Abstract:
Cette thèse a pour objet l'étude thermique d'un réacteur à Décharge à Barrière Diélectrique (DBD) de laboratoire. Une étude expérimentale a d'abord été réalisée afin de mesurer les températures en différents points du réacteur à l'aide de fibres optiques. Ces mesures ont été effectuées en régime transitoire et en régime stationnaire. Afin d'examiner l'influence des pertes thermiques, nous avons en outre considéré des réacteurs non isolés et des réacteurs isolés thermiquement. L'influence de la nature et de la forme des tensions appliquées a également été considérée. C'est ainsi que des expériences ont été menées avec une tension sinusoïdale et avec une alimentation pulsée qui permet une meilleure dynamique de dépôt de puissance. D'une manière générale, l'élévation de la température dans le réacteur est toujours de l'ordre de quelques dizaines de degrés. Cependant, un phénomène intéressant qui semble ne pas avoir été reporté par ailleurs a été mis en évidence. En effet un phénomène d'emballement thermique se produit dans certaines situations conduisant à une élévation de la température de plus de 150°C et ouvrant ainsi de nouvelles perspectives telles que l'association plasma/catalyse, puisque l'on atteint ici des niveaux de température suffisants pour enclencher le phénomène de catalyse. Différents modèles conductifs numériques et analytiques ont par ailleurs été développés. Les modèles analytiques simplifiés, qui ont été proposés, ont été validés et peuvent servir à mieux dimensionner les réacteurs à décharge à barrière diélectrique
This thesis aims to study the thermal behaviour of a laboratory Dielectric Barrier Discharge (DBD) reactor. An experimental study was first realized to measure temperatures at different points of the reactor by using optic fibres. These measurements were performed in transient and steady states. To examine the influence of heat losses , not insulated and insulated reactors were considered. The influence of the nature and the form of the applied voltage was else considered. Experiments were conducted with a sinusoidal voltage and a pulsed power supply

L’objectif de ma thèse est de montrer la grande originalité de l’œuvre de Jean-Louis Dubut de Laforest et de rendre accessibles au lecteur contemporain ses orientations idéologiques et ses choix esthétiques.L’appartenance de ses romans à une littérature dite « populaire » s’explique par le primat qu’ils ac-cordent à l’action et surtout par les modalités de leur diffusion : un grand nombre d’entre eux ont en effet d’abord été publiés sous la forme de romans-feuilletons dans la presse à grand tirage de son époque. Cependant, chacune de ses créations est profondément ancrée dans un environnement historique que l’auteur reflète, commente et sur lequel il cherche à peser au moyen de l’invention narrative, en vertu d’une ambition sociale plusieurs fois exprimée. Une partie de mon travail consiste donc également à mettre au jour le contexte social et politique de la période de rédaction des romans, entre 1880 et 1902, en m’appuyant essentiellement sur des travaux d’historiens.Après une présentation détaillée des quelque quatre-vingts volumes qui composent l’œuvre de Jean-Louis Dubut de Laforest, il s’agit donc d’étudier son ensemble en privilégiant des axes thématiques où les choix de l’auteur sont particulièrement sensibles. Les motifs étudiés dans la seconde partie donnent leur titre aux différents chapitres : la condition féminine, la prostitution, l’homosexualité, l’antisémitisme, la presse, les relations entre science et religion, l’action politique, les syndicats et mutuelles, l’anarchisme et l’utopie, et le souvenir de la guerre de 1870. Le dernier chapitre envisage la réception de cette œuvre monumentale et le statut du lecteur dans l’ensemble des romans
The aim of my thesis is to show the great originality of Jean-Louis Dubut de Laforest’s literary work and to make accessible his ideological convictions and aesthetic choices to the contemporary reader.The inscription of his novels in the so-called popular literature can be explained by the importance given to the action and mostly by the means of their distribution: a great number of them were first published in serial form in the popular press of his time. However, each of his creations is deeply rooted in its historical background, which is reflected in it and commented on by the author who tries to influence his era through the narrative invention by virtue of his social ambition so frequently expressed. Therefore, a part of my study consists in analyzing the social and political context of the period corresponding to the writing of the novels, between 1880 and 1902, by using history researches. After a detailed presentation of the some eighty volumes of which is composed Jean-Louis Dubut de Laforest’s work, the matter is to study the whole by privileging certain thematic axes where the author’s choices are particularly meaningful. The motifs which are examined in the second part of the study give the name of the different chapters : women’s condition, prostitution, homosexuality, anti-Semitism, the press, science and religion relationship, political action, trade unions and mutual benefit societies, anarchism and Utopia, and the memory of the Franco-Prussian War. The last chapter deals with the reception of this monumental work and the reader’s status in the novels

Funções Wavelet de suporte compacto têm sido recentemente aplicadas na resolução numérica de equações diferenciais com resultados bastante promissores. A partir do sucesso do uso das wavelets de Daubechies em diversos métodos como o de Galerkin, surgiram novas famílias de wavelets para a resolução de problemas específicos. Nesse contexto, vale destacar uma família de wavelets com características de funções interpoladoras chamadas Interpolets. Este trabalho tem como uma de suas contribuições a formulação de elementos finitos baseados em funções wavelet de Daubechies e interpolets de Deslauriers-Dubuc para sua utilização em problemas dinâmicos como a propagação de ondas em estruturas, além de problemas não-lineares como o cálculo de cargas críticas de flambagem para colunas e pórticos. A partir dessa formulação, o Método de Wavelet-Galerkin foi adaptado para a solução direta das equações diferenciais através de uma implementação que não depende da discretização do sistema em graus de liberdade (formulação sem-malha ou meshless). Este tipo de abordagem permite também explorar ao máximo as propriedades de multirresolução das wavelets. Diversos exemplos com descontinuidades e não-linearidades foram estudados com êxito.
The use of compactly supported wavelet functions has become increasingly popular in the development of numerical solutions for differential equations. Daubechies wavelets have been successfully used as base functions in several schemes like the Galerkin Method. Meanwhile, Deslauriers and Dubuc developed a new wavelet family with interpolating characteristics called Interpolets. One important contribution presented in this work is the formulation of Finite Elements based in Daubechies wavelets and Deslauriers-Dubuc interpolets. Dynamic problems like wave propagation and structural stability problems were used as examples for the validation of the Finite Elements. The Wavelet-Galerkin Method has then been adapted for the direct solution of differential equations in a meshless formulation. This approach enables the use of a multiresolution analysis. Several examples with discontinuities and nonlinearities were studied successfully.

