Abstract:
Last year, a new book by Dr. Thomas Durell Young was published by the British publisher Bloomsbury, entitled THE ANATOMY OF POST-COMMUNIST EUROPEAN DEFENCE INSTITUTIONS: THE MIRAGE OF MILITARY MODERNITY. The author probably does not need any special introduction for those whose work is related to security and defence matters. For all others, though, let me mention that he is a long-time lecturer at the Center for Civil-Military Relations (CCMR) from Monterey, California, USA. Prior to joining CCMR, Dr Young was a Research Professor at the Strategic Studies Institute of the US Army War College, where he focused on European politico-military issues, as well as on joint planning, execution and management of systems and procedures. He holds a Ph.D. in international economics from Geneva, and has authored several monographs. From 1989 to 2017 he was an editor of Small War and Insurgencies, and he is currently an editor of Defence and Security Analyses. An important fact is that his work has led him to travel to the majority of European countries and, as such, he is very well acquainted with their respective defence systems, defence and military history, as well as many defence and military decision-makers and experts, with whom he had shared his views and experiences. In most of these countries he has also acted as a defence adviser. Dr Young is an author whose field of research is extremely rich, both in theory and practice. It is therefore no surprise that his new book reflects his thorough knowledge of the historical background of the former communist states, their defence systems and their armed forces. The work is divided into nine chapters. In addition to the introduction and conclusion, it includes the following thematic sections: The State of Communist Defense Institutions and Armed Forces, circa 1990; NATO Exports its “New Model Army”: Why It Did Not Take; Former Soviet Republics' Defense Institutions; Former Warsaw Pact Republics' Defense Institutions; Former Yugoslav Republics' Defense Institutions; Building Defense Institutions: Sharpening the Western Mind; Reforming Western Policy and Management of Defense Reform. The concluding chapter is entitled Getting to Honest Defense. Young is very systematic and analytical when presenting the topic, and uses all the characteristics of comparative analysis. Wherever historical, political, defence and military bases are identical or highly similar, he provides a thorough explanation of the reasons for significant differences in further development, as well as those who had prevented the occurrence of changes or their sufficient implementation. He is very insightful in his attempts. The basic principle of his research or the criterion for the evaluation of defence institutions in the former communist states is the “Western Mind”, as he calls it. In short, a comparison of western and eastern thinking reveals key differences in planning, decision-making, leadership and implementation of the state’s defence function. The important criteria Young considers when implementing these policies are a review of defence institutions, the public policy framework and compliance of concepts, defence planning techniques, national-level command, the military decision-making process, and the concepts of operations, logistics and professionalism. According to his findings, these are the major areas where the former communist states differ. The differences between them are normally related to the past regimes and mind-sets in the respective countries. With time, some of the countries have managed to successfully overcome those differences and implement the “Western Mind”, while others have not been so successful. Although the author uses the Western Mind as the criterion to establish the level of development of defence systems in eastern countries, he does not claim this to be the only and the best criterion everyone should aim for. Quite the opposite: in his chapter “NATO Exports its ‘New Model Army’: Why It Did Not Take”, he provides a thorough explanation of where, when and why critical mistakes or flaws have been made, which still prevent some NATO members to perform up to their potential within the Alliance. He does not, however, miss the fact that the western democratic concepts of defence management cannot serve just as a means to simply follow the model of others. Rather, these areas must be functional and effective, as well as adapted to each separate country. In his book, the author lists several examples of bad practice, highlighting at the same time some of the unique and good solutions put forward by some countries. Nevertheless, individual chapters reveal some of the characteristics specific to a subject area or a country, which are hard to classify as good or bad. They are just special features that require new solutions with a higher level of functionality. In the conclusion, the latest monograph by Thomas Young lists proposals on how to build defence institutions using the Western Mind. These institutions first must be upgraded in order to be more effective in responding to new security threats. For young countries, which some twenty or twenty-five years ago still used communist concepts, Young proposes a new approach called “Honest Defense”, which is generally intended for all those building collective defence. The approach brings something new for amateurs of such topics, and at the same time serves as a broadly useful and helpful study material for experts who wish and are obliged to find concrete solutions and opportunities for a new and safe future. The monograph includes interesting concrete examples from individual countries, which the author professionally and critically comments on. The discussed examples comprise both good and bad practice. Additionally, they include an analysis of the development of the defence system and institutions in Slovenia, which enables a neutral external observer to understand our development as well as identify their own strengths and deficiencies.

