Abstract:
This thesis presents, in Part One, fifty poems in an unpublished collection entitled Hold Your Breath and, in Part Two, close readings of four poems published in Louis MacNeice’s final collection The Burning Perch. The introduction to Part Two provides a detailed analysis of particular aspects of MacNeice’s life and work that have informed the close readings of his poems, with special attention to his work in radio. This study of poems from his final collection will establish that, far from being detrimental as some commentators have argued, his radio work at the BBC had a positive influence on his poetry. Rather than acting in opposition, his work at the BBC and his writing of poetry benefited creatively from each other. By resisting any temptation to ‘pigeon-hole’ his work, and reading the poems with reference to the wider contexts in which he wrote, it is possible to gain a greater understanding and appreciation of MacNeice’s work and his ability to extend boundaries. The readings are a personal response which recognises what MacNeice himself would have regarded as the ‘impure’ nature of the poems. The title of the collection could serve as the title for both parts of the thesis, as it can be taken to express both an awareness of the fragility of life and the sense of excitement and anticipation that such an awareness brings to living. In essence it is this awareness that is explored in both the collection and the commentary.

Abstract:
Worldwide, more than 1,000,000 women develop breast cancer each year, and more than 400,000 die because of it. Basal-like breast cancers, which account for 8% to 20% of all breast cancer cases, represent the most aggressive of breast cancers with the majority resistant to existing targeted therapies. Accumulating lines of evidence implicate the Notch pathway in the aetiology of these basal-like breast cancers; while current work in our lab supports the notion that signalling through the Ectodermal Dysplasin Receptor (EDAR) pathway is also important. Notch signalling functions in normal development to control cell fate decisions and is mediated primarily, although not exclusively, through the CBF1 / RBP-Jĸ transcription factor. Aberrant Notch signalling leads to mammary tumourigenesis in mice; however at the outset of this work it was unclear whether signalling through RBP-Jĸ and / or through alternative pathways is required. This thesis presents novel data showing that elevated Notch signalling through the RBP-Jĸ-dependent pathway alone in murine mammary glands causes a number of developmental defects, including reduced ductal outgrowth, increased ductal side branching at puberty and, most significantly, is sufficient to induce mammary tumourigenesis. The data presented also provide supporting evidence that Notch signalling through RBP-Jĸ likely contributes to tumourigenesis, at least in part, via the suppression of apoptosis. At the outset of this thesis far less was known regarding the role(s) of the EDAR pathway within the mammary gland. Despite its recognised function in the development of ectodermal appendages it has been predominantly studied in the context of hair and tooth development. We show here that elevated Edar signalling affects the morphology of numerous ectodermally-derived glands, including the mammary gland, where in general, it results in glands that are enlarged or more elaborately branched. Most significantly, we show that elevated Edar signalling causes mammary tumourigenesis in mice, and provide data to support the hypothesis that elevated EDAR signalling might also be important in a subset of basal-like breast cancers in humans. The murine mammary gland phenotypes seen in response to elevated Edar signalling, including the squamous metaplasia within Edar-induced tumours, are very similar to those observed when Wnt signalling is increased. We provide data to support a positive correlation between activation of the EDAR and Wnt pathways in murine mammary tumourigenesis and provide data to support a comparable interaction in the aetiology of basal-like breast cancer showing squamous differentiation in humans. In summary, this thesis identifies the EDAR pathway as a novel potential therapeutic target in the treatment of a subset of basal-like breast cancers, and provides evidence that signalling through the RBP-Jĸ-dependent Notch pathway is sufficient to induce mammary tumourigenesis, most likely through the suppression of apoptosis.