Abstract:
The study analyses growth and development of Northern red oak stands in the Czech Republic. The Northern red oak stands under study have the age span between 17 to 159 years. The study also investigated the influence of the species on soil upper horizons. The results show the high wood production potential of red oak when compare to sessile oak being at the same or higher quality and health. Data show slower decomposition of biomass in upper soil horizons under red oak causing acid reaction of soils. The data propose the red oak to be a good additional species in lowland stands and it could be a species which substitutes domestic oaks where tracheomycosis diseases can occur

Abstract:
Bark beetles are major pests of pine forests in North America that can inflict considerable damage and cause severe economic loss. The checkered beetle Thanasimus dubius Fabricius (Coleoptera: Cleridae), an abundant predator in the Eastern USA and Canada, has been suggested to influence the dynamics of several bark beetles, including the economically important southern pine beetle (SPB), Dendroctonus frontalis Zimmermann (Coleoptera: Curculionidae). Because of the importance of T. dubius in pine forests, I studied several aspects of T. dubius biology including both its chemical and dispersal ecology. I first present an experiment that was conducted to improve the rearing system of T. dubius so that sufficient numbers of predators could be produced more efficiently for both behavioral and ecological studies. Improvements to the rearing method potentially could be achieved by adding a preservative to increase the shelf-life of the diet used to feed larval T. dubius thereby allowing feeding intervals to be extended. To evaluate this, I added sorbic acid, a preservative, to the larval diet at three different concentrations (0, 0.1 and 0.2%) and for two different intervals between feedings (2-3 vs. 5 days). Additionally, I measured potential deleterious effects of this preservative on predator performance (i.e., female fecundity). I then assessed the effect of this predator on SPB survival by releasing newly hatched T. dubius larvae at several densities (0, 50 100, and 200) on pine logs infested by SPB. I also studied the chemotactic response of this predator toward various volatiles such as prey pheromones or tree volatiles in a wind tunnel. I developed a method that associated upwind flight behavior and antennal behavior to quantify the relative attractiveness of each tested source of chemical. I then conducted three experiments to investigate the ability of individual T. dubius to respond to different prey olfactory signals (bark beetle pheromones) and assess their potential for learning. First, I examined individual predator preferences toward three major prey pheromones (ipsenol, ipsdienol, and frontalin) in a study that combined a mark-release-recapture experiment with a field choice test. I also measured the responses of individual wild T. dubius in a wind tunnel, which allowed the testing of a wider range of semiochemicals (frontalin, ipsenol, ipsdienol, sulcatol, and α-pinene). A final wind tunnel experiment tested the ability of naive predators to learn two prey pheromones, frontalin and ipsenol, that were presented alone or associated with a reward. In the last chapter, I quantified the dispersal of this predator and its behavior along an edge separating a pine forest from a non-suitable habitat. I analyzed T. dubius distribution and movement in the field relative to its bark beetle prey Ips grandicollis Eichhoff (Coleoptera: Curculionidae) and to the root feeder Dendroctonus valens LeConte (Coleoptera: Curculionidae). These three insects are associated with the phenomenon of red pine decline in the Great Lakes area. Thus, understanding the relative pattern of movements of these three beetles could provide considerable knowledge on the spatial and temporal progression of red pine decline by, for instance, assessing the connection between existing pockets. I estimated the dispersal quantiles for this predator relative to its bark beetle prey and determined whether dispersal behavior was relatively homogeneous (one kind of disperser) or heterogeneous (two kinds). In a second part, I sampled the abundance of T. dubius, I. grandicollis, and D. valens along transects set between a matrix area constituted by a clearing and a habitat zone composed of red pine forest. I further developed a diffusion model, including a constant k (corresponds to the ratio of densities along the edge), that permits characterization of beetle behavior around the boundary. Studies developed in the present dissertation, therefore, investigated several aspects of T. dubius ecology: predation on SPB, ability to respond to an olfactory signal, and dispersal traits. I first showed that augmentation with larval T. dubius can successfully reduce the SPB ratio of increase in infested pine logs. I also developed a system of rearing T. dubius that was more efficient and could allow larger numbers of predators to be produced. Sorbic acid did not reduce adult lifetime or size but did affect female fecundity (20 - 40% reduction). Increasing feeding time interval to 5 days (instead of 2-3 days) improved the efficiency of the system, even when sorbic acid was not added in the diet. Wind tunnel experiments present a methodology that could enable future quantification of the relative attractiveness of various semiochemical treatments. I identified a particular antennal behavior, which I called "stand up antennal" that was typically produced when the insect was attracted to a semiochemical. The choice tests in the field and the wind tunnel experiment using wild adults both showed that T. dubius individuals are generalists that are flexible in their response to a broad range of semiochemical signals (frontalin, ipsenol, ipsdienol, sulcatol, and α-pinene). I demonstrate that naive predators are more attracted toward ipsenol after having experienced conditioning with this kairomone via a reward. Finally, experiments on dispersal and edge behavior revealed that T. dubius and one of its common prey, the bark beetle I. grandicollis, exhibit a different pattern of movement within red pine stands and surrounding habitats. First, adult T. dubius can disperse substantially further (50% dispersed beyond 1.54 km) than I. grandicollis adults (50% dispersed beyond 0.13 km). Second, T. dubius was highly affected by the presence of an edge, showing a step-like pattern with very predators caught in unsuitable areas. Conversely, the behavior of I. grandicollis was less limited by such a boundary. As with T. dubius, the root colonizer D. valens showed a strong edge behavior that was characterized by avoiding matrix zones.