Right after the construction of Islamabad, National Capital of Pakistan, different foreign architectswere hired by the State to undertake landmark projects in the city. Most notable among them were Doxiadis, Kenzo Tange, Edward Durell Stone. The preference given to these foreign architects over the local architect was tied to the inherited ideas of Modernist Supermacy, ‘glorified western cultures’ and their understanding of the traditional architecture specifically the so called, Islamic architecture. It was assumed that the architecture, foreign architects will produce would be technologically advanced, help in the International recognition of national architecture and become a symbol of Nation’s progression. The paper focusses on a comparative critique of the two landmark projects namely Presidential Estate and Pakistan Atomic Energy Commission building (PAEC) named as Pakistan Institute of Nuclear Science and Technology (PINSTECH), both situated in Islamabad and designed by Edward Durell Stone. These two buildings have been analysedwith reference to the background forces, in line to Foucault’s term,narrated as political power, subjectivity and resistance. The research has anchored on the idea that in line with the official brief, the former project was not only inspired byInternationalstyle,but also adapted to the Mughal gardens in terms of grandeur and spatial construction. On the other hand, PINSTECH building can be read as an effort of the forceful marriage of states religious subjectivities and Modernity. The paper concludes with ‘folk architecture’ a term coined by Doxiadis and reading it in conjunction with the architectural anomalies.

Since the seminal study of Steinbeck and Durell (1968), few epidemiological studies have attempted to replicate whether psychosocial stress precipitates the onset of a first psychotic episode. Our aim was to support or refute the finding of elevated psychosis incidence in the first month of army induction and to examine factors impacting the timing of onset. Data were collected from medical files of 186 army conscripts, hospitalized with a diagnosis of First Episode Psychosis (FEP) between 2005 and 2014 in the Psychiatric Military Hospital in Athens, Greece. FEP rates were at least 4.5 times higher in the first month of military service, compared with any other month. Earlier FEP onset was associated with rural environment at the time of birth, multiple drug use and service away from home. Psychosocial stress precipitates FEP, particularly in those exposed to other risk factors.

Resumo Este estudo teve como objetivo proporcionar evidências de validade e fidedignidade da Escala de Gratidão (G-20) para a população de adultos brasileiros. Especificamente, verificar a estrutura do questionário através da análise fatorial confirmatória, explorar a consistência interna da escala e as validades de critério concorrente e convergente. Trata-se de um estudo quantitativo, correlacional com delineamento transversal. A amostra por conveniência está composta por 348 adultos entre 18 e 59 anos das cinco regiões brasileiras. Os instrumentos utilizados foram questionário de dados sociodemográficos, G-20, GQ-6, DASS-21, PANAS e P-DURELL. A escala total demonstrou confiabilidade satisfatória (α = 0,92 e Ω = 0,71) e em todas as dimensões do instrumento. Além disso, a análise do ajustamento dos dados para a estrutura de quatro fatores apontou valor aceitável para uma boa adequação do modelo (RMSEA = 0,068, CFI= 0,956 e TLI= 0,949) e a predominância das correlações entre os instrumentos utilizados no estudo mostrou-se significativas.

This article explores the visual abundance found in a number of early projects by Leinweber, Yamasaki and Hellmuth (LYH) and Minoru Yamasaki and Associates (MYA), which stands in stark contrast to the austere character of architectural form during the interwar period. Although Yamasaki received his architectural training in the 1930s, he was neither a true modernist, nor a fully postmodern architect. His aesthetic, and his firm’s work, lies in the interstices between these two distinct architectural moments, in company with contemporaries Edward Durell Stone and Paul Rudolph, among others. The work of these architects embraced a kind of visual and formal excess but stopped short of approaching the playful linguistic games of postmodern architecture. With themes of visual and material excess in mind, I examine two early commissions from the U.S. federal government that put into play ideas of global exchange, power, and extravagance in architecture as the United States emerged as a major world power in the aftermath of World War II, including the U.S. Consulate in Kobe, Japan (1954–1955) and the Federal Science Pavilion at the Seattle World’s Fair (1962).