Abstract:
As mães de recém-nascidos prematuros vivenciam a ansiedade oriunda da fragilidade e incerteza sobre as condições de vida de seu filho e a convivência com o estressante ambiente hospitalar, com implicações no aleitamento materno. Na tentativa de amenizar esta situação, o objetivo deste estudo foi avaliar o efeito da acupuntura na ansiedade de mães de recém-nascidos de muito baixo peso (RNMBP). Trata-se de um ensaio clínico randomizado realizado em um hospital universitário do norte do Paraná. A pesquisa foi aprovada pelo Comitê de Ética e Pesquisa da instituição na qual foi realizado o estudo e foi submetida a registro em base de dados para ensaios clínicos Australian New Zealand Clinical Trials Registry. Todas as participantes assinaram o Termo de Consentimento Livre e Esclarecido. A coleta de dados ocorreu no Banco de Leite Humano da instituição, entre agosto de 2011 e novembro de 2012. As participantes foram alocadas em dois grupos: grupo acupuntura (GA) e grupo placebo (GP) e receberam sessões semanais de acupuntura auricular chinesa nos pontos Shenmen, Tensão, Ansiedade 1 e 2 e Relaxamento Muscular. A acupuntura placebo foi realizada com agulhas auriculares adaptadas, sem perfurar a pele. Houve cegamento entre as participantes e o estatístico que realizou a análise dos dados. Utilizaram-se o Inventário de Ansiedade Traço-Estado e a mensuração do cortisol salivar, antes e após a intervenção. Participaram do estudo 29 mães de RNMBP, 14 pertencentes ao GA e 15 pertencentes ao GP. A idade das participantes variou entre 16 e 40 anos, com mediana de 27 anos. A maioria tinha escolaridade até o ensino médio (57%), vivia com o companheiro (72%) e possuía trabalho remunerado (65%). Quanto às variáveis de aleitamento materno, 48% relataram experiência prévia com amamentação, sendo que 13% apresentaram intercorrências durante este período. A incidência de aleitamento materno exclusivo entre as participantes, na data da alta do recém-nascido, foi de 76%, nove pertencentes ao GA (64%) e 13, ao GP (87%), sem relevância estatística entre estes valores (p=0,215). Os resultados para o cortisol salivar, antes da intervenção, variaram entre 0,02 e 0,33 ug/dL para o total de participantes, com média de 0,13 para ambos os grupos (DP=0,08). Após a intervenção, ambos os grupos apresentaram média de 0,14 ug/dL (DP=0,12), variando entre 0,02 e 0,49 ug/dL. A diferença média entre as medidas de cortisol salivar, antes e após a intervenção, foi de -0,02 ug/dL (DP=0,11), mostrando que os níveis de cortisol aumentaram após a intervenção em ambos os grupos, sem significância estatística (p=0,480), e contrariando o esperado. Após a intervenção, o escore de ansiedade-estado das participantes teve redução média de oito pontos, não havendo diferença estatística entre os grupos (p=0,888), com a possibilidade de que tanto a acupuntura real quanto a placebo tenham produzido resultados positivos na redução da ansiedade. Sugere-se que estudos com maior amostra e um terceiro grupo controle sejam conduzidos para melhor esclarecer os efeitos da acupuntura para a redução da ansiedade nesta população.<br>Mothers of preterm infants experience anxiety due to the child\'s frail and uncertain health condition. Spending time in a stressful hospital environment has implications to lactation and breastfeeding. In the attempt to diminish this situation, the aim of this study was to assess the effect of acupuncture in mothers with very low birth weight infants (VLBW). This is a randomized, patient-assessor blind, sham-controlled clinical trial, conducted in a school hospital in northern Paraná. This research has been approved by the ethics review board of the institution where it took place. The trial is registered at Australian New Zealand Clinical Trials Registry, number 12611000025932. All participants signed the written informed consent. Data was collected at the institution\'s Human Milk Bank from August 2011 to November 2012. Subjects were allocated into two groups: acupuncture - AG or placebo-acupuncture - PG and treatment sessions occurred once a week. The Chinese ear acupoints used were Shenmen, Tension, Anxiety 1 and 2, Muscle relaxation. For placebo- acupuncture, custom-designed noninvasive ear needles were used. Participants were submitted to the STAI-Y scale and salivary cortisol measurements before and after treatment. Twenty-nine mother\'s with VLBW infants took part in the study (GA = 14, GP = 15). Participants\' age ranged from 16 to 40, median age was 27 years old. Most subjects had high school educational levels (57%); lived with their partners (72%) and had paid jobs (65%). 