Resumo: A hialuronidase (Hyase) de peçonha contribui para a difusão da peçonha do local da inoculação. Neste trabalho, foi purificada e caracterizada a Hyase da peçonha da Vitalius dubius (Araneae, Theraphosidae), uma aranha caranguejeira encontrada no sudeste do Brasil. A peçonha obtida por estimulação elétrica de machos e fêmeas adultas foi fracionada por cromatografia em gel filtração em uma coluna Superdex 75 (tampão acetato de sódio 0,1 M, pH 6, contendo 0,15 M de NaCl), a um fluxo de 1 ml/min. O perfil de eluição foi monitorado a 280 nm e frações de 1,5 ml foram coletadas e analisadas para Hyase. As frações ativas foram reunidas e aplicadas em uma coluna de afinidade heparin-Sepharose (em acetato de sódio 0,01 M, pH 6). A coluna foi lavada com o mesmo tampão e as proteínas foram eluídas com um gradiente linear de NaCl (0-1 M). As frações ativas foram agrupadas e avaliadas quanto à pureza por SDS-PAGE e RP-HPLC. O pH ótimo, a estabilidade térmica, a presença das isoformas, e a neutralização por flavonóides e antivenenos comerciais também foram avaliadas. A Hyase foi purificada em duas etapas cromatográficas, com atividade específica de 148 unidades de redução de turbidez (TRU)/mg (peçonha: 36 TRU/mg; fator de purificação de ~ 4). A Hyase apresentou uma massa molecular estimada em 43 kDa por SDS-PAGE, que não foi afetada por ?-mercaptoetanol. Zimografia em gel contendo ácido hialurônico confirmou atividade sobre o substrato. O pH ótimo foi de 4 a 5, com atividade ótima à 37°C. A Hyase manteve estabilidade até 60°C, mas perdeu rapidamente a atividade em altas temperaturas; a atividade se manteve após vários ciclos de congelamento e descongelamento. A atividade enzimática foi totalmente inibida por dois (apigenina e naringina) dos cinco flavonóides testados. A concentração de NaCl (0,05-1 M) não influenciou a atividade. A enzima apresentou maior atividade com ácido hialurônico em relação ao sulfato de condroitina A e foi totalmente neutralizada pelo antiveneno aracnídeo polivalente contra peçonha de aranha-marrom (Loxosceles sp.), aranha armadeira (Phoneutria nigriventer) e escorpião amarelo (Tityus serrulatus), mas não por antivenenos contra peçonha de lagarta (Lonomia obliqua), escorpião (T. serrulatus e Tityus bahiensis) ou serpente (Bothrops, Crotalus e Micrurus). A Hyase aumentou a permeabilidade vascular na pele dorsal de ratos. As propriedades bioquímicas da Hyase foram semelhantes às Hyases encontradas em outras peçonhas. A neutralização pelo antiveneno aracnídeo (mas não pelo escorpiônico) indicou que esta enzima compartilha epítopos antigênicos com enzimas semelhantes em peçonhas de outras aranhas
Abstract: Venom hyaluronidase (Hyase) contributes to the diffusion of venom from the site of inoculation. In this work, we purified and characterized Hyase from the venom of Vitalius dubius (Araneae, Theraphosidae), a large theraphosid found in southeastern Brazil. Venom obtained by electrical stimulation of adult male and female V. dubius was initially fractionated by gel filtration on Superdex 75 (in 0.1 M sodium acetate, pH 6, containing 0.15 M NaCl), at a flow rate of 1 ml/min. The elution profile was monitored at 280 nm and 1 ml fractions were collected and assayed for Hyase. Active fractions were pooled and applied to an affinity column of heparin-Sepharose (in 0.01 M sodium acetate, pH 6). The column was washed with the same buffer and proteins were eluted with a linear gradient of NaCl (0-1 M). Active fractions were pooled and assessed for purity by SDS-PAGE and RP-HPLC. The pH optimum, heat stability, presence of isoforms and neutralization by flavonoids and commercial antivenoms were also assessed. Hyase was purified in two chromatographic steps with a specific activity of 148 turbidity reducing units (TRU)/mg (venom: 36 TRU/mg; purification factor of ~4). Hyase had a molecular mass of 43 kDa by SDS-PAGE that was unaffected by ?-mercaptoethanol. Zymography in gels containing hyaluronic acid indicated that there were no isoforms. The pH optimum was 4-5, with optimal activity at 37°C. Hyase was stable up to 60°C, but rapidly lost activity at higher temperatures and maintained activity after several freeze-thaw cycles. Enzyme activity was completely inhibited by two (apigenin and naringin) of five flavonoids tested. The NaCl concentration (0.05-1 M) did not influence activity. Hyase had greater activity towards hyaluronic acid compared to chondroitin sulphate and was completely neutralized by polyvalent arachnid antivenom raised against brown spider (Loxosceles spp.), banana spider (Phoneutria nigriventer) and yellow scorpion (Tityus serrulatus) venoms, but not by antivenoms to caterpillar (Lonomia obliqua), scorpion (T. serrulatus and Tityus bahiensis) or snake (Bothrops, Crotalus and Micrurus species) venoms. Hyase increased vascular permeability in rat dorsal skin. The biochemical properties of this Hyase were similar to other venom Hyases. The neutralization by arachnid but not scorpion antivenom indicated that this enzyme shared antigenic epitopes with similar enzymes in other spider venoms

Abstract:
The thesis follows the bachelor thesis on comparison of morphological and economic parameters of various types of seedlings, which resulted in the planting of containerized and bare root seedlings of the European beech (Fagus sylvativa Linne) and the English oak (Quercus robur Linnaeus) in a forest regeneration in September 2011 and April 2012. The measurement of the height and thickness of the root neck of the seedlings was carried out on designated plots in March 2015. I also investigated possible mortality from the afforestation inspection in July 2012. Furthermore, 154 seedlings, 84 containerized seedlings a 70 bare root seedlings, in order to evalutate the development of the root system, its possible deformation and subsequently detection of the volume by using xylometric method. The data collected in the field were processed into tables and compared with the results observed in the bachelor thesis. The average figures for the height, the thickness of the root neck and the volume of the root system showed almost identical development of bare root and containerized seedlings in the forest cover, even though the figures of individual seedlings were highly variable. When reviewing the development of the root system, I found unacceptable deformation of the main taproot at 25% containerized seedlings and 20% bare root seedlings. The remaining seedlings were without deformation or with acceptable deformation of the taproot (wavy taproot, unbalanced taproot with the axis in the above-ground part, an outgrowth). It was discovered that containerized seedlings were more easily removable from the ground, the seedlings were not sufficiently fixed in the ground and its root system was in some cases less developed than it was with bare root seedlings. This may be influenced by several factors, e.g. the influence of richness of the root system on fine roots, the obstruction of development of the root system by cementing the surrounding soil when using the slit method. The bare root seedlings, the English oak in particular, showed root development to a greater depth, which is very important not only for sealing but also as an important factor in the drought season and extreme summer temperatures when there is rapid exhaustion of the moisture in the upper horizon of the soil due to the vapor. Unfortunately, it was not possible to verify these findings in a larger sample size, as there had been major damage of the forest.

Abstract:
Ethnobotanical and dietary questionnaires were used to assess the utilization of plants to treat the signs and symptoms of anemia by women in La Chapelle, Haiti. The usual diet of respondents was found to be low to intermediate in iron bioavailability. The ethnobotanical questionnaire showed that most respondents (82%) used plant-based home remedies to treat anemia Amaranthus dubius, Citrus aurantium, Corchorus olitorius, Moringa oleifera, Phaseolus vulgaris and Portulaca oleracea used in the diet and as remedies for anemia were analyzed. Amaranthus dubius was found to have the higher in availability by in-vitro dialysis, 30%. The intra species variation in iron availability was influenced by storage and cooking times. The interspecies variation in iron availability was explained by the acidity of the plant species' cooked homogenate (r = 0.4168, p = 0.007).

