Full textAcetabular dysplasia causes articular instability and the reduction of the loading area in the hip. This causes the of articular function instability and the reduction of the loading area in the early onset of osteoarthritis which leads to pain and the eventual loss Acetabular dysplasia is commonly treated by periacetabular osteotomy, but the precise effects of this correction on intraarticular biomechanics is not yet well known. Using discrete element analysis, pressure distribution has been calculated in the dysplastic hips of 9 patients before and after periacetabular osteotomy. A variety of radiographic measurements and numerical metrics have been evaluated in order to q uantify of acetabular correction and excessive loading. the degree Correlations between these measures and metrics have been established in order to reveal any causal effects. correction s of the lateral centeredge angle, acetabular inde x and In particular, the femoral head extrusion index to are correlated to the rise of average contact area during gait (p 0.50).value < 0.05, R 2 = 0.22 Additionally, the influence of cartilage layer reconstruction method (population based healthy vs. dysplastic thicknesses) and loading parameters (healthy vs. dysplastic gait profiles) was significant for a majority of evaluated metrics. These results factors can help to assist with surgical planning , provide a basis for inand influencing vitro validation of surgical optimi zation acetabular dysplasia. and act as a standard for designing new orthopedic treatments for acetabular dysplasia.

Full textDog health constitutes a major concern for breeders, owners, as well as the general public, all the more since several study and reports have recently underlined potential impacts of breeding practices on dog health and fitness (APGAW 2009, Nicholas 2011). According to Online Mendelian Inheritance in Animals (OMIA, omia.angis.org.au) more than 586 disorders/traits have been reported in dogs, with various prevalence and consequences for canine health (Collins et al. 2011, Nicholas et al. 2011). As an exemple, Orthopedic Foundation for Animals (OFA 2011, http://www.offa.org) consider that Hip Dysplasia, a polygenetic trait affected by environmental factors, with variable impact on welfare, affects at least 163 dog breeds, with prevalence ranging from 1.2 to 72.1%. Implementation of breeding plans in order to reduce incidence of inherited disorders and their impact on welfare should be a priority for breeders and breeding organizations. Efficiency of such strategies is however highly dependant on several factors such as inheritance pattern, availability of efficient clinical/genetic test, and specific context conditions (prevalence, demography, existence of other disorders, cooperation of breeders…). For instance, it has been showed that for a monogenic recessive disorder with the same frequency, impact of a given strategy on genetic diversity will be completely different depending on the breed (Leroy and Rognon 2012). It is also important to underline that breeding context and breeding rules are very different according to countries. As an exemple, in Sweden the large proportion of pets insured (about 50%) allows the settlement of large surveys on dog health (Bonnett et al. 2005), leading to the identification of disorders critical to breed welfare. Depending on countries, the control of inherited disorders is implemented through various measures, from breeding recommandations to mating ban. In the case of hip dysplasia, a genetic evaluation system has been implemented in some countries (Germany, Sweden, UK) for a few breeds, while in some other countries, it is still under development. The fact that for many breeds there is an exchange of breeding animals between several countries with different breeding policies constitutes also a critical point to be taken into account, when settling a breeding strategy. Moreover it has been showed that efficiency of genetic evaluation for a polygenic trait such a hip dysplasia could be improved by joint evaluation between different countries (Fikse et al. 2012). For this purpose, a preliminary project, starting 2013 in Sweden, will investigate the interest of exchanging pedigree and health data within the framework of Nordic Kennel Union. The aim of this project is to provide operational tools to improve breed health in an international context, concerning both genetic evaluation and implementation of breeding policies

Full textThe goals of the study were to analyze and assess the motor function after hip arthroplasty according to the type of implants used with a clinical and radiological analysis, a biomechanical reconstruction study and functionals tests. The hip resurfacing group has a better stability and a stronger motor pattern than the conventionnal hip arthroplasty. According to the stabilometry datas, the postural coordination appears stable and regular whatever the level of constraints imposed. The proprioception test was developped and analysed according to two levels of measures : eyes opened or closed. The dynamic pattern applied during a vertical jump on the gravity center assessed the muscular power. The two groups of patients has significant differencies during hop test and proprioception test. In the hip resurfacing group, all the patients has done the tests. The motor comportment of the hip resurfacing patients was with very low outliers for the hop test. The hip arthroplasty group patients never reached a normal gait and functional motor situation. In the hip resurfacing group, the motor function, the gait walking datas and the proprioception results were equivalents of those observed in a control group