48% mothers had previous experiences with breastfeeding and 13% mentioned some problems while doing so. Exclusive breastfeeding rate at infant\'s hospital discharge was 76%, AG = 9 (64%), PG = 13 (87%), not significant (p=0,215). Salivary cortisol levels before intervention ranged from 0,02 to 0,33 ug/dL and both groups presented mean 0,13 ug/dL (SD = 0,08). After treatment both groups had 0,14 ug/dL (SD = 0,12) mean levels, with a 0,02 - 0,49 ug/dL range. Mean difference between salivary cortisol levels before and after treatment was -0,02 ug/dL (SD = 0,11), showing increase in cortisol levels after treatment for both groups, not significant and contrary to expected. After treatment, mean STAI-Y1 (state) scores dropped 8 points, not significant between groups (p=0,888). This result indicate that both real and placebo acupuncture may have produced positive effects in anxiety reduction. We suggest other studies are conducted with bigger sample sizes and that include a third arm with routine care as control group to better understand the effects of acupuncture for anxiety in this population.

Abstract:
This study investigates the values expressed and implemented through local cereal and cereal-related products such as bread and flour in the alpine region of Valle d’Aosta (north-west Italy), contributing to the existing body of literature on food values. It is based on anthropological fieldwork among people engaging with cereal both professionally and non-professionally (such as bakers, farmers, agronomists and other categories of people involved in the cereal sector) and on theories drawn from food and economic anthropology, anthropological theories of value and literature on social movements. This research aims at understanding the values that inform cereal-related practices in Valle d’Aosta and that precede the relationships its inhabitants generate around cereal. Such values are intended as moral standpoints from which people engaging with cereal organise their action and conceptualise their own understanding of their practices. Values of tradition, community and individual place identity, health, environmental and socio-economic values serve as spectacles through which to grasp the vision that people engaging with cereal in Valle d’Aosta have of society, of the role of the economy, of the relationship between the community and the individual. Ultimately, cereal-related practices, based on a particular conception of the economy which puts into question the neoliberal system, are represented as tools bridging past, present and future, as the past serves as a source of inspiration to bring about a better future and to materialise it into the present, through a deeply moral endeavour.

Doctor of Philosophy<br>Department of Diagnostic Medicine/Pathobiology<br>Douglas Powell<br>Randall Phebus<br>Not-ready-to-eat entrées purchased at retail and prepared in the home have been identified as a risk factor for salmonellosis. From 1998 to 2010, ten outbreaks implicated undercooked not-ready-to-eat entrées. In each outbreak, affected individuals prepared entrées in a microwave oven, did not follow recommended cooking instructions, and failed to take the internal temperature of the cooked product. This dissertation surveyed grocery stores for product availability, evaluated consumers’ preparation practices of raw, breaded, frozen chicken entrées, and validated on-package label instructions. The survey of retail revealed that several manufacturers fail to provide consumers clear preparation instructions. A video capture system was used to observe food preparation practices of 41 consumers–21 primary meal preparers and 20 adolescents–in a mock domestic kitchen using uncooked, frozen, breaded chicken products, and determined if differences exist between consumers’ reported safe food handling practices and actual food handling behavior as prescribed on product labels. Differences between self-report and observed food safety behaviors were identified between groups. Many participants reported owning a food thermometer (73 percent) and reported using one when cooking raw, breaded chicken entrées (19.5 percent); however, only five participants (12.2 percent) were observed measuring the final internal temperature with a food thermometer despite instructions on the product packaging to do so. Food handling errors identified during the meal preparation sessions were then mimicked in a controlled laboratory setting to determine the impact of such deviations on end-product temperature. For all products, highly variable internal temperatures were recorded across entrées when prepared in a 600W microwave oven. Microwave cooking of raw breaded poultry products is unpredictable in achieving uniform target end-point temperatures; however, a 1000W microwave oven consistently produced a safe end product. Data collected through direct observation more accurately reflected consumer food handling behaviors than data collected through self-reported surveys. Low wattage microwave ovens failed to produce a safe end product. Processors should validate instructions for not-ready-eat entrées using a range of microwave ovens rather than a single wattage, develop a unique set of instructions for entrées, and provide consumers clear cooking instructions that result in a safe end product.