Resumo: A aranha Vitalius dubius pertence à família Theraphosidae, que compreende todas as caranguejeiras. Sua distribuição é restrita à região Sudeste do Brasil, principalmente em São Paulo e Minas Gerais. Trata-se de uma espécie urbana, pouco agressiva e que não apresenta registros de acidentes com relevância clínica. O presente trabalho teve como objetivo descrever o aparato venenífero e caracterizar bioquímica e farmacologicamente a peçonha desta aranha. A estrutura do aparato venenífero consiste em duas robustas quelíceras anteriores, com grandes ferrões para imobilização de presas e inoculação de peçonha. Cada quelícera é preenchida por musculatura estriada, responsável pela mobilidade dos ferrões, além de abrigar a glândula de peçonha que, por sua vez, encontra-se imersa nesta musculatura, sem estar ligada à mesma. A glândula é envolta por duas túnicas musculares helicoidais entrelaçadas, responsáveis pela contração da glândula para expulsão da peçonha através de um duto interno ao ferrão. Interna à musculatura, uma camada basal rica em elastina sustenta uma complexa rede epitelial produtora de peçonha. Este epitélio extratificado possui células anastomosadas, com núcleos periféricos e prolongamentos citoplasmáticos voltados para a luz da glândula, organizado principalmente por filamentos de actina. Estes prolongamentos formam reservatórios envolvidos na biossíntese de peçonha. A análise ultra-estrutural deste epitélio revelou a presença de organelas envolvidas na síntese protéica, como núcleo com abundante eucromatina, nucléolo, retículo endoplasmático rugoso, polissomos, além de abundantes: retículo liso, aparato de Golgi, mitocôndrias e vacúolos autofágicos. Durante a fase de produção de peçonha, as principais alterações ultra-estruturais observadas foram compactação e remodelamento do epitélio, abundância de organelas e deformidade de núcleos, bem como a presença de hemócitos nos últimos estágios. A extração de peçonha revelou que as fêmeas possuem rendimento maior, em quantidade absoluta [23,1 ± 2,3 (n=11) e 12,5 ± 0,7 (n=16) mg de peçonha/aranha/extração, para fêmeas e machos, respectivamente]. No entanto, quando a quantidade de peçonha é relacionada ao o peso do animal, os machos possuem maior índice. Além disso, as fêmeas adultas com maior peso apresentaram rendimento de peçonha menor que as mais jovens. A caracterização bioquímicofarmacológica da peçonha revelou através de eletroforese que a peçonha de V. dubius possui poucas proteínas de elevada massa molecular, mas é rica em peptídeos e pequenas moléculas. A peçonha contém bastante hialuronidase, mas é destituída de atividade proteolítica, assim como não foi observada ação hemolítica. No imunoensaio de ELISA, a peçonha apresentou reatividade cruzada com soro antiaracnídico do Instituto Butantan, preparado contra as peçonhas de Loxosceles gaucho, Phoneutria nigriventer e Tityus serrulatus. No entanto, o immunoblotting demonstrou que apenas componentes da peçonha maiores que 30 kDa foram responsáveis por esta reação. A peçonha de V. dubius (10 - 300 µg/mL) foi citotóxica para células leucêmicas em cultura e exerceu forte ação edematogênica (10 µg/sítio causou o extravasamento de 90 ± 20 µL de plasma, n = 4) em pele de ratos, mediada em parte por receptores serotoninérgicos e a formação do óxido nítrico. A peçonha não apresentou significativa toxicidade quando testada em coração semi-isolado de barata. Em músculo liso (íleo de cobaia) a peçonha não causou contratura, mas produziu uma pequena potencialização da ação da bradicinina, e o bloqueio parcial de contrações eletroestimuladas em músculo anococígeo de ratos mediado por uma ação pré-sináptica da peçonha. Em preparação de nervo frênico-diafragma de camundongo, a peçonha (80 µg/mL) bloqueou as contrações por estímulos indiretos (95 ± 6% de bloqueio em 56 ± 28 minutos, n = 3). Uma maior potência de bloqueio foi observada em preparação de biventer cervicis de pintainho, onde 25 µg/mL causou 85 ± 4% de bloqueio de contrações indiretamente estimuladas em 52 ± 6 minutos (n = 4); a peçonha também produziu contratura estável. Além disso, as respostas contráteis desta preparação à Ach e KCl foram diminuídas de forma concentração-dependente pela peçonha. O fracionamento da peçonha resultou em dois grupos de proteínas ativas e um inativo tanto no edema quanto na atividade neuromuscular. Estes dados sugerem que ao menos duas toxinas são responsáveis pelo aumento da permeabilidade vascular local. Além disso, o fracionamento da peçonha indicou a presença de, no mínimo, duas toxinas neuromusculares com ações possivelmente de antagonismo nicotínico reversível e promoção de danos à estrutura funcional muscular, respectivamente.
Abstract: Vitalius dubius is a medium-sized, non-aggressive tarantula found in southeastern Brazil. In this work, we examined the histological organization of the venom apparatus of V. dubius and investigated some biochemical and pharmacological properties of this spider's venom. The venom apparatus consisted of two chelicerae fitted with large fangs for prey immobilization and venom injection. Each chelicera contained bundles of striated muscle involved in fang movement and also housed one venom gland that was surrounded by, but unattached to, these muscle bundles. The gland was closely associated with a helicoidally arranged muscle layer responsible for gland compression and venom extrusion through an inner fang duct that opened on the anterior face of the gland. Within the gland, an elastic basal membrane attached to muscles supported the secretory epithelium. This epithelium consisted of anastomosed cells, with peripheral nuclei and cytoplasmic elongations extending towards the gland lumen in an arrangement organized mainly by F-actin filaments. These elongations supported a complex network of vesicles and cisternae involved in venom biosynthesis. Ultrastructural analysis showed an abundance of organelles involved in protein synthesis, including smooth and rough endoplasmic reticulum, Golgi apparatus, mitochondria and lysosomes. Nuclei with evident nucleoli were seen. During venom production, the main ultrastructural changes were epithelial compactation, greater organelle abundance and alterations in nuclear shape. Venom was milked regularly by electrical stimulation. In absolute terms, females yielded more venom than males [23.1+2.3 mg (n=11) versus 12.5+0.7 mg (n=16) of venom/spider/milking], although when expressed in terms of body weight, males had slightly but significantly greater yields. Venom yield decreased with successive milkings and with spider age. SDS-PAGE showed that the venom contained few high molecular mass proteins, but a variety of small molecules (peptides). The venom was devoid of proteases but contained considerable hyaluronidase activity (estimated molecular mass: 45 kDa). ELISA showed that the venom reacted with immunoglobulins from commercial anti-arachnid antivenom raised against the venoms of Loxosceles gaucho (spider), Phoneutria nigriventer (spider) and Tityus serrulatus (scorpion). Immunoblotting showed that only venom components >30 kDa were responsible for this immunoreactivity. Vitalius dubius venom had no hemolytic activity but was cytotoxic to cultured leukemic cells (up to 300 µg/mL). The venom caused potent, dose-dependent (0.1-100 µg/site) dorsal skin edema in rats that was mediated by serotonin and nitric oxide but not by bradykinin or histamine. The edema-forming activity was not neutralized by commercial anti-arachnid antivenom. The venom (100 µg) was weakly cardiotoxic in the cockroach isolated heart and did not contract non-vascular smooth muscle (isolated guinea pig ileum and rat anoccocygeus), nor did it significantly alter the contractile responses to a variety of agonists in these preparations. However, electrically-induced muscle contractures in the anoccocygeus were attenuated by co-incubation with the venom. Reversible neuromuscular blockade was seen in indirectly stimulated chick biventer cervicis preparations (10-50 µg/mL), with a less potent action in mouse phrenic nerve-diaphragm preparations (at 80 µg/mL). Attenuation of the responses to exogenous acetylcholine (110 µM) and KCl (20 mM) in avian preparations suggested that the venom affected postsynaptic nicotinic receptors and had a direct action on muscle. Marked muscle contracture was also seen with the venom. Fractionation of the venom by reverse-phase chromatography yielded three major groups of proteins, two of which produced both edema and neuromuscular blockade while the third was inactive in these assays. These findings indicate that V. dubius venom contains at least two toxins that cause edema and/or produce neuromuscular blockade.

