrazol dan dosis SP36 tedapat pada parameter tinggi tanaman dan jumlah bunga.

Abstract:
Insufisiensi laktasi menjadi masalah kesehatan masyarakat karena meningkatkan risiko kesakitan dan kematian bayi. Plectranthus amboinicus (L.) telah dilaporkan efeknya pada peningkatan produksi susu. Dalam beberapa tahun terakhir, metode ekstraksi basah menggunakan pelarut air belum dilaporkan pengaruhnya terhadap produksi susu. Penelitian ini bertujuan untuk 1) mendapatkan dosis ekstrak basah yang efektif terhadap produksi susu, konsumsi pakan, pertumbuhan berat badan tikus dan anakan, 2) memperoleh waktu optimum dalam meningkatkan produksi susu, konsumsi pakan, pertumbuhan berat badan tikus dan anakan, dan 3) mengetahui hubungan produksi susu, konsumsi pakan, pertumbuhan berat badan tikus dan anakan. Penelitian dilakukan di Tropical Biopharmaca Research Center, IPB University, pada Februari 2019 hingga Agustus 2019. Lima belas ekor tikus betina (Rattus norvegicus) galur Sprague-Dawley (SD) dibagi menjadi tiga kelompok. Kelompok pertama sebagai kontrol, kelompok kedua adalah tikus yang diberi dosis 162 mg/kgBB (dosis 1), dan kelompok ketiga adalah tikus yang diberi dosis 324 mg/kgBB (dosis 2). Hasil penelitian menunjukkan bahwa dosis 324 mg/kgBB dapat meningkatkan produksi susu tikus (2,81 ± 0,47 g), pertumbuhan bobot badan anak tikus (18,43 ± 0,21 g), dan konsumsi pakan tikus (18,86 ± 0,18 g) dibandingkan dengan kontrol (masing-masing 1,79 ± 0,89 g, 13,39 ± 0,29 g, 13,14 ± 0,33 g). Pertumbuhan bobot badan tikus pada dosis 2 mengalami penurunan 1,79 ± 0,94 g dibandingkan dengan kelompok kontrol 0,43 ± 0,49 g. Waktu optimal untuk peningkatan produksi susu diperoleh pada hari ke 9-16. Peningkatan produksi susu berkorelasi positif dengan pertumbuhan bobot badan anak tikus dan konsumsi pakan tikus. Di sisi lain, produksi susu berkorelasi negatif dengan pertumbuhan berat badan tikus.

Pupuk kandang ayam dan larutan mikroorganisme lokal daun gamal dapat meningkatkan hasil panen kacang hijau. Penelitian dosis pupuk kandang ayam dan konsentrasi larutan mikro organisme lokal (MOL) daun gamal yang optimal untuk meningkatkan hasil panen kacang hijau sangat diperlukan. Penelitian dilaksanakan di Kebun Percobaan dan Laboratorium Teknologi Benih Politeknik Negeri Jember pada Agustus 2015 sampai Desember 2015. Tujuan penelitian mengetahui pengaruh dosis pupuk kandang ayam dan konsentrasi MOL daun gamal terhadap hasil panen kacang hijau varietas Vima-1. Penelitian menggunakan RAK Faktorial. Faktor pertama adalah dosis pupuk kandang ayam terdiri atas tiga taraf yaitu dosis 250, 500 dan 750 gram/polybag. Faktor kedua adalah konsentrasi larutan MOL daun gamal terdiri atas tiga taraf yaitu 60, 90 dan 120 ml/liter. Kombinasi perlakuan diulang tiga kali. Analisis menggunakan ANOVA, dilanjutkan dengan uji DMRT. Hasil penelitian menunjukkan terdapat pengaruh interaksi antara dosis pupuk kandang ayam dan (MOL) daun gamal terhadap bobot basah, bobot kering, jumlah polong, bobot biji per tanaman, sedangkan Berat 100 Butir, daya Kecambah, kecepatan tumbuh dan keserempakan tumbuh menunjukkan berbeda tidak nyata. Dosis pupuk kandang ayam 250 g/polybag dengan konsentrasi MOL daun gamal 120 ml/l menunjukkan bobot basah, bobot kering, jumlah polong tertinggi. Aplikasi pupuk kandang ayam dengan MOL daun gamal dapat meningkatkan berat 100 Butir, daya Kecambah, kecepatan tumbuh dan keserempakan tumbuh benih kacang hijau.