Em produtos cárneos, além de atuar sobre aspectos sensoriais e de textura, o sal desempenha papel chave sobre sua estabilidade microbiológica. Uma simples redução pode resultar em produtos instáveis durante sua vida de prateleira. Logo, para que estratégias de redução de sal sejam implementadas de maneira segura, faz-se necessário a aplicação de tecnologias ou agentes de preservação complementares. Tecnologias emergentes não-térmicas de inativação microbiana como o processamento à Alta Pressão (HPP - High Pressure Processing) surgem como opções factíveis para tal propósito, eliminando a dependência de conservantes químicos em face à redução de sal. A adição de agentes antimicrobianos naturais como barreiras adicionais à HPP (Tecnologia de Múltiplas Barreiras), visando potencializar os efeitos conservadores, têm sido apontada como uma promissora tendência na literatura moderna. Diante deste exposto, após determinação da formulação de trabalho com teor reduzido de sódio, a pesquisa objetivou ajustar as variáveis de processo "carga de pressão" e "tempo de residência" (utilizando baixas temperaturas de 25°C) para o processamento de embutido de peito peru fatiado embalado a vácuo (tipo "Blanquet"). Foram avaliadas respostas de inativação microbiana pós-processo (reduções logarítmicas) para populações inoculadas do patógeno Listeria monocytogenes e bactérias ácido láticas deterioradoras incluindo as espécies Leuconostoc mesenteroides e Lactobacillus sakei; também foram avaliados os atributos de qualidade incluindo cor, atividade de água, pH, sinerese, textura e oxidação lipídica. Em adição, visando à utilização de tecnologia de múltiplas barreiras para conservação, foi estudado o efeito da adição do componente bioativo carvacrol em concentrações de 200ppm, na formulação do produto. Empregando estratégia de simples redução, sem a adição sais substitutos, após avaliação de aspectos físico-químicos, sensoriais e microbiológicos, concluiu-se que níveis de 30% de redução de sal (cerca de 25% menos sódio) sobre o controle formulado com 20g/kg, são viáveis; entretanto, foi evidenciada redução da estabilidade microbiológica. Após avaliação de diferentes faixas de pressão e tempo de processo, seguindo os critérios exigidos de desempenho para inativação pós-processo de Listeria monocytogenes (4-5 reduções logarítmicas), um valor de 600MPa/180 segundos a 25°C se apresentou como um tratamento adequado para o produto com baixo teor de sódio estudado, promovendo reduções logarítmicas eficazes contra o patógeno-alvo estudado e a microbiota deteriorante. Alterações instrumentais (p<0.05) em alguns dos atributos de qualidade avaliados puderam ser destacadas principalmente sinerese, oxidação lipídica e textura; porém são necessários estudos mais amplos de análise sensorial. Adicionalmente, dados apresentados demonstraram que os consumidores não foram capazes de diferir o produto low-sodium processado por alta pressão isostática (600MPa/180sec/25°C) de um não pressurizado; de fato, apesar dos efeitos instrumentais significativos, estas alterações parecem não ser identificadas pelos consumidores. A adição de uma barreira natural complementar, o carvacrol, em níveis sensorialmente aceitáveis, foi capaz de potencializar os efeitos de inativação pós-processo (reduções logarítimicas) dos grupos microbianos alvo; efeitos benéficos combinados ao longo da estocagem refrigerada do produto também foram evidenciados por meio de reduzidas taxas de crescimento e fase lag aumentada, maximizando assim o potencial de conservação. A utilização combinada de HPP e carvacrol representou uma alternativa promissora contra fenômenos de injúrias sub-letais e recuperação celular. Sugere-se que a intensidade do processo HPP (em termos de carga de pressão, tempo de processo e temperatura) necessária para inativar microrganismos em níveis requeridos, possa ser reduzida na presença de barreiras antimicrobianas adicionais, garantindo a qualidade total do produto processado em condições mais amenas. Em adição, uma série de vantagens industriais podem ser destacadas tais como a redução de custos para a instalação de equipamento inicial e de manutenção (equipamentos operando em