Abstract:
Arthropod predators are able to influence the population dynamics of forest insects such as bark beetles. Two common predators of bark beetles are Temnochila virescens (F.) (Coleoptera: Trogositidae) and Thanasimus dubius (F.) (Coleoptera: Cleridae). Studies on T. virescens ecology have been few and not quantitative. Also, in recent years there has been increased focus on multiple-predator effects on the population dynamics of a single prey species, which better describes a natural system. Therefore, a series of three studies was conducted to provide a general overview of T. virescens ecology and its potential use for biological control. The first study examined the functional response of T. virescens toward its Ips prey. The second evaluated the impacts of intraguild competition between T. virescens and T. dubius adults on predation of a mutual prey species, I. grandicollis. The third determined if there were differences in pheromone preference among individual T. virescens adults. To evaluate the functional response, T. virescens and Ips grandicollis (Eichhoff) adults were combined in varying densities. Ips grandicollis survival was assessed by examining the number of prey eaten in relation to predator and prey densities. I found that a type II functional response model incorporating interference provided a good fit for the data. The interference was strong enough that a ratio-dependent model fit the data just as well as the full model. The effect of competition between T. dubius and T. virescens adults on I. grandicollis survival was determined by placing varying numbers of each predator with a set prey number. I found a significant interaction between densities of the two predators, which indicates possible emergent predator effects. The combined effect of the predators was risk reducing for prey, likely because of intraguild predation. The predator T. dubius alone was more efficient at reducing the number of prey adults successfully entering the log. This study indicates the importance of low density releases of these predators for augmentative biological control, to reduce the effects of intraspecific competition. To determine if there were differences in pheromone preference among individual T. virescens adults, a mark-release-recapture experiment was combined in the field to provide a choice test between ipsenol and ipsdienol pheromones. Temnochila virescens adults that were initially captured on ipsenol were more frequently recaptured with this pheromone, while those first captured on ipsdienol were recaptured on both pheromones. This suggests populations of T. virescens are composed of specialists attracted only to ipsenol and generalists attracted to both ipsdienol and ipsenol pheromones.

Composed in the first half of the thirteenth century in Lombardy, by an anonymous belonging to the church of Desenzano or also known as Church of albanenses, the composition figures as an outline on dualistic theology. In it, we find an effort by the heresiarch, still nourished by Catholicism, in defining the belief of the two principles - a good, the creator of the spiritual world and the eternal and the other bad, the creator of the material and transitional world – based mostly on the Catholic texts written at the time, but also on the intellectual methods in vogue in university circles. We intend in this paper to analyze the nature of dualism exposed by such dissident as well as its implications on scholastic methods

Cancer may arise as a result of deregulation of oncogenes and/or tumor suppressors. Although much progress has been made for the identification of such cancer related genes, our understanding of the complex tumorigenesis pathways is still not complete. Therefore, to improve our understanding of how certain basic mechanisms work in normal and in cancer cells, we aimed to characterize two different breast cancer related genes. First part of the study focused on subcellular localization USP32 (Ubiquitin Specific Protease 32) to help understand the function of this uncharacterized gene. USP32 is a member of deubiquitinating enzymes (DUBs) and the gene maps to a gene rich region on 17q23. Genes on 17q23 are known to undergo amplification and overexpression in a subset of breast cancer cells and tumors. DUBs are known to be implicated in a variety of cellular functions including protein degradation, receptor endocytosis and vesicle trafficking. Therefore to elucidate the function of USP32, we localized the full length USP32 protein fused to GFP, in HeLa cells, using Fluorescence Protease Protection (FPP) assay and confocal microscopy. Results suggested a Golgi localization for USP32 as confirmed by co-localization study via BODIPY-TR, a Golgi specific marker. Additional investigations to find the role of USP32 in Golgi will further clarify the function of this candidate oncogene. Second part of the study focused on a potential tumor suppressor. For this purpose, we functionally characterized miR-125b, a microRNA gene as a potential tumor suppressor in breast cancer. microRNAs are regulators of gene expression and their deregulation is detected in cancer cells. miR-125b is reported as a down regulated microRNA in breast cancers. In this study, we investigated the expression, function and possible targets of miR-125b in breast cancer cell lines (BCCLs). Our results revealed a dramatic down regulation of miR-125b in a panel of BCCLs. Restoring the expression of miR-125b in low miR-125b expressing cells decreased the cell proliferation and migration as well as cytoplasmic protrusions, detected by staining of actin filaments. While connection of miR-125b and cell motility based on ERBB2 targeting has been reported earlier, here we present data on ERBB2 independent effects of miR-125b on cell migration in non-ERBB2 overexpressing breast cancer cells. Our results showed involvement of a miR-125b target, ARID3B, in cell motility and migration. Our findings showed miR-125b to be an important regulator of cell proliferation and migration in ERBB2 negative breast cancer cells, possibly through regulating multiple targets.