Full textOdonto-onycho-derma/ dysplasia is a rare autosomal recessive ectodermal dysplasia in which the presenting phenotype is dry hair, severe hypodontia, smooth depapillated tongue, onychodysplasia, keratoderma and hyperhidrosis of pa/ms and soles, and hyperkeratosis of the skin. Using a homozygosity mapping strategy, we assigned the disease locus to a 9 cMregion at chromosome 2q3S-q36. 2, located between markers rs168S3834 and D2S3S3 with a maximum multipoint LOD score of 5. 7, in 3 Lebanese consanguineous Muslim Shiite families. Using a candidate gene approach, we identify the same c. 697G> T (p. Glu233X) homozygous nonsense mutation in ail patients in exon 3 of the WNT10A gene. At the protein level, the mutation is predicted to result in a premature truncated protein of 232 aa instead of 417 aa. Finally we started functional studies to try to prove the implication of WnV/3 catenin pathway in this disease

Full textRationaleBronchopulmonary dysplasia is the most common chronic respiratory disease in prematureinfants. Genetic factors might contribute to bronchopulmonary dysplasia susceptibility.ObjectivesTo identify genetic variants involved in bronchopulmonary dysplasia through a genome-wideassociation study.MethodsWe prospectively evaluated 418 premature neonates (gestational age below 28 weeks), ofwhom 22% developed bronchopulmonary dysplasia. Two discovery series were created, usinga DNA pooling strategy in neonates from Caucasian and African ancestry. Polymorphismsassociated with the disease were confirmed in an independent replication population. Geneswere then explored by fine mapping and associations were replicated in an external Finnishpopulation of 213 neonates. Validated genes expression patterns were studied in rat lung,following air or hyperoxia exposure.Measurements and Main ResultsSPOCK2 gene was identified by both discovery series. The most significant polymorphism(rs1245560, p=1.66x10-7) was confirmed by individual genotyping, and in the replicationpopulation (p=0.002). Fine mapping confirmed the association of rs1245560 withbronchopulmonary dysplasia in both Caucasian and African populations with adjusted oddsratios of 2.96 (95% CI [1.37-6.40]) and 4.87 [1.88-12.63] respectively. In Caucasian neonates,rs1049269 was also associated with the disease (OR=3.21 [1.51-6.82]). These associationswere replicated in the Finnish population. In newborn rat lungs, SPOCK2 mRNA levelsmarkedly increased during the alveolar stage of lung development. After rat exposure tohyperoxia, SPOCK2 expression increased relative to air-exposed controls

Full textThe aim of this study is to elaborate a mechanical model of the healthy hip joint. The main objective is to use it as a reference model in order to analyse pathological hip joint mechanical behaviours and evaluate prescribed treatment by comparison of pre vs. Post simulated behaviours. Therefore, a generic finite element model of the hip joint is developed. Experimental acquisitions of kinematical and kinetic data are implemented in a musculo-skeletal model to calculate muscular forces acting at the hip joint. Our approach consists in coupling an inverse dynamic method with static optimisation. From the evaluation of each individual hip muscle force, the contact reaction force is computed. The contact reaction force is then implemented in the finite element model in order to perform simulations for several instants of the stance phase of gait and stair climbing. Contact pressure distributions during the stance phase of gait are compared to those available in the literature, measured in vivo by mean of instrumented prosthesis or cadaveric experiments simulating gait cycles. A clinical application concerns the evaluation of the effect of foot orthoses on the mechanical behaviour of an osteoarthritic hip joint. The loading conditions at the hip joint, when the patient is walking or climbing a step successively with and without foot orthoses, is determined in each experimental conditions using the approach previously described