cargas mais baixas); e maximização da produção de processamento por ciclos efetivamente encurtados (ciclos de produtividade mais elevados por hora)<br>Abstract: The reduction of sodium chloride in processed food formulations represents a great challenge for modern industry, considering the relationship established between high sodium intake and the occurrence of arterial hypertension, the major risk factor associated with the occurrence of cardiovascular diseases and strokes. In meat products, in addition to acting on sensory quality and texture, salt plays a key role on the microbiological stability. A simple reduction can result in unstable products during their shelf-life. Therefore, for the success of salt reduction strategies implemented safely, it is necessary the application of complementary technologies or alternative preservatives. Non-thermal emerging technologies for microbial inactivation, such as High Pressure Processing (HPP), appear as feasible alternative for this purpose, eliminating the dependence of chemical preservatives considering the reduction of salt barrier. The addition of natural antimicrobial agents as additional hurdles (Multi Hurdle Technology) to the HPP, aiming to enhance the preservative effects, has been identified as a promising trend in modern literature. Given this exposed, after determination of low-sodium working formulation, this research aimed adjust the processing variables "pressure load" and "dwell time" (using low-processing temperatures of 25°C) for sliced vacuum-packaged ready-to-eat turkey breast. Post-processing microbial inactivation responses were evaluated (by logarithmic reductions) against inoculated populations of the target pathogen Listeria monocytogenes and spoilage lactic acid bacteria including Leuconostoc mesenteroides and Lactobacillus sakei; were also evaluated quality-attributes including color, water activity, pH, syneresis, texture and lipid oxidation. In addition, in order to use multiple barriers for preservation technology, was studied the effect of adding bioactive component, carvacrol at concentrations of 200ppm in the product formulation. Employing a simple reduction strategy, without adding salt substitutes, after evaluation of physicochemical aspects, sensory and microbiological, it was concluded that levels of 30% NaCl reduction (aproximatelly 25% less sodium) on the control made with 20g/kg, are viable; however, problems with microbiological stability were evidenced. After evaluation of different pressure ranges and processing time, following the performance criteria required for post-inactivation process Listeria monocytogenes (4-5 log reduction), a value of 600MPa/180 seconds at 25°C was presented with an adequate treatment for studied product with low sodium content, promoting effective logarithmic reductions against the studied target pathogen and spoilage LAB. Instrumental changes (p<0.05) in some of the evaluated quality attributes could be revealed mainly in syneresis, lipid oxidation and texture; however, a sensory confirmation needs to be established. Additionally, data presented showed that consumers have not been able to differ low-sodium products processed by HPP (600MPa/180sec/25°C) and unpressurised; in fact, despite significant instrumental effects, these changes seems to be not identified by consumers. The addition of natural barrier, such as carvacrol at acceptable sensory levels (200ppm), was able to potentiate the effects of post-process inactivation (logarithmic reductions) of the target microbial groups; combined benefit effects along the refrigerated product storage were also evidenced by reduced growth rates and increased lag phases, thus maximizing their potential for conservation. The combination of HPP and carvacrol represented a promising weapon against phenomena of sub-lethal injuries and cell recovery. It is suggested that the intensity of the HPP process (in terms of pressure load, process time and temperature) required to inactivate microorganisms in required levels, may be reduced in the presence of additional microbial barriers, ensuring the overall quality of the processed product under mild processing conditions.