Ubiquitylation is an important post-translational modification and can be reversed by the action of deubiquitinating (DUB) enzymes. The ubiquitylation and deubiquitylation of target proteins are significant in terms of regulating cellular events such as protein degradation, signal transduction, vesicle trafficking, DNA repair and apoptosis. Chromosomal band 17q23 is frequently amplified in breast cancers and harbors a predicted ubiquitin specific protease gene, USP32 (ubiquitin specific protease 32). Given its potential role in breast cancer, we aimed to characterize USP32 for its potential DUB activity. Bioinformatic analysis of USP32 and known yeast and mouse DUBs suggested presence of Cys-His domains which are common in active DUBs of the USP superfamily. Our in vivo and in vitro DUB activity assays revealed that USP32 was indeed an active deubiquitinating enzyme. To investigate its substrate specificity and kinetic properties, USP32 was expressed in insect cell culture to be isolated and purified. Using isolated USP32 protein, diubiquitin assay was performed with all seven types of diubiquitin (K6, K11, K27, K29, K33, K48 and K63) as well as linear diubiquitin. Results showed that USP32 was able to cleave all seven types of ubiquitin linkages with higher cleavage efficiency for K6, K11, K48 and K63-linked diubiquitin. Moreover, kinetic parameters, Km, kcat and kcat/ Km, suggested that full length protein had lower affinity for potential substrates and lower catalytic activity compared to the catalytic domain alone. These data suggested the importance of USP32 tertiary structure and possible role of other non DUB domains (e.g. EF hand domain) which may be regulated by an as of unknown mechanism in cells. Further investigations are underway to understand the functions of USP32 in cells and how it may contribute to breast tumorigenesis.

La thèse se propose de faire une incursion dans la pensée esthétique du fin XVIIe siècle-début XVIIIe, c'est-à-dire dans les débuts de cette nouvelle discipline en tant que science à part. Les contributions de Jean-Baptiste Dubos à la naissance de cette discipline sont massives, donc ce philosophe de l'art retient et mérite toute notre attention. D'abord, grâce à la Querelle des Anciens et des Modernes, son esprit s'enrichit et se forme, ses connaissances se multiplient de façon à lui permettre de réaliser une synthèse, dans son système d'esthétique, de la riche palette eidétique déployée jusqu'à lui. Ayant comme point de départ le cartésianisme, dont il se distingue entièrement, et dont il hérite uniquement le dogmatisme (l'universalité), cette fois-ci appliqué au sixième sens, celui qui juge de la valeur d'une œuvre artistique, il aboutit à s'ériger en auteur d'un courant opposé, apparenté au sensualisme. L'éloge des sens, au détriment de la raison, qui sont impliqués dans le jugement de goût du public invité à la diffusion de l'art, témoigne d'une coupure entre les époques ; la nouvelle ère appartient à Dubos et à ses disciples, qui répondent même des territoires étrangers, de l'Angleterre et même d'Allemagne. Les cinq parties de la thèse contiennent la description de la marche en avant, des pas de géant effectués dans le domaine des idées esthétiques par l'abbé Dubos. Si, dans la première partie, à l'occasion de la Querelle des Anciens et des Modernes, on découvre une rupture au sein des notions ; la notion de climat étant le noyau d'une théorie esthétique formulée avec le plus grand soin de détail par Dubos ; on découvre donc aussi une focalisation sur le sujet, créateur ou récepteur, siège du sentiment, la deuxième partie vient pour continuer à relever les mérites de Dubos, qui rend possible une attitude différente du public, public qui ne cherche plus l'instruction dans l'art, mais le plaisir, né de la satisfaction du besoin d'être occupé. Les trois autres parties explorent d'une façon plus profonde l'apport esthétique de Dubos : la critique psychologique, la critique scientifique et la critique comparée poésie/peinture. Les qualités de bon psychologue de Dubos, qui plonge dans l'âme du public au moment où il est attendri, rendent possible une meilleure création du génie, qui sait désormais s'adresser au public, de façon émouvante. À cet usage, Dubos formule une théorie du plaisir de l'art, du beau artistique et de l'émotion, qu'il incorpore de façon soudée à son système. Dans la quatrième partie, la critique scientifique, Dubos sonde l'âme du génie, le créateur surpris dans l'enthousiasme nécessaire à la naissance de l'œuvre d'art véritable, créée en vue de plaire. Date Palm (Phoenix dactylifera L. Family: Arecaceae; sub-family: Poaceae) is thought to be the oldest fruit tree grown in the Sultanate of Oman and is a major crop in terms of the number of trees and their distribution. Dates are widely considered to be a strategic source of food security as well as an important cash crop, and have always been looked upon as a key source of stability for oasis agro-ecosystems. Several insect and mite pests and plant pathogens attack Date Palm trees. The Dubas bug, Ommatissus lybicus DeBergevin (Homoptera: Tropiduchidae), has been considered the key pest of Date Palm plantations in Oman for 40 years due to the total area infested, the severity of infestations and the scale of the crop losses. Current control programmes for Dubas bug are reliant on chemical pesticides, applied by air and from the ground over an area ranging from 12,000-20,000 ha annually. These pesticides are currently the only feasible control measure against this pest, although research is underway on bio-control technologies. The cropping environment resulting from traditional Date Palm cultivation - high planting density, high temperature and humidity, topography of the plantations, intercropping and variability in cultivars and plant height - has an influence on pest and disease incidence and also affects the control options available. The present research attempts to determine the effects of technical, environmental and biological factors on the control of Dubas Bug in order to develop a more integrated control strategy. Data have emerged from this study on: 1) optimal spray droplet spectrum and application rates for improved coverage and placement of spray deposits, including under leaf deposition; 2) information on how Dubas bug daily movement can influence control efficacy; 3) on the selective use of insecticides against Dubas bug at economic threshold levels as a supplement to other IPM methods, including cultural practices such as thinning to maintain optimal distances between trees, irrigation management, the removal of old leaves including pruning cuts and surrounding tissues that may support and harbour Dubas bug.

This study concerns the declaimed performance of recitative in early French opera. Because the dramatic use of the voice was crucial to the opera genre, this investigation begins with a survey of historical definitions of declamation. Once the topic has been described, the thesis proceeds to thoroughly study three treatises dealing with sung recitation: Bacilly's Remarques curieuses, Grimarest's Traité de recitatif, and Dubos' Reflexions critiques. Principles from these sources are then applied to representative scenes from the literature. The paper closes with a commentary on the relationship between spoken and sung delivery and on the development of different declamatory styles.