Pengujian pupuk Fertila (12-6-24 +2MgO+3CaO+14SO3+unsur mikro) bertujuan untuk mengetahui pengaruh dosis pupuk terhadap kadar N, P, K daun, kadar sari buah, kadar asam dan total padatan terlarut, serta efektivitas pupuk terhadap hasil buah jeruk keprok Terigas. Percobaan ini dilaksanakan di IP2TP Kliran dan Laboratorium Ekofisiologi, Balai Penelitian Tanaman Jeruk dan Buah Subtropika, pada Agustus 2017. Tanaman yang digunakan adalah keprok Terigas yang diokulasi pada batang bawah Japansche Citroen berusia 4 tahun, dengan produksi tahun sebelumnya adalah 20 kg/pohon. Percobaan disusun menggunakan rancangan acak kelompok dengan 6 perlakuan, yaitu kontrol (tanpa pupuk), pupuk rekomendasi (0,60 kg Urea (45% N) + 0,50 kg SP36 (36% P2O5) + 0,10 kg KCl (60% K2O)/pohon), serta pupuk Fertila dengan dosis 1,50, 2,25, 3,00, dan 3,75 kg/pohon. Setiap perlakuan diulang 4 kali. Hasil penelitian menunjukkan bahwa aplikasi pupuk Fertila pada Inceptisol dengan status N tanah rendah, P tersedia dan K dapat ditukar sedang, berpengaruh nyata terhadap kadar N dan K daun jeruk. Fertila pada dosis tertinggi menghasilkan sari buah dengan total padatan terlarut lebih besar dibandingkan dengan pupuk rekomendasi, tetapi tidak berbeda nyata pada kandungan sari buah dan total asam tertitrasi. Kecuali dosis 1,5 kg/pohon (7,5% dari bobot panen sebelumnya), pupuk Fertila menghasilkan efektivitas hasil buah lebih dari 100% dan dosis pupuk optimum yang dianjurkan adalah 3 kg/pohon setahun.

Pengujian Pupuk Ammonium Klorida BUMI IJO ini dilakukan di Kebun Percobaan Ciparanje, Fakultas Pertanian Universitas Padjadjaran, Kampus Jatinangor, Kabupaten Sumedang, Jawa Barat. Lokasi penelitian memiliki ketinggian 820 m di atas permukaan laut, dan analisis laboratorium dilaksanakan di Laboratorium Kimia Tanah Fakultas Pertanian Universitas Padjadjaran, Jatinangor. Bahan yang digunakan dalam pengujian ini adalah media tanam berupa tanah mineral Inceptisols. Benih jagung manis (Zea mays L.); Pupuk Anorganik Hara Makro “BUMI IJO” Ammonium Klorida; Pupuk Anorganik Tunggal Urea (45 % N), SP-36 (36 % P2O5), dan KCl (60 % K2O). Dari hal tersebut dapat disimpulkan bahwa pengaplikasian pupuk anorganik “BUMI IJO” dengan ¾ dosis anjuran menujukan pengaruh nyata terhadap pertumbuhan vegetatif tinggi tanaman, jumlah daun, diameter batang dengan perlakuan kontrol. Berdasarkan analisis statistik bobot tongkol dan konversi bobot hasil per hektar tertinggi ditunjukkan oleh perlakuan E (3/4 dosis anjuran) dibandingkan dengan perlakuan lainnya. Nilai Nilai Relativitas Agronomi (RAE) tertinggi 171% dicapai oleh perlakuan Pupuk Anorganik “BUMI IJO” ¾ dosis anjuran. Dilihat dari perhitungan “index benefit cost ratio” (IBCR), yaitu berdasarkan bandingan antara penerimaan yang memperoleh perlakuan dikurangi control terhadap pengeluaran yang memperoleh perlakuan dikurangi kontrol, maka pemberian Pupuk Anorganik “BUMI IJO” memperlihatkan nilai-nilai yang relatif tinggi dibandingkan dengan perlakuan kontrol. Berdasarkan perhitungan B/C, usahatani jagung manis dengan penggunaan Pupuk Anorganik “BUMI IJO” dapat memberikan keuntungan bersih rata-rata 108 sampai 296 persen. Bila dihitung dengan biaya produksi per hektar pada musim penghujan sejak Februari - Juni 2016 dapat memberikan keuntungan bersih 1,08 – 2,96 kali dari modal yang ditanam. Pendapatan usahatani yang relatif tinggi diperoleh pada perlakuan Pupuk Anorganik “BUMI IJO” ¾ dosis anjuran (E) diikuti oleh perlakuan Pupuk Anorganik “BUMI IJO” 1 ¼ dosis anjuran (F) dan kemudian Pupuk Anorganik “BUMI IJO” 1 ½ dosis anjuran (G) lalu yang terakhir adalah perlakuan NPK Standard (B). Masingmasing sebesar Rp 60.480.000; Rp 54.038.880; Rp 53.760.000; dan Rp 47.488.560, selama 4 sampai 5 bulan (termasuk waktu persiapan tanam).

Esta nota de clase presenta al enfermero en formación la importancia y la responsabilidad de su quehacer en el día a día en relación con el cuidado de los pacientes ante una situación de emergencia o un estado de salud complejo, que requiera la administración de medicamentos de alto riesgo tales como inotrópicos, vasopresores, vasodilatadores, antiarrítmicos, insulinas, potasio, etc. Ese compromiso enfermero-paciente implica conocimiento científico basado en la evidencia, capacitación continua y adquisición de habilidades para realizar cálculos matemáticos sencillos, preparación de mezclas, programación de equipos o bombas de infusión y, desde luego, la atención cálida y humana que distingue a nuestra profesión, basada en los principios éticos y valores fundamentales de la sociedad. Los medicamentos anteriormente mencionados, no solo pueden salvar la vida de los enfermos, sino que también pueden poderla en riesgo, si se cometen errores en la preparación de la mezcla, en la programación de la velocidad o dosis de la infusión o debido a la falta de conocimiento del enfermero acerca del tipo y características del medicamento que está manejando.