ABSTRACT We have examined mutations in the ectodomain of the human immunodeficiency virus type 1 transmembrane glycoprotein gp41 within a region immediately adjacent to the membrane-spanning domain for their effect on the outcome of the fusion cascade. Using the recently developed three-color assay (I. Muñoz-Barroso, S. Durell, K. Sakaguchi, E. Appella, and R. Blumenthal, J. Cell Biol. 140:315–323, 1998), we have assessed the ability of the mutant gp41s to transfer lipid and small solutes from susceptible target cells to the gp120-gp41-expressing cells. The results were compared with the syncytium-inducing capabilities of these gp41 mutants. Two mutant proteins were incapable of mediating both dye transfer and syncytium formation. Two mutant proteins mediated dye transfer but were less effective at inducing syncytium formation than was wild-type gp41. The most interesting mutant proteins were those that were not capable of inducing syncytium formation but still mediated dye transfer, indicating that the fusion cascade was blocked beyond the stage of small fusion pore formation. Fusion mediated by the mutant gp41s was inhibited by the peptides DP178 and C34.

Le malformazioni durali artero-venose intracraniche o fistole durali sono lesioni rare (10–15% di tutte le malformazioni artero-venose intracraniche). Per quanto a nostra conoscenza, non sono riportate in letteratura associazioni tra MAV durali ed aneurisma intracranico. Riportiamo un caso di associazione di una fistola durale del seno cavernoso con un aneurisma dell'arteria carotide interna omolaterale. La paziente è stata sottoposta a trattamento embolizzante della fistola durale e, successivamente, a «clippaggio» dell'aneurisma, ottenendo un buon risultato. Vengono discusse le ipotesi eziopatogenetiche e l'inquadramento nosografico di tale rara associazione nell'ambito di una breve revisione della letteratura.

Vari esami RM dimostrarono la presenza di un ispessimento durale con accentuazione dopo contrasto, sia in sede endocranica che a livello cervicale, ed anche un sottile e circoscritto scollamento durale con lievi fenomeni emorragici; alla remissione clinica si accompagnò la normalizzazione del quadro neuroradiologico. Vengono discussi i possibili meccanismi patogenetici delle alterazioni durali alla luce dell'anatomia degli strati profondi della dura e ne vengono sottolineati gli aspetti radiologici.

Durerea viscerală are particularităţi faţă de cea somatică: receptorii viscerali au grade diferite de angajare funcţională, nivelul nocicepţiei diferă de la un organ la altul, aferenţele viscerale puţine sunt predominant nemielinizate şi au o divergenţă extensivă la nivelul sistemului nervos central; răspunsurile antrenează activări autonomice. Matricea durerii viscerale abdominale, funcţională la naştere, este suprasolicitată prin impulsuri mecanice, chimice, osmotice; suprapunerea infecţiilor, inflamaţiilor prin mediatorii eliberaţi creează condiţiile „sensibilizării“ periferice şi centrale şi ale dobândirii „memoriei durerii“; factori comportamentali şi emoţionali pot agrava hiperactivitatea matricei durerii viscerale. În acest context patogenetic se delimitează durerea nonorganică şi forma ei severă, hiperalgezia viscerală. Se adaugă particularităţi genetice care influenţează pragul de sensibilitate la durere. Având la bază modelele biopsihosociale ale tulburărilor funcţionale, criteriile Roma III au etichetat durerile abdominale funcţionale şi sindromul de intestin iritabil ca posibile diagnostice directe şi nu doar diagnostice de excludere. Durerea non-organică, asociată frecvent cu dureri abdominale recurente, este printre cele mai frecvente probleme medicale întâlnite în pediatrie. Majoritatea pacienţilor cu dureri uşoare au o evoluţie favorabilă în timp; un procent redus prezintă simptome grave şi uneori dizabilitante. Pediatrii se confruntă frecvent cu delimitarea diagnosticului de durere non-organică şi, uneori, cu eşecuri terapeutice. Autorii prezintă date epidemiologice, mecanismele fiziopatologice implicate, abordarea clinică şi opţiunile terapeutice cu privire la durerea nonorganică şi hiperalgezia viscerală la copii şi adolescenţi.