Seasonal fluctuations, development and phylogeny of Dubas bug Ommatissus lybicus de Bergevin (Hemiptera: Tropiduchidae) and its native natural enemy Pseudoligosita babylonica Viggiani (Hymenoptera: Trichogrammatidae) were studied on date palm Phoenix dactylifera Linnaeus (Arecales: Arecaceae) in the Sultanate of Oman. P. babylonica emergence and 0. lybicus egg hatching were significantly influenced by temperature whereas relative humidity had no significant effect. P. babylonica developed successfully at temperatures between 22.3°C and 32.4°C with an optimum at 30.0°C whilst no development occurred at temperatures of 17.6,34.4 and 37.2°C. 0. lybicus eggs developed at temperatures between 17.6°C and 32.4°C with an optimum at 27.5°C, whereas no development occurred at 34.4 and 37.2°C. Linear regression estimated the lower thermal threshold (Tzower) and thermal constant (DD) whereas non-linear models (Briere l " and Briere 2nd equations) estimated TZowen upper (Tmax) and optimum thermal threshold (Topt). P. babylonica successfully parasitised 0. lybicus eggs of 1 to 42 days old with embryogenesis of 1.3% to 57.2%. There was a negative response of P. babylonica female density on parasitism %. P. babylonica showed arrhenotokous parthenogenesis where male progeny developed from unfertilised eggs and females from fertilised eggs. Three sex forms were recorded (female, normal kale and deformed male). Deformed males successfully mated with females and this resulted in the production of all three forms. The results of phylogenic analysis on samples of 0. lybicus and P. babylonica collected from four different geographical locations in Oman revealed 98.1-100% nucleotide similarity among 0. lybicus samples with 8 different genotypes of which two were dominant based on sequences of the Cytochrome Oxidase Subunit I (COl) and showed 85% nucleotide similarity with Hemiptera species. P. babylonica samples shared 96.8-100% nucleotide similarity among each other with 10 different genotypes based on sequences of Cytochrome B (CytB) and shared 86% nucleotide similarity with Achrysocharoides sp. Predictive models were developed and tested for P. babylonica emergence and 0. lybicus egg hatching and showed promise for increasing the efficiency of P. babylonica in suppressing 0. lybicus populations and suggested improved timings for the applications of non-persistent insecticides used in an rPM programme. La degradació de proteïnes mitjançant el sistema de les ubiquitines i el proteasoma (UPS) és un procés altament regulat, en el qual diverses famílies enzimàtiques s'encarreguen de la correcte conjugació i deconjugació de molècules d'ubiquitina a un determinat substrat. Una d'aquestes famílies és la dels enzims deubiquitinitzants (DUBs), de la qual se n'han descrit, en humans, més de 90 membres.
USP25 (21q11.2) codifica per un enzim deubiquitinitzant i produeix tres isoformes per splicing alternatiu. Una d'elles, generada per la inclusió de l'exó 19a entre el 18 i el 19b, presenta una expressió restringida a múscul esquelètic i cor (USP25m). Mitjançant un assaig de dos híbrids en llevat varem determinar que USP25m interacciona amb una proteïna d'unió a la miosina (MyBPC1), amb l'actina 1 (ACTA1) i amb la filamina C (FLNC). Totes aquestes proteïnes són sarcomèriques, regulen la diferenciació i el manteniment de les cèl.lules musculars i han estat implicades en diverses patologies humanes.
Assajos bioquímics realitzats per aprofundir en la rellevància funcional d'aquestes interaccions ens mostren que els nivells de MyBPC1 estan regulats pel proteasoma, i que USP25m és capaç d'abolir seva degradació. En canvi, ACTA1 i FLNC són proteïnes estables, suggerint que la seva interacció amb USP25m està relacionada amb mecanismes reguladors de l'estructura i la funció del sarcòmer, i no amb la seva degradació.
USP25 conté tres dominis d'unió a ubiqüitina implicats en el reconeixement de substrats i pot ser modificada per diversos tipus de modificació posttraduccional com la ubiqüitinació, la sumoïlació, la fosforilació i l'acetilació. El fet que sigui una proteïna multimodificada suggereix que la seva activitat enzimàtica ha d'estar finament regulada.
Per últim, la sobreexpressió d'USP25 és capaç d'induïr una aturada en el cicle cel·lular, per mecanismes encara desconeguts.
USP25 is a deubiquitinating enzyme (DUB) that generates a muscle-specific isoform, USP25m, by alternative splicing. USP25m interacts with several sarcomeric proteins, but similarly to most DUBs, the contribution of different protein domains to enzymatic activity and substrate recognition remains to be ascertained. In silico analysis revealed the position of the catalytic signatures and predicted three ubiquitin binding domains (UBDs): one ubiquitin-associated domain (UBA) and 2 ubiquitin-interacting motifs (UIMs). By generating serial and combinatorial deletions of these domains, we analyzed their contribution to USP25m catalytic activity, substrate recognition, subcellular localization, and post-translational modifications, such as ubiquitination and sumoylation. Our results indicate that ablation of these motifs do not abrogate USP25 deubiquitinating activity, neither promoted any significant alteration in subcellular localization of the enzyme nor ubiquitin. However, USP25 UBDs contribute to specific substrate recognition, as interaction with each substrate requires a different combination or number of UBDs, in an additive or synergistic manner. USP25m is sumoylated in vitro and ubiquitinated, although the UBDs are not required for these post-translational modifications. In addition, USP25m is phosphorylated in Tyr, Ser/Thr residues and acetylated. The physiological relevance of all these modifications remains to be ascertained. USP25 overexpression causes cell cycle arrest, most probably at the G1/S transition.

Abstract:
This thesis aims to shed new light on the Chinese character xia 侠 and the literature and history of the Republican Era (1912-1949) that revolves around it. Xia refers to either a concept (identifiable with kindness, altruism, righteousness, etc.) or to a person who practices this concept. Ever since its arrival in Chinese texts in the sixth century BC, it has created controversy for some and sympathy for others. In Modern China, xia became the central aspect of a literary genre that reached its zenith in production and consumption in the Republican Era, i.e. wuxia fiction 武侠小说, which can be translated as “using martial arts (wu 武) to obtain xia”. The concept of xia was an integral part of presumably the most widespread literary genre of the time, but why were Republican-era readers so interested in it? Why did they relate to xia and what do the themes of these novels say about the chaotic Republican Era? To answer these questions, this thesis presents a case study of a wuxia pentalogy written by Wang Dulu at the end of the Republican Era and attempts to identify the topics and aspects most reflective of that historical period, showing that, despite the heavy criticism of intellectuals of that time, these “easy” popular novels contain innovative and modern aspects and can become today of great historical importance. The thesis starts with two literature reviews. The first determines that the term xia has not received enough scholarly attention, calling for a reassessment. The second literature review focuses on Republican Era wuxia fiction, showing how there is a gap in scholarship on this period. This is followed by a discussion of the methodology used for the analysis of the case study on Wang Dulu’s Crane-Iron Series written in Qingdao (1938-1944), presented in the final three chapters of the thesis. Chapter one analyses the origins of the term xia in texts from the Warring States Period (475-221 BC) and Han Dynasty (206 BC-AD 220), presenting new interpretations for a more comprehensive understanding of the term. Chapter two gives a historical overview of xia¬-related literature and addresses the historical reasons for the changes that xia underwent throughout Chinese history. Chapter three includes a historiography of the Republican Era in combination with the life of the author Wang Dulu and identifies the aspects of the author’s life that will become important in the textual analysis in the chapters to follow. Chapter four focuses on xia in the Crane-Iron Series. After having collected the terms and identified the semantic spheres that include the Chinese character xia, the chapter demonstrates how the story of one of the series’ protagonist can be seen as an personification of Republican-era China, proving the historical dimension and value of these novels. Chapter five analyses yi 义 (righteousness) and represents the virtuous aspect of xia, concluding that, according to Wang Dulu, for the concept of xia, virtue is more important than being trained in martial arts (wu). Chapter six focuses on the literary figure of the baobiao 保镖 (protector) and is seen as the commercialisation of martial arts not necessarily linked to xia, showing how entrepreneurship and violence were characteristics of the time.