The aim of this study was to determine the chemical characterization and antimicrobial properties of the essential oil of Romanian Origanum vulgare. The oil was isolated by hydro distillation. The chemical composition was characterized by Gas chromatography-Mass spectroscopy. Antibacterial activity was evaluated by disk diffusion method and determination of the minimum inhibitory concentration (MIC).Thirty-two volatile constituents were identified in the oil studied and the major compounds were thymol (35.51%), g-terpinen (19.19%), durenol (17.99%), durene (11.40) and carvacrol (2.69%). It showed a bactericidal activity towards all tested reference strains: Klebsiella pneumoniae, Pseudomonas aeruginosa, Staphylococcus aureus, Streptococcus pyogenes and Candida albicans.

Per la diagnosi ed il trattamento delle MAV durali e spinali è necessaria l'angiografia spinale selettiva. Poiché è opportuno un bilancio angiografico il più completo possibile e poiché talora l'esplorazione deve essere estesa al di fuori del rachide, preferiamo avere una ragionevole certezza della presenza di una MAV prima di iniziare l'angiografia. Questa certezza la cerchiamo con la mielografia. In determinate condizioni anche la RM può fare diagnosi di MAV ed è augurabile che in futuro questo esame possa sostituire la mielografia o quanto meno rappresentare un mezzo valido e non invasivo per il controllo dopo trattamento. Da quando è stata resa nota la natura di queste MAV durali, il trattamento, sia di embolizzazione che chirurgico, dovrebbe limitarsi ad interrompere il flusso tra MAV durale e plesso perimidollare. Come materiale d'embolizzazione abbiamo usato inizialmente il Bucrilato, attualmente preferiamo le particelle di Ivalon, di più facile impiego rispetto alle colle. Sottoponiamo al chirurgo i casi di recidiva e i casi in cui la situazione dell'arteria spinale anteriore rappresenta un pericolo per l'embolizzazione.

Lani je pri britanski založbi Bloomsbury izšla nova knjiga dr. Thomasa Durella Younga z naslovom Anatomy of Post-Communist European Defence Institutions: The Mirage of Military Modernity. Avtorja verjetno ni treba posebej predstavljati vsem tistim, katerih delo je povezano z varnostnimi in obrambnimi zadevami. Za vse druge pa naj navedem, da gre za dolgoletnega predavatelja Centra za civilno-vojaške odnose iz Montereya v Kaliforniji v Združenih državah Amerike. Pred tem je bil raziskovalec na inštitutu za strateške študije na ameriški Army War College, kjer se je posvečal področju nacionalne obrambe in politično-vojaškim vprašanjem ter tudi združenemu načrtovanju, izvajanju sistemov in opravljanju postopkov. Doktoriral je iz mednarodne ekonomije v Ženevi. Je avtor več monografij. Od leta 1989 do 2017 je bil urednik publikacije Small War and Insurgencies, zdaj pa je urednik pri Defence & Security Analyses. Pomembno je, da je v okviru svojega dela v centru prepotoval večino evropskih držav in zelo dobro pozna njihove obrambne sisteme, obrambno in vojaško zgodovino ter številne odločevalce in strokovnjake na obrambnem in vojaškem področju, s katerimi je delil svoje poglede in izkušnje, v večini držav pa je bil tudi v funkciji obrambnega svetovalca. Gre torej za avtorja, čigar raziskovalno področje je zelo bogato tako v teoretičnem kot tudi v aplikativnem smislu, zato ne preseneča, da nova knjiga odraža odlično poznavanje zgodovinskega ozadja nekdanjih komunističnih držav, njihovih obrambnih sistemov in oboroženih sil. Delo je razdeljeno na devet poglavij. Poleg uvodnega in sklepnega poglavja si sledijo naslednji vsebinski sklopi: Stanje komunističnih obrambnih institucij in oboroženih sil leta 1990 (The State of Communist Defence Institutions and Armed Forces, circa 1990), Nato izvaža svoj »novi vojaški model«: Zakaj ne deluje (NATO Exports Its »New Model Army«: Why It Did Not Take), Obrambne institucije nekdanjih sovjetskih republik (Former Soviet Republics' Defence Institutions), Obrambne institucije nekdanjih republik varšavskega pakta (Former Warsaw Pact Republics' Defence Institutions, Obrambne institucije nekdanjih jugoslovanskih republik (Former Yugoslav Republics' Defence Institutions, Vzpostavitev obrambnih institucij: vpeljevanje zahodnega načina (Building Defence Instituions: Sharpening the Western Mind) ter Spreminjanje zahodne politike in menedžmenta obrambnih reform (Reforming Western Policy and Management of Defense Reform). Zadnje poglavje ima naslov Prehod k »pošteni obrambi« (Getting to Honest Deffense). Pri podajanju vsebine je Young zelo sistematičen in analitičen ter uporablja vse zakonitosti primerjalne analize. Povsod tam, kjer so zgodovinska, politična, obrambna in vojaška izhodišča identična ali zelo podrobna, pojasni, kaj je povzročilo pomembne razlike v nadaljnjem razvoju oziroma zakaj do sprememb ni prišlo ali pa te razlike niso bile dovolj obsežne. Pri tem je zelo pronicljiv. Glavno vodilo pri njegovem raziskovanju ali merilo za ocenjevanje obrambnih institucij nekdanjih komunističnih držav je tako imenovano zahodno načelo ali, kot ga imenuje sam, »western mind«. Na kratko bi zahodno načelo v odnosu do vzhodnega lahko opisali kot razlike pri načrtovanju, odločanju, vodenju in izvajanju obrambne funkcije države. Pri izvajanju teh politik Young kot pomembna merila upošteva pregled obrambnih institucij, javni politični okvir in konceptualno usklajenost, tehnike obrambnega načrtovanja, poveljevanje na nacionalni ravni, proces vojaškega odločanja, koncept operacij, logistično podporo in profesionalizem. Kot ugotavlja, so to glavna področja, na katerih prihaja med nekdanjimi komunističnimi državami do odločujočih razlik, ki so sicer povezane s preteklim režimom in miselnostjo. Nekatere države so razlike skozi čas zelo uspešno premagale in uvedle zahodno načelo, druge pa pri tem niso bile tako uspešne. Čeprav je avtor vzel zahodno načelo kot merilo za ugotavljanje stopnje razvoja obrambnih sistemov vzhodnih držav, ne trdi, da je edino in najboljše. Ravno nasprotno. V poglavju Nato izvaža svoj »novi vojaški model«: Zakaj ne deluje podrobno razloži, kje, kdaj in zakaj so se zgodile napake ali pomanjkljivosti, ki še danes vplivajo na to, da nekaterim državam članicam zavezništva nikakor ne uspe v njem odigrati svoje vloge. Ne zaobide pa dejstva, da zahodni demokratični koncepti upravljanja obrambnega področja ne morejo biti le nekaj, kar enostavno naredimo po modelu, kakršnega imajo drugi, temveč morajo biti ta področja funkcionalna in učinkovita ter prilagojena vsak državi posebej. V knjigi avtor navaja kar nekaj primerov slabih praks, hkrati pa izpostavi nekatere unikatne dobre rešitve posameznih držav. Kljub vsemu je v posameznih poglavjih mogoče najti različne posebnosti po posameznih področjih in državah, ki jih je težko uvrstiti glede na dobre ali slabe, ostajajo le posebnosti, ki potrebujejo nove rešitve z višjo stopnjo funkcionalnosti. Najnovejša monografija Thomasa Younga v sklepnem delu daje predloge za vzpostavitev obrambnih institucij po zahodnem načelu. Institucije je treba najprej nadgraditi, da bodo bolj prilagojene za učinkovitejše odzivanje na nova varnostna tveganja. Za mlade države, ki so bile pred približno 20 oziroma 25 leti še v konceptu komunističnega delovanja, pa predlaga nov pristop, ki ga imenuje poštena obramba. Ta je sicer namenjen vsem, ki gradijo kolektivno obrambo. Prinaša dobrodošlo novost za ljubitelje tovrstnih vsebin, hkrati pa je vsestransko uporabno in koristno študijsko gradivo za strokovnjake, ki želijo in morajo poiskati rešitve in priložnosti za novo varno prihodnost. V monografiji so zanimivi konkretni primeri iz posameznih držav, ki jih avtor na strokovno kritičen način komentira. Med njimi so primeri dobrih in slabih praks. Med njimi je tudi analiza razvoja obrambnega sistema in institucij v Sloveniji, ki omogoča razumevanje našega razvoja in prepoznavanje lastnih pomanjkljivosti, pa tudi prednosti, s strani nevtralnega zunanjega opazovalca.