L'activació sostinguda de Hog1 porta a una inhibició del creixement cel·lular. En aquest treball, hem observat que el fenotip de letalitat causat per l'activació sostinguda de Hog1 és parcialment inhibida per la mutació del complexe SCFCDC4. La inhibició de la mort causada per l'activació sostinguda de Hog1 depèn de la via d'extensió de la vida. Quan Hog1 s'activa de manera sostinguda, la mutació al complexe SCFCDC4 fa que augmenti l'expressió gènica depenent de Msn2 i Msn4 que condueix a una sobreexpressió del gen PNC1 i a una hiperactivació de la deacetilassa Sir2. La hiperactivació de Sir2 és capaç d'inhibir la mort causada per l'activació sostinguda de Hog1.
També hem observat que la mort cel·lular causada per l'activació sostinguda de Hog1 és deguda a una inducció d'apoptosi. L'apoptosi induïda per Hog1 és inhibida per la mutació al complexe SCFCDC4. Per tant, la via d'extensió de la vida és capaç de prevenir l'apoptosi a través d'un mecanisme desconegut.
Sustained Hog1 activation leads to an inhibition of cell growth. In this work, we have observed that the lethal phenotype caused by sustained Hog1 activation is prevented by SCFCDC4 mutants. The prevention of Hog1-induced cell death by SCFCDC4 mutation depends on the lifespan extension pathway. Upon sustained Hog1 activation, SCFCDC4 mutation increases Msn2 and Msn4 dependent gene expression that leads to a PNC1 overexpression and a Sir2 deacetylase hyperactivation. Then, hyperactivation of Sir2 is able to prevent cell death caused by sustained Hog1 activation.
We have also observed that cell death upon sustained Hog1 activation is due to an induction of apoptosis. The apoptosis induced by Hog1 is decreased by SCFCDC4 mutation. Therefore, lifespan extension pathway is able to prevent apoptosis by an unknown mechanism.

Phytoremediation is an emerging technology where specially selected and engineered metal-accumulating plants are used for bioremediation. Amaranthus dubius (marog or wild spinach) is a popular nutritious leafy vegetable crop which is widespread especially in the continents of Africa, Asia and South America. Their rapid growth and great biomass makes them some of the highest yielding leafy crops which may be beneficial for phytoremediation. This study was undertaken to evaluate the potential of A. dubius for the phytoremediation of Chromium (Cr), Mercury (Hg), Arsenic (As), Lead (Pb), Copper (Cu) and Nickel (Ni). Locally gathered soil and plants of A. dubius were investigated for the metals from a regularly cultivated area, a landfill site and a sewage site. Metals were extracted from the samples using microwave-digestion and analyzed using Inductively Coupled Plasma – Mass Spectroscopy (ICP-MS). Further experiments were conducted with plants from locally collected seeds of A. dubius, in a tunnel house under controlled conditions. The mode of phytoremediation, the effect of the metals on the plants, the ability of the plant to extract metals from soil (Bioconcentration Factor - BCF), and the ability of the plants to move the metals to the aerial parts of the plants (Translocation Factor - TF) were evaluated for the different metals. Finally, A. dubius was micro-propagated in a tissue culture system with and without exposure to the metal, and the effect was studied by electron microscopy.

The aim of this thesis was to inventory the natural regeneration of pedunculate oak on floodplain sites (FE Židlochovice, forest district Židlochovice), analysis of the conditions where the regeneration of oak under mother canopy shows a determination of potential natural regeneration of oak on the FE Židlochovice. It had been inventoried 8 (7.96 hectares) stands. These forests occupy approximately 0.24 % of the total area of interest (3355 ha). From this number fulfills the criteria ensuring the 5 forest stands (6.02 ha). The determining factor for regeneration under canopy of pedunculate oak is gap fraction (GFr) and the limiting factors are the state of the soil cover and damages by game. Natural oak regeneration can be achieved only at large-scale regeneration cuttings, but also in smaller areas such as border cutting and successfully grow it to established state.

The aim of the thesis was to characterize the state of the coppice on research plots Usakov and Hradisko. Describe resprouting of Sessile oak (Quercus petraea) and European hornbeam (Carpinus betulus) in partial areas that were previously cutted down. Find out whether it would be theoretically possible under the conditions of the Czech Republic to manage the oak and hornbeam coppice using the selection system. Conducted to evaluate impact of executed felling on radial increment and high increment and establish basic principles of coppice tending. Within the thesis was characterized present state in both research plots. It revealed a high probability of resprouting and expressed its dependence on type of felling and polycormon parameters. The results shows that in Czech Republic is use of selection system in Sessile oak and European hornbeam coppice possible. Fellings carried out on research plots had a statistically significant effect on the radial increases of Sessile oak and European hornbeam. From the detected biomass increses was developed a model, that is possible to use when proceeding with selection system in coppice (determination of returnable time).

Abstract:
The study was conducted on spoil heap Podkrušnohorská (50 ř 14'09 S, 12 ř 39'05 V) near Sokolov. Several factors that influence the establishment of oak were studied: the distance from the source of diaspors, type of habitat (succession vs alder reclamation), position of seedling on the wave and impact of animal grazing. Four alder restoration areas and three spontaneous succession areas have been mapped with GPS equipment, the occurence of mature oaks, which could serve as a source of acornsg was also mapped on the dump and in surroundings. Most seedlings (657) were found in the succession and only 98 in alder reclamation, and the occurence in succession was statistically significantly more frequent. The number of seedlings from sources diaspors decreased, the longest distance was over 1300 meters. For three habitats (spontaneous succession, alder reclamation, open space with grass vegetation (Calamagrostis epigeios)) were planted 12 oak seedlings in fence and 12 seedlings outsider of the fence. Annual growth rate and survival of oak were evaluated using two-way ANOVA. Results showed that the area has a significant effect on the growth of oaks. In alder reclamation and the open area had a high mortality of seedlings and small increase, the fence does not have significant influence on these areas....