If truth hurts, this is no doubt because it is often enough forced on us. And the question as to whether the reception of “nice,” “easy” truths is similarly an outcome of coercion negates itself in its very formulation—we do not ask “why are things the way they are?” from a feeling of comfort; the plaintiff cry of “how, then, shall we live?” does not come to us out of a sense of security. Indeed, insofar as truth overtakes us and interrupts the conventions of our lives, it occurs to us quite apart from our ordinary desires and wants. We are thus faced with a paradox: what claim can truth make on a being that “doesn’t need it and doesn’t care about it—since it doesn’t at all concern his needs”? When one considers that the awareness of truth is indexed to lived experience, the paradox is only heightened.

Abstract The tumor suppressor p53 is mutated (mt-p53) in over 50% of human cancers causing gain-of-function oncogenic effects, including metabolic changes that reduce tumor responsiveness to radio/chemotherapy. Common hot-spot mutations within the DNA-binding domain can be categorized as conformational (R175H) or DNA binding (R248W). NSC59984 has been characterized as a small molecule that targets mt-p53 for degradation and restores wt-p53 signaling. Using esophageal adenocarcinoma cells and CRISPR generated isogenic cell lines bearing matching hot-spot p53 mutations, we aim to understand how the molecular features of mt-p53 affect drug efficiency and enable the development of targeted therapies to limit cancer cell growth. We found that NSC59984 covalently modifies p53 by Michael addition at cysteine residues 124 and 229, which promote interactions that would stabilize the protein/DNA complex leading to increased p53 transcriptional activity. In cells, the effects of NSC59984 were substantially greater in cells harboring the R248W mutation compared with the R175H mutation. Treatment with NSC59984 reduced proliferation and increased apoptosis via the intrinsic mitochondrial pathway. It also induced changes in OXPHOS, ATP level, mitochondrial membrane potential, glycolysis, and lactate production. Furthermore, treatment of cells with NSC59984 increased reactive oxygen species production and decreased glutathione levels; effects were enhanced by the addition of buthionine sulfoximine and inhibited by N-acetyl cysteine. NSC59984 treatment increased G6PD activity, total NADPH levels, and expression of TIGAR. Knockout of TIGAR partially removed the antiproliferative effects of the drug and reduced G6PD levels in the p53-R248W cells. Incorporation of [13C6] into cellular metabolites suggests that p53-regulated transcription of TIGAR increased utilization of the pentose phosphate pathway and inhibited glycolysis at the fructose-6-P fructose-1,6-bisphosphate junction, supported by an increase in Hexokinase 2 and a decrease of phosphofructokinase-1. Thermal proteome profiling identified TIGAR as an additional reaction target of NSC59984, suggesting increased involvement in modulating these metabolic effects. Combining currently available therapeutic metabolic inhibitors with NSC59984 enhanced the antiproliferative effects in cells harboring p53-R248W creating a therapeutic window when compared to the wt-p53 expressing cells. This suggests these combinations could be used in a clinically relevant setting. Overall, this work has identified a distinctive mode of action for p53 reactivation resulting in not only transcriptional activity, but also a unique effect on cellular energetics. This study shows evidence of variation in responsiveness of different mt-p53 forms and allows the development of specific therapeutics directed to individuals for patient-centered precision medicine. Importantly, we have shown that targeting p53 signaling has significant effects on the metabolic profiles of cancer cells rendering them more vulnerable to neoadjuvant therapy.
